Objective:

To develop an HPLC method for the separation of perindopril tert-butylamine and its epimer [(±)-1”-epi-perindopril tert-butylamine] and the determination of the epimer.

Methods:

Perindopril and (±)-1”-epi-perindopril were separated on an Agilent Poroshell CS-C18 column (100 mm×3.0 mm, 2.7 μm) maintained at 50 ℃ with the mobile phase containing a mixture of 0.15% sodium heptanesulfonate solution (adjusted to pH 2.0 with phosphoric acid) and acetonitrile-pentanol(217∶3)(82∶18, V/V) at 0.8 mL·min-1, and the detection wavelength was set at 215 nm. The injection volume was 2 μL.

Results:

(±)-1”-epi-perindopril and perindopril were separated successfully in 25 min with peak to valley ratio more than 3.0 or a resolution factor of 1.7. Good linear relationships were established between the peak response and the concentration in the range of 2-2 000 μg·mL-1 for the epimer and perindopril tert-butylamine(r>0.999). The quantitative limits(S/N= 10) were both about 1.0 μg·mL-1, and the detection limits(S/N=3) were both 0.3 μg·mL-1. The spiked recovery of the epimerer was 97.2% (RSD=1.8%, n=9). The content of (±)-1”-epi-perindopril tert-butylamine in 10 batches of samples ranged from 0.025% to 0.078%.

Conclusion:

The proposed method enhances the resolution efficiency, shows high accuracy, repeatability and stability. It can be effectively employed for the quality control of perindopril tert-butylamine.

目的:

建立一种快速液相色谱方法分离培哚普利叔丁胺及(±)-1”-差向-培哚普利叔丁胺(差向异构体),并对该差向异构体进行测定。

方法:

采用Agilent Poroshell CS-C18(100 mm×3.0 mm,2.7 μm)色谱柱,以0.15%庚烷磺酸钠溶液(磷酸调节pH 2.0)-乙腈/戊醇(217∶3)(82∶18)为流动相,流速0.8 mL·min-1,柱温50 ℃,进样量2 μL,在215 nm波长处分离培哚普利及(±)-1”-差向-培哚普利。

结果:

培哚普利及(±)-1”-差向-培哚普利在25 min内依次出峰,峰谷比和分离度满足测定要求。培哚普利叔丁胺及其差向异构体均在2~2 000 μg·mL-1范围内线性关系良好(r>0.999),定量限(S/N约10)和检测限(S/N约3)均分别约为1.0 μg·mL-1和0.3 μg·mL-1。差向异构体低、中、高3个浓度的平均加样回收率为97.2%(RSD=1.8%,n=9)。10批样品中(±)-1”差向-培哚普利叔丁胺含量均在0.025%~0.078%。

结论:

建立的培哚普利叔丁胺中(±)-1”-差向-培哚普利叔丁胺的液相色谱测定方法,与现行药典收载色谱方法相比可极大缩短平衡与分离时间,提高分离效率和检测灵敏度,重现性好,可有效用于培哚普利叔丁胺的生产质量控制。

目的:

建立一种快速液相色谱方法分离培哚普利叔丁胺及(±)-1”-差向-培哚普利叔丁胺(差向异构体),并对该差向异构体进行测定。

方法:

采用Agilent Poroshell CS-C18(100 mm×3.0 mm,2.7 μm)色谱柱,以0.15%庚烷磺酸钠溶液(磷酸调节pH 2.0)-乙腈/戊醇(217∶3)(82∶18)为流动相,流速0.8 mL·min-1,柱温50 ℃,进样量2 μL,在215 nm波长处分离培哚普利及(±)-1”-差向-培哚普利。

结果:

培哚普利及(±)-1”-差向-培哚普利在25 min内依次出峰,峰谷比和分离度满足测定要求。培哚普利叔丁胺及其差向异构体均在2~2 000 μg·mL-1范围内线性关系良好(r>0.999),定量限(S/N约10)和检测限(S/N约3)均分别约为1.0 μg·mL-1和0.3 μg·mL-1。差向异构体低、中、高3个浓度的平均加样回收率为97.2%(RSD=1.8%,n=9)。10批样品中(±)-1”差向-培哚普利叔丁胺含量均在0.025%~0.078%。

结论:

建立的培哚普利叔丁胺中(±)-1”-差向-培哚普利叔丁胺的液相色谱测定方法,与现行药典收载色谱方法相比可极大缩短平衡与分离时间,提高分离效率和检测灵敏度,重现性好,可有效用于培哚普利叔丁胺的生产质量控制。

Objective:

To develop an HPLC method for the separation of perindopril tert-butylamine and its epimer [(±)-1”-epi-perindopril tert-butylamine] and the determination of the epimer.

Methods:

Perindopril and (±)-1”-epi-perindopril were separated on an Agilent Poroshell CS-C18 column (100 mm×3.0 mm, 2.7 μm) maintained at 50 ℃ with the mobile phase containing a mixture of 0.15% sodium heptanesulfonate solution (adjusted to pH 2.0 with phosphoric acid) and acetonitrile-pentanol(217∶3)(82∶18, V/V) at 0.8 mL·min-1, and the detection wavelength was set at 215 nm. The injection volume was 2 μL.

Results:

(±)-1”-epi-perindopril and perindopril were separated successfully in 25 min with peak to valley ratio more than 3.0 or a resolution factor of 1.7. Good linear relationships were established between the peak response and the concentration in the range of 2-2 000 μg·mL-1 for the epimer and perindopril tert-butylamine(r>0.999). The quantitative limits(S/N= 10) were both about 1.0 μg·mL-1, and the detection limits(S/N=3) were both 0.3 μg·mL-1. The spiked recovery of the epimerer was 97.2% (RSD=1.8%, n=9). The content of (±)-1”-epi-perindopril tert-butylamine in 10 batches of samples ranged from 0.025% to 0.078%.

Conclusion:

The proposed method enhances the resolution efficiency, shows high accuracy, repeatability and stability. It can be effectively employed for the quality control of perindopril tert-butylamine.

培哚普利叔丁胺是一种不含巯基的长效血管紧张素转换酶抑制剂,用于治疗高血压和充血性心衰,临床疗效佳,安全性高,副作用小。培哚普利叔丁胺为手性化合物,具有5个手性中心。5个手性中心的碳原子均为S构型[1-2]。分子结构式详见图1。原研企业为法国施维雅(LES LABORATOIRES SERVIER)。国内有上药东英(江苏)药业有限公司、浙江美诺华药物化学有限公司、江苏永达药业有限公司等原料供应商。
《中华人民共和国药典》(以下简称《中国药典》)、欧洲药典(EP)和英国药典(BP)均收载了培哚普利叔丁胺,均需要对包括对映异构体和非对映异构体在内的一系列立体异构体进行质量控制,难度较大。2020年版《中国药典》、EP11和BP 2023的色谱条件一致,均采用HPLC等度洗脱,培哚普利峰的保留时间约为100 min,分析时间约为150 min[3-5],此外,EP 11版和BP 2023还要求色谱系统至少平衡4 h。由于分离难度大,系统适用性要求差向异构体杂质峰的峰高和该异构体与主成分峰之间的峰谷比(peak to valley ratio)应>3。目前培哚普利质量控制方面的文献集中在溶出度、有关物质检查和含量测定[6-14],而包括差向异构体在内的异构体方面的研究报道[15]很少。为克服现有立体异构体杂质检测方法存在的分析时间长,分离难度大,系统稳定性差等不足,本研究开发了一种适用于碱性化合物分析的基于表面带正电荷的杂化颗粒小粒径十八烷基硅烷键合表面多孔型硅胶色谱柱的液相色谱紫外检测方法,该方法分离时间短,分离良好,系统稳定,具有实际应用价值。
Agilent 1260液相色谱仪,紫外检测器;Agilent 1290液相色谱仪,二极管阵列检测器;Agilent OpenLAB CDS v2.6计算机化系统(安捷伦科技公司)。Waters Acquity Arc液相色谱仪,Empower 3数据库系统(沃特斯公司)。BP211D型十万分之一精密天平(Sartorius公司)。Bluepard DHG-9140A型电热鼓风干燥箱(上海一恒科学仪器有限公司),Binder KBF 240恒温恒湿箱(Binder公司),STARITECH光照箱(北京天星科仪科技有限公司)。Labonova Ultra Pro超纯水系统(Think-Lab公司)。
立体异构体检查用培哚普利叔丁胺对照品(含0.1%的(±)-1”-差向-培哚普利叔丁胺),批号2.1,由EDQM提供。差向异构体[(±)-1”-差向-培哚普利叔丁胺]对照品,批号C4X-12521-1807,纯度90.8%,由CATO提供。培哚普利叔丁胺对照品,批号101072-201902,纯度99.3%,由中国食品药品检定研究院提供。
培哚普利叔丁胺原料药7批,批号分别为802008401、801700531、801609823、801700518(Glenmark Pharmaceuticals Limited公司)和DY-PDP-2020036、DY-PDP-2020037、DY-PDP-2020038(上药东英(江苏)药业有限公司)。培哚普利叔丁胺原料药粗品3批,批号分别为DY-7-23-2020021、DY-7-23-2020024、DY-7-23-2020025(上药东英(江苏)药业有限公司)。
乙腈、甲醇、乙醇、异丙醇(色谱纯,默克公司)。庚烷磺酸钠(色谱纯,东京化成公司)。戊醇、磷酸、高氯酸、盐酸、冰醋酸(分析纯,上海凌峰化学试剂有限公司)。

称取立体异构体检查用培哚普利叔丁胺对照品(含0.1%的(±)-1”-差向-培哚普利叔丁胺)适量,或者分别称取培哚普利叔丁胺与(±)-1”-差向-培哚普利叔丁胺的对照品适量,加乙醇适量溶解并稀释制成每1 mL含培哚普利叔丁胺2 mg和(±)-1”-差向-培哚普利叔丁胺2 μg的溶液。
取(±)-1”-差向-培哚普利叔丁胺对照品适量,精密称定,加乙醇溶解并定量稀释制成每1 mL中约含2 mg的差向异构体对照品储备液。
分别取培哚普利叔丁胺和(±)-1”-差向-培哚普利叔丁胺的对照品适量,精密称定,加乙醇溶解并定量稀释制成每1 mL中含培哚普利叔丁胺和(±)-1”-差向-培哚普利叔丁胺各约2 mg的混合对照品储备液。
取培哚普利叔丁胺原料药或粗品约20 mg,精密称定,置10 mL量瓶中,加乙醇溶解并稀释至刻度,制得每1 mL约为2 mg的供试品溶液。
取系统适用性溶液和空白溶液(乙醇)分别进样2 μL,典型色谱图见图2,(±)-1”-差向-培哚普利与培哚普利在Agilent 1260、Agilent 1290上的峰谷比分别为4.89和6.18,满足峰谷比>3的分离度要求。在Waters Acquity Arc上的分离度为1.71,可实现基线分离。空白乙醇对分离检测无干扰。
分别精密量取混合对照品储备液适量,用乙醇定量稀释成含差向异构体和含培哚普利叔丁胺质量浓度为2、10、20、40、100、400、1 000和2 000 μg·mL-1的混合对照品溶液。照“2.1”项色谱条件测定,以浓度X为横坐标,峰面积Y为纵坐标,分别进行线性回归。差向异构体和培哚普利叔丁胺质量浓度在2~2 000 μg·mL-1范围内的线性回归方程分别为

由上述差向异构体线性回归方程斜率/培哚普利叔丁胺线性回归方程线斜率,计算得到差向异构体的相对响应因子(F)分别为1.00和0.98。此外,根据二者低、中、高浓度对照品溶液的相对响应值,计算得到F平均值为0.99。2种计算得到的F一致,可以采用峰面积归一化方法进行定量。
将混合对照品储备液用乙醇逐步稀释后进样,得到差向异构体和培哚普利叔丁胺的定量限(S/N约为10)为0.9和1.2 μg·mL-1(相当于供试品溶液浓度的0.045%和0.06%),检测限(S/N约为3)均为0.3 μg·mL-1(相当于供试品溶液浓度的0.015%)。
分别取“2.4”项下差向异构体和培哚普利叔丁胺均分别为2、10和20 μg·mL-1的混合对照品溶液,各连续进样6次,差向异构体低、中、高浓度的(±)-1”-差向-培哚普利峰面积的RSD(n=6)分别为5.5%、4.2%和1.4%,培哚普利叔丁胺低、中、高浓度的培哚普利峰面积的RSD(n=6)分别为5.9%、5.4%和2.5%。
精密称取样品(批号802008401)6份,按“2.2.4”项方法制备供试品溶液,进行测定,按照峰面积外标法和归一化法计算,差向异构体含量(n=6)分别为0.044%和0.042%,均略小于定量限(0.045%),不再考察样品测定重复性。
取原料(批号802008401)9份各6 mg,精密称定,分别精密加入2、10和20 μg·mL-1浓度水平的差向异构体对照品溶液3 mL,每个浓度各3份,定量溶解制成每1 mL含培哚普利叔丁胺2 mg和差向异构体1.88、9.39和18.78 μg·mL-1的回收率试验溶液,进样测定。按外标法计算回收率,结果见表1。低、中、高3个浓度的平均加样回收率(n=9)为97.2%,RSD为1.8%。
考虑到乙醇溶液的挥发性,分别取“2.6”项下低、中、高浓度回收率试验溶液,在12 ℃控温进样器中放置,分别在放置后0、31和55 h各进样2 μL,低、中、高浓度回收率试验溶液的(±)-1”-差向-培哚普利峰面积的RSD(n=9)分别为9.4%、3.4%和1.2%,在12 ℃控温进样器中连续放置55 h稳定性较好。
按照“2.1”项下色谱条件,分别对有机相比例、柱温和流速进行小的变动,考察10 μg·mL-1差向异构体的回收率试验溶液不受影响的承受程度。结果见表2
略微改变柱温、流速和有机相比例,差向异构体峰面积归一化法测得结果无明显差异,系统适用性分离满足要求。改变流动相中有机相比例对分离影响较大,建议流动相中有机相仅在17%和18%的比例之间调整。
取供试品溶液,按照“2.1”项下色谱条件进行检测,样品的典型色谱图见图3,按照外标法和归一化法计算,10批样品中差向异构体含量均在0.025%~0.078%,测定结果如表3。2种计算结果基本一致。
本研究以0.15%庚烷磺酸钠溶液(磷酸调节pH 2.0)-乙腈/戊醇(217∶3)(75~82∶25~18)为初筛流动相,流速0.8 mL·min-1,柱温50 ℃,取系统适用性溶液2 μL进样,在Poroshell HPH-C18(100 mm×3.0 mm,2.7 μm)色谱柱上基本无分离,在Pursuit XRs Ultra(150 mm×3.0 mm,2.8 μm)、Poroshell CS-C18(100 mm×3.0 mm,2.7 μm)、Poroshell Bonus-RP(100 mm×3.0 mm,2.7 μm)和Zorbax SB-C18(100 mm×3.0 mm,1.8 μm)色谱柱上有可分离的趋势,以Poroshell CS-C18(100 mm×3.0 mm,2.7 μm)色谱柱上分离效果为最好。经定位,(±)-1”-差向-培哚普利和培哚普利依次出峰。故采用Poroshell CS-C18(100 mm×3.0 mm,2.7 μm)色谱柱继续进行色谱条件优化。详见色谱图4。Poroshell CS-C18为表面带正电荷的十八烷基硅烷键合表面多孔型杂化颗粒硅胶色谱柱,表面多孔型硅胶传质效率和柱效高;表面带正电荷对分离碱性药物可改善峰形,提高分析效率。
照“2.1”项下色谱条件,仅改变有机相分别为18%的甲醇、乙腈或乙腈/戊醇(217∶3),分别取系统适用性溶液进样,如图5。有机相为18%的乙腈/戊醇(217∶3)时,(±)-1”-差向-培哚普利和培哚普利2个异构体的峰谷比>3,保留时间适中,满足分离要求。而选择甲醇时40 min未出峰。
仅改变有机相中乙腈/戊醇的比例分别为217∶3、217∶5和217∶8,取系统适用性溶液进样。结果乙腈/戊醇比例为217∶3时,峰谷比最高;比例为217∶5和217∶8时,流动相极性增加,保留减弱,峰谷比<3。故本文选择有机相为乙腈/戊醇(217∶3)。
仅改变有机相比例,分别为14%、16%、18%、20%和22%,取系统适用性溶液进样。结果有机相比例增加,流动相极性增加,保留减弱,峰谷比减小。有机相比例为14%时,2个异构体保留太强,40 min未出峰;有机相比例为16%时,峰谷比>7,2个异构体保留时间较长,分别为36和39 min;有机相比例为18%时,保留时间适中,峰谷比>3。本文选择有机相比例为18%。
离子对试剂作为添加剂分离生物碱时,兼具离子交换模式与离子对模式的效果。2种分离模式均随着离子对试剂浓度的增加而增强对待测成分的保留[16]。分别以0.05%、0.10%和0.15%庚烷磺酸钠溶液(磷酸调节pH 2.0)-乙腈/戊醇(217∶3) (82∶18)为流动相,其他照“2.1”项下色谱条件,取系统适用性溶液进样。结果庚烷磺酸钠溶液浓度增加,2个异构体保留增加,分离度和峰谷比增加。以0.10%或0.15%庚烷磺酸钠溶液(磷酸调节pH 2.0)-乙腈/戊醇(217∶3)(82∶18)为流动相均可满足峰谷比>3的要求。
分别选择高氯酸、磷酸和盐酸、醋酸为pH调节剂,与不调节pH的0.10%庚烷磺酸钠溶液进行比较。结果,在其他条件相同情况下,用高氯酸和磷酸调节pH 2.0,两异构体保留时间和峰谷比基本一致,峰谷比均>3;以磷酸为pH调节剂得到的信噪比更高。离子对试剂溶液pH>2.0时,峰谷比均<3;离子对试剂溶液不调节pH,40 min未见峰洗脱。故本文选择调节离子对试剂溶液pH为2.0。
照“2.1”项下色谱条件,分别在40、45、50、55和60 ℃柱温时,取系统适用性溶液2 μL进样。结果上述柱温下2个异构体峰谷比均>3。随着柱温升高,2个异构体保留时间缩短,峰谷比增加。综合考虑,本文选择柱温50 ℃。
照“2.1”项下色谱条件,分别取系统适用性溶液1、2、5、8和10 μL进样。结果进样量增加,分离度和峰谷比下降,信噪比增加。进样量为1和2 μL时,峰谷比均>6。综合考虑,本文选择2 μL进样量。
照“2.1”项下色谱条件,分别在0.6、0.8和1.0 mL·min-1流速时,取系统适用性溶液2 μL进样。结果流速增加,保留减弱,分离度和峰谷比下降。流速为0.6和0.8 mL·min-1时,峰谷比均>3。综合考虑,本文选择0.8 mL·min-1的流速。
照“2.1”项下色谱条件,分别取2和5 mg·mL-1的系统适用性溶液2 μL进样。结果随着样品浓度增加,分离度和峰谷比略有下降,信噪比增加,均能满足测定要求。综合考虑,本文选择2 mg·mL-1的样品浓度。
取原料样品(批号802008401、DY-PDP-2020036)和原料粗品(批号DY-7-23-2020025)各约1 g,一式3份共9份,置称量瓶中,分别于(4 500±500) 1x光强、60 ℃恒温箱,25 ℃/(90±5)%RH条件下放置,于第5天和第10天进行取样配制供试品溶液。照“2.1”项色谱条件测定,不同放置条件差向异构体测定结果基本一致;与常规条件含量相比基本未发生变化。培哚普利叔丁胺样品在光、60 ℃以及25 ℃/(90±5)%RH环境中基本稳定,不会发生差向异构体的转化。
本研究建立的培哚普利叔丁胺原料中培哚普利叔丁胺差向异构体测定方法快速准确,操作简便,耐用性好;与《中国药典》和欧洲药典150 min的分析时间和分析方法相比,分离时间显著缩短,分离良好,系统稳定,无须柱前长时间平衡时间,在不同液相色谱仪和不同批号的该类型带正电荷杂化颗粒表面多孔C18色谱柱上均可以重现,可有效用于培哚普利叔丁胺的实际生产质量控制,同时,作为《中国药典》方法修订的依据,可以提高培哚普利叔丁胺中立体异构体的质量控制水平。
