Objective:

To establish an HPLC fingerprint of Zuoyu’an San and conduct quality control by combining chemometrics and quantitative analysis of multi-components with a single-marker(QAMS).

Methods:

HPLC was used to establish the fingerprint spectra of 12 batches of Zuoyu’an San and perform similarity evaluation. Cluster analysis (CA),principal component analysis (PCA),and orthogonal partial least squares discriminant analysis (OPLS-DA) were combined to perform chemical pattern recognition analysis on different batches of Zuoyu’an San. At the same time,QAMS method was established using rhein as the internal standard reference. The relative correction factors between the internal reference and jatrorrhizine,palmatine,berberine,aloe-emodin,emodin,chrysophanol,and physcion were established. The external standard method (ESM) and QAMS method were used to determine jatrorrhizine,palmatine,berberine,aloe-emodin,rhein,emodin,chrysophanol,and physcion in Zuoyu’an San,respectively. To verify the reliability of QAMS metod results,the contents of 8 components of physcion were determined.

Results:

A total of 23 common peaks were calibrated in the fingerprint,and 8 components were identified by comparing with the mixed reference solution. The similarity between the samples and the control spectrum was between 0.952 and 0.994;CA divided 12 batches of Zuoyu’an San samples into 3 categories;PCA showed that there were certain differences in the chemical composition of different batches of Zuoyu’an San,and extracted 5 principal components that affected the quality evaluation of Zuoyu’an San;OPLS-DA screened out 13 potential marker components (VIP>1) among 23 common peaks that caused quality differences between different batches of Zuoyu’an San samples;There was no significant difference between the QAMS method calculated value and the ESM measured value.

Conclusion:

The established HPLC fingerprint and multi-index component determination method was accurate and simple,and combined with chemometric methods,could provide reference for the quality control and evaluation of Zuoyu’an San.

目的:

建立坐浴安散的HPLC指纹图谱,并结合化学计量学和一测多评法开展坐浴安散质量控制研究。

方法:

采用HPLC法建立12批坐浴安散的指纹图谱并进行相似度评价,结合聚类分析(CA)、主成分分析(PCA)、正交偏最小二乘-判别分析(OPLS-DA),对不同批次坐浴安散进行化学模式识别分析。同时以大黄酸为内标参照物建立一测多评(QAMS)法,建立内参物与药根碱、巴马汀、小檗碱、芦荟大黄素、大黄素、大黄酚、大黄素甲醚的相对校正因子,采用外标法(ESM)与QAMS法分别测定坐浴安散中药根碱、巴马汀、小檗碱、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚8个成分的含量,以验证QAMS结果的可靠性。

结果:

指纹图谱共标定23个共有峰,通过与混合对照品溶液比对指认出8个成分,样品与对照图谱相似度在0.952~0.994;CA将12批坐浴安散样品分为3类;PCA显示不同批次的坐浴安散化学成分存在一定的差异,并提取影响坐浴安散质量评价的主成分5个;OPLS-DA在23个共有峰中筛选出13个导致不同批次坐浴安散样品间质量差异的潜在标志成分(VIP>1);QAMS计算值与ESM测定值无明显差异。

结论:

建立的HPLC指纹图谱及多指标成分测定方法准确、简便,结合化学计量学方法可为坐浴安散的质量控制与评价提供参考。

目的:

建立坐浴安散的HPLC指纹图谱,并结合化学计量学和一测多评法开展坐浴安散质量控制研究。

方法:

采用HPLC法建立12批坐浴安散的指纹图谱并进行相似度评价,结合聚类分析(CA)、主成分分析(PCA)、正交偏最小二乘-判别分析(OPLS-DA),对不同批次坐浴安散进行化学模式识别分析。同时以大黄酸为内标参照物建立一测多评(QAMS)法,建立内参物与药根碱、巴马汀、小檗碱、芦荟大黄素、大黄素、大黄酚、大黄素甲醚的相对校正因子,采用外标法(ESM)与QAMS法分别测定坐浴安散中药根碱、巴马汀、小檗碱、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚8个成分的含量,以验证QAMS结果的可靠性。

结果:

指纹图谱共标定23个共有峰,通过与混合对照品溶液比对指认出8个成分,样品与对照图谱相似度在0.952~0.994;CA将12批坐浴安散样品分为3类;PCA显示不同批次的坐浴安散化学成分存在一定的差异,并提取影响坐浴安散质量评价的主成分5个;OPLS-DA在23个共有峰中筛选出13个导致不同批次坐浴安散样品间质量差异的潜在标志成分(VIP>1);QAMS计算值与ESM测定值无明显差异。

结论:

建立的HPLC指纹图谱及多指标成分测定方法准确、简便,结合化学计量学方法可为坐浴安散的质量控制与评价提供参考。

Objective:

To establish an HPLC fingerprint of Zuoyu’an San and conduct quality control by combining chemometrics and quantitative analysis of multi-components with a single-marker(QAMS).

Methods:

HPLC was used to establish the fingerprint spectra of 12 batches of Zuoyu’an San and perform similarity evaluation. Cluster analysis (CA),principal component analysis (PCA),and orthogonal partial least squares discriminant analysis (OPLS-DA) were combined to perform chemical pattern recognition analysis on different batches of Zuoyu’an San. At the same time,QAMS method was established using rhein as the internal standard reference. The relative correction factors between the internal reference and jatrorrhizine,palmatine,berberine,aloe-emodin,emodin,chrysophanol,and physcion were established. The external standard method (ESM) and QAMS method were used to determine jatrorrhizine,palmatine,berberine,aloe-emodin,rhein,emodin,chrysophanol,and physcion in Zuoyu’an San,respectively. To verify the reliability of QAMS metod results,the contents of 8 components of physcion were determined.

Results:

A total of 23 common peaks were calibrated in the fingerprint,and 8 components were identified by comparing with the mixed reference solution. The similarity between the samples and the control spectrum was between 0.952 and 0.994;CA divided 12 batches of Zuoyu’an San samples into 3 categories;PCA showed that there were certain differences in the chemical composition of different batches of Zuoyu’an San,and extracted 5 principal components that affected the quality evaluation of Zuoyu’an San;OPLS-DA screened out 13 potential marker components (VIP>1) among 23 common peaks that caused quality differences between different batches of Zuoyu’an San samples;There was no significant difference between the QAMS method calculated value and the ESM measured value.

Conclusion:

The established HPLC fingerprint and multi-index component determination method was accurate and simple,and combined with chemometric methods,could provide reference for the quality control and evaluation of Zuoyu’an San.

坐浴安散是广西中医药大学附属瑞康医院研发的中药制剂(桂药制字Z20050001),处方由大黄、功劳木、苦参、五倍子、两面针、白矾、芒硝、冰片组成,具有清热解毒、消肿止痛的功效,用于治疗内外痔发炎、痔瘘手术后、肛瘘肿痛。该方临床应用已有20余载,疗效显著[1-2]。该制剂现行质量标准仅有2020年版《中华人民共和国药典》中散剂项下规定的检查项及部分鉴别项,对制剂的质量控制较弱。近年来,通过多指标同步质量控制反映中药质量已成为当前中药乃至复方中药制剂质量评价领域的共识。中药指纹图谱具有整体性强、专属性高、分析手段丰富等特点,能表征被测样品主要化学成分的整体特征,确保其内在质量的均一和稳定等,现已广泛应用于中药及中成药制剂的整体质量评价[3];化学计量学是通过统计学或数学方法结合计算机科学,将对化学体系的测量值与体系的状态之间建立联系[4];一测多评(QAMS)法与外标法(ESM)相比,只需要采用1种对照,就能实现多种成分的同时测定,降低了对照品的使用成本,从而使检测更加简便、经济、快速[5]。本研究采用HPLC指纹图谱,结合化学计量学对坐浴安散进行全面分析,并运用QAMS法同时测定坐浴安散中多个成分,以期为坐浴安散的整体质量控制提供参考。
UV-1800PC紫外可见分光光度计(上海美谱达仪器有限公司);LC-2030(Plus)高效液相色谱仪(Shimadzu公司,包括真空脱气机、四元泵、自动进样器、UV检测器);Shim-pack VP-ODS色谱柱(Shimadzu公司,250 mm×4.6 mm,5μm);ZORBAX SB-Aq色谱柱(Agilent公司,250 mm×4.6 mm,5 μm);Symmetry Shield RP18色谱柱(Waters公司,250 mm×4.6 mm,5 μm);ES 225SM-DR(E)电子天平(上海精密科学仪器有限公司,十万分之一);SYU-3-100超声波清洗机(郑州生元仪器有限公司)。
对照品盐酸药根碱(批号110733-202110,纯度90.3%)、盐酸巴马汀(批号110732-201913,纯度85.7%)、盐酸小檗碱(批号110713-202015,纯度85.9%)、芦荟大黄素(批号110795-201710,纯度98.3%)、大黄酸(批号110757-201607,纯度99.3%)、大黄素(批号110756-201913,纯度96.0%)、大黄酚(批号110796-201922,纯度99.4%)、大黄素甲醚(批号110758-201616,纯度99.0%),均购自中国食品药品检定研究院;无水乙醇(天津市富宇精细化工有限公司,分析纯);甲醇(赛默飞世尔科技有限公司,色谱纯);乙腈(赛默飞世尔科技有限公司,色谱纯);水(屈臣氏蒸馏水,批号20230704);坐浴安散12批(编号S1~S12,批号分别为20231123、20230919、20230807、20230728、20230717、20230629、20230620、20230511、20230328、20230306、20221215、20221010),广西中医药大学附属瑞康医院制剂室生产。
取坐浴安散约3 g,精密称定,置具塞锥形瓶中,精密加入甲醇30 mL,称量,超声处理(功率100 W,频率40 kHz)30 min,取出,放冷,再称量,用甲醇补足减失的量,滤过,取续滤液,微孔滤膜(0.45 μm)滤过,即得。
精密称取芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚、盐酸药根碱、盐酸巴马汀、盐酸小檗碱的对照品适量,置100 mL量瓶中,加甲醇溶解,摇匀,制成含芦荟大黄素27.52 μg·mL-1、大黄酸50.64 μg·mL-1、大黄素30.43 μg·mL-1、大黄酚40.56 μg·mL-1、大黄素甲醚48.02 μg·mL-1、盐酸药根碱130.21 μg·mL-1、盐酸巴马汀89.90 μg·mL-1、盐酸小檗碱34.02 μg·mL-1的混合对照品溶液。
分别取处方缺大黄及功劳木的其余药材,按处方相同制备工艺制得缺大黄和缺功劳木的阴性样品各3 g,精密称定,照供试品溶液制备方法操作,即得。
采用Shim-pack VP-ODS(250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱(0~10 min,20%A;10~20 min,20%A→45%A;20~35 min,45%A→60%A;35~48 min,60%A→65%A;48~60 min,65%A→45%A),流速1.0 mL·min-1,柱温30 ℃,进样量10 μL,检测波长为345 nm(0~25 min,检测药根碱、巴马汀、小檗碱)、254 nm(25~70 min,检测芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚);理论塔板数按大黄酸峰计算应不低于3 000。
精密度试验:取坐浴安散样品(S2),按“2.1.1”项下方法制备供试品溶液,按“2.2”项下色谱条件连续6次进样测定,以大黄酸峰为参照峰,测得各共有峰相对峰面积的RSD<2.0%,相对保留时间的RSD<2.0%,表明仪器精密度良好。
重复性试验:取坐浴安散样品(S2),按“2.1.1”项下方法平行制备6份供试品溶液,按“2.2”项下色谱条件进样测定,以大黄酸峰为参照峰,测得各共有峰相对峰面积的RSD<2.0%,相对保留时间的RSD<2.0%,表明该方法重复性良好。
稳定性试验:取坐浴安散样品(S2),按“2.1.1”项下方法制备供试品溶液,分别于室温下放置0、4、10、18、24 h后,按“2.2”项下色谱条件进样测定,以大黄酸峰为参照峰,测得各共有峰相对峰面积的RSD<2.0%,相对保留时间的RSD<2.0%,表明溶液在24 h稳定性良好。
按“2.1.1”项下方法制备12批坐浴安散供试品溶液,按“2.2”项下色谱条件进样测定,将12批坐浴安散HPLC图谱色谱峰积分转化为AIA格式,利用《中药色谱指纹图谱评价系统(2.0版)》对数据进行处理,以样品S1图谱为参照图谱,时间窗宽度设置为0.1 min,采用中位数法,通过多点校正进行全谱峰匹配,建立12批坐浴安散HPLC指纹图谱,在此基础上生成对照指纹图谱(见图1),并进行相似度评价。结果12批坐浴安散样品(S1~S12)的相似度分别为0.974、0.978、0.979、0.993、0.992、0.991、0.994、0.984、0.986、0.985、0.952、0.970,与其对照图谱的相似度在0.952~0.994,表明不同批次的坐浴安散样品一致性及稳定性良好。经过全峰匹配,确定了23个共有峰,与混合对照品溶液图谱比对,指认了8个共有峰,根据保留时间先后顺序依次为峰6(药根碱)、峰8(巴马汀)、峰9(小檗碱)、峰18(芦荟大黄素)、峰19(大黄酸)、峰21(大黄素)、峰22(大黄酚)、峰23(大黄素甲醚),其中峰19(大黄酸)分离度较好,响应较高,故选择其作为参照峰。
聚类分析(CA)能够从整体角度分析并比较不同样品间的相似性,排除相关性较差变量的影响[7]。使用SPSS 27.0软件,以12批坐浴安散的23个共有峰的峰面积为变量,选择平方欧式距离为测度,采用组间均联法进行系统聚类,对其进行CA。结果显示,当平方欧式距离为10时,12批样品可以被分为3类,S1、S2、S3、S4、S5、S6、S8、S9、S10为第1类,S7为第2类,S11、S12为第3类,CA谱系树状图见图2
主成分分析(PCA)能将多个指标转化为少数几个主成分,这些主成分是原始变量的线性组合,且彼此之间互不相关,可以直观显示出不同样品之间的整体差异性,进一步寻找不同批坐浴安散质量的差异性变量[8]。使用SPSS 27.0软件,以12批坐浴安散的23个共有峰的峰面积为变量,将其Zscore标准化处理,再进行PCA。以特征值>1,自动得到5个主成分,累积的方差贡献率为94.097%,相关信息见表1。结合总方差解释结果,观察碎石图(见图3),当折线由陡峭突然变得平稳时,陡峭到平稳对应的因子个数即为参考提取因子个数。结果表明,5个主成分能够充分体现23个共有峰的基本特征值和主要信息,可以作为不同批次坐浴安散质量的差异性变量反映其整体质量,具有进一步研究的意义。
主成分载荷矩阵反映了各变量对主成分的贡献大小和作用方向。从主成分矩阵(见表2)看到,23个共有峰中,峰2、峰4、峰5、峰6(药根碱)、峰7、峰8(巴马汀)、峰9(小檗碱)、峰11~峰13、峰16、峰17、峰18(芦荟大黄素)、峰19(大黄酸)、峰21(大黄素)、峰22(大黄酚)、峰23(大黄素甲醚)位于第1个主成分上,有较高的载荷;峰9(小檗碱)、峰13、峰20位于第2个主成分上,有较高的载荷;峰1、峰3位于第3个主成分上,有较高的载荷;峰23(大黄素甲醚)位于第4个主成分上,有较高的载荷;峰14位于第5个主成分上,有较高的载荷,说明影响样品质量差异的并不是单一成分,而是由多个成分聚集产生作用,对应复方中药制剂治疗疾病多成分、多靶点的特点。进一步对表2中主成分的载荷值进行计算,得出各主成分因子的线性模型[9],其表达式见表3。其中,Y1~Y5分别代表5个主成分的得分,X1~X23分别代表23个共有峰的峰面积。将各批次样品数据代入上述表达式后,可得其主成分得分。结合方差贡献率进行综合评价,按公式Y=(0.528 70×Y1+0.175 40×Y2+0.121 18×Y3+0.064 10×Y4+0.051 60×Y5)/0.940 97计算,结果见表4。综合评分的范围为-3.046~2.856,表明不同批次的坐浴安散化学成分存在一定的差异,产生差异的原因可能与饮片质量、制备过程等因素相关。
PCA为无监督的模型验证方法,并不能够对所有样本加以区分以使得每个样本对模型都有相同的贡献,所以,当样本组间差异大而组内差异较小时,PCA难以区分和发现组内差异,因此,有必要采用一种有监督模式的分析—正交偏最小二乘-判别分析(OPLS-DA)[10]。为了进一步探寻引起不同批次坐浴安散质量差异的标志成分,将12批坐浴安散的23个共有峰峰面积作为变量导入SIMCA 14.1软件进行OPLS-DA,结果见图4R2X=0.640,R2Y=0.859,Q2=0.643,>0.5,说明建立的模型稳定可靠,具有较强的预测能力,12批坐浴安散共有峰峰面积均落在95%置信区间内。为防止数据出现过拟合现象而影响结果的准确性,利用200次置换试验进行检验,结果R2=(0.0,0.456),Q2=(0.0,-0.573),<0.05,由于Q2拟合直线Y轴截距为-0.573,为负数,说明模型未出现过度拟合的情况,所构建的OPLS-DA模型可以判别12批坐浴安散质量成分的差异,可进行进一步分析。以变量重要性投影(VIP)值>1为标准,筛选贡献较大的差异性成分,结果显示VIP>1的色谱峰有峰2、峰4、峰6(药根碱)、峰8(巴马汀)、峰9(小檗碱)、峰11、峰12、峰16、峰17、峰18(芦荟大黄素)、峰19(大黄酸)、峰21(大黄素)、峰22(大黄酚),表明这些成分可能是导致不同批次坐浴安散样品间质量差异的潜在标志成分,可作为坐浴安散含量测定的参考成分。
取“2.1”项下混合对照品溶液、供试品溶液、阴性对照溶液,按“2.2”项下色谱条件分别进样测定,记录色谱图(图5)。结果对照品溶液和供试品溶液的色谱图中芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚、药根碱、巴马汀、小檗碱的色谱峰峰形对称,理论塔板数按大黄酸峰计均>3 000,阴性对照溶液对相应的待测成分测定无干扰,表明方法专属性较强。
取“2.1.2”项下混合对照品溶液各1、2、4、6、8、9 mL,置10 mL量瓶中,加甲醇稀释至刻度,得6个梯度混合对照品溶液,按“2.2”项下色谱条件分别进样测定,以峰面积Y为纵坐标,溶液浓度X(μg·mL-1)为横坐标,进行线性回归,得各成分的回归方程、相关系数及线性范围(见表5)。结果表明,该色谱条件下,待测成分线性关系良好。
取坐浴安散样品(S1),按“2.1.1”项下方法制备供试品溶液,按“2.2”项下色谱条件连续进样6次进行测定,结果药根碱、巴马汀、小檗碱、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚峰面积的RSD分别为1.1%、1.5%、1.3%、0.27%、1.0%、0.24%、0.28%、0.90%,表明仪器的精密度良好。
取坐浴安散样品(S1),按“2.1.1”项下方法平行制备6份供试品溶液,再按“2.2”项下色谱条件分别进样测定,结果样品中药根碱(以盐酸药根碱计)、巴马汀(以盐酸巴马汀计)、小檗碱(以盐酸小檗碱计)、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚的平均含量(n=6)分别为233.76、217.13、112.88、86.52、219.02、190.27、145.39、180.74 μg·g-1,RSD分别为0.67%、0.65%、1.6%、0.60%、0.55%、0.57%、0.58%、0.93%,表明本方法的重复性良好。
取坐浴安散样品(S1),按“2.1.1”项下方法制备供试品溶液,分别于室温下放置0、4、10、16、20、24 h后进样测定,结果药根碱、巴马汀、小檗碱、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚峰面积的RSD分别为0.47%、1.4%、1.2%、0.77%、0.47%、0.66%、0.71%、1.5%,表明供试品溶液在24 h内稳定性良好。
取已知药根碱、巴马汀、小檗碱、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚8个成分含量的坐浴安散6份,每份1 g,精密称定,置具塞锥形瓶中,精密加入“2.1.2”项下配制的混合对照品溶液2 mL,按“2.1.1”项下自“称量”起操作,制备加样供试溶液6份,在“2.2”项色谱条件下分别进样测定,计算加样回收率和RSD。结果药根碱(以盐酸药根碱计)、巴马汀(以盐酸巴马汀计)、小檗碱(以盐酸小檗碱计)、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚的平均加样回收率(n=6)分别为97.6%、97.7%、98.1%、104.1%、104.6%、105.2%、102.8%、102.0%,RSD分别为0.87%、1.6%、1.9%、1.8%、1.5%、2.2%、0.99%、1.2%。表明方法的加样回收率(准确度)良好。
取12批(S1~S12)样品,按“2.1.1”项下方法制备供试品溶液各3份,按“2.2”项下色谱条件分别进样测定,结果药根碱(以盐酸药根碱计)、巴马汀(以盐酸巴马汀计)、小檗碱(以盐酸小檗碱计)、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚的平均含量(n=3)分别为234.86、209.90、112.01、85.15、218.51、187.70、143.23、157.02 μg·g-1,RSD分别为2.4%、3.7%、3.0%、3.1%、3.9%、3.4%、3.1%、9.2%。
以大黄酸为内参物,测定其他7个指标成分相对于大黄酸的校正因子。取“2.1.2”项下混合对照品溶液1、2、5、8、10、20 μL,依次注入高效液相色谱仪,测定峰面积,每个取样量平行测定2份。分析结果,峰面积以平均值计,按公式fk/s=fk/fs=(Ck/Ak)/(Cs/As)计算各成分的相对校正因子[11],式中Ck为待测成分的浓度,Ak为待测成分的色谱峰峰面积,Cs为大黄酸的浓度,As为大黄酸的色谱峰峰面积,结果见表6
固定其他色谱条件不变,考察了体积流量(0.8、0.9、1.0、1.1、1.2 mL·min-1)对相对校正因子的影响,结果见表7,可知均无明显影响(RSD<5%)。
固定其他色谱条件不变,考察了柱温(20、25、30、35 ℃)对相对校正因子的影响,结果见表8,可知均无明显影响(RSD<5%)。
固定其他色谱条件不变,考察了色谱柱(Shim-pack VP-ODS、Symmetry Shield RP18、ZORBAX SB-Aq)对相对校正因子的影响,结果见表9,可知均无明显影响(RSD<5%)。
采用相对保留时间法,分别考察内标和其他待测成分在不同色谱柱下的相对保留时间,采用待测成分保留时间/大黄酸保留时间计算,结果均无明显差异(RSD<5%)。
3个不同实验人员采用“2.5.8”项下方法对相对校正因子进行测定并计算,结果见表10。RSD均<3%,相对校正因子值具有较好的重现性。因此,采用3个实验人员测定的平均相对校正因子作为本次QAMS试验的校正因子。
取12批坐浴安散样品(S1~S12),分别按“2.1.1”下方法制备供试品溶液,依法进样测定,记录峰面积,分别用ESM和QAMS法计算8个成分的含量并进行比较,结果见表11。2种方法所得含量的相对平均偏差(RAD)均<3%,表明QAMS法的计算值与ESM测定值无明显差异,本研究建立的QAMS法稳定可行。
本研究比较了不同浓度甲醇(50%、70%、100%)及不同提取工艺(超声和加热回流)对坐浴安散中成分提取结果的影响。结果显示,以100%甲醇为提取溶剂,色谱峰具有较好的峰形和分离度,基线噪声较小;2种提取工艺提取效果差异较小,为了实验简便及节省能源,将超声方法用于供试品溶液的制备。
含量测定中8个待测成分最大吸收波长有差异,用单一波长检测,很难兼顾每个成分的最佳响应值。使用紫外可见分光光度计,对8个化合物在200~400 nm范围内进行全波长扫描,在不考虑有较大干扰的末端吸收下,根据各成分的紫外吸收图谱来确定检测波长[12]。观察各成分光谱的吸收曲线,结合2020年版《中华人民共和国药典》及相关文献报道[13-14],最终确定芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚的检测波长为254 nm,药根碱、巴马汀、小檗碱的检测波长为345 nm。在HPLC实验中进行波长切换,采用分段波长法检测,兼顾含量测定及指纹图谱的整体性。
流动相体系考察了乙腈-水和乙腈-0.1%磷酸溶液,结果显示,以乙腈-0.1%磷酸溶液为流动相时,基线平稳,色谱峰出峰情况较好,无拖尾现象。同时对梯度洗脱的时间程序进行了大量摸索,最终在文中选定的洗脱程序下进样时,色谱峰较多且分离度好,指纹图谱整体性得到保证[15]
本研究建立了12批坐浴安散的指纹图谱,相似度以对照指纹图谱为参考在0.952~0.994,表明这12批样品质量相对稳定。结合CA、PCA、OPLS-DA一系列化学计量学方法对其进行分析,结果不同批次样品出现一定的差异,筛选出13个VIP值>1的共有峰,分别为峰2、峰4、峰6(药根碱)、峰8(巴马汀)、峰9(小檗碱)、峰11、峰12、峰16、峰17、峰18(芦荟大黄素)、峰19(大黄酸)、峰21(大黄素)、峰22(大黄酚)。本方中大黄主要化学成分包括结合型和游离型蒽醌类,具有清热泻火、凉血解毒和逐瘀通经等功效[16];功劳木主要含有小檗碱、巴马汀、氧化小檗碱等生物碱类化合物,具有清热燥湿、泻火解毒的功效,药理活性主要为抗氧化、抗炎和抑菌等[17]。依据本方君臣佐使配伍原理,结合OPLS-DA结果和主要药效成分,选择大黄酸等8个成分进行定量。含量测定QAMS法选择大黄酸为内参物,建立了药根碱等其他7个待测成分的相对校正因子,同时以ESM验证QAMS法的准确性,结果表明2种方法准确度无明显差异。但是从测定结果来看,不同批次的坐浴安散8个成分含量存在一定的批间差异,可能与所用饮片产地、批次、厂家和制剂加工等因素有关,提示在今后的生产中饮片产地、厂家、控制生产参数等因素可以相对固定,以保证制剂质量的稳定[18]
综上所述,本研究建立的指纹图谱和含量测定方法稳定可行,化学计量学分析可为坐浴安散的质量评价提供依据。
