Objective:

To establish an HPLC method of the content and related substances of compound amino acid injection(3AA).

Methods:

RP-HPLC was adopted to determine compound amino acid injection(3AA), combining with the use of two-dimensional column switching-LC/MSn method was applied to separate and identify the impurities. The determination was performed on Capcell PAK AQ C18(250 mm×4.6 mm, 3 μm) column with 0.2 mol·L-1 sodium dihydrogen phosphate solution (adjusted pH to 2.8 with phosphoric acid) -acetonitrile (98∶2) as mobile phase at the flow rate of 1.0 mL·min-1. The column temperature was 40 ℃, and the detection wavelength was 210 nm. And the injection volume was 20 μL. The LC/MSn method was performed on a Thermo Accucore AQ C18 (100 mm×4.6 mm, 2.6 μm) column with 0.1% formic acid solution as mobile phase A, 0.1% formic acid solution-acetonitrile as mobile phase B, at a flow rate of 0.4 mL·min-1, and at a column temperature of 40 ℃. The mass spectrometry conditions were performed using an ESI ionisation source in the positive-ion scanning mode with a scan range of m/z 100-1 000, and the secondary mass spectrum was carried out by data-dependent scanning.

Results:

The related substances were completely separated from the main constituents in RP-HPLC. The standard curve of valine was linear over the range of 1.263-5.050 mg·mL-1, with the average recovery of 99.0% (n=9). The standard curve of isoleucine was linear over the range of 1.350-5.402 mg·mL-1, with the average recovery of 99.4%(n=9). The standard curve of leucine was linear over the range of 1.647-6.588 mg·mL-1, with the average recovery of 99.5%(n=9). The main impurities in the three batches of samples were all process impurities introduced from the raw materials, with methionine content of 4.344 μg·mL-1, 3.751 μg·mL-1, 4.503 μg·mL-1, respectively, phenylalanine content of 4.636 μg·mL-1, 4.889 μg·mL-1, 4.753 μg·mL-1, respectively. The maximum single impurity contents were 0.01%, 0.02% and 0.01%, respectively.

Conclusion:

The method is proved by the methodology validation that it can be used for the quality control of compound amino acid injection(3AA).

目的:

建立复方氨基酸注射液(3AA)的主成分和有关物质测定的高效液相色谱方法。

方法:

采用反相高效液相色谱法并结合二维柱切换LC/MSn法对制剂中主要杂质进行分离和结构鉴定,建立复方氨基酸注射液(3AA)中缬氨酸、异亮氨酸和亮氨酸及其有关物质含量检测方法。采用Capcell PAK AQ C18(250 mm×4.6 mm,3 μm)色谱柱,以0.2 mol·L-1磷酸二氢钠溶液(用磷酸调pH至2.8)-乙腈(98∶2)为流动相,流速1.0 mL·min-1,柱温40 ℃,检测波长210 nm,进样量20 μL。采用Thermo Accucore AQ C18(100 mm×4.6 mm,2.6 μm)色谱柱,以0.1%甲酸溶液为流动相A,以0.1%甲酸溶液-乙腈为流动相B,流速0.4 mL·min-1,柱温40 ℃;质谱条件采用ESI电离源,正离子扫描模式,扫描范围为m/z 100~1 000,二级质谱采用数据依赖型扫描开展。

结果:

主峰和相邻杂质峰分离度良好,缬氨酸的线性范围为1.263~5.050 mg·mL-1,平均回收率(n=9)为99.0%;异亮氨酸的线性范围为1.350~5.402 mg·mL-1,平均回收率(n=9)为99.4%;亮氨酸的线性范围为1.647~6.588 mg·mL-1,平均回收率(n=9)为99.5%。3批样品中主要杂质均为原料引入的工艺杂质,其中甲硫氨酸含量分别为4.344、3.751、4.503 μg·mL-1,苯丙氨酸含量分别为4.636、4.889、4.753 μg·mL-1。其他单个未知杂质分别为0.01%、0.02%、0.01%。

结论:

经方法学验证,本方法可用于复方氨基酸注射液(3AA)的质量控制。

目的:

建立复方氨基酸注射液(3AA)的主成分和有关物质测定的高效液相色谱方法。

方法:

采用反相高效液相色谱法并结合二维柱切换LC/MSn法对制剂中主要杂质进行分离和结构鉴定,建立复方氨基酸注射液(3AA)中缬氨酸、异亮氨酸和亮氨酸及其有关物质含量检测方法。采用Capcell PAK AQ C18(250 mm×4.6 mm,3 μm)色谱柱,以0.2 mol·L-1磷酸二氢钠溶液(用磷酸调pH至2.8)-乙腈(98∶2)为流动相,流速1.0 mL·min-1,柱温40 ℃,检测波长210 nm,进样量20 μL。采用Thermo Accucore AQ C18(100 mm×4.6 mm,2.6 μm)色谱柱,以0.1%甲酸溶液为流动相A,以0.1%甲酸溶液-乙腈为流动相B,流速0.4 mL·min-1,柱温40 ℃;质谱条件采用ESI电离源,正离子扫描模式,扫描范围为m/z 100~1 000,二级质谱采用数据依赖型扫描开展。

结果:

主峰和相邻杂质峰分离度良好,缬氨酸的线性范围为1.263~5.050 mg·mL-1,平均回收率(n=9)为99.0%;异亮氨酸的线性范围为1.350~5.402 mg·mL-1,平均回收率(n=9)为99.4%;亮氨酸的线性范围为1.647~6.588 mg·mL-1,平均回收率(n=9)为99.5%。3批样品中主要杂质均为原料引入的工艺杂质,其中甲硫氨酸含量分别为4.344、3.751、4.503 μg·mL-1,苯丙氨酸含量分别为4.636、4.889、4.753 μg·mL-1。其他单个未知杂质分别为0.01%、0.02%、0.01%。

结论:

经方法学验证,本方法可用于复方氨基酸注射液(3AA)的质量控制。

Objective:

To establish an HPLC method of the content and related substances of compound amino acid injection(3AA).

Methods:

RP-HPLC was adopted to determine compound amino acid injection(3AA), combining with the use of two-dimensional column switching-LC/MSn method was applied to separate and identify the impurities. The determination was performed on Capcell PAK AQ C18(250 mm×4.6 mm, 3 μm) column with 0.2 mol·L-1 sodium dihydrogen phosphate solution (adjusted pH to 2.8 with phosphoric acid) -acetonitrile (98∶2) as mobile phase at the flow rate of 1.0 mL·min-1. The column temperature was 40 ℃, and the detection wavelength was 210 nm. And the injection volume was 20 μL. The LC/MSn method was performed on a Thermo Accucore AQ C18 (100 mm×4.6 mm, 2.6 μm) column with 0.1% formic acid solution as mobile phase A, 0.1% formic acid solution-acetonitrile as mobile phase B, at a flow rate of 0.4 mL·min-1, and at a column temperature of 40 ℃. The mass spectrometry conditions were performed using an ESI ionisation source in the positive-ion scanning mode with a scan range of m/z 100-1 000, and the secondary mass spectrum was carried out by data-dependent scanning.

Results:

The related substances were completely separated from the main constituents in RP-HPLC. The standard curve of valine was linear over the range of 1.263-5.050 mg·mL-1, with the average recovery of 99.0% (n=9). The standard curve of isoleucine was linear over the range of 1.350-5.402 mg·mL-1, with the average recovery of 99.4%(n=9). The standard curve of leucine was linear over the range of 1.647-6.588 mg·mL-1, with the average recovery of 99.5%(n=9). The main impurities in the three batches of samples were all process impurities introduced from the raw materials, with methionine content of 4.344 μg·mL-1, 3.751 μg·mL-1, 4.503 μg·mL-1, respectively, phenylalanine content of 4.636 μg·mL-1, 4.889 μg·mL-1, 4.753 μg·mL-1, respectively. The maximum single impurity contents were 0.01%, 0.02% and 0.01%, respectively.

Conclusion:

The method is proved by the methodology validation that it can be used for the quality control of compound amino acid injection(3AA).

复方氨基酸注射液(3AA)是由3种支链氨基酸缬氨酸(Val)、亮氨酸(Leu)和异亮氨酸(Ile)配制而成的灭菌水溶液,输注后能纠正血浆中支链氨基酸和芳香氨基酸失衡,防止因脑内芳香氨基酸浓度过高引起的肝昏迷[1-2],促进蛋白质合成以及减少蛋白质分解。主要用于肝胆外科手术前后以及其他原因引起的肝性脑病、重症肝炎以及肝硬化、慢性活动性肝炎[3-4]。该品种在各国药典中均未收载,现行标准收载于《卫生部药品标准(二部)》第六册(生化药品第一分册)[5]。标准中采用氨基酸分析仪或高效液相色谱法进行含量测定,未给出具体色谱条件,且标准中未收载有关物质检查项,存在一定安全风险。2020年版《中华人民共和国药典》、EP 11.0、USP 43及JP 18均收载了包括缬氨酸、亮氨酸和异亮氨酸原料的其他氨基酸(茚三酮阳性物质、有关物质)检查方法[6-9],多采用薄层色谱法(茚三酮显色)或氨基酸分析仪法(柱后茚三酮衍生)检测,方法仅针对氨基基团显色,无法全面反映制剂中杂质的整体情况[7,10]
近年来,针对多种氨基酸同时检测的方法大多仍采用衍生化法,分为柱前衍生化和柱后衍生化,通常使用邻苯二甲醛(OPA)、异硫氰酸苯酯(PITC)、2,4-二硝基氟苯(DNFB)及茚三酮等与氨基酸的氨基进行反应[11-13],但各种衍生化反应均存在一定的不足,如衍生产物不稳定、前处理复杂及存在对色谱柱损伤等[14-16]。本文中采用反相高效液相色谱技术,建立了同时测定复方氨基酸注射液(3AA)主成分含量及有关物质的新方法。通过柱切换的二维液相色谱联用高分辨质谱技术,对产品中主要有关物质进行结构鉴定。经方法验证显示,建立的方法灵敏度高,专属性强,准确度高,可有效用于复方氨基酸注射液(3AA)主成分及有关物质含量测定要求,对进一步保障产品质量及临床用药的安全有效提供方法参考。
UltiMate-3000高效液相色谱仪(Thermo Fisher公司),Thermo Orbitrap XL高分辨质谱仪(Thermo Fisher公司),XP205型十万分之一电子天平(METTLER TOLEDO公司),Milli-Q超纯水器(IQ7000,Merck Millipore公司),pH计(Seven Excellence,Mettler Toledo公司)。
乙腈(北京百灵威科技有限公司,纯度99.9%,批号L850U47),磷酸二氢钠(北京市通广精细化工公司,纯度99.0%,批号20200723),磷酸(TEDIA公司,批号19080735),水为超纯水(实验室自制,大于18.2 MΩ·cm)。
对照品:缬氨酸、异亮氨酸、亮氨酸、甲硫氨酸、苯丙氨酸(纯度均为99.9%,批号为140624-201506,中国食品药品检定研究院);市售复方氨基酸注射液(3AA)(批号为1020010101、1020010102、1020010103)。
采用Capcell PAK AQ C18 (250 mm×4.6 mm,3 μm)色谱柱,以0.2 mol·L-1磷酸二氢钠溶液(用磷酸调pH至2.8)-乙腈(98∶2)为流动相,流速1.0 mL·min-1,柱温40 ℃,检测波长210 nm,进样量20 μL。
精密称取缬氨酸、异亮氨酸、亮氨酸的对照品各适量,加流动相溶解并稀释制成每1 mL约含缬氨酸2.5 mg、异亮氨酸2.7 mg、亮氨酸3.3 mg的混合溶液,作为含量测定用对照品溶液。精密称取甲硫氨酸、苯丙氨酸对照品各适量,加流动相溶解并稀释制成每1 mL中甲硫氨酸、苯丙氨酸各含25 μg的混合溶液,作为有关物质检查用对照品溶液。
精密量取复方氨基酸注射液(3AA)2 mL,置10 mL量瓶中,用流动相稀释至刻度,摇匀,滤过,取续滤液作为含量测定用供试品溶液。取复方氨基酸注射液(3AA)滤过,取续滤液作为有关物质检查用供试品溶液。
精密量取复方氨基酸注射液(3AA)1 mL,置100 mL量瓶中,用流动相稀释至刻度,摇匀,滤过,取续滤液,即得。
取流动相作为空白溶液,滤过,即得。
一维液相色谱条件同“2.1”项。
二维液相色谱条件:采用Thermo Accucore AQ C18(100 mm×4.6 mm,2.6 μm)色谱柱,流动相A为0.1%甲酸溶液,流动相B为0.1%甲酸溶液-乙腈,梯度洗脱[0~(tR+1) min(tR为目标杂质在一维色谱系统的保留时间),2% B(脱盐处理);(tR+1) min~(tR+5) min,90%B;(tR+5) min~(tR+7) min,90%B;(tR+7.1) min~(tR+10) min,2%B],流速0.4 mL·min-1,柱温40 ℃。
二维柱切换构型图如图1所示,其色谱系统工作流程:①将切换阀设为位置A,采用一维液相色谱系统(泵1及色谱柱1)分离供试品中相关杂质,同时二维液相色谱系统(泵2)对二维分析色谱柱(色谱柱2)进行预平衡;②当采用一维液相色谱系统分离的目标杂质出峰后,将切换阀设为位置B,色谱柱1流出组分进入切换阀1中定量环(loop),出峰结束随即切回初始位置A;③采用二维液相色谱系统将定量环中的目标组分反冲至第二维分析色谱柱(色谱柱2)中,并通过切换阀2脱盐处理后进入下一级质谱检测器进行相应的结构分析。
采用ESI电离源,正离子扫描模式,喷雾电压4.5 kV,加热电压25 V,离子导入电压(skimmer电压) 20 V,鞘气流速30 arb,吹扫气流速10 arb,离子传输毛细管温度375 ℃,扫描范围为m/z 100~1 000;二级质谱采用数据依赖型扫描(data dependent MS/MS),用于监测一级质谱全扫描中已确定质量数离子的二级质谱,二级质谱碰撞诱导解离归一化能量值设置为35%。
采用Thermo U3000-Orbitrap XL二维液质联用系统,一维液相色谱采用“2.1”项色谱条件进行,将一维液相色谱图中目标杂质切入二维液相色谱柱,使用“2.3.1”项的二维液相色谱条件进行分析,洗脱液进Orbitrap XL质谱检测器,使用“2.3.2”项的质谱条件进行目标化合物的一级、二级质谱数据采集。目标杂质色谱图见图2
采用建立的高效液相色谱方法并结合二维柱切换LC/MSn法对样品进行有关物质检查,样品中较大杂质分别标注为杂质Ⅰ和杂质Ⅱ,结合强酸(2 mol·L-1盐酸溶液)、强碱(2 mol·L-1氢氧化钠溶液)、氧化(10%过氧化氢溶液)、高温(105 ℃加热)及光照(紫外光365 nm)条件强制降解试验分析,2个杂质均为工艺杂质而非降解生成,首先对复方氨基酸注射液(3AA)杂质Ⅰ的一、二级质谱图进行分析,确定加合物及二级碎片裂解规律。如图3所示,杂质Ⅰ所得m/z 150.058 15分子离子峰,根据高分辨数据推测元素组成与[M+H]+ C5H12NO2S相匹配(偏差值-1.17×10-6)。二级质谱图主要碎片离子包括m/z 133.11、87.89离子,其中m/z 133.11离子为母离子丢失1分子NH3后的产物,该离子进一步分别丢失HCOOH后得到m/z 87.89离子,根据高分辨质量数元素匹配及碎片信息推测,鉴定杂质Ⅰ为甲硫氨酸。杂质Ⅱ所得m/z 166.086 10分子离子峰,根据高分辨数据推测元素组成与[M+H]+ C9H12NO2相匹配(偏差值-0.93×10-6)。二级质谱图主要碎片离子包括m/z 149.04、120.00离子,在该正离子模式下,m/z 149.04离子为母离子丢失1分子NH3后的产物,另外母离子丢失HCOOH后得到m/z 120.00离子,推测鉴定杂质Ⅱ为苯丙氨酸,相关质谱图见图4
分别量取空白溶液、混合对照品溶液和供试品溶液,按“2.1”项下色谱条件进样测定,记录色谱图。在该色谱条件下,缬氨酸、异亮氨酸和亮氨酸分别在3.82、6.39和6.94 min出峰(图5)。取复方氨基酸注射液(3AA) (批号1020010101)各2 mL置10 mL量瓶中,分别进行强酸降解(2 mol·L-1盐酸溶液2 mL,破坏2 h)、强碱降解(2 mol·L-1氢氧化钠溶液2 mL,破坏2 h)、氧化降解(10%过氧化氢溶液2 mL,放置2 h)、高温降解(105 ℃加热破坏2 h)和光照降解(紫外光365 nm下照射24 h),酸碱降解后进行中和,用流动相稀释至刻度,摇匀,滤过。取各强制降解后供试溶液测定,结果表明,3个氨基酸完全分离且峰形良好,空白溶液不干扰待测组分测定,供试溶液在各降解条件下较为稳定,无较明显杂质生成。色谱图中杂质峰与主成分峰的分离度良好。
精密称取缬氨酸、异亮氨酸和亮氨酸的对照品各适量,加流动相溶解并稀释得到系列浓度的混合对照品溶液(缬氨酸质量浓度为1.263、1.512、2.016、2.525、5.050 mg·mL-1,异亮氨酸质量浓度为1.350、1.621、2.161、2.701、5.402 mg·mL-1,亮氨酸质量浓度为1.647、1.976、2.635、3.294、6.588 mg·mL-1)。取系列浓度的混合对照品溶液各20 μL注入液相色谱仪,记录峰面积,以浓度(X)为横坐标、色谱峰面积(Y)为纵坐标进行线性回归,得复方氨基酸注射液(3AA)中缬氨酸、异亮氨酸和亮氨酸的线性回归方程。见表1
精密称取对照品缬氨酸505.02 mg、亮氨酸658.81 mg、异亮氨酸540.20 mg,置同一100 mL量瓶中,加流动相溶解并稀释至刻度,摇匀。精密量取5 mL置10 mL量瓶中,用流动相稀释至刻度,摇匀。按“2.1”项色谱条件进样分析。3个主成分连续进样测定6次,峰面积的RSD分别为0.31%、0.29%和0.31%,表明仪器的精密度良好。
取复方氨基酸注射液(3AA) (批号1020010101)6份,分别按“2.2.2”项下的方法制备供试品溶液,进样测定,结果缬氨酸、亮氨酸、异亮氨酸的平均含量分别为98.13%、97.91%和97.30%,RSD分别为0.89%、0.89%、0.87%,表明该方法重复性良好。
取复方氨基酸注射液(3AA) (批号1020010101),按“2.2.2”项下方法制备供试品溶液,在室温下分别于0、2、4、6、8、12 h进行测定。结果供试品溶液中缬氨酸、异亮氨酸、亮氨酸浓度的RSD分别为0.13%、0.11%、0.11%,表明供试品溶液在室温条件下放置12 h,稳定性良好。
分别精密称取对照品缬氨酸252.13 mg、亮氨酸269.71 mg、异亮氨酸330.23 mg,置同一100 mL量瓶中,加流动相溶解并稀释至刻度,摇匀,作为混合对照品溶液。精密量取复方氨基酸注射液(3AA)(批号1020010101)1 mL,9份,每3份为一组,分别置10 mL量瓶中,每组分别精密加入混合对照品溶液4、5、6 mL,用流动相稀释至刻度,摇匀,滤过,取续滤液,即得低、中、高浓度的供试溶液各3份,按“2.1”项下条件进样测定,计算平均回收率和RSD。结果显示,在标示浓度的80%~120%范围内,缬氨酸平均回收率(n=9)为99.0%,RSD=0.90%;异亮氨酸平均回收率(n=9)为99.4%,RSD=0.82%;亮氨酸平均回收率(n=9)为99.5%,RSD=0.77%,表明该方法准确度良好。
精密称取甲硫氨酸、苯丙氨酸的对照品各约25 mg,置同一100 mL量瓶中,用流动相溶解并稀释至刻度,摇匀,作为混合对照品储备液。精密量取上述储备液逐级进行稀释,得到质量浓度为2.5、5、10、25、50 μg·mL-1的系列混合对照品溶液。取各浓度混合对照品溶液20 μL注入液相色谱仪,记录峰面积,以质量浓度(X)为横坐标、色谱峰面积(Y)为纵坐标进行线性回归,得甲硫氨酸和苯丙氨酸的线性回归方程。将混合对照品储备液加流动相逐级稀释成所需的不同浓度,进样测定,以信噪比(S/N)为10∶1时的浓度为定量限,以信噪比(S/N)为3∶1时的浓度为检测限。结果见表2
精密称取甲硫氨酸、苯丙氨酸的对照品各适量,加流动相溶解并稀释制成每1 mL中均含25 μg的混合对照品溶液,连续进样6次,记录色谱图,甲硫氨酸、苯丙氨酸色谱峰面积的RSD(n=6)分别为0.74%和0.47%。精密量取复方氨基酸注射液(3AA)(批号1020010101) 6份,分别滤过,取续滤液作为供试品溶液,按“2.1”项下条件进样测定,甲硫氨酸、苯丙氨酸的含量均值(n=6)分别为3.92、4.83 μg·mL-1,其RSD分别为1.2%和0.60%。表明仪器精密度及方法重复性均良好。
取由复方氨基酸注射液(3AA) (批号1020010101)制备的供试品溶液,在室温下分别于0、2、4、6、8、12 h进行测定,记录色谱图。供试品溶液中甲硫氨酸峰面积的RSD为0.92%,苯丙氨酸峰面积的RSD为0.62%,表明供试品溶液在室温条件下稳定性良好。
精密量取复方氨基酸注射液(3AA) (批号1020010101)5 mL,9份,每3份为一组,分别置10 mL量瓶中,每组分别精密加入“2.6.1”项下混合对照品储备液0.8、1、1.2 mL,用流动相稀释至刻度,摇匀,滤过,取续滤液,即得低、中、高浓度供试溶液各3份。按“2.1”项下色谱条件分别进样,测定甲硫氨酸、苯丙氨酸的含量,计算回收率。在限度浓度的80%~120%范围内,甲硫氨酸的平均回收率(n=9)为102.2%(RSD=0.67%),苯丙氨酸的平均回收率(n=9)为99.8%(RSD=0.70%),表明方法准确度良好。
取不同批号的复方氨基酸注射液(3AA),按“2.2”项方法制备供试品溶液、对照品溶液及对照溶液,并按“2.1”项色谱条件进行分析,测定峰面积。采用外标法计算缬氨酸、异亮氨酸、亮氨酸以及杂质甲硫氨酸、苯丙氨酸的含量,未知杂质含量采用异亮氨酸主成分自身对照法计算,结果见表3
参考美USP 43中收载的缬氨酸、亮氨酸和异亮氨酸原料药有关物质检测方法,缬氨酸原料有关物质检测方法采用氨基柱,在基于磷酸盐-乙腈体系下填料流失较为严重,稳定性低,方法耐用性有一定限制;亮氨酸和异亮氨酸原料有关物质检测方法均采用反相C18柱,经实验测定各成分峰形较好,本研究在此基础上进行方法优化。
本实验中通过优化流动相组成、pH、柱温及色谱柱规格等,对各实验条件下主成分分离保留及杂质分析进行了评估,结果显示,在磷酸盐缓冲液(pH 2.8)-乙腈条件下,各色谱峰对称性更优;柱温升高至40 ℃,在聚合物包被型AQ色谱柱(250 mm×4.6 mm,3 μm)条件下,亮氨酸和异亮氨酸分离度可达到2.5以上,各主成分及杂质分离效果更优,并且在高水相条件下该方法色谱柱耐用性更好。方法使用紫外末端吸收,不需要特殊设备,通用性较强。
由于建立的液相色谱方法中采用磷酸盐为流动相,不适用于直接进行LC/MS分析。二维液相色谱-质谱联用技术通过在线联用的自动化操作,可在一维液相色谱体系中使用质谱不兼容流动相对目标杂质进行分析,通过柱切换技术实现对组分的捕集和脱盐,并转移至二维液相色谱中进行质谱采集,使用适宜的流动相进行组分结构鉴定。本研究采用在线二维液相质谱联用技术对复方氨基酸注射液(3AA)中2个主要杂质峰进行了结构解析,确定了产品中2个主要杂质为原料药的工艺杂质甲硫氨酸和苯丙氨酸。
本文建立了反相高效液相色谱法并利用柱切换技术联合高分辨质谱鉴定了复方氨基酸注射液(3AA)中2个主要杂质,对复方氨基酸注射液(3AA)进行含量及有关物质检测。该方法专属性强,准确度高,可有效地控制复方氨基酸注射液(3AA)的产品质量。
