Objective:

To establish the determination methods for chromatographic fingerprint and multiple constituents,and then evaluate the quality grade of 103 batches of Banlangen granules that were from 55 manufacturers based on the reference drug.

Methods:

Ultra-high-performance liquid chromatography (UPLC) analysis was used to establish the fingerprint of Banlangen granules and quantitate the contents of 7 components-uridine,adenine,vernine,(RS)-goitrin,adenosine,syringin and clemastanin B. The test was performed on the Waters ACQUITY UPLC HSS T3 (100 mm×2.1 mm,1.8 μm) column with gradient elution using methanol-water at a flow rate of 0.2 mL·min-1. The injection volume was 2 μL,and the column temperature was constant at 30 ℃. Then,the results were detected using adopting a multi-wavelength detection mode. Next,the characteristic peaks and their belonging were clarified by comparing them with the reference drug. The sample fingerprint similarity and determination of contents were finally calculated,and the quality grades of Banlangen granules in each item were evaluated by using the reference drug.

Results:

The fingerprint was established,and 10 main characteristic peaks were identified. The similarities of the sample’s fingerprints ranged from 0.541 to 0.993,of which 101 batches were over 0.75 that were reached the second level,and 81 batches were over 0.90 that were reached the first level. The linear ranges of 7 components had good linear relationships within their respective ranges. The average recoveries (n=9) were 97.0%-104.7% with RSDs all below 3%. The precision,stability,and repeatability of methods were all good with RSDs all below 3%. Unified the sample specification to 5 g per bag,the content range of Banlangen granules samples was between 0.189-10.347 mg per bag. As a result,the content of 75 batches of samples reached the second level,and the content of 59 batches of samples reached the first level.

Conclusion:

The established methods are simple,accurate and rapid,which can be used for the quality control and grade evaluation of Banlangen granules,as well as the research paradigm of other traditional Chinese medicine preparations.

目的:

以对照制剂为随行对照,分别建立指纹图谱和多成分指标含量测定方法,对55个厂家103批板蓝根颗粒质量进行分级评价。

方法:

采用超高效液相色谱法,建立板蓝根颗粒指纹图谱,同时测定7个成分(尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、紫丁香酚苷、直铁线连宁B)含量。采用Waters ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8 μm)色谱柱,以甲醇-水为流动相,梯度洗脱,流速0.2 mL·min-1,进样量2 μL,柱温30 ℃,采用多波长检测模式。与对照品比较,确定特征峰及其化合物归属。以对照制剂为参照,计算样品指纹图谱相似度及成分含量,并初步分级评价其质量。

结果:

建立了板蓝根颗粒特征指纹图谱,共指认10个特征峰,样品指纹图谱相似度在0.541~0.993范围,101批制剂相似度>0.75,达到二级限度,81批制剂相似度≥0.90,达到一级限度。7个成分在各自范围内线性关系良好,平均回收率(n=9)为97.0%~104.7% (RSD均<3%),精密度、稳定性、重复性良好(RSD均<3%)。以5 g·袋-1计,7个成分总含量范围为0.189~10.347 mg·袋-1。结果显示,75批次样品的含量达到二级限度,59批次样品的含量达到一级限度。

结论:

建立的方法简便、准确、快速,可用于板蓝根颗粒的质量控制和等级评价,也可为其他中成药质量等级评价研究提供参考。

目的:

以对照制剂为随行对照,分别建立指纹图谱和多成分指标含量测定方法,对55个厂家103批板蓝根颗粒质量进行分级评价。

方法:

采用超高效液相色谱法,建立板蓝根颗粒指纹图谱,同时测定7个成分(尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、紫丁香酚苷、直铁线连宁B)含量。采用Waters ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8 μm)色谱柱,以甲醇-水为流动相,梯度洗脱,流速0.2 mL·min-1,进样量2 μL,柱温30 ℃,采用多波长检测模式。与对照品比较,确定特征峰及其化合物归属。以对照制剂为参照,计算样品指纹图谱相似度及成分含量,并初步分级评价其质量。

结果:

建立了板蓝根颗粒特征指纹图谱,共指认10个特征峰,样品指纹图谱相似度在0.541~0.993范围,101批制剂相似度>0.75,达到二级限度,81批制剂相似度≥0.90,达到一级限度。7个成分在各自范围内线性关系良好,平均回收率(n=9)为97.0%~104.7% (RSD均<3%),精密度、稳定性、重复性良好(RSD均<3%)。以5 g·袋-1计,7个成分总含量范围为0.189~10.347 mg·袋-1。结果显示,75批次样品的含量达到二级限度,59批次样品的含量达到一级限度。

结论:

建立的方法简便、准确、快速,可用于板蓝根颗粒的质量控制和等级评价,也可为其他中成药质量等级评价研究提供参考。

Objective:

To establish the determination methods for chromatographic fingerprint and multiple constituents,and then evaluate the quality grade of 103 batches of Banlangen granules that were from 55 manufacturers based on the reference drug.

Methods:

Ultra-high-performance liquid chromatography (UPLC) analysis was used to establish the fingerprint of Banlangen granules and quantitate the contents of 7 components-uridine,adenine,vernine,(RS)-goitrin,adenosine,syringin and clemastanin B. The test was performed on the Waters ACQUITY UPLC HSS T3 (100 mm×2.1 mm,1.8 μm) column with gradient elution using methanol-water at a flow rate of 0.2 mL·min-1. The injection volume was 2 μL,and the column temperature was constant at 30 ℃. Then,the results were detected using adopting a multi-wavelength detection mode. Next,the characteristic peaks and their belonging were clarified by comparing them with the reference drug. The sample fingerprint similarity and determination of contents were finally calculated,and the quality grades of Banlangen granules in each item were evaluated by using the reference drug.

Results:

The fingerprint was established,and 10 main characteristic peaks were identified. The similarities of the sample’s fingerprints ranged from 0.541 to 0.993,of which 101 batches were over 0.75 that were reached the second level,and 81 batches were over 0.90 that were reached the first level. The linear ranges of 7 components had good linear relationships within their respective ranges. The average recoveries (n=9) were 97.0%-104.7% with RSDs all below 3%. The precision,stability,and repeatability of methods were all good with RSDs all below 3%. Unified the sample specification to 5 g per bag,the content range of Banlangen granules samples was between 0.189-10.347 mg per bag. As a result,the content of 75 batches of samples reached the second level,and the content of 59 batches of samples reached the first level.

Conclusion:

The established methods are simple,accurate and rapid,which can be used for the quality control and grade evaluation of Banlangen granules,as well as the research paradigm of other traditional Chinese medicine preparations.

板蓝根颗粒现行标准收载于2020年版《中华人民共和国药典》(以下简称《中国药典》)一部[1],是由板蓝根单味药材制成的中药成方制剂,具有清热解毒、凉血利咽的功效,用于肺胃热盛所致的咽喉肿痛、口咽干燥、腮部肿胀、急性扁桃体炎、腮腺炎等症状[2-6]。板蓝根颗粒临床用药需求大,市面上不同厂家的板蓝根颗粒,仅从外观色泽就存在着很大差异,提示产品质量间的差异值得关注。板蓝根颗粒现行标准以尿苷、鸟苷和腺苷3个成分含量为定量指标,是需要满足的最低标准,但难以全面反映板蓝根颗粒整体质量。板蓝根颗粒具有多成分、多靶点的特点[7-9],化学质量指标应尽可能体现中药专属性、有效性与安全性。此外,因缺少绝对的质量优劣评价标准或者相对的质量参照物,板蓝根颗粒质量分级评价困难,需要进一步深入研究。为了更加全面评价其质量,区分质量优劣,需建立新的质量评价方法,创新质量等级评价模式。指纹图谱技术及其相似度评价方法可以呈现中药色谱或光谱等性质的图像信息[10-13],更加整体性反映中药化学信息,描绘出更加全面的“化学轮廓”,较单一的或指标成分的质控方法,更能体现整体性特征,是控制中药质量的有效手段之一。在指纹图谱定性分析基础上,选择分离度好、质量相关的指标成分,同时进行含量测定,即实现“定量指纹图谱”,可增加质量分析维度,有效提高检验效率[14]
中药对照制剂是指采用道地、优质、规范加工的原料药材(饮片)和辅料,严格按照制法和生产工艺规程并遵循药品生产质量管理规范制备的实物参照,是中药对照物质的创新形式[15-23]。将其引入质量评价体系中,可提供丰富的化学信息,消除因仪器、色谱柱等实验条件引起的误差,提高方法的重现性[22],并且作为“标尺”比对未知样品质量,为质量等级划分提供客观依据,为判断中成药质量优劣提供了可行模式。目前,基于对照制剂的中药制剂质量评价还处于探索阶段。据文献报道[23-26],已有将对照制剂应用于牛黄清胃丸、复方鲜竹沥液、大七厘丸等多个中成药质量评价的报道,但未见板蓝根颗粒有关的研究报道。
本研究研制了板蓝根颗粒对照制剂,创新性地将其引入质量评价研究中,建立了指纹图谱和多成分指标含量测定评价方法,并以对照制剂为实物参照,对板蓝根颗粒质量进行了初步分级评价,也可为其他中成药质量等级评价研究提供参考。
Waters ACQUITY UPLC超高效液相色谱仪(配有PDA检测器、Empower色谱工作站),AE-240电子分析天平(万分之一,Mettler公司)、XPE105电子分析天平(十万分之一,Mettler公司),Mili-Q纯水仪(Millipore公司),KQ-300DA型数控超声波清洗器(昆山市超声仪器有限公司),Allegra 25R离心机(贝克曼公司)。
对照品尿苷(批号110887-201803,含量99.5%)、腺嘌呤(批号110886-201102,含量99.4%)、鸟苷(批号111977-201501,含量93.6%)、(RS)-告依春(批号111753-201706,含量100.0%)、腺苷(批号110879-201703,含量99.7%)和次黄嘌呤(批号140661-201704,含量99.4%)来自中国食品药品检定研究院,对照品紫丁香酚苷(批号111760-201502,含量98%)、落叶松树脂醇(批号T27A11Z122494,含量98%)和吲哚乙酸(批号S26N9Y75964,含量98%)购于上海源叶生物科技有限公司。对照品直铁线连宁B(批号STC6500105,含量98%)购于上海诗丹德标准技术服务有限公司。蔗糖购自广州市华侨糖厂,糊精购自天津华盛化学试剂有限公司。乙醇(国药集团化学试剂有限公司)为分析纯,甲醇(Thermo Fisher公司)、甲酸(Merck公司)、乙酸(北京迪马科技有限公司)和醋酸铵(Sigma-Aldrich公司)均为色谱纯,水为超纯水。
试验共收集55个生产企业的103批板蓝根颗粒样品,为省/市药检所(安徽、江西、广西、广州、青海、云南、湖南)药品监督抽样药品或市售药品。3批对照制剂(批号为F20F035、F20F036、F20F037)采用课题组前期板蓝根药材质量评价研究被判定为优质的原料药材(产地甘肃,批号为B-59、B60、B61),按《中国药典》规定的处方制法委托某板蓝根颗粒厂家以实际生产规模制成。
采用Waters ACQUITY UPLC HSS T3 (100 mm×2.1 mm,1.8 μm)色谱柱,以甲醇(A)-水(B)为流动相,梯度洗脱(0~7 min,0%A→5%A;7~15 min,5%A→8%A;15~20 min,8%A→20%A;20~25 min,20%A→30%A;25~30 min,30%A→40%A),柱温30 ℃,流速0.2 mL·min-1,进样量2 μL。指纹图谱检测波长254 nm;含量测定采用多波长扫描,分别在220 nm波长下检测紫丁香酚苷,240 nm波长下检测(RS)-告依春,250 nm波长下检测鸟苷,260 nm波长下检测尿苷、腺嘌呤和腺苷,275 nm波长下检测直铁线连宁B。
取尿苷、次黄嘌呤、腺嘌呤、鸟苷、(RS)-告依春、腺苷、吲哚乙酸、紫丁香酚苷、直铁线连宁B、落叶松树脂醇的对照品适量,精密称定,用5%甲醇溶解,分别制备得质量浓度为0.180 6、0.133 3、0.103 1、0.4 02 8、1.014 4、0.546 0、0.113 3、0.156 4、0.090 0、0.557 5 mg·mL-1的单一对照品储备液。取适量单一对照储备液,用5%甲醇制备得6个系列稀释浓度的混合对照品溶液(表1)。
取装量差异项下的本品,研细,称取5 g,精密称定,置具塞锥形瓶中,精密加入5%甲醇10 mL,密塞,称量,超声(功率300 W,频率100 kHz)处理5 min,放冷,再称量,用5%甲醇补足减失的量,摇匀,滤过,取续滤液,即得。
精密称取蔗糖0.737 g及糊精0.041 g(相当于1 g板蓝根颗粒的辅料量),混匀,即得阴性样品;同“2.2.2”项下方法操作,制备阴性对照溶液。
根据课题组前期建立的板蓝根药材质量评价综合指数模型,采用性状特征、显微特征、成分含量及生物活性4个维度指标评价板蓝根药材质量,达到一级质量限度的板蓝根药材被认定为优质药材。选择3批次板蓝根优质药材(产地甘肃,批号为B-59、B60、B61)作为3批对照制剂原料药材,严格按照《中国药典》规定的投料量和制法,在生产车间以实际规模分别制备得3批对照制剂(批号为F20F035、F20F036、F20F037)。
取制备的3批对照制剂,采用本试验建立的方法标定指纹图谱和成分含量,比较以不同批次对照制剂为参照指纹图谱相似度结果差异,并且比较3批对照制剂含量测定值。结果表明,分别采用不同批次对照制剂为参照图谱评价同一批次样品,相似度结果接近;选定同一批次对照制剂为参照图谱评价其余对照对照制剂,相似度结果接近,且均>0.90;3批对照制剂成分含量均符合《中国药典》板蓝根颗粒含量测定项下要求,且测定的成分总含量接近,其中F20F036号对照制剂的成分总含量最接近3批对照制剂总含量测定结果平均值。综上,选择F20F036号对照制剂为本试验实物对照制剂。
对照制剂溶液制备方法同“2.2.2”项下方法。
取“2.2.2”项下供试品溶液(厂家1,样品批号20190402),按“2.1”项下条件进样测定,得板蓝根颗粒样品色谱图。观察色谱图(图1),选择信号较强,分离较好的色谱峰进行归属,经与对照品溶液色谱图和PDA光谱图比对,指认出10个色谱峰为主要特征峰,化合物归属见图1。其中腺苷色谱峰(峰6)分离度好,保留时间适中,相对峰面积大,故选择腺苷色谱峰为参照峰。
取供试品溶液(厂家1,样品批号20190402)连续进样6次,计算腺苷色谱峰峰面积和保留时间的RSD分别为0.76%和0.10%,表明仪器精密度良好。
取同一供试品溶液(厂家1,样品批号20190402),分别于溶液制备后0、2、4、8、12和24 h测定其指纹图谱,计算得各特征峰峰面积的RSD<5%,保留时间的RSD<1%,表明供试品溶液在试验温度下24 h内稳定。
取同一批样品(厂家1,样品批号20190402),平行制备6份供试品溶液,分别测定其指纹图谱,以腺苷色谱峰为参照峰,计算得各特征峰峰面积的RSD<5%,保留时间的RSD<1%,表明该方法重复性良好。
对103批次样品和对照制剂的色谱图进行分析(其中55个厂家与对照制剂的色谱图见图2),直观分析结果表明,除厂家4、5、6、26外,其余厂家不同批次的样品指纹图谱整体轮廓基本一致,说明批间一致性良好。对全部图谱进行聚类热图二维分析(见图3),其中横向聚类样品,纵向聚类色谱峰,热图颜色代表归一化峰面积百分比。色谱峰聚类结果表明,尿苷、腺苷色谱峰单独聚为一类,提示样品不同,二者峰面积差异较大,因此二者是强特征峰。直观分析和聚类分析均表明,在不同样品中鸟苷色谱峰和(RS)-告依春色谱峰峰面积差异明显,也是样品指纹图谱的主要差异色谱峰,在进行质量控制时需重点关注。
以腺苷色谱峰为参照峰,对保留时间在5~30 min,相对峰面积>0.1%的色谱峰进行积分。将各积分后的色谱图导入“中药色谱指纹图谱相似度评价软件(2012版)”,以对照制剂色谱图为参照图谱,多点校正,计算样品相似度,结果见图4。在“2.4.5”项下,结合样品指纹图谱比较分析,拟定相似度0.75为二等制剂限度,相似度0.90为一等制剂限度。结果101批制剂相似度>0.75,达到二级限度;81批制剂相似度≥0.90,达到一级限度。
取“2.2.1”项下混合对照品溶液、供试品溶液(厂家1,样品批号20190402)和阴性对照溶液,分别按照“2.1”项下方法测定。结果表明,以各成分色谱峰计,理论塔板数>5 000,分离度>1.5,且阴性无干扰,方法专属性好。对照品及阴性色谱图见图5
取“2.2.1”项下对照品储备液适量,用5%甲醇制备为6个系列稀释浓度的混合对照品溶液。精密吸取混合对照品溶液1 μL,按照“2.1”项下方法测定,记录峰面积。以对照品质量浓度(X,mg·mL-1)为横坐标,峰面积(Y)为纵坐标,绘制标准曲线,并进行线性回归。结果见表2,表明7个成分在各自浓度范围内线性关系良好。
取同一份板蓝根颗粒供试品溶液(厂家1,样品批号20190402),按照“2.1”项下条件连续进样6次,记录峰面积并计算RSD。结果表明,尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、紫丁香酚苷、直铁线莲宁B峰面积的RSD均<2%,表明方法精密度良好。
取一份板蓝根颗粒供试品溶液(厂家1,样品批号20190402),分别于溶液制备后0、2、4、8、12和24 h按“2.1”项下谱条件进样测定,记录峰面积并计算各成分含量的RSD。结果表明,尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、紫丁香酚苷、直铁线莲宁B的含量的RSD均<3%,表明供试品溶液在24 h内稳定。
取同一批号板蓝根颗粒样品(厂家1,样品批号20190402),按“2.2.2”项下方法平行制备6份供试品溶液,按“2.1”项下条件进样,记录峰面积并计算各待测物的含量及RSD。结果表明,尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、紫丁香酚苷、直铁线莲宁B的平均含量分别为0.18、0.02、0.17、0.16、0.18、0.02、0.06 mg·g-1,RSD均<3%,表明方法的重复性良好。
取同一批已测知含量的板蓝根颗粒样品(厂家1,样品批号20190402),研细,称取5 g,精密称定,共称取9份,每3份为1组。分别向上述9份样品中精密加入混合对照品溶液0.8、1.0、1.2 mL,各平行3份,制得加标量为低、中、高水平的样品,按“2.2.2”项下方法制备供试溶液,按“2.1”项下条件进样测定,计算加样回收率。7个成分的加样回收率在97.0%~104.7%,RSD均<3%,表明方法的准确度良好。
取样品按“2.2.2”项下方法制备供试品溶液,并在“2.1”项色谱条件下进样测定,外标法计算含量,每批样品平行测定2次,结果见表3
根据《中国药典》的规定、对照制剂和样品实测含量,对55个厂家103批样品的质量进行初步分级。2020年版《中国药典》一部中板蓝根颗粒含量测定项下规定,每袋含板蓝根以尿苷、鸟苷、腺苷的总量计,不得少于0.70 mg(以每袋5 g计)。本试验方法的提取溶剂、色谱系统与《中国药典》方法一致。根据《中国药典》含量要求判定样品测定结果,共有29批样品3个分总含量低于0.70 mg,被判定为不合格样品。这29批样品中,26批样品以尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、紫丁香酚苷、直铁线莲宁B 7个成分总含量计低于1.0 mg。因此,根据《中国药典》规定限度和本试验7个成分含量测定结果折算,以7个成分总含量计,拟定二级质量限度为1.0 mg(以每袋5 g计)。3批对照制剂7个成分总含量相近,以对照制剂测定结果的平均值为参照,拟定一级质量限度为1.6 mg(以每袋5 g计)。按照上述拟定限度判断,75批次样品达到二级质量限度,59批次样品达到一级质量限度。
板蓝根颗粒药理活性明确,具有抗菌、抗病毒(流感病毒、乙型肝炎病毒)[27-28]、抗炎[29]、免疫调节作用。板蓝根颗粒化学成分种类多样,主要化学成分有生物碱[30]、有机酸类[31]、糖类[32]、氨基酸[33]、硫代葡萄糖苷及其代谢产物[34]、核苷[35]、苯丙素及其苷类[36],其中生物碱[37]又包括吲哚类生物碱、喹唑酮类生物碱、喹啉类生物碱等。据文献报道,板蓝根颗粒中(RS)-告依春和直铁线莲宁B具有明显的抗病毒活性[38]。(RS)-告依春在体外细胞模型与体内鸡胚模型中均表现出抗病毒活性[39],直铁线莲宁B对人流感病毒(甲型、乙型)多种分型(H1N1、H7N3、H3N2、H6N2、H9N2)具有抗病毒活性[40]。现行2020年版《中国药典》板蓝根颗粒含量测定项下以尿苷、鸟苷、腺苷3个成分含量总量为质量指标,虽然3个成分是板蓝根颗粒中含量较高成分,但不是板蓝根颗粒专属成分,且与药效相关性小,还需考虑增加板蓝根颗粒有效成分指标,增加质量控制整体性和有效性。经课题组对板蓝根颗粒活性成分进行筛选研究,(RS)-告依春、紫丁香酚苷、直铁线莲宁B是板蓝根颗粒抗流感强活性组分中的3个差异性成分[41],提示是板蓝根颗粒抗流感病毒活性相关的物质基础,且经含量初步考察,在制剂中上述3个差异性成分的含量较高,能够被UPLC定量检测,因此确定它们为含量测定指标。此外,腺嘌呤色谱峰相对面积较大,经含量考察,其在制剂中含量水平与紫丁香酚甘相当,因此增加腺嘌呤为含量测定指标。最终,本研究除保留《中国药典》中3个含量测定指标,还增加腺嘌呤、(RS)-告依春、紫丁香酚苷和直铁线莲宁B为含量测定指标,以7个成分总含量为质量评价和分级依据,增加了方法的有效性和整体性。
试验考察了Waters ACQUITY UPLC CSH C18 (100 mm×2.1 mm,1.7 μm)、CORTECS UPLC T3 (100 mm×2.1 mm,1.6 μm)、Atlantis Premier BEH C18 (100 mm×2.1 mm,1.7 μm)、ACQUITY UPLC HSS T3 (100 mm×2.1 mm,1.8 μm)4种不同色谱柱对供试品溶液各色谱峰分离效果,结果表明ACQUITY UPLC HSS T3分离效果最好;保持其他条件不变,考察在甲醇-水、甲醇-0.1%甲酸、甲醇-0.15%乙酸、含10 mmol·L-1醋酸铵甲醇-含10 mmol·L-1醋酸铵水4种流动相系统下样品图谱中各色谱峰的分离效果。结果表明,甲醇-水流动相系统下基线平稳,主要色谱峰分离效果较好,故选择以甲醇-水为流动相进行梯度洗脱。
在190~400 nm波长范围内,对尿苷、腺嘌呤、鸟苷、(RS)-告依春、腺苷、紫丁香酚苷、直铁线莲宁B 7个成分的对照品溶液进行扫描。结果显示,在254 nm波长下检出峰数量最多、特征明显,适用于指纹图谱分析。紫丁香酚苷在220 nm,(RS)-告依春在240 nm,鸟苷在250 nm,尿苷、腺嘌呤和腺苷在260 nm,直铁线连宁B在275 nm处有最大吸收。因此采用多波长切换测定方法,在各自最大吸收波长下测定各成分含量。
对照制剂是中药对照的新形式,是采用已知道地、优质、规范加工原料药材(饮片)和辅料,严格制法和生产工艺规程并遵循药品生产质量管理规范制备,因此质量可控且相对优质。本研究研制出板蓝根颗粒对照制剂,将其分别作为指纹图谱相似度评价和多成分含量测定的参照,区分了不同厂家、不同批次板蓝根颗粒的质量差异,初步探索出基于对照制剂的质量分级评价模式。但因本研究研制的对照制剂批次有限,对照制剂的重复性、稳定性、批间一致性还需进一步深入研究和探讨。
研究结果表明,有101批次样品的指纹图谱相似度达二级限度,81批次样品达一级限度;有75批次样品的含量达二级限度,59批次样品达到一级限度。有4个厂家(厂家4、11、15、18)的3批样品含量均被判为二级限度以下,说明生产厂家投料存在原料药材(饮片)以次充好、偷工减料、生产工艺不规范等质量问题。含量被判定为一级的59批次样品中,指纹图谱相似度达到一级限度的有50批次,达到二级限度的有9批次,说明2种方法判定结果具有较好的一致性,均可作为板蓝根颗粒质量等级评价的有力手段。2个厂家(厂家33、47)的样品指纹图谱相似度和成分含量均被判定为二级以下,提示需改进生产工艺,提升其药品质量。
本研究引入对照制剂作为实物参照,建立了基于对照制剂的板蓝根颗粒指纹图谱和多成分含量测定质量评价方法,提高了方法的整体性与有效性。结果表明,2种评价方法结合对照制剂,可以有效区分不同厂家不同批次产品质量,初步实现板蓝根颗粒质量分级,为提升药品质量标准,引导厂家提升产品质量,促进优质优价提供参考。
