The change of analytical procedure, a part of the entire lifecycle of a method, is increasingly valued with the emergence of new technologies, the increasing demand for product quality, 3R, environmental protection, and cost reduction requirements. This paper refers to the latest progress in the study of counterpart law at home and abroad, systematically classifies changes of analytical procedures from the perspective of methodology, and discusses the differences between different types of changes and the evaluation criteria and implementation approaches of analytical procedures after procedure changes.

方法变更作为方法全生命周期中的一环,随着新技术的不断涌现,对产品质量需求的不断提高,3R和环境保护以及降低成本等要求的不断增加,越来越受到重视。本文参考国内外对方法学研究的最新进展,从方法学角度对方法变更进行系统的类别划分,并对不同方法变更类型间的差异和对变更后方法的评价标准和实施方式进行探讨。

The change of analytical procedure, a part of the entire lifecycle of a method, is increasingly valued with the emergence of new technologies, the increasing demand for product quality, 3R, environmental protection, and cost reduction requirements. This paper refers to the latest progress in the study of counterpart law at home and abroad, systematically classifies changes of analytical procedures from the perspective of methodology, and discusses the differences between different types of changes and the evaluation criteria and implementation approaches of analytical procedures after procedure changes.

分析方法的全生命周期(lifecycle)包括方法的设计和开发(design and development)、方法验证(validation)、方法的持续确认(ongoing verification)和方法变更(change of analytical procedure)等环节,详见图1[1]。方法变更是指对方法经批准使用后所做改变的一个统称,是方法全生命周期中的重要环节。
同产品上市后的变更一样,分析方法在整个产品生命周期内均有可能发生变更,如采用更先进的分析技术建立起来的方法,采用更能保护环境(包括3R)技术方法,以及可以节省更多时间等降低试验成本的方法等。这类变更可能涉及到现有方法的改进,方法进行实验室间的转移,更新检测技术手段,某些情况是对产品检测属性的改变,其中对产品检测属性改变的方法则需要对分析目标概况(ATP)进行修改并建立新方法或进行重新评价。
一般来说,方法变更的目的是保证变更后的方法始终满足其对产品的质量控制,同时更简便,更节省时间或成本,更符合3R原则和环境保护,更易于规范、自动化或连续检测等[1]
由于方法变更情形或类型较多,不同的变更类型需要采用不同的评估方式和评估标准,判断变更后的方法是否达到对产品质量控制的预期目的,目前的指导原则和法规并未对不同变更类型的评估方式有系统和详细的介绍,且未对变更是否成功给出明确的评估标准。
本文主要从方法学角度对变更的类型进行分类探讨,并结合分类阐述所需的评估方式和接受标准。
方法完成验证并被批准投入使用后,随着对所检验产品认识的深入,科学技术的发展和一些方法所需条件的变化,会有改进的需求。这些改进即属于方法变更。根据检测方法发生改变的程度和使用环境条件等的不同,可将其变更大致分为3类情况,方法转移、方法改进和方法替代。
方法转移和方法改进对产品的检测属性、分析技术和检测标准均未改变,仅分析环境条件[如实验室环境、试剂、仪器、检测药品(处方)等]发生变更。这类变更仅对原方法参数和/或操作条件进行风险评估即可。
根据所检测属性是否相关,将方法替代划分为3种类型,①相同属性完全替代的方法。对同一产品相同属性采用同属性、相同技术的优化或不同技术及操作步骤的检测方法。这类变更属于检测属性相同但检测技术更新而发生的变更,也可称为完全相同属性的方法替代。如对某具有紫外吸收的药品成分,采用兼具分离和分析功能的液相色谱方法替代分光光度方法,具有更好的专属性等(并非指全部HPLC技术的专属性没问题),对该种类型的替代,风险来源主要是2类方法之间的各个环节,如人、机和料。②2个属性有完全因果关系的方法替代。2种方法的检验原理和检验的机制不同,但是所检测的2个属性之间有完全的层级因果(递进)关系,如中小分子的含量与效应。这类方法替代,应该特别注意是否有别的影响因素造成2个属性间不能建立良好的模型关系。③属性间无完全因果关系的方法替代。检测原理和机制完全不同的方法,同时2种方法所检测的属性间也没有必然的因果关系。如对新冠疫苗活性的检测,从最初的检测动物体内产生抗体方法,转变为使用ELISA法在体外检测疫苗抗原活性;脊髓灰质炎灭活疫苗最初纳入欧洲药典时采用免疫后大鼠或鸡血清中抗体的体内检测方法,转变为使用D抗原含量检测法[2];化妆品中采用体外3T3中性红成纤维细胞光毒试验(3T3NRU光毒试验)法替代动物的皮肤光毒试验[3]等。其分析方法的ATP已从根本上发生变化,需重新开始新的方法探索。

对于方法改进、方法转移和方法替代3类变更方式,每一类都有其专属的定义和适用情况。
方法改进,是一种基于风险管理,在不影响方法预期用途的情况下,对已确定分析方法相关参数的改变,主要包括它在方法开发期间获得的数据(例如,实验设计(DOE)的耐用性实验)、既定条件(EC)、经证实的分析方法可接受范围(PAR)或方法可操作设计区域(MODR)内或外的方法相关参数[1]。对于改进的方法,其各操作步骤和实验地点没变,仅是MODR相关参数的变化,如改变色谱柱的品牌而造成的分离效果不佳,需调整色谱参数以达到良好的分离效能。
方法转移是指一个实验室(接收方)能够使用由另一个实验室(转移方,也称作转出方)所开发检验方法的过程,并确保接收的实验室明确检验的程序并有能力按规定进行检验[4]。方法转移的目的就是保证检验结果的准确可靠,保证实验室间检验结果的一致性,同时也是对实验室检验能力的重要评估[5]。方法转移常见的情况有生产企业的研发实验室转移到质控实验室;同一生产企业转移到外包生产企业等[6]
目前各国的法规和药典中给出的定义各有不同,但其核心内容可简单概括为“方法替代是用一种新的经过验证的方法(新方法),替代现有的法定方法(原方法)”。有关替代方法3种类型的实际应用如液相色谱方法替代分光光度方法、细胞实验替代动物实验等在本文“1”项中已详述,在此不再赘述。
方法成功改进的标准:改进后的方法是否能够满足(药典或法定)有关方法验证形成的判定标准[47]
方法改进成功的判定视修订情况的不同存在差异。①对于方法在既定条件(EC)、经证实的分析方法可接受范围(PAR)或方法可操作设计区域(MODR)内的方法修订,无需进行前后比较,仅从风险评估的角度进行评估即可;②对于超出上述范围的修订,需对修订后的方法进行方法验证[8],具体的评价标准参照药典或法定标准。
方法成功转移的标准是转移后方法的检测结果(报告值)是否处于主研发实验室方法验证给出的报告值预测区间或容忍区间范围内,或至少等效于原实验室的报告结果,即使用主研发实验室的验证标准进行判断。这里一般采用部分验证或共同验证。
参考文章“分析方法确认和转移的评估标准探讨”[9]中对于转移是否成功判定方式的介绍,确定转移后的方法与转移前方法检测能力相当的评价方式有方法的总变异、预测区间或容忍区间,方法误判率或方法能力指数等。在方法转移是否成功的判定中,可以通过与主研发实验室已建立起的上述验证标准去比较。过去一般使用准确度和精密度的判断方式,需要进行独自验证或部分验证,目前一种简单的方式,就是把转移后实验室的检测报告值与原实验室方法验证标准中预测区间或容忍区间去比较即可。这里需注意,容忍区间较预测区间更宽(大);使用预测区间对转移后的方法检测结果进行判断,当满足后,可保证方法更可靠或风险更小。但在目前国内方法验证所得数据集不容易计算该类区间的情况下,该方式难以实施。
总之,方法转移可采用上述方式,直接与已有的验证标准对比即可。有关详细的概念和运算可参考相关文献[9-10]
替代成功的接受标准为等效或适用。
图1可见,替代分为3类。一类是检测属性和/或检测技术发生变化(ATP发生变化),但2个属性间存在完全的因果关系;一类是检测属性和/或检测技术发生变化(ATP发生变化),但2个属性间无(完全)因果关系;一类是检测属性没变(ATP没变),仅检测技术发生变化。
检测属性和/或检测技术发生变化(ATP发生变化),但2个属性间存在完全因果关系的情形,因为对产品的检测属性有完全的因果关系,可对检验结果进行量值转换,确定新方法和原方法间是否存在一定的模型关系来判断新方法是否能够替代原方法。
检测属性发生变化的情形,由于检测属性发生变化,2种方法难以进行直接比较。这时,需要从保证产品质量的初心出发,判断新方法是否能够替代原有检测属性的方法。有时可能需要使用多种新属性的检测方法方可替代原有方法,如原检测抗原活性的动物体内方法,当用体外方法替代时,需要检测抗原含量和抗原结构等体外方法组合来替代。对于这类方法,很难进行直接比较,一般需从产品放行的质量风险角度加以考虑,也可通过对前期放行检验结果的批次进行检验,通过质反应数据的结论比较或质反应-量反应的ROC曲线方式找出替代方法检测连续性结果保持一致结论的量值方式[10],这都需要通过对更大量数据样本计算后方可得到可靠结论。
对检测属性没变,仅检测技术发生变化的替代方式,首先应该对替代方法进行验证,确定替代方法是否能够满足其预期的用途,然后按等效或者优于的方式进行2种方法的比较研究[411-12]
关于新方法的验证,可参考现有国内外药典[4-5]和ICH相关指导原则[17]
变更后的方法是否能够满足预期检验用途,首先与方法本身验证结果的稳健可靠密切相关。因此,需要对新方法的验证进行科学严谨的实验设计(样本量、Ⅰ类和Ⅱ类错误的确定,以及实验次数等),以从整体把控方法,获得与方法变异或方法能力相关的参数,包括预测区间和容忍区间等,为方法变更后提供变更成功的判定标准。
t检验有许多的局限性[13-14]。首先其最大的缺陷是即便差异具有显著性,也不能反映2组数据之间是否有有意义的差别(即不等效)。其次,当采用双样本t检验来判定2组样本是否等效时,其结果受检验的精密度以及样本量的影响。当传统的t检验是采用较差的精密度和/或小的样本量来进行比较时,较易出现假阴性结果,即将2种有显著差异的方法,判断为无差异,或误认为二者等效;而当二者的样本量较大时,又易出现将2个没有差异的方法判定为有显著差异,即不等效的情况。因此在判定2种方法检验数据集是否等效时推荐采用等效性检验[15],而非差异性检验[16-17]
知识管理和风险管理贯穿分析方法的全生命周期。知识管理用于定期评估科学且合适的分析技术;风险管理可用于优化影响分析方法性能的方法参数、评估分析方法参数对分析方法性能的潜在影响,以及确定并优先考虑需进行实验研究的分析参数。因此,通过知识管理和对影响因素的周期性评估,可更好地发现变更的方向,使检测方法更加科学可靠。此外,通过对变更相关风险的评估以及既往知识的累积,能够确保分析方法在变更后始终符合目的。
