Objective:

To establish a method for the rapid identification of the authenticity of Gastrodiae Rhizoma herbs based on enzymatic recombinase amplification (ERA) technique.

Methods:

Following the principle of ERA primer design, Oligo 7.0 software was applied to screen and optimize the ERA-specific primers of Tianma based on the ITS2 genome sequences of Gastrodia elata and its common artifacts. Primer Premier 5.0 software was applied to design the specific PCR primers for the identification of Gastrodia elata, and through the optimization of ERA and PCR reaction system, the optimal reaction time for ERA was finally determined to be 17 min, the optimal reaction temperature was 40 ℃, the optimal annealing temperature for PCR was 57 ℃, and the cycle was 32 times, and the established method was verified for sensitivity and specificity, and the samples of asparagus available in the traditional Chinese medicine The sensitivity and specificity of the established method were verified, and the commercially available asparagus samples in the market were selected for testing.

Results:

The designed primers for the specific identification of ERA in Gastrodia elata did not cross-react with its common forgeries, and the specificity was good, a repeatability results showed that the three repeatability tests were consistent, with no false positives or false negatives;the sensitivity of this method for Gastrodia elata genomic DNA was 1 pg·μL-1, which was higher than that of conventional PCR;the ERA technique can be used for the rapid identification of commercially available samples of Gastrodia elata and the results of the assay are the same as those of the PCR method.

Conclusion:

The established detection method is simple, rapid, with high specificity and sensitivity, and provides a new means for the authentication of the Gastrodiae Rhizoma.

目的:

基于酶促恒温扩增(enzymatic recombinase amplification,ERA)技术建立一种可快速定性鉴别道地药材天麻真伪的方法。

方法:

遵循ERA引物设计原则,应用Oligo 7.0软件根据天麻及其常见伪品的ITS2基因组序列,筛选优化天麻ERA特异性引物,应用Primer Premier 5.0软件设计天麻特异性PCR鉴别引物,通过对ERA、PCR反应体系的优化,最终确定ERA最佳反应时间为17 min,最适反应温度为40 ℃;PCR最优退火温度为57 ℃,循环为32次,对所建立的方法进行灵敏度、特异性验证,并选取中药市场市售天麻样品进行检测。

结果:

所设计的天麻ERA鉴别引物与其常见伪品间无交叉反应,特异性良好;重复性结果显示,3次重复性检测结果一致,未出现假阳性与假阴性;该方法对天麻基因组DNA的灵敏度为1 pg·μL-1,比传统PCR灵敏性高;ERA技术能够用于天麻市售样品的快速鉴定,且检测结果与PCR方法相同。

结论:

所建立的检测方法操作简单、快速,具有较高的特异性和灵敏度,为道地药材天麻的真伪鉴别提供了一种新手段。

目的:

基于酶促恒温扩增(enzymatic recombinase amplification,ERA)技术建立一种可快速定性鉴别道地药材天麻真伪的方法。

方法:

遵循ERA引物设计原则,应用Oligo 7.0软件根据天麻及其常见伪品的ITS2基因组序列,筛选优化天麻ERA特异性引物,应用Primer Premier 5.0软件设计天麻特异性PCR鉴别引物,通过对ERA、PCR反应体系的优化,最终确定ERA最佳反应时间为17 min,最适反应温度为40 ℃;PCR最优退火温度为57 ℃,循环为32次,对所建立的方法进行灵敏度、特异性验证,并选取中药市场市售天麻样品进行检测。

结果:

所设计的天麻ERA鉴别引物与其常见伪品间无交叉反应,特异性良好;重复性结果显示,3次重复性检测结果一致,未出现假阳性与假阴性;该方法对天麻基因组DNA的灵敏度为1 pg·μL-1,比传统PCR灵敏性高;ERA技术能够用于天麻市售样品的快速鉴定,且检测结果与PCR方法相同。

结论:

所建立的检测方法操作简单、快速,具有较高的特异性和灵敏度,为道地药材天麻的真伪鉴别提供了一种新手段。

Objective:

To establish a method for the rapid identification of the authenticity of Gastrodiae Rhizoma herbs based on enzymatic recombinase amplification (ERA) technique.

Methods:

Following the principle of ERA primer design, Oligo 7.0 software was applied to screen and optimize the ERA-specific primers of Tianma based on the ITS2 genome sequences of Gastrodia elata and its common artifacts. Primer Premier 5.0 software was applied to design the specific PCR primers for the identification of Gastrodia elata, and through the optimization of ERA and PCR reaction system, the optimal reaction time for ERA was finally determined to be 17 min, the optimal reaction temperature was 40 ℃, the optimal annealing temperature for PCR was 57 ℃, and the cycle was 32 times, and the established method was verified for sensitivity and specificity, and the samples of asparagus available in the traditional Chinese medicine The sensitivity and specificity of the established method were verified, and the commercially available asparagus samples in the market were selected for testing.

Results:

The designed primers for the specific identification of ERA in Gastrodia elata did not cross-react with its common forgeries, and the specificity was good, a repeatability results showed that the three repeatability tests were consistent, with no false positives or false negatives;the sensitivity of this method for Gastrodia elata genomic DNA was 1 pg·μL-1, which was higher than that of conventional PCR;the ERA technique can be used for the rapid identification of commercially available samples of Gastrodia elata and the results of the assay are the same as those of the PCR method.

Conclusion:

The established detection method is simple, rapid, with high specificity and sensitivity, and provides a new means for the authentication of the Gastrodiae Rhizoma.

天麻(Gastrodiae Rhizoma)是我国常用的名贵中药材,为兰科植物天麻(Gastrodia elata Bl.)的干燥块茎,有平抑肝阳、息风止痉的功效,临床上对抗惊厥、镇痛、保护神经元系统[1-4]等具有重要意义。天麻在2021年被评为吉林省首批道地药材的优势品种,因其药用保健价值较高,得到社会各界的广泛认可,价格逐年升高,不法商人为谋取利润常以价格低廉的天花粉(Trichosanthis Radix)、紫茉莉根(Mirabills jalapa L.)以及大丽菊(Dahlia pinnata Cav.)的块茎等来冒充天麻,严重危害公众健康。吉林省道地药材天麻出售量大,保证中药材的质量安全问题关乎到民生。目前,对于天麻真伪鉴定的研究不再局限于性状、显微、理化鉴别,随着科技的发展,天麻水提物HPLC指纹图谱[5]、稳定同位素技术[6]、非线性化学指纹图谱[7]以及DNA条形码技术[8]等都应用于天麻的鉴定中,但这些技术方法需要专业人员及昂贵的仪器设备,在一定程度上限制了推广应用。
近几年,等温扩增技术(isothermal amplification technology,IAT)发展迅速,以环介导等温扩增(loop-mediated isothermal amplification,LAMP)技术、重组酶聚合酶扩增(recombinase polymerase amplification,RPA)技术、重组酶介导链替换核酸扩增(recombinase aided amplification,RAA)技术和酶促恒温扩增(enzymatic recombinase amplification,ERA)技术为代表的IAT在食品[9-10]、中药[11]、病原[12-13]鉴定等方面得到广泛应用。
其中,国产RAA试剂盒、ERA试剂盒是进口RPA试剂盒的替代品,ERA技术是我国于继彬等[14]在2019年研发的具有全球自主知识产权的全新的IAT方法,是对Piepenburg等[15]研发的RPA技术的改造和升级。目前,该技术方法已申请了国家专利和国际专利。ERA技术与RPA检测原理相似,是在重组酶、单链DNA结合蛋白以及DNA聚合酶的作用下,在37~42 ℃恒温条件下,10~20 min将靶DNA特异性、指数级的扩增,相较于LAMP需要设计4条引物的烦琐性,ERA技术只需要设计2条引物的简便优势显的尤为突出。目前尚没有见到将ERA技术应用于天麻真伪鉴定的研究,为此,本研究以天麻ITS2序列为靶基因,应用ERA技术初步建立一种可快速、定性鉴别天麻真伪的方法。
UV WHITE2020D凝胶成像分析系统(Bio-rad公司),NanoDrop One微量核酸蛋白测定仪(Thermo公司),JY300E通用型电泳仪(北京君意东方电泳设备有限公司),台式高速冷冻离心机(长沙湘仪离心机仪器有限公司),Q6000微量紫外分光光度计(DeNovix公司),K30系列干式恒温器(杭州奥盛仪器有限公司),Digital Vortex Mixer (Thermo Fisher公司)。
天麻对照药材(中国食品药品检定研究院,批号120944-202112)、紫茉莉根对照药材(乐美天医药|德思特生物,批号DST201211-072)、大丽菊块茎(吉林国安药业有限公司)、天花粉(安徽亳州药材市场),所有样品均经吉林国安药业有限公司曲永梅中药师鉴定。编号S1-S15为市售天麻样品,用于评价中药市场天麻的合格率。
基础型核酸扩增试剂盒KS101(苏州先达基因科技有限公司)、核酸染料(SuperRed公司)、100 bp DNA Marker(上海生物工程有限公司)、2×Taq PCR Master Mix酶(南京诺唯赞生物科技股份有限公司)、琼脂糖(西班牙Agrose公司)。
采用改良CTAB法提取植物基因组DNA,分别取各样品100 mg经液氮研磨呈粉末状,放置于2 mL离心管中,加入P1溶液(洗脱缓冲液)1 000 μL,P2溶液(β-巯基乙醇)3 μL,13 000 r·min-1离心6 min,弃上清;加入65 ℃预热的P3溶液(4×CTAB)750 μL,P2溶液(β-巯基乙醇)2.25 μL,涡旋震荡混匀,封口膜封口后65 ℃水浴震荡1 h。冷却至室温,加入P4[三氯甲烷-异戊醇(24∶1)]溶液750 μL,颠倒混匀,使溶液呈乳浊状,不可震荡,12 000 r·min-1离心5 min;取上清,加入等体积P5溶液(异丙醇),于-20 ℃沉淀30 min,12 000 r·min-1离心5 min,弃去上清;加入P6溶液(70%乙醇)600 μL,13 000 r·min-1离心5 min,弃上清(重复此步骤1次),室温晾干;加入P7溶液(ddH2O) 30 μL,即得样品的DNA溶液。
将样品的DNA溶液用微量紫外分光光度计测定浓度,每个溶液重复测定3次,取测量结果的平均值,根据A260 nmA280 nm的比值鉴定纯度。结果如表1所示,样品纯度均在1.70~2.10,符合扩增要求。
从NCBI数据库GenBank中下载天麻(EU135912.1)及其常见伪品的ITS2基因组序列进行多序列比对分析,找到差异较大的区域,参考苏州先达基因有限公司ERA引物与探针设计指南,应用Oligo 7.0软件设计出6对天麻特异性ERA鉴别引物,供后续试验筛选优化;根据天麻DNA分子标记序列,应用Primer Premier 5.0软件设计1对天麻PCR特异性引物,引物序列均由生工生物工程(长春)股份有限公司合成,所设计的引物序列及预期产物长度如表2所示。
通过优化PCR反应体系和条件,确定最终PCR反应体系为25 μL,其中2×Taq PCR Master Mix 12.5 μL,上、下游引物(10 μmol·L-1)各1 μL,检测时将天麻对照药材与伪品的DNA溶液(100 ng·μL-1)1 μL加入各自反应管,灭菌ddH2O补足至25 μL。另取灭菌ddH2O作为阴性对照。PCR反应程序:95 ℃预变性5 min,循环反应32次(95 ℃,30 s;57 ℃,30 s;72 ℃,30 s),72 ℃延伸5 min,4 ℃保存。反应结束后取电泳上样量6 μL,DNA分子标记上样量4 μL点样于2%琼脂糖凝胶电泳板上,120 V·cm-1凝胶电泳40 min,电泳结束后将凝胶置于紫外分析仪上观察结果。结果如图1所示,所设计的天麻特异性PCR引物在178 bp处出现单一明亮条带,而其常见伪品没有条带,表明该PCR引物具有良好特异性。
根据基础型核酸扩增试剂盒KS101说明书配制48 μL预混液,其预混液中包括溶解剂20 μL,10 μmol·L-1的上、下游引物(F1/R1)各2 μL,10 ng·μL-1的样品DNA溶液1 μL,无菌ddH2O补足至48 μL,将预混液充分涡旋振荡混匀,6 000 r·min-1离心3 s;将预混液加入到基础扩增试剂中,以3 000 r·min-1的速度混匀3~5 s,直到扩增试剂重悬,再次6 000 r·min-1离心3 s。将激活剂2 μL加在反应管管盖上,6 000 r·min-1短暂离心3 s,使激活剂进入预混液中,3 000 r·min-1混匀3 s,再次以6 000 r·min-1的速度离心3 s。将反应管放入恒温仪中,40 ℃孵育17 min,反应结束后立即终止反应,取ERA扩增产物溶液10 μL加Loading Buffer 1.5 μL于60 ℃孵育5 min。
称取0.8 g琼脂糖粉末置于锥形瓶中,加入1×TBE 40 mL,在微波炉中中火加热4 min溶解,加入核酸染料GelRed 4 μL,并用腕力摇匀,得2%琼脂糖凝胶。取电泳上样量6 μL,DNA分子标记上样量4 μL点样于琼脂糖凝胶电泳板上,电压120 V,当蓝色指示剂距点样孔2~3 cm时结束电泳,将凝胶置于紫外分析仪上观察结果。
分别以10 ng·μL-1的天麻对照药材、紫茉莉根对照药材、天花粉、大丽菊块茎的DNA溶液作为模板,同时以无菌ddH2O作为阴性对照,采用“2.5”项下的反应体系和反应条件进行恒温扩增,反应结束后采用“2.6”项下方法观察结果,验证引物特异性。结果如图2所示,只有1泳道出现大小为180 bp的目的条带,伪品及阴性对照未出现目的条带,表明F1/R1这对引物在该检测体系中特异性良好。
以10 ng·μL-1的天麻对照药材、紫茉莉根对照药材、天花粉、大丽菊块茎的DNA溶液以及无菌ddH2O作为模板,采用“2.5”项下检测体系,由同一操作人员在相同的检测体系与反应条件下,在同一实验环境中的不同时间段重复试验3次,反应结束后采用“2.6”项下方法观察结果。结果如图3所示,经3次重复性试验,天麻对照药材均在180 bp处出现目的条带,而伪品及阴性对照无目的条带,未出现假阴性及假阳性,且3次试验结果一致,表明该引物具有良好的重复性。
将天麻对照药材基因组DNA溶液按1∶10进行倍比稀释,分别稀释到100 ng·μL-1、10 ng·μL-1、1 ng·μL-1、100 pg·μL-1、10 pg·μL-1、1 pg·μL-1、100 fg·μL-1、10 fg·μL-1,以稀释的DNA溶液作为模板,并同时采用试验中所建立的ERA、PCR检测体系进行平行测定,比较2种检测方法的灵敏度。由于ERA试剂盒具有较高的灵敏度,故本试验将ERA灵敏度从10 ng·μL-1开始进行检测,PCR灵敏度从100 ng·μL-1开始进行检测。
图4-A可见,在第5泳道可以看到清晰目的条带,第6泳道时有目的条带但不清晰,由此确定ERA的灵敏度为1 pg·μL-1。由图4-B可见,在第3泳道可见清晰目的条带,第4泳道时未出现目的条带,故确定PCR的灵敏度为1 ng·μL-1,表明ERA的灵敏度优于PCR。
按照“2.1”项下方法提取购于中药市场的市售天麻样品DNA,微量核酸蛋白仪测定DNA溶液浓度,并将市售样品DNA溶液分别用灭菌ddH2O稀释至100 ng·μL-1和10 ng·μL-1,按照“2.4”项和“2.5”项下检测体系进行扩增检测,比较2种方法检测结果是否一致。结果如图5所示,ERA检测方法在180 bp处出现单一明亮条带的为正品,而PCR检测方法在178 bp处出现单一明亮条带的为正品,无扩增条带的为伪品以及阴性对照,2种方法所得的检测结果一致。
根据所设计的6对ERA引物进行组合筛选,分别建立不同的扩增体系,采用“2.5”项下检测体系进行扩增,选取能扩增出单一明亮条带的引物进行试验摸索和特异性验证。结果图6所示,引物F1/R2未出现目的条带且出现非特异性扩增,F2/R2出现目的条带,但也出现非特异性扩增,故舍弃这2对引物,而引物F1/R1、F2/R1、F3/R3、F3/R4均出现1条单一目的条带,由于引物F1/R1扩增效率较高,条带更为明亮,故本试验优先选择扩增产物长度为180 bp的引物即F1/R1进行后续试验验证。
将天麻对照药材DNA溶液浓度稀释至10 ng·μL-1,采用“2.5”项下检测体系,将反应管分别至于33、37、39、40、41、43、45 ℃的恒温仪中恒温孵育,反应结束后立即终止反应,并取ERA扩增产物溶液10 μL加入试剂盒中自带的Loading Buffer 1.5 μL于60 ℃孵育5 min。反应结束后采用“2.6”项下方法观察结果,选择最适反应温度。
图7所示,在37~45 ℃下均扩增出目的条带,1~3泳道由于温度较低,导致ERA扩增效率不高;4、5泳道扩增产物相对更稳定,条带单一明亮,但在43 ℃以后,随着温度的升高扩增效率逐渐降低,所以确定天麻ERA最适反应温度为40 ℃。
采用“2.5”项下反应体系,分别在扩增时间为10、15、17、19、20 min时取出反应管,将反应管放置于冰盒中终止反应,取ERA扩增产物溶液10 μL加Loading Buffer 1.5 μL于60 ℃孵育5 min。反应结束后采用“2.6”项下方法观察结果,选择最佳反应时间。
图8所示,在反应时间为10 min时,可见目的条带但不清晰,在15 min以后,随着时间的延长ERA产物扩增效率变化不大,在第3泳道时条带更为明亮,故确定最佳反应时间为17 min。
采用“2.5”项下检测体系,在说明书推荐的引物量范围内,设置1.6、1.8、2.0、2.2、2.4、2.5 μL共6个引物变量,将反应管置于恒温仪中孵育,反应结束后取ERA扩增产物溶液10 μL加入Loading Buffer 1.5 μL于60 ℃孵育5 min。采用“2.6”项下方法观察结果,以确定体系的最佳引物量。结果如图9所示,随着引物量的升高扩增效率无太大变化,但在第3泳道时条带更为明亮,故确定最佳引物量为2.0 μL。
天麻又名赤箭、合离草、水洋芋等,无根无叶,常以地下块茎入药,市场上的天麻混伪品种类繁多,伪品的掺入严重贻误了疾病治疗。ERA因其具备进口RPA所具有的鉴定优势且成本较低,因而备受关注。本研究基于天麻及其常见伪品的ITS2基因组序列设计多对ERA鉴别引物,由于目前尚没有专门的软件用于ERA的引物设计[16-17],所以需要经过大量的试验进行引物筛选优化,最终找到特异性较高的F1/R1这对引物。通过对ERA反应体系的优化,最终确定该体系的最适反应温度为40 ℃,最佳反应时间为17 min,与试剂盒推荐的温度、时间稍有不同,但相对于普通PCR,ERA技术使检测时间大幅缩短。
本研究基于PCR、ERA 2种方法平行检测,特异性结果表明,所设计的天麻PCR、ERA引物与常见伪品紫茉莉根、天花粉、大丽菊块茎间无交叉反应,这2种方法均表现出良好的特异性。敏感性结果表明,ERA检测灵敏度比PCR高1 000倍,可达pg级。通过这2种检测方法分别对市售的15份天麻样品进行检测,其中样品3、7、12未出现目的条带,判定为伪品,2种检测方法均与吉林国安药业有限公司质量研发部曲永梅中药师鉴定结果完全一致,表明所建立的ERA方法准确率较高。本试验应用的是基础型ERA核酸扩增试剂盒,需要通过琼脂糖凝胶电泳观察结果,由于ERA是一类多酶反应体系,在进行核酸电泳观察结果时需要进行纯化,否则会堵塞胶孔,导致核酸无法跑出。本试验使用的是试剂盒中自带的Loading Buffer进行纯化处理,效果略次于试剂盒法的纯化。由于所设计的PCR与ERA产物片段大小相近,只相差2 bp,在结合终点法琼脂糖凝胶电泳定性观察检测结果时,根据目的条带出现的位置并不能将2者明显的区分开,但PCR与ERA是2种不同的检测方法,本试验中ERA的扩增产物会在目的条带的下方出现弥散条带,而PCR的产物条带单一,根据这一特征可以确定ERA与PCR产物。试剂盒的基础扩增试剂是八连管的设计,在混匀离心的过程中,如未紧密扣严,易产生污染,这也是该技术的弊端,所以在使用ERA技术结合终点法进行检测时,必须遵循严格的污染控制措施,在打开反应管盖时务必对扩增产物做降温处理,平衡内外大气压差以降低气溶胶污染引起的假阳性风险。为了避免交叉污染,建议将试剂配置区与扩增分析区分隔开,本试验过程中就有一伪品曾多次出现假阳性的情况。当试验环境已存在污染,建议多次使用消毒液或者75%的酒精对试验区域进行彻底消毒,同时使用新的扩增试剂。
综上,本试验基于ERA技术初步建立了一种可快速、定性鉴别天麻真伪的方法,该技术不需要昂贵的仪器设备,且操作程序简单。后续考虑设计引物探针,将ERA技术与侧向流试纸条技术相结合,实现偏远地区现场的快速检测,或应用RT-ERA方法来规避气溶胶污染的问题,继续为道地药材天麻的真伪鉴别提供新的技术手段。
