Objective:

To explore the feasibility and potential applications of high-resolution cyclic ion mobility separation mass spectrometry (Cyclic IMS) for detecting the conformations (topological isomers) of complex medicines.

Methods:

Using MccJ25 and Rubrinodin lasso peptides as models,the compounds were subjected to heat treatment at 0,50,and 80 ℃ for 4 h,respectively. The samples were then directly injected for separation and detection using high-resolution Cyclic IMS.

Results:

The two model compounds exhibited different topological structural changes at three temperatures tested. MccJ25 showed no significant conformational changes and exhibited a relatively stable topological structure across all temperatures. In contrast,Rubrinodin demonstrated significant conformational changes at each temperature. The cyclic ion mobility cell in Cyclic IMS was able to accurately identify conformational changes,such as folding and unfolding,in the complex drugs.

Conclusion:

Cyclic IMS can precisely analyze the conformation (topological isomers) of complex drugs,detect small conformational changes,and offers strong operability,good reproducibility,and high accuracy. It holds potential for use in the conformational determination and quality control of complex drug molecules.

目的:

探究高分辨环形离子淌度质谱(Cyclic IMS)检测套索多肽类复杂药物构象(拓扑异构体)的可行性及潜在应用。

方法:

以Mccj25及Rubrinodin套索多肽为药物分析模型,将2种化合物分别于0、50、80 ℃处理4 h,采用高分辨Cyclic IMS直接进样的方式对样品进行分离、检测。

结果:

2种模型药物在3个温度下呈现不同的拓扑结构变化,Mccj25在3个温度下构象未发生明显变化,具有较稳定的拓扑结构,而Rubrinodin在3个温度下构象发生了明显变化;采用Cyclic IMS中的环形淌度可精准识别复杂药物的折叠、解体等构象变化。

Cyclic IMS可对复杂药物的构象(拓扑异构体)进行精准分析,可识别药物微小的构象变化,方法可操作性强,重现性较好,准确度高,可用于复杂药物的构象测定及质量控制。

目的:

探究高分辨环形离子淌度质谱(Cyclic IMS)检测套索多肽类复杂药物构象(拓扑异构体)的可行性及潜在应用。

方法:

以Mccj25及Rubrinodin套索多肽为药物分析模型,将2种化合物分别于0、50、80 ℃处理4 h,采用高分辨Cyclic IMS直接进样的方式对样品进行分离、检测。

结果:

2种模型药物在3个温度下呈现不同的拓扑结构变化,Mccj25在3个温度下构象未发生明显变化,具有较稳定的拓扑结构,而Rubrinodin在3个温度下构象发生了明显变化;采用Cyclic IMS中的环形淌度可精准识别复杂药物的折叠、解体等构象变化。

结论:

Cyclic IMS可对复杂药物的构象(拓扑异构体)进行精准分析,可识别药物微小的构象变化,方法可操作性强,重现性较好,准确度高,可用于复杂药物的构象测定及质量控制。

Objective:

To explore the feasibility and potential applications of high-resolution cyclic ion mobility separation mass spectrometry (Cyclic IMS) for detecting the conformations (topological isomers) of complex medicines.

Methods:

Using MccJ25 and Rubrinodin lasso peptides as models,the compounds were subjected to heat treatment at 0,50,and 80 ℃ for 4 h,respectively. The samples were then directly injected for separation and detection using high-resolution Cyclic IMS.

Results:

The two model compounds exhibited different topological structural changes at three temperatures tested. MccJ25 showed no significant conformational changes and exhibited a relatively stable topological structure across all temperatures. In contrast,Rubrinodin demonstrated significant conformational changes at each temperature. The cyclic ion mobility cell in Cyclic IMS was able to accurately identify conformational changes,such as folding and unfolding,in the complex drugs.

Conclusion:

Cyclic IMS can precisely analyze the conformation (topological isomers) of complex drugs,detect small conformational changes,and offers strong operability,good reproducibility,and high accuracy. It holds potential for use in the conformational determination and quality control of complex drug molecules.

离子淌度质谱是近年来飞速发展的一种新型质谱技术,依靠离子在电场中不同的迁移率,可以实现药物的高精度分离与检测[1]。环形离子淌度质谱(Cyclic IMS)是在传统线性离子淌度的基础上进一步升级的产品,通过增加紧凑型的环形离子导向装置,Cyclic IMS可以控制离子进行一圈或多圈的淌度分离,从而大大提高药物分离分析的分辨率[2-3]。随着分辨率的大幅提高,Cyclic IMS可以广泛应用于分离小分子药物的顺反异构、非对映异构、对映异构体及大分子的拓扑异构体等,可以帮助研究人员更好地理解化合物的结构、构象等特征[2]
复杂大分子药物是近年来创新医药领域发展迅速的一类分支,包括多肽、多肽-小分子偶联药物、单克隆抗体、蛋白质药物、核酸药物等,在治疗肿瘤、感染、自身免疫病、代谢疾病等领域发挥着越来越重要的作用[4]。该类药物往往具有副作用低,特异性强,生物活性高等优点,但同时也有结构复杂,稳定性低,易变性等缺点[5]。这类药物的高级结构对药物生物活性起到至关重要的作用,也是考察药物质量可控性需要重点关注的领域[6]。各国药典都对大分子药物的质量控制进行了相关描述,包括一级结构、高级结构、生物活性等维度进行结构确认,确保产品的有效性和安全性,相关的技术手段涉及圆二色谱法、差示扫描量热法、傅里叶变换红外光谱法、X射线晶体衍射、核磁共振等[7]。与小分子药物不同,大分子药物的结构分析往往较为复杂,同时其变质过程往往还涉及到自身微小构象的变化,这些变质属性很难被传统分析方法精准识别,给药品的质量控制及监管带来了非常大的挑战,亟需开发新的质量控制方法。如何利用前沿分析技术如离子淌度质谱等对这类药物进行质量分析也逐渐成为近年来的研究热点[8-10]
套索多肽是一类来源于微生物的核糖体合成并翻译后修饰的多肽类天然药物,具有广泛的生物活性,如抗肿瘤、抗菌及受体拮抗剂等[11]。套索多肽具有独特的套索拓扑3D结构,其N端通常由7~9个氨基酸通过N端氨基酸的α-氨基与天冬氨酸或谷氨酸的羧酸侧链形成大环内酰胺,C端多肽尾巴通过穿入N端的大环内酰胺从而形成互锁套索样结构(图1[12]。这种独特的套索结构使这类分子具有良好的稳定性并被应用于药物支架设计[11]。例如,Microcin J25套索多肽已被开发用于展示RGD(Arg-Gly-Asp)肽,用于特异性识别在癌细胞中过表达的αvβ3整合素[13]。此外,美国初创药物公司Lassogen已获得450万美元种子轮融资,致力于研究套索多肽作为开发新药的一种方法。这类药物介于小分子化学药与大分子生物药之间,具有独特的化学性质及拓扑结构,一旦上市,将给相关药物的质量控制带来挑战。
本文以Mccj25套索多肽及Rubrinodin套索多肽为模型[14-15],采用Cyclic IMS对2种化合物的拓扑结构进行研究。通过对2种化合物进行不同的热处理,通过环形离子淌度直接进样的方式,考察此类药物在热处理变性条件下微小的构型变化,相关分析技术的开发为具有复杂拓扑结构药物的质量控制提供了新方法参考。
UltiMate 3000高效液相色谱仪(Thermo Fisher Scientific公司);Select Series Cyclic IMS环形离子淌度质谱仪(Waters Technologies公司)。
Mccj25套索多肽(5 mg)与Rubrinodin套索多肽(5 mg)均为中国食品药品检定研究院抗肿瘤和放射性药品室自制。三氟乙酸为色谱纯(购自Sigma-Aldrich);甲醇、乙腈均为色谱纯(购自百灵威科技有限公司)。
采用资生堂CAPCELL CORE C18(250 mm×4.6 mm,2.7 μm)色谱柱,流动相为0.1%三氟乙酸水溶液(A)-0.1%三氟乙酸乙腈溶液(B),梯度洗脱(0~2 min,10%B;2~15 min,10%B→50%B;15~25 min,50%B→95%B;25~30 min,95%B;30~31 min,95%B→10%B;31~35 min,10%B),流速1 mL·min-1,紫外检测波长220 nm,柱温30 ℃,进样量10 μL。
采用电喷雾正离子化,碰撞能量6 eV,碰撞能量转移4 eV,四极杆质量驻留时间10 ms,毛细管电压50 V,脱溶剂气流量9 L·min-1,脱溶剂温度450 ℃,扫描范围m/z 50~2 000。Cyclic IMS参数见表1
传统高效液相色谱方法可以对大分子药物构象进行分离分析。本文选用结构较为稳定的Mccj25套索多肽及结构相对不稳定的Rubrinodin套索多肽作为分析模型进行研究。精密称取Mccj25套索多肽及Rubrinodin套索多肽各约1 mg,置2 mL离心管中,用50%甲醇1 mL溶解,制成约1 mg·mL-1的套索多肽溶液各3份,分别置0、50、80 ℃金属浴处理4 h,将样品稀释10倍后转移至进样小瓶并进行液相色谱梯度洗脱分析。如图2-A所示,Mccj25在3个温度处理后,在液相色谱图上均呈现单一色谱峰(保留时间约为12.1 min),表明Mccj25套索多肽的构象在高温处理条件下未发生明显变化。与之相反,Rubrinodin呈现出明显不同的变化(图2-B):在0 ℃处理条件下,Rubrinodin在10.0 min处呈现单一主峰,在约8.5 min处有少量拓扑异构体产生。这些异构体主要是Rubrinodin在生产制备纯化过程产生的[14];Rubrinodin在50 ℃处理4 h后,大部分套索结构发生解折叠,产生新的伸展态的Rubrinodin(保留时间约8.5 min);Rubrinodin在80 ℃处理4 h后,液相色谱结果显示折叠态的Rubrinodin已完全转化为伸展态的多肽。高效液相色谱分析结果初步证明了Mccj25套索多肽相对稳定,在热处理条件下保持构象稳定,而Rubrinodin套索多肽在热处理条件下发生了明显的构象变化,推测套索多肽的C端尾巴从N端内酰胺大环中挣脱出来,形成伸展态多肽,此结论与文献[14]结论相符。
采用直接进样的方式对经过热处理的2种套索多肽进行高分辨Cyclic IMS分析。首先对Mccj25在80 ℃热处理4 h的样品进行质谱全扫描分析,发现Mccj25在质谱中显示出一系列带有不同钠离子的准分子离子峰,其中观测到的Mccj25[M+2H+Na]3+离子m/z为1 065.071,与理论预测值([M+2H]2+m/z 1 054.018)相符(图3-A)。对Rubrinodin在50 ℃处理4 h的样品进行质谱全扫描分析,发现Rubrinodin在质谱中同样显示出一系列带有不同钠离子的准分子离子峰,证明不同套索多肽在直接进样时带有不同数量钠离子是普遍现象。其中,观测到Rubrinodin[M+3H+5Na]8+离子m/z为795.049,与预测值([M+3H]3+ m/z 758.381)相符。
由于Cyclic IMS质谱采用环形淌度池,在测量时可以通过电场对离子的控制,实现离子在淌度池中运行较多的圈数,从而实现最优的分离效果。首先对Mccj25套索多肽进行分析,通过考察不同准分子离子峰,最终选择Mccj25的[M+2H+Na]3+进行优化。如图4所示,Mccj25通过环形淌度池1圈时,仅呈现单一质谱峰,这与HPLC的分析结果一致,显示Mccj25具有较为稳定的构象结构;将离子淌度条件提高到7圈时,Mccj25中有少量的拓扑异构体得到分离,从化学角度分析,Mccj25利用19位的苯丙氨酸和20位的酪氨酸为锁扣结构,结构极为稳定,但由于Mccj25套索含有2个脯氨酸,导致其有少量不同构象的套索多肽产生,在之前的研究中,已经有报告证明Mccj25套索多肽有Pro7/Pro16的顺式/反式异构化现象[16];进一步将离子淌度条件增加到9圈,发现2个拓扑异构体的分离度进一步增加。此实验证明,即使像Mccj25这种极其稳定的套索结构,其在溶液中也并非具有完全的构象均一性。Cyclic IMS提供了一种强大的分析工具,可以精准地将这些微少拓扑异构体分离分析出来。通过优化,最终选择运行7圈这个条件继续研究,探索Mccj25在热稳定性样品中的构象变化。
采用同样的考察策略,取50 ℃处理4 h的Rubrinodin套索多肽,选择[M+3H+5Na]8+ m/z 795.049的准分子离子峰进行环形离子淌度分析。如图5所示,当离子淌度条件为1圈时,同样仅呈现单一质谱峰;当离子淌度条件提高到5圈时,可以明显看到Rubrinodin已经分裂成双组分,推测其一为折叠态的套索多肽,另一为伸展态的多肽,与液相色谱结果一致;当进一步将离子淌度条件提高到11圈时,可以看到之前推测为单一构象的折叠态套索多肽进一步分裂成2个峰,证实Rubrinodin套索多肽在50 ℃变性过程中还产生了其他位置的拓扑异构体,推测其为C端尾巴未完全脱离N端大环内酰胺的中间态。通过高分辨的Cyclic IMS,证实Rubrinodin套索多肽在解折叠过程中产生了不同构象,并可分步精准监测,这种独特的分析方法为相关复杂药物的分析提供了一种可选方法。通过优化,最终选择运行11圈作为进一步分析Rubrinodin热稳定性行为的分析条件。
图6所示,Mccj25在0、50、80 ℃ 3个条件下分别处理4 h后,在环形离子淌度池中运行7圈,其构象分布基本没有变化。实验证明Mccj25具有非常好的热稳定性,同时证明了天然纯化的Mccj25套索多肽存在少量的拓扑异构体。对测定样品进行成像分析,结果Mccj25套索多肽在3个温度下表现一致,再次证明了套索多肽有良好的锁扣结构(具有大分子排阻的侧链),可为套索提供优越的热稳定性。
与Mccj25采用酪氨酸为锁扣结构不同,Rubrinodin采用侧链相对较小的精氨酸为锁扣,同时Rubrinodin的N端大环由9个氨基酸组成。这种大环套索叠加较小的锁扣氨基酸,使得Rubrinodin热稳定性较差。如图7-A所示,Rubrinodin在0 ℃时,Cyclic IMS显示其仍有3个拓扑异构体。在之前液相色谱分析中,已经观察到少量完全伸展态的多肽(保留时间8.5 min)。出乎意料的是,在液相色谱图中为单一色谱峰的套索多肽(保留时间10.0 min)混有部分拓扑异构体,提示在Rubrinodin的纯化或者保存过程中,已有部分套索多肽在拓扑结构上发生了变质,而这种变质是常规液相色谱无法检测到的。Rubrinodin在50 ℃处理4 h后(图7-B),可以明显看到套索已变成3个构象的混合物,完全伸展及解折叠中间态的套索多肽比例显著增加。Rubrinodin套索多肽在80 ℃处理4 h后(图7-C),大部分套索多肽已经完全转化成伸展态。对测定样品进行成像分析,结果证明Rubrinodin套索多肽在3个温度下经历了明显的分步构象变化。
传统小分子药物质量控制主要集中在有关物质的检测,涉及同分异构体、降解杂质等[17]。通过传统的高效液相色谱结合色谱柱等手段,可对相关杂质进行有效控制[18]。随着药物不断向中分子、大分子药物发展,涉及药物拓扑异构体的杂质给药品质量监管带来了更多挑战。在众多复杂药物的设计过程中,药物的空间拓扑结构对药物的生物活性至关重要,例如各种抗体及多肽类靶向药物[19],在构象上发生变质往往难以检测,潜在的生物危害难以察觉,还大大降低了药物的生物活性,亟需开发新的分析手段对此类杂质进行检测[20]
本文以Mccj25套索多肽及Rubrinodin套索多肽为模型,探究了复杂药物拓扑异构体的定性分析方法。以Rubrinodin套索多肽为例,当此类分子经历了较大的构象变化时(如折叠态变为完全伸展态),高效液相色谱结合色谱柱可进行有效地分离及分析;当药物经历微小构象变化时,高效液相色谱已无法精准判断样品的变质情况,在液相色谱图上显示为色谱纯的Rubrinodin样品在Cyclic IMS中呈现出多种构象的混合物。在Rubrinodin热处理过程中,可以通过环形离子淌度技术精准地观测到样品的解折叠中间态及伸展态的变化,Cyclic IMS为复杂药物在拓扑维度上的质量控制提供了强大的分析手段。
在Mccj25套索多肽的分析过程中发现,即使Mccj25是相对较为稳定的骨架结构,在Cyclic IMS中仍能观测到小部分拓扑异构体的存在,揭示了其结构的复杂性。提示在开发类似具有拓扑空间结构的药品时,需要同时关注此类分子的拓扑结构均一性。
