Objective:

To establish a chromatogram-effect correlated quality evaluation system for the antibacterial activity of Flos Lonicerae extract by data analysis of components.

Methods:

The best liquid chromatographic conditions were optimized by investigating the conditions of mobile phase, wavelength and flow rate, and the quantitative fingerprint of 50 batches of Flos Lonicerae extract was established. The best antibacterial concentration of the extract was obtained by microdilution method, and the antibacterial rates of 50 batches of samples were determined. Similarity analysis, cluster analysis, principal component analysis, grey correlation analysis and mathematical model establishment by support vector machine were used to analyze the quantitative fingerprints and antibacterial rates of 50 batches of samples.

Results:

Eighteen common peaks were selected from the 50 quantitative fingerprints of Flos Lonicerae extract, and the similarities were between 0.608-1. Six chemical components were identified (peak 4: neochlorogenic acid, peak 8: chlorogenic acid, peak 9: cryptochlorogenic acid, peak 16: isochlorogenic acid B, peak 17: isochlorogenic acid A and peak 18: isochlorogenic acid C). The average antibacterial rates of 50 batches of extracts were between 3.93%-70.50%. The results of principal component analysis and cluster analysis were highly consistent, and the results of grey correlation analysis showed that all components were positively correlated with antibacterial effect. The relative deviations between the predicted data of the mathematical model and the experimental data were all below 2%.

Conclusion:

The HPLC conditions of Flos Lonicerae extract are stable and reliable, and the quality evaluation system of antibacterial activity of Flos Lonicerae extract established can evaluate its quality based on antibacterial rate.

目的:

对金银花提取物的化学成分进行数据分析,建立谱-效相关金银花提取物抑菌作用质量评价系统。

方法:

通过考察流动相、波长、流速等条件,确定最佳液相色谱条件,建立50批金银花提取物的定量指纹图谱;通过微量稀释法得出金银花提取物的最佳抑菌浓度,并对50批样品进行抑菌测定,得出抑菌率;采用相似度分析、聚类分析、主成分分析、灰色关联度分析及通过支持向量机法建立数学模型等方法对50批定量指纹图谱及抑菌率结果进行分析。

结果:

建立的50批金银花提取物定量指纹图谱中选择了18个共有峰,相似度在0.608-1,确立了6个化学成分(峰4:新绿原酸、峰8:绿原酸、峰9:隐绿原酸、峰16:异绿原酸B、峰17:异绿原酸A、峰18:异绿原酸C),50批金银花药材的的抑菌率平均值在3.93%~70.50%。主成分分析与聚类分析的结果吻合度很高,灰色关联度分析结果所有成分均与抑菌作用呈正相关。数学模型的拟合数据与实验数据的相对偏差均在2%以下。

结论:

建立的金银花提取物液相色谱条件稳定可靠,建立的金银花提取物抑菌作用质量评价系统能够达到从抑菌率评价药物质量的目的。

目的:

对金银花提取物的化学成分进行数据分析,建立谱-效相关金银花提取物抑菌作用质量评价系统。

方法:

通过考察流动相、波长、流速等条件,确定最佳液相色谱条件,建立50批金银花提取物的定量指纹图谱;通过微量稀释法得出金银花提取物的最佳抑菌浓度,并对50批样品进行抑菌测定,得出抑菌率;采用相似度分析、聚类分析、主成分分析、灰色关联度分析及通过支持向量机法建立数学模型等方法对50批定量指纹图谱及抑菌率结果进行分析。

结果:

建立的50批金银花提取物定量指纹图谱中选择了18个共有峰,相似度在0.608-1,确立了6个化学成分(峰4:新绿原酸、峰8:绿原酸、峰9:隐绿原酸、峰16:异绿原酸B、峰17:异绿原酸A、峰18:异绿原酸C),50批金银花药材的的抑菌率平均值在3.93%~70.50%。主成分分析与聚类分析的结果吻合度很高,灰色关联度分析结果所有成分均与抑菌作用呈正相关。数学模型的拟合数据与实验数据的相对偏差均在2%以下。

结论:

建立的金银花提取物液相色谱条件稳定可靠,建立的金银花提取物抑菌作用质量评价系统能够达到从抑菌率评价药物质量的目的。

Objective:

To establish a chromatogram-effect correlated quality evaluation system for the antibacterial activity of Flos Lonicerae extract by data analysis of components.

Methods:

The best liquid chromatographic conditions were optimized by investigating the conditions of mobile phase, wavelength and flow rate, and the quantitative fingerprint of 50 batches of Flos Lonicerae extract was established. The best antibacterial concentration of the extract was obtained by microdilution method, and the antibacterial rates of 50 batches of samples were determined. Similarity analysis, cluster analysis, principal component analysis, grey correlation analysis and mathematical model establishment by support vector machine were used to analyze the quantitative fingerprints and antibacterial rates of 50 batches of samples.

Results:

Eighteen common peaks were selected from the 50 quantitative fingerprints of Flos Lonicerae extract, and the similarities were between 0.608-1. Six chemical components were identified (peak 4: neochlorogenic acid, peak 8: chlorogenic acid, peak 9: cryptochlorogenic acid, peak 16: isochlorogenic acid B, peak 17: isochlorogenic acid A and peak 18: isochlorogenic acid C). The average antibacterial rates of 50 batches of extracts were between 3.93%-70.50%. The results of principal component analysis and cluster analysis were highly consistent, and the results of grey correlation analysis showed that all components were positively correlated with antibacterial effect. The relative deviations between the predicted data of the mathematical model and the experimental data were all below 2%.

Conclusion:

The HPLC conditions of Flos Lonicerae extract are stable and reliable, and the quality evaluation system of antibacterial activity of Flos Lonicerae extract established can evaluate its quality based on antibacterial rate.

金银花来源于忍冬科植物忍冬(Lonicera japonica Thunb)的干燥花蕾或带初开的花[1],具有疏散风热、清热解毒的作用,主要治疗风热感冒、热毒血痢、丹毒等症[2-4],黄酮类化合物、有机酸类、环烯醚萜类、三萜皂苷类及挥发油类等成分[5-7]是金银花药材中的典型有效成分,具有解热抗炎、抗菌、抗病毒、抗紫外、抗氧化、增强免疫力[8-10]等作用。目前金银花提取物用途广泛,是中药制剂常用的一种成分,然而市面上金银花提取物的质量确参差不齐,为了能够更好地选择高质量的金银花提取物,本文通过建立金银花提取物的定量指纹图谱,采用微量稀释法测定50批金银花提取物样品抑菌率;采用相似度分析、聚类分析、主成分分析、灰色关联度分析及通过支持向量机法建立数学模型等方法[11-15]对50批金银花提取物定量指纹图谱及抑菌率结果进行分析。并以抑菌率为药效指标建立了一套能够评价其质量的数学模型,为更好的应用于临床提供参考。
Waters e2695高效液相色谱仪,配有Waters 2998紫外检测器(Waters公司),Milli-Q超纯水系统(密理博公司),KDM-A数显恒温电热套(山东博科再生医学有限公司),酶标仪(BIO-RAD公司),CP225D十万分之一电子天平(Sartorius公司),HWS智能型恒温恒湿培养箱(宁波江南仪器厂),LDZF-75L-I立式高压蒸汽灭菌器(上海习仁科学仪器有限公司)。
乙腈(Honeywell公司,色谱纯);磷酸(天津市科密欧化学试剂有限公司,色谱纯);水为超纯水。
胰酪大豆胨琼脂培养基(上海源培生物);胰酪大豆胨液体培养基(上海源培生物);抑菌实验用水均为蒸馏水。
葛根素对照品(中国食品药品检定研究院,批号110752-202217);阳性药物:阿莫西林胶囊(石药控股集团有限公司,批号400170311)。
从全国各地收集金银花药材50批,由山东省中医药研究院林慧彬教授鉴定鉴定,均为忍冬科植物忍冬(Lonicera japonica Thunb)的干燥花蕾。照2020年版《中华人民共和国药典》一部中金银花提取物质量标准项下【制法】制备金银花提取物50批。
取制备好的金银花提取物约0.2 g,精密称定后置于圆底烧瓶中,精密加入100 mL蒸馏水,称量,回流提取30 min,放冷至室温后用蒸馏水补足称量,过滤,取续滤液与内标溶液1∶1混匀,即得。
采用CAPCELL PAK C18(250 mm×4.6 mm,5 μm)和 Venusil MP C18(250 mm×4.6 mm,3 μm)色谱柱,流动相为乙腈-0.3%磷酸,梯度洗脱(见表1),流速为0.7 mL·min-1,检测波长为235 nm。
对金银花提取物进行方法学考察(精密度、稳定性、重复性),以葛根素为内标峰采集指纹图谱,计算相对峰面积值和相对保留时间值,结果显示各共有峰的相对峰面积RSD<2%,相对保留时间RSD<2%,表明仪器精密度良好,金银花提取物供试品在30 h内稳定性且重复性均良好。
按照“2.1.3”方法,对50批金银花提取物进行测定并确定其指纹图谱共有峰,记录指纹图谱峰面积值及保留时间,按照以下转换公式:金银花提取物共有峰相对峰面积值=(金银花提取物共有峰峰面积/供试品溶液浓度)/(内标峰峰面积/内标浓度)×抑菌浓度,将各成分相对峰面积值转换成抑菌浓度下的相对峰面积值,以便与抑菌作用建立联系。通过对照品比对以及LCMS-8050液-质联用仪(岛津公司)对金银花提取物的共有峰成分进行鉴别研究,确定6种化学成分,见图1
随机取10批金银花提取物水提液,过0.22 μm微孔滤膜,经灭菌后稀释6个浓度(1∶32,1∶16,1∶8,1∶4,1∶2,1∶1)采用96孔板法,每个浓度平行4次,每孔加入液体培养基50 μL、金银花提取物水提液50 μL和金葡菌液10 μL(浓度为1×10 5 cfu·mL-1),将各组置于37 ℃的恒温恒湿培养箱内培养24 h,采用酶标仪在波长570 nm下分别测定培养前及培养后的吸收度(A),最终得出金银花提取物最佳抑菌浓度为40 μg·mL-1。在此浓度下考察50批金银花提取物的抑菌效果。结果见表2

采用《中药色谱指纹图谱相似度评价系统(2004 A版)》中位数法对50批金银花提取物定量指纹图谱进行分析,并生成标准指纹图谱R,计算各批次样品指纹图谱与对照指纹图谱的相似度,结果表明,金银花提取物各批次间的相似度在0.608~1,由此可以看出金银花提取物各批次之间差异较大,50批相似度指纹图谱见图2
采用SPSS 19.0软件对50批金银花提取物抑菌浓度下的共有峰相对峰面积进行聚类分析,见图3
采用SPSS 19.0对50批金银花提取物的18个变量进行主成分分析。按照以下步骤对各批次进行综合评价。
采用SPSS 19.0软件,对标准化数据进行相关检验,结果KMO统计量=0.682>0.5,球形检验卡方统计量=1 121.095,单侧P=0.000<0.01,说明样本适宜进行主成分分析。
图4碎石图可见图中曲线存在1个明显的拐点,拐点前4个点代表前4个主成分,说明前4个主成分的分析结果基本显示出了不同批次金银花提取物之间的差异性。提取前4个主成分,进行主成分分析,分析结果见表3
表3可知,特征值大于1的成分有4个,累积贡献率高达82.946%。第1主成分的贡献共有峰包含3、4(新绿原酸)、7、8(绿原酸)、9(隐绿原酸)、10、11、13、14、16(异绿原酸C),第2主成分的贡献共有峰包含1、6、12,第3主成分的贡献共有峰包含2、3、5、15,第4主成分的贡献共有峰包含8(隐绿原酸)、17(异绿原酸A)、18(异绿原酸C)。计算各主成分F得分,F得分越高,表明样品质量越好。结果见表4
在多元统计分析中,由于变量复杂多样,所以一般都需要采用2种或2种以上的多元统计学方法对变量进行分析,以增加结论的可信度。主成分分析虽然比较直观,但是无法对各对象所属类别进行准确划分,因此进一步通过聚类分析消除类别划分的随意性。由聚类分析结果可知,可将金银花提取物分为2个大类。样品37单独为Ⅰ类,其余样品为Ⅱ类。Ⅱ类中可分为3小类,其中12、31、34、2为一类,40为一类,19、29、18、43、16、28、23、17、21、8、14、15、24、25、26、27、30、20、39为一类。除个别批次外,大部分样品与主成分分析中的分析结果一致。
利用灰色关联分析法对金银花提取物共有峰的指纹图谱数据和抑菌率进行分析,通过计算母序列(抑菌率)与子序列(共有峰的指纹图谱数据)的关联程度来体现金银花提取物中化学成分对金葡菌的抑菌作用强弱。通过确定系统特征、无量纲化处理等步骤[11]后,按以下公式计算Yi(k),(i=1,2,…,18)与Y0的关联系数与关联度及关联极性。

如果signi)=sign0),则YiY0呈正相关,说明YiY0是增强作用;如果signi)=sign0),说明YiY0负相关,即YiY0是削弱作用,i=1,2,…,18。这里sign(x)为符号函数,即金银花提取物各化学成分对金葡菌抑菌作用灰色关联分析见表5
若关联度大于0.9,则子序列对母序列起显著作用;若关联度在0.8~0.9,表示有相对显著作用;若关联度在0.7~0.8,表示有明显作用;若关联度在0.6~0.7,表示有较小作用;若关联度小于0.6,表示有非常小的作用。
根据表6可以将金银花提取物各化学成分抑菌作用由强到弱排列:5>8(绿原酸)>18(异绿原酸C)>17(异绿原酸A)>7>16(异绿原酸B)>9(隐绿原酸)>11>10>4(新绿原酸)>1>13>14>15>2>6>12>3。其中,成分5、8(隐绿原酸)、18(异绿原酸C)、17(异绿原酸A)、7(绿原酸)、16(异绿原酸B)、9、11、10、4(新绿原酸)、1、13、14、15、2有显著抑菌作用;成分6、12、3有相对显著抑菌作用。由关联极性可知,所有化学成分均有抑菌作用。
通过LS-SVM Lab工具采用最小二乘支持向量机法随机对金银花提取物的47组数据(谱-效药效指标值)进行数学建模,当γ=1 309.26,σ2=1.68时,其拟合与预测效果最好。结果见表6图5
由上述图表可以看出,该数学模型拟合数据与实验数据的相对偏差均在1%以下,平均相对偏差低至0.036%,说明数学模型的拟合值与实验数据的吻合度很好。
为了验证金银花提取物数学模型是否准确,将其他3组金银花提取物指纹图谱数据代入建立的数学模型中,计算出预测值抑菌率,再将此数据与实验值抑菌率进行比较,结果见表7
表7可知,该模型的实验数据与预测数据的平均相对偏差在2%以下。由此说明,建立的金银花提取物指纹图谱-抑菌药效指标数学模型,预测值与实验值误差很小,因此可以通过输入指纹图谱数据对金银花提取物的抑菌作用进行准确预测。
本文通过自制金银花提取物,建立高效液相定量指纹图谱检测方法,采用微量稀释法测定金银花提取物的抑菌作用,利用灰色关联分析及最小二乘支持向量机法对谱(指纹图谱数据)-效(抑菌率)数据进行关联分析,结果显示,金银花提取物的18个共有成分均与抑菌率呈正相关,建立的数学模型的拟合数据与实验数据的相对偏差均在2%以下,能够通过将多种化学成分与药效相结合的方式实现仅输入指纹图谱数据就能预测金银花提取物质量(抑菌率)的目的,此外,若不同批次的金银花药材提取条件相同,宜可采用此评价系统通过金银花提取物的抑菌率大小判断金银花药材的优劣,实现从药材源头及提取物两个环节对质量的控制。金银花提取物谱-效相关质量评价系统的建立,可为金银花在抑菌作用上的药效成分有更深入的认知,不仅可全面、客观、科学地评价金银花提取物的质量优劣,也可以为含金银花提取物的成方制剂提供相应的质量评价手段,为丰富中药质量评价模式,提高质量评价的现代化、科学化水平提供相应的技术支撑。因此,建立的金银花提取物抑菌作用谱-效相关质量评价系统相比较于传统的仅由个别成分、多个指标及指纹图谱评价药材优劣更加全面、客观、有说服力。本文建立的评价系统仅从抑菌方面评价药物优劣,药效指标单一,制定更全面更科学的药效指标将是我们后期研究的重点。
