Objective: To establish a multi-component content determination and characteristic chromatogram method for Jieyu Anshen granules, combined with chemometrics, to comprehensively evaluate the quality of Jieyu Anshen granules. Methods: The separation was performed on a CAPCELL PAK MG Ⅱ C18(250 mm×4.6 mm, 5 μm) column using acetonitrile (A) and 0.1% phosphoric acid aqueous solution (B) as the mobile phases with gradient elution at a flow rate of 1.0 mL·min-1. The column temperature was 30 ℃ and the detection wavelengths were set at 237 nm and 335 nm. The analysis method was validated. Based on the multi-component content determination method, characteristic chromatogram was established. The samples were analyzed and evaluated through chemometrics to identify the main factors affecting their quality. Results: A multi-component content determination method was established for Jieyu Anshen granules. The five components had good linear relationships within their respective ranges, with recovery rates ranging from 95.6% to 100.0%. And 145 batches of samples were tested, and a total of 54 batches were found to be unqualified, with a total unqualified rate of 37.2%. Establishing a characteristic chromatogram of Anshen granules, 12 characteristic peaks were attributed to 6 medicinal herbs. Through chemometric analysis, it was found that there were certain differences in the quality of samples from 10 manufacturers. The quality of fried gardenia and stir-baked licorice was the key factor. Conclusion: The established multi-component content determination method and characteristic chromatogram method for Jieyu Anshen granules are convenient and reliable, and can be used for comprehensive evaluation of the quality of Jieyu Anshen granules.

目的:建立解郁安神颗粒的多组分含量测定及特征图谱方法与化学计量学结合,对解郁安神颗粒质量进行整体评价。方法:采用CAPCELL PAK MGⅡC18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL·min-1,柱温30 ℃,检测波长在237 nm(栀子苷、甘草苷、甘草酸铵)和335 nm(斯皮诺素及3,6’-二芥子酰基蔗糖)5个成分进行含量测定并进行方法学验证。基于多组分含量测定方法,建立样品的特征图谱,并通过化学计量学对样品进行分析评价,找出影响样品质量的主要因素。结果:建立了解郁安神颗粒的多组分含量测定方法,5个成分在各自范围内线性关系良好,回收率在95.6%~100.0%,对145批次样品进行测定,不合格样品共54批,总不合格率为37.2%;建立了解郁安神颗粒的特征图谱,标定了12个特征峰分别归属于6味药材,通过化学计量学软件分析,10个厂家样品质量存在一定差异,炒栀子与炙甘草质量的优劣是影响样品质量的关键因素。结论:建立的解郁安神颗粒多组分含量测定方法及特征图谱方法便捷可靠,可用于解郁安神颗粒品质的综合评价。

Objective: To establish a multi-component content determination and characteristic chromatogram method for Jieyu Anshen granules, combined with chemometrics, to comprehensively evaluate the quality of Jieyu Anshen granules. Methods: The separation was performed on a CAPCELL PAK MG Ⅱ C18(250 mm×4.6 mm, 5 μm) column using acetonitrile (A) and 0.1% phosphoric acid aqueous solution (B) as the mobile phases with gradient elution at a flow rate of 1.0 mL·min-1. The column temperature was 30 ℃ and the detection wavelengths were set at 237 nm and 335 nm. The analysis method was validated. Based on the multi-component content determination method, characteristic chromatogram was established. The samples were analyzed and evaluated through chemometrics to identify the main factors affecting their quality. Results: A multi-component content determination method was established for Jieyu Anshen granules. The five components had good linear relationships within their respective ranges, with recovery rates ranging from 95.6% to 100.0%. And 145 batches of samples were tested, and a total of 54 batches were found to be unqualified, with a total unqualified rate of 37.2%. Establishing a characteristic chromatogram of Anshen granules, 12 characteristic peaks were attributed to 6 medicinal herbs. Through chemometric analysis, it was found that there were certain differences in the quality of samples from 10 manufacturers. The quality of fried gardenia and stir-baked licorice was the key factor. Conclusion: The established multi-component content determination method and characteristic chromatogram method for Jieyu Anshen granules are convenient and reliable, and can be used for comprehensive evaluation of the quality of Jieyu Anshen granules.

解郁安神颗粒为临床经验方,由四平康乐制药厂与北京中医研究院西苑医院联合研制,收载于2020年版《中华人民共和国药典》一部,具有舒肝解郁、安神定志的功效,用于情志不畅、肝郁气滞所致的失眠、心烦、焦虑、健忘,神经官能症、更年期综合征见上述证候者。本品由柴胡、大枣、石菖蒲、姜半夏、炒白术、浮小麦、制远志、炙甘草、炒栀子、百合、胆南星、郁金、龙齿、炒酸枣仁、茯苓、当归16味药组成,经临床研究发现,解郁安神颗粒对中风后抑郁[1]、脑卒中后抑郁[2]、更年期抑郁症[3]等各种抑郁症及盆腔炎清栓治疗慢性盆腔痛[4]等均有显著疗效,其联合西药还有治疗老年失眠伴抑郁[5]、老年冠心病稳定型心绞痛伴抑郁[6]、提高精神分裂症伴抑郁认知功能[7]等功效。
现行解郁安神颗粒法定标准仅定量了栀子苷,定性了制远志、柴胡和炙甘草3味药材,而现有文献[8-12]也仅针对其中单一药味进行研究。解郁安神颗粒由16味药组成,成分复杂,仅依靠单一成分测定难以控制其整体的质量,通过多组分含测及化学计量学多元统计等方法,可以较全面地控制制剂的内在质量。中药特征图谱具有信息量大,特征性强,整体性和模糊性等特点,是公认的现代中药质量评价最为实用的分析方法之一,可宏观反映中药化学信息的整体状况,实现中药内在质量的有效表征,从整体上提高中药质量控制水平[13-14]。为实现从整体控制产品质量,本实验建立多组分含量测定方法同时对3,6’-二芥子酰基蔗糖、斯皮诺素、甘草苷、甘草酸、栀子苷的含量进行测定,以考察解郁安神颗粒中制远志、炒酸枣仁、甘草与炒栀子的质量情况,并建立解郁安神颗粒的特征图谱,结合多元统计的分析方法找出影响产品质量的关键因素,为解郁安神颗粒质量标准的建立提供参考。
Waters高效液相色谱仪(Waters公司);ML204型万分之一电子分析天平(梅特勒-托利多仪器上海有限公司);XP26型百万分之一电子分析天平(梅特勒-托利多仪器上海有限公司)。Milli-Q超纯水系统(Millipore公司)。
对照品栀子苷(批号110749-201919,含量97.1%)、甘草酸铵(批号110731-202122,含量94.4%)、甘草苷(批号111610-201908,含量95.0%)、斯皮诺素(批号111869-202005,含量94.8%)和3,6’-二芥子酰基蔗糖(批号111848-202006,含量96.5%)购于中国食品药品检定研究院提供,乙腈为色谱纯,其他试剂均为分析纯。
145批次解郁安神颗粒均为2023年国家药品抽检的样品。
采用CAPCELL PAK MGⅡC18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)和0.1%磷酸水溶液(B)为流动相,梯度洗脱(0~34 min,3% A→21% A;34~40 min,21% A→25% A;40~41 min,25% A→28% A;41~85 min,28% A→53% A),流速1.0 mL·min-1,柱温30 ℃,栀子苷、甘草酸、甘草苷检测波长为237 nm;3,6’-二芥子酰基蔗糖和斯皮诺素检测波长为335 nm,进样量10 μL。理论板数以栀子苷峰计算应不得低于5 000。
精密称取对照品栀子苷、甘草苷、甘草酸铵、3,6’-二芥子酰基蔗糖适量,加70%甲醇水制成每1 mL含栀子苷100 μg、甘草苷20 μg、甘草酸铵50 μg(甘草酸质量=甘草酸铵质量/1.0207)、3,6’-二芥子酰基蔗糖5 μg的混合对照品溶液。
取样品(编号样1)适量,研细,取约2 g,精密称定,置具塞锥形瓶中,精密加入70%甲醇水20 mL,称量,超声(功率600 W,频率40 kHz)处理30 min,放冷,再称量,用70%甲醇水补足减失的量,摇匀,滤过,取续滤液,即得。
阴性样品溶液按照2020年版《中华人民共和国药典》一部中解郁安神颗粒的处方工艺,分别制备缺炒栀子、缺炙甘草、缺制远志和缺酸枣仁的阴性样品,按照 “2.1.2.2”项方法制备,阴性样品溶液。
分别吸取混合对照品溶液、供试品溶液、阴性样品溶液各10 μL,按照“2.1.1”项下色谱条件测定,结果阴性样品在供试品待测成分色谱峰相应位置无对应色谱峰,表明阴性无干扰,本法专属性良好。见图1~3
精密称取对照品栀子苷5.501 mg、甘草苷5.121 mg分别置5 mL量瓶中,对照品甘草酸铵5.174 mg、斯皮诺素5.538 mg、3,6’-二芥子酰基蔗糖5.716 mg分别置10 mL量瓶中,加70%甲醇水稀释至刻度,摇匀,即得(栀子苷1.068 3 mg·mL-1、甘草苷0.973 0 mg·mL-1、甘草酸0.4785 mg·mL-1、斯皮诺素0.525 0 mg·mL-1、3,6’-二芥子酰基蔗糖0.551 6 mg·mL-1)。
精密量取栀子苷对照品储备液1.00 mL、甘草苷对照品储备液200μL、甘草酸铵对照品储备液500 μL、斯皮诺素对照品储备液及3,6’-二芥子酰基蔗糖对照品储备液各100 μL置同一5 mL量瓶中,加70%甲醇水稀释至刻度,作为混合对照品溶液①。精密量取混合对照品溶液①1.00 mL,置2 mL量瓶,加70%甲醇水稀释至刻度,作为混合对照品溶液②。精密量取混合对照品溶液②1.00 mL,置2 mL量瓶中,加70%甲醇水稀释至刻度,作为混合对照品溶液③。
分别精密吸取对照品混合溶液①2、10和20 μL;混合对照品溶液②1、2和10 μL;混合对照品溶液③1 μL,注入液相色谱仪、测定,以峰面积Y为纵坐标,对照品进样量对数值X为横坐标,绘制标准曲线,结果见表1,表中各成分在各自范围内线性关系良好。
取样品(样1),按照“2.1.2.2”项下方法制备供试品溶液,在“2.1.1”项下色谱条件连续进样6次,计算各成分含量,计算RSD,结果栀子苷、甘草苷、甘草酸、斯皮诺素和3,6’-二芥子酰基蔗糖的RSD分别为0.25%、0.33%、0.22%、0.25%和0.51%,表明该法精密度良好。
取样品(样1),按照“2.1.2.2”项下方法制备供试品溶液,在“2.1.1”项色谱条件下分别于0、3、8、13、17和24 h进样测定,计算各成分含量,计算RSD,结果栀子苷、甘草苷、甘草酸、斯皮诺素和3,6’-二芥子酰基蔗糖的RSD分别为0.47%、2.6%、1.6%、0.38%和0.36%,表明该法稳定性良好,供试品溶液在24 h内稳定。
取样品(样1)6份,按照“2.1.2.2”项方法,平行制备供试品溶液6份,在“2.1.1”项色谱条件下进样,测定峰面积,计算各成分含量,栀子苷、甘草苷、甘草酸、斯皮诺素和3,6’-二芥子酰基蔗糖的平均含量分别为4.357 5、1.002 5、1.235 9、0.194 0和0.290 3 mg·袋-1。计算RSD,结果栀子苷、甘草苷、甘草酸、斯皮诺素和3,6’-二芥子酰基蔗糖的RSD值分别为1.66%、0.74%、0.64%、0.22%和0.43%,表明该法重复性良好。
取已知含量解郁安神颗粒(样1)研碎,取约1 g,精密称定,分别精密加入根据样品含量另行制备的加样用对照品溶液(栀子苷50.39 μg·mL-1、甘草苷9.73 μg·mL-1、甘草酸23.92 μg·mL-1、斯皮诺素1.95 μg·mL-1、3,6’-二芥子酰基蔗糖3.47 μg·mL-1)20 mL,称量,按“2.1.2.2”项制成加样样品溶液,并按上述色谱条件测定,计算加样回收率。结果样品中栀子苷、甘草苷、甘草酸、斯皮诺素、3,6’-二芥子酰基蔗糖的回收率分别为100.0%、97.0%、96.0%、97.6%和99.7%,RSD分别为0.50%、1.3%、1.2%、0.63%和0.57%。表明该方法准确度良好,符合方法学要求。
按照“2.1.2.2”项方法对10家生产企业去重批号后的145批样品进行制备,在“2.1.1”项色谱条件下进行测定,结果甘草含量45批不符合规定,不合格率为31.03%;制远志含量29批不符合规定,不合格率为20%。不合格样品共54批,总不合格率为37.2%。
检测波长为210 nm,其余同“2.1.1”项下。
根据处方按样品取样量为2 g时计算出各药材用量,故取对照药材石菖蒲0.16 g、甘草0.12 g、远志 0.16 g、栀子0.16 g、百合0.4 g、酸枣仁0.2 g,照“2.1.2.2”项方法制备供试品溶液,即得。
除不加甘草酸铵外,其余对照品同“2.1.2.1”项方法制备混合对照品溶液。
同“2.1.2.2”项方法。
按处方比例称取一定量的缺百合和缺石菖蒲的阴性样品,并按“2.1.2.2”项方法制备缺百合阴性样品溶液和缺石菖蒲阴性样品溶液,其余同“2.1.2.3”项方法。
本研究对解郁安神颗粒中的炒栀子、炒酸枣仁、百合、炙甘草、制远志、石菖蒲等6味药材的特征峰进行研究(图45)。其中炒栀子中栀子苷(2号峰)、炒酸枣仁中斯皮诺素(5号峰)、炙甘草中甘草苷(6号峰)、制远志中的3,6’-二芥子酰基蔗糖(8号峰)均为2020年版《中华人民共和国药典》中各药材所规定的指标成份。
对收集的药材3批炒栀子、10批炒酸枣仁、3批百合、19批炙甘草、10批制远志、19批石菖蒲与各自对照药材按“2.2.2.3”项下方法制备成供试品溶液并进样分析。将各单味药材样品溶液与供试品溶液色谱图(图6)比对分析,明确样品中1号特征峰(未知)来源于制栀子药材,3号特征峰(未知)来自于酸枣仁药材,4、7号特征峰(未知)来自于百合药材,9号特征峰(未知)来源于甘草药材,10、11号特征峰(未知)来自于制远志药材,12号特征峰(未知)来自于石菖蒲药材。
取“2.2.2.4”项下阴性样品溶液,按“2.2.1”项下色谱条件进样分析,各阴性样品溶液的色谱中均不呈现与各自对照药材溶液保留时间相同的谱峰,表明该方法的专属性良好,见图7
取供试品溶液(样1),按“2.2.1”项下色谱条件连续进样6次,记录特征图谱,计算特征峰的保留时间。结果各特征峰保留时间的RSD在0.090%~0.23%,表明仪器精密度良好。
取样品(样1)6份,每份约2 g,按照“2.1.2.2”项下方法,平行制备供试品溶液6份,按“2.2.1”项下色谱条件,溶液注入HPLC仪,记录特征图谱,计算特征峰的保留时间,结果各特征峰的保留时间RSD在0.030%~0.16%,表明该方法的重复性良好。
取供试品溶液(样1),按“2.2.1”项下色谱条件,分别于0、4、8、12、18和24 h测定,记录特征图谱计算特征峰的保留时间,结果各特征峰的保留时间RSD在0.09%~0.26%,表明供试品溶液在24 h内稳定。
取每个厂家2批共20批解郁安神颗粒样品,按“2.1.2.2”项方法制备供试品溶液,按“2.2.1”项色谱条件进样并记录色谱图。使用《中药色谱指纹图谱相似度评价系统》(2012版)进行共有峰匹配,选择重复性好、分离度较高,可归属到处方药味的 12个色谱峰作为共有峰,生成 20批样品的HPLC 特征图谱,见图8
对本次国家药品抽检去重复批号样品共145批样品进行测定,使用科迈恩化学计量学软件对其进行主成分分析(PCA),发现相同厂家间样品较为集中,表明同企业样品质量较稳定;不同厂家间样品较为分散,其中SP企业的产品与其他企业生产的产品差异较大。从主成分得分图(图9)可知这PCA1和PCA2贡献值分别为71.41%和19.10%。
通过调研了解到,不同企业对原料药的进货渠道基本固定,相对固定的原料来源使得企业对产品的质量控制更加稳定;而SP企业与其他企业不同的是,该企业原料药材多为在产地直接收购,产地收购省去中间商环节,直接收购优质药材,更直观地把控了药材品质,实现从源头上对药品质量控制。
采用SIMCA 14.1开展SP企业特征图谱测定值与其他厂家产品样本关系的正交偏最小二乘法判别分析(OPLS-DA),将SP企业产品与其余厂家产品分为2组,采用Par法,得到归一化数据,模式选择OPLS-DA进行自动调整建模,得到SP企业和与其他厂家样本之间的分析判别模型,该模型拟合参数R2X为0.675、R2Y为0.764、Q2为0.722。上述参数越接近于1说明模型越优秀,一般大于0.5即认为模型稳定且预测能力较好。所有批次样本的得分图如图10所示。
采用置换检验的方法对所构建的模型进行验证,在打乱建模的分组后,进行200次数据循环,每次得到新的R2和 Q2数据,与构建模型的参数R2和Q2数据进行比较,如图11所示。图中可见Q2数据所形成的回归线Y轴截距小于0,表明所建立的模型为可靠的监督判别分析模型。
在构建的模型可行可靠的前提下,可以利用 OPLS-DA的优点,找到最具差异性的变量,进而确定解郁安神颗粒的质量标志物。变量重要性投影值(variable importance in projection,VIP)代表每一个变量的重要性程度,一般认为该值大于1即对模型有较大影响。通过数据分析,得到VIP差异图及VIP值,如图12所示。 其中,峰2、峰6、峰1的VIP值大于1,峰1、峰2来自炒栀子,峰6来自炙甘草,提示炒栀子与炙甘草的质量差异是使SP企业与其他厂家产品产生较大差异的主要原因。
解郁安神颗粒由16味药材组成,根据处方分析功效作用及查阅相关文献发现栀子苷[15]为炒栀子的主要成分,可通过对布鲁顿酪氨酸激酶/Janus激酶/信号转导与转录激活因子1(BTK/JAK2/STAT1) 进行抑制,发挥抗抑郁功效;其还能调控高血糖素样肽1受体/蛋白激酶B通路缓解抑郁。甘草始载于《神农本草经》,以甘草苷和甘草酸(质控指标为甘草酸铵)为代表的三萜类物质是甘草中的主要活性成分,研究表明其具有解毒、抗炎、抗病毒、抗肿瘤及增加免疫等作用[16],并且甘草苷具有较好的抗抑郁作用[17]。3,6’-二芥子酰基蔗糖作为制远志的代表性成分,具有良好的抗抑郁和神经保护作用[18]。酸枣仁对多种精神疾病有疗效,可以改善睡眠状况、抗焦虑、抗抑郁,改善学习记忆、延缓阿尔兹海默症等,同时对心血管系统也有保护作用,对心律失常、心肌缺血缺氧等症均有疗效[19-20],依据2020年版《中华人民共和国药典》酸枣仁的含量测定,选择斯皮诺素作为控制酸枣仁质量的指标性成分进行测定。故将栀子苷、甘草苷、甘草酸铵、斯皮诺素和3,6’-二芥子酰基蔗糖作为对解郁安神颗粒整体质量进行控制的标志物。
实验中对不同溶剂(不同浓度的甲醇和乙醇)、不同提取方式(超声和加热回流)、不同提取时间(15、30、60 min)、不同提取体积(10、20、40 mL)进行考察,发现使用70%甲醇水20 mL超声提取30 min时即可将目标成分提取完全,故使用该方法作为提取方法。同时采用DAD检测器对在200~400 nm对供试品溶液进行全波长扫描,结果发现在210 nm处即能反映较为丰富的色谱信息,且基线较为平整,故选择210 nm作为特征图谱的检测波长;对于含量测定,各成分最大吸收波长差异较大,兼顾各组分最大吸收波长、分离度、灵敏度等因素,最终选定栀子苷、甘草酸、甘草苷检测波长为237 nm;3,6’-二芥子酰基蔗糖与斯皮诺素检测波长为335 nm。
本实验使用HPLC法同时测定炒栀子中栀子苷,炙甘草中甘草苷和甘草酸,炒酸枣仁中斯皮诺素和制远志中,3,6’-二芥子酰基蔗糖(C36H46O17)的含量,同时结合特征图谱对样品进行分析,与2020年版《中华人民共和国药典》中仅测定栀子苷含量的方法相比,能更全面地反映药品的质量。
依据145批次样品测定结果,除栀子苷外对其每个成分结果绘制正态分布曲线,取95%置信区间范围内数据求平均值,甘草苷为0.71 mg·袋-1,甘草酸铵为2.2 mg·袋-1,斯皮诺素为0.094 mg·袋-1,3,6’-二芥子酰基蔗糖为0.24 mg·袋-1,由于斯皮诺素含量的平均值在万分之一左右,含量低,故不拟定限度。其他成分按平均值的80%定为限度。
在收集药材与调研的过程中炒酸枣仁与制远志是处方中价格较高的2味药材,推测部分企业可能存在少投料或者投质量较差药材的情况。通过测定数据,制远志含量29批不符合规定,不合格率为20.0%;而斯皮诺素平均含量在万分之一左右,含量较低,与预测结果较为一致。在检测中还发现,炙甘草的不合格率也比较高,这说明这部分企业对原料药的质量把控不够严格,企业为了追求经济效益而放松对法定标准未控制的药材质量的监控。望各生产企业引以为戒真正做到“好药品是生产出来的”,继续加强对药材质量的监控,严格按照处方与制法生产合格的品药品。
