Objective:

To establish a method of thin-layer chromatographic identification,HPLC fingerprints and a method of the content determination of 13 chemical constituents (chlorogenic acid,caffeic acid,ferulic acid,senkyunolide I,senkyunolide H,coniferyl ferulate,senkyunolide A,butylphthalide,sedanolide,Z-ligustralactone,butenylylphthalide,levistilide A,and linoleic acid) in Ligusticum sinense Oliv. cv. Chaxiong Mss. and to provide a basis for the quality control of Ligusticum sinense Oliv. cv. Chaxiong Mss.

Methods:

Thin-layer chromatography was used for the qualitative identification of senkyunolide I,scopoletin,ferulic acid,levistilide A and Z-ligustilide in Ligusticum sinense Oliv. cv. Chaxiong Mss. using n-hexane,ethyl acetate and acetic acid (10:5:0.5) as the mobile phase. The separation was carried out on an Acutfex PA-C18 column (250 mm×4.6 mm,5 μm),with gradient elution of 0.1% aqueous phosphoric acid solution and acetonitrile at a flow rate of 1 mL·min-1. The column temperature was 30 ℃,the injection volume was 10 μL,and the detection wavelengths were 210 nm and 285 nm. The fingerprints of 54 batches of Ligusticum sinense Oliv. cv. Chaxiong Mss. were constructed. SPSS and SIMCA software were used for the chemical pattern recognition analysis,and 13 chemical components were determined at the same time.

Results:

A thin-layer chromatographic method was established for the identification of senkyunolide I,scopoletin,ferulic acid,levistilide A and Z-ligustilide in Ligusticum sinense Oliv. cv. Chaxiong Mss. The HPLC fingerprints of Ligusticum sinense Oliv. cv. Chaxiong Mss. were established,21 common peaks were found,13 common peaks were identified,and the similarities of all batches of the herbs were above 0.9,except sample S47. The herbs could be roughly divided into four groups according to the collection time and altitude by clustering analysis. OPLS-DA screened out seven differential constituents:peak 17(Z-ligustilide),peak 13 (senkyunolide A),peak 12 (coniferyl ferulate),peak 5 (ferulic acid),peak 6,peak 16 (sedanolide),and peak 2 (chlorogenic acid). A method was established for the determination of chlorogenic acid,caffeic acid,ferulic acid,senkyunolide I,senkyunolide H,coniferyl ferulate,senkyunolide A,butylphthalide,sedanolide,Z-ligustralactone,butenylylphthalide,levistilide A and linoleic acid in Ligusticum sinense Oliv. cv. Chaxiong Mss. with good linearity,precision,stability,reproducibility,and spiked recoveries. And the content ranges of the above-mentioned compounds in the herbs were 0.03-3.80 mg·g-1,0.004-0.055 mg·g-1,0.17-0.82 mg·g-1,0.08-0.57 mg·g-1,0.01-0.19 mg·g-1,0.04-2.51 mg·g-1,1.37-11.64 mg·g-1,0.15-0.58 mg·g-1,0.18-4.22 mg·g-1,2.69-9.53 mg·g-1,0.15-0.59 mg·g-1,0.07-0.62 mg·g-1 and 0.31-6.04 mg·g-1,respectively.

Conclusion:

The method of thin-layer chromatography,HPLC fingerprints,and the determination method of the 13 constituents in Ligusticum sinense Oliv. cv. Chaxiong Mss. are simple,accurate and reliable,and can provide a basis for the evaluation of the quality of Ligusticum sinense Oliv. cv. Chaxiong Mss..

目的:

建立茶芎的薄层色谱鉴别方法、HPLC指纹图谱以及绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸13个化学成分含量测定方法,为茶芎药材的质量控制提供依据。

方法:

采用薄层色谱法,以正己烷-乙酸乙酯-冰醋酸(10:5:0.5)为展开剂,对茶芎中洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯进行鉴别;采用Acutfex PA-C18 (250 mm×4.6 mm,5 μm)色谱柱,以0.1%磷酸水溶液-乙腈为流动相,进行梯度洗脱,体积流量1 mL·min-1,柱温30 ℃,进样量10 μL,检测波长210、285 nm,对54批茶芎药材进行指纹图谱构建;采用SPSS和SIMCA软件进行化学模式识别分析,同时测定13个化学成分的含量。

结果:

建立了茶芎药材中洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯的薄层色谱鉴别方法;建立了茶芎的HPLC指纹图谱,标定了21个共有峰,指认出13个共有峰,除S47号药材外,其他批次药材相似度均>0.9;聚类分析后可以按照采集时间和海拔分为4类;OPLS-DA筛选出峰17(Z-藁本内酯)、峰13(洋川芎内酯A)、峰12(阿魏酸松柏酯)、峰5(阿魏酸)、峰6、峰16(新蛇床内酯)、峰2(绿原酸)7个差异性成分;建立了茶芎中绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸的含量测定方法,其线性、精密度、稳定性、重复性、加样回收率均良好,上述各成分在药材中含量范围分别为0.03~3.80、0.004~0.055、0.17~0.82、0.08~0.57、0.01~0.19、0.04~2.51、1.37~11.64、0.15~0.58、0.18~4.22、2.69~9.53、0.15~0.59、0.07~0.62、0.31~6.04 mg·g-1

目的:

建立茶芎的薄层色谱鉴别方法、HPLC指纹图谱以及绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸13个化学成分含量测定方法,为茶芎药材的质量控制提供依据。

方法:

采用薄层色谱法,以正己烷-乙酸乙酯-冰醋酸(10:5:0.5)为展开剂,对茶芎中洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯进行鉴别;采用Acutfex PA-C18 (250 mm×4.6 mm,5 μm)色谱柱,以0.1%磷酸水溶液-乙腈为流动相,进行梯度洗脱,体积流量1 mL·min-1,柱温30 ℃,进样量10 μL,检测波长210、285 nm,对54批茶芎药材进行指纹图谱构建;采用SPSS和SIMCA软件进行化学模式识别分析,同时测定13个化学成分的含量。

结果:

建立了茶芎药材中洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯的薄层色谱鉴别方法;建立了茶芎的HPLC指纹图谱,标定了21个共有峰,指认出13个共有峰,除S47号药材外,其他批次药材相似度均>0.9;聚类分析后可以按照采集时间和海拔分为4类;OPLS-DA筛选出峰17(Z-藁本内酯)、峰13(洋川芎内酯A)、峰12(阿魏酸松柏酯)、峰5(阿魏酸)、峰6、峰16(新蛇床内酯)、峰2(绿原酸)7个差异性成分;建立了茶芎中绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸的含量测定方法,其线性、精密度、稳定性、重复性、加样回收率均良好,上述各成分在药材中含量范围分别为0.03~3.80、0.004~0.055、0.17~0.82、0.08~0.57、0.01~0.19、0.04~2.51、1.37~11.64、0.15~0.58、0.18~4.22、2.69~9.53、0.15~0.59、0.07~0.62、0.31~6.04 mg·g-1

结论:

建立的茶芎薄层色谱鉴别、HPLC指纹图谱及13个成分的含量测定方法操作简便,准确可靠,可为茶芎药材质量评价提供依据。

Objective:

To establish a method of thin-layer chromatographic identification,HPLC fingerprints and a method of the content determination of 13 chemical constituents (chlorogenic acid,caffeic acid,ferulic acid,senkyunolide I,senkyunolide H,coniferyl ferulate,senkyunolide A,butylphthalide,sedanolide,Z-ligustralactone,butenylylphthalide,levistilide A,and linoleic acid) in Ligusticum sinense Oliv. cv. Chaxiong Mss. and to provide a basis for the quality control of Ligusticum sinense Oliv. cv. Chaxiong Mss.

Methods:

Thin-layer chromatography was used for the qualitative identification of senkyunolide I,scopoletin,ferulic acid,levistilide A and Z-ligustilide in Ligusticum sinense Oliv. cv. Chaxiong Mss. using n-hexane,ethyl acetate and acetic acid (10:5:0.5) as the mobile phase. The separation was carried out on an Acutfex PA-C18 column (250 mm×4.6 mm,5 μm),with gradient elution of 0.1% aqueous phosphoric acid solution and acetonitrile at a flow rate of 1 mL·min-1. The column temperature was 30 ℃,the injection volume was 10 μL,and the detection wavelengths were 210 nm and 285 nm. The fingerprints of 54 batches of Ligusticum sinense Oliv. cv. Chaxiong Mss. were constructed. SPSS and SIMCA software were used for the chemical pattern recognition analysis,and 13 chemical components were determined at the same time.

Results:

A thin-layer chromatographic method was established for the identification of senkyunolide I,scopoletin,ferulic acid,levistilide A and Z-ligustilide in Ligusticum sinense Oliv. cv. Chaxiong Mss. The HPLC fingerprints of Ligusticum sinense Oliv. cv. Chaxiong Mss. were established,21 common peaks were found,13 common peaks were identified,and the similarities of all batches of the herbs were above 0.9,except sample S47. The herbs could be roughly divided into four groups according to the collection time and altitude by clustering analysis. OPLS-DA screened out seven differential constituents:peak 17(Z-ligustilide),peak 13 (senkyunolide A),peak 12 (coniferyl ferulate),peak 5 (ferulic acid),peak 6,peak 16 (sedanolide),and peak 2 (chlorogenic acid). A method was established for the determination of chlorogenic acid,caffeic acid,ferulic acid,senkyunolide I,senkyunolide H,coniferyl ferulate,senkyunolide A,butylphthalide,sedanolide,Z-ligustralactone,butenylylphthalide,levistilide A and linoleic acid in Ligusticum sinense Oliv. cv. Chaxiong Mss. with good linearity,precision,stability,reproducibility,and spiked recoveries. And the content ranges of the above-mentioned compounds in the herbs were 0.03-3.80 mg·g-1,0.004-0.055 mg·g-1,0.17-0.82 mg·g-1,0.08-0.57 mg·g-1,0.01-0.19 mg·g-1,0.04-2.51 mg·g-1,1.37-11.64 mg·g-1,0.15-0.58 mg·g-1,0.18-4.22 mg·g-1,2.69-9.53 mg·g-1,0.15-0.59 mg·g-1,0.07-0.62 mg·g-1 and 0.31-6.04 mg·g-1,respectively.

Conclusion:

The method of thin-layer chromatography,HPLC fingerprints,and the determination method of the 13 constituents in Ligusticum sinense Oliv. cv. Chaxiong Mss. are simple,accurate and reliable,and can provide a basis for the evaluation of the quality of Ligusticum sinense Oliv. cv. Chaxiong Mss..

茶芎来源于伞形科植物茶芎Ligusticum sinense Oliv. cv. Chaxiong Mss.的干燥根茎[1],是江西省特色中药材之一,主产于江西九江地区,其味辛,性温,归肝、心经,具有活血行气、祛风止痛等功效[2]。茶芎古代名抚芎,与川芎、西芎等合称为芎,最早出现于《神农本草经》之中,当地人将其与茶叶一起泡水饮用,可治疗感冒头痛,故名茶芎[3-4]。目前从茶芎中分离得到苯酞类、有机酸类、黄酮类、香豆素类、挥发油、生物碱类等化学成分,现代药理研究表明,茶芎在中枢神经系统、心血管系统、子宫平滑肌等方面具有显著的药理作用,可用于治疗头痛、风湿痹痛、痛经、闭经、产后淤阻腹痛等疾病。有研究经过染色体核型分析发现茶芎是与川芎同源的三倍体植物[5],临床上常与川芎混用。
茶芎现收载于《江西省中药材标准》(2014年版),其中仅对欧当归内酯A进行了薄层色谱鉴别,以正丁基苯酞为含量测定指标[6],该标准对茶芎药材质量评价具有一定的局限性。指纹图谱作为一种中药质量控制模式,可以结合化学模式识别分析对药材进行有效分析,并结合薄层色谱鉴别及多种化学成分的含量测定对药材质量进行评价[7]。因此,本研究基于对茶芎质量评价的完善,收集来自江西九江地区的茶芎样品54批,采用薄层色谱对茶芎中洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯5个成分进行鉴别,构建了茶芎的HPLC指纹图谱并建立了绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸13个化学成分的含量测定方法,为茶芎药材的整体质量评价提供依据。
MS 105DU型万分之一分析天平(METTLER TOLEDO公司);KQ-250V型超声波清洗器(昆山市超声仪器有限公司);REPROSTAR-3薄层色谱成像仪(卡玛公司);岛津LC-20 AT高效液相色谱仪(岛津公司);HC-3016R高速冷冻离心机(安徽中科中佳科学仪器有限公司);RT-02 B粉碎机(泓荃制药机械公司)。
对照品绿原酸(批号110753-202119)、阿魏酸(批号110773-201915)、咖啡酸(批号110885-201703)、欧当归内酯A(批号111826-202107)、亚油酸(批号111622-202105)购自中国食品药品检定研究院,洋川芎内酯I(批号Y08501904030),阿魏酸松柏酯(批号A00111811003)、洋川芎内酯A(批号RFS-Y08301903021)、正丁基苯酞(批号RFS-Z03702108010)、新蛇床内酯(批号X05802102001)、丁烯基苯酞(批号RFS-D05701905006)购自成都瑞芬思生物科技有限公司,洋川芎内酯H(批号5846)购自上海诗丹德标准技术服务有限公司,Z-藁本内酯(批号DST230307-007)、东莨菪内酯(批号DST210304-064)购自成都乐美天医药科技有限公司,以上对照品质量分数均≥98%;乙腈为色谱纯,其余试剂为分析纯,水为娃哈哈矿泉水。
54批药材均采集或收集于江西德安、武宁、瑞昌3个县,经江西中医药大学钟国跃教授鉴定为茶芎Ligusticum sinense Oliv. cv. Chaxiong Mss.的根茎。去除泥沙和须根后烘干,样品来源信息见表1
称取S54号茶芎药材粉末(过4号筛)约1 g,加入甲醇25 mL超声(功率250 W,频率40 kHz)提取30 min,滤过,滤液浓缩至干,残渣加入甲醇1 mL溶解,即得。
称取洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯的对照品适量,分别加甲醇制成质量浓度分别为1、0.1、1、1、3 mg·mL-1的对照品溶液;依次吸取上述各对照品溶液1、0.2、1、1、2 mL,混合均匀,即得混合对照品溶液。
参照2020年版《中华人民共和国药典》四部0502薄层色谱法,分别吸取“2.1.2”项下洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A的对照品溶液各1 μL,Z-藁本内酯对照品溶液、混合对照品溶液、供试品溶液3 μL,点于同一GF254薄层板上,以正己烷-乙酸乙酯-冰醋酸(10:5:0.5)为展开剂,饱和10 min,展开8~9 cm,取出,晾干,分别置于紫外光254 nm和365 nm下检视,结果如图1所示。样品色谱中,在与对照品色谱相对应的位置上显示相同颜色的荧光斑点。54批药材薄层图谱见图2
取S54号茶芎药材粉末(过4号筛)约0.5 g,精密称定,加入甲醇25 mL,称量,超声(功率250 W,频率40 kHz)处理45 min,放冷后,再称量,用甲醇补足减失量,5 000 r·min-1离心5 min,0.45 μm微孔滤膜滤过,取续滤液,即得。
分别取绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸的对照品,精密称定,置于10 mL量瓶中,加甲醇制成质量浓度分别含3.15、2.13、25.80、19.03、7.04、78.38、405.83、32.69、252.75、421.12、25.69、17.27、188.42 μg·mL-1的混合对照品溶液。
采用Acutfex PA-C18 (250 mm×4.6 mm,5 μm)色谱柱,以0.1%磷酸水溶液(A)-乙腈(B)为流动相,梯度洗脱(0~35 min,10%B→30%B;35~40 min,30%B→50%B;40~65 min,50%B;65~70 min,50%B→80%B;70~80 min,80%B→90%B;80~81 min,90%B→100%B;81~90 min,100%B),体积流量1.0 mL·min-1,柱温30 ℃,进样量10 μL,检测波长210 nm和285 nm。
取茶芎药材(编号S54),按照“2.2.1”项下方法制备供试品溶液,按照“2.2.3”项下色谱条件连续进样6次,检测,记录色谱图。各成分中Z-藁本内酯的响应值最高,峰形及分离度良好,故以Z-藁本内酯的保留时间和峰面积为参照,计算各共有峰的相对保留时间及相对峰面积[8]。相对保留时间的RSD均<0.13%,相对峰面积的RSD均<3.5%,表明仪器精密度良好。
取茶芎药材(编号S54),按照“2.2.1”项下方法制备供试品溶液,分别于溶液制备后0、2、4、8、16、24 h,按照“2.2.3”项下色谱条件进样检测,记录色谱图,以Z-藁本内酯的保留时间和峰面积为参照,计算各共有峰的相对保留时间及相对峰面积。相对保留时间的RSD均<0.53%,相对峰面积的RSD均<2.7%,表明茶芎供试品溶液稳定性良好。
取同一批茶芎药材(编号S54),按照“2.2.1”项下方法平行制备6份供试品溶液,按照“2.2.3”项下色谱条件进样检测,记录色谱图,以Z-藁本内酯的保留时间和峰面积为参照,计算各共有峰的相对保留时间及相对峰面积。相对保留时间的RSD均<0.063%,相对峰面积的RSD均<3.8%,表明该试验重复性良好。
将54批次(S1~S54号)的茶芎药材,按照“2.2.1”项下方法制备供试品溶液,按照“2.2.3”项下色谱条件进样检测,记录色谱图,将色谱图数据(AIA文件)导入《中药色谱指纹图谱相似度评价系统(2012版)》软件中,分别输出各批次色谱图(TXT文件)[9],0~55 min选择285 nm波长下的色谱图数据,55 min后选择210 nm波长下的色谱图数据,将整合后的54批次茶芎药材色谱图数据再导入软件,设S54号样品色谱图为参照图谱,时间窗宽度0.1 min,采用平均数法,经多点校正、峰匹配后得到54批次茶芎药材HPLC叠加指纹图谱及对照指纹图谱R,共确定了21个共有峰,如图3。通过文献查阅以及与对照品的紫外吸收光谱图和保留时间进行比对[10],共识别了13个峰,其中2号峰为绿原酸,3号峰为咖啡酸,5号峰为阿魏酸,8号峰为洋川芎内酯I,9号峰为洋川芎内酯H,12号峰为阿魏酸松柏酯,13号峰为洋川芎内酯A,14号峰为正丁基苯酞,16号峰为新蛇床内酯,17号峰为Z-藁本内酯,18号峰为丁烯基苯酞,20号峰为欧当归内酯A,21号峰为亚油酸。
将54批次茶芎药材的绝对峰面积数据导入《中药色谱指纹图谱相似度评价系统(2012版)》软件中,计算各批次茶芎药材指纹图谱与所生成的对照指纹图谱R的相似度[11],54批茶芎样品相似度在0.872~0.999,除S47外,其他批次药材相似度均>0.9,表明不同批次茶芎药材相似度较高,质量相对稳定。
将S1~S54号茶芎药材的共有峰绝对峰面积导入SPSS 27.0数据统计软件,以21个共有峰为变量,采用组间聚类方法,选择平方欧氏距离,以Z得分标准化后进行系统聚类分析,结果见图4所示。当欧式距离为5时,大致可以分为6类,S45、S47、S48、S53号药材为一类,S1~S7、S9号药材为一类,S36、S40~S42、S44、S46、S49、S51、S54号药材为一类,S50号药材为一类,S52号药材为一类,其他药材为一类。结合采集信息来看,茶芎药材按照采集时间和产地呈现差异性。
将S1~S54号茶芎药材的共有峰绝对峰面积导入SPSS 27.0数据统计软件,进行主成分分析,以特征值>1为标准,共得到4个主成分,累计方差贡献率达到82.449%,详见表2。由主成分矩阵(表3)可知各个共有峰对主成分的贡献率,峰2、6、7、11、13、15~17、19对主成分1贡献较大,峰3、4、8、9、18、20、21对主成分2贡献较大,峰1、5、10、12对主成分3贡献较大,峰14对主成分4贡献较大[12]
以S1~S54号茶芎药材中共有峰的绝对峰面积为变量,导入SIMCA统计软件,进行正交最小二乘法判别分析(OPLS-DA),建立OPLS-DA模型,见图5,该模型R2X=0.922,R2Y=0.642,Q2=0.513,均>0.5,说明该模型稳定可靠[13]。为防止该模型出现假阳性结果,对其进行200次交叉验证,结果显示R2截距为0.11,Q2截距为-0.266,说明该模型可靠,未出现过拟合现象[14],见图6。对OPLS-DA得分图进行分析,54批茶芎药材根据采集时间和海拔大致分为4类,别是低海拔(300 m)、中海拔(370~400 m)、高海拔(470 m以上)以及非当季茶芎,该结果与SPSS聚类结果较为相似。以变量重要性投影(variable importance in the projection,VIP)>1为标准,筛选出7个差异性标志物,见图7,它们的色谱峰分别是峰17、峰13、峰12、峰5、峰6、峰16、峰2,该7个成分可能是影响茶芎质量的主要化学成分。

精密吸取“2.4.2”项下混合对照品溶液0.4、1、2、5、10 mL,分别置于10 mL量瓶中,加甲醇稀释得到不同质量浓度的混合对照品溶液,按“2.4.3”项下色谱条件进样检测,以待测成分的质量浓度(X)为横坐标,峰面积(Y)为纵坐标,进行线性回归,得线性回归方程。结果表明各对照品线性关系良好,r均在0.999以上,结果见表4
取“2.4.4”项下稀释5倍的混合对照品溶液,按照“2.4.3”项下色谱条件连续进样6次,记录峰面积,计算各成分峰面积的RSD。结果绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸峰面积的RSD分别为1.6%、1.9%、1.1%、1.0%、1.2%、1.1%、0.85%、2.0%、0.86%、0.82%、1.6%、0.84%、1.9%,表明仪器精密度良好。
取茶芎药材(编号S54),按照“2.4.1”项下方法制备供试品溶液,分别于室温下放置0、2、4、8、12、24、48 h,按照“2.4.3”项下色谱条件进样检测,记录峰面积,计算各成分峰面积的RSD。结果绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸峰面积的RSD分别为3.6%、2.6%、2.5%、2.2%、2.4%、1.9%、2.0%、2.8%、1.9%、1.9%、2.7%、2.0%、3.9%,表明供试品溶液在48 h内稳定性良好。
取茶芎药材(编号S54),按照“2.4.1”项下方法平行制备6份供试品溶液,按照“2.4.3”项下色谱条件进样检测,记录峰面积,分别计算各成分的含量及其RSD。结果表明绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸平均含量分别为0.04、0.03、0.27、0.26、0.08、0.93、4.83、0.31、1.61、4.86、0.35、0.26、2.25 mg·g-1,RSD分别为2.9%、3.1%、3.5%、2.1%、1.6%、3.7%、1.7%、1.9%、1.7%、1.7%、2.4%、2.0%、3.2%,表明试验重复性良好。
取已知含量的茶芎药材(编号S54)粉末约0.25 g,精密称定,共6份,分别加入与样品中待测成分含有量相当的各对照品溶液,按照“2.4.1”项下方法制备供试溶液,按照“2.4.3”项下色谱条件进样检测,记录峰面积,计算各成分的加样回收率。结果显示绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸的平均回收率分别为93.5%、93.9%、101.0%、103.5%、94.7%、96.8%、102.1%、95.1%、97.9%、94.1%、94.3%、104.0%、97.2%,其RSD分别为2.4%、1.5%、2.9%、2.4%、2.3%、3.3%、2.9%、1.8%、2.9%、3.0%、2.6%、1.7%、2.6%,表明该实验方法良好。
将54批次茶芎药材按照“2.4.1”项下方法制备供试品溶液,按照“2.4.3”项下色谱条件进样检测,记录峰面积,根据线性回归方程计算各成分含量,见表5
为建立茶芎的薄层色谱鉴别方法,分别考察了甲醇、乙酸乙酯、乙醚、正己烷4种不同提取溶剂的提取效果,结果表明甲醇提取的样品斑点更加清晰。同时也考察了甲苯-冰醋酸-甲醇、石油醚-乙酸乙酯、正己烷-乙酸乙酯、正己烷-乙酸乙酯-甲酸、正己烷-乙酸乙酯-冰醋酸等不同类型及比例的展开剂,结果表明当展开剂为正己烷-乙酸乙酯-冰醋酸(10:5:0.5)时展开效果最好。此外,也对温度(4、25 ℃)、湿度(18%、58%、88%)、点样量(1、2、3、4、5 μL)等进行了考察,确定建立了茶芎中洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯的薄层色谱鉴别方法。
为建立茶芎药材的指纹图谱及13个成分的含量测定方法,考察了甲醇-水、乙腈-水、甲醇-0.1%磷酸水、乙腈-0.1%磷酸水等不同流动相体系,从出峰数量、峰分离度、基线、拖尾因子等因素判断,最终选择乙腈-0.1%磷酸水为流动相体系;因13个成分在单一检测波长下不能同时出峰,根据最大吸收波长大致可分为2类,故选择285 nm和210 nm 2个波长同时检测;通过对洗脱梯度的优化,确定了分离度、峰形良好的梯度洗脱程序;同时考察了茶芎药材的提取溶剂(水、25%甲醇、50%甲醇、75%甲醇、甲醇)、提取方式(超声提取、回流提取)、提取时间(15、30、45、60 min),以Z-藁本内酯的含量为评价指标,确定了茶芎药材的最佳处理方式为用甲醇超声提取45 min。建立了茶芎药材的指纹图谱,共发现了21个共有峰,通过与对照品进行比对,共识别了13个峰;相似度评价结果显示除个别批次药材外,其余相似度均>0.9,表明不同批次茶芎药材具有高度相似性。然后建立了13个成分的含量测定方法。
据前期实地考察得知,茶芎一般于冬至前后种植,次年小暑大暑前后采挖,通过采用SPSS和SIMCA软件对检测结果进行分析,54批茶芎药材根据采集时间和地点大致可以分为4类,分别是低海拔(300 m)、中海拔(370~400 m)、高海拔(470 m以上)以及非当季采摘茶芎。通过OPLS-DA筛选出峰17(Z-藁本内酯)、峰13(洋川芎内酯A)、峰12(阿魏酸松柏酯)、峰5(阿魏酸)、峰6、峰16(新蛇床内酯)、峰2(绿原酸)这7个化学成分可能是影响茶芎药材质量的主要成分。文献报道苯酞类化合物是茶芎的主要化学成分,包括Z-藁本内酯、正丁基苯酞、丁烯基苯酞、新蛇床内酯等。有研究表明[15-16],苯酞类化合物在抗血栓、改善局部脑缺血引起的脑水肿、抗惊厥、治疗脑卒中等方面有良好疗效。此外,阿魏酸、绿原酸等有机酸类化合物在茶芎中同样有较高的含量,研究表明该类化合物是活血化瘀的主要有效成分[17],因此本研究最终确定了以洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯作为茶芎的薄层色谱鉴别的指标性成分,以绿原酸、咖啡酸、阿魏酸、洋川芎内酯I、洋川芎内酯H、阿魏酸松柏酯、洋川芎内酯A、正丁基苯酞、新蛇床内酯、Z-藁本内酯、丁烯基苯酞、欧当归内酯A、亚油酸作为茶芎药材含量测定的指标性成分。
本实验建立了茶芎中洋川芎内酯I、东莨菪内酯、阿魏酸、欧当归内酯A、Z-藁本内酯5个成分的薄层色谱鉴别方法,建立了茶芎的指纹图谱及13个成分的含量测定方法,方法稳定可靠,可用于茶芎药材质量评价和资源开发利用。
