Objective: To establish an HPLC method for the simultaneous determination of six components (uridine, adenine, adenosine (R, S)-goitrine, guanosine, clemastanin B) in Radix Isatidis, and to investigate the linear calibration with two reference substances (LCTRS) method for the qualitative analysis of multiple components in Radix Isatidis. Methods: HPLC method was used, with methanol as mobile phase A and water as mobile phase B. Gradient elution (0-3 min, 3% A; 3-18 min, 3% A→14% A; 18-25 min, 14% A→26% A; 25-34 min, 26% A; 34-40 min, 26% A→46% A; 40-60 min, 46% A→90% A) was performed at a flow rate of 0.8 mL·min-1. The column temperature was 30 ℃, the detection wavelengths were 254 nm (0-32 min) and 230 nm (32-60 min). The injection volume was 10 μL. The actual retention time of 6 components in Radix Isatidis was determined on 13 C18 chromatographic columns of different brands and models. Guanosine and clemastanin B were used as double reference compounds, and LCTRS method was used to locate the chromatographic peak of each component. Three unknown chromatographic columns were used for method validation. Using guanosine as a reference substance, the relative retention time method was used to predict the retention time of the other 5 components. The predictive accuracy and column coincidence of these two methods were compared. Results: The LCTRS method could effectively predict and qualitatively analyze the retention time of six indicator components. Compared with the relative retention time method, the LCTRS method had higher accuracy in predicting results and better column universality. Conclusion: The LCTRS method for simultaneous determination of multiple components in Radix Isatidis is feasible and accurate, with simple operation and good durability, and has promotional value.

目的:建立同时检测板蓝根中6个成分(尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、直铁线莲宁B)的HPLC方法,考察双标线性校正(LCTRS)法对于板蓝根多成分定性分析的可行性。方法:采用HPLC法,以甲醇为流动相A,水为流动相B,梯度洗脱(0~3 min,3% A;3~18 min,3% A→14% A;18~25 min,14% A→26% A;25~34 min,26% A;34~40 min,26% A→46% A;40~60 min,46% A→90% A),流速0.8 mL·min-1,柱温30 ℃,检测波长为0~32 min,254 nm;32~60 min,230 nm,进样量10 μL;测定板蓝根中6个成分在16根不同品牌和型号的C18色谱柱的实际保留时间,以鸟苷和直铁线莲宁B 2个成分作为双标化合物,采用LCTRS法定位各成分色谱峰,并用3根未知色谱柱进行方法验证。同时以鸟苷为参照物,采用RRT法预测其他5个成分的保留时间。比较这2种方法的预测准确性和色谱柱符合率。结果:LCTRS法能够较好地对6个指标性成分进行保留时间的预测和定性分析。与RRT法相比较,LCTRS法所预测结果准确性更高,色谱柱普适性更优。结论:LCTRS法用于同时测定板蓝根中多成分可行且准确,操作简便,耐用性好,具有推广价值。

目的:建立同时检测板蓝根中6个成分(尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、直铁线莲宁B)的HPLC方法,考察双标线性校正(LCTRS)法对于板蓝根多成分定性分析的可行性。方法:采用HPLC法,以甲醇为流动相A,水为流动相B,梯度洗脱(0~3 min,3% A;3~18 min,3% A→14% A;18~25 min,14% A→26% A;25~34 min,26% A;34~40 min,26% A→46% A;40~60 min,46% A→90% A),流速0.8 mL·min-1,柱温30 ℃,检测波长为0~32 min,254 nm;32~60 min,230 nm,进样量10 μL;测定板蓝根中6个成分在16根不同品牌和型号的C18色谱柱的实际保留时间,以鸟苷和直铁线莲宁B 2个成分作为双标化合物,采用LCTRS法定位各成分色谱峰,并用3根未知色谱柱进行方法验证。同时以鸟苷为参照物,采用RRT法预测其他5个成分的保留时间。比较这2种方法的预测准确性和色谱柱符合率。结果:LCTRS法能够较好地对6个指标性成分进行保留时间的预测和定性分析。与RRT法相比较,LCTRS法所预测结果准确性更高,色谱柱普适性更优。结论:LCTRS法用于同时测定板蓝根中多成分可行且准确,操作简便,耐用性好,具有推广价值。

Objective: To establish an HPLC method for the simultaneous determination of six components (uridine, adenine, adenosine (R, S)-goitrine, guanosine, clemastanin B) in Radix Isatidis, and to investigate the linear calibration with two reference substances (LCTRS) method for the qualitative analysis of multiple components in Radix Isatidis. Methods: HPLC method was used, with methanol as mobile phase A and water as mobile phase B. Gradient elution (0-3 min, 3% A; 3-18 min, 3% A→14% A; 18-25 min, 14% A→26% A; 25-34 min, 26% A; 34-40 min, 26% A→46% A; 40-60 min, 46% A→90% A) was performed at a flow rate of 0.8 mL·min-1. The column temperature was 30 ℃, the detection wavelengths were 254 nm (0-32 min) and 230 nm (32-60 min). The injection volume was 10 μL. The actual retention time of 6 components in Radix Isatidis was determined on 13 C18 chromatographic columns of different brands and models. Guanosine and clemastanin B were used as double reference compounds, and LCTRS method was used to locate the chromatographic peak of each component. Three unknown chromatographic columns were used for method validation. Using guanosine as a reference substance, the relative retention time method was used to predict the retention time of the other 5 components. The predictive accuracy and column coincidence of these two methods were compared. Results: The LCTRS method could effectively predict and qualitatively analyze the retention time of six indicator components. Compared with the relative retention time method, the LCTRS method had higher accuracy in predicting results and better column universality. Conclusion: The LCTRS method for simultaneous determination of multiple components in Radix Isatidis is feasible and accurate, with simple operation and good durability, and has promotional value.

中药的质量控制对于中药的疗效和安全性,促进中药的规范化、标准化和现代化发展具有重要意义[1]。中药成分复杂,通常包括多种有效成分,在进行多指标成分分析时,需要多种对照品,检测成本高昂[2]。有学者提出了一测多评法和替代对照品法等对多个目标成分进行测定,但多存在局限性或色谱柱耐用性不佳的问题[3-6]。孙磊等[7]提出了双标线性校正法(linear calibration with two reference substances,LCTRS),即选取2个参照物对多组分色谱峰进行定性分析,拥有较高的准确度和色谱柱适用性,同时可降低检测成本,现已成功地运用于中药材、中药饮片、配方颗粒及中成药的多成分定性分析[8-14]
板蓝根为十字花科草本植物菘蓝 Isatis indigotica Fort.的干燥根,为大宗药材之一,含有生物碱、木脂素、核苷、有机酸等类活性成分,主要用于感冒发热、温病斑疹、咽喉肿痛、痄腮、丹毒等病证[15-16]。现代研究证明,木脂素类作为板蓝根主要活性成分之一,具有抗炎、抗病毒、抑菌、抗肿瘤等多种药理作用;板蓝根中的尿苷、鸟苷、腺苷、腺嘌呤等核苷类成分能干扰病毒核酸的合成,是抗病毒的活性成分,在板蓝根中含量较高,被广泛应用于板蓝根的质量评价;板蓝根中的生物碱类具有抗炎、抗氧化、抗肿瘤、抗病毒等药理作用[17]
2020年版《中华人民共和国药典》中板蓝根的质量控制指标成分仅为(RS)-告依春[18],难以体现板蓝根的整体质量。因此,建立多成分、多指标的质量控制方法,对于快速、全面地评价药理作用广泛的板蓝根的质量具有重要意义。本实验采用HPLC,选取了板蓝根中主要的活性成分:生物碱类化合物(RS)-告依春、木脂素类化合物直铁线莲宁B,以及核苷类化合物尿苷、鸟苷、腺苷、腺嘌呤,建立同时测定板蓝根中6个成分的方法,并首次采用LCTRS法用于板蓝根多指标色谱峰定性分析,与传统的相对保留时间法(relative retention time,RRT)相比,LCTRS法的准确性和普适性更佳。本研究为全面快速地评价板蓝根质量提供了准确可靠的检验方法,同时拓展了LCTRS法在中药多指标成分定性分析方面的应用。
Agilent 1100高效液相色谱仪(Agilent公司),梅特勒托利多 XS205DU十万分之一电子天平(Mettler Toledo公司),KQ-800KDB超声波清洗机(昆山舒美司),Milli-Q Advantage A超纯水机(Millipore公司);甲醇为色谱纯(Merck公司),其余试剂皆为分析纯;对照品尿苷(批号110887-202104,含量99.6%)、腺嘌呤(批号11088-202203,含量99.8%)、鸟苷(批号111977-201501,含量93.6%)、(RS)-告依春(批号111753-202007,含量100%)、腺苷(批号110879-201703,含量99.7%)购于中国食品药品检定研究院,直铁线莲宁B(批号A04HB190505,含量98%)购于上海源叶生物科技有限公司;19根色谱柱(250 mm×4.6 mm,5 μm)信息详见表1
共收集不同产地的10批板蓝根样品,编号分别为BLG01~BLG20,经黑龙江省药品检验研究院笔雪艳主任药师鉴定为十字花科植物菘蓝Isatis indigotica Fort.的干燥根。
采用WELCH ULTIMATE PG C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇(A)-水(B)为流动相,梯度洗脱(0~3 min,3% A;3~18 min,3% A→14% A;18~25 min,14% A→26% A;25~34 min,26% A;34~40 min,26% A→46% A;40~60 min,46% A→90% A),流速为0.8 mL·min-1,柱温为30 ℃,检测波长分别为254 nm(0~32 min)和230 nm(32~60 min),进样量为10 μL。
分别取对照品尿苷、腺嘌呤、鸟苷、(R,S)-告依春、腺苷、直铁线莲宁B适量,加5%甲醇制成每1 mL含尿苷5.26 μg、腺嘌呤2.56 μg、鸟苷7.35 μg、(R,S)-告依春72.31 μg、腺苷7.56 μg、直铁线莲宁B 5.02 μg的混合溶液。
取本品粉末(过4号筛)约1 g,精密称定,置于具塞锥形瓶中,精密加入5%甲醇50 mL,密塞,称量,超声(功率600 W,频率40 kHz)处理30 min,放冷,再称量,用5%甲醇补足减失的量,摇匀,滤过,取续滤液,即得。
取“2.2.1”项下对照品溶液适量,按“2.1”项下色谱条件连续进样测定6次,分别记录尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、直铁线莲宁B的保留时间。结果表明6个成分色谱峰的保留时间RSD均<1.0%,表明仪器精密度良好。
取样品(BLG06)适量,精密称定,按“2.2.2”项下方法制备供试品溶液,分别于0、8、12、16、20和24 h时按“2.1”项下色谱条件进样测定,记录保留时间。结果6个成分的保留时间RSD均<1.0%,表明供试品溶液在24 h内稳定性良好。
取板蓝根样品(样品BLG06)6份,按“2.2.2”项下方法制备供试品溶液,在“2.1”项色谱条件下进样测定,记录每份样品中6个成分的保留时间。结果6个成分的保留时间RSD均<2.0%,表明该法重复性良好。
按照“2.2.2”项方法制备供试品溶液,在“2.1”项色谱条件下,分别采用Col 1~Col 16共16根色谱柱进样测定,Col 4、Col 9、Col 14色谱柱色谱图中存在色谱柱分离度较差或出峰时间颠倒的问题,因此将上述3根色谱柱删除,其余色谱柱均可将6个对照品色谱峰较好分离。对照品和样品色谱图见图1
LCTRS法原理是基于待测成分的保留时间在不同色谱柱上呈线性关系,在多指标成分分析中,选择其中两种成分作为参照物,将实测保留时间代入确定的线性方程来预测其余待测成分的保留时间,从而确定待测成分色谱峰峰位[19]
研究流程为将采集到的不同品牌和型号色谱柱的色谱图数据以“.cdf”格式导入 DRS Origin软件,关联每张色谱图中的对照品,考察选取合适的双标化合物,以分析得到不同色谱柱保留时间预测准确率和色谱柱符合率。同时采用RRT法预测各成分的保留时间,对LCTRS法和RRT法的准确性和耐用性进行比较和评价。
以13根色谱柱中6个成分保留时间的均值作为标准保留时间(standard retention time,SRT),SRT分别为尿苷10.02 min、腺嘌呤14.56 min、鸟苷15.84 min、(RS)-告依春19.51 min、腺苷23.41 min、直铁线莲宁B 36.78 min,以6个成分的SRT(X)为横坐标,以实际保留时间(Y)为纵坐标,得到13根色谱柱的保留时间拟合结果,各色谱柱的线性方程和相关系数见表2。结果表明6个成分在13根色谱柱上得到的线性方程r均大于0.999,线性关系和拟合效果较好。
双标化合物的选择有多种方案,使用DRS Origin软件通过计算得到10种双标化合物选择方案,其中由于峰3(鸟苷)及峰6(直铁线莲宁B)作为双标化合物进行组合时,保留时间预测正确率和色谱柱符合率均为100%,且保留时间回归偏差最小,因此选取峰3(鸟苷)及峰6(直铁线莲宁B)作为双标化合物。
将13根不同色谱柱按照上述色谱条件分析后得到的色谱图谱数据以“.cdf”的格式导入 DRS Origin软件,选定RRT法作为分析方法,得到各个成分作为内参物的分析结果,结果表明选择鸟苷作为内参物时,平均保留时间偏差最小且色谱肯定率最高,且鸟苷在该色谱条件下出峰时间适中,符合RRT法选择依据,因此选择鸟苷作为内参物。2种保留时间预测方法的结果见表3。结果表明,RRT法的预测保留时间与实际值偏差较大,13个色谱柱中,仅有6个色谱柱对于6个色谱峰保留时间的预测值偏差<1.0 min,且保留时间偏差(ΔtR)均较大;而LCTRS法在13根色谱柱的预测值偏差均<0.3 min。由此可见,LCTRS法的预测准确度更高,适用的色谱柱数量更多,显著优于RRT法。
再选择3根色谱柱(250 mm×4.6 mm,5 μm),按“2.2.2”项下方法制备供试品溶液,按“2.1”项下色谱条件进样分析,比较LCTRS法和RRT法的定性结果,见表45。结果表明,LCTRS法能较好地预测板蓝根中6个成分在各色谱柱上的保留时间,然而RRT法在3根色谱柱上的预测误差较大,结果表明LCTRS法比RRT法预测更加准确。
本研究建立了同时分离检测板蓝根中6个成分的HPLC方法,在检测波长选择中,由于尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷5个成分在254 nm处有较好地吸收,而直铁线莲宁B在254 nm下峰面积较小,峰指认困难,且在230 nm处有最大吸收,因此选用254 nm及230 nm双波长作为检测波长。该法重复性好、指标较为全面,可作为板蓝根的质量评价方法,同时也可为后续深入研究板蓝根的质量标准提供理论依据。
本研究建立的LCTRS法用于板蓝根中多组分定性分析结果表明,相比于RRT法,LCTRS法能够更加准确地预测板蓝根中6个成分在各色谱柱上的保留时间,且色谱柱普适性更优。分析原因可能是RRT法中预测峰与参照物的保留时间间隔较大,造成预测结果的误差较大。
采用LCTRS法对板蓝根中多成分进行分析,具有良好的准确性、耐用性,同时有助于降低检验成本,并有利于更为全面地对板蓝根质量进行控制和评价。因此,LCTRS法在中药多指标成分定性方面有更好地应用前景和实用价值。
