Objective:

To establish a laser diffraction method for the determination of particle size distribution of budesonide suspension for inhalation.

Methods:

The Master sizer 3000 laser size analyzer with the Hydro SV wet sampler was used. The sample solution should be immediately measured after ultrasonication. The measurement conditions were as follows:refractive index of budesonide was 1.592,absorbency of budesonide was 0.01,dispersing medium was saturated solution of budesonide containing 0.5% Tween 80 with refractive index of 1.33,stirring rate was 1 000 r·min-1,and laser obscurations were 6%-12%.

Results:

RSD of D50 was lower than 2%,RSDs of D10 and D90 were both lower than 5%.The particle size distribution D90 values of 3 batches of budesonide suspension for inhalation were less than 5 μm.

Conclusion:

The method is simple,accurate and producible,which is suitable for the particle size detection of budesonide suspension for inhalation.

建立激光衍射法用于测定吸入用布地奈德混悬液的粒度分布。

方法:

采用Mastersizer 3000激光粒度分析仪和Hydro SV型湿法进样器,样品超声混匀后立即注入样品池,折射率为1.592,吸收率为0.01,分散介质为0.5%吐温-80的布地奈德饱和溶液,其折射率为1.33,搅拌速率为1 000 r·min-1,遮光度为6%~12%。

结果:

方法学精密度考察结果D50的RSD小于2.0%,D10D90的RSD均小于5.0%;3批吸入用布地奈德混悬液粒度分布的D90均小于5 μm。

结论:

该方法操作简便,准确度高,重复性好,适用于吸入用布地奈德混悬液的粒度分布测定。

目的:

建立激光衍射法用于测定吸入用布地奈德混悬液的粒度分布。

方法:

采用Mastersizer 3000激光粒度分析仪和Hydro SV型湿法进样器,样品超声混匀后立即注入样品池,折射率为1.592,吸收率为0.01,分散介质为0.5%吐温-80的布地奈德饱和溶液,其折射率为1.33,搅拌速率为1 000 r·min-1,遮光度为6%~12%。

结果:

方法学精密度考察结果D50的RSD小于2.0%,D10D90的RSD均小于5.0%;3批吸入用布地奈德混悬液粒度分布的D90均小于5 μm。

结论:

该方法操作简便,准确度高,重复性好,适用于吸入用布地奈德混悬液的粒度分布测定。

Objective:

To establish a laser diffraction method for the determination of particle size distribution of budesonide suspension for inhalation.

Methods:

The Master sizer 3000 laser size analyzer with the Hydro SV wet sampler was used. The sample solution should be immediately measured after ultrasonication. The measurement conditions were as follows:refractive index of budesonide was 1.592,absorbency of budesonide was 0.01,dispersing medium was saturated solution of budesonide containing 0.5% Tween 80 with refractive index of 1.33,stirring rate was 1 000 r·min-1,and laser obscurations were 6%-12%.

Results:

RSD of D50 was lower than 2%,RSDs of D10 and D90 were both lower than 5%.The particle size distribution D90 values of 3 batches of budesonide suspension for inhalation were less than 5 μm.

Conclusion:

The method is simple,accurate and producible,which is suitable for the particle size detection of budesonide suspension for inhalation.

布地奈德为糖皮质激素,通过局部抗炎作用治疗哮喘。临床上吸入用布地奈德混悬液经雾化器雾化后,患者经潮式呼吸吸入,主要用于小儿哮喘的维持治疗及预防[1-4]。吸入用布地奈德混悬液的原料药不溶于水,以微粒状态分散于液体介质中,因此,粒度和粒度分布是吸入用布地奈德混悬液的关键质量属性,关系到本品的安全性和有效性。国内外对药品的粒度控制有相关要求,如ICH Q6A提出对于固体制剂或含不溶性药物的液体制剂,若原料药的粒度对溶解度或生物利用度等有关键影响,均需制订粒度标准[5];2020年版《中华人民共和国药典》(简称《中国药典》)四部0111吸入制剂项下规定,吸入制剂中原料药物粒度大小通常应控制在10 μm以下,其中大多数应在5 μm以下[6]12。因此,有必要对吸入用布地奈德混悬液的粒度分布进行研究。
粒度分析方法主要有筛分法、显微镜法、激光衍射法、电阻法(库尔特计数法)等[7]。2020年版《中国药典》四部0982粒度和粒度分布测定法收载了3种方法,一法为显微镜法,二法为筛分法,三法为光散射法(激光衍射法)[6]145。吸入用布地奈德混悬液的原研药为AstraZeneca公司产品,执行标准(JX20180163)发布较早,采用库尔特计数器(电阻法)测定粒度,该方法在2020年版《中国药典》四部通则“0982粒度和粒度分布法”中暂未收载。BP 1973收载了电阻法,该方法分辨率高,重复性较好,但动态范围较窄,测量下限不够小,且容易发生堵孔故障[7]。与显微镜法、筛分法相比,激光衍射法具有测试精度高,速度快,重复性好,动态范围宽等优点,目前已被用于原料药及制剂的粒度分布研究[8-10]。本研究拟基于激光衍射法建立吸入用布地奈德混悬液的粒度分布测定方法。
本研究对不同的分散介质、光学参数和分散条件进行全面考察,建立了测定吸入用布地奈德混悬液粒度分布的激光衍射法,并对建立的方法进行验证。本方法简单快速,重现性良好,适用于吸入用布地奈德混悬液粒度分布的测定,也可为其他混悬型药物的粒度分布测定方法的建立及验证提供参考。
Malvern Mastersizer 3000型激光粒度分析仪、Hydro SV型湿法进样器、Mastersizer 3000工作站,Malvern公司;KQ-300DE型数控超声仪,昆山市超声仪器有限公司。
吸入用布地奈德混悬液(批号 KA22186、KA23041、KA23056)、布地奈德原料药(批号 22053002),四川普瑞特药业有限公司;吐温-80(批号20180119),天津市美琳工贸有限公司;聚乙二醇辛基苯基醚(Triton X-100,批号20190900301)、氯化钠(批号2022051702),成都市科隆化学品有限公司;水为纯化水。
将布地奈德原料药约50 mg,缓缓加入500 mL 0.5%吐温-80溶液中,边加边搅拌约12 h使饱和,用0.22 μm滤膜滤过,得布地奈德饱和溶液,以其为分散介质,搅拌速率1 000 r·min-1,搅拌时间2 min作为分散条件;样品折射率为1.592,样品吸收率为0.01;分散介质折射率为1.33;分析模式为通用模式;测量次数为3;样品测量时间为10 s;背景测量时间为10 s。将分散介质缓慢加入样品池,避免产生气泡,照粒度和粒度分布测定法(2020年版《中国药典》四部通则0982第三法)(Malvern Master Sizer 3000,Hydro SV型湿法进样器)测量背景。取本品3支,充分振摇后取全部内容物,超声5 min使混匀,作为供试品溶液,取供试品溶液适量立即加入样品池,使检测器遮光度在6%~12%,依法测定。采用Mastersizer 3000型工作站采集数据并分析。
取布地奈德原料药,缓缓加入含不同表面活性剂的水溶液中,边加边搅拌约12 h使饱和,用0.22 μm滤膜滤过,配制布地奈德饱和溶液作为分散介质。配制的分散介质见表1。取本品适量,按“2.1”项下方法进行测定,统计3次结果的D10D50D90、残差的平均值。结果见表1
在其他参数一致的测定条件下,采用Triton X-100为表面活性剂的介质1(原研标准中的分散介质)与采用吐温-80为表面活性剂的介质3,D10D50D90和残差的平均值无明显区别,但Triton X-100的细胞通透性强,根据全球化学品统一分类和标签制度(GHS)危险性分类[11-12],Triton X-100的安全技术说明书指出其具有严重眼损伤的危险,且对水生环境具有长期危险,因此在实验数据无明显区别的前提下,确定吐温-80为分散介质的表面活性剂。
采用含氯化钠和吐温-80为分散介质的介质3与采用不含氯化钠仅使用吐温-80为分散介质的介质4,测定结果无差别。原研标准采用的电阻法要求待测样品含一定的电解质,且达到规定的电阻率才能进行测定,原研标准中的氯化钠可能是为了满足电阻法测定需求而加入的电解质。本研究确定采用不含氯化钠的吐温-80为分散介质,更为简单便捷。
采用含0.2%~2.0%的吐温-80为分散介质,D10D50D90、残差的平均值并无明显变化,但吐温-80的浓度为2.0%的介质7,通过滤膜的速度很慢,因此不宜采用。经观察,吐温-80浓度越大,产生的气泡越多,且越不容易消散,最终确定0.5%吐温-80的布地奈德饱和溶液作为分散介质,该浓度耐用性好,对样品的分散好,产生的气泡较少。
本研究采用激光衍射粒度分析法的湿法模式进行测定,需要设定的光学参数:分散介质的折射率、样品的折射率和吸收率。分散介质中的表面活性剂和布地奈德的量很低,折射率与水相当,为1.33。布地奈德的折射率和吸收率目前没有准确的值,需进行考察,对拟合曲线的形状、残差大小等方面综合考虑,从而确定参数值。其中,残差是指基于光学参数,理论光强数据与实际测量光强的最小二乘拟合差值,残差越小,表示测定结果越接近真实状态[13-15]
对不同数据库提供的布地奈德折射率(1.459、1.520、1.592)进行考察,样品吸收率均设为0.01。结果(表2图1)显示:样品折射率对粒度分布曲线的拟合形态有较大影响,当折射率分别为1.459和1.520时,粒度分布的拟合曲线均未呈正态分布,表明以上参数不适用;当折射率为1.592时,粒度分布的拟合曲线接近正态分布,残差小于1,最终确定样品折射率为1.592。
对未知光学参数的样品进行湿法方法的粒度测定,吸收率可以根据颗粒的材质、颜色来做近似判断,并根据拟合曲线进行修正,如黑色样品为1+,透明到白色样品为0~0.01,浅色至深色样品为0.1~1,该估算值仅需精确到10的倍数。本实验分别选择吸收率值为0、0.001、0.01和0.1进行考察。结果显示,吸收率为0.1时,粒度分布的拟合曲线未呈正态分布;吸收率为0、0.001、0.01时,粒度分布的拟合曲线均接近正态分布,D10D50D90、残差均接近,结合本品为白色细微颗粒的混悬液,最终确定样品吸收率为0. 01。结果见表2图2
样品的代表性和分散程度决定了激光粒度法测试结果的准确度和重复性[16-17]。对样品前处理过程中的超声时间以及样品的遮光度、搅拌速率和搅拌时间进行考察,确定分散条件。
吸入用布地奈德混悬液非常易沉降,取样需要代表性才能确保结果的准确性。取本品3支,充分振摇后取全部内容物,进行超声混匀时间的考察,在其他参数一致的情况下,分别考察超声2、5、10 min后样品的D10D50D90平均值及RSD,结果(表3)各超声时间下测定结果无显著差异,。在考察的时间范围内超声不会对粒径测定结果造成影响,最终确定超声时间为5 min。
遮光度反映了样品浓度,考察遮光度为3%、6%、9%、12%、15%、18%时的D10D50D90、残差的平均值及RSD。遮光度为6%~12%时,D10D50D90无明显变化,残差均小于1%;遮光度为3%时,残差略大,可能是因为加入样品量较少,粒子的衍射光强较弱,检测信号强度小且不稳定,理论光强数据与实际测量光强数据相差太大,导致表征测量结果和理论值拟合差值的残差变大[18];遮光度为15%时,残差略大;遮光度增至18%后,D10D50D90降低,残差增大,可能是因为加入样品量较大,产生了多重散射,前段的小颗粒比例明显增强(图3)。样品量过低或过高均会影响结果的准确性。最终确定遮光度应在6%~12%范围,在该范围内的测定结果一致。结果见表4
样品加入分散介质后,分别考察了500、800、1 000、1 200、1 500 r·min-1搅拌速率下的D10D50D90,以及搅拌0.5、2、5 min后的D10D50D90,结果基本无差异。为保证测定条件具有一定的耐用性,最终确定搅拌速率1 000 r·min-1和搅拌时间2 min作为试验条件。
由2名分析人员分别平行制备6份样品,采用“2.1”项下方法进行测定。D10D50D90的平均值及RSD见表5。2名分析人员分析测定6份样品,D50的RSD均<2%,D10D90的RSD均<5%;2名分析人员测定12份样品,D50的RSD均<2%,D10D90的RSD均<5%,满足USP<429>的相关要求(D50小于10%,D10D90小于15%),表明该方法精密度良好。
取本品适量,按“2.1”项下方法制备供试品溶液,分别放置0(立即进样)、5、10、20 min,加入样品池进行测定,结果D10D50D90均随放置时间的增加而降低,见图4。表明随着供试品溶液放置时间的延长,药物颗粒会发生沉降。建议在测定过程中,供试品溶液应临用现配,即待分散体系稳定后,再制备供试品溶液,并立即加入样品池进行分析测定。
采用“2.1”项下方法,对3批样品进行测定,统计各批次样品的D10D50D90平均值,结果见表6。3批样品的D90均在5 μm以下,符合2020年版《中国药典》四部0111吸入制剂的一般要求。
布地奈德在水中几乎不溶,首选水为分散介质,但样品在纯水中容易发生团聚现象,不易分散均匀,因此需要在水中加入表面活性剂,以使样品能够均匀分散在介质中。各种表面活性剂对布地奈德在水中的溶解均有不同程度的促进作用,仅含表面活性剂的分散介质(不加布地奈德进行饱和),可能由于原料药不同程度的溶解导致无法评价各介质的真实分散效果,也无法反映样品的真实粒度分布情况。因此,需配制含表面活性剂的布地奈德饱和溶液作为分散介质,以使布地奈德能够均匀分散在介质中的同时不再发生溶解。含表面活性剂的原料药饱和溶液同时解决了样品的分散问题和溶解问题,尤其适用于含不溶性药物的液体制剂的粒度测定,可以作为混悬液粒度检测的优选分散介质。
ISO13320:2020引入了质量源于设计(quality by design,QbD)的理念指导粒度方法开发,绘制了完整的粒度分析方法的影响因素风险评估图,影响因素包括分析、环境、材料、仪器、方法等变量[19]。其中,取样方法是分析变量中的重要影响因素,取样的科学合理性、代表性直接影响检验结果的真实性[20]。混悬液是难溶性固体原料药物分散在液体介质中制成的两相制剂,在放置过程中均会发生不同程度的沉降,也可能产生絮凝现象,如何合理且有代表性地取样关系着结果的真实性。吸入用布地奈德混悬液性状为:细微颗粒混悬液,静置后有细微颗粒沉淀,振摇后呈白色或类白色混悬液。说明书中详细列出使用步骤,包括开盖前振摇,药液不能部分使用,需全部用尽等。注意事项也明确:应按规定方法振摇,重复至少3次,直至内容物完全混合均匀。故此,测定方法中取样前的“充分振摇后取全部内容物”对本品的粒度检测至关重要,关系着取样的代表性。本研究采取“超声5 min”的前处理方式,也是为了破除混悬剂的絮凝现象,同时使样品分散更为均匀。样品稳定性考察结果也表明,供试品溶液制备后需立即进样,否则供试品溶液静置后即发生沉降,有细微颗粒沉淀,与样品的性状描述一致。本研究也提示混悬液粒度测定方法开发中应考虑样品中的药物沉降问题,以保证取样的代表性。
