Objective:

To establish a hydrogen nuclear magnetic quantitative method which can rapidly determine the contents of linalool and protocatechuate in Homalomena occulta.

Methods:

Using dimethyl sulfoxide-d6(DMSO-d6)as the test solvent and pyrazine -as the internal standard, 1H-q NMR measurement was performed on a 400 MHz spectrometer. The quantitative resonance peaks of pyrazine, Linalool and protocatechuate were δ 8.66, δ 5.90 and δ 8.60, respectively.

Results:

The linear correlation coefficients of linalool and protocatechuate were good with r of 0.999 7 and 0.998 6, respectively. The RSDs of precision were 0.30% and 0.40%, and the recoveries were 98.8%-100.7%(RSD=3.7%) and 99.4%-100.6%(RSD=4.4%), respectively, which indicated that the established method was accurate, stable and feasible. And the whole detection process was completed in about three minutes. The contents of 2 components in 4 batches of Homalomena occulta were 0.328-0.398 mg·g-1(linalool)and 0.559-0.630 mg·g-1 (protocatechuate), respectively.

Conclusion:

The 1H-q NMR method has the advantages of simple operation, fast analysis speed and specificity, and can be used for the simultaneous determination of two active components in Homalomena occulta, providing a scientific basis for the overall quality evaluation and quality control of Homalomena occulta.

目的:

采用氢核磁定量方法快速测定千年健中主要成分芳樟醇、原儿茶酸的含量。

方法:

以氘代二甲基亚砜为测试溶剂,吡嗪为内标物,在400 MHz光谱仪上进行1H-qNMR测量,其中吡嗪、芳樟醇、原儿茶酸的定量共振峰分别为δ 8.66、δ 5.85~5.95、δ 6.78~6.80。

结果:

芳樟醇、原儿茶酸的线性关系良好,相关系数分别为0.999 7、0.998 5,精密度RSD分别为0.30%、0.40%,回收率分别为98.8%~100.7%(RSD为3.7%)和99.4%~100.6%(RSD为4.4%),表明所建立的氢核磁定量方法,结果准确,稳定可行,且整个检测过程在3 min左右完成。4批千年健药材中芳樟醇、原儿茶酸的含量范围分别为0.328~0.398 mg·g-1、0.559~0.630 mg·g-1

结论:

该方法操作简单,分析速度快,专属性强,可用于千年健中2个有效成分的同步测定,为千年健药材的全面质量评价和质量控制提供科学依据。

目的:

采用氢核磁定量方法快速测定千年健中主要成分芳樟醇、原儿茶酸的含量。

方法:

以氘代二甲基亚砜为测试溶剂,吡嗪为内标物,在400 MHz光谱仪上进行1H-qNMR测量,其中吡嗪、芳樟醇、原儿茶酸的定量共振峰分别为δ 8.66、δ 5.85~5.95、δ 6.78~6.80。

结果:

芳樟醇、原儿茶酸的线性关系良好,相关系数分别为0.999 7、0.998 5,精密度RSD分别为0.30%、0.40%,回收率分别为98.8%~100.7%(RSD为3.7%)和99.4%~100.6%(RSD为4.4%),表明所建立的氢核磁定量方法,结果准确,稳定可行,且整个检测过程在3 min左右完成。4批千年健药材中芳樟醇、原儿茶酸的含量范围分别为0.328~0.398 mg·g-1、0.559~0.630 mg·g-1

结论:

该方法操作简单,分析速度快,专属性强,可用于千年健中2个有效成分的同步测定,为千年健药材的全面质量评价和质量控制提供科学依据。

Objective:

To establish a hydrogen nuclear magnetic quantitative method which can rapidly determine the contents of linalool and protocatechuate in Homalomena occulta.

Methods:

Using dimethyl sulfoxide-d6(DMSO-d6)as the test solvent and pyrazine -as the internal standard, 1H-q NMR measurement was performed on a 400 MHz spectrometer. The quantitative resonance peaks of pyrazine, Linalool and protocatechuate were δ 8.66, δ 5.90 and δ 8.60, respectively.

Results:

The linear correlation coefficients of linalool and protocatechuate were good with r of 0.999 7 and 0.998 6, respectively. The RSDs of precision were 0.30% and 0.40%, and the recoveries were 98.8%-100.7%(RSD=3.7%) and 99.4%-100.6%(RSD=4.4%), respectively, which indicated that the established method was accurate, stable and feasible. And the whole detection process was completed in about three minutes. The contents of 2 components in 4 batches of Homalomena occulta were 0.328-0.398 mg·g-1(linalool)and 0.559-0.630 mg·g-1 (protocatechuate), respectively.

Conclusion:

The 1H-q NMR method has the advantages of simple operation, fast analysis speed and specificity, and can be used for the simultaneous determination of two active components in Homalomena occulta, providing a scientific basis for the overall quality evaluation and quality control of Homalomena occulta.

千年健始载于《本草纲目拾遗》,具有祛风湿、壮筋骨的功效[1-2]。千年健(Homalomena occulta (Lour.) Schott.)作为天南星科千年健属植物,其味苦、辛,性温,归肝、肾经,主产于广西、广东、云南等地,主治风寒湿痹、腰膝冷痛、 拘挛麻木、筋骨痿软,临床上常用于治疗风湿腰腿痛、风湿及类风湿性关节炎、骨折、跌打损伤等,主要化学成分为挥发油和倍半萜类化合物,此外还有酚酸类、生物碱、脂肪酸等[3-6]。药理研究表明其具有抗炎镇痛、抗阿尔兹海默症、抗骨质疏松、抗病原微生物、抗氧化、抗肿瘤等作用,具有广阔的开发应用前景。2020年版《中华人民共和国药典》采用气相色谱法测定芳樟醇的含量及其他常规鉴别项目作为控制千年健质量的手段和指标,存在耗时长,样品预处理复杂等缺点,且单一指标难以全面、科学地评价千年健药材的质量[7-9]
近年来,氢核磁定量(1H-qNMR)方法由于测试时间短,专属性强,样品前处理简单,不破坏样品自身结构,且定量过程中通过选用合适的内标物避免了对照品的使用,对于稳定性差的化合物是很好的定量分析方法,已被广泛应用于中药有效成分含量的测定等[10-18]。尚未见千年健相关氢核磁定量方法的报道。本文以吡嗪为内标物,采用1H-qNMR 法同时测定千年健药材中芳樟醇、原儿茶酸的含量,该法快速、简便,不需要对照品,测定结果准确,为千年健的全面质量评价和质量控制提供依据。2个成分的结构见图1
Bruker Advance Ⅲ 400 核磁共振波谱仪(Bruker公司),AL-104 电子分析天平(精度 0.000 1,慈溪市天东衡器厂),XO-5200DT超声波清洗机(南京先欧仪器制造有限公司),RE-2000A旋转蒸发仪(上海亚荣生化仪器厂),粉碎机(天津市泰斯特仪器有限公司)。
对照品芳樟醇(含量≥98%,购自希恩思化学技术有限公司)、原儿茶酸(含量≥98%,购自希恩思化学技术有限公司)、吡嗪(含量≥99%,购自上海麦克林生化科技有限公司),DMSO-d6(含量 99.8%,购自萨恩化学技术有限公司)。千年健药材购自当地各大药房并经鉴定,4批药材分别标记为1、2、3、4。
内标法为QNMR分析最常用的方法。测定制备的样品和内标物混合溶液,通过比较样品特征峰面积与内标峰面积计算样品含量。积分时,先将NMR谱图基线调平,选定峰积分区间,每个峰积分3次,RSD<1%时取平均值。用绝对定量公式计算待测组分的量:Ws=Wr×(As/Ar)×(Es/Er)其中Wr为内标物的量,AsAr分别为供试品特征峰和内标峰的峰面积,EsEr分别为供试品和内标物的质子当量重量(质量)(以分子量除以特征峰的质子数计算得到),然后根据称样量计算组分在样品中的含量[10-18]
精密称取10 mg内标物吡嗪,溶解至 DMSO-d6 1 mL中,即得。
取药材粉末(过40 目筛)5.00 g,精密称定,置于200 mL 锥形瓶中,加入80%乙醇100 mL,45 ℃超声(功率180 W,频率40 kHz)处理30 min,抽滤,重复2次,合并滤液,于50 ℃减压回收旋干得粗浸膏。精密称定10 mg提取物浸膏于质谱管中,用移液枪精密加入DMSO-d6490 μL和已配制好的内标溶液10 μL,超声溶解,移至核磁管待测。
精密称定芳樟醇和原儿茶酸各10 mg,用DMSO-d6配制浓度为10 mg·mL-1 储备液,分别取2个成分的储备液各13、25、50、100、200、400 μL于核磁管中,分别加入10 mg·mL-1内标液50 μL(吡嗪),后加DMSO-d6至500 μL,混合均匀,得芳樟醇和原儿茶酸质量浓度分别为0.26、0.5、1、2、4、8 mg·mL-1单一系列对照品溶液,按照“2.3”条件测定1H-qNMR波谱。
采用zg30脉冲序列,谱宽(SWH)为11 904.8 Hz,弛豫时间(D1)为1 s,脉冲宽度(P1)14.90 s,样品扫描次数(NS)32。所有数据均使用MestReNova软件处理。得到的检测图谱采用手动调整相位和基线,并进行手动积分。在本测试条件下,内标物吡嗪和原儿茶酸、芳樟醇对照品的1H-qNMR波谱图见图2
分别取“2.2.3”项芳樟醇和原儿茶酸系列对照品溶液,分别转移至5 mm核磁管中,按照“2.3”项条件下测定察,分别以(芳樟醇/吡嗪、原儿茶酸/吡嗪)质量比X为横坐标,相应1H-qNMR定量峰面积比Y为纵坐标,得芳樟醇、原儿茶酸的线性回归方程:
芳樟醇、原儿茶酸与内标物吡嗪质量比值在0.10~4.0内与两者定量峰面积比值呈良好的线性关系。
取“2.2.2”项中的供试品溶液(样品1),按照“2.3”项条件连续测试6次,连续测试3 d,分别考察仪器日内和日间精密度,结果芳樟醇峰面积和内标峰面比日内RSD为0.30%,日间RSD为1.5%;原儿茶酸峰面积和内标峰面积比的日内RSD分别为0.40%,日间RSD为1.8%,精密度好,也说明样品稳定性好。
取“2.2.2”项中的同一份供试品溶液(样品1),按照“2.3”项下条件分别在0、2、4、8、12、24 h进行1H-qNMR测试,记录用于定量的目标峰的积分面积。结果芳樟醇、原儿茶酸积分面积的RSD为1.3%和0.80%。表明样品的供试溶液24 h内稳定性良好。
取“2.2.2”项中的供试品溶液(样品1),平行制备6份,按照“2.3”项下条件进行测试。经测定芳樟醇、原儿茶酸的平均含量分别为0.340、0.570 mg·g-1,RSD分别为3.8%、3.5%。表明重复性良好。
精密称取6份已知芳樟醇、原儿茶酸含量的浸膏样品(样品1)10 mg,按照“2.2.2”项的方法制备供试品溶液且分别精密加入一定量的芳樟醇、原儿茶酸对照品,按照“2.3”项的条件进行测定,计算加样回收率,芳樟醇的平均回收率为99.6%,RSD为3.7%;原儿茶酸的平均回收率为100.1%,RSD为4.4%(见表2),表明试验方法准确度可靠。
按照“2.2”项下方法制备供试品溶液,照“2.3”项条件,分别对4批千年健药材的2个有效成分含量进行测定,对每一样品重复测定3次,并计算其含量。结果见表3,波谱图见图3。4批千年健药材中芳樟醇和原儿茶酸含量分别为0.328~0.398 mg·g-1和0.559~0.630 mg·g-1
千年健粉末分别用80%甲醇水溶液、70%甲醇水溶液、60%甲醇水溶液、丙酮、95%乙醇、水、80%乙醇水溶液、60%乙醇水溶液等溶剂进行提取。结果显示80%乙醇水溶液提取的浸膏2个有效成分含量均较高,且在DMSO-d6中的溶解性比较好,所以选择80%的乙醇水溶液作为提取溶剂。
内标物的选择考察了苯甲酸、3.5-二硝基苯甲酸、对苯二酚、顺丁烯二酸、苯甲酸苄酯、吡嗪,结果显示吡嗪性质稳定且不与样品发生反应,有1个易于识别的单峰,且信号峰不与待测峰重叠,所以选择吡嗪作为内标物。千年健浸膏和吡嗪在 DMSO-d6中溶解性最好,并且信号峰不与样品峰重叠,也没有干扰。因此试验选择 DMSO-d6作为氘代试剂。
通过对芳樟醇、原儿茶酸和吡嗪的谱峰分析,优先选择耦合裂分数最少的,且分离度较好的峰,根据以上要求,以δ 5.85~5.95为芳樟醇、δ 6.78~6.80原儿茶酸的峰为定量峰,以δ 8.66为内标吡嗪定量峰(图2)。
扫描次数考察了16、32、64、128次,测试结果显示,当扫描次数≥32 次时,积分面积稳定,RSD<1%,所以选择扫描32次。
1H-qNMR法具有专属性强,准确快捷,重现性好,不破坏化合物结构,且不需要使用对照品,仅使用内标物质即可同时对多种成分进行含量测定和鉴别等优点,已广泛应用于中药有效成分含量的测定[10-18]。本实验建立同时测定千年健中芳樟醇、原儿茶酸含量的1H-qNMR法,其结果表明从各药店购买的千年健药材都符合2020年版《中华人民共和国药典》芳樟醇不少于0.20%的标准,说明药材质量都符合要求,各产地的千年健药材中芳樟醇和原儿茶酸的含量差异不明显。
本实验建立了一种快速、简便的同时测定千年健中芳樟醇、原儿茶酸含量的1H-qNMR法。该方法操作简单,、速度快,、特异性强,、样品不丢失,不需要标准物质,同时只需用内标物就可以对多种化学成分进行定量,为千年健药材质量控制提供了一种快速、有效的新方法。
