Objective:

To establish an HPLC method for the ratio of the enantiomer in fluralaner.

Methods:

The chromatographic separation was performed on chiral chromatography column CHIRALPAK AD-H (250 mm×4.6 mm,5 μm). The mobile phase consisted of n-hexane-anhydrous ethanol (60:40) and the flow rate was 1.0 mL·min-1. The detection wavelength was 265 nm and the column temperature was room temperature.

Results:

Under this chromatographic condition,three batches of samples were determined,the proportions of R-fluralaner and S-fluralaner were 1:1. The calibration curves of R-fluralaner and S-fluralaner good linearities at the range of 80.288-187.338 μg·mL-1(r=0.999 7) and 81.902-191.104 μg·mL-1 (r=0.999 9),respectively. The average recoveries were 100.6% and 100.8%.

Conclusion:

The method is accurate,reproducible and can be used for the ratio of enantiomers in fluralaner racemate.

目的:

建立氟雷拉纳对映异构体比例的高效液相色谱测定方法。

方法:

采用CHIRALPAK AD-H手性色谱柱(250 mm×4.6 mm,5 μm);以正己烷-无水乙醇(60:40)为流动相,流速1.0 mL·min-1,检测波长265 nm,柱温室温,进样量20 μL。

结果:

在本文色谱条件下,测定的3批样品,R-氟雷拉纳与S-氟雷拉纳峰面积的比例均为1:1,R-氟雷拉纳与S-氟雷拉纳质量浓度分别在80.288~187.338 μg·mL-1 (r=0.999 7)和81.902~191.104 μg·mL-1(r=0.999 9)范围内与峰面积呈良好的线性关系,平均回收率分别为100.6%和100.8%。

结论:

本方法准确度高,重复性好,可用于氟雷拉纳外消旋体中对映异构体的比例测定方法。

Objective:

To establish an HPLC method for the ratio of the enantiomer in fluralaner.

Methods:

The chromatographic separation was performed on chiral chromatography column CHIRALPAK AD-H (250 mm×4.6 mm,5 μm). The mobile phase consisted of n-hexane-anhydrous ethanol (60:40) and the flow rate was 1.0 mL·min-1. The detection wavelength was 265 nm and the column temperature was room temperature.

Results:

Under this chromatographic condition,three batches of samples were determined,the proportions of R-fluralaner and S-fluralaner were 1:1. The calibration curves of R-fluralaner and S-fluralaner good linearities at the range of 80.288-187.338 μg·mL-1(r=0.999 7) and 81.902-191.104 μg·mL-1 (r=0.999 9),respectively. The average recoveries were 100.6% and 100.8%.

Conclusion:

The method is accurate,reproducible and can be used for the ratio of enantiomers in fluralaner racemate.

氟雷拉纳(fluralaner)在2004年由日本日产化学和美国杜邦公司联合研发并获得化合物专利[1]。该化合物具有1对对映异构体,药物以右旋异构体和左旋异构体组成的消旋体上市,结构式见图1。2019—2023年英特威国际股份有限公司在我国相继对氟雷拉纳咀嚼片、溶液剂及滴剂进行了进口注册,主要应用于犬、猫体外寄生虫及鸡螨虫的防治[2]。Thomas等[3-6]研究得出,氟雷拉纳溶液可有效杀灭鸡皮刺螨,对蛋鸡体重无影响,且能提高产蛋率,耐受性、安全性良好。Walther等[7]在犬上的研究得出,氟雷拉纳宜与食物同服。此外,Rohdich等[8-11]通过研究证实,氟雷拉纳对跳蚤和蜱的杀灭作用持效期达12周。单次口服和静脉注射治疗后112 d内,氟雷拉纳在血浆中均可测出。局部用药对预防网纹革蜱传播的巴贝斯虫病在12周内均有效。口服该药对犬实验感染长角血蜱的有效期可长达114 d。
目前上市的药物成分为RS构型的外消旋体,国内氟雷拉纳仅见其消旋体合成工艺[12]、生物效应[13]的文献,但是通过查询相关文献、企业标准和药典中均未有针对氟雷拉纳的异构体测定方法的报道,近年来S-氟雷拉纳的手性拆分制备研究热度高涨[14-15],因此,建立氟雷拉纳消旋体中异构体的分离检测方法是非常必要的。本研究建立了高效液相色谱法测定氟雷拉纳中对映异构体的检测方法,并进行了完整的方法学研究,结果表明该方法专属性强,可有效分离及快速检出对映异构体,方法简便可行,为后续开发S-氟雷拉纳上市,并且为氟雷拉纳的质量控制提供重要的依据。
LC-20A高效液相色谱仪(岛津公司);ES 225SM-DR十万分之一电子天平(Precisa公司);UV-1900i紫外可见分光光度计(岛津公司)。
R-氟雷拉纳对照品(委托药明康德公司分离纯化制得,批号R-FL-001-P1,纯度99.652%);S-氟雷拉纳对照品(委托药明康德公司分离纯化制得,批号R-FL-001-P2,纯度99.448%);氟雷拉纳(公司自制,批号分别为FL-Z-200901、FL-Z-200910、FL-Z-200921);正己烷、无水乙醇(HPLC级,天津川普伟业科技有限公司);异丙醇(HPLC级,西陇科学股份有限公司)。
用直链淀粉-三(3,5-二甲基苯基氨基甲酸酯)硅胶为填充剂(大赛璐CHIRALPAK AD-H柱,4.6 mm×250 mm,5 μm或效能相当的色谱柱)[16],以正己烷-无水乙醇(60:40)为流动相,流速1.0 mL·min-1,检测波长265 nm,柱温室温,进样体积20 μL。
以无水乙醇为空白溶剂。
精密称取R-氟雷拉纳的对照品约125 mg,置50 mL量瓶中,加无水乙醇使溶解并稀释至刻度,摇匀,即得R-氟雷拉纳对照品储备液。精密量取R-氟雷拉纳对照品储备液5 mL,置100 mL量瓶中,加无水乙醇稀释至刻度,摇匀,即得。
精密称取S-氟雷拉纳对照品约125 mg,置50 mL量瓶中,加无水乙醇使溶解并稀释至刻度,摇匀,即得S-氟雷拉纳对照品储备液。精密量取S-氟雷拉纳对照品储备液5 mL,置100 mL量瓶中,加无水乙醇稀释至刻度,摇匀,即得。
取氟雷拉纳适量,精密称定,加无水乙醇溶解并稀释制成每1 mL中约含0.25 mg的溶液,即得。
分别取空白溶剂、R-氟雷拉纳对照品溶液、S-氟雷拉纳对照品溶液、供试品溶液各20 μL分别注入液相色谱仪,记录色谱图及峰面积。结果表明,R-氟雷拉纳与S-氟雷拉纳之间良好分离,溶剂无水乙醇不干扰本品异构体检查(见图2)。
精密量取R-氟雷拉纳对照品储备液3、4、5、6、7 mL,分别置100 mL量瓶中,加无水乙醇稀释至刻度,摇匀,即得R-氟雷拉纳系列浓度对照品溶液。精密量取S-氟雷拉纳对照品储备液3、4、5、6、7 mL,分别置100 mL量瓶中,加无水乙醇稀释至刻度,摇匀,即得S-氟雷拉纳系列浓度对照品溶液。精密量取上述系列对照品溶液各20 μL,分别按“2.1”项条件进样检测,记录色谱图及峰面积。以峰面积对浓度进行线性回归。求得R-氟雷拉纳、S-氟雷拉纳回归方程分别为
结果表明,R-氟雷拉纳质量浓度在80.288~187.338 μg·mL-1S-氟雷拉纳质量浓度在81.902~191.104 μg·mL-1范围内线性关系良好。
精密量取供试品溶液20 μL,按“2.1”项条件连续进样6次,记录色谱图及峰面积。结果R-氟雷拉纳的平均峰面积为5 831 672,RSD为0.030%;S-氟雷拉纳的平均峰面积为5 830 189,RSD为0.039%。表明进样精密度均良好。
精密量取供试品溶液20 μL,按“2.1”项条件进样测定,记录色谱图,按面积归一化法计算2个异构体的比例。设计变化因素为柱温、流动相比例、流速,考察各变化因素对2个异构体比例的影响。变化柱温为20、30 ℃,流动相比例调整为62:38、58:42,变化流速为0.9、1.1 mL·min-1。结果轻微改变测定条件,2个异构体峰分离度均>2.0,2个异构体峰面积的比值平均为0.998,RSD为0.079%,即2个异构体的比例为1:1,且基本不变,表明本方法异构体测定的耐用性良好。
取供试品溶液于室温下放置6 h,分别于第0、1、2、4、6 h,精密量取20 μL进样,记录色谱图及峰面积。结果R-氟雷拉纳的平均峰面积为5 795 890,RSD为0.065%;S-氟雷拉纳的平均峰面积为5 799 140,RSD为0.094%。表明供试品溶液在室温6 h内稳定。
分别精密量取R-氟雷拉纳、S-氟雷拉纳的对照品储备液各5 mL置同一100 mL量瓶中,用无水乙醇稀释至刻度,摇匀,即得氟雷拉纳异构体混合对照品溶液。取氟雷拉纳约15 mg,精密称定,置200 mL量瓶中,分别精密加入R-氟雷拉纳、S-氟雷拉纳的对照品储备液各5 mL,振摇使溶解,用无水乙醇稀释至刻度,摇匀,即得回收率80%组供试溶液,重复制备3份。取氟雷拉纳约25 mg,精密称定,置200 mL量瓶中,分别精密加入R-氟雷拉纳、S-氟雷拉纳的对照品储备液各5 mL,振摇使溶解,用无水乙醇稀释至刻度,摇匀,即得回收率100%组供试品溶液,重复制备3份。取氟雷拉纳约35 mg,精密称定,置200 mL量瓶中,分别精密加入R-氟雷拉纳、S-氟雷拉纳的对照品储备液各5 mL,振摇使溶解,用无水乙醇稀释至刻度,摇匀,即得回收率120%组供试品溶液,重复制备3份。取上述溶液各20 μL,分别注入液相色谱仪,记录色谱图及峰面积。按外标法以峰面积计算回收率。结果见表12
取氟雷拉纳3批,按“2.2.4”项下方法制备供试品溶液,按照“2.1”项下色谱条件,分别取20 μL注入液相色谱仪,记录色谱图及峰面积。计算R-氟雷拉纳与S-氟雷拉纳峰面积之间的比值,结果分别为0.998 7、0.998 8、0.999 0,即3批样品R-氟雷拉纳与S-氟雷拉纳峰面积的比例均为1:1。
委托药明康德公司采用手性制备柱分离纯化得到了氟雷拉纳2个对映异构体对照品,后通过比旋度的测定得到了左旋与右旋信息。
R-氟雷拉纳对照品溶液、S-氟雷拉纳对照品溶液及氟雷拉纳供试品溶液分别进行紫外全波长扫描,结果显示均在265 nm波长处有最大吸收,因此选用265 nm检测波长进行检测。
采用大赛璐AD-H(250 mm×4.6 mm,5 μm)色谱柱,首先以正己烷-异丙醇(50:50)为流动相,结果R-氟雷拉纳与S-氟雷拉纳良好分离,但S-氟雷拉纳拖尾因子略大;更换异丙醇为无水乙醇,改变流动相为正己烷-无水乙醇(50:50),结果R-氟雷拉纳与S-氟雷拉纳良好分离,S-氟雷拉纳峰形明显改善,但是2个对映异构体保留时间变近;降低无水乙醇比例,即调整流动相为正己烷-无水乙醇(60:40),结果R-氟雷拉纳与S-氟雷拉纳良好分离,2个对映异构体保留时间合理,且峰形对称。
氟雷拉纳在N-N二甲基甲酰胺、甲醇、乙醇、异丙醇、乙腈中溶解度好,在水、正己烷中几乎不溶。由于此液相色谱系统为正相模式,优选乙醇、异丙醇、正己烷等有机溶剂溶解样品。根据流动相为正己烷-无水乙醇(60:40),且氟雷拉纳在正己烷中几乎不溶,因此选择无水乙醇作为配制溶剂,与流动相的兼容性更好,且溶解性能也较好。
本研究以消旋体氟雷拉纳为原料,通过手性制备柱分离纯化后,经比旋度测定得到R-氟雷拉纳与S-氟雷拉纳。以正相手性色谱柱开发出了HPLC法检测氟雷拉纳中对映异构体比例的方法,并进行了专属性、线性、精密度、耐用性、溶液稳定性、回收率等详细的方法学验证试验。经验证得出,本方法检测氟雷拉纳中对映异构体的比例可行。同时本方法,也为推进S-氟雷拉纳上市,建立检测R-氟雷拉纳异构体的方法提供支持。
