Objective: To establish the HPLC fingerprint of Suhexiang pills and determine the contents of gallic acid, piperine, costunolide, eugenol and cinnamic acid simultaneously, and to provide reference for the quality control method of Suhexiang pills. Methods: The separation was performed on a Swell Chromplus C18 column (250 mm×4.6 mm, 5 μm) with methanol (A)-0.05% phosphoric acid solution (B) as the mobile phase at a flow rate of 1.0 mL·min-1. The detection wavelength was 220 nm and the column temperature was 30 ℃. Fifteen batches of Suhexiang pills were analyzed. The “Traditional Chinese Medicine Fingerprint Similarity Evalution System” was used to establish the reference fingerprint and the similarity analysis was evaluated in combination with cluster analysis and principal component analysis. Results: The fingerprint of Suhexiang pills was established,and the similarities of 15 batches of samples were above 0.9. Thirteen common peaks were identified,and five components including galliic acid,cinnamic acid,eugenol,piperine and costunolide were identified. The contents of the five components were 2.506-3.652 mg·g-1,0.666-0.876 mg·g-1,3.834-5.140 mg·g-1,0.884-1.306 mg·g-1 and 19.908-55.704 mg·g-1,respectively. Using cluster analysis,15 batches of Suhexiang pills were divided into four categories. Three principal components were selected,and the cumulative variance contribution rate was 76.102%,indicating that the principal components could contain most of the information of the original data. Conclusion: The fingerprint and multi-component content determination method of Suhexiang pills are stable and reliable,which can provide reference for the quality control and clinical application of Suhexiang pills.

目的:建立温通开窍药苏合香丸高效液相色谱指纹图谱以及同时测定没食子酸、胡椒碱、木香烃内酯、丁香酚和肉桂酸5个成分含量的方法,为苏合香丸质量控制方法提供参考。方法:采用Swell Chromplus C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇(A)-0.05%磷酸水溶液(B)为流动相,梯度洗脱,流速 1.0 mL·min-1,检测波长220 nm,柱温30 ℃,对15批苏合香丸进样分析;采用“中药指纹图谱相似度评价系统”生成对照指纹图谱,并进行相似度分析,结合聚类分析、主成分分析进行评价。结果:建立苏合香丸指纹图谱,15 批样品的相似度均大于0.9。共标定了13个共有峰,并指认了没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯5个成分,并建立5个成分的含量测定方法,测定其含量分别为2.506~3.652 mg·g-1、0.666~0.876 mg·g-1、3.834~5.140 mg·g-1、0.884~1.306 mg·g-1和19.908~55.704 mg·g-1。采用聚类分析将15 批苏合香丸分为4 类,主成分分析筛选出3个主成分,累计方差贡献率为76.102%,说明主成分可包含原有数据的绝大部分信息。结论:建立的苏合香丸指纹图谱及多成分含量测定方法稳定、可靠,可为苏合香丸的质量控制及临床应用提供参考。

目的:建立温通开窍药苏合香丸高效液相色谱指纹图谱以及同时测定没食子酸、胡椒碱、木香烃内酯、丁香酚和肉桂酸5个成分含量的方法,为苏合香丸质量控制方法提供参考。方法:采用Swell Chromplus C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇(A)-0.05%磷酸水溶液(B)为流动相,梯度洗脱,流速 1.0 mL·min-1,检测波长220 nm,柱温30 ℃,对15批苏合香丸进样分析;采用“中药指纹图谱相似度评价系统”生成对照指纹图谱,并进行相似度分析,结合聚类分析、主成分分析进行评价。结果:建立苏合香丸指纹图谱,15 批样品的相似度均大于0.9。共标定了13个共有峰,并指认了没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯5个成分,并建立5个成分的含量测定方法,测定其含量分别为2.506~3.652 mg·g-1、0.666~0.876 mg·g-1、3.834~5.140 mg·g-1、0.884~1.306 mg·g-1和19.908~55.704 mg·g-1。采用聚类分析将15 批苏合香丸分为4 类,主成分分析筛选出3个主成分,累计方差贡献率为76.102%,说明主成分可包含原有数据的绝大部分信息。结论:建立的苏合香丸指纹图谱及多成分含量测定方法稳定、可靠,可为苏合香丸的质量控制及临床应用提供参考。

Objective: To establish the HPLC fingerprint of Suhexiang pills and determine the contents of gallic acid, piperine, costunolide, eugenol and cinnamic acid simultaneously, and to provide reference for the quality control method of Suhexiang pills. Methods: The separation was performed on a Swell Chromplus C18 column (250 mm×4.6 mm, 5 μm) with methanol (A)-0.05% phosphoric acid solution (B) as the mobile phase at a flow rate of 1.0 mL·min-1. The detection wavelength was 220 nm and the column temperature was 30 ℃. Fifteen batches of Suhexiang pills were analyzed. The “Traditional Chinese Medicine Fingerprint Similarity Evalution System” was used to establish the reference fingerprint and the similarity analysis was evaluated in combination with cluster analysis and principal component analysis. Results: The fingerprint of Suhexiang pills was established,and the similarities of 15 batches of samples were above 0.9. Thirteen common peaks were identified,and five components including galliic acid,cinnamic acid,eugenol,piperine and costunolide were identified. The contents of the five components were 2.506-3.652 mg·g-1,0.666-0.876 mg·g-1,3.834-5.140 mg·g-1,0.884-1.306 mg·g-1 and 19.908-55.704 mg·g-1,respectively. Using cluster analysis,15 batches of Suhexiang pills were divided into four categories. Three principal components were selected,and the cumulative variance contribution rate was 76.102%,indicating that the principal components could contain most of the information of the original data. Conclusion: The fingerprint and multi-component content determination method of Suhexiang pills are stable and reliable,which can provide reference for the quality control and clinical application of Suhexiang pills.

苏合香丸为著名的温通开窍药,其用药历史悠久,最早见于唐·《玄宗开元广济方》,原名“吃力伽(白术丸)”,后来的《千金要方》和《外台秘要》均有记载[1],是中医临床抢救急危重症的有效药物,国家重点新药冠心苏合丸、麝香保心丸都是在其处方基础上精简研制而成。苏合香丸由苏合香、安息香、冰片、水牛角浓缩粉、人工麝香、檀香、沉香、丁香、香附、木香、乳香(制)、荜茇、白术、诃子肉、朱砂等15味药材配伍组成[2],其中苏合香、安息香共为君药,辟秽化痰、通行气血;木香、白檀香、沉香、乳香、丁香、香附为臣药,助行气解郁、散寒止痛、辟秽化浊、活血化痰。诸药相合共奏行气开窍、温中止痛之功,临床上用于痰迷心窍所致的痰厥昏迷、中风偏瘫、肢体不利,以及中暑、心胃气痛[3],有关研究指出在中风、新型冠状病毒感染、脑梗死、突发性耳聋、脑动脉硬化等疾病方面有确切疗效[4-11]
目前2020年版《中华人民共和国药典》仅采用气相色谱法对苏合香丸中的龙脑和丁香酚进行含量测定,而中药指纹图谱技术的信息量大,特征性强,能够更科学地显示中药质量控制标准的整体性与模糊性[12-14]。故本研究拟建立苏合香丸的HPLC指纹图谱,并采用化学计量学分析方法比较不同样品中化学成分的差异[15-19],分析影响苏合香丸质量的主要色谱峰,同时建立了苏合香丸中 5 个活性成分的含量测定方法,以期为苏合香丸的质量控制提供参考。
苏合香丸15批,批号分别为230601、230602、230603、230604、230701、230702、230703、230704、230801、230802、230803、231001、231002、231003和231004,规格为3 g·丸-1,均由成都九芝堂金鼎药业有限公司提供。对照品肉桂酸(含量99.96%,批号21032206)、丁香酚(含量99.15%,批号23033107)购自成都普菲德生物技术有限公司;胡椒碱(含量98%,批号23033101)、木香烃内酯(含量98%,批号23031307)购自成都康邦生物科技有限公司;没食子酸(含量91.5%,批号110831-201906)购自中国食品药品检定研究院。HPLC级甲醇、分析纯磷酸、硅藻土等均由成都市科隆化学品有限公司提供;水为超纯水,自制。
Waters 2695型高效液相色谱仪,Waters Empower色谱工作站;普利赛斯ES225SM-DR(E)十万分之一电子天平;赛多利斯BS224S万分之一电子天平;KQ-300E型超声清洗仪;上海安亭TGL-16G台式高速离心机。
采用Swell Chromplus C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇为流动相A,以0.05%磷酸水溶液流动相B,梯度洗脱(0~15 min,5% A→20% A;15~40 min,20% A→45% A;40~70 min,45% A→85% A;70~80 min,85% A→5% A;80~85 min,5% A),流速1.0 mL·min-1,检测波长220 nm,进样体积5 μL,柱温30 ℃。
分别取没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯的对照品适量,精密称定,置量瓶中,加80%甲醇水制成质量浓度分别为1.169、1.035、1.052、1.021、4.998 mg·mL-1的对照品储备溶液。
取苏合香丸10丸,精密称定,剪碎,取3 g,精密称定,加入硅藻土3 g,研匀,移至具塞锥形瓶中,加入80%甲醇水50 mL,称量,超声(功率300 W,频率40 kHz)处理15 min,取出放冷,再称量,用80%甲醇水补足减失的量,摇匀,10 000 r·min-1离心5 min,取上清液用0.45 μm有机微孔滤膜滤过,取滤液,即得。
取苏合香丸(批号230702),按“2.3”项下方法制备供试品溶液,按“2.1”项下条件连续进样6针,记录色谱图。以没食子酸峰(2号峰)为参照峰,计算各共有峰的相对保留时间(relative retention time,RRT)和相对峰面积(relative common peak area,RPA)的RSD。各共有峰RRT的RSD为0.14%~0.19%,RPA的RSD为0.20%~2.5%,均<3.0%,表明仪器精密度良好。
取苏合香丸(批号230702),按“2.3”项下方法制备供试品溶液,按“2.1”项条件分别在0、4、6、8、12 h进样分析,记录色谱图。以没食子酸色谱峰为参照峰,计算得各共有峰的RRT及RPA的RSD分别为0.20%~0.38%和0.35%~2.9%,均<3%,表明供试品溶液在室温条件下12 h内稳定性良好。
取苏合香丸(批号230702),按“2.3”项下方法平行制备供试品溶液6份,按“2.1”项下条件进样分析,记录色谱图。以没食子酸色谱峰为参照峰,计算得各共有峰的RRT及RPA的RSD分别为0.070%~0.15%和0.46%~2.5%,表明所建方法的重复性良好。
取15批苏合香丸,按“2.3”项下方法制备苏合香丸供试品溶液,按 “2.1”项下色谱条件进样检测,采用“中药色谱指纹图谱相似度评价系统”2012版,以S1为参照图谱,时间窗宽度为0.1,通过多点校正、Mark峰匹配,得15批苏合香丸指纹图谱共有模式及对照谱图谱,见图12表1。匹配后标定了13个共有峰,经过与对照品谱图及保留时间对比指认出5个色谱峰(见表2),分别为 2 号峰没食子酸、7 号峰肉桂酸、8号峰丁香酚、10 号峰胡椒碱、11号峰木香烃内酯。
以对照图谱为参照,对15批苏合香丸指纹图谱进行相似度评价,结果15批样品的相似度分别为0.992、0.999、0.992、0.998、0.997、0.996、0.999、0.978、0.991、0.998、0.995、0.992、0.994、0.995和0.996,均>0.9,表明相似度良好,不同生产批次间苏合香丸化学成分一致,质量稳定。
应用SPSS 22.0软件,以共有峰的峰面积为变量,进行系统聚类分析,见图3。 15 批苏合香丸可分为4类:Ⅰ类包括1~8号样品,10号样品,为2306、2307和2308批次样品,由此说明临近生产日期不同批次样品间差异较小,且各个批次的药品质量较为相似;Ⅱ类包括11~14号样品,为2310批次样品;15号样品与9号样品被单独分为两类,分别为第Ⅲ类及第Ⅳ类,分别为批号为230801和231004的样品,推测可能与该批样品的加工方法等有关。
利用 SPSS 22.0软件对数据标准化后进行主成分分析,由特征值大于1且贡献率大于75%的标准来看,当提取前3个主成分时,前3个特征值均大于1,其累计贡献率大于75%,即前3个主成分包含了12个共有成分的76.102% 的信息,符合主成分分析条件,结果见表3。结果苏合香丸第一主成分特征值为4.940,贡献率为41.169%,对应的载荷量较大者为肉桂酸、胡椒碱、丁香酚,说明第一主成分受这几个成分的影响较大,为多成分定量提供了依据;第二主成分特征值为3.090,贡献率为25.749%,对应的载荷量较大者为没食子酸、胡椒碱和木香烃内酯,说明第二主成分受这几个成分的影响比较大;第三主成分特征值为1.102,贡献率为9.184%,对应的载荷量较大者为4号峰、6号峰;苏合香丸前 3个主成分的累计方差贡献率则为 76.102%,说明了前3 个主成分能够概括原有数据的绝大部分信息。碎石图显示,提取出来的3个主成分的坡度较陡,表明前 3 个主成分是评价苏合香丸质量的代表性成分,结果见图4
精密移取各对照品溶液适量,置于10 mL量瓶中,加80%甲醇水稀释至刻度,配制没食子酸、肉桂酸、丁香酚、胡椒碱和木香烃内酯质量浓度分别为0.409 2、0.103 5、0.631 5、0.255 2和3.999 mg·mL-1的混合对照品溶液,作为标曲最高浓度。精密移取混合对照品溶液0.5、1.0、1.5、2.5和3.5 mL置于5 mL量瓶中,加80%甲醇水稀释并定容,混匀,制备得到不同浓度的对照品工作溶液,按“2.1”项下条件进样分析,以峰面积Y为纵坐标,对照品质量浓度X为横坐标,绘制标准曲线并计算回归方程,结果见表4
取苏合香丸(批号230702),按“2.3”项下方法制备供试品溶液,按“2.1”项下条件连续进样6针,计算没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯的峰面积的RSD分别为0.19%、0.49%、0.38%、0.23%和0.98%。表明仪器精密度良好。
取苏合香丸(批号230701),按“2.3”项下方法制备供试品溶液,按“2.1”项条件下,分别在0、4、6、8、12 h进样分析,计算没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯的峰面积的RSD分别1.7%、1.8%、2.6%、0.67%和2.2%,表明供试品溶液12 h内稳定。
取苏合香丸(批号230702),按“2.3”项下方法平行制备供试品溶液6份,按“2.1”项下条件进样分析,计算没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯平均含量分别为3.067、0.799、4.703、1.060、26.620 mg·g-1,RSD分别为2.8%、2.7%、2.9%、2.9%和2.2%,表明方法重复性良好。
精密称取已知含量的同一批次苏合香丸(批号230702)9份,平均分为3组,每组按5个成分已知含量的50%、100%和150%精密加入对照品。按“2.3”项下方法制备供试溶液,按“2.1”项下条件进样分析,计算各组分的加样回收率,结果没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯50%加标量的平均加样回收率分别为101.5%、101.9%、97.4%、100.1%和95.1%,RSD分别为1.3%、2.4%、2.4%、0.90%和0.90%;按100%加标量的平均加样回收率分别为96.3%、103.5%、102.0%、97.2%和 99.5%,RSD分别为0.80%、1.5%、0.80%、1.8%和0.60%;按150%加标量的平均加样回收率分别为102.4%、102.0%、104.4%、97.6%和97.0%,RSD分别为0.20%、0.60%、0.30%、0.50%和0.70%,符合测定要求。
分别取15批苏合香丸,按“2.3”项下方法制备供试品溶液,按“2.1”项下条件进样分析,记录各成分峰面积并计算含量,结果(表5)表明,15批苏合香丸中没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯含量范围分别为2.506~3.352、0.666~0.876、3.834~5.140、0.884~1.306和19.908~55.704 mg·g-1
本实验前期对供试品溶液的制备方法以及HPLC色谱条件筛选进行单因素考察,分别考察了不同体积分数(0%、20%、50%、80%、100%) 甲醇、不同提取时间(15、30、60 min)、不同测定波长(220、254、270、290、310、350、370 nm)、流动相中不同比例(0.1%、0.2%、0.05%)磷酸、不同色谱柱[Swell Chromplus C18(250 mm×4.6 mm,5 μm),Hypersil BDS C18(200 mm×4.6 mm,5 μm)和Spursil C18(250 mm×4.6 mm,5 μm)]对苏合香丸的指纹图谱及多成分含量的影响,以实验简便化,含量最优化进行优选,确定了提取方法为超声,提取溶剂为80%甲醇水,提取时间为15 min,测定波长为220 nm,流动相中含磷酸0.05%,色谱柱选择Swell Chromplus C18(250 mm×4.6 mm,5 μm)。
通过分析15批苏合香丸指纹图谱,指认了5个成分,并对这5个成分进行含量测定,采用聚类分析将15批苏合香丸分为4类,其中个别样品与其他批次样品差异较大,这可能与样品贮存运输等条件有关。采用主成分分析,前3个主成分包含了共有成分的76.102%,说明了前3个主成分能够概括原有数据的绝大部分信息,其中肉桂酸、胡椒碱、丁香酚、没食子酸、木香烃内酯等对主成分有较大影响,为多成分定量提供了依据。
本研究采用指纹图谱结合多成分含量测定及化学模式识别的分析方法,对15批苏合香丸进行了初步研究,经过与对照品的比对,指认了其中的5个共有峰分别为峰2(没食子酸)、峰7(肉桂酸)、峰8(丁香酚)、峰10(胡椒碱)、峰11(木香烃内酯),其中没食子酸是有机酚酸的一种,广泛存在于五倍子、锁阳、地榆、石榴等草药和水果中,已被证实具有抗菌、抗炎、抗氧化、抗病毒及抗癌等生物学作用[20-22];肉桂酸为肉桂及其挥发油中主要有效成分,具有解痉止痛、抗菌、抗肿瘤及促进免疫等药理作用[23-25];丁香酚是丁香油的主要成分,具有抗菌、抗病毒、抗炎、镇痛和抗氧化等生物学特性[26-28];胡椒碱是一种酰胺类生物碱成分,具有保护心血管、调节糖脂代谢、抗肿瘤、抗炎等药理作用[29-30];木香烃内酯为倍半萜内酯类化合物,是中药木香的主要化学成分,具有广泛的药理活性,包括抗肿瘤、降血糖、抗菌、抗炎等作用[31-32]。同时本研究也建立了苏合香丸中没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯等5个成分的含量测定方法,并对15批苏合香丸进行测定,结果15批样品中没食子酸、肉桂酸、丁香酚、胡椒碱、木香烃内酯含量范围分别为2.506~3.352、0.666~0.876、3.834~5.140、0.884~1.306和19.908~55.704 mg·g-1。该方法可以较快地筛选出影响不同批次苏合香丸质量的关键色谱峰,可为苏合香丸的质量控制提供依据。
