Objective:

To establish a quantitative analysis of multi-components by single marker (QAMS) method for the simultaneous determination of nine effective components including phenolic acids and flavonoids (neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, rutoside, isoquercitrin, kaemperol-3-O-rutinoside, 3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid), in Solidaginis Herba.

Methods:

Ultra-high performance liquid chromatography was developed on an ACQUITY UPLC® HSS T3 (100 mm×2.1 mm, 1.8 μm) column with acetonitrile (A)-0.1% phosphoric acid solution (B) as mobile phase in gradient elution. The flow rate was 0.3 mL·min-1, the detection wavelength was 327 nm, the column temperature was 30 ℃, and the injection volume was 1 μL. Chlorogenic acid and rutoside were chosen as the internal standards for phenolic acids and flavonoids respectively to establish the determine correction factors of neochlorogenic acid, cryptochlorogenic acid,3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid isoquercitrin and kaemperol-3-O-rutinoside, respectively, by multi-point correction method and their contents were calculated. The results were compared with the results of external standard method to verify their accuracy.

Results:

Nine components showed good linear relationships within their own ranges(r≥0.999 5), whose average recoveries were 98.2%-101.7% with the RSDs of 1.0%-1.7%. The contents of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, rutoside, isoquercitrin, kaemperol-3-O-rutinoside, 3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid and 4,5-O-dicaffeoylquinic acid, in nine batches of samples were 0.008%-0.014%, 0.055%-0.097%, 0.010%-0.019%, 0.191%-0.412%, 0.018%-0.035%, 0.116%-0.50%, 0.028%-0.048%, 0.111%-0.235% and 0.113%-0.182%. The results obtained by QAMS were consistent with those of external standard method(relative average deviations less than 2.8%).

Conclusion:

The simple, accurate and reproducible method can be used for the quality control of Solidaginis Herba.

目的:

建立一测多评法同时测定一枝黄花中酚酸类及黄酮类共9个成分(新绿原酸、绿原酸、隐绿原酸、芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸和4,5-O-二咖啡酰奎宁酸)的含量。

方法:

采用超高效液相色谱法,采用ACQUITY UPLC® HSS T3 (100 mm×2.1 mm,1.8 μm)色谱柱,以乙腈(A)-0.1%磷酸溶液(B)为流动相进行梯度洗脱,流速0.3 mL·min-1,检测波长327 nm,柱温30 ℃,进样量1 μL。酚酸类成分以绿原酸为内标,黄酮类成分以芦丁为内标,采用多点校正法,分别建立新绿原酸、隐绿原酸、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸和异槲皮苷、山柰酚-3-O-芸香糖苷的相对校正因子,并计算其含量。测定结果与外标法结果进行比较,验证其准确性。

结果:

9个成分在各自范围内线性关系良好(r≥0.999 5);平均加样回收率分别为98.2%~101.7%,RSD分别为1.0%~1.7%。9批样品中新绿原酸、绿原酸、隐绿原酸、芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸和4,5-O-二咖啡酰奎宁酸的含量范围分别为0.008%~0.014%、0.055%~0.097%、0.010%~0.019%、0.191%~0.412%、0.018%~0.035%、0.116%~0.250%、0.028%~0.048%、0.111%~0.235%、0.113%~0.182%。一测多评法测定结果与外标法结果一致(相对平均偏差<2.8%)。

结论:

该方法简便、准确,重复性好,可用于一枝黄花药材的质量控制。

目的:

建立一测多评法同时测定一枝黄花中酚酸类及黄酮类共9个成分(新绿原酸、绿原酸、隐绿原酸、芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸和4,5-O-二咖啡酰奎宁酸)的含量。

方法:

采用超高效液相色谱法,采用ACQUITY UPLC® HSS T3 (100 mm×2.1 mm,1.8 μm)色谱柱,以乙腈(A)-0.1%磷酸溶液(B)为流动相进行梯度洗脱,流速0.3 mL·min-1,检测波长327 nm,柱温30 ℃,进样量1 μL。酚酸类成分以绿原酸为内标,黄酮类成分以芦丁为内标,采用多点校正法,分别建立新绿原酸、隐绿原酸、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸和异槲皮苷、山柰酚-3-O-芸香糖苷的相对校正因子,并计算其含量。测定结果与外标法结果进行比较,验证其准确性。

结果:

9个成分在各自范围内线性关系良好(r≥0.999 5);平均加样回收率分别为98.2%~101.7%,RSD分别为1.0%~1.7%。9批样品中新绿原酸、绿原酸、隐绿原酸、芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸和4,5-O-二咖啡酰奎宁酸的含量范围分别为0.008%~0.014%、0.055%~0.097%、0.010%~0.019%、0.191%~0.412%、0.018%~0.035%、0.116%~0.250%、0.028%~0.048%、0.111%~0.235%、0.113%~0.182%。一测多评法测定结果与外标法结果一致(相对平均偏差<2.8%)。

结论:

该方法简便、准确,重复性好,可用于一枝黄花药材的质量控制。

Objective:

To establish a quantitative analysis of multi-components by single marker (QAMS) method for the simultaneous determination of nine effective components including phenolic acids and flavonoids (neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, rutoside, isoquercitrin, kaemperol-3-O-rutinoside, 3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid), in Solidaginis Herba.

Methods:

Ultra-high performance liquid chromatography was developed on an ACQUITY UPLC® HSS T3 (100 mm×2.1 mm, 1.8 μm) column with acetonitrile (A)-0.1% phosphoric acid solution (B) as mobile phase in gradient elution. The flow rate was 0.3 mL·min-1, the detection wavelength was 327 nm, the column temperature was 30 ℃, and the injection volume was 1 μL. Chlorogenic acid and rutoside were chosen as the internal standards for phenolic acids and flavonoids respectively to establish the determine correction factors of neochlorogenic acid, cryptochlorogenic acid,3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid isoquercitrin and kaemperol-3-O-rutinoside, respectively, by multi-point correction method and their contents were calculated. The results were compared with the results of external standard method to verify their accuracy.

Results:

Nine components showed good linear relationships within their own ranges(r≥0.999 5), whose average recoveries were 98.2%-101.7% with the RSDs of 1.0%-1.7%. The contents of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, rutoside, isoquercitrin, kaemperol-3-O-rutinoside, 3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid and 4,5-O-dicaffeoylquinic acid, in nine batches of samples were 0.008%-0.014%, 0.055%-0.097%, 0.010%-0.019%, 0.191%-0.412%, 0.018%-0.035%, 0.116%-0.50%, 0.028%-0.048%, 0.111%-0.235% and 0.113%-0.182%. The results obtained by QAMS were consistent with those of external standard method(relative average deviations less than 2.8%).

Conclusion:

The simple, accurate and reproducible method can be used for the quality control of Solidaginis Herba.

一枝黄花为菊科植物一枝黄花Solidago decurrens Lour.的干燥全草。收载于2020年版《中华人民共和国药典》一部,其性凉,味辛、苦,具有清热解毒、疏散风热之功效。用于喉痹、乳蛾、咽喉肿痛、疮疖肿毒、风热感冒[1]。多产于我国南方,浙江、福建、江西、四川、贵州、湖南、湖北、广西、贵州、云南等地广为分布。因地域不同,一枝黄花别名很多,如江浙通名金锁钥,福建名百根草,江西又有一支枪、一支箭等名称[2]。作为民间常用的清热解毒传统中药,一枝黄花组方众多,常用的中成药有复方一枝黄花喷雾剂、鼻宁喷雾剂、姜黄消痤搽剂、妇平胶囊、穿黄消炎片、肾炎片、感冒炎咳灵片(糖浆)、复方桑菊感冒片、感冒解毒(片)颗粒、清热感冒颗粒、复方野菊感冒颗粒、罗浮山百草油等。现代研究表明,一枝黄花具有抗菌、消炎、利尿、祛痰平喘、抗病毒、抗氧化等多种药理作用。其主要化学成分有黄酮类、苯丙酸类、皂苷类等[3-6]。大量研究表明以芦丁为代表的黄酮类成分和以绿原酸为代表的酚酸类成分是其主要药效成分[7-13]。目前2020年版《中华人民共和国药典》一部仅以芦丁单个成分作为含量测定指标,对中药材质量评价具有一定的局限性。已有文献报道的多成分含量测定研究也仅以2、3个代表性成分作为指标[14-16],且均为外标法(external standard method,ESM),ESM测定对对照品的需求较大,部分对照品不易得到,检测成本较高,且相对耗时,限制了一枝黄花多指标质量控制的建立和应用推广。
一测多评(quantitative analysis of multi-components by single marker,QAMS)的方法是基于中药同类有效成分间可能存在内在关系和规律性,利用有效成分内在的函数关系和比例关系,通过单一成分来获得多个成分的同步定量结果的中药质量评价方法[17]。自2006年首次提出以来,在中药多指标质量评价方面,得到广泛应用和认可[18-21],2020年版《中华人民共和国药典》一部已有丹参、黄连、淫羊藿、银杏叶提取物、银杏叶系列制剂、咳特灵片(胶囊)、宫血宁胶囊、镇脑宁胶囊等17个中药材、饮片、中药提取物及制剂采用QAMS法控制其质量。目前未见一枝黄花药材的相关研究报道。因此,本研究参考QAMS法,酚酸类成分以绿原酸为内标,黄酮类成分以芦丁为内标,同时测定一枝黄花中9个成分的含量,以期为进一步完善一枝黄花药材及其制剂的质量评价方法提供参考。
1290 Infinity超高效液相色谱仪(Agilent公司,含OpenLab CDS色谱工作站、四元泵、自动进样器和DAD检测器);Mettler Toledo ML204电子天平(梅特勒-托利多仪器(上海)有限公司,万分之一);Sartorius BT 25S电子天平(赛多利斯科学仪器北京有限公司,十万分之一);KQ-500DE型数控超声波清洗器(昆山市超声仪器有限公司);Milli-Q Direct 16纯水系统(Millipore公司)。
对照品新绿原酸(批号240008-202005,纯度≥98%),上海鸿永生物科技公司;隐绿原酸(批号AF9080321,纯度≥98%),成都埃法生物科技有限公司;3,4-O-二咖啡酰奎宁酸(批号3089,纯度≥98%),上海诗丹德科技有限公司;3,5-O-二咖啡酰奎宁酸(批号111782-202208,纯度95.9%)、4,5-O-二咖啡酰奎宁酸(批号111894-202205,纯度94.9%)、山柰酚-3-O-芸香糖苷(批号112007-202103,纯度94.0%)、异槲皮苷(批号111809-201804,纯度97.2%)、芦丁(批号100080-202012,纯度91.6%)、绿原酸(批号110753-202119,纯度96.3%),中国食品药品检定研究院。
乙腈(Sigma-Aldrich公司)、磷酸(上海麦克林生化科技股份有限公司)均为色谱纯,水为Milli-Q制备的超纯水,其余试剂均为分析纯。
9批一枝黄花药材来源广西、云南、贵州、江西等地,经江西省药品检验检测研究院陈伟康主任中药师鉴定为菊科植物一枝黄花Solidago decurrens Lour.的干燥全草。样品编号为S1~S9。
精密称取对照品新绿原酸13.60 mg、绿原酸15.58 mg、隐绿原酸31.05 mg、芦丁65.68 mg、异槲皮苷16.16 mg、山柰酚-3-O-芸香糖苷61.29 mg、3,4-O-二咖啡酰奎宁酸14.14 mg、3,5-O-二咖啡酰奎宁酸61.32 mg、4,5-O-二咖啡酰奎宁酸对照品33.28 mg,分别置5、5、10、20、5、25、5、20、10 mL量瓶中,加75%甲醇水使溶解并稀释至刻度,摇匀,即得质量浓度分别为2.665 6、3.000 7、3.042 9、3.008 1、3.141 5、2.304 5、2.771 4、2.940 3和3.158 3 mg·mL-1的单一成分溶液,即得。
精密吸取新绿原酸、绿原酸、隐绿原酸、芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸的对照品储备液适量,置25 mL量瓶中,加75%甲醇水稀释至刻度,摇匀,即得上述成分质量浓度分别为13.328 0、60.014 0、20.326 6、300.810 0、31.415 0、230.450 0、36.915 0、176.418 0和157.915 0 μg·mL-1的混合对照品溶液。
取一枝黄花粉末(过3号筛)约1 g,精密称定,置具塞锥形瓶中,精密加入75%甲醇水10 mL,称量,超声(功率500 W,频率40 kHz)处理1 h,放冷,用75%甲醇水补足减失的量,摇匀,过0.22 μm微孔滤膜,取续滤液,即得。
采用ACQUITY UPLC®HSS T3 (100 mm×2.1 mm,1.8 μm)色谱柱,以乙腈(A)-0.1%磷酸溶液(B)为流动相,梯度洗脱(0~7 min,5%A→20%A;7~15 min,20%A→30%A),流速0.3 mL·min-1,检测波长327 nm,柱温30 ℃,进样量1 μL。在上述条件下,一枝黄花中9个化学成分峰与相邻峰的分离度均>1.5,结果见图1
精密吸取“2.1.1”项下新绿原酸对照品储备液0.025、0.062、0.100、0.125、0.188、0.250、0.125 mL,绿原酸对照品储备液0.100、0.250、0.400、0.500、0.750、1.000、0.500 mL,隐绿原酸对照品储备液0.033、0.083、0.133、0.167、0.250、0.333、0.167 mL,芦丁对照品储备液0.500、1.250、2.000、2.500、3.750、5.000、2.500 mL,异槲皮苷对照品储备液0.050、0.125、0.200、0.250、0.375、0.500、0.250 mL,山柰酚-3-O-芸香糖苷对照品储备液0.500、1.250、2.000、2.500、3.750、5.000、2.500 mL,3,4-O-二咖啡酰奎宁酸对照品储备液0.067、0.167、0.267、0.333、0.500、0.667、0.333 mL、3,5-O-二咖啡酰奎宁酸对照品储备液0.300、0.750、1.200、1.500、2.250、3.000、1.500 mL,4,5-O-二咖啡酰奎宁酸对照品储备液0.250、0.625、1.000、1.250、1.875、2.500、1.250 mL,一一对应分别置25、25、25、25、25、25、10 mL量瓶中,加75%甲醇水稀释至刻度,摇匀,即得7个浓度点的混合对照品溶液;分别按“2.2”项下色谱条件进样分析,以各对照品的质量浓度(X,μg·mL-1)为横坐标,相应峰面积(Y)为纵坐标,绘制标准曲线并进行回归计算,得9个成分的线性回归方程,结果见表1,可知9个成分在各自范围内线性关系良好。
精密吸取“2.1.2”项下混合对照品溶液1 μL,按“2.2”项下色谱条件连续进样测定6次,测得新绿原酸、绿原酸、隐绿原酸、芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸峰面积的RSD(n=6)为0.10%~0.42%,表明仪器精密度良好。
取同一批一枝黄花药材(S9号样品)粉末(过3号筛)6份,每份约1 g,精密称定,分别按“2.1.3”项下方法制备供试品溶液,按“2.2”项下色谱条件进行测定,测得新绿原酸、绿原酸、隐绿原酸、芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸的平均含量(n=6)分别为0.014%、0.089%、0.018%、0.368%、0.031%、0.230%、0.045%、0.210%和0.169%,RSD(n=6)为0.09%~0.56%,结果表明该方法重复性良好。
精密吸取同一供试品溶液(S9号样品)1 μL,于溶液制备后0、1、2、4、8、12、24 h分别按“2.2”项下色谱条件进样测定,测得新绿原酸、绿原酸、隐绿原酸、芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸峰面积的RSD(n=6)为0.20%~0.82%,结果表明供试品溶液在24 h内稳定。
精密称取已测知含量的S9号一枝黄花药材的粉末(过3号筛)6份,每份约0.5 g,精密称定,分别精密加入含新绿原酸0.006 8 mg·mL-1、绿原酸0.045 6 mg·mL-1、隐绿原酸0.009 0 mg·mL-1、芦丁0.189 0 mg·mL-1、异槲皮苷0.017 0 mg·mL-1、山柰酚-3-O-芸香糖苷0.121 8 mg·mL-1、3,4-O-二咖啡酰奎宁酸0.023 7 mg·mL-1、3,5-O-二咖啡酰奎宁酸0.106 6 mg·mL-1、4,5-O-二咖啡酰奎宁酸0.086 8 mg·mL-1的混合对照品溶液10 mL,按“2.1.3”项下方法制备供试溶液,并按“2.2”项下色谱条件进样测定,计算回收率,结果见表2
本研究采用多点校正法计算相对校正因子,即取“2.3”项下9个成分7个浓度点的进样浓度和对应峰面积,酚酸类成分以绿原酸为内标,黄酮类成分以芦丁为内标,按公式Fi=(As×Ci)/(Cs×Ai)计算另外7个待测成分的相对校正因子。
其中,Fi为待测成分的相对校正因子,CsCi分别为内标物和待测成分的浓度,AsAi分别为内标物和待测成分的峰面积。结果,新绿原酸、隐绿原酸、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸的平均相对校正因子(n=7)分别为1.142 1、1.362 5、0.820 1、1.193 7、1.070 2、0.764 2、0.809 5,RSD均<1.0%。
取“2.1.2”项下混合对照品溶液,考察3个型号色谱仪(Agilent 1290 Infinity、Agilent 1290 Infinity Ⅱ、Waters ACQUITY UPLCTM)和3根色谱柱[ACQUITY UPLC® HSS T3(100 mm×2.1 mm,1.8 μm)、ACQUITY UPLC® BEH C18(100 mm×2.1 mm,1.7 μm)、InfinityLab Poroshell 120 Aq-C18(100 mm×2.1 mm,2.7 μm)]对新绿原酸、隐绿原酸、异槲皮苷、山柰酚-3-O-芸香糖苷、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸以及4,5-O-二咖啡酰奎宁酸相对校正因子的影响,结果见表3。由表3可知,新绿原酸、异槲皮苷等7个成分相对校正因子的RSD(n=9)均<1.0%,表明不同色谱仪和不同色谱柱对新绿原酸、异槲皮苷等7个成分相对校正因子的影响较小。
本实验考察了各成分相对保留时间值(ri/s=tRi/tRs)在不同仪器、色谱柱下的情况,结果见表4。由此可知,各成分相对保留时间的RSD均<3.5%,故本试验采用相对保留时间进行色谱峰定位。
取不同批号的9批一枝黄花药材粉末(过3号筛)各2份,分别按“2.1.3”项下方法制备供试品溶液,并按“2.2”项下色谱条件进样测定,采用ESM和QAMS法计算含量,QAMS法计算公式为Ci=(Fi×[(Cs×Ai)/As),其中CiCs分别为待测成分及内标物的浓度,Fi为待测成分的相对校正因子,AiAs分别为待测成分及内标物的峰面积,结果见表5。由此可知,ESM和QAMS法所得结果相近,相对平均偏差(relative average deviation,RD)<2.8%,表明QAMS法可用于一枝黄花的含量测定。
QAMS法提出者王智民课题组认为QAMS可应用于中药复杂体系中同类成分之间的评价,但在不同结构母核的化学成分类别之间,应用QAMS法仍需探讨[22]。经结构分析,一枝黄花中的6个酚酸类成分具有相同的结构母核,3个黄酮类成分也具有相同的结构母核。同时对9个成分进行光谱扫描,6个酚酸类成分光谱图基本一致,3个黄酮类成分光谱图也基本一致。基于此,选择性质较稳定,对照品易得,含量较大的绿原酸和芦丁分别作为酚酸类和黄酮类的内标物,保证结果的准确性,试验结果也证实以2个物质作为内标物的测定结果和ESM的结果非常接近。
将混合对照品溶液于200~400 nm波长范围内进行光谱扫描,结果6个酚酸类成分在327 nm附近有最大吸收,黄酮类成分在354 nm附近有最大吸收,样品中新绿原酸和隐绿原酸含量较低,在354 nm附近响应低,黄酮类成分在327 nm处响应相对较好,因此选择酚酸类成分的最大吸收波长327 nm作为检测波长。
考察了不同提取方式(加热回流提取和超声提取),不同提取溶剂(30%甲醇水、70%甲醇水、甲醇、乙醇),不同提取时间(0.5、1、1.5 h),不同料液比(1∶5、1∶10、1∶20)。结果加热回流提取时温度较高,酚酸类化合物受热不稳定,2类化合物的70%甲醇水提取效率明显高于甲醇和乙醇,超声0.5 h的含量偏低,超声1 h和1.5 h的含量基本一致,1∶10和1∶20的料液比结果无明显区别,但料液比为1∶20时,供试品溶液中含量低的待测成分峰面积较小,故最终选择采用料液比1∶10,70%甲醇水超声处理1 h作为供试品溶液的制备方法。
考察了4个流动相系统(乙腈-水、乙腈-0.1%磷酸溶液、乙腈-0.1%醋酸溶液、甲醇-水),结果以乙腈-0.1%磷酸溶液为流动相时,能够获得较好的色谱峰峰形及分离效果,同时分析时间也最短。
综上,本研究采用QAMS法同时测定一枝黄花中新绿原酸、绿原酸、隐绿原酸、3,4-O-二咖啡酰奎宁酸、3,5-O-二咖啡酰奎宁酸、4,5-O-二咖啡酰奎宁酸6个酚酸类和芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷3个黄酮类成分的含量,方法操作简便高效,结果准确可靠,减少了试剂试药的消耗,节约了研究检测成本,可为进一步完善和提高一枝黄花药材的质量标准提供参考依据。
