Objective:

To establish an amino solid-phase extraction-HPLC method for the determination of three related substances in eldecalcitol soft capsules.

Methods:

The eldecalcitol soft capsules were extracted by an amino solid-phase extraction method, followed by eluting with ethyl acetate: n-hexane and anhydrous ethanol. The residue was then dissolved with acetonitrile and water after vacuum evaporation. An ODS column (250 mm×4.6 mm, 5 μm) was used. Water-acetonitrile with gradient elution was employed. The flow rate was 1.0 mL·min-1, the injection volume was 50 μL, and the detection wavelength was set at 265 nm.

Results:

Interference from blank excipients was eliminated with pretreatment. Baseline separation and good selectivity of eldecalcitol and other impurities was achieved. The stability of the test and reference solution was investigated at 4 ℃. The area change percentage of the impurities in the test solution were 98.6%-102.3% at 24 h. The area change percentage of the impurities in the reference solution were 101.2%, 96.9%, 98.5% and 96.2% at 24 h, respectively. The quantitation limits of eldecalcitol, trans impurity, pre-eldecalcitol and tachy impurity were 0.107 μg·mL-1, 0.102 μg·mL-1, 0.128 μg·mL-1 and 0.063 μg·mL-1, respectively. The detection limits were 0.054 μg·mL-1, 0.051 μg·mL-1, 0.064 μg·mL-1, 0.031 μg·mL-1, respectively. The linear ranges of 4 components were 0.1-6.3 μg·mL-, 0.1-1.2 μg·mL-, 0.1-5.6 μg·mL-and 0.1-1.1 μg·mL-1, respectively. The correlation coefficients(n=5) were 0.999 9, 1.000, 1.000 and 0.999 4, respectively. The average recoveries (n=6)of tachy impurity, pre-eldecalcitol and trans impurity were 106.6%, 88.6% and 102.8% with RSDs of 6.7%, 3.1% and 2.1%, respectively. In the three batches of samples, no impurities were detected except pre-eldecalcitol. The contents of pre-eldecalcitol were 5.0%, 5.0% and 5.1%, respectively.

Conclusion:

The method is simple to operate, economical and practical with high safety, strong specificity, good repeatability and high accuracy. The method provides a reliable basis for the study of drug quality such as eldecalcitol soft capsules and drugs with similar oily substrate.

目的:

建立一种氨基固相萃取-HPLC法测定艾地骨化醇软胶囊中3个同分异构体的有关物质。

方法:

采用氨基固相萃取法,经乙酸乙酯-正己烷洗脱,无水乙醇提取,提取液经减压蒸干后用乙腈复溶,经ODS色谱柱(250 mm×4.6 mm,5 μm)分析,以水-乙腈为流动相,梯度洗脱,流速为1.0 mL·min-1,进样量为50 μL,采用紫外检测器,在检测波长265 nm处检测有关物质。

结果:

固相萃取前处理可有效去除空白油干扰,艾地骨化醇与各杂质均可基线分离;在低温4 ℃条件下考察溶液稳定性,24 h时供试品溶液中各杂质面积百分比为98.6%~102.3%;杂质对照品溶液中,24 h时Tachy杂质、前体、艾地骨化醇和Trans杂质的面积百分比分别为101.2%、96.9%、98.5%和96.2%;艾地骨化醇、trans杂质、前体和tachy杂质的定量限分别为0.107、0.102、0.128、0.063 μg·mL-1,检测限分别为0.054、0.051、0.064、0.031 μg·mL-1;四者质量浓度分别在0.1~6.3 μg·mL-1、0.1~1.2 μg·mL-1、0.1~5.6 μg·mL-1和0.1~1.1 μg·mL-1范围内与峰面积呈良好线性关系,相关系数(n=5)分别为0.999 9、1.000、1.000和0.999 4;tachy杂质、前体和trans杂质回收率(n=6)分别为106.6%、88.6%和102.8%,RSD(n=6)分别为6.7%、3.1%和2.1%。3批样品含前体5.0%、5.0%和5.1%,杂质tachy、杂质Trans和未知单杂均未检出。

结论:

该方法操作简单,经济实用,安全性高,专属性强,重复性好,准确度高,为艾地骨化醇软胶囊及类似油性基质的药物质量研究提供可靠依据。

目的:

建立一种氨基固相萃取-HPLC法测定艾地骨化醇软胶囊中3个同分异构体的有关物质。

方法:

采用氨基固相萃取法,经乙酸乙酯-正己烷洗脱,无水乙醇提取,提取液经减压蒸干后用乙腈复溶,经ODS色谱柱(250 mm×4.6 mm,5 μm)分析,以水-乙腈为流动相,梯度洗脱,流速为1.0 mL·min-1,进样量为50 μL,采用紫外检测器,在检测波长265 nm处检测有关物质。

结果:

固相萃取前处理可有效去除空白油干扰,艾地骨化醇与各杂质均可基线分离;在低温4 ℃条件下考察溶液稳定性,24 h时供试品溶液中各杂质面积百分比为98.6%~102.3%;杂质对照品溶液中,24 h时Tachy杂质、前体、艾地骨化醇和Trans杂质的面积百分比分别为101.2%、96.9%、98.5%和96.2%;艾地骨化醇、trans杂质、前体和tachy杂质的定量限分别为0.107、0.102、0.128、0.063 μg·mL-1,检测限分别为0.054、0.051、0.064、0.031 μg·mL-1;四者质量浓度分别在0.1~6.3 μg·mL-1、0.1~1.2 μg·mL-1、0.1~5.6 μg·mL-1和0.1~1.1 μg·mL-1范围内与峰面积呈良好线性关系,相关系数(n=5)分别为0.999 9、1.000、1.000和0.999 4;tachy杂质、前体和trans杂质回收率(n=6)分别为106.6%、88.6%和102.8%,RSD(n=6)分别为6.7%、3.1%和2.1%。3批样品含前体5.0%、5.0%和5.1%,杂质tachy、杂质Trans和未知单杂均未检出。

结论:

该方法操作简单,经济实用,安全性高,专属性强,重复性好,准确度高,为艾地骨化醇软胶囊及类似油性基质的药物质量研究提供可靠依据。

Objective:

To establish an amino solid-phase extraction-HPLC method for the determination of three related substances in eldecalcitol soft capsules.

Methods:

The eldecalcitol soft capsules were extracted by an amino solid-phase extraction method, followed by eluting with ethyl acetate: n-hexane and anhydrous ethanol. The residue was then dissolved with acetonitrile and water after vacuum evaporation. An ODS column (250 mm×4.6 mm, 5 μm) was used. Water-acetonitrile with gradient elution was employed. The flow rate was 1.0 mL·min-1, the injection volume was 50 μL, and the detection wavelength was set at 265 nm.

Results:

Interference from blank excipients was eliminated with pretreatment. Baseline separation and good selectivity of eldecalcitol and other impurities was achieved. The stability of the test and reference solution was investigated at 4 ℃. The area change percentage of the impurities in the test solution were 98.6%-102.3% at 24 h. The area change percentage of the impurities in the reference solution were 101.2%, 96.9%, 98.5% and 96.2% at 24 h, respectively. The quantitation limits of eldecalcitol, trans impurity, pre-eldecalcitol and tachy impurity were 0.107 μg·mL-1, 0.102 μg·mL-1, 0.128 μg·mL-1 and 0.063 μg·mL-1, respectively. The detection limits were 0.054 μg·mL-1, 0.051 μg·mL-1, 0.064 μg·mL-1, 0.031 μg·mL-1, respectively. The linear ranges of 4 components were 0.1-6.3 μg·mL-, 0.1-1.2 μg·mL-, 0.1-5.6 μg·mL-and 0.1-1.1 μg·mL-1, respectively. The correlation coefficients(n=5) were 0.999 9, 1.000, 1.000 and 0.999 4, respectively. The average recoveries (n=6)of tachy impurity, pre-eldecalcitol and trans impurity were 106.6%, 88.6% and 102.8% with RSDs of 6.7%, 3.1% and 2.1%, respectively. In the three batches of samples, no impurities were detected except pre-eldecalcitol. The contents of pre-eldecalcitol were 5.0%, 5.0% and 5.1%, respectively.

Conclusion:

The method is simple to operate, economical and practical with high safety, strong specificity, good repeatability and high accuracy. The method provides a reliable basis for the study of drug quality such as eldecalcitol soft capsules and drugs with similar oily substrate.

代谢性骨病是继糖尿病和甲状腺疾病之后的第三大类内分泌代谢疾病。维生素D活性代谢物是一类具有生物活性的脂溶性类固醇衍生物,在体内发挥广泛的药理活性,包括调节钙磷平衡、骨重建、肠道和肾脏钙的吸收等作用[1],临床上对骨质疏松症疗效显著。艾地骨化醇(eldecalcitol)是一种新型活性维生素D衍生物[2-3],由日本中外制药株式会社与大正制药株式会社联合开发,于2001年1月在日本上市,用于治疗骨质疏松症,疗效优于阿法骨化醇,显示出广阔的使用前景。
艾地骨化醇分子式为C30H50O5,化学名为(1R,2R,3R,5Z,7E)-2-(3-羟基丙氧基)-9,10-开环胆甾-5,7,10(19)-三烯-1,3,25-三醇,结构式见图1。与骨化三醇相比,艾地骨化醇在2β位引入3-羟基丙氧基,使其与维生素D结合蛋白的亲和力更强,更难被24-羟化酶降解[4]。艾地骨化醇具有抑制骨吸收的作用[5-6],促进骨微塑建的效力也大大高于骨化三醇。此外,在促进肠道钙吸收方面,艾地骨化醇也优于阿法骨化醇[7-11]
目前《中华人民共和国药典》、美国药典、欧洲药典、日本药局方等均未收载艾地骨化醇及其相关制剂的质量控制方法,相关文献报道也很少。Wang等[12-13]报道,骨化三醇性质不稳定,光照和高温都会引起异构化,甚至产生降解的杂质,降低其功效,故将骨化三醇溶解在合适的脂溶性基质(如中链甘油三酯)中制备成软胶囊,保证骨化三醇稳定性,提高生物利用度。
据此,艾地骨化醇溶解于脂溶性基质制备的软胶囊中,主成分浓度极低(约占0.000 5%),而中链甘油三酯(辅料)占比极高,基质干扰显著,不仅影响了主成分浓度测定,更是大大影响了艾地骨化醇软胶囊的有关物质检查。因此,探索高效去除油性基质的样品前处理方法对于产品的质控至关重要。
本文采用氨基固相萃取法去除艾地骨化醇软胶囊基质干扰,并对萃取液进行浓缩,提高进样浓度后,采用反相液相色谱法进行艾地骨化醇软胶囊有关物质的测定,并进行方法验证。结果表明,此方法可有效去除基质干扰,灵敏度和准确度符合要求,研究结果为艾地骨化醇软胶囊及类似基质的药物质量研究提供了可靠依据。
艾地骨化醇与前体、tachy杂质(图2)互为可逆异构体,2对异构体均对温度敏感,故在验证时,采用临用前合成并标定的方法确定杂质纯度。且为避免杂质间互转造成干扰,对方法验证中的溶液稳定性、灵敏度、线性与范围和准确度验证进行了分组验证。
高效液相色谱仪(紫外检测器,岛津公司),YMC-Pack ODS-AM 色谱柱(250 mm×4.6 mm,5 μm);LUX-12水浴锅(北京陆希科技有限公司),PMK224ZH型万分之一电子天平(OHAUS公司),N-1210BV-W旋转蒸发仪(Heidolph公司),100 mg·mL-1氨基固相萃取小柱、100 mg·mL-1C18固相萃取小柱、100 mg·mL-1Silica固相萃取小柱(Welchrom公司)。乙腈、无水乙醇、乙酸乙酯和正己烷等试剂均为色谱纯,屈臣氏纯化水。
艾地骨化醇对照品(正大制药(青岛)有限公司,批号QCL-20210802,含量99.7%),艾地骨化醇软胶囊样品(正大制药(青岛)有限公司,批号2212281、2212291、2212301,规格0.75 μg·粒-1),艾地骨化醇前体(深圳远扬医药科技有限公司,批号27-JUN-2229,含量90.58%),trans杂质(深圳远扬医药科技有限公司,批号29-JUN-2229,含量97.80%),tachy杂质(甘肃皓天医药科技有限责任公司,批号HT018-45-14-2206001,含量89.6%),中链甘油三酯(湖北葛店人福药用辅料有限责任公司,批号F347C230401)。
取艾地骨化醇软胶囊内容物适量(约相当于艾地骨化醇12 μg),置10 mL量瓶中,用乙酸乙酯-正己烷(1∶10)溶解并稀释至刻度,摇匀,将所得溶液缓慢通过氨基固相萃取小柱[Welchrom®NH2,100 mg·mL-1或其他相当产品,预先用无水乙醇5 mL清洗,再用乙酸乙酯-正己烷(1∶10)清洗2次,每次5 mL],待液面近干时,用乙酸乙酯-正己烷(1∶10)25~30 mL洗脱,弃去洗脱液,继续用无水乙醇2 mL洗脱,收集洗脱液,30 ℃水浴减压蒸干,残留物加乙腈0.1 mL超声(功率500 W,频率40 kHz)使溶解,再加水0.1 mL,超声混合均匀(所有试验均避光操作)。
取中链甘油三酸酯约1.6 g,置10 mL量瓶中,加乙酸乙酯-正己烷(1∶10)溶解并稀释至刻度,摇匀,照“2.1.1”项下自“将所得溶液缓慢通过氨基固相萃取小柱……”处理。
取艾地骨化醇对照品适量,用乙腈溶解并稀释成每1 mL中约含60 μg的溶液,于80 ℃水浴中加热1 h(注意密封,防止溶剂挥发),放冷至室温,即得。
精密量取系统适用性溶液1 mL,置100 mL量瓶中,用乙腈稀释至刻度,摇匀,再精密量取1 mL,置10 mL量瓶中,用乙腈稀释至刻度,摇匀,即得。
取艾地骨化醇对照品和各杂质对照品各适量,精密称定,用50%乙腈溶解并稀释制成每1 mL含艾地骨化醇约60 μg、tachy杂质约0.6 μg、艾地骨化醇前体杂质约4.2 μg、trans杂质约0.3 μg的混合溶液。
取tachy杂质对照品适量,精密称定,用50%乙腈溶解并稀释制成每1 mL含tachy杂质约0.6 μg的溶液。
取艾地骨化醇前体杂质对照品适量,精密称定,用50%乙腈溶解并稀释制成每1 mL含艾地骨化醇前体杂质约4.2 μg的溶液。
取艾地骨化醇和trans杂质对照品适量,精密称定,用50%乙腈溶解并稀释制成每1 mL含艾地骨化醇和trans杂质各约0.3 μg的溶液。
trans杂质对照品适量,精密称定,用50%乙腈溶解并稀释制成每1 mL含trans杂质约0.3 μg的溶液。
用十八烷基硅烷键合硅胶为填充剂(250 mm×4.6 mm,5 μm),以水为流动相A,乙腈为流动相B,梯度洗脱(见表1),流速为1.0 mL·min-1,检测波长为265 nm,柱温为10 ℃,样品盘温度为4 ℃,进样体积50 μL。
精密量取50%乙腈50 μL,注入液相色谱仪,结果显示,在艾地骨化醇及相关杂质位置无干扰峰,表明乙腈不影响测定结果。
按“2.1.2”项下方法制备空白辅料溶液,按“2.2”项下色谱条件进样,结果色谱图见图3。空白辅料在艾地骨化醇及相关杂质位置无干扰峰产生,表明空白辅料不影响艾地骨化醇软胶囊的有关物质检测。
按“2.1.5”项下方法制备混合溶液,按“2.2”项下色谱条件进样,结果色谱图见图4。主成分与相邻杂质以及各已知杂质间均可基线分离,专属性良好。
取供试品溶液、tachy杂质对照品溶液、艾地骨化醇前体杂质对照品溶液、艾地骨化醇和trans杂质对照品溶液,按“2.2”项下色谱条件,分别在室温和低温(4 ℃)下于0、2、4、8、12、18、24、36和48 h进样。以0 h峰面积为100%,计算各时间点峰面积与0 h峰面积的百分比,结果表明,在室温条件下,18 h时供试品溶液中各杂质面积百分比为102.8%~103.8%,表明供试品溶液室温下18 h内稳定;杂质对照品溶液中,3 h时tachy杂质、前体、艾地骨化醇和trans杂质的面积百分比分别为105.5%、95.7%、102.0%和102.4%,表明室温条件下tachy杂质和前体杂质对照品溶液不稳定;在低温4 ℃条件下,24 h时供试品溶液中各杂质面积百分比为98.6%~102.3%;杂质对照品溶液中,24 h时tachy杂质、前体、艾地骨化醇和trans杂质的面积百分比分别为101.2%、96.9%、98.5%和96.2%,表明供试品和各杂质均24 h稳定。
分别精密量取“2.1.6~2.1.8”项下各对照品溶液适量,用50%乙腈逐级定量稀释配制定量限摸索溶液,照“2.2项下”色谱条件进样,当信噪比(S/N)为10∶1时对应的样品浓度即为定量限,待定量限确定后,再进一步稀释配制检测限溶液。当信噪比为3∶1时对应的样品浓度即为检测限。测定结果见表2
将艾地骨化醇及其杂质分3组进行线性与范围的验证,艾地骨化醇和trans杂质为1组,前体为1组,tachy杂质为1组。取艾地骨化醇及杂质对照品适量,加50%乙腈溶解并稀释至刻度,摇匀,逐级稀释,制成不同线性浓度的艾地骨化醇及杂质溶液。按照“2.2”项下色谱条件进样,记录峰面积。以浓度为横坐标,峰面积为纵坐标绘制标准曲线,回归方程及线性范围见表3。结果说明线性良好。
精密量取tachy杂质对照品溶液、艾地骨化醇前体杂质对照品溶液、trans杂质对照品溶液各1.0 mL,一一对应分别置于5 mL量瓶中,用乙酸乙酯-正己烷(1∶10)稀释至刻度摇匀,作为杂质对照品储备液;取本品内容物1.6 g(约相当于艾地骨化醇12 μg),精密称定,置10 mL量瓶中,精密加入杂质对照品储备液1 mL,照“2.1.1”项自“将所得溶液缓慢通过氨基固相萃取小柱……”处理。
按照外标法计算各杂质的回收率,结果表明,tachy杂质、前体杂质、trans杂质和主成分艾地骨化醇平均回收率(n=6)分别为93.1%、89.7%、75.7%和79.0%,RSD(n=6)分别为3.4%、4.5%、10.1%和2.9%。
按照加校正因子的面积归一化法计算各杂质回收率,结果表明,tachy杂质、前体杂质和trans杂质平均回收率(n=6)分别为106.6%、88.6%和102.8%,RSD(n=6)分别为6.7%、3.1%和2.1%。准确度良好,优于外标法,故采用加校正因子的面积归一化法作为本品有关物质的定量方式。
取样品(批号2212281),照“2.1.1”项方法配制供试品溶液,平行配制6份。按“2.2”项下色谱条件进样,计算各杂质的含量。结果表明,tachy杂质和trans杂质均未检出,前体杂质和总杂(不包括前体)平均含量(n=6)分别为6.6%和0.06%,RSD(n=6)分别为0.50%和2.1%。
由不同人员在不同日期采用不同仪器进行重现性试验。结果表明,tachy杂质和trans杂质均未检出,前体杂质和总杂(不包括前体)平均含量(n=12)分别为6.5%和0.08%,RSD(n=12)分别为1.6%和26.0%。主检测结果均无明显差异,该方法中间精密度良好。
更换不同批次色谱柱,改变柱温、流速和流动相比例等,照“2.2”项下色谱条件检测系统适用性溶液和供试品溶液,结果表明,各物质分离效果良好,供试品的检测结果无明显差异,方法耐用性良好。
取3批艾地骨化醇软胶囊,采用“2.1.1”项方法制备供试品溶液,按照“2.2”项条件检测,结果见表4
目前样品前处理方法主要有皂化回流、液-液萃取、固相萃取、超临界流体萃取等[14]。李攻科等[15]在测定去皮(壳)的鱼或海洋生物样品中的多氯联苯时采用了微波皂化萃取技术,处理过程中多次使用到浓硫酸等强腐蚀性试剂,实验过程繁琐复杂。因此,随着各种新技术的发展,皂化回流技术逐渐被更加环保高效的方法取代。液-液萃取成为当前最成熟且最常用的前处理方法,马海峰等[16]在测定食品中黄曲霉毒素B1时采用液液萃取的前处理方法,但该方法操作步骤烦琐,样品损失量大,成本高。而超临界流体萃取法属于新兴技术手段,方法不够完善成熟,且设备成本较高。
固相萃取是一种利用固体材料的选择性吸附对样品进行纯化富集的前处理手段,在食品、环境样品和生物制品等方面应用较多,具有有机试剂用量少,操作简单,富集纯化效率高且成本低廉诸多优势[17]。但固相萃取法在骨化醇类药物中的应用较少,目前报道中对油性基质药物的有关物质考察多为固相萃取-正相液相色谱法。
吴金平等[17]在建立骨化三醇软胶囊有关物质分析方法时,采用Online SPE-HPLC法考察了硅胶、硅藻土和C18固相萃取小柱对骨化三醇软胶囊内容物的提取分离效果,确定使用Hypersil Gold silica硅胶柱对样品进行前处理,并建立正相色谱条件考察有关物质,但正相色谱条件采用挥发性较强的正己烷和乙酸乙酯等试剂,且等度洗脱,针对艾地骨化醇的多个异构体杂质分离效果不理想。
Gianluca Bartolucci[18]等采用氨基固相萃取法对鱼肝油进行前处理,并联合LC-APCI-MS法对鱼肝油中的维生素D3的含量进行考察,采用氨基固相萃取柱前处理效果良好,但仅考察了主成分的含量。可为油基质的艾地骨化醇软胶囊多种异构体杂质检测的前处理方法提供参考。
结合上述文献,本文选择采用C18固相萃取小柱、氨基固相萃取小柱和Silica固相萃取小柱对艾地骨化醇软胶囊的内容物进行前处理,比较不同填料萃取柱的提取富集效果。结果显示,C18固相萃取小柱专属性差,Silica固相萃取小柱重复性和回收率较差,而氨基固相萃取小柱重复性、回收率和耐用性均较好。
氨基固相萃取小柱的填料,当用正己烷等非极性溶剂进行活化处理时,它可与任何含-OH,-NH或-SH基团的分子发生氢键作用,对结构异构体的分离和脂类样品中带极性基团的化合物提取具有良好的效果。恰好可以解决骨化醇类软胶囊在有关物质检查中存在的难点:①规格小,主成分浓度极低(约占0.000 5%)。②油性基质,中链甘油三酯占比极高,基质干扰问题严重。
因此,选用氨基固相萃取小柱进行艾地骨化醇前处理,采用反相高效液相色谱系统进行有关物质考察。
本文用乙酸乙酯-正己烷(1∶10)作为中链甘油三酯洗脱溶剂,分别对固相萃取小柱上样后的洗脱体积进行15、20、25及30 mL的考察。结果见表5图5。当乙酸乙酯-正己烷(1∶10)洗脱液用量为15~30 mL时,本品有关物质检测结果均无明显差异,为充分洗脱,拟定采用乙酸乙酯-正己烷(1∶10)洗脱液25~30 mL洗脱。
本文用乙醇作为艾地骨化醇洗脱溶剂,分别对固相萃取小柱上样经乙酸乙酯-正己烷(1∶10)淋洗后的艾地骨化醇洗脱体积进行1.5、2.0、2.5 mL考察。结果见表6图6。当无水乙醇洗脱体积为1.5~2.5 mL时,本品有关物质检测结果均无明显差异,拟定采用无水乙醇2 mL洗脱。
采用二极管阵列检测器,对艾地骨化醇溶液及各个杂质溶液的紫外吸收图谱进行考察,结果表明艾地骨化醇及各杂质在265 nm处均具有较强的紫外吸收,且峰形良好,故最终采用265 nm波长作为艾地骨化醇有关物质考察的检测波长。
按外标法计算时,tachy杂质、前体、trans杂质和艾地骨化醇回收率结果提示,前处理过程中主成分和杂质存在损失。主要原因是复溶体积小,导致转移过程中的体积损失比例增大,影响了回收率。按加校正因子的面积归一化法计算时,可以避免转移体积损失对结果的影响。
因此,本文选用了加校正因子的面积归一化法进行定量。
本文建立了艾地骨化醇软胶囊有关物质的氨基固相萃取-反相液相色谱检查方法,固相萃取法去除油基质的干扰,并对艾地骨化醇进行富集,提高了检测的灵敏度,采用反相色谱法对有关物质进行考察,避免了使用正相色谱的弊端如正相流动相挥发性较强,基线不易平衡,系统硬件要求高等。对该方法进行系统验证,保证方法的专属性、灵敏度及准确性均符合要求,为艾地骨化醇软胶囊及类似基质的药物质量研究提供可靠依据。
