Objective:

To establish an HPLC method for determination of related substances and purity in pregabalin API.

Methods:

The analytical column was an Inertsil ODS-3 (150 mm×4.0 mm,3 μm),the mobile phase A was buffer (7.05 g ammonium dihydrogen phosphate and 1.45 g diammonium hydrogen phosphate dissolved in 1 000 mL water)-methanol-acetonitrile(900:80:20) and the mobile phase B was acetonitrile,the whole carried out by gradient elution at a flow rate of 0.8 mL·min-1,the detection wavelength was set at 210 nm,the column temperature was 30 ℃ and the injection volume was 50 μL.

Results:

Pregabalin was separated completely from the impurities and degradation products (the resolution>2.0). The test solution was stable for at least 48 h. The LOQs of other impurities (verification was performed by using pregabalin),lactam,4-alkene pregabalin,5-alkene pregabalin,trimer,3-isobutylglutaric acid (PGB-3),3-isobutyl glutaric acid monoamide (PGB-5),R-phenethylamine,4-isobutyl-2,6-piperidinedione (PGB-5B),monoamide phenylethylamine (PGB-5C) and phenylethylamine adipate (PGB-5D) were 0.05%,0.01%,0.01%,0.03%,0.01%,0.05%,0.03%,0.01%,0.01%,0.01% and 0.01%. The linear correlation coefficients of other impurities (verification was performed by using pregabalin),lactam,4-alkene pregabalin,5-alkene pregabalin,trimer,PGB-3 and PGB-5 were all more than 0.99. The linear ranges were LOQ-150% of impurities’ specification,respectively. The average recoveries (n=9) of lactam,4-alkene pregabalin,5-alkene pregabalin,trimer,PGB-3 and PGB-5 were 100.6% (RSD=0.56%),100.2% (RSD=0.38%),100.5% (RSD=0.46%),101.1% (RSD=1.1%),100.0% (RSD=0.63%),100.0% (RSD=0.54%). The repeatability and intermediate precision completely met the requirements. The impurities contents in three batches of pregabalin API 6 months accelerate and 60 months long-term stability test completely met the requirements,respectively.

Conclusion:

This method is simple,rapid,sensitive and specific to be used for the determination of related substances and purity in phenethylamine process pregabalin API.

目的:

建立HPLC法检测苯乙胺工艺普瑞巴林原料药中的有关物质及纯度。

方法:

普瑞巴林峰与各杂质及降解产物峰能够完全分离(分离度>2.0),供试品溶液在48 h内稳定性良好;其他单杂(用普瑞巴林进行验证)、内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、3-异丁基戊二酸(PGB-3)、3-异丁基戊二酸单酰胺(PGB-5)、R-苯乙胺、4-异丁基-2,6-哌啶二酮(PGB-5B)、单酰胺苯乙胺(PGB-5C)、二酸苯乙胺(PGB-5D)的定量限分别为0.05%、0.01%、0.01%、0.03%、0.01%、0.05%、0.03%、0.01%、0.01%、0.01%和0.01%;其他单杂(用普瑞巴林进行验证)、内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5线性相关系数均>0.99,范围为LOQ~各杂质指标150%;内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5平均回收率(n=9)分别为100.6%(RSD=0.56%)、100.2%(RSD=0.38%)、100.5%(RSD=0.46%)、101.1%(RSD=1.1%)、100.0%(RSD=0.63%)、100.0%(RSD=0.54%);重复性和中间精密度符合规定。经检测,3批普瑞巴林原料药6个月加速和60个月长期稳定性各个杂质检测结果均符合质量标准。

结论:

该方法简便快速,灵敏度高,专属性强,可用于苯乙胺工艺普瑞巴林原料药中有关物质及纯度的检测。

目的:

建立HPLC法检测苯乙胺工艺普瑞巴林原料药中的有关物质及纯度。

方法:

采用Inertsil ODS-3(150 mm×4.0 mm,3 μm)色谱柱,以缓冲液(7.05 g磷酸二氢铵和1.45 g磷酸氢二铵溶于1 000 mL水中)-甲醇-乙腈(900:80:20)为流动相A,乙腈为流动相B,进行梯度洗脱,流速0.8 mL·min-1,检测波长210 nm,柱温30 ℃,进样量50 μL。

结果:

普瑞巴林峰与各杂质及降解产物峰能够完全分离(分离度>2.0),供试品溶液在48 h内稳定性良好;其他单杂(用普瑞巴林进行验证)、内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、3-异丁基戊二酸(PGB-3)、3-异丁基戊二酸单酰胺(PGB-5)、R-苯乙胺、4-异丁基-2,6-哌啶二酮(PGB-5B)、单酰胺苯乙胺(PGB-5C)、二酸苯乙胺(PGB-5D)的定量限分别为0.05%、0.01%、0.01%、0.03%、0.01%、0.05%、0.03%、0.01%、0.01%、0.01%和0.01%;其他单杂(用普瑞巴林进行验证)、内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5线性相关系数均>0.99,范围为LOQ~各杂质指标150%;内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5平均回收率(n=9)分别为100.6%(RSD=0.56%)、100.2%(RSD=0.38%)、100.5%(RSD=0.46%)、101.1%(RSD=1.1%)、100.0%(RSD=0.63%)、100.0%(RSD=0.54%);重复性和中间精密度符合规定。经检测,3批普瑞巴林原料药6个月加速和60个月长期稳定性各个杂质检测结果均符合质量标准。

结论:

该方法简便快速,灵敏度高,专属性强,可用于苯乙胺工艺普瑞巴林原料药中有关物质及纯度的检测。

Objective:

To establish an HPLC method for determination of related substances and purity in pregabalin API.

Methods:

The analytical column was an Inertsil ODS-3 (150 mm×4.0 mm,3 μm),the mobile phase A was buffer (7.05 g ammonium dihydrogen phosphate and 1.45 g diammonium hydrogen phosphate dissolved in 1 000 mL water)-methanol-acetonitrile(900:80:20) and the mobile phase B was acetonitrile,the whole carried out by gradient elution at a flow rate of 0.8 mL·min-1,the detection wavelength was set at 210 nm,the column temperature was 30 ℃ and the injection volume was 50 μL.

Results:

Pregabalin was separated completely from the impurities and degradation products (the resolution>2.0). The test solution was stable for at least 48 h. The LOQs of other impurities (verification was performed by using pregabalin),lactam,4-alkene pregabalin,5-alkene pregabalin,trimer,3-isobutylglutaric acid (PGB-3),3-isobutyl glutaric acid monoamide (PGB-5),R-phenethylamine,4-isobutyl-2,6-piperidinedione (PGB-5B),monoamide phenylethylamine (PGB-5C) and phenylethylamine adipate (PGB-5D) were 0.05%,0.01%,0.01%,0.03%,0.01%,0.05%,0.03%,0.01%,0.01%,0.01% and 0.01%. The linear correlation coefficients of other impurities (verification was performed by using pregabalin),lactam,4-alkene pregabalin,5-alkene pregabalin,trimer,PGB-3 and PGB-5 were all more than 0.99. The linear ranges were LOQ-150% of impurities’ specification,respectively. The average recoveries (n=9) of lactam,4-alkene pregabalin,5-alkene pregabalin,trimer,PGB-3 and PGB-5 were 100.6% (RSD=0.56%),100.2% (RSD=0.38%),100.5% (RSD=0.46%),101.1% (RSD=1.1%),100.0% (RSD=0.63%),100.0% (RSD=0.54%). The repeatability and intermediate precision completely met the requirements. The impurities contents in three batches of pregabalin API 6 months accelerate and 60 months long-term stability test completely met the requirements,respectively.

Conclusion:

This method is simple,rapid,sensitive and specific to be used for the determination of related substances and purity in phenethylamine process pregabalin API.

普瑞巴林(pregabalin)是一种γ-氨基丁酸(GABA)受体激动剂,能阻断电压依赖性钙通道,减少神经递质的释放,临床常用于治疗带状疱疹神经痛、纤维肌痛综合征等神经病理性疼痛,以及成年患者局部不全性癫痫发作,广泛性焦虑症等疾病[1-6]。普瑞巴林是全球最畅销的镇痛药,FDA批准用于治疗与糖尿病周围神经病变神经痛、带状疱疹后神经痛、脊髓损伤相关神经痛、纤维肌痛相关神经痛以及特定成人癫痫患者部分性发作[7]
目前《中华人民共和国药典》尚无普瑞巴林原料药的质量控制方法,USP[8]、EP[9]中收载了普瑞巴林原料药的质量标准。然而,USP、EP中的有关物质分析方法均不能有效地检测苯乙胺工艺产生的副产物、降解杂质等。用高效液相色谱法测定普瑞巴林中有关物质已有文献报道,但并未列全降解产物及可能存在的杂质[10-15]。基于产品质量控制的需求,迫切需要开发有关物质检测方法。
质量研究中涉及的物质有活性成分普瑞巴林,工艺杂质4-烯普瑞巴林、5-烯普瑞巴林、三聚物、3-异丁基戊二酸(PGB-3)、4-异丁基-2,6-哌啶二酮(PGB-5B)、单酰胺苯乙胺(PGB-5C)、二酸苯乙胺(PGB-5D)、R-普瑞巴林、3-异丁基戊二酸单酰胺(PGB-5,中间体),R-苯乙胺(拆分剂),内酰胺(降解杂质)等。主要杂质信息见表1
苯乙胺工艺中各个杂质的指标:内酰胺,杂质含量≤0.15%;4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5、R-苯乙胺、PGB-5B、PGB-5C、PGB-5D,杂质含量均≤0.10%;R-普瑞巴林,杂质含量≤0.15%,可直接引用USP分析方法控制。指标制定的依据来源于ICH Q3A[16],界定阈值≤0.15%,鉴定阈值≤0.10%。
内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5在工艺中残留的可能性较大,定为已知杂质,进行全套方法学验证;R-苯乙胺、PGB-5B、PGB-5C、PGB-5D在工艺中去除效果较好,不定为已知杂质,进行限度验证,验证内容包括专属性、LOD、LOQ。
本文建立了苯乙胺工艺普瑞巴林原料药有关物质高效液相色谱法,并进行方法学验证试验,通过试验结论判定该方法用于苯乙胺工艺原料药普瑞巴林的质量控制。
Agilent 1260高效液相色谱仪(Agilent公司);XP205十万分之一电子天平(Mettler Toledo公司);XP26百万分之一电子天平(Mettler Toledo公司);S400 pH计(Mettler Toledo公司);磷酸二氢铵(ACS,百灵威科技有限公司);磷酸氢二铵(色谱纯,ACROS公司);乙腈(色谱纯,MERCK公司);甲醇(色谱纯,MERCK公司);超纯水(MiLLi-Q纯水仪制备)。
对照品普瑞巴林(批号1.0,含量99.8%,EDQM),内酰胺(批号1-NMA-39-2,含量99.92%,TRC),4-烯普瑞巴林(3-氨基甲基-5-甲基-4-烯酸)、5-烯普瑞巴林(3-氨基甲基-5-甲基-5-烯酸)、三聚物、PGB-3、PGB-5、R-苯乙胺、PGB-5B、PGB-5C、PGB-5D(含量分别为99.4%、96.2%、96.8%、98.8%、99.4%、99.4%、96.2%、96.8%、98.8%、99.4%,均来自浙江华海药业股份有限公司)。
普瑞巴林原料药(批号D5248-16-053、D5248-17-086、087、088,浙江华海药业股份有限公司)。
采用Inertsil ODS-3(150 mm×4.0 mm,3 μm)色谱柱,以缓冲液(磷酸二氢铵7.05 g和磷酸氢二铵1.45 g溶于1 000 mL水中)-甲醇-乙腈(900:80:20)为流动相A,乙腈为流动相B,按表2的条件进行梯度洗脱,流速0.8 mL·min-1,检测波长210 nm,柱温30 ℃,进样量50 μL。
精密称取内酰胺对照品30 mg,置于20 mL量瓶中,加乙腈溶解并稀释至刻度,摇匀,制成每1 mL含1.5 mg的对照品储备液。精密称取R-苯乙胺对照品20 mg,置于20 mL量瓶中,加乙腈20 mL溶解并稀释至刻度,摇匀,再移取1 mL至100 mL量瓶中,加流动相A稀释至刻度,摇匀,制成每1 mL含0.01 mg的对照品储备液。精密称取4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5、PGB-5B、PGB-5C、PGB-5D的对照品各20 mg,分别置于20 mL量瓶中,加乙腈溶解并稀释至刻度,摇匀,制成每1 mL含1 mg的各个杂质对照品储备液。
精密称取普瑞巴林对照品100 mg,置于10 mL量瓶中,以流动相A溶解并稀释至刻度,摇匀,制成每1 mL含10 mg普瑞巴林的溶液,即得。
精密称取普瑞巴林对照品10 mg,置于100 mL量瓶中,以流动相A溶解并稀释至刻度,摇匀,再精密量取1 mL到10 mL量瓶中,用流动相A稀释至刻度,摇匀,制成每1 mL含10 μg普瑞巴林的溶液,即得。
精密称取普瑞巴林原料药100 mg,置于10 mL量瓶中,以流动相A溶解并稀释至刻度,摇匀,制成每1 mL含10 mg普瑞巴林的溶液,即得。
精密称取内酰胺对照品20 mg,置于20 mL量瓶中,加乙腈溶解并稀释至刻度,摇匀,制成每1 mL含1 mg的溶液,即得。
精密称取普瑞巴林对照品20 mg,置于20 mL量瓶中,用流动相A溶解并稀释至刻度,摇匀,再精密量取适量,以流动相A稀释,制成每1 mL含0.05 mg普瑞巴林的溶液,即得。
精密量取“2.2.1”项下内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5的杂质对照品储备液各5 mL,置于同一100 mL量瓶中,加流动相A稀释至刻度,摇匀,再精密量取2 mL至10 mL量瓶中,精密加入普瑞巴林对照品100 mg及R-苯乙胺杂质对照品储备液1 mL,加流动相A溶解并稀释至刻度,摇匀,制成每1 mL含普瑞巴林10 mg、内酰胺15 μg、4-烯普瑞巴林10 μg、5-烯普瑞巴林10 μg、三聚物10 μg、PGB-3 10 μg、PGB-5 10 μg、R-苯乙胺1 μg的混合溶液,即得。
精密量取“2.2.1”项下R-苯乙胺、PGB-5B、PGB-5C、PGB-5D的杂质储备液各5 mL,置于100 mL量瓶中,加流动相A稀释至刻度,摇匀,再精密量取2 mL至10 mL量瓶中,精密加入普瑞巴林对照品100 mg,加流动相A溶解并稀释至刻度,摇匀,制成每1 mL含普瑞巴林10 mg、R-苯乙胺10 μg、PGB-5B 10 μg、PGB-5C 10 μg、PGB-5D 10 μg的混合溶液,即得。
分别称取普瑞巴林样品1 000 mg,置于100 mL量瓶中,通过酸降解(1 mol·L-1盐酸溶液,60 ℃,20 mL,2 h)、碱降解(1 mol·L-1氢氧化钠溶液,60 ℃,20 mL,2 h)、氧化降解(10%过氧化氢溶液,室温,20 mL,2 h)、金属降解(500 mmol·L-1硫酸铜溶液,室温,20 mL,8 h),酸碱降解后进行中和,中和溶液分别为酸降解所用盐酸溶液及碱降解所用氢氧化钠溶液,用流动相A稀释至刻度,摇匀,即得酸降解溶液、碱降解溶液、氧化降解溶液、金属降解溶液。将固体样品裸露于高温高湿(60 ℃,RH 75%,7 d)、高温低湿(60 ℃,RH 30%,7 d)、光降解(证实性:强白光总照度≥1.2×106 lx·h,紫外光能量≥200 W·h·m-2;强制性为3倍证实性)环境中进行降解;分别称取降解产物100 mg,置于10 mL量瓶中,用流动相A溶解并稀释至刻度,摇匀,即得高温高湿降解溶液、高温低湿降解溶液、光降解溶液。取上述各降解溶液进样分析,由研究结果可知,最容易由普瑞巴林降解产生的杂质为内酰胺,而在碱破坏条件下降解程度最大,降解产生的内酰胺量最多;通过改变碱降解的时间,可得降解产生的内酰胺增长曲线,试验数据证实内酰胺在增长到一定量后,会达到上限,与主成分普瑞巴林处于平衡状态,详见图12
结合工艺分析可得,苯乙胺工艺可能产生的工艺杂质为4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5B、PGB-5C、PGB-5D、内酰胺(降解杂质)、R-苯乙胺(拆分剂)、PGB-5(中间体)。现对以上杂质进行定位研究。
在“2.1”项色谱条件下,精密量取选择性溶液1和选择性溶液2各50 μL分别注入高效液相色谱仪,记录图谱。普瑞巴林与各杂质分离度均>2.0。空白溶液(流动相A)、选择性溶液1和选择性溶液2图谱见图3
精密量取各杂质对照品储备液、未知杂质储备液、内酰胺LOQ储备液适量,分别用流动相A进行逐级稀释,初始配制浓度为0.05%,以0.04%、0.03%、0.02%、0.01%浓度往下稀释,LOQ最小不低于0.01%。按“2.1”项下色谱条件进样分析,以信噪比约为10时普瑞巴林及各杂质的浓度作为定量限,以信噪比约为3时普瑞巴林及各杂质的浓度作为检测限,结果见表3
精密量取内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5的杂质对照品储备液各5 mL,置于同一100 mL量瓶中,加流动相A稀释至刻度,即得母液。精密量取母液3 mL,置于25 mL量瓶中,加入未知杂质储备液3 mL,用流动相A稀释至刻度,摇匀,即得60%指标浓度线性溶液;精密量取母液4 mL,置于25 mL量瓶中,加入未知杂质储备液4 mL,用流动相A稀释至刻度,摇匀,即得80%指标浓度线性溶液;精密量取母液2 mL,置于10 mL量瓶中,加入未知杂质储备液2 mL,用流动相A稀释至刻度,摇匀,即得100%指标浓度线性溶液;精密量取母液6 mL,置于25 mL量瓶中,加入未知杂质储备液6 mL,用流动相A稀释至刻度,摇匀,即得120%指标浓度线性溶液;精密量取母液3 mL,置于10 mL量瓶中,加入未知杂质储备液3 mL,用流动相A稀释至刻度,摇匀,即得150%指标浓度线性溶液。分别精密量取4-烯普瑞巴林杂质对照品储备液1 mL、5-烯普瑞巴林杂质对照品储备液3 mL、三聚物杂质对照品储备液1 mL、PGB-3杂质对照品储备液5、PGB-5杂质对照品储备液3 mL以及未知杂质储备液5 mL和内酰胺LOQ储备液1 mL,置于同一100 mL量瓶中,加流动相A稀释至刻度,再精密量取1 mL至10 mL量瓶中,用流动相A稀释至刻度,摇匀,得各杂质LOQ浓度线性溶液。
各线性浓度溶液,按“2.1”项下色谱条件进样,记录色谱图。以普瑞巴林、各杂质浓度(X)为横坐标,峰面积(Y)为纵坐标,绘制标准曲线,回归方程及线性范围见表4,结果说明线性关系良好。
精密量取内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5的杂质对照品储备液各5 mL,置于同一100 mL量瓶中,加流动相A稀释至刻度,摇匀,再精密量取2 mL至10 mL量瓶中,精密加入样品(D5248-16-053)100 mg,加流动相A溶解并稀释至刻度,摇匀,制成每1 mL含10 mg的普瑞巴林以及其他各杂质均为指标的100%溶液(每1 mL含15 μg内酰胺,每1 mL含10 μg其他各杂质),同法配制6份,按“2.1”项下色谱条件分别各进样1次。结果显示,内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5峰面积的RSD分别为0.34%、0.28%、0.34%、0.70%、0.45%、0.47%。更换人员、色谱柱(部件号:1A7144718、6AI81910)、仪器(Agilent公司:Agilent1260、Agilent1260 InfinityII)、试验日期,同法配制中间精密度溶液6份,按“2.1”项下色谱条件分别各进样1次。结果显示,内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5峰面积的RSD(n=12)分别为0.46%、0.57%、0.40%、1.8%、0.54%、0.40%,表明方法精密度好。
取参比溶液以及供试品溶液,室温下分别于0、24、48 h按“2.1”项下色谱条件进样。结果48 h和0 h参比溶液中普瑞巴林峰面积比为1.03,供试品溶液中普瑞巴林、内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3和PGB-5的结果无明显变化,表明参比溶液及供试品溶液在48 h内稳定性良好。
精密量取内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5的杂质对照品储备液各5 mL,置于同一100 mL量瓶中,加流动相A稀释至刻度,摇匀,即得母液。精密量取母液4 mL至25 mL量瓶中,精密加入普瑞巴林样品(D5248-16-053)250 mg,加流动相A溶解并稀至刻度,摇匀,制成每1 mL含普瑞巴林10 mg、其他各杂质浓度分别为指标的80%的溶液(每1 mL含12 μg内酰胺,每1 mL含8 μg其他各杂质);精密量取母液2 mL至10 mL量瓶中,精密加入普瑞巴林供试品(D5248-16-053)100 mg,加流动相A溶解并稀释至刻度,摇匀,制成每1 mL含普瑞巴林10 mg、其他各杂质浓度分别为指标的100%的溶液(每1 mL含15 μg内酰胺,每1 mL含10 μg其他各杂质);精密量取母液6 mL至25 mL量瓶中,精密加入普瑞巴林样品(D5248-16-053)250 mg,加流动相A溶解并稀释至刻度,摇匀,制成每1 mL含普瑞巴林10 mg、其他各杂质浓度分别为指标的120%的溶液(每1 mL含18 μg内酰胺,每1 mL含12 μg其他各杂质)。上述每个浓度点的溶液同法配制3份,共9份。按“2.1”项下色谱条件进样测定并记录色谱图,计算试验结果,内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5的平均回收率(n=9)分别为100.6%(RSD=0.56%)、100.2%(RSD=0.38%)、100.5%(RSD=0.46%)、101.1%(RSD=1.1%)、100.0%(RSD=0.63%)、100.0%(RSD=0.54%),说明方法准确度高。
更换不同批号的色谱柱(部件号:1A7144718、6AI81910)、不同编号的高效液相色谱仪(Agilent公司:Agilent1260、Agilent1260 InfinityII),柱温30±2 ℃、流速0.8±0.1 mL·min-1,在“2.1”项下的色谱条件下检测选择性溶液1,结果普瑞巴林、内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5的分离情况及峰面积无明显差别,说明方法耐用性好。
对不同批次普瑞巴林原料药的加速稳定性样品(放置条件:温度(40±2)℃,相对湿度75%±5%;放置时间:0 d、1月、2月、3月、6月)以及长期稳定性样品(放置条件:温度(25±2)℃,相对湿度60%±5%;放置时间:0 d及3、6、9、12、18、24、36、48、60月),按照“2.2.4”项下方法制备供试品溶液,在“2.1”项下色谱条件进行测定。检测结果见表5,稳定性样品各个杂质结果均未见明显增长。
按照本文建立的方法,对D5248-17-086、087、088 3批样品进行检测,结果显示,各个杂质结果均符合质量标准,数据见表5稳定性试验结果0 d。
各强制降解条件下产生的杂质主要为内酰胺;加速样品检验数据表明普瑞巴林原料药各个杂质结果均未见明显增长;此外,通过USP衍生化方式,检测各批次加速试验样品中对映异构体含量,结果均为未检出。加速稳定性及长期稳定性数据表明,本品性质稳定,杂质限度符合要求。
通过对USP、EP中普瑞巴林有关物质检测方法进行试验比较,结果显示,USP、EP方法均不适用于苯乙胺工艺普瑞巴林原料药有关物质检测。其中USP方法下工艺杂质PGB-3出峰时间受主峰干扰,工艺杂质三聚物无法在的梯度洗脱时间内出峰,内酰胺受溶剂峰干扰;EP方法下中间体PGB-5与主峰无法分离,工艺杂质三聚物保留时间较长(100 min以上)等。故舍弃2个药典的方法,重新开发适用于苯乙胺工艺的普瑞巴林原料药有关物质方法。
参考USP中有关物质分析方法给出的pH,调整流动相比例。通过理论分析可知,将普瑞巴林及内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5、R-苯乙胺、PGB-5B、PGB-5C、PGB-5D 10个杂质在短时间内全部洗脱并有效分离,需要用到梯度洗脱的方法。由于10个杂质极性跨度较大,在洗脱初始阶段,采取缓冲液加甲醇和乙腈的等度方式,保留并分离强极性杂质,后与乙腈进行梯度洗脱,保留并分离弱极性杂质。通过摸索,最终定流动相A为磷酸盐缓冲液-乙腈-甲醇(900:20:80),流动相B为乙腈,进行梯度洗脱。
参考USP中有关物质分析方法给出的pH,在进行杂质加样回收过程中,发现PGB-3杂质在单独配制与加入大浓度普瑞巴林时,出峰时间相差较大。通过缓冲液pH摸索可得,在缓冲液偏酸性条件下,PGB-3的出峰不再受到高浓度普瑞巴林的影响,而在此条件下R-苯乙胺容易产生溶剂效应,出现分叉现象。再次通过缓冲液pH摸索后,最终将缓冲液的pH定为6.0左右,在此条件下,各个杂质均能有效分离,且未出现峰飘及溶剂效应。
综合分析,苯乙胺工艺普瑞巴林原料药涉及降解杂质、副产物、拆分剂性质差别较大,色谱峰较多,建立能将所有物质有效分离的分析方法,是普瑞巴林原料药有关物质方法开发的关键;通过对不同型号色谱柱、流动相pH以及流动相比例等色谱条件的反复摸索,建立了普瑞巴林苯乙胺工艺有关物质分析方法,并通过酸、碱、氧、光照等破坏试验以及各个杂质的加标试验,验证了该分析方法的专属性,保证了该方法在原料药稳定性样品检测过程中的耐用性;本试验对于普瑞巴林工艺杂质进行了检测能力的确认(定量限、检测限),对工艺中去除效果较好的R-苯乙胺、PGB-5B、PGB-5C、PGB-5D进行限度验证,对于苯乙胺工艺各指标杂质内酰胺、4-烯普瑞巴林、5-烯普瑞巴林、三聚物、PGB-3、PGB-5进行了定量限、检测限、线性关系、回收率等一系列方法学验证,结果证明该分析方法精密度高,重复性好,稳定无干扰,可用于普瑞巴林原料药的质量控制。
