Objective: To establish the UHPLC fingerprint and quantitative analysis of CBDA, CBD, THC, and THCA for hemp, and provides a basis for the quantity control of hemp. Methods: UHPLC was launched on a Phenomenex C18 (150 mm×4.6 mm, 2.6 μm) with mobile phase of acetonitrile-0.2% phosphoric acid aqueous solution at a flow rate of 1.0 mL · min-1, column temperature of 35 ℃, detection wavelength of 220 nm, and an injection volume of 2 μL by gradient elution. 15 batches of hemp samples from Daxing ’anling region and Qiqihar were analyzed to establish the fingerprint. Cluster analysis, principal component analysis and orthogonal partial least squares discriminant analysis were adopted to assess of UHPLC fingerprint of hemp. Results: the UHPLC fingerprint of hemp was established, and 26 common peaks were selected the 26 common peaks were compared with the reference substance. The structures of four chromatographic peaks were identified and simultaneously analyzed for content determination. four chromatographic peaks were identified as CBDA, CBD, THC and THCA.The similarity of fingerprint of them was greater than 0.950. The samples from Daxing’anling were clustered into class Ⅰ, with the best quality. The results show that hemp samples from different regions were quite different. The variable importance projection (VIP) showed that the four compounds to be tested were the main components leading to the difference of hemp. Conclusion: The established method is stable and reliable and can be used for qualitative and quantitative analysis of hemp. The findings of this study are expected to provide a scientific basis for quality control of hemp.

目的:建立汉麻超高效液相色谱(UHPLC)指纹图谱及大麻二酚酸(CBDA)、大麻二酚(CBD)、四氢大麻酚(THC)、四氢大麻酚酸(THCA)4个成分含量同时测定的分析方法,为汉麻的质量评价提供依据。方法:采用Phenomenex C18 色谱柱(150 mm×4.6 mm,2.6 μm),流动相为乙腈-0.2%磷酸水溶液,流速为1.0 mL · min-1,柱温为35 ℃,检测波长为220 nm,进样量为2 μL,梯度洗脱;使用《中药色谱指纹图谱相似度评价系统(2012版)》对大兴安岭地区和齐齐哈尔地区的15批汉麻进行指纹图谱分析,采用聚类分析、主成分分析和正交偏最小二乘判别分析,对汉麻UHPLC指纹图谱进行分析评价。结果:建立了汉麻UHPLC指纹图谱,标定了26个共有峰,指认了其中4个色谱峰并进行了含量测定,分别为CBDA、CBD、THC、THCA,其相似度均> 0.950。其中,大兴安岭地区样品聚为Ⅰ类,质量最好。表明不同产地汉麻样品有较大差异。26个共有峰经对照品比对,且由变量重要性投影可知,4个被指认成分均是导致汉麻差异的主要成分。结论:该方法稳定可靠,可用于汉麻的定性定量分析,研究结果为汉麻的质量评价提供依据。

目的:建立汉麻超高效液相色谱(UHPLC)指纹图谱及大麻二酚酸(CBDA)、大麻二酚(CBD)、四氢大麻酚(THC)、四氢大麻酚酸(THCA)4个成分含量同时测定的分析方法,为汉麻的质量评价提供依据。方法:采用Phenomenex C18 色谱柱(150 mm×4.6 mm,2.6 μm),流动相为乙腈-0.2%磷酸水溶液,流速为1.0 mL · min-1,柱温为35 ℃,检测波长为220 nm,进样量为2 μL,梯度洗脱;使用《中药色谱指纹图谱相似度评价系统(2012版)》对大兴安岭地区和齐齐哈尔地区的15批汉麻进行指纹图谱分析,采用聚类分析、主成分分析和正交偏最小二乘判别分析,对汉麻UHPLC指纹图谱进行分析评价。结果:建立了汉麻UHPLC指纹图谱,标定了26个共有峰,指认了其中4个色谱峰并进行了含量测定,分别为CBDA、CBD、THC、THCA,其相似度均> 0.950。其中,大兴安岭地区样品聚为Ⅰ类,质量最好。表明不同产地汉麻样品有较大差异。26个共有峰经对照品比对,且由变量重要性投影可知,4个被指认成分均是导致汉麻差异的主要成分。结论:该方法稳定可靠,可用于汉麻的定性定量分析,研究结果为汉麻的质量评价提供依据。

Objective: To establish the UHPLC fingerprint and quantitative analysis of CBDA, CBD, THC, and THCA for hemp, and provides a basis for the quantity control of hemp. Methods: UHPLC was launched on a Phenomenex C18 (150 mm×4.6 mm, 2.6 μm) with mobile phase of acetonitrile-0.2% phosphoric acid aqueous solution at a flow rate of 1.0 mL · min-1, column temperature of 35 ℃, detection wavelength of 220 nm, and an injection volume of 2 μL by gradient elution. 15 batches of hemp samples from Daxing ’anling region and Qiqihar were analyzed to establish the fingerprint. Cluster analysis, principal component analysis and orthogonal partial least squares discriminant analysis were adopted to assess of UHPLC fingerprint of hemp. Results: the UHPLC fingerprint of hemp was established, and 26 common peaks were selected the 26 common peaks were compared with the reference substance. The structures of four chromatographic peaks were identified and simultaneously analyzed for content determination. four chromatographic peaks were identified as CBDA, CBD, THC and THCA.The similarity of fingerprint of them was greater than 0.950. The samples from Daxing’anling were clustered into class Ⅰ, with the best quality. The results show that hemp samples from different regions were quite different. The variable importance projection (VIP) showed that the four compounds to be tested were the main components leading to the difference of hemp. Conclusion: The established method is stable and reliable and can be used for qualitative and quantitative analysis of hemp. The findings of this study are expected to provide a scientific basis for quality control of hemp.

汉麻为桑科(Moraceae)大麻属(Cannabis)一年生草本植物,多雌雄异株[1]。药用汉麻植株中四氢大麻酚(THC)含量低于0.3%,不具备提取THC或直接作为毒品吸食的价值[2],在我国各地均有分布,但仅黑龙江、云南两省允许规模化种植[3]。汉麻的药理作用研究历史悠久,早在《神农本草经》中记载“麻蕡,一名麻勃,主治五劳七伤,利五藏,下血,寒气,久服通神明,轻身”[4]。汉麻含有大麻素、黄酮、萜、生物碱等多种类成分。大麻素类成分在镇痛[5]、抗肿瘤[6]、神经保护[7]、抗菌[8]、抗炎[9]等方面具有良好的药理活性,因大麻二酚(CBD)对治疗神经系统和心血管疾病等具有显著疗效,所以国内外对于CBD的研究最为深入[10]
汉麻品种复杂多样且含有多种成分,受地理及生长环境等因素的影响,汉麻质量参差不齐。目前,有通过重测序技术构建汉麻指纹图谱,评价不同汉麻样品间的遗传多样性[11]。通过SDS-PAGE电泳技术构建火麻仁蛋白指纹图谱,用于分析抗氧化能力与植物栽培品种和环境的关系[12],而关于工业大麻HPLC指纹图谱的研究相对较少,但有关于工业大麻化学成分检测方法非常丰富[13-15],这为工业大麻的质量控制和指纹图谱的建立提供了技术支持。因此,为了探究不同产地汉麻质量差异,客观评价汉麻整体质量,本实验采用UHPLC方法,建立15批不同地区汉麻的指纹图谱,并应用聚类分析(CA)、主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)等分析方法对大兴安岭地区和齐齐哈尔地区汉麻样品进行统计分析。通过与照品比对,对汉麻中的主要特征成分CBDA、CBD、THC 和THCA 进行含量测定。通过汉麻指纹图谱的建立及上述4个主成分的含量测定,比较不同产地汉麻品质的差异,为汉麻指纹图谱的建立及质量评价提供依据。
Agilent 1290型超高效液相色谱仪(安捷伦公司),色谱柱为Phenomenex C18(150 mm×4.6 mm,2.6 μm)(飞诺美公司),ZA220R4 型电子天平(万分之一)(上海赞维衡器有限公司),JP-060型超声波清洗器(深圳洁盟技术股份有限公司),中草药粉碎机FW177(天津市泰斯特仪器有限公司),L530台式低速离心机(湖南湘仪实验室仪器开发有限公司)。
对照品 CBDA(批号FE04301903)、CBD(批号FE10071912)、Δ9-THC(批号FE02072001)、Δ9-THCA(批号FE01142103)购于西格玛公司(纯度98%),15批样品,其中黑龙江省大兴安岭地区11批(C1、C2、C3、C4、C5、C6、C7、C8、C10、C11和C14),齐齐哈尔市4批(C9、C12、C13和C15),采收后自然阴干经齐齐哈尔大学天然产物研究室张树军教授鉴定为汉麻(hemp),标本保存在齐齐哈尔大学天然产物研究室。乙腈、甲醇为色谱纯,北京伊诺凯科技有限公司;磷酸为分析纯,上海阿拉丁生化科技股份有限公司;水为屈臣氏蒸馏水。
采用Phenomenex C18色谱柱(150 mm×4.6 mm,2.6 µm),流动相为乙腈(A)-0.2% 磷酸水溶液(B),梯度洗脱(0~20 min,13%A→28%A;20~26 min,28%A→45%A;26~35 min,45%A→50%A;35~45 min,50%A;45~70 min,50%A→70%A;70~75 min,70%A→73%A,75~85 min,13%A),流速为 1.0 mL · min-1,柱温为 35 ℃,检测波长为 220 nm,进样量为 2 μL。
将CBD、THC和THCA标准品溶液加甲醇分别稀释成制成对照品储备液,对照品储备液浓度分别为CBDA 1 000 µg · mL-1、CBD 500 µg · mL-1、THC 40 µg · mL-1、THCA 100 µg · mL-1
取15批汉麻样品粉末(过5号筛)约1 g,精密称定后分别置于具塞锥形瓶中,加入甲醇50 mL,称定质量,超声(300 W,40 kHz)提取30 min,甲醇补足减失的量,在3 600 r · min-1离心5 min,取上清液用0.22 μm 滤膜过滤,取续滤液,即得。
取C1样品,按“2.2.2”项下方法制备供试品溶液,在“2.1”项色谱条件下进样6次测定,测得各共有峰相对峰面积(relative common peak area,RPA)的RSD<2.2%,相对保留时间(relative retention time,RRT)的RSD<0.22%,结果表明该仪器精密度良好。
取C1样品6份,按“2.2.2”项下方法制备供试品溶液,在“2.1”项色谱条件下进样测定,测得各共有峰RPA的RSD<2.1%,RRT的RSD<0.14%,结果表明该检测方法重复性良好。
取C1样品,按“2.2.2”项下方法制备供试品溶液,室温下暗处放置0、2、4、8、12、24 h,在“2.1”项色谱条件下进样测定,测得各共有峰RPA的RSD<2.6%,RRT的RSD<0.13%,结果表明供试品溶液在24 h内稳定性良好。
取15批汉麻样品,按“2.2.2”项下方法制备供试品溶液,在“2.1”项色谱条件下进样测定得UHPLC图谱,将UHPLC数据依次导入《中药色谱指纹图谱相似度评价系统(2012版)》,建立汉麻指纹图谱,以C1为参照图谱,设定时间窗宽度为0.1 min,通过Mark峰匹配与多点校正,建立汉麻指纹图谱,见图1。15批汉麻样品指纹图谱共有26个共有峰,各共有峰相对保留时间的RSD<1.0%。通过与对照品保留时间相比,指认了其中四个色谱峰分别为峰16(CBDA)、峰17(CBD)、峰21(Δ9-THC)、峰24(Δ9-THCA)。
以“2.3.4”项建立的汉麻指纹图谱C1的UHPLC谱图为参照,经多点校正和Mark峰匹配后计算相似度,各批样品UHPLC色谱图相似度在0.951~0.999,表明2个地区汉麻样品化学成分组成基本相同,差异较小,建立的指纹图谱具有代表性,可用于汉麻质量评价。
以15批汉麻样品指纹图谱共有峰的峰面积为变量,使用Origin软件进行聚类分析(CA)。使用Z-得分对数据进行标准化处理后,采用Ward聚类方法和欧氏距离进行CA,结果见图2。当分类距离为10时,C1、C2、C3、C4、C5和C6聚为第1类,C7、C8、C10、C11、C14聚为第2类,是大兴安岭地区汉麻样品;C9、C12、C13和C15聚为第3类,是齐齐哈尔产大麻样品。说明不同产区汉麻样品差异相对较大,可通过指纹图谱辨别不同产区汉麻。
为客观反映15批汉麻样品产区间的差异性,以15批汉麻样品指纹图谱的共有峰峰面积为变量,将数据结果导入SIMCA 14.1软件进行主成分分析(PCA)。将汉麻样品峰面积归一化处理后,采用PCA模型自动拟合为PCA得分图,见图3。由图3可知,汉麻样品可明显分为3类:C1、C2、C3、C4、C5和C6聚为第1类,C7、C8、C10、C11、C14聚为第2类,是大兴安岭地区汉麻样品;C9、C12、C13和C15聚为第3类,是齐齐哈尔产大麻样品。PCA结果与CA结果一致,由PCA得分图的分离程度可知,不同产区的汉麻指纹图谱之间存在一定差异,可根据PCA得分图辨识两地汉麻产区。
正交偏最小二乘判别分析(OPLS-DA)可用于寻找样品间差异的变量。因此进一步分析不同产地汉麻样品之间的差异,以15批汉麻指纹图谱的26个共有峰峰面积为变量,将数据导入SIMCA 14.1软件进行OPLS-DA,见图4。C9、C12、C13和C15为齐齐哈尔产区汉麻,其余为大兴安岭产区汉麻。由此可见,不同地区汉麻样品具有一定差异,为进一步确认对样品分类贡献度较大的成分,通过变量重要性投影(VIP)>1筛选出导致汉麻差异的标志性成分,VIP是一种在多变量统计分析中常用的方法,用于评估不同自变量对因变量的贡献程度,VIP越大,表明该化合物对组间差异的贡献越大。标记出19个贡献较大的成分,依次为16、24、17、14、10、21、18、20、22、15、19、11、7、13、5、8、2、6和12号色谱峰,结果见图5。经对照品比对指认出色谱峰16、17、21和24分别为CBDA、CBD、THC和THCA,并对这4个指标成分进行了含量测定,由此证明,这4个成分的含量会影响汉麻的质量差异。
取C1样品,按“2.2.2”项下方法制备供试品溶液。分别取“2.2.1”项下制备的对照品储备液适量,均匀混合后,制成混合对照品溶液,在“2.1”项色谱条件下进样测定,结果见图6。发现各色谱峰均具有良好的分离度和峰的对称性,并且与对照品一一对应,表示该方法适合汉麻检测。
取各对照品储备液,加甲醇逐级稀释至CBDA(1~1 000)µg · mL-1;CBD(1~500)µg · mL-1;Δ9-THC(0.1~40)µg · mL-1;Δ9-THCA(0.1~100)µg · mL-1的系列5个浓度溶液,在“2.1”项色谱条件下进样测定。以对照品质量浓度为横坐标X,峰面积为纵坐标Y,使用Excel软件绘制标准曲线,得到各指标成分回归方程。将最低浓度的对照品溶液继续逐级稀释,在“2.1”项色谱条件下进样测定,按信噪比(S/N)=10计算定量限,按信噪比(S/N)=3计算检测限,结果见表1,可知各成分在各自范围内线性关系良好。
取C1样品,按“2.2.2”项下方法制备供试品溶液,在“2.1”项色谱条件下进样测定6次,测得汉麻CBDA、CBD、THC、THCA峰面积的RSD分别为0.11%、0.12%、1.4%、0.60%,表明仪器精密度良好。
取C1样品,按“2.2.2”项下方法平行制备6份供试品溶液,在“2.1”项色谱条件下进样测定,记录峰面积,测得汉麻中CBDA、CBD、THC、THCA的平均含量分别为45.7841、23.9652、0.0650、0.1972 mg · g-1。RSD分别为0.10%、0.24%、0.69%和0.14%,表明方法的重复性良好。
取C1样品,按“2.2.2”项下方法制备供试品溶液,分别于0、2、4、8、12、24 h在“2.1”项色谱条件下进样测定,测得汉麻中4个指标成分的峰面积RSD分别为0.14%、0.64%、2.4%和0.60%,表明溶液在24 h内稳定性良好。
精密称取C9汉麻样品约0.1 g 6份,按表2中加标量加入对照品,按“2.2.2”项下方法平行制备供试品溶液。在“2.1”项色谱条件下进样测定,记录峰面积,计算各成分回收率,结果见表2,表明该方法准确性良好。
取15批样品,按“2.2.2”项下方法制备供试品溶液,在“2.1”项色谱条件下进行测定,计算含量,结果见表3
本文考察了甲醇体积分数为80%、90%、100%作为提取溶剂时对汉麻色谱图的影响,结果表明甲醇作为提取溶剂时,汉麻色谱图中色谱峰数量相对较多且峰面积最大,故选择甲醇为汉麻样品提取溶剂;考察了超声和加热回流2种提取方法,结果表明超声提取效果较好;考察了料液比为1 ∶ 40、1 ∶ 50和1 ∶ 60的提取效果,结果发现料液比1 ∶ 50提取效果最好;另外,考察了提取时间分别为20、30、40 min的提取效果,结果表明,30 min 即可充分提取汉麻主要成分,故选择超声提取时间为30 min。
通过紫外吸收光谱全波长扫描,发现检测波长为220 nm时,汉麻样品有最大吸收,且在220 nm时汉麻样品色谱图出峰数目较多,基线平稳,故选择220 nm 作为UHPLC检测波长。同时对色谱柱进行优化,比较COSMOSIL(100 mm×2.1 mm,2.6 µm)和Phenomenex C18(150 mm×4.6 mm,2.6 μm)2种色谱柱,结果Phenomenex C18色谱柱达到分离要求;考察乙腈-0.1%甲酸水、乙腈-0.1%磷酸水、乙腈-0.2%磷酸水3个不同的流动相体系,结果发现,乙腈-0.2%磷酸水体系分离效果最好。
本研究发现,15批汉麻UHPLC指纹图谱之间的相似度较高,通过对其中4个主成分进行定量测定,发现15批汉麻中CBDA(17.241 1~46.527 5 mg · g–1)、CBD(4.929 3~29.741 0 mg · g–1)是含量相对较高的2个成分,对汉麻的相关报道以及本研究的结果表明,CBDA和CBD是汉麻重要的差异性成分。THC(0.016 5~0.075 5 mg · g–1)、THCA(0.072~0.202 6 mg · g–1)符合国家法律标准,在国家允许种植范围内。CA、PCA与OPLS-DA 3种分析方法均可将汉麻样品分为3类,C9、C12、C13和C15为齐齐哈尔产区汉麻,其余为大兴安岭产区汉麻,结果一致。通过VIP分析,色谱峰9、10、14、15、16、17、18、19、20、21、22、24是影响汉麻成分差异的指标成分,因此本文选择16、17、21、24号色谱峰进行定量测定是合理的。结果发现,大兴安岭地区汉麻样品中CBDA、CBD含量高于齐齐哈尔产汉麻,可能与地区气候、土壤条件有关。汉麻指纹图谱与化学模式识别法相结合,不仅可有效评价汉麻品质,诠释影响汉麻质量差异的主要因素,还可有效辨识不同产区汉麻。
通过多种化学计量学分析方法,对不同产地汉麻进行质量评价,发现不同产地汉麻具有差异。通过对4个主成分进行含量测定,发现不同产地样品主成分含量有较大差别。综上所述,本实验建立汉麻UHPLC指纹图谱,并对4个大麻素类化合物含量进行了测定,该方法重复性好,专属性强,可为汉麻的质量评价和开发利用提供一定的依据。
