Objective: To develop a method to assess bioactivity of trasfer factor (TF) based on their protective effcets on Jurkat E6-1 cells. Methods: Proliferative effects of different concentrations of TF on 6-mercaptopurine(6-MP) treated Jurkat E6-1 cells detected by CCK-8 assay and the precision was validated. Results: TF exhibited protective activity to change the inhibitory concentration (IC50) value of 6-MP from(0.46±0.10)μg·mL-1 to (1.11±0.30)μg·mL-1 when treated with 20 μg·mL-1 TF. When cells were treated with 6-MP, there is a good linear relationship between the concentration of TF and cell survival rate, r>0.95. At each concentration level of the dose-response curve, the RSD was less than 20%. The median effective concentration (EC50) of TF was (28.49±9.60)μg·mL-1, and the confidence limit was less than 20%. Conclusions: TF significantly improved Jurkat E6-1 cell survival rate in dose dependent manner when challenged with 6-MP. It may be suitable for evaluate bioactivity of TF.

目的:建立基于人白血病T细胞株Jurkat E6-1的转移因子(TF)生物活性检测方法。方法:采用细胞毒性和增殖(CCK-8)法探索不同浓度TF对6-巯基嘌呤(6-MP)处理Jurkat E6-1细胞增殖的影响,并对该方法的精密度进行验证。结果:20 μg·mL-1 TF可使细胞的生存率明显提高,使6-MP的 IC50由(0.46±0.10)μg·mL-1右移至(1.11±0.30)μg·mL-1;当1.0 μg·mL-1 6-MP处理细胞时,TF的浓度和细胞生存率具有良好的线性关系,r>0.95。不同批次间在剂量-反应曲线的每个浓度水平上,RSD均小于20%。TF的半数有效浓度(EC50)为(28.49±9.60)mg·mL-1,可信限率小于20%。结论:TF可以显著提高暴露于6-MP的Jurkat E6-1细胞生存率,该作用呈剂量依赖性,可为TF生物活性测定提供客观、准确的评价手段。

Objective: To develop a method to assess bioactivity of trasfer factor (TF) based on their protective effcets on Jurkat E6-1 cells. Methods: Proliferative effects of different concentrations of TF on 6-mercaptopurine(6-MP) treated Jurkat E6-1 cells detected by CCK-8 assay and the precision was validated. Results: TF exhibited protective activity to change the inhibitory concentration (IC50) value of 6-MP from(0.46±0.10)μg·mL-1 to (1.11±0.30)μg·mL-1 when treated with 20 μg·mL-1 TF. When cells were treated with 6-MP, there is a good linear relationship between the concentration of TF and cell survival rate, r>0.95. At each concentration level of the dose-response curve, the RSD was less than 20%. The median effective concentration (EC50) of TF was (28.49±9.60)μg·mL-1, and the confidence limit was less than 20%. Conclusions: TF significantly improved Jurkat E6-1 cell survival rate in dose dependent manner when challenged with 6-MP. It may be suitable for evaluate bioactivity of TF.

转移因子(transfer factors,TF)又称Lawrence TF或可透析白细胞提取物(dialyzable leukocyte extracts,DLE),因其能够将致敏捐赠者的延迟性过敏反应和细胞介导的免疫力传递给非免疫接受者而得名[1]。TF是一种含多肽氨基酸和多核苷酸混合物的免疫调节药,具有不引起过敏反应、不产生抗体、无明显毒副作用等特点[2]。其主要作用机制可能是细胞介导的免疫反应,产生巨噬细胞游走抑制因子(macrophage migration inhibitory factor,MIF)和干扰素(interferon-γ,IFN-γ),抑制核因子κB(nuclear factor kappa-B,NF-κB),减少肿瘤坏死因子(tumor necrosis factorα,TNFα)和白介素4 (Interleukin-4,IL-4)水平。TF中加入核糖核酸酶(RNAse)或脱氧核糖核酸酶(DNAse)并不改变其转移能力[3]。TF主要来源于人或牛的白细胞,蛋黄,淋巴组织,动物的脾细胞及牛初乳等。临床用于感染、过敏、自身免疫等疾病,某些抗生素难以控制的病毒性或霉菌性细胞内感染的辅助治疗;亦对恶性肿瘤、湿疹、血小板减少、多次感染综合征以及慢性皮肤黏膜真菌病等免疫缺陷疾病有一定的治疗作用[4-5]
已知的TF活性测定方法,包括:e-玫瑰花结法、小鼠延迟型超敏反应的诱导、白细胞迁移法和IFN-γ分泌测定法[6-9],然而,这些方法费时耗力,又缺乏可靠性和重复性。硫唑嘌呤、6-巯基嘌呤(6-MP)、硫鸟嘌呤(6-thioguanine,6-TG)等嘌呤抗代谢药物可通过干扰核糖核酸(ribonucleic acid,RNA)、脱氧核糖核酸(deoxyribonucleic acid,DNA) 和蛋白质合成,从而促进T淋巴细胞的凋亡[10]。而TF担负重要的调节免疫应答功能,可以促进 T 淋巴细胞中胸腺细胞的成熟和分化。Jurkat E6-1细胞常用于研究T淋巴细胞增殖、功能以及信号传导等分子作用机制[11-13]。本文使用Jurkat E6-1细胞和6-MP建立了来源于猪脾的TF的生物活性测定方法。该方法的细胞易获得,且可准确测定TF的生物活性,以期为TF产品的生物活性提供更为便利的检测手段。
CCL-7-B-8二氧化碳细胞培养箱(ESCO公司);ACB-A1垂直流超净工作台(ESCO公司);DMIL倒置相差显微镜(LEICA公司);AXTDL5M台式冷冻离心机(盐城市安信实验仪器有限公司);K2双荧光细胞计数仪(Nexcelom公司);Spectramax plus 384型酶标仪(Molecular Device公司)。
TF[批号20240204、20240205、20240206,规格10 mL∶10 mg(多肽)∶300 μg(核酸)]购自大连百利天华制药有限公司,6-MP(CAS 50-44-2,批号K1827074)购自阿拉丁公司,RPMI 1640培养基购自北纳创联生物科技有限公司,胎牛血清购自Gibco公司;细胞增殖及毒性检测试剂盒(CCK-8)购自博士德公司,二甲基亚砜(DMSO)购自Sigma公司,磷酸盐缓冲溶液(PBS)购自麦克林公司。
Jurkat CloneE6-1人T淋巴细胞白血病细胞购自北纳创联生物科技有限公司。
将Jurkat E6-1细胞培养于含15%胎牛血清,并补充青霉素100 U·mL-1,链霉素0.1 mg·mL-1 RPMI 1640 培养基的25 mL培养瓶中,于37 ℃、5%CO2的培养箱内培养,细胞密度达到2×106个·mL-1时传代,待细胞长至对数生长期用于实验。
6-MP用DMSO溶解后,用灭菌注射用水稀释至所需浓度(DMSO的终浓度≤0.1%);TF用灭菌注射用水稀释至所需浓度;Jurkat E6-1细胞用PBS清洗2次,用含10%胎牛血清的RPMI 1640培养基调整所需细胞密度。以200 μL·孔-1(细胞数为2×104个·孔-1或4×104个·孔-1)接种于无菌96孔培养板中,按组别加入6-MP、TF处理细胞,TF组加入TF处理2 h后,再加入6-MP,同时设置空白培养基组和正常对照组,然后在37 ℃,5% CO2条件下继续培养48 h或72 h。取出培养板,每孔加入CCK-8溶液20 μL,放回培养箱内继续培养1~2 h,取出、轻轻混匀,用酶标仪在450/630 nm波长处测定吸收度(A)。计算细胞生存率[细胞生存率=(A样品组-A空白组)/(A正常对照组-A空白组)×100%]。
采用GrapPad Prism 8对数据进行统计学分析,计量资料以均数±标准差()表示,按文献方法计算EC50[14]。以P<0.05为差异显著,具有统计学意义。
6-MP对Jurkat E6-1细胞的毒性呈剂量、时间和细胞密度依赖性。如图1所示,不同培养时间和细胞密度均在6-MP质量浓度约为1.0 μg·mL-1时,细胞存活率变化趋于稳定。6-MP浓度在0.2~3.0 μg·mL-1,细胞接种数为2×104个·孔-1时,不同培养时间均可降低细胞生存率,且随时间变化较小。细胞接种数为4×104个·孔-1,培养48 h时,细胞生存率变化较小,在6-MP质量浓度为3.0 μg·mL-1时,细胞生存率降低50%左右;与培养48 h比较,72 h时在6-MP浓度约为1.0 μg·mL-1细胞生存率变化显著(P>0.05)。
细胞接种数为4×104个·孔-1,培养72 h时,TF可显著提高细胞生存率。结果见图2-A,50 μg·mL-1 TF使细胞生存率提高了(50.41±1.27)%(与6-MP组比较,P<0.000 1),TF仅使正常细胞生存率提高了(7.51±3.95)%(与正常组比较P>0.05)。结果表明,TF对正常的Jurkat E6-1细胞无增殖作用,但能有效提高受损的Jurkat E6-1细胞的生存率。
6-MP抑制Jurkat E6-1细胞增殖,半数抑制浓度(IC50)值为(0.46±0.10) μg·mL-1。20 μg·mL-1 TF可有效降低6-MP在0.3~5.0 μg·mL-1浓度范围的细胞毒性,使6-MP的IC50值右移至(1.11±0.30)μg·mL-1P<0.000 1),曲线下面积由166.1±2.7(6-MP)增加到228.9±10.0(TF+6-MP),说明TF可对抗6-MP对Jurkat E6-1细胞毒性作用,见图2-B
以200 μL·孔-1(细胞数为4×104个·孔-1)接种于无菌96孔培养板中,按组别加入1.0 μg·mL-1的6-MP及不同浓度的TF处理细胞,TF组加入TF 处理2 h后,再加入6-MP,同时设置空白培养基组和正常对照组,然后在37 ℃,5% CO2条件下继续培养72 h。测定及计算同方法“2.2”项。结果如图3显示,TF质量浓度在2.33~50.0 μg·mL-1范围内和细胞生存率具有良好的线性关系,r>0.95,符合线性与范围要求。
采用“3.3”建立的方法,以半数有效浓度(EC50)为指标,考察3批TF的生物活性及精密度。结果如表1所示,TF的EC50为(28.49±9.60) μg·mL-1n=3),其可信限均低于20%;经单因素方差分析(one-ANOVA)比较组间差异,3批EC50无显著性差异(P>0.05);不同批次独立样本在剂量-反应曲线的每个浓度水平上,细胞生存率的RSD均低于20%;曲线的斜率或截距之间没有显著差异(P>0.05),说明此方法具有良好的精密度和重复性,满足生物活性测定精密度要求。
TF是由多肽、氨基酸和多核苷酸组成的免疫调节药,其免疫调节活性测定存在以下几个难点:①免疫调节药的作用机制复杂多样;②不可能同时测定不同的作用机制,容易出现假阴性和/或假阳性的结果;③无法分别评估先天免疫和适应性免疫的影响以及它们之间的联系[15-18],这些原因导致免疫调节活性的测定变得很复杂。因此,检查免疫调节活性最有效的方法是单独测试某种特定的效果[19]。基于以上难点,有文献报道了人白细胞来源的DLE影响硫唑嘌呤作用下的Jurkat E6-1细胞增殖,用于DLE的生物活性评价[15,20]
Jurkat E6-1细胞系来源于人急性T淋巴细胞白血病细胞/人淋巴细胞瘤细胞,是一类永生化的T淋巴细胞系,它保留了人周围血T淋巴细胞的特性[21]。6-MP可通过多种机制诱导Jurkat E6-1细胞凋亡,由于6-MP的细胞内水平越高,代谢酶和多药耐药相关蛋白4(multidrug resistance-associated protein 4,MRP4)的活性就越高[22],这导致6-MP的抗增殖作用是非线性的浓度依赖性[23],同本研究中,6-MP的质量浓度从1.0到3.0 μg·mL-1时其细胞存活率无明显变化,结果一致。6-MP诱导Jurkat E6-1细胞凋亡具有时间依赖性,6-MP在孵育72 h后会诱导自噬体积累,从而促进凋亡[24]
根据本研究结果,方法最终选取细胞接种数为4×104个·孔-1,6-MP质量浓度为1.0 μg·mL-1,培养72 h时为实验条件。本研究的结果表明TF对正常的Jurkat E6-1细胞无促增殖作用,但能有效提高受损的Jurkat E6-1细胞的生存率;在一定的浓度范围内呈剂量反应相关,r>0.95;不同批次间重复性的RSD在20%以内,3批TF测定的EC50为(28.49±9.60) μg·mL-1,可信限均在20%以下,且3批样品的EC50无明显差异,精密度和重复性满足生物活性测定方法要求。
本研究以传代人淋巴细胞Jurkat E6-1作为靶细胞,基于TF调节T淋巴细胞的生物活性为原理,能很好地测定TF的生物活性。2020年版《中国药典》四部收录了多种利用细胞增殖/MTT比色测定生物活性的方法,都需要用活性质控参考品对结果进行校正。对于TF而言,目前缺少合适的活性质控参考品。本研究以TF抗6-MP损伤Jurkat E6-1细胞的EC50为指标,可有效评价TF的生物活性。综上所述,本研究开发的TF生物活性测定方法,靶细胞易得,重复性好,为后续分析TF的生物活性质量控制提供了研究思路和数据支持。
