Objective:

To establish a fingerprint of Gualou Guizhi decoction and a method for determination of multi-component to clarify the transfer rule of quantities and quality from decoction pieces and substance benchmarks.

Methods:

Fifteen batches of Gualou Guizhi decoction substance benchmarks were prepared and analyzed by the method of HPLC. Traditional Chinese medicine (TCM) Chromatographic Fingerprint Similarity Evaluation Software (2012) and hierarchical cluster analysis (HCA), principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA) was used to evaluate the quality differences between batches of substance benchmarks, and screen the main chemical compositions that were led to quality discrepancy. The main differential components were determined by HPLC. And their contents, yields and transfer rates were analyzed for transfer rule of quantities and quality.

Results:

HPLC fingerprint of Gualou Guizhi decoction was established. A total of 30 common peaks in the fingerprint of Gualou Guizhi decoction substance benchmarks were assigned. Compared with the reference standards, 9 components were identified as gallic acid, albiflorin, paeoniflorin, liquiritin, ononin, liquiritigenin, cinnamic acid, isoliquiritigenin and glycyrrhizic acid. The similarities of 15 batches of Gualou Guizhi decoction substance benchmarks samples were all above 0.920. All of them were divided into two categories by three chemical recognition modes for 5 major differential components, including glycyrrhizic acid, liquiritigenin, paeoniflorin, iquiritin, and ononin. The HPLC was also applied to determine the contents of multi-components and the method validation results were good. The average transfer rates of five index components were 51.65%, 40.46%, 74.74%, 60.06%, and 34.54%, respectively and their average extraction rate was 14.20%.

Conclusion:

The critical quality properties of Gualou Guizhi decoction can be stably transferred from decoction pieces to substance benchmarks. The method of fingerprint and content determination is accurate and reliable, which is able to provide technique for quality control of Gualou Guizhi decoction formulation.

目的:

建立栝楼桂枝汤(Gualou Guizhi decoction)物质基准指纹图谱和多成分含量测定方法,探究饮片-物质基准关键质量属性的量值传递规律。

方法:

制备15批栝楼桂枝汤物质基准,采用ChromCore 120 C18 (250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)-0.1%甲酸水(B)为流动相,进行梯度洗脱,流速0.8 mL·min-1,柱温30 ℃,检测波长254 nm,采用中药色谱指纹图谱相似度评价软件(2012年版)建立指纹图谱,结合层次聚类分析(hierarchical cluster analysis,HCA)、主成分分析(principal component an alysis,PCA)与正交偏最小二乘法-判别分析(orthogonal partial least squares-discriminant analysis,OPLS-DA),筛选不同批次栝楼桂枝汤物质基准质量差异的主要化学成分。建立HPLC法测定主要差异性成分含量,以含量、出膏率和转移率分析栝楼桂枝汤的量值传递规律。

结果:

建立了栝楼桂枝汤物质基准HPLC指纹图谱,确定30个共有峰,与对照品比对共指认出其中9个成分,分别为没食子酸、芍药内酯苷、芍药苷、甘草苷、芒柄花苷、甘草素、肉桂酸、异甘草素、甘草酸。15批栝楼桂枝汤物质基准指纹图谱相似度均>0.920。3种化学识别模式均将样品分为2类,造成主要差异化学成分共5个,分别为甘草酸、甘草素、芍药苷、甘草苷、芒柄花苷。建立了多成分HPLC含量测定方法,符合方法学要求。饮片-物质基准之间5个差异成分的平均转移率分别为51.65%、40.46%、74.74%、60.06%、34.54%,平均出膏率为14.20%。

结论:

栝楼桂枝汤物质基准的关键质量属性在饮片-物质基准间稳定传递。建立的栝楼桂枝汤指纹图谱和含量测定方法准确、可靠,可为栝楼桂枝制剂的质量控制提供技术参考。

目的:

建立栝楼桂枝汤(Gualou Guizhi decoction)物质基准指纹图谱和多成分含量测定方法,探究饮片-物质基准关键质量属性的量值传递规律。

方法:

制备15批栝楼桂枝汤物质基准,采用ChromCore 120 C18 (250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)-0.1%甲酸水(B)为流动相,进行梯度洗脱,流速0.8 mL·min-1,柱温30 ℃,检测波长254 nm,采用中药色谱指纹图谱相似度评价软件(2012年版)建立指纹图谱,结合层次聚类分析(hierarchical cluster analysis,HCA)、主成分分析(principal component an alysis,PCA)与正交偏最小二乘法-判别分析(orthogonal partial least squares-discriminant analysis,OPLS-DA),筛选不同批次栝楼桂枝汤物质基准质量差异的主要化学成分。建立HPLC法测定主要差异性成分含量,以含量、出膏率和转移率分析栝楼桂枝汤的量值传递规律。

结果:

建立了栝楼桂枝汤物质基准HPLC指纹图谱,确定30个共有峰,与对照品比对共指认出其中9个成分,分别为没食子酸、芍药内酯苷、芍药苷、甘草苷、芒柄花苷、甘草素、肉桂酸、异甘草素、甘草酸。15批栝楼桂枝汤物质基准指纹图谱相似度均>0.920。3种化学识别模式均将样品分为2类,造成主要差异化学成分共5个,分别为甘草酸、甘草素、芍药苷、甘草苷、芒柄花苷。建立了多成分HPLC含量测定方法,符合方法学要求。饮片-物质基准之间5个差异成分的平均转移率分别为51.65%、40.46%、74.74%、60.06%、34.54%,平均出膏率为14.20%。

结论:

栝楼桂枝汤物质基准的关键质量属性在饮片-物质基准间稳定传递。建立的栝楼桂枝汤指纹图谱和含量测定方法准确、可靠,可为栝楼桂枝制剂的质量控制提供技术参考。

Objective:

To establish a fingerprint of Gualou Guizhi decoction and a method for determination of multi-component to clarify the transfer rule of quantities and quality from decoction pieces and substance benchmarks.

Methods:

Fifteen batches of Gualou Guizhi decoction substance benchmarks were prepared and analyzed by the method of HPLC. Traditional Chinese medicine (TCM) Chromatographic Fingerprint Similarity Evaluation Software (2012) and hierarchical cluster analysis (HCA), principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA) was used to evaluate the quality differences between batches of substance benchmarks, and screen the main chemical compositions that were led to quality discrepancy. The main differential components were determined by HPLC. And their contents, yields and transfer rates were analyzed for transfer rule of quantities and quality.

Results:

HPLC fingerprint of Gualou Guizhi decoction was established. A total of 30 common peaks in the fingerprint of Gualou Guizhi decoction substance benchmarks were assigned. Compared with the reference standards, 9 components were identified as gallic acid, albiflorin, paeoniflorin, liquiritin, ononin, liquiritigenin, cinnamic acid, isoliquiritigenin and glycyrrhizic acid. The similarities of 15 batches of Gualou Guizhi decoction substance benchmarks samples were all above 0.920. All of them were divided into two categories by three chemical recognition modes for 5 major differential components, including glycyrrhizic acid, liquiritigenin, paeoniflorin, iquiritin, and ononin. The HPLC was also applied to determine the contents of multi-components and the method validation results were good. The average transfer rates of five index components were 51.65%, 40.46%, 74.74%, 60.06%, and 34.54%, respectively and their average extraction rate was 14.20%.

Conclusion:

The critical quality properties of Gualou Guizhi decoction can be stably transferred from decoction pieces to substance benchmarks. The method of fingerprint and content determination is accurate and reliable, which is able to provide technique for quality control of Gualou Guizhi decoction formulation.

栝楼桂枝汤出自东汉·张仲景《金匮要略》,由天花粉、桂枝、白芍、生姜、大枣和甘草6味药材组成,具有滋养津液、解肌发表、柔润筋脉等功效,主治太阳病[1-2]。现代研究表明,栝楼桂枝汤对脑缺血再灌注损伤具有显著的神经保护作用[3-4],临床上主要用于治疗脑卒中后肢体痉挛[5]。目前,尚未有对栝楼桂枝汤整体质量控制方面的研究,因此,建立与完善其质量标准具有重要的意义。
2021年国家药典委员会颁布的《中药配方颗粒质量控制与标准制定技术要求》中明确提出“中药配方颗粒所有药效学研究均须与标准汤剂对比,以保证与标准汤剂质量一致性”,中药标准汤剂已成为中药配方颗粒剂[6]、经典名方制剂[7]质量控制的重要模式之一。标准汤剂作为复方制剂的质量基准和标准参照物,可实现中药整体质量控制。然而中药成分的复杂性使得保持汤剂-颗粒剂质量一致性成为质量控制的难点[8]。中药指纹图谱或特征图谱结合化学计量学方法能科学、全面、定量反映中药所包含的化学信息,表征从原料到中间体到成品的量质传递情况,是标准汤剂研究常用的技术手段[9-11]
为进一步完善栝楼桂枝汤物质基准的质量标准及后续颗粒制剂的开发,本研究拟建立栝楼桂枝汤指纹图谱并结合化学模式识别开展质量评价和筛选主要质量差异物,同时建立含量测定方法,测定15批栝楼桂枝汤物质基准指纹图谱、主要差异性成分含量、转移率和出膏率,明确关键质量属性在饮片-物质基准的传递规律,初步建立栝楼桂枝汤的过程质量控制体系。
LC-20AT高效液相色谱仪(岛津公司),配置LC-20AT型二元梯度泵和SPD-20AV型DAD检测器;EX225DZH型十万分之一电子天平(奥豪斯仪器有限公司);KQ-500DE型超声波清洗器(昆山市超声仪器有限公司);HWS-12型电热恒温水浴锅(上海-恒科学仪器有限公司);MILLI-Q Direct16超纯水机(Millipore公司)。
对照品没食子酸(批号110831-201906,纯度91.5%)、甘草苷(批号111610-202209,纯度95.2%)、肉桂酸(批号119786-201604,纯度98.8%)、芍药苷(批号110736-202145,纯度94.6%)均购自中国食品药品检定研究院;芍药内酯苷(批号MUST-22050618,纯度≥91.4%)、甘草素(批号MUST-22092613,纯度99.51%)均购自成都曼斯特生物科技有限公司;甘草酸(批号Y02J11L113432,纯度≥98%)购自上海源叶生物科技有限公司;异甘草素(批号C179013,纯度≥98%)、芒柄花苷(批号O138900,纯度≥98%)均购自上海阿拉丁试剂有限公司;乙腈、甲醇均为色谱纯,购自Thermo Fisher公司;水为超纯水,其他试剂均为分析纯。
栝楼、桂枝、白芍、大枣、生姜、甘草等6味饮片均由闽侯县上街至诚医药商店代为购买,经福建省食品药品质量检定研究院金鸣主任中药师检测均符合2020年版《中华人民共和国药典》一部各饮片项下性状鉴别项规定,其余项目经课题组检测亦符合规定。饮片产地信息见表1。采用随机数法对表1中15批次各饮片进行随机组合并排序,制得15批栝楼桂枝汤物质基准,组合信息见表2
《金匮要略》中记载栝楼桂枝汤原方为“天花粉、甘草各二两,桂枝、芍药、生姜各三两,大枣十二枚。以水九升,煮取三升,分温三服,取微汗。汗不出,食顷,啜热粥发之”。通过考证及度量换算[15-16],基于福建中医药大学附属第二人民医院[17]治疗脑卒中临床用药,参照《医疗机构中药煎药室管理规范》[18]拟定栝楼桂枝汤标准汤剂制备方法。按处方剂量称取天花粉30 g,桂枝9 g,白芍9 g,生姜9 g,大枣9 g,甘草6 g,加8倍量水,放置浸泡30 min,以武火煮沸后,转文火继续煎煮30 min,200目标准筛趁热过滤,滤渣再加入7倍量水煎煮20 min,滤过,合并滤液,60 ℃水浴浓缩至600 mL,冷冻干燥48 h,即得物质基准样品(S1~S15)。
采用ChromCore 120 C18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)-0.1%甲酸水溶液(B)为流动相,梯度洗脱(0~50 min,5%A→32%A,50~60 min,32%A→48%A,60~65 min,48%A→5%A,65~72 min,5%A),流速0.8 mL·min-1,柱温30 ℃,进样量10 μL,检测波长254 nm。理论板数按没食子酸峰计算应不低于4 000。
取没食子酸、芍药内酯苷、芍药苷、甘草苷、芒柄花苷、甘草素、肉桂酸、甘草酸、异甘草素的对照品适量,精密称定,加甲醇溶解制成每1 mL含1.01、1.98、19.97、2.49、0.36、2.00、1.96、4.01、2.00 mg的单一对照品储备液。分别精密量取各单一对照品储备液适量,加甲醇制成每1 mL含100.97、198.02、199.68、248.96、35.80、200.20、195.88、400.64、200.00 μg的混合对照品溶液,保存于4 ℃冰箱中备用。
精密称取“2.1”项下的栝楼桂枝汤物质基准样品3.00 g,置具塞锥形瓶中,加入50%甲醇水溶液30 mL,超声(功率250 W,频率40 kHz)提取30 min,放冷,过滤,用50%甲醇水溶液约10 mL洗涤锥形瓶及残渣。重复上述操作2次,合并滤液与洗液,60 ℃水浴浓缩,50%甲醇水溶液定容至50 mL,0.45 μm微孔滤膜过滤,取续滤液,冷藏备用。
取同一供试品溶液(样品编号S6),按“2.2.1”项色谱条件重复连续测定6次,记录色谱图。以1号峰(没食子酸)为参照峰,计算特征峰的相对保留时间及相对峰面积,结果相对保留时间的RSD均<1.1%,相对峰面积的RSD均<2.9%,表明仪器精密度良好。
取同一批栝楼桂枝汤样品(样品编号S6),按“2.2.2.2”项方法平行制备6份供试品溶液,按“2.2.1”项色谱条件分别进样,记录色谱图。以1号峰(没食子酸)为参照峰,计算特征峰的相对保留时间及相对峰面积,结果相对保留时间的RSD均<1.8%,相对峰面积的RSD均<2.3%,表明本方法重复性良好。
取同一供试品溶液(样品编号S6),按“2.2.1”项色谱条件分别在0、2、4、8、12、24 h进样,记录色谱图。以1号峰(没食子酸)为参照峰,计算特征峰的相对保留时间及相对峰面积,结果相对保留时间的RSD均<2.3%,相对峰面积的RSD均<3.0%,表明供试品溶液在24 h内稳定性良好。
按“2.1”项下方法制备15批栝楼桂枝汤物质基准、各单味药及阴性样品,按“2.2.2.2”项下方法制备供试品溶液、单味药供试溶液及阴性样品溶液,照“2.2.1”项下色谱条件测定。采用国家药典委员会“中药色谱指纹图谱相似度评价系统(2012版)”软件进行分析。以S6号样品的指纹图谱作为参照图谱进行指纹匹配,确定了30个共有峰,并生成对照指纹图谱(R),物质基准指纹图谱叠加图见图1。栝楼桂枝汤物质基准指纹图谱与对照指纹图谱的相似度分别为0.984、0.971、0.967、0.990、0.987、0.970、0.952、0.980、0.977、0.926、0.963、0.991、0.959、0.950、0.961,相似度均>0.920,表明所建立的指纹图谱稳定,可以反映物质基准的指纹特征。但不同批次之间物质基准因饮片产地不同存在质量差异。比较对照品色谱图和紫外吸收光谱图,共标定出9个色谱峰所属的化学成分,分别为没食子酸(峰1)、芍药内酯苷(峰11)、芍药苷(峰12)、甘草苷(峰15)、芒柄花苷(峰20)、甘草素(峰21)、肉桂酸(峰22)、异甘草素(峰28)和甘草酸(峰29)。物质基准特征峰及其归属如图2所示,其中4、5、8号峰来自天花粉,3、16、17、19、21、22、30号峰来自桂枝,1、7、10、11、12、16、17、18号峰来自白芍。在30个共有峰中使药甘草占据15个,分别为2、6、9、10、13、14、15、20、23、24、25、26、27、28、29和30号峰,表明使药甘草对该方的质量控制较为重要。
层次聚类分析(hierarchical cluster analysis,HCA)属于无监督分析方法,直接对数据进行建模分类,可以明显区分组间差异,常用于数据的初步探索性分析[19-20]。本研究应用SPSS 23.0数据分析软件对15批栝楼桂枝汤物质基准的共有峰进行系统聚类,采用组间连接法,测量区间为平方欧氏距离,聚类结果见图3。结果表明,当欧氏距离约为25时,样品可分为2类,S1~S10、S14~S15号样品归为第1类;S11~S13号样品为第2类,不同样品间存在一定的差异性。含量测定结果发现,S11~S13号中甘草酸含量高于其他批次,提示甘草酸可能为造成差异的原因。
将15批物质基准的30个共有峰的峰面积为变量导入SIMCA-P+14.1软件进行正交偏最小二乘法-判别分析(orthogonal partial least squares-discriminant analysis,OPLS-DA)处理,得分矩阵图见图4-B。15批样品聚为2类,S1~S10、S14~S15号样品归为第1类,S11~S13号样品为第2类。结果累积解释能力参数(R2XR2Y)分别为0.967和0.982,预测能力参数Q2为0.901,均>0.5,表明所建立的模型稳定可靠且具有良好的预测性。图4-C中的点离原点越远,表明对应变量(色谱峰)权重值越大。以变量重要性投影(VIP)值>1为判定标准,筛选差异组分,结果共找到了5个差异标志物,按VIP值从大到小排序分别为甘草酸(峰29)、甘草素(峰21)、芍药苷(峰12)、甘草苷(峰15)、芒柄花苷(峰20);其中芍药苷来源于白芍,其余4个差异成分均来源于甘草,说明严格控制芍药、甘草药材质量是确保栝楼桂枝汤物质基准质量稳定的关键。VIP结果见图4-D
同“2.2.1”项下条件。
参考2020年版《中华人民共和国药典》一部方法制备白芍、甘草的供试溶液。
取甘草饮片粉末(过60目筛)约0.2 g,精密称定,置具塞锥形瓶中,精密加入70%乙醇水溶液100 mL,称量,超声(功率250 W,频率40 kHz)处理30 min,放冷,用70%乙醇水溶液补足减失的量,摇匀,滤过,取续滤液,3 600 r·min-1离心10 min,取上清液过微孔滤膜(0.45 μm),即得甘草供试溶液,冷藏备用。
取白芍饮片粉末(过60目筛)约0.1 g,精密称定,置具塞锥形瓶中,加70%乙醇水溶液35 mL,称量,超声(功率240 W,频率45 kHz)处理30 min,放冷,用70%乙醇水溶液补足减失的量,摇匀,滤过,取续滤液,3 600 r·min-1离心10 min,取上清液过微孔滤膜(0.45 μm),即得白芍供试溶液,冷藏备用。
同“2.2.2.2”项下方法操作,即得。
分别精密吸取“2.2.2.1”项下的芍药苷、甘草苷、芒柄花苷、甘草素、甘草酸的单一对照品储备液适量,加甲醇制成每1 mL含芍药苷、甘草苷、芒柄花苷、甘草素、甘草酸998.40、62.24、8.96、50.08、100.16 μg的混合对照品溶液,保存于4 ℃冰箱中备用。
以70%乙醇水溶液、甲醇为空白溶剂,分别吸取混合对照品溶液、白芍供试溶液、甘草供试溶液、栝楼桂枝汤物质基准供试品溶液和空白溶剂注入HPLC分析仪,按“2.3.1”项色谱条件进行测定。如图5所示,5个成分色谱峰与相邻色谱峰的分离度均>1.5,栝楼桂枝汤物质基准样品供试品溶液、白芍供试溶液、甘草供试溶液与混合对照品溶液保留时间相同位置均出现色谱峰,空白溶剂在与混合对照品溶液相同保留时间位置上无色谱峰,表明空白溶剂对待测各成分的测定无干扰,表明该方法专属性良好。
精密量取混合对照品溶液适量,加甲醇逐级稀释1、2、4、8、16倍,摇匀,制得系列浓度混合对照品溶液,按“2.3.1”项色谱条件测定各成分的峰面积,以质量浓度(X)为横坐标,峰面积积分值(Y)为纵坐标进行线性回归。结果见表3,表明各成分在各自质量浓度范围内线性关系良好。通过注入不同浓度的一系列混合对照品稀释溶液,以信噪比(S/N)为3∶1时的浓度为检测限(LOD),S/N为10∶1时的浓度为定量限(LOQ),结果见表3
取混合对照品溶液适量,按“2.3.1”项色谱条件连续进样6次,分别记录峰面积。结果芍药苷、甘草苷、芒柄花苷、甘草素、甘草酸5个成分峰面积的RSD分别为2.8%、2.7%、0.52%、0.40%、0.64%,仪器精密度良好。
取S6号样品的供试品溶液,分别于0、4、8、10、14、18、24 h按“2.3.1”项色谱条件进样,测定峰面积,芍药苷、甘草苷、芒柄花苷、甘草素、甘草酸5个成分峰面积的RSD分别为2.1%、0.60%、0.84%、1.8%、1.3%,结果表明供试品溶液24 h内稳定。
取S6样品6份,按“2.3.2.2”项方法制备供试品溶液,按“2.3.1”项色谱条件进样测定,记录色谱峰面积,计算含量。芍药苷、甘草苷、芒柄花苷、甘草素、甘草酸5个成分的平均含量分别为4.38、0.64、0.69、0.45、2.15 mg·g-1,RSD分别为2.0%、1.2%、1.1%、0.78%、0.82%,表明该方法重复性良好。
精密称取已知含量的样品(样品编号S6)约1.00 g,分别加入相当于样品浓度100%的混合对照品溶液,按照“2.3.2.2”项下方法平行制备6份供试溶液,按“2.3.1”项色谱条件进样测定,芍药苷、甘草苷、芒柄花苷、甘草素、甘草酸的加样回收率分别为100.7%、99.6%、102.2%、99.4%、98.6%,RSD分别为1.9%、3.7%、4.8%、3.7%、2.5%,表明该方法回收率良好。
取甘草、白芍单味饮片的供试溶液和栝楼桂枝汤物质基准(S1~S15)供试品溶液,按照“2.3.1”项色谱条件进样分析,计算栝楼桂枝汤物质基准和单味饮片中5个差异性成分的含量,结果见表4。芍药苷、甘草苷、芒柄花苷、甘草素、甘草酸在饮片-物质基准之间的平均转移率分别为74.74%、60.06%、34.54%、40.46%、51.65%,结果见表5。转移率均在均值±3SD范围内,这些成分在饮片-物质基准的转移率较为稳定。
参照2020年版《中华人民共和国药典》四部通则2201浸出物测定法中“热浸法”测定,精密吸取“2.1”项下水浴浓缩后的滤液25 mL置已干燥至恒重的蒸发皿中,水浴蒸干后,105 ℃干燥3 h,置干燥器中冷却30 min,迅速精密称量。
出膏率=mV/Mv×100%,其中m表示干膏质量(g),M表示饮片质量(g),v代表取样体积(mL),V代表汤液总体积(mL)。各批次栝楼桂枝汤出膏率见表6。出膏率均值为14.53%,RSD为4.6%,未出现离散数据,说明栝楼桂枝汤物质基准制备工艺较合理,稳定可行,未造成明显的批间差异。
本研究通过指纹图谱结合化学模式识别方法筛选出甘草酸、甘草素、芍药苷、芒柄花苷、甘草苷为栝楼桂枝汤量值传递关键指标成分。方中天花粉(栝楼)为君药,现代研究表明栝楼中氨基酸类成分为栝楼桂枝汤发挥抗缺血性脑卒中的主要活性成分群之一[21-22],但氨基酸类成分极性较强,与其他药材化学成分极性差异较大,较难使用C18色谱柱同时分析包括君药在内的主要活性成分,后续将开展君药活性成分在栝楼桂枝汤中的量值规律传递研究,完善质量体系。白芍酸寒苦泄,益阴敛营,柔肝止痛,为臣药;甘草用于治疗四肢挛急疼痛,为使药。研究表明白芍中的芍药苷对缺血性脑损伤有保护作用[23-24],甘草中甘草酸、甘草素、甘草苷能分别通过抑制炎症反应、抗氧化、抗凋亡等途径对局灶性脑缺血/再灌注起到神经保护作用[25-28]。课题组前期通过研究大脑中动脉栓塞(MCAO)大鼠口服栝楼桂枝汤后主要化学成分发现甘草酸、甘草素、芍药苷、芒柄花苷和甘草苷均为汤剂主要的入血原型成分。此外,白芍和甘草的主要有效成分芍药苷、甘草苷等,均为2020年版《中华人民共和国药典》单味药项下规定的质控成分[29]。综合考虑全方和VIP排序筛选结果,选择具有代表性且与药效相关性强的甘草酸、甘草素、芍药苷、芒柄花苷、甘草苷作为汤剂质控成分,可较为准确地反映原方质量。
本实验采用光电二极管阵列检测器对供试品溶液和混合对照品溶液进行全波长扫描,发现在254 nm波长下混合对照品溶液中5个成分响应值较高;供试品溶液基线平稳,色谱信息丰富,故选择254 nm作为栝楼桂枝汤物质基准指纹图谱和多成分含量测定的检测波长。在流动相系统考察中,分别考察了甲醇-水、乙腈-水和乙腈-0.1%甲酸水3个流动相系统对样品指纹图谱分离的影响,结果发现乙腈-0.1%甲酸水的分离效果最好,拖尾因子满足多成分含量测定的要求,且分析方法准确、可靠,重复性良好。
本实验分别考察了不同类型提取溶剂(纯水、甲醇、乙醇)、不同浓度提取溶剂(10%、30%、50%、70%、90%甲醇水溶液,10%、30%、50%、70%、90%乙醇水溶液)的提取效率。结果采用50%甲醇水溶液提取效率最高,故选择50%甲醇水溶液作为提取溶剂。此外,本研究也考察了50%甲醇水溶液用量(30、50、70 mL)、提取时间(20、30、40 min)和提取次数(1次、2次、3次)对提取效率的影响,最终选择50%甲醇水溶液用量30 mL,超声提取时间30 min,常温提取2次时提取效率较高。
芍药苷、甘草苷、芒柄花苷、甘草酸、甘草素5个成分转移率的高低反映了各成分水溶性的差异,也可能由于不同药味成分间相互作用从而导致其溶解性发生变化[30]。其中芍药苷和甘草酸转移率较高且基本稳定,而芒柄花苷的转移率较低,可能因为芒柄花苷在水中溶解度较低,经过前煮等处理过程含量损失较大,导致含量的下降。
15批栝楼桂枝汤物质基准的出膏率为13.32%~15.59%,平均出膏率为14.59%,全部批次出膏率都处于均值的90%~110%,整体上较为稳定。
综上,本研究建立了栝楼桂枝汤指纹图谱和含量测定方法,出膏率、指标成分含量和转移率等关键质量属性均能在栝楼桂枝汤饮片-物质基准之间稳定传递,可较好反映栝楼桂枝汤汤剂的整体性,为完善栝楼桂枝汤质量控制体系及后期复方颗粒剂的研发提供参考。
