Objective:

To analyze the chemical constituents of Quyusanjie capsules by LC/MS, and establish a method for the determination of active ingredients in Quyusanjie capsules.

Methods:

Using UPLC-Q TOF MS/MS technology, the Hypersil Gold C18 column(100 mm×2.1 mm, 1.9 μm) was used, the mobile phase was acetonitrile(A) and 0.1% formic acid in water(B) with gradient elution, at a flow rate of 0.4 mL·min-1, the the column temperature was 40.0 ℃, and the mass spectrometry data was collected by negative ions mode scanning. Through database matching, elemental composition and fragment structure analysis, the main chemical substances in Quyusanjie capsules were identified. HPLC was used to qualitatively analyze the chemical components of Quyusanjie capsules. The Ultimate® AQ-C18 column(250 mm×4.6 mm, 5 μm) was used, the mobile phase was acetonitrile(A) -0.1% phosphoric acid(B) with gradient elution at the flow rate of 1.0 mL·min-1, the column temperature was 25 ℃, and the detection wavelength was 203 nm. The content of naringin, neohesperidin, notoginsenoside R1, ginsenoside Rg1, and ginsenoside Rb1 in 11 different batches of Quyusanjie capsules were determined using external standard method. QAMS method was established using ginsenoside Rg1 as the internal reference.

Results:

Twenty-nine compounds were identified from Quyusanjie capsule. The contents of naringin, neohesperidin, notoginsenoside R1, ginsenoside Rg1 and ginsenoside Rb1 measured by external standard method were 0.484-1.097 mg·g-1, 0.341-0.618 mg·g-1, 1.685-2.399 mg·g-1, 5.748-8.386 mg·g-1, 3.868-5.898 mg·g -1, respectively. Measured with the QAMS method, the contents of naringin, neohesperidin, notoginsenoside R1 and ginsenoside Rb1 were 0.516-1.153 mg·g-1, 0.372-0.667 mg·g-1, 1.794-2.580 mg·g-1, 4.373-6.690 mg·g-1, respectively. The relative error between the calculated values of the QAMS method and the measured value of the external standard method was less than 8.9%.

Conclusion:

UPLC-Q TOF MS/MS method can quickly identify the chemical components of Quyusanjie capsules. The established external standard method is stable and reliable, and can be used for the quality control of Quyusanjie capsules. The method of QAMS has good feasibility and is suitable for the determination of the daily production of Quyusanjie capsules.

目的:

应用液质联用法分析祛瘀散结胶囊的化学成分,构建祛瘀散结胶囊中有效成分的含量测定方法。

方法:

采用超高效液相色谱-三重四极杆飞行时间质谱联用(UPLC-Q TOF MS/MS)技术,色谱柱为Hypersil Gold C18(100 mm×2.1 mm,1.9 μm),以乙腈(A)-0.1%甲酸水(B)为流动相,梯度洗脱,体积流量0.4 mL·min-1,柱温40.0 ℃,质谱数据采集为负离子模式扫描;通过数据库匹配、元素组成和碎片结构分析,鉴定祛瘀散结胶囊中的主要化学成分。另外,采用HPLC,色谱柱为Ultimate®AQ-C18(250 mm×4.6 mm,5 μm),以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱,体积流量1 mL·min-1,柱温25 ℃,检测波长203 nm;以外标法计算,测定了11个不同批次祛瘀散结胶囊中的柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量,并以人参皂苷Rg1为内参物,建立一测多评法。

结果:

从祛瘀散结胶囊中鉴定出29个化合物;外标法测得柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量分别为0.484~1.097、0.341~0.618、1.685~2.399、5.748~8.386、3.868~5.898 mg·g-1,一测多评法测得的柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rb1的含量分别为0.516~1.153、0.372~0.667、1.794~2.580、4.373~6.690mg·g-1,一测多评法计算值与外标法实测值之间相对误差≤8.9%。

结论:

UPLC-Q TOF MS/MS法能快速鉴定祛瘀散结胶囊的化学成分;建立的外标法稳定可靠,可用于祛瘀散结胶囊的质量控制;以人参皂苷Rg1为内参物建立的一测多评法有较好的可行性,适用于祛瘀散结胶囊日常生产的含量测定。

目的:

应用液质联用法分析祛瘀散结胶囊的化学成分,构建祛瘀散结胶囊中有效成分的含量测定方法。

方法:

采用超高效液相色谱-三重四极杆飞行时间质谱联用(UPLC-Q TOF MS/MS)技术,色谱柱为Hypersil Gold C18(100 mm×2.1 mm,1.9 μm),以乙腈(A)-0.1%甲酸水(B)为流动相,梯度洗脱,体积流量0.4 mL·min-1,柱温40.0 ℃,质谱数据采集为负离子模式扫描;通过数据库匹配、元素组成和碎片结构分析,鉴定祛瘀散结胶囊中的主要化学成分。另外,采用HPLC,色谱柱为Ultimate®AQ-C18(250 mm×4.6 mm,5 μm),以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱,体积流量1 mL·min-1,柱温25 ℃,检测波长203 nm;以外标法计算,测定了11个不同批次祛瘀散结胶囊中的柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量,并以人参皂苷Rg1为内参物,建立一测多评法。

结果:

从祛瘀散结胶囊中鉴定出29个化合物;外标法测得柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量分别为0.484~1.097、0.341~0.618、1.685~2.399、5.748~8.386、3.868~5.898 mg·g-1,一测多评法测得的柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rb1的含量分别为0.516~1.153、0.372~0.667、1.794~2.580、4.373~6.690mg·g-1,一测多评法计算值与外标法实测值之间相对误差≤8.9%。

结论:

UPLC-Q TOF MS/MS法能快速鉴定祛瘀散结胶囊的化学成分;建立的外标法稳定可靠,可用于祛瘀散结胶囊的质量控制;以人参皂苷Rg1为内参物建立的一测多评法有较好的可行性,适用于祛瘀散结胶囊日常生产的含量测定。

Objective:

To analyze the chemical constituents of Quyusanjie capsules by LC/MS, and establish a method for the determination of active ingredients in Quyusanjie capsules.

Methods:

Using UPLC-Q TOF MS/MS technology, the Hypersil Gold C18 column(100 mm×2.1 mm, 1.9 μm) was used, the mobile phase was acetonitrile(A) and 0.1% formic acid in water(B) with gradient elution, at a flow rate of 0.4 mL·min-1, the the column temperature was 40.0 ℃, and the mass spectrometry data was collected by negative ions mode scanning. Through database matching, elemental composition and fragment structure analysis, the main chemical substances in Quyusanjie capsules were identified. HPLC was used to qualitatively analyze the chemical components of Quyusanjie capsules. The Ultimate® AQ-C18 column(250 mm×4.6 mm, 5 μm) was used, the mobile phase was acetonitrile(A) -0.1% phosphoric acid(B) with gradient elution at the flow rate of 1.0 mL·min-1, the column temperature was 25 ℃, and the detection wavelength was 203 nm. The content of naringin, neohesperidin, notoginsenoside R1, ginsenoside Rg1, and ginsenoside Rb1 in 11 different batches of Quyusanjie capsules were determined using external standard method. QAMS method was established using ginsenoside Rg1 as the internal reference.

Results:

Twenty-nine compounds were identified from Quyusanjie capsule. The contents of naringin, neohesperidin, notoginsenoside R1, ginsenoside Rg1 and ginsenoside Rb1 measured by external standard method were 0.484-1.097 mg·g-1, 0.341-0.618 mg·g-1, 1.685-2.399 mg·g-1, 5.748-8.386 mg·g-1, 3.868-5.898 mg·g -1, respectively. Measured with the QAMS method, the contents of naringin, neohesperidin, notoginsenoside R1 and ginsenoside Rb1 were 0.516-1.153 mg·g-1, 0.372-0.667 mg·g-1, 1.794-2.580 mg·g-1, 4.373-6.690 mg·g-1, respectively. The relative error between the calculated values of the QAMS method and the measured value of the external standard method was less than 8.9%.

Conclusion:

UPLC-Q TOF MS/MS method can quickly identify the chemical components of Quyusanjie capsules. The established external standard method is stable and reliable, and can be used for the quality control of Quyusanjie capsules. The method of QAMS has good feasibility and is suitable for the determination of the daily production of Quyusanjie capsules.

祛瘀散结胶囊原名块结消(抑瘤宝)胶囊,由白花蛇舌草、白英、山慈菇、夏枯草、三七、土鳖虫、蜈蚣、山楂、枳壳、仙鹤草、苦楝皮等15味药材组成,具有清热解毒、活血化瘀、软坚散结、扶正消瘤等功效[1],临床上已用于治疗乳癖、乳腺囊性增生病,无明显的毒副作用,疗效明显[2-3]。目前,对于祛瘀散结胶囊的研究主要集中在临床疗效药理作用和单个或2个成分的含量测定方面[4-6],但其化学成分研究及使用一测多评法测定多成分含量的研究尚未见报道[7-9],该制剂组成复杂,产品质量控制极为重要,而在祛瘀散结胶囊现行标准中,仅使用薄层色谱法测定人参皂苷Rg1和人参皂苷Rb1的含量[10],而利用薄层色谱法进行定量分析的操作较复杂,重复性和稳定性不及HPLC。因此,本文先通过UPLC-Q TOF MS/MS技术对祛瘀散结胶囊化学成分进行定性分析,其次,基于HPLC技术采用外标法和一测多评法测定祛瘀散结胶囊中5个成分的含量,进一步为药效物质基础研究及质量控制提供科学依据。
Sciex X500R Q-TOF液质联用仪(Sciex公司),用于定性分析;Waters e2695高效液相色谱仪(沃特世科技(上海)有限公司),用于耐用性分析;Acquity Arc超高效液相色谱仪(沃特世科技(上海)有限公司),用于含量测定;SB-800DTD超声波清洗机(宁波新芝生物科技股份有限公司);SQP十万分之一电子分析天平(赛多利斯科学仪器有限公司)。
对照品人参皂苷Rb1(批号PS000787,含量≥98.0%)、人参皂苷Rg1(批号PS010958,含量≥98.0%)、三七皂苷R1(批号PS010384,含量≥98.0%)、柚皮苷(批号PS001113,含量≥98.0%)、新橙皮苷(批号PS010413,含量≥99.0%)均购于成都普思生物科技股份有限公司;实验溶剂有屈臣氏蒸馏水、色谱级甲醇、色谱级乙腈、色谱级磷酸;编号为S1~S11的11批祛瘀散结胶囊均由恒拓集团广西圣康制药有限公司提供,批号分别为200301、200401、200402、200403、200404、200901、200902、200903、200904、200906、200905(规格:每粒0.48 g)。夏枯草、山慈菇、白花蛇舌草、白英、土鳖虫、三七、山楂、仙鹤草、黄芪、蜈蚣、枳壳、苦楝皮、冰片、甘草、麦芽均购自广西南宁老百姓大药房,经广西中医药大学药学院奉建芳教授鉴定均为正品。
将同一批次的10粒祛瘀散结胶囊内容物混合均匀,取祛瘀散结胶囊粉末约0.5 g,精密称定,置于50 mL具塞锥形瓶中,精密移取50%甲醇水溶液20 mL,密塞,称量,超声(功率508 W,频率40 kHz)处理1 h,放冷,再称量,用50%甲醇水溶液补足减失的量,滤过,取续滤液,过0.22 μm微孔滤膜,即得。
精密称取对照品柚皮苷2.64 mg、新橙皮苷1.20 mg,分别置于2 mL量瓶中,三七皂苷R1 9.35 mg置于5 mL量瓶中,人参皂苷Rg1 13.50 mg、人参皂苷Rb1 10.70 mg,分别置于10 mL量瓶中,加甲醇定容至刻度,最终制成上述成分质量浓度分别为1.32、0.6、1.87、1.35、1.07 mg·mL-1的对照品溶液。精密吸取各对照品溶液适量,置于25 mL量瓶中,用50%甲醇水溶液定容,得到柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1质量浓度分别为0.052 8、0.024 0、0.149 6、0.324 0、0.214 0 mg·mL-1的混合对照品溶液,保存于4 ℃冰箱中,备用。
按照处方比例及制剂工艺,分别制成缺三七、缺枳壳的阴性样品,按“2.1.1”项方法操作,即得。
采用Hypersil Gold C18(100 mm×2.1 mm,1.9 μm)色谱柱,以乙腈(A)-0.1%甲酸水(B)为流动相,梯度洗脱(0~1 min,5% A;1~5min,5% A→28% A;5~10 min,28% A→50% A;10~15 min,50% A→75% A;15~20 min,75% A→95% A),体积流量0.4 mL·min-1,柱温40.0 ℃,进样量3 μL。
电喷雾电离源(ESI),负离子模式;雾化气压(GS1):3.79×105 Pa;扫描范围为m/z 100~1 500;碰撞能量为-35 eV;离子源温度为600 ℃;喷雾电压为-4 500 V。
采用UPLC-Q TOF MS/MS对祛瘀散结胶囊进行分析,选择负离子模式对供试品溶液进行扫描,总离子流图(total ions current,TIC)见图1。通过SCIX OS数据处理系统对数据进行分析处理,根据一级质谱信息确定精确相对分子质量,二级质谱获得裂解信息,综合分析色谱峰保留时间及质谱碎裂规律,并结合文献相关信息[11-20],共鉴定出29个化学成分,见表1
采用Ultimate®AQ-C18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱(0~12 min,15% A;12~60 min,15% A→36% A;60~65 min,36% A→90% A),体积流量1 mL·min-1,柱温25 ℃,检测波长203 nm,进样量20 μL。取混合对照品溶液、供试品溶液及各阴性样品溶液,分别按上述色谱条件进样,记录色谱图,结果表明,阴性样品溶液色谱图中相应位置上无干扰峰出现,见图2
分别精密吸取“2.1.2”项的混合对照品溶液1、2、4、5、6、8 mL于10 mL量瓶中,加50%甲醇水定容至刻度,稀释制成6个线性标准溶液1~6,按“2.3.1”项色谱条件进样测定,以峰面积(Y)为纵坐标,质量浓度(X)为横坐标进行回归。结果见表2,表明各成分在各自线性范围内线性关系良好。
取“2.3.2”项的3号混合对照品溶液,按“2.3.1”项下条件,连续进样6次。结果柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1峰面积RSD的分别为1.6%、1.7%、1.8%、1.8%、1.9%,表明仪器精密度良好。
取同一批样品(S2),按“2.1.1”项下方法制备供试品溶液,室温放置0、2、4、8、12、24 h,按“2.3.1”项下条件进样测定,结果柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1峰面积的RSD分别为1.5%、0.40%、2.2%、0.65%、0.47%,表明供试品溶液在24 h内稳定性良好。
取同一批样品(S2),按“2.1.1”项下方法制备供试品溶液6份,按“2.3.1”项下条件进样测定,结果表明,在203 nm下,柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的平均含量分别为0.77、0.55、1.8、6.2、4.2 mg·g-1,对应的RSD分别为2.2%、2.3%、1.6%、1.1%、1.0%,表明该方法重复性良好。
取已知各成分含量的同一批样品(S2)6份,每份约0.25 g,精密称定,每份精密加入柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的质量浓度分别为4.224、2.160、6.579、23.244、21.449 μg·mL-1的对照品溶液各1 mL,按“2.1.1”项下方法制备供试溶液,按“2.3.1”项下条件进样,计算回收率。结果见表3,显示回收率符合《中华人民共和国药典》要求。
取“2.3.2”项下1~6号共6个不同浓度的混合线性标准溶液,按“2.3.1”项下条件进样测定,以人参皂苷Rg1为内参物,用公式1计算其他4个成分的相对校正因子,结果见表4
As为内参物对照品s的峰面积,Cs为内参物对照品s的质量浓度,Ai为某待测成分对照品i的峰面积,Ci为某待测成分对照品i的质量浓度。
分别考察不同仪器(Waters e2695和ACQUITY Arc高效液相色谱仪)和不同色谱柱(Ultimate® AQ-C18、Ultimate LP-C18、Phenomenex Kinetex XB-C18,规格均为250 mm×4.6 mm,5 μm)对fs/i的影响,结果见表5。结果表明在更换不同色谱柱和不同仪器时各成分的相对校正因子重现性良好(RSD<5%)。
本实验考察了不同柱温(23、25、27、29 ℃)对各成分fs/i的影响。结果 (表6) 表明,改变柱温对各成分的fs/i值无明显的影响(RSD<5%)。
本实验考察了不同体积流量(0.8、1.0、1.2 mL·min-1)对各成分fs/i的影响,结果见表7。结果表明改变体积流量对各成分的fs/i值无明显的影响(RSD<5%)。
色谱峰的准确定位是保证一测多评法应用的前提[21-22]。为保证色谱峰的准确定位,本实验中用混合对照品溶液考察待测成分在2台不同仪器、3根不同色谱柱中的保留时间差值和相对保留时间。结果见表89,从结果可以看出,用保留时间差值定位色谱法波动较大,而相对保留时间波动较小(RSD均<5%),所以用相对保留时间定位色谱峰比较合理。
分别精密称取11批次的祛瘀散结胶囊内容物,分别按“2.1.1”项下方法制备供试品溶液,按“2.3.1”项下条件进样测定,分别采用外标法、一测多评法计算含量,结果见表10。11批祛瘀散结胶囊中5个化学成分(柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1)的一测多评法计算值与外标法实测值之间的相对误差(RD)≤8.9%。
现代药理学研究表明,人参皂苷Rg1、人参皂苷Rb1、三七皂苷R1等具有活血、消炎止痛、通脉活络、抑制血小板聚集等作用[23-26];柚皮苷和新橙皮苷是枳壳药材中含量最高的黄酮类物质,有理气宽中、行滞消胀的功效[27-29],且这些成分是2020年版《中华人民共和国药典》三七和枳壳检测的指标性成分。各成分相互协同,共同发挥药效。因此,选用这些指标性成分对祛瘀散结胶囊进行多成分分析,能为其药效物质基础研究和质量控制提供参考。
本实验应用UPLC-Q TOF MS/MS技术,通过高分辨率质谱获得化合物精确相对分子质量,再根据色谱峰保留时间及质谱碎裂规律,结合文献相关信息,共鉴定出29个化学成分。在对祛瘀散结胶囊进行化学成分分析过程中,选择了正离子和负离子模式对样品进行了扫描,与正离子模式相比,样品中的化学成分在负离子模式下的响应信号更好,所呈现的碎片信息更丰富。并且,为了使化学成分有更好的峰形,在流动相中加入了0.1%的甲酸,在负离子模式下,一级质谱更容易形成[M-H]-和[M+HCOO]-的准分子离子峰,故最终选择分析负离子检测模式下样品的化学成分。
采用HPLC测定祛瘀散结的含量时,分别考察了Ultimate® AQ-C18、Ultimate® PG-C18、Ultimate® LP-C18、Phenomenex Kinetex® XB-C18色谱柱的洗脱能力和分离效能,最终选用Ultimate® AQ-C18色谱柱。实验采用梯度洗脱,通过比较不同流动相系统(甲醇-水、乙腈-水),发现乙腈-水系统洗脱能力较强而峰形存在拖尾现象。在此基础上,加入酸(0.1%磷酸、0.1%甲酸)改善峰形,最终发现使用0.1%磷酸水-乙腈洗脱系统峰形最好。实验前期对检测波长(203、210、345 nm)进行了考察,发现203 nm下色谱峰的整体响应度较好。本实验还考察了不同提取溶剂(甲醇、70%甲醇水、50%甲醇水)对实验的影响,发现用50%的甲醇水超声提取时,反映的色谱信息更丰富。
本文使用一测多评法和外标法同时对祛瘀散结胶囊中柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1 5个成分的含量进行测定,其中,外标法具有稳定可靠的特点,而一测多评法的含量测定结果却与外标法的含量测定结果有3.5%~8.9%的相对误差,产生较大误差的原因可能是相对校正因子的影响,相对校正因子的重现性考察内容包括不同实验室、不同仪器及色谱柱、不同流速、不同检测波长、不同柱温等,本文中已经进行了不同仪器和色谱柱、不同流速、不同柱温的考察,相对校正因子的RSD均<5%,但实际测量值和计算值还是出现了偏差,说明一测多评法仍不适用于祛瘀散结胶囊中成分的精准含量测定,但比较适合于日常生产的含量计算。本文建立祛瘀散结胶囊多指标质量评价方法对稳定该制剂产品质量以及确保临床疗效一致性具有重要意义。含量测定结果表明不同批次的5个成分存在一定的差异,RSD为13%~29%,可能是原料药材批次不同所导致的,但这也是中药复方普遍存在的问题,所以,企业在进行生产的时候,应加强原料药材的质量控制,尽可能保证原料药材为品质优良的道地药材,或者尽量保证药材来源的一致性。
采用一测多评法和外标法测定11批祛瘀散结胶囊中5个成分的含量,为全面提升祛瘀散结胶囊的质量提供了新方法,在缺少对照品的情况下,也能够实现对祛瘀散结胶囊多指标成分的含量测定。本实验结果表明,外标法适用于祛瘀散结胶囊中柚皮苷、新橙皮苷、三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1 的5个成分的精准含量测定,一测多评法比较适合于对精准度要求不苛刻的日常生产的含量计算。
