Objective:

To establish a liquid chromatography mass spectrometry (LC-MS) method for the detection of nepasaikosaponin K in Bupleurum marginatum var. stenophyllum, which could be used to quickly identify whether Bupleurum marginatum var. stenophyllum was substituted for Bupleurum or was mixed with Bupleurum. At the same time, nepasaikosaponin K was detected in 16 batches of Bupleurum marginatum var. stenophyllum and 157 batches of bupleurum decoction pieces.

Methods:

Ultra-high performance liquid chromatography (UPLC) coupled with triple quadruple mass spectrometry (QqQ MS) was used to detect nepasaikosaponin K in Bupleurum and Bupleurum marginatum var. stenophyllum. Determination was carried out with the application of a Phenomenex Kinetex C18 (2.1 mm×100 mm, 1.7 μm) column at temperature of 25 ℃. The mobile phase was composed of acetonitrile(A) and water(B) with gradient elution(0-5 min 15% A→25% A; 5-0 min, 25% A→30% A; 30-35 min, 30% A→90% A; 35-36 min, 90% A→15% A; 36-40 min, 15% A) at a flow rate of 0.3 mL·min-1. The electrospray ionization(ESI-) source was performed in multiple reaction monitoring (MRM) mode of the transitions of m/z 943.5→797.3, m/z 943.5→635.3, and m/z 943.5→781.3.

Results:

The contents of nepasaikosaponin K detected in 16 batches of Bupleurum marginatum var. stenophyllum were much higher than minimum requirements. Nepasaikosaponin K was detected in 8 out of 157 batches of Bupleurum.

Conclusion:

The established method is verified by methodology to be specific, so as to develop a supplementary test method for the components of Bupleurum and Bupleurum marginatum var. stenophyllum. to improve the quality control standard and authentic identification investigation, and to effectively control the quality of Bupleurum decoction pieces and guarantee its clinical efficacy.

目的:

建立检测柴胡及藏柴胡中柴胡皂苷K成分的液相色谱-质谱法(LC-MS)检查方法,用以快速鉴别北柴胡是否为藏柴胡或混有藏柴胡。同时对16批藏柴胡和157批柴胡饮片中的柴胡皂苷K进行检测。

方法:

采用超高效液相色谱-三重四极杆串联质谱(UPLC-QqQ-MS)对柴胡及藏柴胡中柴胡皂苷K进行检测,色谱柱为Phenomenex Kinetex C18(100 mm×2.1 mm,1.7 μm),流动相为乙腈(A)-水(B),梯度洗脱(0~5 min,15% A→25% A;5~30 min,25% A→30% A;30~35 min,30% A→90% A;35~36 min,90% A→15% A;36~40 min,15% A),流速0.3 mL·min-1,柱温25 ℃,进样量5 μL;采用电喷雾离子源(ESI-),负离子模式扫描,多反应监测(MRM)模式检测,选择柴胡皂苷K特征分子离子峰m/z 943.5→797.3、m/z 943.5→635.3和m/z 943.5→781.3作为检测离子对。

结果:

16批藏柴胡中柴胡皂苷K的量远高于限度要求,157批柴胡饮片中有8批样品中均检出藏柴胡类成分柴胡皂苷K。

结论:

所建立的方法经方法学验证,专属性强,以期制定柴胡中藏柴胡成分检查项补充检验方法,提升柴胡饮片质量控制标准及真伪鉴别研究,可进一步有效控制其质量,保障临床疗效。

目的:

建立检测柴胡及藏柴胡中柴胡皂苷K成分的液相色谱-质谱法(LC-MS)检查方法,用以快速鉴别北柴胡是否为藏柴胡或混有藏柴胡。同时对16批藏柴胡和157批柴胡饮片中的柴胡皂苷K进行检测。

方法:

采用超高效液相色谱-三重四极杆串联质谱(UPLC-QqQ-MS)对柴胡及藏柴胡中柴胡皂苷K进行检测,色谱柱为Phenomenex Kinetex C18(100 mm×2.1 mm,1.7 μm),流动相为乙腈(A)-水(B),梯度洗脱(0~5 min,15% A→25% A;5~30 min,25% A→30% A;30~35 min,30% A→90% A;35~36 min,90% A→15% A;36~40 min,15% A),流速0.3 mL·min-1,柱温25 ℃,进样量5 μL;采用电喷雾离子源(ESI-),负离子模式扫描,多反应监测(MRM)模式检测,选择柴胡皂苷K特征分子离子峰m/z 943.5→797.3、m/z 943.5→635.3和m/z 943.5→781.3作为检测离子对。

结果:

16批藏柴胡中柴胡皂苷K的量远高于限度要求,157批柴胡饮片中有8批样品中均检出藏柴胡类成分柴胡皂苷K。

结论:

所建立的方法经方法学验证,专属性强,以期制定柴胡中藏柴胡成分检查项补充检验方法,提升柴胡饮片质量控制标准及真伪鉴别研究,可进一步有效控制其质量,保障临床疗效。

Objective:

To establish a liquid chromatography mass spectrometry (LC-MS) method for the detection of nepasaikosaponin K in Bupleurum marginatum var. stenophyllum, which could be used to quickly identify whether Bupleurum marginatum var. stenophyllum was substituted for Bupleurum or was mixed with Bupleurum. At the same time, nepasaikosaponin K was detected in 16 batches of Bupleurum marginatum var. stenophyllum and 157 batches of bupleurum decoction pieces.

Methods:

Ultra-high performance liquid chromatography (UPLC) coupled with triple quadruple mass spectrometry (QqQ MS) was used to detect nepasaikosaponin K in Bupleurum and Bupleurum marginatum var. stenophyllum. Determination was carried out with the application of a Phenomenex Kinetex C18 (2.1 mm×100 mm, 1.7 μm) column at temperature of 25 ℃. The mobile phase was composed of acetonitrile(A) and water(B) with gradient elution(0-5 min 15% A→25% A; 5-0 min, 25% A→30% A; 30-35 min, 30% A→90% A; 35-36 min, 90% A→15% A; 36-40 min, 15% A) at a flow rate of 0.3 mL·min-1. The electrospray ionization(ESI-) source was performed in multiple reaction monitoring (MRM) mode of the transitions of m/z 943.5→797.3, m/z 943.5→635.3, and m/z 943.5→781.3.

Results:

The contents of nepasaikosaponin K detected in 16 batches of Bupleurum marginatum var. stenophyllum were much higher than minimum requirements. Nepasaikosaponin K was detected in 8 out of 157 batches of Bupleurum.

Conclusion:

The established method is verified by methodology to be specific, so as to develop a supplementary test method for the components of Bupleurum and Bupleurum marginatum var. stenophyllum. to improve the quality control standard and authentic identification investigation, and to effectively control the quality of Bupleurum decoction pieces and guarantee its clinical efficacy.

柴胡始载于《神农本草经》,为中医常用清虚热药,既是传统中药,又是现代临床常用中药,主产于辽宁、河北、山西、河南、陕西及甘肃等地[1],具有疏肝解郁、发表退热和升举阳气等功效,主要用于治疗感冒发热、寒热往来、月经不调、肝郁气滞等病证[2-4]。其化学成分主要包括皂苷、黄酮、挥发油、多糖和炔类等[5-7]。现代药理研究表明,柴胡具有抗癌、抗炎、抗抑郁、降血脂和保护肝肺损伤等药理作用[8-12]。柴胡作为大宗中药材品种,市场需求量巨大,市场上有柴胡的栽培品、野生品和部分伪品,近年来随着过度采挖,野生柴胡资源急剧短缺,所以市场上大部分为栽培品,然而栽培品存在问题较多,包括药材基原多、药用状况混乱、质量参差不齐等[13-15]
柴胡药源的混乱给柴胡及其制剂的质量带来隐患,据调查显示,我国有42种柴胡,其中包含17个变种和7种变型[16]。市场的主流柴胡药材有北柴胡、竹叶柴胡、藏柴胡和锥叶柴胡。由于柴胡属植物的性状特征十分接近,在采集和使用时很难进行有效鉴别,加之正品柴胡药材资源匮乏等因素,使得多种混伪品与正品柴胡混用的现象普遍[17-20],临床用药的安全性和有效性受到很大的威胁。
藏柴胡作为柴胡近年来出现的主要伪品,由于其价格较低且柴胡皂苷a、d含量很高,达2.0%以上,很受相关制剂生产企业青睐,导致市场上藏柴胡流通量较大[21-22],柴胡中藏柴胡参伪研究是近年来研究的热点,因此建立快速、稳定的鉴别柴胡掺伪藏柴胡的方法尤为重要。目前在药材鉴别中常用的方法包括理化性质鉴定、性状鉴定和显微鉴定等,虽然能够在一定程度上鉴定中药的良莠[21-22],但采用这些传统的鉴别手段鉴别中药材对人员的专业能力要求较高[23-24],而且理化鉴定耗时耗力。近年来,有许多学者采用灵敏度高、选择性强的液质联用(LC-MS)技术对柴胡掺伪问题进行研究,赵丹彤等[25-26]采用高效液相色谱-串联质谱(HPLC-MS/MS)技术,对柴胡属饮片及含柴胡的中成药中的藏柴胡掺伪检测方法进行了研究。本文以中国食品药品检定研究院联合甘肃省级药检机构对市场及多家企业抽检的157批柴胡为样本,利用具有快速高效分析能力的UPLC-QqQ-MS(MRM)技术手段来建立柴胡掺伪藏柴胡的鉴别方法,以期制定柴胡药材及饮片中藏柴胡成分检查项的补充检验方法,完善质量控制技术和提高用药安全性。

柴胡皂苷K对照品(7285)购自上海诗丹德标准技术服务有限公司;柴胡对照药材(120992-201509)、竹叶柴胡对照药材(121343-201903)均购自中国食品药品检定研究院;甲醇、乙腈为色谱纯(默克公司);氨水为分析纯(国药集团化学试剂有限公司);水为超纯水。
北柴胡药材6批,南柴胡药材3批,藏柴胡药材22批,锥叶柴胡、竹叶柴胡、小叶黑柴胡药材各2批,银柴胡、三岛柴胡、红柴胡药材各1批,其中北柴胡、竹叶柴胡、南柴胡、黑柴胡、三岛柴胡、红柴胡等大部分样品均为产地采集的样品,马潇主任中药师将以上样品与标本及植物志核对后确定为对应的品种。样品信息详见表1
采用Phenomenex Kinetex C18(100 mm×2.1 mm,1.7 μm)色谱柱,流动相为乙腈(A)-水(B),梯度洗脱(0~5 min,15% A→25% A;5~30 min,25% A→30% A;30~35 min,30% A→90% A),流速为 0.3 mL·min-1,柱温为25 ℃,进样量为5 μL。
离子化模式为ESI-,进行多反应监测(MRM),选择柴胡皂苷K特征分子离子峰m/z 943.5→797.3(定量)、m/z 943.5→635.3和m/z 943.5→781.3(定性)作为检测离子对。
取柴胡皂苷K对照品适量,精密称定,加甲醇制成每1 mL含1.0 μg的溶液,即得。
取藏柴胡0.06 g、北柴胡对照药材1.94 g(掺伪3%),精密称定,精密加入含5%浓氨试液的甲醇溶液50 mL,称量,超声(功率500 W,频率80 kHz)处理30 min,放冷,再称量,用含5%浓氨试液的甲醇溶液补足减失的量,摇匀,用0.22 μm微孔滤膜滤过,即得。
表1中柴胡药材(南柴胡、锥叶柴胡、银柴胡、竹叶柴胡、三岛柴胡、黑柴胡、红柴胡)细粉0.5 g,精密称定,精密加入含5%浓氨试液的甲醇溶液50 mL,称量,超声(功率500 W,频率80 kHz)处理30 min,放冷,再称量,用含5%浓氨试液的甲醇溶液补足减失的量,摇匀,用0.22 μm微孔滤膜滤过,即得。
分别取藏柴胡(编号Z1)0.06、0.1、0.2、0.5、1、2 g,对应北柴胡(批号B1)1.94、1.9、1.8、1.5、1、0 g(相当于北柴胡中掺入藏柴胡分别为3%、5%、10%、25%、50%、100%),精密称定,精密加入含5%浓氨试液的甲醇溶液50 mL,称量,超声(功率500 W,频率80 kHz)处理30 min,放冷,再称量,用含5%浓氨试液的甲醇溶液补足减失的量,摇匀,用0.22 μm微孔滤膜滤过,即得。
取北柴胡(B1、B2、B3、B4、B5、B6)阴性样品0.5 g,精密称定,精密加入含5%浓氨试液的甲醇溶液50 mL,称定重量,超声(功率500 W,频率80 kHz)处理30 min,放冷,再称定量,用含5%浓氨试液的甲醇溶液补足减失的量,摇匀,用0.22 μm微孔滤膜滤过,即得。
取藏柴胡(Z1、Z2、Z3、Z4、Z5、Z6)0.5 g,按“2.4.1.1”项方法制备藏柴胡阳性样品溶液。
分别取对照品溶液、阴性样品溶液、阳性样品溶液、“2.3.3”项下各柴胡药材供试品溶液各5 μL进样,按“2.1”项下液相色谱条件及“2.2”项下质谱条件进行测定。结果不同种柴胡中均含有柴胡皂苷K,但藏柴胡中柴胡皂苷K含量均比其他几种柴胡药材含量高约10~30倍,其中以柴胡皂苷K特征离子平均峰面积计,约为北柴胡中柴胡皂苷K特征离子的近40倍。结果见图1
北柴胡阴性样品(B1、B2、B3、B4、B5、B6)中柴胡皂苷K特征离子平均峰面积为482 650,藏柴胡阳性样品(Z1、Z2、Z3、Z4、Z5、Z6)中柴胡皂苷K特征离子平均峰面积为21 113 271,数据高出阴性样品2个数量级,说明该方法专属性好,阴性对目标成分测定基本无干扰。
按“2.3.4”项下方法制备供试品溶液,按“2.1”项下液相色谱条件及“2.2”项下质谱条件进行测定。以样品中柴胡皂苷K(m/z 943.5→797.3)的峰面积Y为纵坐标,藏柴胡掺入量X为横坐标,绘制标准曲线:
结果见图2。且藏柴胡中柴胡皂苷K特征离子对m/z 943.5→797.3的色谱峰面积与其掺伪量相等的北柴胡药材差异较小,基质效应影响较小,结果见图3
取藏柴胡饮片(编号Z1),按“2.3.3”项下方法制备供试品溶液,按“2.1”项下液相条件及“2.2”项下质谱条件进行测定,连续进样6次,记录供试品溶液中柴胡皂苷K特征离子对的MRM色谱峰峰面积(m/z 943.5→797.3)。结果供试品溶液中柴胡皂苷K的MRM色谱峰峰面积的RSD为1.4%,小于3.0%,说明仪器精密度良好。
取藏柴胡饮片(编号Z1),按“2.3.3”项下方法平行制备6份供试品溶液,按“2.1”项下液相条件及“2.2”项下质谱条件进行测定,记录供试品溶液中柴胡皂苷K特征离子对的MRM色谱峰峰面积(m/z 943.5→797.3)。结果供试品溶液中柴胡皂苷K平均含量为0.956 mg·g-1,RSD为1.8%,小于3%,说明方法重复性良好。
取藏柴胡饮片(编号Z1),按“2.3.3”项下方法制备供试品溶液,按“2.1”项下液相条件及“2.2”项下质谱条件进行测定,分别于0、2、6、12、20、24 h进样,记录供试品溶液中柴胡皂苷K特征离子对的MRM色谱峰峰面积(m/z 943.5→797.3)。结果供试品溶液中柴胡皂苷K的MRM色谱峰峰面积的RSD为1.1%,小于3%,说明供试品溶液在24 h内稳定性良好。
取藏柴胡(编号Z1)0.02 g,北柴胡(编号B1)1.98 g,相当于北柴胡中掺入藏柴胡为1%,精密称定,精密加入含5%浓氨试液的甲醇溶液50 mL,称量,超声(功率500 W,频率80 kHz)处理30 min,放冷,再称量,用含5%浓氨试液的甲醇溶液补足减失的量,摇匀,用0.22 μm微孔滤膜滤过,即得。
将相当于北柴胡中掺1%藏柴胡的供试品溶液,按“2.1”项下液相条件及“2.2”项下质谱条件进行测定,结果MRM色谱图中柴胡皂苷K的检测离子对m/z 943.5→797.3、m/z 943.5→635.3和m/z 943.5→781.3的色谱峰的信噪比均大于10。该方法检测限为相当于北柴胡中掺入藏柴胡为1%,即可检出。
精密称取柴胡皂苷K对照品2.11 mg,置100 mL量瓶中,加甲醇溶解并稀释至刻度,制成每1 mL含21.1 μg的溶液,作为对照品储备液;将储备液逐级稀释,即得系列浓度的对照品溶液,质量浓度分别为0.105 5、0.211 0、0.527 5、1.012 8、2.025 6、5.064 0 μg·mL-1,按“2.1”项下液相条件及“2.2”项下质谱条件进行测定,以柴胡皂苷K的943.5→797.3的色谱峰峰面积为纵坐标,浓度为横坐标,绘制标准曲线,得回归方程:
结果表明柴胡皂苷K在0.105 5~5.064 0 μg·mL-1线性关系良好。
按照2020年版《中华人民共和国药典》四部通则0212“药材和饮片检定”项,杂质通常不得过3%。严格按标准要求进行控制,拟定北柴胡中掺伪藏柴胡的限度为3%。按“2.3.2”项下藏柴胡参比溶液的制备方法平行制备供试品样品5份,按“2.1”项下液相条件及“2.2”项下质谱条件进行测定,在对照品柴胡皂苷K的线性范围内,计算藏柴胡掺伪3%的供试品中柴胡皂苷K峰面积对应的浓度,以该柴胡皂苷K对照品溶液浓度作为柴胡药材及饮片中藏柴胡的检出限度。
5份藏柴胡掺伪3%北柴胡中柴胡皂苷K的m/z 943.5→797.3的色谱峰峰面积平均值4 257 338,根据柴胡皂苷K的m/z 943.5→797.3的色谱峰线性关系,计算藏柴胡掺伪3%时北柴胡中柴胡皂苷K的浓度为1.373 1 μg·mL-1,故将柴胡皂苷K对照品溶液的限度拟定为1.0 μg·mL-1
精密称取编号为Z001~Z016的16批藏柴胡样品粉末各0.5 g,按“2.3.3”项下方法制备供试品溶液,按“2.1”项下液相条件及“2.2”项下质谱条件进行测定。结果16批样品中均检出柴胡皂苷K成分,对照品溶液浓度1.012 8 μg·mL-1,其峰面积为3 521 846,16批藏柴胡中柴胡皂苷K色谱峰峰面积远高于本研究拟定的掺伪限度。结果见图4
柴胡属药材和饮片的主要活性成分是柴胡皂苷,近年来,研究者多采用液相色谱法对柴胡中的柴胡皂苷b1、b2、a、c、d等化学成分进行研究[24,27];而对其他柴胡皂苷类成分研究极少,且现有方法灵敏度低、各皂苷类成分多以同分异构体的形式存在,增加了分析检测工作的难度。柴胡皂苷K在北柴胡等品种中含量低,但其在藏柴胡中含量高,有助于提高掺伪后目标成分检测的灵敏度;但柴胡皂苷K的结构和多种皂苷极为相似,研究发现液相分离难度大、薄层色谱图中藏柴胡与柴胡的色谱斑点与颜色无较大差别[28]。所以采用液质联用法可以提高检测的灵敏度,使得柴胡属饮片中藏柴胡掺伪检测方法的准确度提高[26]
本实验对乙腈-水,甲醇-水,乙腈-0.1%甲酸水溶液等多个流动相系统进行了考察,结果表明,流动相系统选择乙腈-水进行梯度洗脱时柴胡皂苷K分离效果及峰形最好。故选择乙腈-水进行梯度洗脱,洗脱时间为40 min,色谱柱温度为25 ℃。通过对柴胡皂苷K(相对分子质量为943.5)对照品溶液进行子离子(daughter scan)扫描,选择3个丰度较高的碎片离子m/z 635.3、797.4、781.4作为柴胡皂苷K的特征离子,结果见图5
为了适应不同灵敏度的仪器,分别考察本方法在岛津LCMS-8060和SCIEX TQ-5500三重四极杆液质联用仪上的重现性,以柴胡皂苷K的m/z 943.5→797.3、m/z 943.5→635.3和m/z 943.5→781.3作为选择离子对进行检测。结果表明柴胡皂苷K在2种类型的仪器上均可检出,且色谱峰峰型和分离度均良好。另外,分别考察了不同型号的色谱柱、不同柱温、不同流动相系统等对样品中柴胡皂苷K检测结果的影响,结果表明,在不同的色谱条件下,检测方法的耐受程度较好,检测结果均符合规定。
目前,由于正品柴胡药材资源长时间短缺,导致多种混伪品与正品柴胡混淆用的问题突出[20],针对这一问题,为保证药品质量,中检院联合甘肃省级药检机构对市场及多家企业的柴胡开展了抽检工作。结果157批样品中8批柴胡饮片(S26、S85、S99、S105、S138、S142、S153、S155)中检出藏柴胡类成分柴胡皂苷K,限度大于定量限,结果见图6
随着中药产业的快速发展,部分中药材及饮片品种出现了资源短缺的状态,中药材以掺伪造假获来取经济利益的现象频发。中药补充检验方法在市场监管中发挥着积极作用,可以有效地打击非法添加、染色、掺伪造假和增重等不法行为。本研究通过对所建立方法的专属性和柴胡中藏柴胡掺入量限度等的研究考察,开发了一种操作简便,精密度高,稳定性好的补充检验方法,适用于检查柴胡掺伪藏柴胡的现象。
