Objective:

To explore the method of pharmacognosic identification of four medicinal seeds, such as Plantaginis Semen from two kinds of original plant,Allii Fistulosi Semen, Allii Tuberosi Semen, and to digitize the identification characteristics.

Methods:

The seeds of the four medicinal plants were studied by means of type microscope and light microscope, including the appearance(positive view, bottom view, hilum view and surface feature), internal morphology(cross profile and longitudinal profile) and the characters of micromorphology(transverse section, powder). The characteristics of the four medicinal seeds were summarized, and the identification points were characterized by digital method.

Results:

The seeds of the four medicinal plants could be identified according to the appearance, internal morphology and the characters of micromorphology.

Conclusion:

This study lays a foundation for the variety identification and provides reference for the standard formulation and quality control of the four medicinal seeds. It also provides ideas for digital characterization of seed medicinal materials.

运用体式显微镜、光学显微镜等仪器设备从外观性状(正面观、底面观、脐面观、表面特征)、内部结构(横剖面、纵剖面)及显微特征(横切面、粉末)对车前子等4种药用植物种子进行深入的性状和显微鉴别研究,初步探讨车前子等4种药用植物种子的鉴别特征,并用数字化的形式对鉴别点进行表征。

结果:

车前子等4种药用植物种子可根据外观性状、内部结构及显微特征等进行鉴别。

结论:

本研究为车前子等4种种子类药材的品种鉴定奠定基础,并为这4种药用植物种子的监督检验和标准修订提供参考,也为种子类药材的数字化表征提供参考。

目的:

探讨车前子2个基源种子、葱子和韭菜子等4种种子类药材生药学鉴定方法,将鉴定特征数字化。

方法:

运用体式显微镜、光学显微镜等仪器设备从外观性状(正面观、底面观、脐面观、表面特征)、内部结构(横剖面、纵剖面)及显微特征(横切面、粉末)对车前子等4种药用植物种子进行深入的性状和显微鉴别研究,初步探讨车前子等4种药用植物种子的鉴别特征,并用数字化的形式对鉴别点进行表征。

结果:

车前子等4种药用植物种子可根据外观性状、内部结构及显微特征等进行鉴别。

结论:

本研究为车前子等4种种子类药材的品种鉴定奠定基础,并为这4种药用植物种子的监督检验和标准修订提供参考,也为种子类药材的数字化表征提供参考。

Objective:

To explore the method of pharmacognosic identification of four medicinal seeds, such as Plantaginis Semen from two kinds of original plant,Allii Fistulosi Semen, Allii Tuberosi Semen, and to digitize the identification characteristics.

Methods:

The seeds of the four medicinal plants were studied by means of type microscope and light microscope, including the appearance(positive view, bottom view, hilum view and surface feature), internal morphology(cross profile and longitudinal profile) and the characters of micromorphology(transverse section, powder). The characteristics of the four medicinal seeds were summarized, and the identification points were characterized by digital method.

Results:

The seeds of the four medicinal plants could be identified according to the appearance, internal morphology and the characters of micromorphology.

Conclusion:

This study lays a foundation for the variety identification and provides reference for the standard formulation and quality control of the four medicinal seeds. It also provides ideas for digital characterization of seed medicinal materials.

车前子首载于《神农本草经》,属甘寒之品,入肝、小肠、肺、肾、膀胱经,具有清热利尿、渗湿通淋、明目祛痰之功效,可用于治疗小便不利、湿热泄泻、伶俐涩痛、眼目昏花、咳嗽痰多、目赤肿痛等症。因具有药食同源之性,国家卫生健康委员会将其列入可用于保健食品的物品名单里。目前,全世界车前科植物共有3个属,我国仅有车前属1属,有17种2变种1变形。我国传统车前子商品分为大粒车前子和小粒车前子,其中大粒车前子为车前科植物车前Plantago asiatica L.的种子,小粒车前子为车前科植物大车前Plantago major L.或平车前Plantago depressa Willd.的种子,市场上小粒车前子的商品来源主要为大车前。2020年版《中华人民共和国药典》(简称《中国药典》)收载车前子为车前科植物车前或平车前的干燥成熟种子。大车前在《维吾尔药材标准》(1993年版)中被作为车前子的来源收载。相关文献报道中,有基于指纹图谱结合化学计量学对车前草与车前子药材的质量评价研究,为车前草和车前子的专属性鉴别以及产地区分提供参考,并可用于车前草和车前子药材质量的综合评价,也有学者利用指纹图谱技术可有效鉴别车前子及其混伪品。此外,还有学者采用中药微性状法对车前子进行真伪鉴别研究,以及采用荧光体式显微技术、扫描电镜技术鉴别车前子的真伪等。
韭菜子为百合科葱属植物,在我国资源较为丰富,河南、安徽、山东等地韭菜子产量较高,始载于《名医别录》,味辛、甘,性温,归肝、肾经,可温补肝肾、壮阳固精,用于肝肾亏虚、腰膝酸痛、阳痿遗精、遗尿尿频、白浊带下。2020年版《中国药典》收载韭菜子为百合科植物韭菜Allium tuberosu Rottl. ex Spreng.的干燥成熟种子。葱子原名葱实,是百合科葱属植物葱Allium fistulosum L.的种子,始载于《神农本草经》,列为中品,味辛,性温,可补中益精、明目散风,用于肾虚、目眩、风寒感冒等。二者性状相近,常互为混淆品,有学者从性状、显微、以及电镜扫描等方面对其鉴别进行了相关研究,具有一定参考价值。此外,也有根据韭菜子的化学反应和紫外光谱等方面的鉴别研究,还有学者基于SERS的挥发物筛选式葱属植物鉴别分类研究方法为葱属植物分类研究提供参考信息。
数字化是现代科学的象征,中医药想跟上时代步伐,走上现代化就必需具备,因此,数字化和智能化技术应该与中医药发展紧密相关。将传统中药数字化是时代发展的必然趋势,越来越多的前沿科技和手段应用于中药材的识别和质量控制中,如显微成像系统、高清数码技术、客户端应用软件等,为中药材鉴别的智能化及数字化发展奠定基础。本文运用高清数码相机、体式显微镜及光学显微镜等设备对车前子2个基源种子、葱子和韭菜子4种药用植物种子的外观性状、剖面构造、横切面及粉末显微特征进行数字化表征,更直观系统地展现其识别特点,为这4种药用种子的日常鉴别与质量评价提供一定的试验依据,同时为相关标准的增修订、质量控制及进一步的数字化研究工作提供参考。

试验样品收集自中药材专业市场,经山西省检验检测中心罗晋萍主任药师鉴定(详见表1)。试验样品保存于山西省检验检测中心中药标本馆。
挑选能够代表种子特征与特征范围的样品,合理摆放,使其呈现自然稳定的状态,对其进行观测并拍摄。正面观:种脐朝向下端,将种子最大表面轮廓的一面朝上,拍摄成像。脐面观:为表现种脐在种子上的位置、比例、性状等,将种子种脐朝上进行拍摄,可视具体情况将种子进行切割。侧面观:种脐朝向下端,将种子的侧面朝上,即种子较窄的一面朝上,拍摄其侧面观。由于种子性状的多样性,可根据实际情况拍摄相应的特征面,如底面观、背面观等。表面特征:最大表面轮廓朝上,观察种子表面的微构造并拍摄成像。
车前子:呈不规则长圆形或三角状长圆形,略扁,长约2 mm,宽约1 mm。表面黄棕色至黑褐色,有细皱纹,一面有灰白色凹点状种脐。质硬,气微,味淡。见图1-A图2-A
车前子(平车前子):呈椭圆形、不规则长圆形,略扁,长约1.0 mm,宽约0.6 mm。表面黄棕色至黑褐色,有细皱纹,一面有灰白色凹点状种脐。质硬,气微,味淡。见图1-B图2-B
葱子:呈类三角状卵形,长3~4 mm,宽2~3 mm。表面黑色,略有光泽。一面微凹,另一面隆起,隆起面有棱线1~2条,光滑或有疏皱缩纹,放大镜下观察表面可见细密纹理。基部有2个小突起,较短的突起为种脐,顶端灰棕色或白色;较长的突起顶端为珠孔。质坚硬,种皮较薄,剖面可见灰白色胚乳,富油性。体轻,质坚硬,气特异。见图3-A图4-A
韭菜子:呈半圆形或半卵圆形,略扁,长2~4 mm,宽1.5~3 mm。表面黑色,一面突起,粗糙,有细密的网状皱纹,另一面微凹,皱纹不甚明显。顶端钝,基部稍尖,有点状突起的种脐。质硬,气特异,味微辛。见图3-B图4-B
纵剖面:沿种子的长轴方向纵向切面。横剖面:沿种子的短轴方向的中部横向切面。观察种子以上2种切面,并拍摄成像。
车前子:纵剖面呈长椭圆形,子叶2,种皮薄。横剖面呈椭圆形。切面可见乳白色的胚乳及子叶。见图5-A

葱子:纵剖面呈半卵圆形。种皮菲薄,胚乳灰白色,富油性。胚白色,卷曲,环形,子叶1枚。胚根细小,位于种脐端。除胚外,胚乳几乎占种子的全部。横剖面呈长条形,可见胚的2个断面。见图5-C
韭菜子:纵剖面呈半圆形或半卵圆形。种皮薄,胚乳灰白色,富油性。胚卷曲,环形;胚根细小,位于种脐端。除胚外,胚乳几乎占种子的全部。横剖面呈长条形,可见胚的2个断面。见图5-D
沿种子长轴方向的中部位置将种子横向切片,装片。横切面制片方法按《中国药典》显微鉴别法进行。应用光学显微镜进行观察并拍摄成像。
车前子:种皮由2层细胞组成;外层为黏液层,外壁薄容易被破坏;内层为色素层,含棕黃色色素,类方形或类长方形,壁略弯曲。胚乳细胞多角形,类方形或类长方形,胞腔内充满糊粉粒。子叶细胞类圆形或类方形,排列整齐,含小糊粉粒和油滴。见图6-A
车前子(平车前子):种皮由2层细胞组成:外层为黏液层,外壁薄容易被破坏:内层为色素层,含棕黃色色素,类方形或类长方形,壁略弯曲。胚乳细胞多角形,类方形或类长方形,胞腔内充满糊粉粒。子叶细胞类圆形或类方形,排列整齐,含小糊粉粒和油滴。见图6-B
葱子:种皮表皮细胞外侧壁均匀地向外突起,细胞棕黑色,长方形,细胞壁厚,被有薄角质层,细胞腔含暗褐色物质。内表皮为数列棕色薄壁细胞,细胞壁弯曲。胚乳占种子大部分,胚乳细胞大,排列紧密,细胞壁念珠状增厚,有大型纹孔,细胞腔内可见淡黄色晶体。胚细胞排列紧密,胞腔内充满糊粉粒。见图7-A
韭菜子:种皮表皮细胞棕黑色,壁厚,外被角质层,内含深棕色物。其下为数列纵横交错排列的棕黄色种皮薄壁细胞,细胞壁弯曲。胚乳占种子大部分,胚乳细胞大,排列紧密,细胞壁念珠状增厚,胞腔内充满糊粉粒及油滴。胚细胞排列紧密,胞腔内充满糊粉粒。见图7-B
粉末制片方法按《中国药典》显微鉴别法进行。应用光学显微镜进行观察并拍摄成像。
车前子:粉末深黄棕色。种皮外表皮细胞断面观类方形或略切向延长,细胞壁黏液质化。种皮内表皮细胞黄棕色,表面观类长方形,直径5~19 μm,长约至83 μm,壁薄,微波状,常作镶嵌状排列。胚乳细胞壁甚厚,充满细小糊粉粒。子叶细胞类圆形或类方形,排列整齐,由薄壁细胞组成,充满小糊粉粒和油滴。见图8-A
车前子(平车前子):粉末深黄棕色。种皮外表皮细胞断面观类方形或略切向延长,细胞壁黏液质化。种皮内表皮细胞表面观类长方形,直径5~15 μm,长11~45 μm,壁薄,常作镶嵌状排列。胚乳细胞壁甚厚,充满细小糊粉粒。子叶细胞类圆形或类方形,排列整齐。见图8-B
葱子:粉末灰黑色。种皮表皮细胞棕褐色,多角形、类圆形或不规则形,表面具网状纹理。内表皮薄壁细胞棕色,纵横交错排列,较大,多破碎。胚乳细胞众多,多破碎,类圆形,有较多大的类圆形或长圆形纹孔,壁增厚,胞腔内可见淡黄色晶体。胚细胞成片,长方形、正方形或类圆形,内含糊粉粒。见图9-A
韭菜子:粉末灰黑色。种皮表皮细胞棕色或棕褐色,长条形、多角形、类圆形或不规则形,长44~132 μm,宽13~45 μm,表面具网状纹理。内表皮薄壁细胞棕色,纵横交错排列,较大,多破碎。胚乳细胞众多,多破碎,有较多大的类圆形或长圆形纹孔,壁增厚,内含糊粉粒及油滴。胚细胞成片,长方形或正方形,内含糊粉粒。见图9-B
2020年版《中国药典》一部收载的车前子有2个基原,为车前科植物车前或平车前的干燥成熟种子。在生活实际应用中,由于车前子细小,极易与一些其他植物来源细小的种子或果实混淆,如荆芥的果实、党参的种子、北柴胡的果实等,肉眼识别困难,掺伪现象时有发生,给大众安全用药带来不便。而药典标准尽管对其性状及粉末显微鉴别做了描述,不够直观,且两基源之间也只对种皮内表皮细胞做了尺寸的区别。本文经过上述研究实验,发现除种皮内表皮细胞尺寸的区别外,二者之间的尺寸以及性状均有较大差别,车前子呈不规则长圆形或三角状长圆形,略扁,个头偏大,平车前子呈椭圆形、不规则长圆形,略扁,个头偏小,可作为主要鉴别点,并图文结合,更好地方便了车前子及平车前子的鉴别研究。
葱子和韭菜子同为百合科,外观极为相似,互为易混品,给生活中实际应用带来不便,但经过上述研究实验,发现二者之间通过不同的鉴别方式是有很多鉴别点的。首先,气味不同,葱子放在嘴里嚼食会有浓郁的葱香,韭菜子嚼食后是浓郁的韭菜香味。其次,外观不同,葱子外观是三角状的卵形,一面微凹另一面隆起,隆起的一面有1到2条棱线,且表面有光泽,具细密纹理。韭菜子则是扁卵形,它的顶部钝,基部稍长,而且隆起面上有网状皱纹。其中,葱子表面的细密纹理与韭菜子表面的网状皱纹在二者的剖面观特征以及横切面显微特征中均可作为显著鉴别点,为其鉴别提供依据。
本文在前期研究基础上,借鉴植物学有关形态鉴定与分类的术语和研究方法,应用体式显微镜和光学显微镜等仪器设备及数码成像技术手段,从正面、侧面、腹面、表面等外观性状,横剖面和纵剖面特征,与横切面、粉末的显微特征对车前子的2种基原、葱子和韭菜子这对易混品进行了生药学研究和数字化表征,对其鉴别要点有了更直观形象的展现和区分,可对这4种种子类药材的日常监管、标准提高、科普宣传等提供参考。
我国中医药文化源远流长,中药材的质量又是中药之根本。中药材性状和显微鉴定作为一种独特的中药质量分析方法,在保证中药质量、保障中药临床用药安全有效中发挥着重大作用。在国家政策的大力支持下,随着显微技术与数字化技术的发展,中国食品药品检定研究院联合多家药品检验检测机构,共同开展了中药民族药数字标本平台的建设工作,尤其是在传统中药材鉴定方法与数字化和智能化结合方面,做了一些探索和尝试。数字化将图像信息与文字描述精准对应,传统中药材鉴定中主观、抽象化的语言描述被转化为直观、真实的图像,有助于信息的保存、知识的普及、文化的传递,可有效缓解资金及时间成本,为生药鉴定传承与创新奠定良好基础。
