Objective:

To establish an ion chromatography method suitable for simultaneous determination of acid radical content in chemical reference substances containing methanesulfonic acid,hydrochloric acid,phosphoric acid,sulfuric acid and butadisulfonic acid.

Methods:

The chromatographic column of Metrosep A Supp 5 250/4.0 (250 mm×4.0 mm,5 μm) was used with 6.4 mmol·L-1 Na2CO3 and 2.0 mmol·L-1 NaHCO3 as the eluent with a flow rate of 0.7 mL·min-1. Column temperature was 30 ℃.

Results:

Methanesulfonate (19.1-71.7 μg·mL-1),chloride ion (2.6-25.8 μg·mL-1),phosphate (7.0-26.3 μg·mL-1),sulfate (6.8-51.0 μg·mL-1),and succinate (21.3-105.5 μg·mL-1) had good linear relationships within their respective ranges (r≥0.999 5). Precision (RSD≤2%),stability (RSD≤2%) and repeatability (RSD≤2%) were good. The average recovery rates (n=9) were 98%-102%. Calculated as salt formation,the content of mesylate in amlodipine mesylate was 23.69%,the content of hydrochloric acid in guanfacine hydrochloride was 12.66%,the content of phosphoric acid in reagliptin phosphate was 17.38%,the content of sulphate in salbutamol sulfate was 16.49%,and the content of butyldisulfonic acid in adenosine butyldisulfonic acid was 45.61%. The deviation between theoretical and measured values was less than 0.50%. The measured values were close to the theoretical values.

Conclusion:

The method is suitable for the screen and determination of acid radicals in chemical reference products containing methanesulfonic acid,hydrochloric acid,phosphoric acid,sulfuric acid and butadisulfonic acid.

目的:

建立适用于同时测定含甲磺酸、盐酸、磷酸、硫酸、丁二磺酸化学对照品中酸根含量的离子色谱法。

方法:

采用Metrosep A Supp 5 250/4.0(250 mm×4.0 mm,5 μm)色谱柱,以6.4 mmol·L-1碳酸钠溶液-2.0 mmol·L-1碳酸氢钠溶液为淋洗液,等度洗脱,流速0.7 mL·min-1,柱温30 ℃。

结果:

甲磺酸根(19.1~71.7 μg·mL-1)、氯离子(2.6~25.8 μg·mL-1)、磷酸根(7.0~26.3 μg·mL-1)、硫酸根(6.8~51.0 μg·mL-1)和丁二磺酸根(21.3~105.5 μg·mL-1)在各自范围内线性关系良好(r≥0.999 5);精密度(RSD≤2%)、稳定性(RSD≤2%)、重复性(RSD≤2%)良好;平均加样回收率(n=9)为98%~102%,以成盐形式计算,甲磺酸氨氯地平中甲磺酸含量为23.69%,盐酸胍法辛中盐酸含量为12.66%,磷酸瑞格列汀中磷酸含量为17.38%,硫酸沙丁胺醇中硫酸含量为16.49%,丁二磺酸腺苷蛋氨酸中丁二磺酸含量为45.61%,理论值与测定值偏差均<0.50%。

结论:

该方法适用性良好,可用于含甲磺酸、盐酸、磷酸、硫酸、丁二磺酸化学对照品中酸根的筛查及含量测定。

建立适用于同时测定含甲磺酸、盐酸、磷酸、硫酸、丁二磺酸化学对照品中酸根含量的离子色谱法。

方法:

采用Metrosep A Supp 5 250/4.0(250 mm×4.0 mm,5 μm)色谱柱,以6.4 mmol·L-1碳酸钠溶液-2.0 mmol·L-1碳酸氢钠溶液为淋洗液,等度洗脱,流速0.7 mL·min-1,柱温30 ℃。

结果:

甲磺酸根(19.1~71.7 μg·mL-1)、氯离子(2.6~25.8 μg·mL-1)、磷酸根(7.0~26.3 μg·mL-1)、硫酸根(6.8~51.0 μg·mL-1)和丁二磺酸根(21.3~105.5 μg·mL-1)在各自范围内线性关系良好(r≥0.999 5);精密度(RSD≤2%)、稳定性(RSD≤2%)、重复性(RSD≤2%)良好;平均加样回收率(n=9)为98%~102%,以成盐形式计算,甲磺酸氨氯地平中甲磺酸含量为23.69%,盐酸胍法辛中盐酸含量为12.66%,磷酸瑞格列汀中磷酸含量为17.38%,硫酸沙丁胺醇中硫酸含量为16.49%,丁二磺酸腺苷蛋氨酸中丁二磺酸含量为45.61%,理论值与测定值偏差均<0.50%。

结论:

该方法适用性良好,可用于含甲磺酸、盐酸、磷酸、硫酸、丁二磺酸化学对照品中酸根的筛查及含量测定。

Objective:

To establish an ion chromatography method suitable for simultaneous determination of acid radical content in chemical reference substances containing methanesulfonic acid,hydrochloric acid,phosphoric acid,sulfuric acid and butadisulfonic acid.

Methods:

The chromatographic column of Metrosep A Supp 5 250/4.0 (250 mm×4.0 mm,5 μm) was used with 6.4 mmol·L-1 Na2CO3 and 2.0 mmol·L-1 NaHCO3 as the eluent with a flow rate of 0.7 mL·min-1. Column temperature was 30 ℃.

Results:

Methanesulfonate (19.1-71.7 μg·mL-1),chloride ion (2.6-25.8 μg·mL-1),phosphate (7.0-26.3 μg·mL-1),sulfate (6.8-51.0 μg·mL-1),and succinate (21.3-105.5 μg·mL-1) had good linear relationships within their respective ranges (r≥0.999 5). Precision (RSD≤2%),stability (RSD≤2%) and repeatability (RSD≤2%) were good. The average recovery rates (n=9) were 98%-102%. Calculated as salt formation,the content of mesylate in amlodipine mesylate was 23.69%,the content of hydrochloric acid in guanfacine hydrochloride was 12.66%,the content of phosphoric acid in reagliptin phosphate was 17.38%,the content of sulphate in salbutamol sulfate was 16.49%,and the content of butyldisulfonic acid in adenosine butyldisulfonic acid was 45.61%. The deviation between theoretical and measured values was less than 0.50%. The measured values were close to the theoretical values.

Conclusion:

The method is suitable for the screen and determination of acid radicals in chemical reference products containing methanesulfonic acid,hydrochloric acid,phosphoric acid,sulfuric acid and butadisulfonic acid.

成盐能够有效改善药物的成药性,提高药物的溶解度、纯度和稳定性[1],2020年版《中华人民共和国药典》中,药物与盐酸成盐品种达四百余个,与磷酸、硝酸、硫酸等酸根成盐药物品种近200种[2],数目众多。中国食品药品检定研究院化学药品标准物质具有化学药的明显特性,即成盐品种比较多,这类含有酸根类成盐的化学对照品存在潜在的不均匀特性以及成盐比例的定值不准确性的风险,随着酸根分子量的增长,以及分子量占比的增加,酸根的定值准确与否影响该品种定值的准确性。化学对照品量值的准确性直接影响我国的药品生产与检验工作,因此成盐药物的酸根准确定值具有重要价值,建立简便、准确、适用性广的酸根定值方法对含酸根药物进行酸根含量及纯度测定具有较高价值和意义。
目前对成盐药物中酸根含量定值的方法包括核磁共振定量法、滴定法、离子色谱法、高效液相色谱法等[2-7],因盐酸、磷酸、硫酸、甲磺酸及丁二磺酸无紫外吸收,不易通过常规高效液相色谱法进行含量测定,且核磁共振定量法成本较高,滴定法精确度较低。离子色谱法具有高效、准确、自动化的优点,能够用于测定化学对照品和制剂中酸根的含量[8-10],且能够同时测定药物中多种阴离子和阳离子的含量[311-12]和药物主成分[13-14]的含量。因此拟建立通用的离子色谱法进行酸根定值,选择盐酸胍法辛、磷酸奥司他韦、硫酸沙丁胺醇、甲磺酸沙非胺、丁二磺酸腺苷蛋氨酸作为研究对象进行方法学考察验证。
Metrohm 940 Professional IC Vario离子色谱仪,配置电导检测器、自动进样器(858 Professional Sample Processor)、二元泵、柱温箱,万通公司;Mettler XPE 206DR,十万分之一电子天平,梅特勒-托利多仪器有限公司;Milli-Q超纯水处理系统,Millipore公司;KS-3000DV液晶超声波清洗器,昆山洁力美超声仪器有限公司。
酸根类对照品:氯离子标准溶液(批号2021C029-2)、硫酸根标准溶液(批号239027)、磷酸根标准溶液(批号236039-2)浓度均为1 000 μg·mL-1,购于国家有色金属及电子材料分析测试中心,丁二磺酸二钠(纯度100%,批号1-F-881)购于广州佳途科技股份有限公司,甲磺酸钠(纯度97%,批号20220913)购于国药集团化学试剂有限公司。
供试品(待测化学对照品):盐酸胍法辛(纯度99.8%,批号430049-202201)、甲磺酸沙非胺(纯度99.7%,批号430001-202201)、硫酸沙丁胺醇(纯度99.9%,批号100328-202205)、磷酸瑞格列汀(纯度98.7%,批号430036-202201)、丁二磺酸腺苷蛋氨酸(纯度99.3%,批号140867-202301)均来源于中国食品药品检定研究院。
试剂:碳酸钠(ACS,批号1.06329.0500)和碳酸氢钠(ACS,批号1.06392.0500)均购于默克公司。
精密称取甲磺酸钠、丁二磺酸钠于量瓶中,量取氯离子、磷酸根、硫酸根标准溶液于同一量瓶中,加纯化水稀释制成每1 mL含甲磺酸根35 μg、氯离子15 μg、磷酸根80 μg、硫酸根40 μg、丁二磺酸根200 μg的混合溶液,作为系统适应性溶液,典型图谱如图2所示。
精密称取甲磺酸钠、丁二磺酸二钠的对照品适量,分别加纯化水溶解并稀释制成每1 mL约含甲磺酸根0.23 μg、丁二磺酸根0.38 μg的对照品溶液。精密量取氯离子标准溶液、硫酸根标准溶液、磷酸根标准溶液适量,分别加纯化水稀释制成每1 mL约含氯离子0.16 μg、硫酸根0.22 μg、磷酸根0.17 μg的对照品溶液。
称取盐酸胍法辛、磷酸奥司他韦、硫酸沙丁胺醇、甲磺酸沙非胺、丁二磺酸腺苷蛋氨酸适量,精密称定,分别加纯化水超声溶解并定容,制成每1 mL约含盐酸胍法辛0.1 mg、磷酸奥司他韦0.1 mg、硫酸沙丁胺醇0.2 mg、含甲磺酸沙非胺0.2 mg、丁二磺酸腺苷蛋氨酸0.1 mg的溶液,典型图谱如图3所示。
取对照品溶液,分别加纯化水稀释制成每1 mL水中含甲磺酸0.035 μg、氯离子0.013 μg、硫酸根0.050 μg、磷酸根0.025 μg、丁二磺酸根0.05 μg的溶液,纯化水作为空白溶液,典型图谱如图4所示。
采用Metrosep A Supp 5-250/4.0(250 mm×4.0 mm,5 μm)色谱柱,检测器为电导检测器,以6.4 mmol·L-1碳酸钠溶液和2.0 mmol·L-1碳酸氢钠溶液为淋洗液,等度洗脱,柱温30 ℃,流速0.7 mL·min-1,进样体积20 μL。甲磺酸根、氯离子、硫酸根、磷酸根、丁二磺酸根出峰顺序及保留时间如图2所示,盐酸胍法辛、磷酸奥司他韦、硫酸沙丁胺醇、甲磺酸沙非胺、丁二磺酸腺苷蛋氨酸的供试品溶液和灵敏度溶液典型图谱如图3图4所示。
精密称取对照品甲磺酸钠30 mg于250 mL量瓶中,加纯化水超声溶解并定容,即得甲磺酸钠对照品储备液;精密称取对照品丁二磺酸二钠24 mg于200 mL量瓶中,加纯化水超声溶解并定容,即得丁二磺酸二钠对照品储备液;精密量取氯离子标准溶液2.6 mL、磷酸根标准溶液3.5 mL各置于100 mL量瓶中,加纯化水稀释并定容,即得氯离子对照品储备液和磷酸根对照品储备液;精密量取硫酸根标准溶液3.4 mL于50 mL量瓶中,加纯化水稀释并定容,即得硫酸根对照品储备液。
分别量取上述5种对照品储备液适量,加纯化水逐级稀释成对照品储备液浓度75%、50%、30%、20%、10%的系列对照品溶液,精密吸取不同浓度对照品溶液20 μL,按“2.2”项下色谱条件进样分析,以质量浓度(X,mg·mL-1)为横坐标,峰面积(Y)为纵坐标,绘制标准曲线并进行回归计算,分别得到5种酸根的线性回归方程,结果见表1。5种酸根在各自浓度范围内线性关系良好。
精密吸取“2.1.2”项下对照品溶液,按“2.2”项下色谱条件连续进样6次,记录峰面积,计算RSD。结果甲磺酸根、氯离子、磷酸根、硫酸根、丁二磺酸根峰面积的RSD(n=6)分别为1.6%、0.40%、1.4%、0.67%、1.1%,表明仪器精密度良好。
精密吸取“2.1.3”项下供试品溶液,于供试品溶液制备后0、2、4、6、12 h分别按“2.2”项下色谱条件进行测定,计算峰面积的RSD。结果甲磺酸根、氯离子、磷酸根、硫酸根、丁二磺酸根峰面积的RSD(n=5)分别为1.9%、1.2%、1.9%、0.89%、0.74%。表明供试品溶液在12 h内稳定。
取盐酸胍法辛、甲磺酸沙非胺、硫酸沙丁胺醇、磷酸瑞格列汀、丁二磺酸腺苷蛋氨酸各6份,分别按“2.1.3”项下方法配制供试品溶液,并按照“2.2”项下色谱条件进行测定。以成盐形式计算,甲磺酸沙非胺中甲磺酸含量为23.69%,RSD为1.9%;盐酸胍法辛中盐酸含量为12.66%,RSD为1.2%;磷酸瑞格列汀中磷酸含量为17.38%,RSD为0.59%;硫酸沙丁胺醇中硫酸的含量为16.49%,RSD为0.24%;丁二磺酸腺苷蛋氨酸中丁二磺酸含量为45.61%,RSD为0.74%。测定结果与样品酸根含量理论值见表2
称取盐酸胍法辛20 mg、甲磺酸沙非胺40 mg、硫酸沙丁胺醇20 mg、磷酸瑞格列汀20 mg、丁二磺酸腺苷蛋氨酸10 mg,精密称定,分别置于20 mL量瓶中,加纯化水超声溶解并定容,得到5个化学对照品各自供试品浓溶液。取“2.3”项下系列对照品溶液中的50%浓度对照品储备液作为对照品溶液。精密量取5个供试品浓溶液1 mL分别于20 mL量瓶中,加入各自的对照品溶液6、10、14 mL,加纯化水稀释至刻度,得到80%、100%、120%浓度的溶液,每种浓度平行制备3份,按照“2.2”项下色谱条件进行测定,按照标准曲线法计算各样品的加标回收率,并计算RSD,如表3所示。
取盐酸胍法辛、甲磺酸沙非胺、硫酸沙丁胺醇、磷酸瑞格列汀、丁二磺酸腺苷蛋氨酸适量,按“2.1.3”项下方法配制供试品溶液,按“2.2”项下色谱条件进行测定,以成盐形式通过标准曲线法计算样品中酸根的含量,甲磺酸沙非胺中甲磺酸含量为23.69%;盐酸胍法辛中盐酸含量为12.66%;磷酸瑞格列汀中磷酸含量为17.38%;硫酸沙丁胺醇中硫酸的含量为16.49%;丁二磺酸腺苷蛋氨酸中丁二磺酸含量为45.61%。测定值与理论值一致,偏差<0.5%。
比较了Thermo DionexTMIonPacTMAS11 (250 mm×4.0 mm,5 μm)色谱柱与Metrosep A Supp 5 250/4.0(250 mm×4.0 mm,5 μm)色谱柱,根据与仪器适配度及色谱峰出峰效果,选择Metrosep A Supp 5 250/4.0(250 mm×4.0 mm,5 μm)色谱柱。根据色谱柱标准淋洗液,比较了1.0 mmol·L-1碳酸氢钠-3.2 mmol·L-1碳酸钠溶液、2.0 mmol·L-1碳酸氢钠-6.4 mmol·L-1碳酸钠溶液、3.0 mmol·L-1碳酸氢钠-9.6 mmol·L-1碳酸钠溶液作为淋洗液时甲磺酸根、氯离子、磷酸根、硫酸根、丁二磺酸根色谱峰的出峰时间及分离度。结果,当淋洗液浓度增加时,5种离子色谱峰出峰时间有不同比例的缩短,以2.0 mmol·L-1碳酸氢钠-6.4 mmol·L-1碳酸钠溶液为最佳。当柱温升高时,柱压降低,出峰时间提前,综合考虑柱压与出峰时间,在25、30、35 ℃中选择30 ℃作为柱温。
因盐酸、磷酸、硫酸、甲磺酸及丁二磺酸无紫外吸收,不易通过常规高效液相色谱法进行含量测定,因此建立离子色谱法用于含盐酸、磷酸、硫酸、甲磺酸及丁二磺酸化学对照品中的酸根含量测定。根据建立的离子色谱方法对甲磺酸沙非胺、盐酸胍法辛、磷酸瑞格列汀、硫酸沙丁胺醇和丁二磺酸腺苷蛋氨酸中酸根进行测定,测定结果与理论值一致,表明对照品定值准确,本方法同时可用于化学原料药中盐酸、磷酸、硫酸、甲磺酸及丁二磺酸的含量测定。因离子色谱检测灵敏度高,本方法也可用于化学对照品中盐酸、磷酸、硫酸、甲磺酸及丁二磺酸的筛查。
但该方法仍存在局限性,因溶剂为水,不适用于在水中溶解性差的药物酸根含量测定。后续可继续开发难溶药物的酸根定值方法,通过对药物进行合适的前处理或借助新仪器新技术解决该问题[15-16],在化学对照品定值过程中,建议将离子色谱法测定值与滴定法、核磁共振定量法等方法测定结果进行对比,以保证化学对照定值的准确性。
