Objective:

To establish a quantitative nuclear magnetic resonance hydrogen spectroscopy(1H qNMR) method for determining the moisture content in several pharmaceutical excipients.

Methods:

Using sodium benzenesulfonate as the internal standard and heavy water as the solvent,the water content in the blank sample was determined by absolute quantification using nuclear magnetic resonance spectroscopy. By comparing with the blank value,a quantitative calculation formula for the water content in the sample was established,and the water content in the sample was tested and calculated.

Results:

Methodological studies had shown that this method had a good linear relationship(r=0.999 1),with a linear range of 0.032-0.561 mg absolute water content in the sample,the precision RSD was 0.039%,repeatability RSD was 0.43%,and the average recovery was 98.7%. The 1H qNMR method measured the water content in DMSO-1, DMSO-2, sodium benzenesulfonate, and sodium acetate to be 0.23%, 0.28%, 0.46%, and 0.28%, respectively, which were essentially consistent with the results obtained from the method described in the Chinese Pharmacopoeia(2020).

Conclusion:

The 1H qNMR method established is fast,simple to operate,with a small sample size and accurate results. It can be used for the determination of water content in some pharmaceutical excipients.

目的:

建立定量核磁共振氢谱(1H qNMR)法测定若干药用辅料中水分。

方法:

以苯磺酸钠为内标,重水为溶剂,采用核磁共振氢谱绝对定量法测定空白试样中水的含量,并在此基础上,与空白值进行比对,建立样品水含量定量计算公式,并对样品中水分进行测试和计算。

结果:

方法学研究表明,该方法具有良好的线性关系(r=0.999 1),线性范围为试样绝对含水量0.032~0.561 mg,精密度试验RSD为0.039%,重复性试验RSD为0.51%,平均加样回收率为98.7%,1H qNMR法测得DMSO-1、DMSO-2、苯磺酸钠、乙酸钠中水分分别为0.23%、0.28%、0.46%、0.28%,与2020年版《中华人民共和国药典》经典方法试验结果基本一致。

结论:

建立的1H qNMR法操作快速、简便,样品用量少,结果较准确,可用于一些药用辅料中水含量的测定。

目的:

建立定量核磁共振氢谱(1H qNMR)法测定若干药用辅料中水分。

方法:

以苯磺酸钠为内标,重水为溶剂,采用核磁共振氢谱绝对定量法测定空白试样中水的含量,并在此基础上,与空白值进行比对,建立样品水含量定量计算公式,并对样品中水分进行测试和计算。

结果:

方法学研究表明,该方法具有良好的线性关系(r=0.999 1),线性范围为试样绝对含水量0.032~0.561 mg,精密度试验RSD为0.039%,重复性试验RSD为0.51%,平均加样回收率为98.7%,1H qNMR法测得DMSO-1、DMSO-2、苯磺酸钠、乙酸钠中水分分别为0.23%、0.28%、0.46%、0.28%,与2020年版《中华人民共和国药典》经典方法试验结果基本一致。

结论:

建立的1H qNMR法操作快速、简便,样品用量少,结果较准确,可用于一些药用辅料中水含量的测定。

Objective:

To establish a quantitative nuclear magnetic resonance hydrogen spectroscopy(1H qNMR) method for determining the moisture content in several pharmaceutical excipients.

Methods:

Using sodium benzenesulfonate as the internal standard and heavy water as the solvent,the water content in the blank sample was determined by absolute quantification using nuclear magnetic resonance spectroscopy. By comparing with the blank value,a quantitative calculation formula for the water content in the sample was established,and the water content in the sample was tested and calculated.

Results:

Methodological studies had shown that this method had a good linear relationship(r=0.999 1),with a linear range of 0.032-0.561 mg absolute water content in the sample,the precision RSD was 0.039%,repeatability RSD was 0.43%,and the average recovery was 98.7%. The 1H qNMR method measured the water content in DMSO-1, DMSO-2, sodium benzenesulfonate, and sodium acetate to be 0.23%, 0.28%, 0.46%, and 0.28%, respectively, which were essentially consistent with the results obtained from the method described in the Chinese Pharmacopoeia(2020).

Conclusion:

The 1H qNMR method established is fast,simple to operate,with a small sample size and accurate results. It can be used for the determination of water content in some pharmaceutical excipients.

在制药工程等领域,制品中水分的存在,会影响制品活性成分的含量、药效、理化稳定性及保质期等多项参数,为了满足生产工艺、质量控制、产品储存等方面的要求,水分的测定显得十分重要[1],也是很多药品辅料质控必需的一个指标。此外,在标准物质的研制方面,水分的测定也极其关键[2-3],这将直接影响到质量平衡法的最终定量纯度值。
针对不同种类的制品及水分要求,2020年版《中华人民共和国药典》(简称《中国药典》)列举了若干类测定方法,如费休氏法、烘干法、减压干燥法、甲苯法等。同时,随着研究开发,近年来亦出现了一些其他水分分析手段,如近红外光谱分析、气相色谱法、微波传感分析等[4-6]。这些方法在不同的领域虽各有优势,但是,目前采用最多的经典方法还是费休氏法。与其他测定水分的方法相比,费休氏法具有检测限低,线性区间宽,适用范围广等优点,可应用于很多场景并被视为标准方法[7-9]。但是,费休氏滴定法也存在一些不足之处,比如,操作较为烦琐,需要专门配制或购买费休氏试液,在采用容量滴定法时还需要对试液进行标定;为了提高结果的准确度,往往要求的样品量较大;有时需要通过卡式炉联用进行测定等[10]
近年来,定量核磁共振波谱(qNMR)技术因其操作简单、无损测定等优点,在各领域得到越来越广泛的应用[11-15],也是质量平衡法的重要补充[16],文献报道也有若干利用qNMR测定样品中水分的研究[17-19]。但这些研究测定的样品对象比较有限,其中有的方法需要化学衍生化,步骤较为烦琐。为了拓展qNMR方法的应用场景,本研究以重水为溶剂,苯磺酸钠为内标,建立了1H qNMR法测定样品中水分绝对含量的方法,对二甲基亚砜(DMSO)等一些不含活泼氢的药品辅料中的水分进行了测定,方法简单快捷,准确性高,样品用量较少,通用性强。
Agilent NMR 600 MHz核磁共振波谱仪(Agilent公司);CP225D分析天平(十万分之一,赛多利斯公司);移液枪(Eppendorf公司);916 Ti-Touch自动电位滴定仪(万通公司)。
重水(批号INBAABAFB,北京伊诺凯科技有限公司,含量99.9%);苯磺酸钠(批号AMUYD-RT,TCI公司,标定含量99.7%);马来酸标准物质(批号BCCC6481,SIGMA-ALDRICH公司,纯度99.94%,用于核磁共振法标定苯磺酸钠);方法验证用若干种实验室常见辅料样品,包括DMSO-1(批号T20100429,国药集团化学试剂有限公司)、DMSO-2(批号F72315773,阿拉丁试剂有限公司)、苯甲酸钠(批号20230505,国药集团化学试剂有限公司,含量99.5%)、乙酸钠(批号BCCJ0024,Sigma-Aldrich公司,含量99. 0%)。
精密称量苯磺酸钠666.88 mg,加入重水20 mL,充分溶解,配成苯磺酸钠内标溶液,密封保存。
移取DMSO样品5~300 μL(体积可根据样品水分适当调整),精密称定移取的样品质量;用移液枪精准加入内标溶液500 μL;溶解混匀并移入核磁管测定。对于固体样品,可精密称定样品(称取质量可根据样品水分适当调整)5~300 mg,其余操作同DMSO样品。
为了尽量减少环境可能的影响,采用与“供试品溶液”配制完全相同的操作步骤,不同之处在于加入DMSO样品的量为0。
采用氢谱的脉冲序列,测试温度25 ℃,谱宽(-2~14)×10-6,脉冲角度45°,脉冲延迟时间60 s,采样时间3 s,扫描次数8次。数据处理使用MestReNova5. 3.1软件,先进行相位和基线校准,然后采用手动积分,计算得到水峰和内标定量峰峰面积的相对比值,采用2020年版《中国药典》四部通则“0441核磁共振波谱法”中绝对定量公式,计算得到空白试样中水分的绝对含量。
将一定量的苯磺酸钠、DMSO样品溶于重水中混合制样,测定试样的1H NMR,得到如图1所示氢谱。图中峰1的两重峰对应苯磺酸钠苯环上磺基邻位的氢质子;峰2为水质子峰;峰3为DMSO质子峰。由图1可以看出,3个谱峰均呈现出良好的线形和分辨率,与其他谱峰基线分离良好且无相互干扰。同时,苯磺酸钠是离子化合物,具有良好的水溶性和稳定性,不易挥发,溶液呈中性状态,没有易电离的氢质子,对样品水分的测定干扰小。因此,苯磺酸钠可作为定量内标,并把δH7.86和4.80处的峰1和峰2作为定量特征峰,对样品中的水分进行测定。
核磁共振定量实验结果的准确性与实验仪器参数的选择紧密相关,仪器参数主要包括延迟时间、采样时间、扫描次数等,其中又以延迟时间的合理设置尤为重要。采用足够的延迟时间,才能保证激发的氢核弛豫完全,从而使氢谱的峰面积与对应的原子数目具有可比性,通常定量核磁要求5T1(纵向弛豫时间)的延迟时间。利用反转恢复法对T1进行测定,结果发现,体系中水质子峰具有最长的纵向弛豫时间,其T1约为11.4 s。除T1外,延迟时间的合理设置还与脉冲角度等多种可能因素相关。因此,为了得到合理的延迟时间,通过改变该参数,考察目标物与内标定量峰面积的比值使之稳定,是一个较为合理的方法,这样就可以消除各种未知因素带来的可能影响。于是分别设定一系列的延迟时间(1、2、5、10、15…80 s),对样品进行氢谱采样。通过考察目标物与内标定量峰面积的比值可知,60 s的延迟时间足以使该比值保持稳定,而这个数值也与5T1基本吻合。在确定延迟时间的前提下,分别设置系列采样时间(1~10 s)和扫描次数(2~32次)进行氢谱测定,结果表明,这2个参数对比值的影响较小,综合考虑谱图信噪比和实验效率,采用采样时间3 s和扫描次数8作为定量实验条件。因此,最后优化的实验参数为:延迟时间60 s,采样时间3 s,扫描次数8。
按照“2.1”项下溶液的制备方法和“2.2”项下检测条件,调整DMSO样品(已标定水分)的加入量为0~116.15 mg,其中空白溶液的样品加入量为0,测定上述溶液的1H NMR并积分。因为DMSO的加入量与其所含水的量成正比,因此,DMSO的加入量可换算为水的加入量,以水的加入量X(mg)为横坐标,以目标物(水)/内标物定量峰面积的比值(AS)为纵坐标Y,得到拟合曲线和线性回归方程
可知目标物质量与峰面积比值在测定范围内线性关系良好,且拟合曲线基本通过空白溶液的坐标点(0,1.817)。因此,在实际测定中为简化步骤,减少工作量,采用扣除空白的单点法定量是合理可行的。在采用绝对定量公式计算空白溶液含水量(mK)的基础上,可得到样品水分计算公式:
式中:ωS为样品中水的质量分数(%),mS为供试品溶液中加入样品的质量(mg),AS为供试品溶液的目标物(水)与内标定量峰面积的比值,AK为空白溶液的目标物(水)与内标定量峰面积的比值,mK为空白溶液中的含水量(mg)。
采用2020年版《中国药典》四部通则“0441核磁共振波谱法”中核磁绝对定量法计算水分,可得到空白溶液含水量为3.02 mg;系列加入DMSO样品的供试品溶液扣除空白后含水量为0.032~0.561 mg,可作为该方法的线性范围。
按照“2.1”项方法配制DMSO样品的供试品溶液,在相同的试验条件下,对其进行连续6次测定,计算水/内标物定量峰面积的比值(AS),可得到RSD为0.039%。因试验是对同一样本连续进行测定,故其RSD与核磁仪器本身的性能相关,试验结果表明,仪器的状态稳定,精密度良好。
按照“2.1”项方法,分别取同一DMSO样品6份配制供试品溶液,在相同试验条件下平行测定6次,同时测定的还有空白溶液,根据积分结果分别计算水的质量及DMSO样品水分(ωS),得到ωS值的RSD为0.51%,具体见表1,由此表明该定量方法具有良好的重复性。
按照“2.1”项方法,分别称取一定量的DMSO样品(已测定水分)于6个试管中,然后分别加入不同质量的重水(H2O含量已经通过核磁绝对定量法进行标定),对上述体系进行含水量测定,DMSO样品和重水溶液中的H2O质量以及体系含水量测定。6次回收率试验包含3种不同加标量,其回收率结果没有显著差异,范围为96.3%~103.3%,平均回收率为98.7%,RSD为2.5%,定量结果较为准确。
在已知“2.5.1”拟合曲线斜率和测定11个空白溶液的基础上,计算得到方法的检测限为供试品溶液中样品水含量8.7 μg,定量限为水含量26 μg,显示了较高的灵敏度。实际检测时,可根据样品中水分的高低,灵活改变加入的样品量,从而可以大大提高方法的通用性,扩展方法的适用范围。
按照测定原理,采用类似的操作步骤,本研究的分析方法同样适用于其他一些药品辅料中水分的测定,其中也包括无机辅料。选择若干种实验室常见的辅料样品,对本方法开展验证试验,同时采用2020年版《中国药典》的经典方法(费休氏法、烘干法)进行对照测试。由表2可以看出,本方法与2020年版《中国药典》的费休氏法(容量滴定法)和烘干法测试结果基本一致(烘干法结果相对偏高,可能源于样品里面包含有其他挥发性物质),也说明了本方法的实用性。
实验室所用的试剂中或多或少都含有一定的水分,因此,方便、快捷、准确地测定样品中的微量水一直是个难题。本研究提出的方法,通过核磁绝对定量法测定并扣除空白溶液中的水分,可以规避试剂含水对测定结果的影响,为采用核磁法测定水分提供了一个可选思路。通过对比发现,本方法与药典方法有较好的一致性,取得了较满意的测试效果,且具有方法简单快捷,样品用量较少,通用性较强的特点。但在实际应用中,有以下几点需要重视。
定量核磁共振测定中,激发的氢核完全弛豫是保证准确定量的基础,而水分子属于小分子,具有较长的纵向驰豫时间,因此,测试条件必须提供足够长的延迟时间,以保证定量的准确性,本研究中的延迟时间设为60 s。
根据测试原理可知,测试样品中不能含有活泼氢,因为活泼氢会和溶剂重水中的氘发生交换,生成H2O,从而使测试结果偏大,这就使得本方法在测定药用辅料水分时具有一定的局限性,限制了其对一些有机辅料的应用(比如醇类、酸类等),但对于很多不含活泼氢的无机辅料该方法却能够较好地使用。为了消除活泼氢的影响,对于这类有机辅料,可采用如氘代DMSO为溶剂(前提是DMSO为其良溶剂),则不会发生上述交换反应;另外,通常氘代DMSO中样品活泼氢与H2O(δH约3.3×10-6)的出峰位置不同,保证了水质子峰积分的一致性。综上,对于含有活泼氢的有机物,按照本文的思路,通过改变溶剂种类(比如氘代DMSO),同样有望开发出类似的方法进行水分测定,从而大大扩展该方法的应用范围。
环境空气湿度的变化可能或多或少会给试验结果带来一些影响,特别是对样品中微量水的测定。空白溶液和供试品溶液采用统一的操作步骤,同步进行测定,可以将这些影响尽可能地降到最低。
数据处理不当可能会对结果带来一些影响,积分处理的过程尤其重要,积分之前需要对相位和基线仔细校准,尽量减少数据处理带来的误差。
