Objective: To investigate the source and quality of commercially available Epimedium based on HPLC fingerprint and multi-index assay, so as to provide reference for its quality evaluation and resource development. Methods: Forty batches of commercially available epimedium were collected. Fingerprint analysis and determination of the total flavonoid glycosides were performed by HPLC method. The total flavonoid glycosides were determined by UV method. And the contents of their extracts were determined. Stoichiometric analysis was employed to perform the comprehensive quality evaluation of different medicinal materials of Epimedium. Results: There were 9 different plant sources of Epimedium in the market. The five authentic Epimedium were of good quality, and the local varieties were coarse Epimedium. The contents were high in Epimedium from Hunan, while the contents were low in Epimedium from Qianling and Sichuan. The comprehensive quality ranking of Epimedium indicated that Epimedium Fletchum and Epimedium Fletchum ranked first by the principal component analysis method. The ranking of the same variety varied greatly with different habitats, and the Qianling Epimedium and adulterated Epimedium ranked last. The results of cluster analysis and principal component analysis were basically consistent. The cluster analysis showed distinguishing significance in plant sources, habitats, processing methods and contents. Based on the common pattern of E. brevicornu, a total of 8 common peaks were identified for 9 species of E. Brevicornu, and their similarities ranged from 0.066 to 0.979. Differences were observed among different species. Conclusion: Both the fingerprint similarity and the chemical model of content determination can be used to evaluate the species and quality of Epimedium.

目的:调查市售淫羊藿来源,采用HPLC指纹图谱及多指标成分含量测定,对市售淫羊藿药材进行质量研究,为其质量评价和资源开发提供参考。方法:收集到40批市售淫羊藿药材,采用HPLC指纹图谱分析,HPLC测定总黄酮醇苷、UV法测定总黄酮含量,并测定浸出物含量,应用化学计量学分析方法,完成不同淫羊藿药材的综合质量评价。结果:市售淫羊藿有9种不同植物来源;5种正品淫羊藿质量良好,地方品种以粗毛淫羊藿、湖南淫羊藿含量较高,而黔岭淫羊藿、四川淫羊藿含量甚微;用主成分分析法进行综合质量排序,淫羊藿、箭叶淫羊藿和柔毛淫羊藿排序靠前,同一品种随产地排序变化较大,黔岭淫羊藿及掺假的淫羊藿商品排序最后;聚类分析显示在植物来源、产地、炮制和含量方面有较好区分意义;以淫羊藿(E. brevicornu)建立共有模式,9种淫羊藿共标定了8个共有峰,相似度范围在0.066~0.979,不同品种间存在一定差异。结论:指纹图谱相似度和含量测定的化学模式均能良好的评价淫羊藿品种和质量。

目的:调查市售淫羊藿来源,采用HPLC指纹图谱及多指标成分含量测定,对市售淫羊藿药材进行质量研究,为其质量评价和资源开发提供参考。方法:收集到40批市售淫羊藿药材,采用HPLC指纹图谱分析,HPLC测定总黄酮醇苷、UV法测定总黄酮含量,并测定浸出物含量,应用化学计量学分析方法,完成不同淫羊藿药材的综合质量评价。结果:市售淫羊藿有9种不同植物来源;5种正品淫羊藿质量良好,地方品种以粗毛淫羊藿、湖南淫羊藿含量较高,而黔岭淫羊藿、四川淫羊藿含量甚微;用主成分分析法进行综合质量排序,淫羊藿、箭叶淫羊藿和柔毛淫羊藿排序靠前,同一品种随产地排序变化较大,黔岭淫羊藿及掺假的淫羊藿商品排序最后;聚类分析显示在植物来源、产地、炮制和含量方面有较好区分意义;以淫羊藿(E. brevicornu)建立共有模式,9种淫羊藿共标定了8个共有峰,相似度范围在0.066~0.979,不同品种间存在一定差异。结论:指纹图谱相似度和含量测定的化学模式均能良好的评价淫羊藿品种和质量。

Objective: To investigate the source and quality of commercially available Epimedium based on HPLC fingerprint and multi-index assay, so as to provide reference for its quality evaluation and resource development. Methods: Forty batches of commercially available epimedium were collected. Fingerprint analysis and determination of the total flavonoid glycosides were performed by HPLC method. The total flavonoid glycosides were determined by UV method. And the contents of their extracts were determined. Stoichiometric analysis was employed to perform the comprehensive quality evaluation of different medicinal materials of Epimedium. Results: There were 9 different plant sources of Epimedium in the market. The five authentic Epimedium were of good quality, and the local varieties were coarse Epimedium. The contents were high in Epimedium from Hunan, while the contents were low in Epimedium from Qianling and Sichuan. The comprehensive quality ranking of Epimedium indicated that Epimedium Fletchum and Epimedium Fletchum ranked first by the principal component analysis method. The ranking of the same variety varied greatly with different habitats, and the Qianling Epimedium and adulterated Epimedium ranked last. The results of cluster analysis and principal component analysis were basically consistent. The cluster analysis showed distinguishing significance in plant sources, habitats, processing methods and contents. Based on the common pattern of E. brevicornu, a total of 8 common peaks were identified for 9 species of E. Brevicornu, and their similarities ranged from 0.066 to 0.979. Differences were observed among different species. Conclusion: Both the fingerprint similarity and the chemical model of content determination can be used to evaluate the species and quality of Epimedium.

淫羊藿为常用中药材,《中华人民共和国药典》(《中国药典》)收载为淫羊藿(Epimedium brevicornu Maxim.)、箭叶淫羊藿(E. sagittatum(Sieb. et. Zucc.)Maxim.)、柔毛淫羊藿(E. pubescens Maxim.)和朝鲜淫羊藿(E. koreanum Nakai)的干燥叶。具有补肾阳、强筋骨和祛风湿的功效,主要用于治疗肾阳虚衰、风湿痹痛和麻木拘挛[1]等。现代研究表明,淫羊藿含有黄酮类、木脂素类、生物碱等成分,具有助孕,抗骨质疏松,控制糖尿病,提高免疫力和抑制肿瘤等药理药效作用[2-5]。淫羊藿广泛应于医药、化妆品、保健品领域,市场需求量很大,是国内最具开发潜力的中药之一[6]
我国淫羊藿属(Epimedium Linn.)植物约40种,药用植物15种[7-8]。淫羊藿属植物没有广布种,4种药用淫羊藿具有明显的地域分布特色,以多种来源的狭布种组成的药用淫羊藿在中药界不多见。我国中部和西南地区的淫羊藿比较丰富,有6种地方习用药材[9]。我国药用淫羊藿品种较多,国内学者主要关注《中国药典》收载的不同品种、不同产地的淫羊藿药材质量[10-13],少有地方品种的研究[14-15]。近年,随着各地发掘利用本省的淫羊藿资源,市售淫羊藿日趋复杂,其品种来源不甚了解,也鲜见质量状况的评价研究。淫羊藿品种的多样性和质量差异性会给临床用药的有效安全带来隐患,成为了药品市场监管的突出问题;与此同时,不同品种的淫羊藿是否具有潜在开发利用价值,也是值得深入研究的课题。
鉴于目前对商品淫羊藿缺乏系统的品种调查和质量研究,2021年以来调查了全国淫羊藿主产区和市售淫羊藿,收集了9个省产地的40份样品,经鉴定为9种不同的植物来源。在此基础上进行了HPLC指纹图谱分析,考虑到不同品种来源的质量评价的等效性与可比性,采用《中国药典》收载的总黄酮醇苷、淫羊藿总黄酮及浸出物含量指标,通过多元统计学进行质量分析,完成淫羊藿的质量研究。为不同品种、不同产地的淫羊藿质量评价、淫羊藿种质资源的开发利用以及了解市售的淫羊藿品种现状提供科学依据。
安捷伦1260型高效液相色谱仪,KQ-300DB数控超声波清洗器(昆山市超声仪器有限公司),UV-2450紫外分光光度仪(岛津公司),QUINTIX224-1CN万分之一分析天平,9240A鼓风干燥箱(北京兰贝石恒温技术有限公司)。
2021—2022年收集了甘肃、吉林、四川、贵州、重庆、湖北、湖南、陕西和安徽等产区或市场流通的淫羊藿40份样品,经甘肃省食品药品检验所宋平顺主任药师鉴定,认为市场上流通的“淫羊藿”来源复杂,除《中国药典》淫羊藿Epimedium brevicornu Maxim.、箭叶淫羊藿E. sagittatum (Sieb. et Zucc.) Maxim.、柔毛淫羊藿E. pubescens Maxim.、朝鲜淫羊藿E. koreanum Nakai.和巫山淫羊藿E. wushanense Ying.外,尚包括地方中药材标准收载的粗毛淫羊藿E. acuminatum Franch.、湖南淫羊藿E.hunanense (Hand.-Mazz.) Hand. Mazz.、黔岭淫羊藿E. leptorrhizum Stearn和四川淫羊藿E.sutchuenense Franch.,样品中包括4批掺假淫羊藿和1批伪品(树木类植物的叶片,品种不详)。40批淫羊藿商品的原植物及产地信息见表1。9种淫羊藿类药材的性状鉴定特征比较,见表2
对照品淫羊藿苷(批号110737-202017,纯度98.1%)、金丝桃苷(批号111521-201809,纯度94.9%)购自中国食品药品检定研究院,对照品朝藿定A(批号 P10J11F118161,纯度≥98%)、朝藿定B(批号 P19A11F121806,纯度≥98%)、朝藿定C(批号R01J11F118161,纯度≥98%)、和宝藿苷Ⅰ(批号 T19D9Z77828,纯度≥98%),乙腈(默克股份两合公司,批号 20240131,色谱纯),乙醇(国药集团化学试剂有限公司,批号 20210810,分析纯),水为屈臣氏水。
采用Aglent ODS(250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)-水(B)为流动相,梯度洗脱(0~30 min,24% A→26% A;30~31 min,26% A→45% A;31~45 min,45% A→47% A),检测波长270 nm,柱温30 ℃,流量1.0 mL·min-1,进样量10 μL。
取淫羊藿苷对照品适量,精密称定,加甲醇制成48 μg·mL-1浓度的对照品溶液。
分别取40批淫羊藿样品的叶片粉末约0.2 g,精密称定,置具塞锥形瓶中,精密加入稀乙醇20 mL,称量,超声(功率400 W,频率50 kHz)处理1 h,放冷,再称量,用稀乙醇补足减失的量,摇匀,滤过,即得。对照品溶液和供试品溶液的HPLC色谱图见图1
分别吸取“2.1.2”项下对照品溶液和“2.1.3”项下供试品溶液10 μL,注入液相色谱仪,测定,记录色谱图,即得。结果见表3
取淫羊藿苷对照品适量,精密称定,加甲醇制成12.5 μg·mL-1浓度的溶液。
取“2.1.3”供试品溶液0.5 mL,置50 mL量瓶中,加甲醇至刻度,摇匀。
分别取对照品溶液和40批供试品溶液,并以稀乙醇为空白,在270 nm测定吸收度,计算含量。40批样品测定结果见表3
取供试品约4 g,精密称定,置250~300 mL的锥形瓶中,精密加水100 mL,密塞,冷浸,前6 h振摇,再静置18 h,用干燥滤器迅速滤过,精密量取续滤液20 mL,置已干燥至恒重的蒸发皿中,在水浴上蒸干后,于105 ℃干燥3 h,置干燥器中冷却30 min,迅速精密称定。除另有规定外,以干燥品计算供试品中水溶性浸出物的含量。结果见表3
采用C18(250 mm×4.6 mm,5 μm)色谱柱,乙腈(A)-水(B)为流动相,洗脱梯度(0~40 min,30% A;40~50 min,30% A→32% A;50~70 min,32% A→35% A;70~75 min,35% A→45% A;75~80 min,30% A),柱温30 ℃,进样量10 μL,流速0.8 mL·min-1,检测波长270 nm。
精密称取金丝桃苷、朝藿定A、朝藿定B、朝藿定C、淫羊藿苷和宝藿苷Ⅰ对照品适量,加甲醇分别制成浓度为20.1、25.5、19.1、31.9、58.1和27.3 μg·mL-1的混合对照品溶液。
分别称取40批淫羊藿样品的叶片(过3号筛)约0.3 g,置具塞锥形瓶中,精密加入甲醇25 mL,称量,超声(功率400 W,频率50 kHz)处理30 min,放冷,再称量,用甲醇补足减失的量,摇匀,滤过。
取同一供试品(S1号)溶液,按“2.4.1”项下色谱条件连续进样6次,记录色谱峰。以淫羊藿苷(6号峰)为参照峰,计算共有峰的相对保留时间及相对峰面积,共有峰相对保留时间RSD均<0.8%,相对峰面积RSD均<1.1%,说明仪器的精密度良好。
取同一供试品(S1号)溶液,按“2.4.1”项下色谱条件分别在0、6、12、18、24 h进样,记录色谱峰。以淫羊藿苷(6号峰)为参照峰,计算共有峰的相对保留时间及相对峰面积,共有峰相对保留时间RSD均<1.2%,相对峰面积RSD均<1.9%,表明供试品溶液在24 h内稳定性良好。
取同一批供试品(S1号),按“2.4.3”项方法平行制备6份供试品溶液,按“2.4.1”项下色谱条件进样检测,记录色谱峰。以淫羊藿苷(6号峰)为参照峰,计算共有峰的相对保留时间及相对峰面积,共有峰相对保留时间RSD均<1.3%,相对峰面积RSD均<2.1%,表明本方法重复性良好。
将40批淫羊藿样品所得色谱图分别导入国家药典委员会“中药色谱指纹图谱相似度评价系统(2012版)”软件进行处理,得到40批淫羊藿药材的HPLC图谱,见图2。以10批淫羊藿E. brevicornu(S1~S6、S8~S11)建立对照图谱,设定S1号样品为参照图谱,时间窗宽度为0.5,按中位数法生成对照图谱,按照峰面积大于淫羊藿苷峰峰面积0.1%和分离度大于1.5的条件共选定了8个共有峰,与对照品指纹图谱比较,指认其中的6个峰,见图3。9种淫羊藿的HPLC图,见图4
将40批淫羊藿的7种含量指标导入SPSS 25.0软件进行主成分分析,经过标准化数据处理,特征值大于1的主成分有2个,累计贡献率为80.66%,见表4。以主成分为变量,得到主成分矩阵,见表5
根据公式;(其中,λ为特征值,α为因子得分,PCA为主成分得分)分别计算出主成分1、2的得分PCA1、PCA2,根据公式F=(61.01%PCA+19.65%PCA2)/80.66%,计算40批样品的综合得分F,得到40批淫羊藿样品的得分排名,如下表6
通过排序认为,9种淫羊藿植物来源中,载入《中国药典》中淫羊藿品种较地方中药材标准中记载其品种相比,整体排名较靠前,质量较优,如箭叶淫羊藿、淫羊藿和柔毛淫羊藿;产于陕西的箭叶淫羊藿排名第1和第3,可能与其总黄酮及浸出物含量较高有关;产于甘肃的淫羊藿排名比较分散,这与其采收季节[17]以及生长年限[18]有关;朝鲜淫羊藿和巫山淫羊藿质量居中;地方习用药材中粗毛淫羊藿质量良好,而湖南淫羊藿、四川淫羊藿质量较次以及黔岭淫羊藿最差,排序靠后;掺假淫羊藿和伪品排序在最后。主成分分析法能够反映出淫羊藿的质量状况,可用于淫羊藿类药材质量的系统性评价,对于不同品种、同品种不同产地淫羊藿有良好的预测性,有助于进一步分析其资源利用价值和筛选质量优良的药材产区。
以测得的40批次不同种淫羊藿样品中7个成分的含量作为评价指标,运用Origin软件进行热图分析,见图5
结果显示,从含量指标分析,S10、S25、S26和S27号4份样品总黄酮醇苷最高,相聚一类;S36(伪品)与S19、S20、S22、S23(4批掺假淫羊藿)、S35、S39、S40(3批总黄酮醇苷很低)的基本相聚一类;从产地分析,甘肃的20批样品中,有12批基本相聚一类;从炮制加工分析,S15、S16(2批羊油制淫羊藿)单独相聚;从淫羊藿品种分析,淫羊藿和箭叶淫羊藿的多数分别相聚一类,4种地方习用药材大致相聚一类。
在品种来源、产地、炮制和含量方面,聚类分析显示较好或一定预见的区分意义,说明淫羊藿的品种、产地以及不同生长环境等都影响了聚类效果。
对40批9种不同淫羊藿来源中所含黄酮醇苷、总黄酮及浸出物测定结果分析,所测定的3个成分含量平均值,淫羊藿分别为2.75%、6.44%和24.73%;柔毛淫羊藿分别为3.56%、7.75%、18.20%;箭叶淫羊藿分别为2.936%、6.59%、25.15%;朝鲜淫羊藿分别为1.19%、5.09%、19.7%;粗毛淫羊藿等4种地方药材所含3个成分含量差异明显。结果不同品种、同一品种的不同产地之间差异较大,需要采用多元统计方法进一步分析。
以生成的对照图谱对40批样品进行分析,结果产于甘肃的淫羊藿(E. brevicornu)相似度为0.979~0.855,大部分样品相似度在0.9以上,柔毛淫羊藿相似度为0.879~0.873,箭叶淫羊藿相似度为0.869~0.726,朝鲜淫羊藿相似度为0.708,巫山淫羊藿相似度为0.887,湖南淫羊藿相似度为0.829,粗毛淫羊藿相似度为0.806,四川淫羊藿相似度为0.124,黔岭淫羊藿相似度为0.146,掺假样品相似度在0.141~0.829,范围变化幅度较大,这与其掺假比例和淫羊藿品种有关联,伪品相似度为0.066。
通过主成分分析,认为淫羊藿(E. brevicornu)、箭叶淫羊藿和柔毛淫羊藿整体质量良好,质量排序在前列,除粗毛淫羊藿外,地方习用药材的整体质量较差,排序在最后。
聚类分析表明,在植物来源、产地、炮制和含量方面,聚类分析都显示较好且有一定预见的区分意义,聚类体现了植物、产地以及不同生长环境等的综合影响。
4批掺假的淫羊藿和1批伪品与含量低的黔岭淫羊藿、四川淫羊藿基本相聚一类,与主成分分析排序结果靠后基本一致。
我国药用淫羊藿野生资源丰富、产区广泛,随着市场需求的增长,各地不断发掘利用本地淫羊藿资源,致使商品淫羊藿更加复杂。调查发现,除《中国药典》收载的5种淫羊藿品种外,尚有粗毛淫羊藿(E. acuminatum) 、黔岭淫羊藿(E. leptorrhizum)、湖南淫羊藿(E. hunanense)和四川淫羊藿(E. sutchuenense),前2种在产地长期被视为“淫羊藿”收购[8],这些属于地方习用药材,在全国范围流通应予高度重视。
通过对9种植物40份样品的含量测定,认为箭叶淫羊藿中朝藿定C、总黄酮醇苷和总黄酮含量整体较高;淫羊藿(E. brevicornu)中淫羊藿苷、朝藿定B的含量整体较高;柔毛淫羊藿中朝藿定C含量整体较高,这与文献报道一致[12-15,19];巫山淫羊藿仅测定了1批,总黄酮含量较高,而不是同属植物中最高的,这与文献报道不一致[20]。同一植物来源的不同产地、不同植物来源的淫羊藿药材的指标成分含量存在较大差异。40批市售淫羊藿中有6批掺有伪品(未知植物),淫羊藿的掺假现象比较突出,应加大我国淫羊藿检验的监管力度。
以淫羊藿(E. brevicornu)建立的HPLC指纹图谱,与其他淫羊藿品种的相似度存在一定差异,对于伪品及其掺假伪品较高时的淫羊藿相似度差异比较明显,可以进行质量的评价,而掺假伪品低时,评价意义不大,需要根据伪品的特征性成分,建立掺假判别函数进行评价。要更加科学全面的分析淫羊藿类药材的质量优劣,需要做大量样品采集工作。本文调查了商品淫羊藿的植物来源,研究和分析了淫羊藿类药材质量,为临床应用和开发利用提供了技术支撑。
实验结果最终表明,淫羊藿类植物分布较广,不同植物来源的淫羊藿含量指标,可能因气候、土壤、地势地貌等因素的影响,导致次生代谢产物也存在一定差异,地理环境不仅造成淫羊藿种类的多样性,也是造成药材质量差异的主要因素。同一种淫羊藿(E.brevicornu)的不同产地相似度也有差异,可能受到采收时期不同或不同地方贮藏的影响所致。由于淫羊藿的品种复杂、采收加工和贮藏等不确定因素的影响,实现指纹图谱对不同品种来源及产地的淫羊藿品种质量评估,仍然需要大量的样品数据和建立不同化学成分指纹图谱等工作的探索研究。
