Objective: To establish a quantitative analysis of multi-components by single marker (QAMS) method to simultaneously determine the contents of scopolamine, anisodamine, scopolin, hyoscyamine and scopoletin in Physochlaina infundibularis Kuang. Methods: Agilent 5 TC-C18 (250 mm×4.6 mm, 5 μm) column was used with methanol-0.1% phosphoric acid aqueous solution as mobile phase for gradient elution at a flow rate of 1.0 mL·min-1. The detection wavelengths were 210 nm and 344 nm, the column temperature was 30 °C, and the injection volume was 10 μL. The relative correction factors for scopolamine, scopolin, hyoscyamine and scopoletin and anisodamine were calculated by using anisodamine as the internal reference substance. The contents were determined and compared with the results of external standard method (ESM). Results: The five components had good linear relationship in their respective ranges(r=1.000). The relative correction factors of scopolamine, anisodamine, scopolin, hyoscyamine and scopoletin were 0.951 8, 0.562 6, 0.830 4 and 0.433 9, respectively, with good repeatability. The contents of scopolamine, anisodamine, scopolin, hyoscyamine and scopoletin in P. infundibularis from 10 habitats calculated by QAMS method were not significantly different from those determined by ESM. Conclusion: The method is accurate, stable and reproducible, and can be used for the quality control of P. infundibularis.

目的:建立一测多评(QAMS)法同时测定华山参中东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯等成分的含量。方法:采用Agilent 5 TC-C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长为210 nm和344 nm,柱温30 ℃,进样量10 μL。以山莨菪碱为内参物,计算东莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的相对校正因子,测定其含量,并与外标法(ESM)测定结果进行比较。结果:5个成分在各自范围内线性关系良好(r=1.000),东莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的相对校正因子分别为0.951 8、0.562 6、0.830 4、0.433 9,重复性较好。QAMS法计算所得10个产地华山参中东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的含量与ESM实测值无显著性差异。结论:该方法准确稳定,重复性好,可用于华山参的质量控制。

目的:建立一测多评(QAMS)法同时测定华山参中东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯等成分的含量。方法:采用Agilent 5 TC-C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长为210 nm和344 nm,柱温30 ℃,进样量10 μL。以山莨菪碱为内参物,计算东莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的相对校正因子,测定其含量,并与外标法(ESM)测定结果进行比较。结果:5个成分在各自范围内线性关系良好(r=1.000),东莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的相对校正因子分别为0.951 8、0.562 6、0.830 4、0.433 9,重复性较好。QAMS法计算所得10个产地华山参中东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的含量与ESM实测值无显著性差异。结论:该方法准确稳定,重复性好,可用于华山参的质量控制。

Objective: To establish a quantitative analysis of multi-components by single marker (QAMS) method to simultaneously determine the contents of scopolamine, anisodamine, scopolin, hyoscyamine and scopoletin in Physochlaina infundibularis Kuang. Methods: Agilent 5 TC-C18 (250 mm×4.6 mm, 5 μm) column was used with methanol-0.1% phosphoric acid aqueous solution as mobile phase for gradient elution at a flow rate of 1.0 mL·min-1. The detection wavelengths were 210 nm and 344 nm, the column temperature was 30 °C, and the injection volume was 10 μL. The relative correction factors for scopolamine, scopolin, hyoscyamine and scopoletin and anisodamine were calculated by using anisodamine as the internal reference substance. The contents were determined and compared with the results of external standard method (ESM). Results: The five components had good linear relationship in their respective ranges(r=1.000). The relative correction factors of scopolamine, anisodamine, scopolin, hyoscyamine and scopoletin were 0.951 8, 0.562 6, 0.830 4 and 0.433 9, respectively, with good repeatability. The contents of scopolamine, anisodamine, scopolin, hyoscyamine and scopoletin in P. infundibularis from 10 habitats calculated by QAMS method were not significantly different from those determined by ESM. Conclusion: The method is accurate, stable and reproducible, and can be used for the quality control of P. infundibularis.

华山参为茄科泡囊草属植物漏斗泡囊草Physochlaina infundibularis Kuang的干燥根,始载于清代赵学敏所著的《本草纲目拾遗》,又名华山人参、热参、秦参、白毛参、醉汉草等,生于山坡、沟谷或草地,主要分布于陕西、河南、山西,已被列为国家二级保护植物[1-2]。具有温肺祛痰、止咳平喘、安神镇惊的功效,用于寒痰喘咳、惊悸失眠[3-4]。2020年版《中华人民共和国药典》分别采用紫外-可见分光光度法和HPLC法测定华山参中莨菪碱和东莨菪内酯的含量,但紫外-可见分光光度法操作复杂,准确性和重现性较差[4-5]。同时,在多种茄科植物如颠茄草、宁夏枸杞及其他科属如光叶丁公藤等植物中均含有东莨菪内酯[6-8],其专属性不足。因此,还需要借助其他分析技术和方法对华山参进行全面的质量控制。
一测多评(multi-components by single marker,QAMS)是近年来应用较广泛的一种中药质量控制方法,它是通过建立内参物与待测成分之间的相对校正因子(relative correction factors,RCF),在仅测定1个成分(内参物)的基础上实现对多种成分的同步检测的方法[9-10],可弥补中药多指标评价模式中存在的对照品数量众多,价格昂贵等不足[11-13]。目前,尚未见关于华山参一测多评法的研究报道,且在华山参质量控制方面还存在定量指标成分较少等问题。因此,本文以山莨菪碱为内参物,建立一测多评法同时测定东莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯和山莨菪碱5个成分的含量,并与外标法测定的结果进行比较,证明QAMS法的可行性,减少了对照品的使用数量,降低了检测成本,为华山参的全面质量控制提供了新思路。
Agilent 1260型高效液相色谱仪(安捷伦公司);SHIMADZU 2010A-HT型高效液相色谱仪(岛津公司);KQ5200DE型数控超声波清洗器(昆山市超声仪器有限公司);DFT-200A型手提式高速粉碎机(温岭市林大机械有限公司);BT25S型电子分析天平(d=0.01 mg),BS210S型电子分析天平(d=0.1 mg)(北京赛多利斯天平有限公司)。
10批不同产地华山参样品来源信息见表1,经陕西省中医药研究院张红研究员鉴定均为茄科泡囊草属植物漏斗泡囊草Physochlaina infundibularis Kuang的干燥根。对照品氢溴酸东莨菪碱(批号CHB210125)、氢溴酸山莨菪碱(批号CHB210118)、东莨菪内酯(批号CHB201202)均购自成都克洛玛生物科技有限公司,纯度均≥98%。东莨菪苷(批号PS010518,纯度≥98%)购自成都普思生物科技股份有限公司、莨菪碱(批号C12946842,纯度≥98%)购自上海麦克林生化科技有限公司。甲醇、乙腈为色谱纯(Thermo Fisher Scientific公司),水为超纯水,磷酸为分析纯。
分别精密称取对照品氢溴酸东莨菪碱、氢溴酸山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯适量,用10%甲醇水溶解并定容,制成质量浓度分别为0.41、0.43、0.33、0.34、0.24 mg·mL-1的对照品储备液;精密吸取上述对照品储备液适量置于同一10 mL量瓶中,加10%甲醇稀释并定容,摇匀,得质量浓度分别为41、43、33、34、24 μg·mL-1的混合对照品溶液。
取华山参药材粉末(过三号筛)约1.0 g,精密称定,置具塞锥形瓶中,精密加入40%甲醇水25 mL,密塞,称量,放置1 h,超声(功率300 W,频率40 kHz)处理1 h,放冷,再称量,用40%甲醇水补足减失的量,摇匀,滤过,取续滤液过0.45 μm微孔滤膜,即得。
采用Agilent 5 TC-C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇为流动相A,0.1%磷酸溶液为流动相B,洗脱梯度(0~5 min,10%A~13%A;5~10 min,13%A~18%A;10~33 min,18%A~26% A;33~40 min,26%A~40%A;40~45 min,40%A~10%A),柱温30 ℃,进样量10 μL,流速1.0 mL·min-1,东莨菪碱、山莨菪碱、莨菪碱检测波长为210 nm,东莨菪苷、东莨菪内酯检测波长为344 nm。在本色谱条件下,华山参中5个成分峰与相邻峰的分离度均>1.5,色谱图见图1
精密吸取混合对照品溶液适量,用10%甲醇依次倍比稀释,制成一系列不同浓度的对照品溶液,将对照品溶液按“2.2”项下色谱条件进行测定。以峰面积Y为纵坐标,质量浓度(X,μg·mL-1)为横坐标,绘制标准曲线,计算回归方程和线性范围(见表2),各成分在各自范围内线性关系良好。
精密吸取“2.1.1”项下的混合对照品溶液,按照“2.2”项下色谱条件连续进样测定6次,记录各个成分的峰面积,并计算RSD。结果表明,东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯峰面积的RSD分别为0.090%、0.22%、0.070%、0.19%、0.11%,表明仪器精密度良好。
精密称取6份S7号样品的粉末,按“2.1.2”项下方法制备供试品溶液,进样测定,测得东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的平均含量(n=6)分别为1.06、3.11、0.99、2.67、0.19 mg·g-1,RSD分别为2.5%、3.3%、1.2%、2.5%、2.2%,表明该方法重复性良好。
精密吸取同一份供试品溶液(样品编号S7),按照“2.2”项下色谱条件分别于0、2、4、6、8、12、24 h进样测定,记录各个成分的峰面积,并计算RSD。结果显示,东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯峰面积的RSD分别为2.6%、1.3%、2.0%、2.0%、0.98%,表明供试品溶液在24 h内稳定性良好。
精密称取各成分含量已知的同一批样品(样品编号S7)0.5 g,共6份,以1∶1的比例加入适量对照品溶液,按“2.1.2”项下方法制备供试品溶液,并按照“2.2”项下色谱条件进样测定,计算各成分的加样回收率及RSD。结果显示,各个成分的平均回收率在90.2%~105.8%,RSD均小于3.0%,表明该方法准确性良好。结果见表3
本试验采用多点校正法计算RCF。精密吸取“2.3.1”项下不同浓度的混合对照品溶液,按照“2.2”项下色谱条件进样,记录各个成分的峰面积,以山莨菪碱为内参物,按照公式[14-15]fs/i=fs/fi=(As/Cs)/(Ai/Ci)=(AsCi)/(AiCs)(式中fs/ifsfi分别为内参物与待测成分的RCF、内参物及待测成分的绝对校正因子,As表示内参物峰面积,Cs表示内参物的浓度,Ai表示待测成分峰面积,Ci表示待测成分的浓度),分别计算各个成分的相对校正因子(fs/i)及其RSD,结果以“均值±标准偏差”表示,见表4。待测成分与内参物RCF的RSD均小于2.0%,表明均值可作为定量用RCF。
精密量取“2.1.1”项下的混合对照品溶液适量,按“2.2”项下色谱条件进样测定,考察Agilent 1260型、岛津SHIMADZU 2010A-HT型高效液相色谱仪及Agilent 5 TC-C18、YMC-Pack ODS-A色谱柱对RCF的影响,结果见表5。测得各成分RCF的RSD均小于2.00%,说明不同色谱柱和不同仪器对所测成分的RCF无显著影响。
精密量取“2.1.1”项下的混合对照品溶液适量,采用Agilent 1260型高效液相色谱仪,Agilent 5 TC-C18色谱柱,分别考察各待测成分在不同柱温(30、35、40 ℃)下的RCF,并计算RSD,结果见表6。测得各成分RCF的RSD均小于1.0%,表明柱温对各成分RCF无显著影响。
精密量取“2.1.1”项下的混合对照品溶液适量,按“2.2”项下色谱条件进样测定,分别考察各待测成分在不同流速(0.8、1.0、1.2 mL·min-1)时的相对校正因子,并计算RSD,结果见表7。各成分RCFs的RSD均小于3.0%,表明流速对各成分RCF无显著影响。
中药多成分含量测定的QAMS,前提是一定要准确定位待测成分色谱峰,本文采用相对保留值(Rs/i=tR(s)/tR(i))法对待测化合物色谱峰进行定位[16-17]。通过测定参照物保留时间tR(s),结合已知的待测成分相对保留时间Rs/i,可求出待测成分的保留时间tR(i),实现对待测成分的准确定位。以山莨菪碱为参照物,精密吸取混合对照品溶液适量,按照“2.2”项下色谱条件进样测定,考察在不同仪器和色谱柱下测得待测成分峰的相对保留时间,结果见表8。表明相对保留时间法在不同仪器、色谱柱下稳定性良好(RSD<5.0%),可用于待测成分色谱峰的定位。
分别取10批华山参药材,按“2.1.2”项下方法制备供试品溶液,进样测定,每份测定3次,分别采用ESM和QAMS法计算含量并根据RSD比较待测成分的含量差异。根据分子式计算,氢溴酸山莨菪碱折算成山莨菪碱系数为0.790 6,氢溴酸东莨菪碱折算成东莨菪碱系数为0.789 4,待测成分质量浓度计算公式[18-19]Ci=fs/iAiCs/As,结果见表9。RSD及配对t检验结果显示,2种方法计算的含量测定结果之间的差异无统计学意义,说明本试验所建立的QAMS法准确、可行,可在只有山莨菪碱1个对照品的情况下,同时测定华山参中这5个成分的含量。此外,不同批次之间的待测成分含量的RSD为34.5%~47.6%,存在一定的差异,可能与原药材的产地、采收等因素有关,故建立多指标成分评价模式有助于确保华山参质量的稳定。
本研究选择了活性明确,含量较高的代表成分作为华山参质量控制的指标性成分进行一测多评法的含量测定,主要包括东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱和东莨菪内酯。其中,莨菪碱、山莨菪碱等莨菪烷类生物碱具有抑制中枢和抗胆碱作用[20],莨菪碱、山莨菪碱、东莨菪碱等还可以扩张支气管,治疗咳嗽、支气管哮喘等疾病[21],东莨菪内酯、东莨菪苷等香豆素类成分具有抗炎镇痛、利尿平喘的药理作用[3],上述成分均为华山参的重要活性成分。因此,所选指标成分均具有与华山参中医理论、药理作用和功能主治相符的药理特性。
内参物不仅要有明确的治疗效果和稳定的化学性质,而且需易获得,价格低廉[22-23]。山莨菪碱是华山参的主要有效成分之一,含量较高,化学性质稳定,保留时间适中,对照品价廉易得,并且色谱峰附近的干扰峰也较少,故选择其作为内参物。
本研究分别采用了40%甲醇水、60%甲醇水、80%甲醇水溶液、甲醇和水为提取溶剂,对同一批华山参药材进行提取,结果显示,40%甲醇水作为提取溶剂时出峰的个数及峰面积大小均最多,故最终选用40%甲醇水作为提取溶剂。
本研究比较了不同的超声提取时间对华山参中莨菪碱、东莨菪内酯等成分提取率的影响:超声提取0.5 h;超声提取1 h;放置1 h后超声提取1 h。结果放置1 h后,超声处理1 h的方法能较充分地将上述5种成分提取出来,因此,选择该方法制备供试品溶液。
本研究考察了甲醇-0.05%磷酸水溶液、甲醇-0.1%磷酸水溶液、甲醇-0.3%磷酸水溶液、乙腈-0.3%磷酸水溶液流动相组成,以测定的华山参中各成分的分离效果作为指标,结果发现以甲醇-0.1%磷酸水溶液洗脱时得到的色谱峰分离效果、塔板数和对称性等方面均较佳,且基线平稳,出峰时间亦较理想,因此,最终采用甲醇-0.1%磷酸水溶液为色谱流动相进行梯度洗脱。
本研究建立了一测多评法同时测定华山参中东莨菪碱、山莨菪碱、东莨菪苷、莨菪碱、东莨菪内酯的含量,通过相对校正因子耐用性的考察、待测目标成分色谱峰的准确定位以及ESM与QAMS法计算结果的对比,确定本文所采用的方法结果准确、可行,重复性好,可用于华山参的多成分定量分析,为华山参药材质量标准的建立和全面质量控制提供新的方法和思路。但不足之处在于并未将定量的5个成分与华山参的临床疗效相关联,后续可开展相关研究,以明确指标成分与疗效之间的关系,为华山参及其制剂的合理用药提供保障。
