Objective: To establish the infrared spectrum identification model to distinguish Mingui from Angelica sinensis from other habitats. Methods: A total of 1 540 batches of A. sinensis were collected from Gansu Province through visits and field investigation, and the mid-infrared spectrum of each sample was collected by Fourier transform infrared spectroscopy. The average spectra of 423 batches of Mingui were calculated as their fingerprint. The identification threshold of Mingui was determined as 0.989 6 (μ-3σ) by mid-infrared spectroscopy combined with the “3σ” criterion. The identification model of Mingui was established. Three batches of Mingui and 97 batches of A. sinensis from other habitats were randomly selected to verify the identification model. Results: The characteristic peaks of samples were observed at 3 560, 3 389, 3 342, 2 931, 1 743, 1 650, 1 459, 1 409, 1 385, 1 374, 1 323, 1 279, 1 239, 1 128, 1 115, 1 104, 1 068, 1 053, 1 013, 1 004, 991 and 920 cm-1, etc. The mid-infrared spectra of 423 batches of Mingui were highly similar, and their peak shape, peak position and peak intensity were similar. The correlation coefficients between Mingui and the fingerprint ranged from 0.992 0 to 0.998 0. The accuracy of identification model test was 97%. Conclusion: The identification model of Mingui could be used to distinguish Mingui from other A. sinensis from other habitats.

目的:建立岷归红外光谱鉴别模型,准确区分岷归和其他产地当归。方法:通过走访和实地调查,从甘肃省采集当归样品共计1 540批次,利用傅里叶变换红外光谱技术采集各样品的中红外光谱。计算423批岷归的平均光谱,作为其指纹图谱。中红外光谱结合“3σ”准则确定岷归的鉴别阈值为(μ-3σ)0.989 6,初步形成岷归的鉴别模型。随机抽取3批岷归及97批其他产地当归对模型进行验证。结果:当归样品特征峰均在3 560、3 389、3 342、2 931、1 743、1 650、1 459、1 409、1 385、1 374、1 323、1 279、1 239、1 128、1 115、1 104、1 068、1 053、1 013、1 004、991、920 cm-1等处,423批岷归的中红外光谱相似度高,其峰形、峰位、峰强相似,岷归样品与指纹图谱的相关系数在0.992 0~0.998 0。鉴别模型检验正确率为97%。结论:建立的岷归红外光谱鉴别模型可用于区别岷归和其他产地当归。

Objective: To establish the infrared spectrum identification model to distinguish Mingui from Angelica sinensis from other habitats. Methods: A total of 1 540 batches of A. sinensis were collected from Gansu Province through visits and field investigation, and the mid-infrared spectrum of each sample was collected by Fourier transform infrared spectroscopy. The average spectra of 423 batches of Mingui were calculated as their fingerprint. The identification threshold of Mingui was determined as 0.989 6 (μ-3σ) by mid-infrared spectroscopy combined with the “3σ” criterion. The identification model of Mingui was established. Three batches of Mingui and 97 batches of A. sinensis from other habitats were randomly selected to verify the identification model. Results: The characteristic peaks of samples were observed at 3 560, 3 389, 3 342, 2 931, 1 743, 1 650, 1 459, 1 409, 1 385, 1 374, 1 323, 1 279, 1 239, 1 128, 1 115, 1 104, 1 068, 1 053, 1 013, 1 004, 991 and 920 cm-1, etc. The mid-infrared spectra of 423 batches of Mingui were highly similar, and their peak shape, peak position and peak intensity were similar. The correlation coefficients between Mingui and the fingerprint ranged from 0.992 0 to 0.998 0. The accuracy of identification model test was 97%. Conclusion: The identification model of Mingui could be used to distinguish Mingui from other A. sinensis from other habitats.

当归为伞形科植物当归Angelica sinensis (Oliv.) Diels的干燥根,在我国主产于甘肃省东南部,另外在云南、湖北、四川、陕西等省份也有栽培[1]。岷县产当归以其独特的加工方法、使用历史悠久、品质佳,被誉为岷归。岷归远销东南亚及欧美等20多个国家和地区,被欧洲人誉为“中国妇科人参”,是世界当归中的上品,素有“中华当归甲天下,岷县当归甲中华”之说。性状鉴别一直是中药材鉴别的常规手段,但需要经验丰富的专业人员。由于专业人员的缺乏,药材集散地和产地大批量样品的快速鉴别困难重重[2]。随着人类保健意识的增强,特别是中国加入WTO后,迎来国际、国内中药材市场竞争的新局面,根据中医药现代化、国际化发展要求,当归的鉴别方法也应从传统性状鉴别向现代化、标准化转变。因此构建一种简便、快捷的岷归鉴别方法对当归药材资源开发利用及保护具有重要意义。红外光谱技术用于药材定性鉴别,具有快速、简便、无损的优点,且能够反映药材的整体特征[3-5]。由于受检测空间声、光、二氧化碳、水分等干扰会产生粗大误差,3σ准则是判别、处理粗大误差的最常用方法[6]。本文基于大样本当归药材的中红外光谱数据,构建了岷归的快速鉴别模型,该模型可快速识别优质当归药材,符合甘肃省经济发展需求,同时也为道地中药材鉴别提供了一种新思路。
FT-Frontier型傅里叶变换红外光谱仪(天津尚勤科技有限责任公司),YP-4压片机(北京新亚恒奥科技有限公司)。压片模具(北京新亚恒奥科技有限公司),模具直径13 mm。BC504型十万分之一电子天平(上海升徽电子有限公司)。
基于前期调查研究基础,本课题组根据当归栽培面积大小以自然村为单位,合理规划,共采集当归1540批次,采样点覆盖甘肃省定西市、甘南藏族自治州、陇南市、临夏回族自治州、武威市、天水市、张掖市、兰州市共计8个市、23个县、129个乡、854个村,当归样品均为课题组自采,样品经甘肃中医药大学晋玲教授鉴定均为药典正品[7]。样品阴干、净制、打粉过6号筛(100目)装入自封袋冷藏保存。溴化钾(KBr)颗粒,光谱纯,批号9209822,购于上海一研生物科技有限公司。
当归样品及溴化钾颗粒临用前在70 ℃干燥1.5 h,转移至干燥器放凉,密封保存。取溴化钾颗粒约100 mg,研磨成细粉,制空白片,用溴化钾空白片扣除背景。取当归粉末约3 mg,溴化钾约100~105 mg,将二者研磨均匀,制样品片。
将制好的样品片放入样品架,采集一维红外光谱图。以透射方式采集样品的FTIR原始光谱数据。技术参数:扫描次数32,扫描时扣除水和二氧化碳干扰,扫描范围4 000~400 cm-1,平行测量4次,光谱分辨率4 cm-1。红外光谱峰的识别阈值设为0.01 A。将采集到的红外光谱进行基线校正和光谱归一化。
红外光谱指纹区吸收峰的特征性强,犹如人的指纹,故称指纹区,指纹区具有多峰性的特点。通过观测423批岷归红外光谱,发现其特征峰集中在4 000~2 500 cm-1及1 800~800 cm-1范围内。2 500~1 800 cm-1范围内未发现吸收峰。800~500 cm-1范围内的吸收峰不同批次样品间差异较大、不稳定。结合文献学习,当归药材800~500 cm-1范围内的吸收峰易受多种因素影响而改变位置[6];4 000~2 500 cm-1范围内的吸收峰易受水分的影响[8];因此本文选择光谱稳定,特征性强的1 800~800 cm-1范围作为指纹区。
为了建立特征性强、评价客观的岷归红外光谱指纹图谱,直观地反映其整体特征,在423批岷归样品中,随机抽取3批作为验证集样品,其余420批为校正集样品,将岷归样品红外光谱的平均光谱作为岷归的指纹图谱。
相似度是用单一数值评价两张谱图的整体相似程度[9]。因此,相关系数(r)大小可以体现中药红外光谱的相似程度,r可用投影向量夹角余弦法计算,计算公式为:
式中n为波数变量的个数,XiYi为光谱Y与光谱X在波数i处的吸收度。
在本文中,先检验420批校正集岷归红外光谱与指纹图谱的相关系数是否服从正态分布,若符从,则参照统计学“3σ”准则,以μ-3σ作为判别岷归真伪的阈值[8]
423批岷归的红外光谱相似,重叠光谱见图1,均在3 560、3 389、3 342、2 931、1 743、1 650、1 459、1 409、1 385、1 374、1 323、1 279、1 239、1 128、1 115、1 104、1 068、1 053、1 013、1 004、991、920 cm-1等处有吸收峰。其中3 560 cm-1为酚羟基吸收峰,3 390 cm-1为羧酸羟基吸收峰,3 340 cm-1为羧酸羟基C-H振动吸收峰,1 322 cm-1主要为酚类分子的O-H弯曲振动吸收峰,主要代表当归药材中有机酸类成分含量[10-11]。1 743 cm-1附近是挥发油、羧酸类、酯羰基和其他混合物中C=O伸缩振动吸收峰,是当归中藁本内酯类化合物的特征峰[12-13]。1 651 cm-1为含有共轭羰基C=O伸缩振动吸收峰,1 458 cm-1为C-H弯曲振动峰,1 410 cm-1为(O)C-H弯曲振动吸收峰,1 278、1 103、1 052、990 cm-1附近的阶梯峰为C-O伸缩振动吸收峰,1 240 cm-1为酯类C-O-C和C-O伸缩振动吸收峰,1 004、921 cm-1分别为糖环对称与不对称伸缩振动吸收峰,即当归中苷类和多糖类物质的特征峰[14-15]。1 129、1 116、1 067 cm-1为当归中纤维素类物质C-O伸缩振动吸收峰[16-17]
420批岷归校正集样品的红外光谱相似度高,与其平均光谱的相关系数在0.992 0~0.998 0,相关系数均值为r=0.995 0,标准差σ=0.001 8。图2为岷归指纹图谱,其指纹区特征峰位于1 740、1 651、1 458、1 410、1 322、1 279、1 239、1 106、1 054、990、920 cm-1等处。
判断420批岷归红外光谱的相关系数是否符合正态分布。正态检验结果P为0.129,大于0.05,岷归与指纹图谱相关系数正态P-P图见图3,图中各点近似呈一条直线,说明相关系数符合正态分布。参照“3σ”准则,以420批岷归相关系数的下限值(μ-3σ)0.989 6,作为岷归鉴别阈值,即:待测样品与当归指纹图谱的r小于0.989 6,判别为不是岷归,否则是岷归。
岷归验证集样品红外光谱见图4,样品的红外光谱相似,均在3 388、2 931、1 738、1 650、1 462、1 408、1 323、1 274、1 237、1 109、1 051、990、916 cm-1处附近有吸收峰。岷归样品与指纹图谱的r分别为0.998、0.995、0.994均大于0.989 6,判别均为岷归。
随机抽取甘肃省其他产地当归的中红外光谱97批,与当归指纹图谱进行比较,不同产地当归红外光谱见图5,均在3 561、3 390、3 344、2 934、1 650、1 459、1 409、1 346、1 323、1 239、1 128、1 115、1 104、1 068、1 053、1 014、1 004、991、920、551 cm-1等处附近有吸收峰。各产地当归与指纹图谱的相关系数在0.921~1.000之间,不同产地当归样品检验结果见表1,张掖市13批,检验正确率100%;天水市18批,检验正确率100%;兰州市2批,检验正确率100%;临夏11批,检验正确率100%;甘南13批,检验正确率92.31%;陇南16批,检验正确率100%;武威24批,检验正确率91.67%。
当归为常用中药,且多为栽培品。不同产地当归的大小、颜色有差异,但它们红外光谱的整体峰形、特征峰的位置及强度相似,说明甘肃省不同来源当归样品的化学组分相似。由岷归样品计算产生的平均红外光谱反映了岷归的整体化学信息和光谱特征,且能与其他产地当归相区别,可作为岷归的红外光谱指纹图谱。
本文基于当归药材的中红外光谱数据,结合统计学、化学计量学方法,构建了岷归的快速鉴别模型。该模型可快速识别优质当归药材,为当归的生产、流通、监管及临床使用带来便利,符合甘肃省经济发展需求。
