Objective:

To establish the substances benchmarks fingerprint of Daqinglong decoction and determine the content and transfer rate of 6 index components,and study the transfer rule of Daqinglong decoction substances benchmarks value,which laid the foundation for the study of Daqinglong decoction compound preparation and quality standard.

Methods:

Fifteen batches of substances benchmarks of Daqinglong decoction were prepared,and the paste rate was determined. To determine the contents of ephedrine,pseudoephedrine,amygdalin,and cinnamic acid,HPLC method was performed on a Dikma Platisil C18 column (250 mm×4.6 mm,5 μm). And 0.15% phosphoric acid water (A) and acetonitrile (B) were used as mobile phases with gradient elution at a flow rate of 1 mL·min-1. The column temperature was 25 ℃,the detection wavelength was 207 nm,and the injection volume was 10 μL. To determine the contents of liquiritin and glycyrrhizic acid,0.05% phosphoric acid water (A) and acetonitrile (B) were used as mobile phases with gradient elution. The detection wavelength was 237 nm. The transfer rate was calculated,and the transfer analysis from the decoction pieces to the substances benchmarks was carried out.

Results:

The fingerprint similarities of 15 batches of Daqinglong decoction substances were ≥0.921. Fifteen common peaks were identified,including 8 from Ephedrae Herba,3 from Cinnamomi Ramulus,2 from Glycyrrhizae Radix et Rhizoma(fried),and 2 from Armeniacae Semen Amarum(here). The contents of ephedrine,pseudoephedrine,amygdalin,liquiritin,cinnamic acid and glycyrrhizic acid were 0.94%-1.30%,0.56%-1.02%,0.70%-1.25%,0.18%-0.32%,0.05%-0.10% and 0.46%-1.23%,respectively. The transfer rates were 41.67%-59.47%,42.66%-59.74%,17.59%-36.34%,21.48%-44.75%,31.06%-48.89% and 12.95%-25.15%,respectively. The paste yields of substances benchmarks were 11.98%-13.38%.

Conclusion:

The fingerprint combined with the determination of paste rates and index component contents are used to study the quantitative value transfer of Daqinglong decoction substance benchmarks,and initially establish a scientific and stable substance benchmarks quality evaluation method,which can provide reference for the future research on compound preparation.

目的:

建立大青龙汤物质基准指纹图谱,测定其6个指标成分的含量及其转移率,研究大青龙汤物质基准量值传递规律,为大青龙汤复方制剂和质量标准研究奠定基础。

方法:

制备15批大青龙汤物质基准,测定出膏率;建立HPLC法,采用Dikma Platisil C18(250 mm×4.6 mm,5 μm)色谱柱,以0.15%磷酸水(A)-乙腈(B)为流动相,梯度洗脱,体积流量1 mL·min-1,柱温25 ℃,检测波长207 nm,进样体积10 μL,测定物质基准麻黄碱、伪麻黄碱、苦杏仁苷、肉桂酸的含量;以0.05%磷酸水(A)-乙腈(B)为流动相,梯度洗脱,检测波长237 nm,测定物质基准甘草苷、甘草酸的含量,计算其转移率,开展饮片到物质基准量值传递分析。

结果:

15批大青龙汤物质基准指纹图谱相似度≥0.921,图谱标定了15个共有峰,其中8个来自麻黄,3个来自桂枝,2个来自炒甘草,2个来自燀苦杏仁;指标性成分麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、肉桂酸、甘草酸的质量分数分别为0.94%~1.30%、0.56%~1.02%、0.70%~1.25%、0.18%~0.32%、0.05%~0.10%、0.46%~1.23%,转移率分别为41.67%~59.47%、42.66%~59.74%、17.59%~36.34%、21.48%~44.75%、31.06%~48.89%、12.95%~25.15%,物质基准出膏率为11.98%~13.38%。

结论:

指纹图谱结合出膏率和指标成分含量测定对大青龙汤物质基准进行量值传递研究,初步建立了科学稳定的物质基准质量评价方法,为后期复方制剂的研究提供参考。

目的:

建立大青龙汤物质基准指纹图谱,测定其6个指标成分的含量及其转移率,研究大青龙汤物质基准量值传递规律,为大青龙汤复方制剂和质量标准研究奠定基础。

方法:

制备15批大青龙汤物质基准,测定出膏率;建立HPLC法,采用Dikma Platisil C18(250 mm×4.6 mm,5 μm)色谱柱,以0.15%磷酸水(A)-乙腈(B)为流动相,梯度洗脱,体积流量1 mL·min-1,柱温25 ℃,检测波长207 nm,进样体积10 μL,测定物质基准麻黄碱、伪麻黄碱、苦杏仁苷、肉桂酸的含量;以0.05%磷酸水(A)-乙腈(B)为流动相,梯度洗脱,检测波长237 nm,测定物质基准甘草苷、甘草酸的含量,计算其转移率,开展饮片到物质基准量值传递分析。

结果:

15批大青龙汤物质基准指纹图谱相似度≥0.921,图谱标定了15个共有峰,其中8个来自麻黄,3个来自桂枝,2个来自炒甘草,2个来自燀苦杏仁;指标性成分麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、肉桂酸、甘草酸的质量分数分别为0.94%~1.30%、0.56%~1.02%、0.70%~1.25%、0.18%~0.32%、0.05%~0.10%、0.46%~1.23%,转移率分别为41.67%~59.47%、42.66%~59.74%、17.59%~36.34%、21.48%~44.75%、31.06%~48.89%、12.95%~25.15%,物质基准出膏率为11.98%~13.38%。

结论:

指纹图谱结合出膏率和指标成分含量测定对大青龙汤物质基准进行量值传递研究,初步建立了科学稳定的物质基准质量评价方法,为后期复方制剂的研究提供参考。

Objective:

To establish the substances benchmarks fingerprint of Daqinglong decoction and determine the content and transfer rate of 6 index components,and study the transfer rule of Daqinglong decoction substances benchmarks value,which laid the foundation for the study of Daqinglong decoction compound preparation and quality standard.

Methods:

Fifteen batches of substances benchmarks of Daqinglong decoction were prepared,and the paste rate was determined. To determine the contents of ephedrine,pseudoephedrine,amygdalin,and cinnamic acid,HPLC method was performed on a Dikma Platisil C18 column (250 mm×4.6 mm,5 μm). And 0.15% phosphoric acid water (A) and acetonitrile (B) were used as mobile phases with gradient elution at a flow rate of 1 mL·min-1. The column temperature was 25 ℃,the detection wavelength was 207 nm,and the injection volume was 10 μL. To determine the contents of liquiritin and glycyrrhizic acid,0.05% phosphoric acid water (A) and acetonitrile (B) were used as mobile phases with gradient elution. The detection wavelength was 237 nm. The transfer rate was calculated,and the transfer analysis from the decoction pieces to the substances benchmarks was carried out.

Results:

The fingerprint similarities of 15 batches of Daqinglong decoction substances were ≥0.921. Fifteen common peaks were identified,including 8 from Ephedrae Herba,3 from Cinnamomi Ramulus,2 from Glycyrrhizae Radix et Rhizoma(fried),and 2 from Armeniacae Semen Amarum(here). The contents of ephedrine,pseudoephedrine,amygdalin,liquiritin,cinnamic acid and glycyrrhizic acid were 0.94%-1.30%,0.56%-1.02%,0.70%-1.25%,0.18%-0.32%,0.05%-0.10% and 0.46%-1.23%,respectively. The transfer rates were 41.67%-59.47%,42.66%-59.74%,17.59%-36.34%,21.48%-44.75%,31.06%-48.89% and 12.95%-25.15%,respectively. The paste yields of substances benchmarks were 11.98%-13.38%.

Conclusion:

The fingerprint combined with the determination of paste rates and index component contents are used to study the quantitative value transfer of Daqinglong decoction substance benchmarks,and initially establish a scientific and stable substance benchmarks quality evaluation method,which can provide reference for the future research on compound preparation.

大青龙汤出自张仲景著《伤寒论》[1],由麻黄(去节)、桂枝(去皮)、甘草(炙)、杏仁(去皮尖)、生姜(切)、大枣(擘)、石膏(碎)组成,临床常用于治疗小儿哮喘[2-3]、小儿急性支气管炎、流感、暑热、急性肾炎、原发性皮肤样变、嗜睡等病症[4-6]。有研究表明[7],大青龙汤可以有效缓解感染新型冠状病毒肺炎人群出现的恶寒、咳嗽、胸闷、乏力、纳差、烦躁不安、肌肉疼痛等不适症状。此方自古至今一直被广泛使用,代代相传,久用不衰,故对其开发研究意义重大。
目前,与大青龙汤有关的研究多集中于临床应用,尚未见有关质量控制研究和物质基准量值传递研究的文献报道。本实验参考文献[8],以古籍记载方法制备15批物质基准煎液,构建其指纹图谱,并对指标性成分麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、甘草酸、肉桂酸进行含量测定,确定其含量及转移率范围,并结合出膏率分析饮片-物质基准量值传递规律,为其质量研究提供科学依据,也为大青龙汤复方制剂进一步研究提供参考。
LC-2030型高效液相色谱仪(岛津公司),AB135-S型十万分之一天平(梅特勒-托利多公司),FA1204B万分之一分析天平(上海精密仪器科技有限公司),KQ-250B型超声清洗机(昆山市超声仪器有限公司),SCIENTZ-12N型冷冻干燥机(宁波新芝生物科技股份有限公司)。
对照品盐酸麻黄碱(批号171241-201809)和盐酸伪麻黄碱(批号 171237-201510)均购于中国食品药品检定研究院,肉桂酸(批号VIP(CT)700287)、苦杏仁苷(批号YJ-110820)、甘草苷(批号YJ-111610)、甘草酸(批号VIP(CT)900069)均购于上海源叶生物科技有限公司,各对照品质量分数均≥98%。乙腈和磷酸均为色谱纯(北京化工厂),水为屈臣氏纯净水,其他试剂均为分析纯。
方中所用饮片信息如表1所示,均购于长春药材市场,并经长春中医药大学高红梅教授鉴定,麻黄为草麻黄Ephedra sinica Stapf的干燥草质茎,桂枝为樟科植物肉桂Cinnamomum cassia Presl的干燥嫩枝,炙甘草为豆科植物甘草Glycyrrhiza uralensis Fisch.的干燥根和根茎的炮制加工品,燀苦杏仁为蔷薇科植物杏Prunus armeniaca L.的干燥成熟种子炮制加工品,大枣为鼠李科植物枣Ziziphus jujuba Mill.的干燥成熟果实,生姜为姜科植物姜Zingiber officinale Rosc.的新鲜根茎。方中甘草原文记载为“炙甘草”,经考证该炮制品为炒甘草[9-12],炮制方法为取甘草片,照清炒法(炮制通则)炒至深黄色。
采用随机数表法,对表1中各饮片进行随机组合并排序,组合信息见表2。经文献考证[13]及度量衡换算,得到大青龙汤物质基准制备方法:称取麻黄82.8 g,桂枝27.6 g,炒甘草27.6 g,燀苦杏仁12 g,生姜41.4 g,大枣30.0 g,石膏60.0 g;1 800 mL水用武火(2 200 W)加热至沸腾,先煮麻黄,后用文火(800 W)减400 mL水,去沫,放入其他饮片,煮取至600 mL,用2层200目纱布趁热滤过,-18 ℃冻存12 h后冷冻干燥72 h,得大青龙汤物质基准冻干粉(J1~J15)。同法制备除石膏外各单味饮片的单煎液冻干粉及缺麻黄、桂枝、甘草(炒)、燀苦杏仁的阴性溶液冻干粉。
采用Dikma Platisil C18色谱柱(250 mm×4.6 mm,5 μm),以0.1%磷酸水(A)-乙腈(B)为流动相,梯度洗脱(0~20 min,6%B→12%B;20~30 min,12%B→18%B;30~45 min,18%B→20%B;45~50 min,20%B→24%B;50~80 min,24%B→65%B),体积流量1 mL·min-1,柱温35 ℃,检测波长207 nm,进样体积10 μL。
精密称取冻干粉0.1 g,置于100 mL具塞锥形瓶中,精密加入80%甲醇水30 mL,超声(功率250 W,频率40 kHz)20 min,放冷后用80%甲醇水补足减失的量,滤过,取续滤液,过0.22 μm微孔滤膜,即得。
精密称取盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、甘草苷、甘草酸、肉桂酸的对照品适量,加甲醇溶解,制成质量浓度分别为19.15、19.53、24.36、20.26、24.57、22.81 μg·mL-1的混合对照品溶液。
取编号J1的物质基准样品,按“2.2.2”项下方法制备供试品溶液,按“2.2.1”项下色谱条件连续测定6次,以甘草苷为参照(s),计算各共有峰的相对保留时间和相对峰面积。结果15个共有峰相对保留时间的RSD均≤0.20%,相对峰面积的RSD均≤2.1%,表明该方法精密度良好。
取编号J1的物质基准样品,按“2.2.2”项下方法制备供试品溶液,按“2.2.1”项下色谱条件,分别于0、2、4、8、12、24 h进样测定,以甘草苷为参照(s),计算各共有峰的相对保留时间和相对峰面积。结果15个共有峰相对保留时间的RSD均≤0.73%,相对峰面积的RSD均≤2.6%,表明供试品溶液在24 h内稳定性良好。
取编号J1的物质基准样品,按“2.2.2”项下方法制备供试品溶液,共6份,按“2.2.1”项下色谱条件进样测定,以甘草苷为参照(s),计算各共有峰的相对保留时间和相对峰面积。结果15个共有峰相对保留时间的RSD均≤0.36%,相对峰面积的RSD均≤2.1%,表明该方法重复性良好。
表2中不同批次的饮片组合,按“2.2.2”项下方法制备15批大青龙汤基准样品的供试品溶液,按“2.2.1”项下色谱条件测定,记录色谱图。将J1~J15基准样品特征图谱以cdf格式分别导入《中药色谱指纹图谱相似度评价系统》(2012版),以J1特征图谱为对照,采用中位数法,时间窗宽度为0.1 min,进行多点校正,生成大青龙汤物质基准样品对照图谱(R),结果见图1。J1~J15批物质基准样品相似度结果见表3
通过比对基准样品与各单味饮片、阴性供试品图谱(图2),将各特征峰进行归属,全处方共归属15个特征峰,其中峰1(麻黄碱)、峰2(伪麻黄碱)、峰3、峰4、峰7、峰8、峰9、峰11归属于麻黄;峰5、峰6(苦杏仁苷)归属于燀苦杏仁;峰12、峰13、峰14(肉桂酸)归属于桂枝;峰10(甘草苷)、峰15(甘草酸)归属于甘草(炒)。
以每味药中的1个特征峰为参照峰,计算相对峰面积,比较物质基准和各单味饮片中特征峰峰面积的比值,结果见表4,大部分特征峰的比值在饮片与基准样品间变化较小,表明饮片单煎与全方合煎时各特征峰间比例关系相似,不同成分间的含量比例关系较稳定,且归属关系清晰,表明饮片中的物质能较为稳定地传递到基准样品。
采用Dikma Platisil C18(250 mm×4.6 mm,5 μm)色谱柱,以0.15%磷酸水(A)-乙腈(B)为流动相,梯度洗脱(0~5 min,4%B→7%B;5~15 min,7%B→8B;15~17 min,8%B→9%B;17~25 min,9%B→11%B;25~35 min,11%B→11.5%B,35~40 min,11.5%B→17%B;40~55 min,17%B→23%B;55~68 min,23%B→28%B;68~75 min,28%B→29%B),检测波长207 nm,测定麻黄碱、伪麻黄碱、苦杏仁苷、肉桂酸;以0.05%磷酸水(A)-乙腈(B)为流动相,梯度洗脱(0~8 min,19%B;8~35 min,19%B→50%B;35~36 min,50%B→100%B;36~40 min,100%B→19%B),检测波长237 nm,测定甘草苷、甘草酸,体积流量1 mL·min-1,柱温25 ℃,进样体积10 μL,物质基准和对饮片的色谱图见图3
(1)取本品冻干粉0.1 g,精密称定,置于100 mL具塞锥形瓶中,密塞,称量,精密加入80%甲醇水30 mL,超声(功率250 W,频率40 kHz)处理20 min,放冷,用80%甲醇水补足减失的量,摇匀,滤过,取续滤液,过0.22 μm微孔滤膜,即得供试品溶液(1),供测定麻黄碱、伪麻黄碱、苦杏仁苷、肉桂酸用。
(2)取本品冻干粉约0.2 g,精密称定,置于150 mL具塞锥形瓶中,密塞,称量,精密加入70%乙醇100 mL,超声(功率250 W,频率40 kHz)处理30 min,放冷,用70%乙醇补足减失的量,摇匀,滤过,取续滤液,过0.22 μm微孔滤膜,即得供试品溶液(2),供测定甘草苷、甘草酸用。
精密称取盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、肉桂酸的对照品适量,用甲醇充分溶解,配成以上4个成分质量浓度分别为0.95、0.95、1.10、1.20 mg·mL-1的混合对照品母液,0.22 μm滤膜滤过,取续滤液,即得对照品溶液(1),供测定麻黄碱、伪麻黄碱、苦杏仁苷、肉桂酸用。
精密称取甘草苷、甘草酸的对照品适量,加入70%乙醇溶解,制得甘草苷、甘草酸质量浓度分别为1.20、1.10 mg·mL-1的混合对照品母液,0.22 μm滤膜滤过,取续滤液,即得对照品溶液(2),供测定甘草苷、甘草酸用。
精密吸取“2.3.3”项下对照品溶液(1),用甲醇稀释,制成麻黄碱质量浓度为4.75、9.5、19、38、57、76 μg·mL-1,伪麻黄碱质量浓度为2.375、4.75、9.5、19、38、57 μg·mL-1,苦杏仁苷质量浓度为1.1、2.75、5.5、11、22、44 μg·mL-1,肉桂酸质量浓度为0.6、1.2、3、6、12、24 μg·mL-1的溶液;精密吸取“2.3.3”项下对照品溶液(2),用70%乙醇进行稀释,制成甘草苷质量浓度为0.6、1.2、3、6、12、24 μg·mL-1,甘草酸质量浓度为0.55、1.1、2.75、5.5、11、22 μg·mL-1的溶液。按照“2.3.1”项下色谱条件进样测定,以对照品质量浓度X为横坐标,峰面积Y为纵坐标,绘制标准曲线,计算回归方程。结果见表5
取混合对照品溶液,按“2.3.1”项下色谱条件,连续进样6次测定,麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、肉桂酸、甘草酸峰面积的RSD分别为0.22%、0.20%、0.54%、0.31%、0.44%、0.38%,表明仪器精密度良好。
取大青龙汤基准样品(J1)供试品溶液,按“2.3.1”项下色谱条件,分别于制备后0、2、4、8、12、24 h进样测定,麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、肉桂酸、甘草酸峰面积的RSD分别为1.1%、1.2%、1.5%、1.2%、0.98%、1.3%,表明供试品溶液24 h内稳定性良好。
取大青龙汤基准样品(J1)供试品溶液,按“2.3.2”项下平行制备6份供试品溶液,按“2.3.1”项下色谱条件进样分析,麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、肉桂酸、甘草酸的平均含量分别为0.96%、0.58%、1.09%、0.19%、0.071%、0.56%,RSD分别为1.1%、1.8%、1.6%、2.0%、2.1%、1.8%,表明该方法重复性良好。
称取已知指标成分含量的大青龙汤物质基准(J1) 6份,按照对照品和样品的含量之比为1:1加入对照品溶液,按“2.3.2”项下方法制备供试溶液,按照“2.3.1”项下色谱条件进样测定各指标成分含量,计算加样回收率,结果麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、肉桂酸、甘草酸的加样回收率分别为98.9%、98.7%、99.6%、100.0%、98.7%、99.4%,RSD分别为0.37%、0.63%、0.51%、0.77%、0.63%、0.79%,表明方法加样回收率良好。
取麻黄、桂枝、炒甘草、燀苦杏仁单味饮片的供试溶液和大青龙汤物质基准(J1~J15)供试品溶液,按照“2.3.1”项下色谱条件进样分析,计算大青龙汤物质基准和单味饮片中6个指标成分的含量,并考察其在饮片-物质基准中的转移率,结果见表67。4个指标性成分的转移率均在均值的70%~130%,无离散数据,说明15批物质基准的制备方法稳定可行。
15批大青龙汤物质基准冻干后记录质量,计算实际出膏率。取15批处方量的麻黄、桂枝、炒甘草、燀苦杏仁、生姜、大枣饮片和石膏,按“2.1”项下方法分别制备各单味饮片水煎液,冻干,记录质量。
式中,m表示冻干粉的质量,M表示各组称取的饮片量。
全方共281.4 g。全方的理论出膏率应为各单味药折算,即理论出膏率=82.8/281.4×对应批次麻黄饮片的出膏率+27.6/281.4×对应批次桂枝饮片的出膏率+27.6/281.4×对应批次炒甘草饮片的出膏率+12/281.4×对应批次燀苦杏仁饮片的出膏率+41.4/281.4×对应批次生姜饮片的出膏率+60.0/281.4×对应批次石膏饮片的出膏率+30.0×对应批次大枣饮片的出膏率,计算结果见表8。15批大青龙汤物质基准出膏率在11.98%~13.38%,平均出膏率为12.76%,各批次出膏率都处于均值的±30%。
实验分别考察了甲醇-0.05%磷酸水溶液、甲醇-0.1%磷酸水溶液、乙腈-0.05%磷酸水溶液以及乙腈- 0.1%磷酸水溶液4种不同的流动相体系和比例下色谱峰的分离情况,结果显示,乙腈-0.1%磷酸水溶液为流动相时,色谱峰分离度和峰形较好,基线平稳。采用DAD检测器对大青龙汤波长(207、210、290、237 nm)进行优选,结果表明,检测波长在207 nm时色谱峰较多,整体信息较为全面。综上,最终选择乙腈-0.1%磷酸水溶液为流动相,207 nm作为指纹图谱的色谱条件。
对15批大青龙汤物质基准进行测定,结果表明各批次物质基准的指纹图谱相似度良好,均大于0.90。全方标定的15个特征峰,能归属到麻黄、桂枝、炒甘草和燀苦杏仁4味饮片,同时指认了麻黄碱、伪麻黄碱、苦杏仁苷、甘草苷、甘草酸、肉桂酸6个成分,而生姜和大枣在全方特征图谱中未见有特征峰的体现。分析原因可能是生姜和大枣在该检测条件下的成分含量低,峰存在响应值太低。课题组后期将采用其他适宜的方法对这2味药进一步进行分析,以弥补这2味药在HPLC研究中的缺失,全方位对大青龙汤质量控制进行研究。
对指标性成分的含量及转移率研究的过程中发现,15批物质基准样品中麻黄碱、伪麻黄碱的转移率较高,而苦杏仁苷、甘草苷、肉桂酸和甘草酸的转移率相对较低,6个成分转移率的高低反映了不同药味成分间可能会相互作用[14-15]。进一步分析发现,麻黄碱、伪麻黄碱、苦杏仁苷和肉桂酸转移率在其均值的70%~130%,说明前期麻黄、桂枝、燀苦杏仁饮片及其物质基准样品的制备工艺相对稳定可行。但是部分批次物质基准的甘草苷和甘草酸质量分数未在其均值的70%~130%。研究表明[16-18],不同产地、不同批次的甘草饮片含量存在较大差异,且甘草不同部位总黄酮含量及甘草苷含量也存在差异。因此,在制备物质基准样品时,建议严格把控甘草饮片的质量,严格按照饮片部位及大小进行分类,或者采取不同产地、不同批次甘草饮片混批投料方式,以确保大青龙汤物质基准质量的稳定。
分析大青龙汤物质基准的出膏率及转移率,平均出膏率为12.76%,平均转移率为64.51%,均处于均值的±30%,较为稳定。物质基准样品实际出膏率均低于对应饮片的理论出膏率,推测原因是单味药单煎与全方合煎水量一致,但全方的药味多,吸水量大,导致全方的实际料液比小于单味药的水量,因此实际出膏率低于理论实际出膏率[19]
本实验采用指纹图谱、指标性成分含量以及出膏率相结合的模式,对大青龙汤物质基准的量值传递过程进行分析,结果15批大青龙汤物质基准指纹图谱相似度较高,大部分指标性成分转移率及出膏率在均值±30%范围内,证明大青龙汤物质基准在制备过程中其6个指标成分的含量、指纹图谱相似度及出膏率均稳定。可为大青龙汤的后续开发及相关制剂的质量控制提供参考依据。
