Objective:

To study different batches of Xiaoer Resuqing granules by combining the method of fingerprint, multi-index component content determination and chemical pattern recognition analysis technology, and to provide the basis for its quality evaluation.

Methods:

The chromatographic SuperLu C18(250 mm×4.6 mm, 5 μm) column was adopted with mobile phase composed of methanol (A)-0.1% phosphoric acid (B) with gradient elution at a flow rate of 1.0 mL·min-1. The column temperature was 30 ℃, and the detection wavelength was 280 nm. Eleven characteristic components were identified, and the contents of seven active components, including puerarin, forsythiaside A, phillyrin, baicalin, wogonoside, baicalein and wogonin were determined. Hierarchical cluster analysis (HCA) and principal component analysis (PCA) were used to classify 15 batches of samples, and orthogonal least partial squares discriminant analysis (OPLS-DA) was performed. To screen out the difference components between batches of Xiaoer Resuqing granules.

Results:

The HPLC fingerprint of Xiaoer Resuqing granules and the content determination method of 7 components were established, the results of the two methods met the requirements. Twenty-eight common peaks were identified, 11 chromatographic peaks were identified, and the similarities of 15 batches of samples ranged from 0.995 to 0.999. HCA and PCA results showed that 15 batches were clustered into two categories, and OPLS-DA screened out 13 markers that caused the differences among the 15 batches of samples. There was little difference in the contents of ingredients.

Conclusion:

The established HPLC fingerprint of Xiaoer Resuqing granules and the determination method of 7 components are specific, accurate and reliable. Combined with chemical pattern recognition, it can be effectively used for the quality control of Xiaoer Resuqing granules.

目的:

采用指纹图谱、多指标成分含量测定与化学模式识别分析技术相结合的方法研究不同批次小儿热速清颗粒,为其质量评价提供依据。

方法:

采用SuperLu C18色谱柱(250 mm×4.6 mm,5 μm),流动相为甲醇(A)-0.1%磷酸水(B),梯度洗脱,体积流量1.0 mL·min-1,柱温30 ℃,检测波长280 nm,对15批小儿热速清颗粒指纹图谱进行相似度评价,指认出11个特征成分,并对葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素7个有效成分进行含量测定,采用聚类分析(HCA)及主成分分析(PCA)对15批样品进行分类并进行正交最小偏二乘法-判别分析(OPLS-DA),筛选出样品批次间差异成分。

结果:

建立了小儿热速清颗粒HPLC指纹图谱及7个成分的含量测定方法,且这2种方法学验证的结果均符合要求,确定了28个共有峰,并指认了11个色谱峰,15个批次相似度范围为0.995~0.999;HCA和PCA结果显示15个批次成分聚为2类,OPLS-DA分析筛选出13个造成样品批次间差异的标志物;多指标成分含量测定表明不同批次间小儿热速清颗粒中化学成分含量差异性较小。

结论:

建立的小儿热速清颗粒HPLC指纹图谱及7个成分含量测定方法的专属性强,准确、可靠,结合化学模式识别可有效的用于小儿热速清颗粒的质量控制。

目的:

采用指纹图谱、多指标成分含量测定与化学模式识别分析技术相结合的方法研究不同批次小儿热速清颗粒,为其质量评价提供依据。

方法:

采用SuperLu C18色谱柱(250 mm×4.6 mm,5 μm),流动相为甲醇(A)-0.1%磷酸水(B),梯度洗脱,体积流量1.0 mL·min-1,柱温30 ℃,检测波长280 nm,对15批小儿热速清颗粒指纹图谱进行相似度评价,指认出11个特征成分,并对葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素7个有效成分进行含量测定,采用聚类分析(HCA)及主成分分析(PCA)对15批样品进行分类并进行正交最小偏二乘法-判别分析(OPLS-DA),筛选出样品批次间差异成分。

结果:

建立了小儿热速清颗粒HPLC指纹图谱及7个成分的含量测定方法,且这2种方法学验证的结果均符合要求,确定了28个共有峰,并指认了11个色谱峰,15个批次相似度范围为0.995~0.999;HCA和PCA结果显示15个批次成分聚为2类,OPLS-DA分析筛选出13个造成样品批次间差异的标志物;多指标成分含量测定表明不同批次间小儿热速清颗粒中化学成分含量差异性较小。

结论:

建立的小儿热速清颗粒HPLC指纹图谱及7个成分含量测定方法的专属性强,准确、可靠,结合化学模式识别可有效的用于小儿热速清颗粒的质量控制。

Objective:

To study different batches of Xiaoer Resuqing granules by combining the method of fingerprint, multi-index component content determination and chemical pattern recognition analysis technology, and to provide the basis for its quality evaluation.

Methods:

The chromatographic SuperLu C18(250 mm×4.6 mm, 5 μm) column was adopted with mobile phase composed of methanol (A)-0.1% phosphoric acid (B) with gradient elution at a flow rate of 1.0 mL·min-1. The column temperature was 30 ℃, and the detection wavelength was 280 nm. Eleven characteristic components were identified, and the contents of seven active components, including puerarin, forsythiaside A, phillyrin, baicalin, wogonoside, baicalein and wogonin were determined. Hierarchical cluster analysis (HCA) and principal component analysis (PCA) were used to classify 15 batches of samples, and orthogonal least partial squares discriminant analysis (OPLS-DA) was performed. To screen out the difference components between batches of Xiaoer Resuqing granules.

Results:

The HPLC fingerprint of Xiaoer Resuqing granules and the content determination method of 7 components were established, the results of the two methods met the requirements. Twenty-eight common peaks were identified, 11 chromatographic peaks were identified, and the similarities of 15 batches of samples ranged from 0.995 to 0.999. HCA and PCA results showed that 15 batches were clustered into two categories, and OPLS-DA screened out 13 markers that caused the differences among the 15 batches of samples. There was little difference in the contents of ingredients.

Conclusion:

The established HPLC fingerprint of Xiaoer Resuqing granules and the determination method of 7 components are specific, accurate and reliable. Combined with chemical pattern recognition, it can be effectively used for the quality control of Xiaoer Resuqing granules.

我国儿童人群基数大,第七次全国人口普查数据显示,0~14岁人口占17.95%[1],儿童作为特殊人群,免疫系统等发育尚未完善,患病率高,因此,安全有效的儿童用药需求迫切,而药物质量是儿童用药安全的有力保障。2020年版《中华人民共和国药典》(简称《中国药典》)一部中收录中成药成方制剂和单味制剂1 457种,而儿科专用品种仅有60种[2],可见儿科专用品种缺乏,而现有的儿科专用品种的质量显得尤为重要。小儿热速清颗粒为儿科专用中成药之一,现收录于2020年版《中国药典》一部[2],其由金银花、柴胡、连翘、葛根、黄芩、大黄、板蓝根、水牛角8味中药组成,具有清热解毒、泻火利咽的功效,用于小儿外感风热所致感冒,症见高热、头痛、咽喉肿痛、鼻塞流涕、咳嗽、大便干结[2]。同时临床研究表明小儿热速清颗粒对儿童甲型流感病毒肺炎[3],小儿急性化脓性扁桃体炎[4],小儿急性上呼吸道感染伴发热[5],手足口病[6]等病症具有辅助治疗的效果。
中药制剂具有多组分、多靶点、多途径相互作用的特点[7],中药指纹图谱能够表征中药及制剂多成分的整体特性,是评价中药材及中药制剂质量的有效方法[8-9]。化学模式识别技术可以对指纹图谱中的信息进行表达、识别和处理[10]。将中药指纹图谱、多成分含量测定与化学模式识别技术结合,可以更加全面、系统地评价中药材以及中药制剂的质量。
产品批间质量稳定是临床等效性和有效性的重要前提[11],现行小儿热速清颗粒质量评价标准仅对黄芩苷、连翘苷2个化学成分进行控制,而多成分协同增效是中药复方发挥作用的特点,因此有必要建立小儿热速清颗粒多成分质量控制标准。小儿热速清颗粒尚无HPLC指纹图谱及化学模式识别相关研究,难以体现制剂整体质量[12-13]。因此,本研究采用高效液相色谱法(HPLC)建立15批小儿热速清颗粒指纹图谱及7个成分的含量测定,在指纹图谱相似度评价的基础上,同时结合聚类分析(HCA)、主成分分析(PCA)及正交最小偏二乘法-判别分析(OPLS-DA)等化学模式识别方法对其进行整体性评价,为全面控制小儿热速清颗粒批次间的稳定性提供了方法,为该制剂建立更系统、全面的质量控制标准提供借鉴与参考。

对照品:黄芩苷(批号21051301,质量分数98.42%)、汉黄芩苷(批号21102106,质量分数99.79%)、黄芩素(批号21090101,质量分数98.18%)、汉黄芩素(批号21051406,质量分数98.04%)、连翘酯苷A(批号21022605,质量分数98.61%)、葛根素(批号21061603,质量分数98.61%)、3-羟基葛根素(批号18102401,质量分数98.6%)、3-甲氧基葛根素(批号21022610,质量分数98.5%),均购于成都普菲生物科技有限公司;连翘苷(批号BCTG-0232,质量分数≥98%),购于江西本草天工科技有限公司;新绿原酸(批号RDD-XO1402112013,质量分数>98%)、异绿原酸B(批号RDD-Y06911801005,质量分数>98%),购于成都瑞芬思生物科技有限公司。
样品:小儿热速清颗粒,均由江西倍肯药业有限公司提供,共15批(规格:6 g·袋-1,批号分别为20210905、20210916、20210917、20210918、20210919、20210922、20210924、20210925、20211010、20211022、20211023、20211024、20211025、20211026、20211027,编号S1~S15)。
甲醇(色谱纯,湖北弗顿科学技术有限公司);甲醇、磷酸(分析纯,西陇科学股份有限公司);水为超纯水。
采用SuperLu C18(250 mm×4.6 mm,5 μm)色谱柱,以甲醇为流动相A,0.1%磷酸溶液为流动相B,梯度洗脱(0~25 min,15% A→37% A;25~38 min,37% A→45% A;38~43 min,45% A;43~56min,45% A→71% A;56~65min,71% A→90% A;65~75min,90% A),检测波长为280 nm,柱温为30 ℃,流速为1.0 mL·min-1,进样量为10 μL。
取样品适量,研细,取约3 g,精密称量,置具塞锥形瓶中,精密加入75%甲醇25 mL,密塞,称量,超声 (功率250 W,频率25 kHz) 处理30 min,放冷,再称量,用75%甲醇补足失去的量,摇匀,以12 000 r·min-1离心5 min,0.22 μm的微孔滤膜过滤,即得。
分别精密称取黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、连翘苷、连翘苷酯苷A、葛根素、3′-羟基葛根素、3′-甲氧基葛根素、新绿原酸、异绿原酸B的对照品适量,加甲醇配制成母液,精密吸取各母液适量,加50%甲醇制成每1 mL分别含以上成分20、30、10、30、50、50、20、30、30、20、20 μg的混合对照品溶液。
按照2020年版《中国药典》一部小儿热速清颗粒制法,分别制备缺金银花、连翘、柴胡、大黄、黄芩、葛根、板蓝根、水牛角的阴性颗粒。各阴性颗粒再按“2.2.1”项下方法制备,即得。
取“2.2”项下的混合对照品溶液及供试品溶液(S1),分别按“2.1”项下色谱条件进样测定,通过与色谱峰的保留时间及紫外吸收光谱图进行对比分析,见图1。结果指认出11个特征峰,分别是3号峰新绿原酸、7号峰3′-羟基葛根素、9号峰葛根素、10号峰3′-甲氧基葛根素、15号峰连翘酯苷A、16号峰异绿原酸、18号峰连翘苷、20号峰黄芩苷、24号峰汉黄芩苷、25号峰黄芩素、27号峰汉黄芩素。
取“2.2”项下的供试品溶液(S1)及各阴性供试品溶液,分别按“2.1”项下色谱条件进样测定,将供试品溶液的色谱峰与8个阴性供试品溶液的色谱峰进行重叠比较,结合色谱峰的保留时间以及紫外吸收光谱图,对指纹图谱的28个共有峰进行归属,见图2。结果指纹图谱中有26个特征峰可以在原药材中找到具体归属(其中2、19号峰来源大黄,3、16~17号峰来源金银花,4~5、14~15、18号峰来源连翘,6~12号峰来源葛根,20~28号峰来源黄芩;另外2个峰为多药材共有峰,其中1号峰为8味药材的共有峰,13号峰同时归属于葛根、连翘2味原药材)。
称取样品(S1)适量,按“2.2.1”项下方法制备供试品溶液,按“2.1”项下色谱条件于0、2、4、8、12、24 h后进样测定,以9号峰(葛根素)作为参照峰,结果测得各共有峰相对保留时间RSD均<0.27%,相对峰面积RSD均<2.8%,表明供试品溶液在24 h稳定。
取同一份样品(S1),按“2.2.1”项下方法制备供试品溶液,按“2.1”项下色谱条件连续进样测定6次,以9号峰(葛根素)作为参照峰,结果测得各共有峰相对保留时间RSD均<1.0%,相对峰面积RSD均<2.6%,表明仪器精密度良好。
称取样品(S1)适量,按“2.2.1”项下方法平行制备6份供试品溶液,按“2.1”项下色谱条件测定,以9号峰(葛根素)作为参照峰,结果测得各共有峰相对保留时间RSD均<0.12%,相对峰面积RSD均<2.9%,表明该方法重复性较好。
取15批样品,按“2.2”项下方法制备供试品溶液,按“2.1”项下色谱条件进样,得到15批样品的HPLC色谱图,导入“中药色谱指纹图谱相似度评价系统(2012.130723版本)”软件进行分析评价,设定样品S1的图谱为参照指纹图谱(R),时间窗宽度为0.1,采用中位数法,结合多点校正并进行Mark峰匹配,生成小儿热速清颗粒特征图谱(见图3)及15批样品HPLC叠加指纹图谱(见图4),共确定28个共有峰。其中9号峰(葛根素)峰面积较大,与相邻色谱峰分离效果好且保留时间稳定,可设为参照峰。结果测得28个共有峰的相对保留时间RSD均<0.21%,相对峰面积RSD在2.4%~53%,说明15批样品中28个共有峰的相对保留时间稳定,相对峰面积具有一定的差异。
将15批小儿热速清颗粒HPLC相关数据导入“中药色谱指纹图谱相似度评价系统(2012.130723版本)”软件,以样品S1色谱图作为对照图谱,计算相似度。由于黄芩苷色谱峰峰面积较大且峰高过高,与其他色谱峰相比差异太大,以致于其在指纹图谱相似度评价中占据了主导作用,使相似度结果均为1。为了不影响计算指纹图谱相似度的准确性,故考虑将黄芩苷色谱峰去除后重新计算相似度[11,14],得15批颗粒指纹图谱相似度分别为0.996、0.996、0.997、0.997、0.996、0.995、0.999、0.998、0.999、0.998、0.997、0.998、0.998、0.997、0.998,说明各批次间具有高度相似性,各批次颗粒工艺制备相对较稳定,质量一致性较好。
以15批样品中28个共有峰的相对峰面积为变量,导入SPSS 21软件,采用组间联接、平方欧式距离法进行系统聚类分析(HCA),见图5。当平方欧式距离介于10时,结果显示样品可聚为2类,即样品S1~S6聚为第1类,样品S7~S15聚为第2类。结果表明15批样品,在制备工艺相同的条件下,不同批次间28个共有峰的相对峰面积存在一定差异,且与相似度结果一致。
以15批小儿热速清颗粒指纹图谱的28个共有峰的峰面积为变量,导入SPSS 21.0统计软件进行主成分分析(PCA)。以特征值>1作为筛选标准,得到5个主成分因子,其累计方差贡献率为93.969%(表1),说明这5个主成分因子可代表小儿热清颗粒指纹图谱28个共有峰的基本特征与绝大部分信息。成分载荷矩阵反映了各主成分与28个共有峰即原始变量之间的相关系数,载荷的绝对值越大,对主成分的贡献越大[15-16],结果由表2可知,主成分1主要反映了峰2、4、5、7~10、13、15、17、19、21~27的信息,主成分2主要反映了峰1、3、6、11、12、14、16、18的信息,主成分3主要反映了峰20的信息,主成分4主要反映了峰1的信息,主成分5主要反映了峰28的信息。将5个主成分的得分分别记为Y1Y2Y3Y4Y5,以各主成分对应的贡献率作为权重系数,建立主成分综合得分模型:
得分结果见表3。得分反映各批次小儿热速清颗粒的质量情况,质量越好,则分数越高。综合得分较高的前7位分别为S11、S10、S14、S15、S8、S7、S12,其综合得分均>1,说明这7个批次的样品质量较好。将15批样品中的28个共有峰的峰面积导入SIMCA 14.1软件进行PCA处理,PCA得分图见图6。结果发现15批样品能分成2类,其中样品S1~S6分为第1类,样品S7~S15分为第2类,与HCA结果一致,指纹图谱相似度结果相符。进一步表明15批样品批次间质量稳定、可靠。
为进一步分析不同批次小儿热速清颗粒之间化学成分的差异,以15批样品中的28个共有峰的峰面积为变量,导入SIMCA 14.1软件进行正交最小偏二乘法-判别分析(OPLS-DA)处理。该模型稳定性参数R2Y=0.989,模型预测参数Q2=0.972,均>0.5,表明建立的OPLS-DA模型具有较高的稳定性与较好的预测能力[17]。为验证该模型没有过拟合现象,随机排列200次做置换检验,得置换检验图,见图7(A)R2回归线在Y轴截距为0.481、Q2回归线在Y轴截距为-0.896,均小于原始值,说明所建立的模型不存在过拟合现象[18],可用于判别分析15批样品的批次间差异。由图7(B)可知,根据OPLS-DA得分,15批样品可聚为2类,其中样品S1~S6聚为1类,S7~S15聚为另1类,与PCA和HCA结果一致,也与去除黄芩苷色谱峰后所得指纹图谱相似度结果相符。VIP能够反映每个共有峰的贡献程度,以VIP>1作为判定标准,筛选造成样品批次间差异的标志性成分[19-20]。结果筛选出13个色谱峰,见图7(C),按VIP大小排序分别为峰7(3′-羟基葛根素)、2、10(3′-甲氧基葛根素)、9(葛根素)、5、4、19、22、27(汉黄芩素)、12、17、21、23。即上述13个色谱峰是引起批次间差异的主要化学成分,即为潜在差异标志物。其中1个来源金银花,2个来源大黄,2个来源连翘,4个来源葛根,4个来源黄芩,进一步说明严格控制黄芩、葛根、大黄、连翘、金银花药材质量是确保小儿热速清颗粒批次间质量稳定的关键。在载荷散点图中,距离原点越远,权重值越大,说明该成分的变化对区分样品各批次间的作用越大[21],作为批次间质量差异性成分的可能性越大。15批样品共有峰载荷散点图见图7(D),结果显示上述13个色谱峰均离原点距离较远,与图7-C结果一致。提示这13个色谱峰的差异对样品分类的影响较大,这些成分可能是导致样品批次间差异的主要标志性成分。
供试品溶液、阴性供试品溶液的制备同“2.2”项方法;精密称取对照品葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素等适量,加50%甲醇配置成质量浓度分别为95.76、183.96、82.88、339.24、212.80、37.30、29.60 μg·mL-1的混合对照品储备液。
称取样品S1适量,取“2.5.1”项下的混合对照品储备液、供试品溶液以及阴性供试品溶液,按“2.1”项下色谱条件进样测定,记录色谱图,见图8。由结果可知,葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素与其相邻色谱峰的分离度均>1.5,且无阴性干扰。
取混合对照品储备液,配制系列标准曲线工作液,以质量浓度(μg·mL-1)为横坐标X,峰面积为纵坐标Y,进行线性回归,结果各成分线性关系良好,见表4
吸取混合对照品储备液适量,按“2.1”项下色谱条件进样测定6次,测得葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素峰面积的RSD在0.39%~2.3%,表明仪器精密度良好。
称取样品S1适量,按“2.2.1”项方法制备供试品溶液,按“2.1”项色谱条件于0、2、4、8、12、24 h后进样测定,测得葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素峰面积的RSD在0.37%~2.6%,表明在24 h内溶液稳定。
称取样品S1适量,按“2.2.1”项下方法平行制备6份供试品溶液,按“2.1”项下色谱条件测定,测得葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素含量的RSD在1.0%~2.8%,表明方法重复性好。
精密称取样品S1粉末1.5 g,共6份,分别加入适量对照品葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素,使加入量与1.5 g粉末中各成分的含量一致,按“2.2.1”项方法制备得到供试品溶液,按“2.1”项色谱条件测定,计算各成分的平均加样回收率。测得葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素的平均加样回收率在95.1%~101.7%,RSD在1.4%~2.6%,说明该方法加样回收率良好。
取15批样品,按“2.2.1”项方法制备供试品溶液,按“2.1”项色谱条件测定,计算葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素含量,结果见表5,不同批次样品中上述7个指标成分含量的RSD在4.2%~11.0%,表明15批样品7个成分含量批间差异性较小。
为了明确所测7个成分含量是否可作为指标性成分用以评价小儿热速清颗粒的质量,采用SPSS 21软件对15批样品中葛根素、连翘酯苷A、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素的总量和指纹图谱的综合得分进行相关性分析,结果表明二者的相关系数为0.746(双尾,P<0.01),说明所测7个成分可以作为指标性成分对小儿热速清颗粒进行质量评价。
中药以多成分作为物质基础发挥药效作用,建立多成分分析检测技术和方法是中药质量评价的有力手段。小儿热速清颗粒处方中黄芩主要有效成分为黄芩苷、黄芩素、汉黄芩苷、汉黄芩素等黄酮类化合物,具有解热抗炎、抗微生物、调节免疫等药理作用[22];连翘中含有的连翘苷、连翘酯苷A具有抗菌、抗病毒、抗炎、免疫调节等药理作用[23];金银花有“中药中的抗生素”之称,含有的新绿原酸、异绿原酸等酚酸类物质具有抗菌、抗病毒的药理活性[24];葛根中含有葛根素、3′-羟基葛根素、3′-甲氧基葛根素,这些异黄酮类成分具有解热镇痛的药理作用[25];在2020年版《中国药典》中,黄芩苷与连翘苷是小儿热速清颗粒的定量指标成分,因此本试验选择标定小儿热速清颗粒中新绿原酸、3′-羟基葛根素、葛根素、3′-甲氧基葛根素、连翘酯苷A、异绿原酸、连翘苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素,这11个成分协同发挥作用共奏清热解毒、泻火利咽的功效。因此,本文建立同时检测11个成分的方法,为更全面的控制小儿热速清颗粒的质量奠定基础。
中药指纹图谱是实现整体鉴别中药真实性、评价质量一致性和产品稳定性的可行模式,在中药质量控制领域被广泛应用。采用指纹图谱及多成分同测方法对15批小儿热速清颗粒质量进行分析,结果显示批间差异较小,质量较为稳定、均一。当前未见采用指纹图谱评价小儿热速清颗粒质量的相关研究,因此本文建立了小儿热速清颗粒的HPLC中药指纹图谱,共标定了28个共有峰,指认了其中11个色谱峰成分,结合化学模式识别技术对其进一步分析,实现对小儿热速清颗粒全面质量的综合评价,可用于小儿热速清颗粒批次间的一致性评价,为全面控制小儿热速清颗粒的质量提供参考依据。
