Objective:

To establish an ultra-high performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) for the simultaneous determination of daidzein, formononetin, genkwanin, quercetin, rutin, apigenin, kaempferol, luteolin and puerarin in Tongfu Jingyaotong tincture.

Methods:

The Waters ACQUITY UPLC HSS T3 Column (100 mm×2.1 mm, 1.8 μm) was used with methanol and 0.1% formic acid (gradient elution) as the mobile phase at a flow rate of 0.3 mL·min-1. Column temperature was 45 ℃, and injection volume was 2.0 μL. Electrospray ion source was adopted with positive and negative ion modes and multi-reaction monitoring and acquisition.

Results:

Apigenin, genkwanin and daidzein had good linearity in the range of 0.2-1 000 ng·mL-1, puerarin, rutin, luteolin, kaempferol and formononetin in the range of 0.2-2 000 ng·mL-1, and quercetin in the range of 1.0-5 000 ng·mL-1. The resolution was good, and the RSDs of precision, repeatability, reproducibility and stability test were all below 5% (n=6). The average recoveries ranged from 82.7% to 111.8% with RSDs (n=6) of 2.0%-5.4%. The contents of daidzein, formononetin, genkwanin, quercetin, rutin, apigenin, kaempferol, luteolin and puerarin in 6 batches of Tongfu Jingyaotong tincture were 8.46-17.97 μg·mL-1, 0.62-1.67 μg·mL-1, 0.03-0.08 μg·mL-1, 1.34-2.22 μg·mL-1, 0.70-1.14 μg·mL-1, 0.48-0.99 μg·mL-1, 0.20-0.90 μg·mL-1, 0.10-0.16 μg·mL-1 and 167.95-227.75 μg·mL-1, respectively.

Conclusion:

This method has strong specificity, wide linear range, accuracy and efficiency, and can detect the contents of 9 active ingredients in Tongfu Jingyaotong tincture at the same time, which offers an effective quality control method for Tongfu Jingyaotong tincture.

目的:

建立一种同时测定熥敷颈腰痛酊中大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素、葛根素含量的超高效液相色谱串联质谱法(UPLC-MS/MS)。

方法:

采用Waters ACQUITY UPLC HSS T3色谱柱(100 mm×2.1 mm,1.8 μm),以0.1%甲酸水溶液为流动相A,甲醇为流动相B,梯度洗脱,流速为0.3 mL·min-1,柱温为45 ℃,进样量为2.0 μL;电喷雾(ESI)离子源,正、负离子模式多反应监测采集。

结果:

芹菜素、芫花素、大豆苷元在0.2~1 000 ng·mL-1范围内线性良好,葛根素、芦丁、木犀草素、山柰酚、芒柄花黄素在0.2~2 000 ng·mL-1范围内线性良好,槲皮素在1.0~5 000 ng·mL-1范围内线性良好;分离度较好,精密度、重复性、重现性、稳定性试验RSD均小于5%(n=6);平均加样回收率在82.7%~111.8%,RSD为2.0%~5.4%(n=6)。6批熥敷颈腰痛酊中大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素、葛根素的含量范围分别为8.46~17.97 μg·mL-1、0.62~1.67 μg·mL-1、0.03~0.08 μg·mL-1、1.34~2.22 μg·mL-1、0.70~1.14 μg·mL-1、0.48~0.99 μg·mL-1、0.20~0.90 μg·mL-1、0.10~0.16 μg·mL-1、167.95~227.75 μg·mL-1

目的:

建立一种同时测定熥敷颈腰痛酊中大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素、葛根素含量的超高效液相色谱串联质谱法(UPLC-MS/MS)。

方法:

采用Waters ACQUITY UPLC HSS T3色谱柱(100 mm×2.1 mm,1.8 μm),以0.1%甲酸水溶液为流动相A,甲醇为流动相B,梯度洗脱,流速为0.3 mL·min-1,柱温为45 ℃,进样量为2.0 μL;电喷雾(ESI)离子源,正、负离子模式多反应监测采集。

结果:

芹菜素、芫花素、大豆苷元在0.2~1 000 ng·mL-1范围内线性良好,葛根素、芦丁、木犀草素、山柰酚、芒柄花黄素在0.2~2 000 ng·mL-1范围内线性良好,槲皮素在1.0~5 000 ng·mL-1范围内线性良好;分离度较好,精密度、重复性、重现性、稳定性试验RSD均小于5%(n=6);平均加样回收率在82.7%~111.8%,RSD为2.0%~5.4%(n=6)。6批熥敷颈腰痛酊中大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素、葛根素的含量范围分别为8.46~17.97 μg·mL-1、0.62~1.67 μg·mL-1、0.03~0.08 μg·mL-1、1.34~2.22 μg·mL-1、0.70~1.14 μg·mL-1、0.48~0.99 μg·mL-1、0.20~0.90 μg·mL-1、0.10~0.16 μg·mL-1、167.95~227.75 μg·mL-1

结论:

本方法专属性强,线性范围宽,准确高效,可同时对熥敷颈腰痛酊中9个有效成分进行含量检测,为该方建立有效的质量控制方法。

Objective:

To establish an ultra-high performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) for the simultaneous determination of daidzein, formononetin, genkwanin, quercetin, rutin, apigenin, kaempferol, luteolin and puerarin in Tongfu Jingyaotong tincture.

Methods:

The Waters ACQUITY UPLC HSS T3 Column (100 mm×2.1 mm, 1.8 μm) was used with methanol and 0.1% formic acid (gradient elution) as the mobile phase at a flow rate of 0.3 mL·min-1. Column temperature was 45 ℃, and injection volume was 2.0 μL. Electrospray ion source was adopted with positive and negative ion modes and multi-reaction monitoring and acquisition.

Results:

Apigenin, genkwanin and daidzein had good linearity in the range of 0.2-1 000 ng·mL-1, puerarin, rutin, luteolin, kaempferol and formononetin in the range of 0.2-2 000 ng·mL-1, and quercetin in the range of 1.0-5 000 ng·mL-1. The resolution was good, and the RSDs of precision, repeatability, reproducibility and stability test were all below 5% (n=6). The average recoveries ranged from 82.7% to 111.8% with RSDs (n=6) of 2.0%-5.4%. The contents of daidzein, formononetin, genkwanin, quercetin, rutin, apigenin, kaempferol, luteolin and puerarin in 6 batches of Tongfu Jingyaotong tincture were 8.46-17.97 μg·mL-1, 0.62-1.67 μg·mL-1, 0.03-0.08 μg·mL-1, 1.34-2.22 μg·mL-1, 0.70-1.14 μg·mL-1, 0.48-0.99 μg·mL-1, 0.20-0.90 μg·mL-1, 0.10-0.16 μg·mL-1 and 167.95-227.75 μg·mL-1, respectively.

Conclusion:

This method has strong specificity, wide linear range, accuracy and efficiency, and can detect the contents of 9 active ingredients in Tongfu Jingyaotong tincture at the same time, which offers an effective quality control method for Tongfu Jingyaotong tincture.

熥敷颈腰痛酊是甘肃省名中医赵继荣教授的经验效方,是由伸筋草、透骨草、威灵仙、葛根等8味药物经酊剂之浸渍法制备而成的外用制剂,方中伸筋草、透骨草为君药,以舒筋通络、活血止痛,两药相须为用增强彼此疗效;威灵仙、木瓜、桂枝、葛根共为臣药,威灵仙和木瓜祛风湿、通经络,桂枝温经通络、散寒止痛,葛根解肌舒筋止痛,桂枝与葛根相配伍,表里同治、寒温相合,温通筋脉,解肌养筋;红花、鸡血藤为佐药,补血活血、通络止痛。以上诸药合用,共奏温经通络、舒筋活血、散寒止痛的之功效[1]
既往研究表明,伸筋草乙醇、正丁醇、三氯甲烷提取物抗炎镇痛作用良好,可降低佐剂性关节炎大鼠血清中白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α (TNF-α)的水平[2]。申鸽等[3]的动物实验研究发现,珍珠透骨草醇提取物通过多组分、多靶点联合发挥抗炎活性。威灵仙中槲皮素、山柰酚、芦丁等多种化合物具有抗炎、抗氧化、镇痛的药物作用[4]。葛根中的葛根素、大豆苷元对神经组织损害和炎症导致的神经性疼痛具有缓解作用,抑制IL-1、IL-6和TNF-α的表达,减轻大鼠背根神经节引起的机械痛觉敏化[5]。桂枝醇提物具有较好的抗炎活性,木瓜中芦丁、槲皮素、山柰酚通过抑制Akt/NF-κB信号通路激活及促炎因子IL-1β、IL-6和TNF-α生成发挥抗炎镇痛活性[6-7]。鸡血藤中的儿茶素、芒柄花黄素具有抗炎、保护血管、改善血液流速的药理作用,红花中的山柰酚、芫花素、姜黄素等通过抑制一氧化氮(NO)的释放而起到抗炎活性[8-9]。综上可知,熥敷颈腰痛酊药物组成中所含共有化合物槲皮素、山柰酚、芒柄花黄素、芦丁等具有广泛的抗炎、镇痛、抗氧化、保护血管、抑制神经病理性疼痛的作用,其机制可能是多药物、多途径、多靶点共同起效,因此值得进一步研究。2020年版《中华人民共和国药典》中关于伸筋草、透骨草等中药的质量控制仅限于高效液相色谱法,但该法的专属性及灵敏性不及液质联用技术[10-11]。本课题组前期研究表明,“熥敷系列”中成药能够改善腰椎间盘突出症患者腰腿疼痛症状,降低炎性指标水平,提高生活质量,其功效为活血化瘀、温通经络、抗炎镇痛,但对其有效成分和作用机制尚未研究[12-14]。故此,本研究采用UPLC-MS/MS同时测定熥敷颈腰痛酊中9个化学成分的含量,旨在为该制剂的质量控制提供方法学参考,为中药酊剂的开发研究提供借鉴。
ACQUITY UPLC I-Class超高液相色谱仪、XEVO TQ-XS串联四极杆质谱系统、Masslynx质谱数据采集软件(Waters公司),Mikro 220R台式高速冷冻离心机(Hettich科学仪器公司),Vortex 2涡旋混匀仪(IKA集团),ME55/155DU十万分之一电子分析天平(METTLER TOLEDO科技有限公司),KQ3200超声波清洗器(昆山市超声仪器有限公司)。
对照品大豆苷元(含量99.99%,批号MUST-22101117)、芒柄花黄素(含量99.03%,批号MUST-23033005)、芫花素(含量98.80%,批号MUST-23061014)、槲皮素(含量98.85%,批号MUST-23042008)、芹菜素(含量98.12%,批号MUST-22120111)、山柰酚(含量99.99%,批号MUST-23030915)购于成都曼思特生物科技有限公司;对照品芦丁(含量≥98%,批号MUST-22039021)、木犀草素(含量≥98%,批号MUST-23010018)、葛根素(含量≥98%,批号MUST-22110405)购于中国食品药品检定研究院;色谱级甲酸和甲醇购于赛默飞世尔科技(中国)有限公司,超纯水(杭州娃哈哈集团有限公司);熥敷颈腰痛酊样品(甘肃省中医院药学部,批号20220101、20220301、20220701、20221001、20230101、20230401)。
采用Waters ACQUITY UPLC I-Class超高效液相色谱系统,色谱柱为Waters ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8 μm),以0.1%甲酸水溶液为流动相A,甲醇为流动相B,梯度洗脱(0~1 min,80% A;1~11 min,80% A→0% A;11~15 min,0% A;15~15.1 min,0% A→80% A;15.1~17 min,80% A),流速为0.3 mL·min-1,柱温为45 ℃,进样量为2.0 μL。
采用Waters XEVO TQ XS串联四极杆质谱系统,电喷雾离子源,正、负离子模式多反应监测(multiple reaction monitoring,MRM)采集,正离子离子源电压为3.0 kV,负离子离子源电压为2.0 kV,去溶剂温度为550 ℃,去溶剂气体流速为1 000 L·h-1,锥孔气体流速为150 L·h-1。各成分的质谱采集参数见表1
精密称取大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素、葛根素的对照品各10 mg,分别置于10 mL量瓶中,加甲醇-水(1∶1)5 mL充分溶解,定容至10 mL,制成质量浓度分别为1.12、1.07、0.98、1.05、0.96、1.01、1.14、0.94、1.02 mg·mL-1的对照品储备液。取上述对照品储备液各1 mL,置于同一20 mL量瓶中,加50%的甲醇水溶液稀释至刻度,摇匀,得到大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素、葛根素的质量浓度分别为56.00、53.50、49.00、52.50、48.00、50.50、57.00、47.00、51.00 μg·mL-1的对照品溶液。
精密量取100 μL样品,置于1.5 mL离心管中,加入甲醇-水(1∶1)900 μL,涡旋混匀10 min后,在4 ℃、12 000 r·min-1离心10 min。再用移液枪移取100 μL上清,置于1.5 mL离心管中,加入甲醇-水(1∶1)700 μL,涡旋混匀10 min后,在4 ℃、12 000 r·min-1离心10 min。取上清液200 μL,置于带250 μL内插管的进样瓶中,进行质谱检测。
精密吸取“2.3.1”项下配制的对照品溶液和“2.3.2”项下制备的供试品溶液、50%甲醇水溶液分别进样,色谱图见图1,各成分均未检测到干扰性成分,表明该方法专属性良好。
精密吸取“2.3.1”项下配制的对照品溶液,逐级稀释,以S/N=10时被测定成分的浓度为定量限(LOQ),S/N=3时被测定成分的浓度为检测限(LOD)。依次进样,以各成分的质量浓度X为横坐标、峰面积Y为纵坐标绘制标准曲线,结果见表2,各成分的线性关系良好。
精密吸取“2.3.1”项下配制的中浓度(100 ng·mL-1)的对照品溶液,连续进样6次,测得大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素和葛根素峰面积的RSD分别是0.63%、0.40%、1.2%、1.4%、1.3%、1.3%、13%、0.66%、0.59%,RSD均小于2%,表明该方法具有良好的精密度。
取同一批号(20220101)样品6份,按“2.3.2”项方法配制供试品溶液,依次进样,计算得大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素和葛根素的平均含量分别为15.32、1.48、0.06、1.52、0.78、0.80、0.66、0.14、196.79 μg·mL-1,各成分RSD分别为0.51%、1.4%、3.6%、4.0%、1.9%、2.1%、3.1%、1.6%、0.77%,RSD均小于5%,表明该方法具有良好的重复性。
取同一批号(20220101)样品6份,按“2.3.2”项方法制备供试品溶液,等分为6份,分别于室温下放置0、2、4、8、12、24 h后测定,记录峰面积,计算得大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素和葛根素峰面积的RSD分别为0.42%、0.76%、2.2%、4.3%、1.7%、1.8%、1.0%、1.2%、0.92%,RSD均小于5%,表明供试品溶液于室温下放置24 h内保持稳定。
精密量取已知目标成分含量的熥敷颈腰痛酊样品溶液6份,按照目标成分约100%的含量精密加入相应质量的对照品溶液,按照“2.3.2”项方法制备供试品溶液并进行测定,记录峰面积。计算加样回收率及RSD,结果见表3,平均回收率范围为82.7%~111.8%,RSD<6%,表明该方法准确。
取6批样品,分别按“2.3.2”项方法制备供试品溶液并进行测定,记录峰面积,计算各成分含量,结果见表4
HPLC法是药物制剂成分测定的常用方法,但对于多组分的制剂检测,同时检测的难度较大,需要使用DAD二极管阵列检测器筛选各自合适的检测波长,早年质谱前端连接的都是HPLC,但近些年发现UPLC有更好的分离效果,更适合作为质谱前端的分离设备[15]。预试验阶段曾尝试以HPLC法对熥敷颈腰痛酊进行定量分析,结果发现复方中各成分含量差异较大,究其原因为HPLC法无法达到同法检测,因此采用分离能力高、选择能力强、灵敏度精准的UPLC-MS/MS技术进行筛选、检测[16]
实验过程中将熥敷颈腰痛酊样品分别稀释10、50、80、100和150倍后上机检测,最后发现80倍浓度时所有待测成分都能够在各自的线性范围内显示,所以对熥敷颈腰痛酊样品的提取方法规定为以50%的甲醇水溶液稀释至80倍浓度。
测试过程中对流动相梯度进行了优化,结果表明梯度条件甲醇从20%升高到100%的时间越长,各色谱峰的分离会越好,但是槲皮素和芦丁2个成分在各种时间的梯度洗脱下都难使其保留时间分离,由于使用的是质谱检测,保留时间虽然区分不开,但是2个成分的检测离子对不同:槲皮素是302.954>69.019,芦丁是611.332>303.352,2个成分的提取离子流图不互相干扰,因此2个成分的含量测定不受影响。检测结果中,样品批号20220101和20220301结果较为相似,批号20220701和20221001结果较为相似,20230101和20230401结果较为相似,这可能与熥敷颈腰痛酊中各中药原料原产地、采收季节和炮制工艺的差异有关。
本研究建立的同时测定熥敷颈腰痛酊中9个成分大豆苷元、芒柄花黄素、芫花素、槲皮素、芦丁、芹菜素、山柰酚、木犀草素、葛根素的UPLC-MS/MS方法,具备简单易操作,准确效率高,专属性强,精密度重复性好,可作为熥敷颈腰痛酊质量评价参考。
