Objective:

To establish a rapid molecular identification method for DNA fingerprinting-immunocolloidal gold test strips of Chinese herbal medicine human placentophagy genetic markers and to develop an integrated quality control gene detection kit.

Methods:

Self-developed method and reagents could be for template DNA extraction from human placentophagy. Species-specific primers were designed according to the human mtDNA cytb gene, and the 5’ ends of the primers were labeled with FAM and Biotin, respectively. The optimal annealing temperature and cycle number of PCR were screened to establish a DNA fingerprinting molecular identification method. To develop PCR gene detection reagent premixes, to prepare DNA clones of control herbs using molecular cloning and gene sequencing techniques and to utilize immunocolloidal gold test strips for result detection. Finally, a rapid gene detection kit integrating DNA extraction reagent, PCR gene detection reagent premixes, control herb DNA clone solution, blank control solution and test strips was developed, and evaluated for specificity, reproducibility, sensitivity and stability.

Results:

The template DNA extracted by self-developed DNA extraction reagents were all in the concentration of 150-280 ng·μL-1, the purities were all in the range of 1.8-1.9, and there were no trail in the agarose gel electrophoresis. And the immunocolloidal gold test strips showed two red bands for human placentophagy control herb and authentic product, and one red band for easy-mix product and blank control at annealing temperature of 59 ℃ and 20 cycles for human placentophagy specific primers. The agarose gel electrophoresis was verified to be correct. The DNA sequencing results of the control herbs were 100% homologous with human mtDNA cytb gene. The self-developed integrated rapid gene detection kit was specific, reproducible, stable and sensitive at 0.1 ng·μL-1.

Conclusion:

The DNA fingerprinting-immunocolloidal gold test strip molecular identification method can specifically, accurately, rapidly and visually identify the authenticity of human placentophagy, and the integrated rapid gene detection kit provides a regulated and standardized testing method for quality control of human placentophagy. Therefore, it is more suitable for on-site field testing and universal promotion.

目的:

建立中药材紫河车遗传标志物DNA指纹-免疫胶体金试纸条快速分子鉴定方法,研发一体化质量控制基因检测试剂盒。

方法:

自主研发紫河车模板DNA提取的方法及试剂,根据人mtDNA cytb基因设计种属特异性引物,引物的5’端分别用FAM、Biotin标记,筛选PCR最佳退火温度及循环次数,建立DNA指纹分子鉴定方法,研发PCR基因检测试剂预混液,采用分子克隆及基因测序技术制备对照药材DNA克隆液,利用免疫胶体金试纸条进行结果检测,开发出集“DNA提取试剂、PCR基因检测试剂预混液、对照药材DNA克隆液、空白对照液、试纸条”为一体的快速基因检测试剂盒,并进行特异性、重现性、灵敏性、稳定性的评价。

结果:

自主研发的DNA提取试剂提取的模板DNA质量浓度均在150~280 ng·μL-1,纯度均在1.8~1.9,琼脂糖凝胶电泳无拖尾。紫河车特异性引物在退火温度59 ℃、循环20次时;免疫胶体金试纸条显示:紫河车对照药材及正品出现2条红色条带,易混品及空白对照出现1条红的条带。琼脂糖凝胶电泳验证正确。对照药材DNA测序结果与人mtDNA cytb基因同源性100%。自主研发的一体化快速基因检测试剂盒的特异性强,重现性好,稳定性好,灵敏度为0.1 ng·μL-1

结论:

DNA指纹-免疫胶体金试纸条分子鉴定方法能够特异、准确、快速、可视化地对紫河车的真伪进行鉴定,一体化快速基因检测试剂盒为紫河车质量控制提供规范化、标准化的检测方式,更适合现场实地检测,可普遍推广使用。

目的:

建立中药材紫河车遗传标志物DNA指纹-免疫胶体金试纸条快速分子鉴定方法,研发一体化质量控制基因检测试剂盒。

方法:

自主研发紫河车模板DNA提取的方法及试剂,根据人mtDNA cytb基因设计种属特异性引物,引物的5’端分别用FAM、Biotin标记,筛选PCR最佳退火温度及循环次数,建立DNA指纹分子鉴定方法,研发PCR基因检测试剂预混液,采用分子克隆及基因测序技术制备对照药材DNA克隆液,利用免疫胶体金试纸条进行结果检测,开发出集“DNA提取试剂、PCR基因检测试剂预混液、对照药材DNA克隆液、空白对照液、试纸条”为一体的快速基因检测试剂盒,并进行特异性、重现性、灵敏性、稳定性的评价。

结果:

自主研发的DNA提取试剂提取的模板DNA质量浓度均在150~280 ng·μL-1,纯度均在1.8~1.9,琼脂糖凝胶电泳无拖尾。紫河车特异性引物在退火温度59 ℃、循环20次时;免疫胶体金试纸条显示:紫河车对照药材及正品出现2条红色条带,易混品及空白对照出现1条红的条带。琼脂糖凝胶电泳验证正确。对照药材DNA测序结果与人mtDNA cytb基因同源性100%。自主研发的一体化快速基因检测试剂盒的特异性强,重现性好,稳定性好,灵敏度为0.1 ng·μL-1

结论:

DNA指纹-免疫胶体金试纸条分子鉴定方法能够特异、准确、快速、可视化地对紫河车的真伪进行鉴定,一体化快速基因检测试剂盒为紫河车质量控制提供规范化、标准化的检测方式,更适合现场实地检测,可普遍推广使用。

Objective:

To establish a rapid molecular identification method for DNA fingerprinting-immunocolloidal gold test strips of Chinese herbal medicine human placentophagy genetic markers and to develop an integrated quality control gene detection kit.

Methods:

Self-developed method and reagents could be for template DNA extraction from human placentophagy. Species-specific primers were designed according to the human mtDNA cytb gene, and the 5’ ends of the primers were labeled with FAM and Biotin, respectively. The optimal annealing temperature and cycle number of PCR were screened to establish a DNA fingerprinting molecular identification method. To develop PCR gene detection reagent premixes, to prepare DNA clones of control herbs using molecular cloning and gene sequencing techniques and to utilize immunocolloidal gold test strips for result detection. Finally, a rapid gene detection kit integrating DNA extraction reagent, PCR gene detection reagent premixes, control herb DNA clone solution, blank control solution and test strips was developed, and evaluated for specificity, reproducibility, sensitivity and stability.

Results:

The template DNA extracted by self-developed DNA extraction reagents were all in the concentration of 150-280 ng·μL-1, the purities were all in the range of 1.8-1.9, and there were no trail in the agarose gel electrophoresis. And the immunocolloidal gold test strips showed two red bands for human placentophagy control herb and authentic product, and one red band for easy-mix product and blank control at annealing temperature of 59 ℃ and 20 cycles for human placentophagy specific primers. The agarose gel electrophoresis was verified to be correct. The DNA sequencing results of the control herbs were 100% homologous with human mtDNA cytb gene. The self-developed integrated rapid gene detection kit was specific, reproducible, stable and sensitive at 0.1 ng·μL-1.

Conclusion:

The DNA fingerprinting-immunocolloidal gold test strip molecular identification method can specifically, accurately, rapidly and visually identify the authenticity of human placentophagy, and the integrated rapid gene detection kit provides a regulated and standardized testing method for quality control of human placentophagy. Therefore, it is more suitable for on-site field testing and universal promotion.

紫河车,为健康产妇娩出之胎盘。具有温肾补精,益气养血的功效,用于虚劳赢廋,阳痿遗精,久咳虚喘,面色萎黄,食少气短。由于紫河车来源紧张,价格不菲,因此市面上出现用羊胎盘、猪胎盘等充当紫河车的现象[1],普通消费者很难从外观进行准确识别。而且临床一般使用紫河车粉入药,不仅给掺假提供了机会,同时也增加了检测方法的难度,严重影响临床用药的有效性。2010年版《中华人民共和国药典》(以下简称《中国药典》)一部仅记载了“性状”项,也许由于检测的标准太低,主观性强,专属性差,2015年版《中国药典》一部不再收载紫河车,目前没有国家质量标准。
中药质量控制是传统中医药得到现代化和全球化发展的前提。中药质量标志物(Q-marker)由2016年刘昌孝院士提出[2]。随着分子生物学技术的发展,学者们逐渐认识到物种的差异归根结底是遗传物质DNA的差异,中药材的质量鉴定从最初通过感官、显微、理化等方面,逐步走向分子鉴定方向。
DNA指纹多采用聚合酶链式反应(PCR)技术对多种生物样品进行人工合成不同的DNA片段,片段的大小和数量在不同生物间不相同,对每一个体的分析都是相对独特的[3],近年来这种基于遗传标志物的技术在遗传分析、鉴定种质等方面得到广泛应用[4]。2020年版《中国药典》一部收录了川贝母、蕲蛇、乌梢蛇以及金钱白花蛇[5]的PCR分子鉴定方法,本团队已经建立了貂心、兔睾丸、鹿心、乳鹿、鹿茸、鹿胎、阿胶[6-15]等PCR快速鉴定方法。但结果都是利用琼脂糖凝胶电泳来检测,费时费力费钱,需要贵重仪器的配合。而免疫胶体金试纸条是一种快速的免疫学测定方法,具有简单、快速,可视化,低成本等特点,不需要贵重仪器的配合,在中药材紫河车真伪鉴定中目前未见文献报道。本研究从DNA水平为紫河车的真伪鉴定提供一种特异灵敏的鉴定方法,为紫河车质量标准的制定提供理论基础,并研发一体化快速检测试剂盒,适合普遍推广应用。
紫河车对照药材(批号121567-201202),购自中国食品药品检定研究院;紫河车正品1~5(20200026~20200030),由吉林市食品药品检验所提供;易混品鹿、猪、马、牛、羊、驴、狗胎盘,由吉林雷宁食品药品检测技术服务有限公司提供。
D3024R高速冷冻离心机(SCILOGEX公司);D1008E掌上离心机(大龙创兴实验仪器有限公司);T100 PCR仪(BIORAD公司);JY300E电泳仪、JY-SPCT电泳槽(北京君意东方电泳设备有限公司);2020D紫外凝胶成像分析仪(BIORAD公司);6000+微量核酸蛋白分析仪(QUAWELL公司);CP214万分之一电子天平(奥豪斯仪器有限公司);ZKW-C恒温水浴锅(上海树立仪器仪表有限公司)。
2×Taq PCR Master Mix、λDNA/Hind Ⅲ、DNA凝胶回收试剂盒、pGM-T载体连接试剂盒、DH5α感受态细胞、质粒DNA小量提取试剂盒、Amp、X-Gal、IPTG、6×DNA上样缓冲液,北京天根生化科技有限公司;100 bp Marker I,北京鼎国生物技术有限公司;引物,上海生物工程有限公司合成。免疫胶体金试纸条,武汉君诺德生物技术有限公司。
随机取各物种胎盘样品5 g,研磨成粉末后各取100 mg置于1.5 mL离心管中。加入细胞裂解缓冲液P1[10%十二烷基磺酸钠(SDS)10 mL,1 mol·L-1三羟甲基氨基甲烷盐酸盐(Tris-HCl,pH 7.5) 1 mL,0.5 mol·L-1乙二胺四乙酸(EDTA,pH 8.0) 2 mL,氯化钠(NaCl)0.58 g,2 0 mg·mL-1蛋白酶K(PK)1 mL,RNA酶(RNase)5 μL,加灭菌ddH2O定容至100 mL]500 μL,充分混匀,55 ℃水浴1 h。室温冷却后倒入DNA层析柱中,10 000 r·min-1离心3 min,弃去过滤液。层析柱放回,加入漂洗缓冲液P2 [5 mol·L-1乙酸钾(KAc) 26 μL,1 mol·L-1 Tris-HCl(pH 7.5) 18 μL,0.5 mol·L-1 EDTA(pH 8.0) 3 μL,无水乙醇480 μL,无菌ddH2O 73 μL]600 μL,10 000 r·min-1离心1 min,弃去过滤液。重复漂洗1次,弃去过滤液。层析柱放回,10 000 r·min-1离心2 min。将层析柱转移入另一干净离心管中,加入100 μL溶解液P3(无菌ddH2O),室温放置20 min后,10 000 r·min-1离心2 min;离心液即各物种胎盘模板DNA溶液,置-20 ℃保存备用。另取紫河车对照药材100 mg,同法制成阳性对照药材模板DNA溶液。
取各物种胎盘模板DNA溶液1 μL,利用微量核酸蛋白分析仪检测260和280 nm处的吸收度,所有样品的DNA质量浓度均在150~280 ng·μL-1,纯度均在1.8~1.9,满足PCR实验要求。取各物种胎盘模板DNA溶液10 μL,与6×DNA上样缓冲液2 μL混匀后,在GelRed染色的0.8%的琼脂糖凝胶中电泳,结果显示,模板DNA扩增条带更清晰明亮,无拖尾,完整性很好(见图1)。表明自主研发的DNA提取试剂较好。
在NCBI数据库中搜索人mtDNA Cytb基因序列(序列号:NC-012920.1),设计特异性引物并进行标记。引物序列及标记物为F:5’(FAM)-GAATGATATTTCCTATTCGC-3’;R:5’(Biotin)-TACTTGTCCAATGATGGTAA- 3’。扩增片段长度为249 bp。
使用紫河车的特异性引物,在退火温度为59 ℃,循环次数为20次条件下对紫河车对照药材及正品进行PCR扩增,琼脂糖凝胶电泳结果显示,紫河车对照药材及正品均出现1条249 bp的特异性扩增条带,而易混品及空白对照均未出现条带,免疫胶体金试纸条结果显示,紫河车对照药材及正品均出现2条红色条带,而易混品及空白对照均出现1条红色条带(见图23)。
因此,建立了紫河车DNA指纹分子鉴定方法,确定PCR最佳反应条件为94 ℃预变性5 min;94 ℃ 30 s,59 ℃ 30 s,72 ℃ 30 s循环次数20次;充分延伸72 ℃ 5 min。确定最佳PCR基因检测试剂为23 μL预混液,包括2×Taq PCR Master Mix 12.5 μL,紫河车上下游引物(10 μmol·L-1)各1 μL,灭菌ddH2O 8.5 μL。
取PCR扩增产物50 μL进行琼脂糖凝胶电泳,结束后紫外透射仪内切下DNA条带并称量,凝胶回收试剂盒回收目的基因;将目的基因与pGM-T载体连接,16 ℃水浴过夜,组成重组DNA分子;次日碎冰上将重组DNA分子转入DH5α感受态细胞中,37 ℃恒温箱中培养,增殖4 h后取菌液30 μL涂布于含Amp、X-Gal和IPTG的LB固体培养基上,37 ℃恒温箱中培养过夜,进行蓝-白斑筛选阳性重组DNA分子;次日挑选阳性白色菌落,接种到含Amp的LB液体培养基中37 ℃恒温培养12~16 h;次日提取质粒DNA进行PCR扩增验证为阳性重组体,送至上海生工进行基因测序。
分子克隆可见氨苄阳性平皿出现蓝-白菌落(见图4)。以白色菌落质粒DNA为模板,退火温度59 ℃、20次循环进行PCR扩增,琼脂糖凝胶电泳显示在249 bp有1条特异性DNA条带(见图5)。序列比对结果显示扩增区域DNA序列与人mtDNA Cytb基因特异性指纹区段DNA序列完全一致,同源性为100%(见图6)。成功制备出紫河车对照药材DNA克隆液。
试剂盒为20次用量,由8个部分组成,包括DNA提取试剂、PCR基因检测试剂、对照药材DNA克隆液、空白对照液、结果检测试纸条(见表1)。
紫河车对照药材DNA克隆液、对照药材、正品及其易混品在59 ℃、20个循环条件下进行PCR扩增,试纸条检测结果显示,紫河车对照药材DNA克隆液、对照药材、正品均出现2条红色条带,阴性对照及空白对照均出现1条红色条带。琼脂糖凝胶电泳验证结果正确(见图7)。表明自主研发的试剂盒特异性强。
紫河车对照药材DNA克隆液、对照药材、正品及其易混品分别在3个实验室,由3位技术人员在59 ℃、20个循环条件下进行PCR扩增,试纸条检测结果显示,紫河车对照药材DNA克隆液、对照药材、正品均出现2条红色条带,阴性对照及空白对照均出现1条红色条带。琼脂糖凝胶电泳验证结果正确(见图8)。表明自主研发的试剂盒重现性良好。
分别在试剂盒配制后第3个月、6个月、9个月和12个月对相同样品进行检测,试纸条检测结果显示,紫河车对照药材DNA克隆液、对照药材、正品出现2条红色条带,空白对照出现1条红色条带。琼脂糖凝胶电泳验证结果正确(见图9)。表明自主研发的试剂盒稳定性良好。
对紫河车对照药材模板DNA溶液100 ng·μL-1分别用灭菌ddH2O进行10倍、100倍、1 000倍、10 000倍稀释,PCR扩增后,试纸条结果显示,模板DNA溶液0.1 ng·μL-1时,仍可见2条红色条带。而琼脂糖凝胶电泳结果显示,模板DNA溶液1 ng·μL-1时,仍可见特异性扩增条带,0.1 ng·μL-1时,无特异性扩增条带。表明核酸试纸条法的灵敏度比琼脂糖凝胶电泳法高10倍(见图10)。
使用自主研发的试剂盒,对吉林雷宁食品药品检测技术服务有限公司提供的9个紫河车样品进行模板DNA的提取,59 ℃、20个循环条件下进行PCR扩增,试纸条检测结果显示,紫河车对照药材DNA克隆液、对照药材及各样品均出现2条红色条带,空白对照出现1条红色条带。琼脂糖凝胶电泳验证结果正确(见图11)。表明各样品均为正品。
紫河车为较常用的中药材,始载于《本草拾遗》,为人出生时所脱掉的胎盘,经过加工干燥而成。中西方均有将紫河车作为保健养生用品的史料记载。目前,以紫河车为原料药的中成药有河车八味丸、河车大造丸、紫河车胶囊、胎产金丹等,并且,紫河车也是临床汤剂的常用药,疗效确切,但由于来源有限,因此以假乱真现象严重。现代研究表明,人胎盘中含有多种生物活性成分,激素类如人绒毛膜促性腺激素(HCG)、促性腺激素释放激素(GnRH)等,还有酶及酶抑制因子、糖蛋白等[16],牛、猪胎盘的促绒毛膜性腺激素、雌激素、孕激素及促乳素等都与人的有一定差异[17],人胎盘中孕酮、HCG、人胎盘泌乳素(HPL)的含量分别为牛和猪胎盘的10倍、100倍、300倍左右[18]。从上述基础化学物质的区别上看,动物胎盘与紫河车的功效完全不同。迄今为止,国内对紫河车真伪鉴别方法的报道较多,主要有高效毛细管电泳法、聚丙烯酰氨凝胶电泳法、硫酸盐沉淀法等[19-21],但都在专属性、重现性或稳定性方面存在一定的缺陷,而未能被广泛应用。因此,建立专属性强,重现性及稳定性均好,简单快速的紫河车真伪鉴定方法,才能确保临床用药的有效性及安全性。
DNA是遗传信息的携带者,不同种生物体种含有的DNA序列是不同的,而同种生物体中,DNA的多态性又使各个体存在差异,因此从分子水平上检测遗传标记物DNA,不易受年龄、环境、采收、储存、加工方式等的影响,最能从本质上反映中药材的自身特征,使中药的鉴别结果更加准确可信。本研究基于遗传标志物建立的紫河车DNA指纹-免疫胶体金试纸条快速鉴定方法,通过设计人的特异性引物,利用PCR技术只能扩增出紫河车的特异性DNA片段,具有专属性强及灵敏度高的特点,PCR扩增产物采用免疫胶体金试纸条进行检测,具有简单快速、稳定可靠、低成本、可视化优势。传统的试纸条检测是基于蛋白质水平,容易出现假阴性,本方法通过强特异、超灵敏的PCR技术,从遗传物质DNA水平鉴定紫河车的真伪,弥补了单一试纸条检测容易出现假阴性的缺点,从源头控制紫河车的质量,保证临床用药的有效性和安全性。
为了使鉴定方法能够规范地普遍应用,本研究研发了一体化基因质量控制试剂盒,从DNA提取、PCR扩增、对照药材与产物检测4个模块进行优化和筛选,以保证在结果准确可靠的前提下,尽量操作简便、快速、规范、标准。一模块优化:提供了DNA提取的全部试剂,只需3步即可简便、快速提取出线粒体DNA,并且质量高,与市售试剂盒相比较至少节省2 h。二模块优化:制备了PCR基因检测试剂为23 μL的预混液,包括紫河车上下游引物(10 μmol·L-1)各1 μL,2×Taq PCR Master Mix 12.5 μL,灭菌ddH2O 8.5 μL。操作者只需将提取的基因组DNA 2 μL加入PCR基因检测试剂预混液中,混匀后即可进行PCR扩增。并且经过反复试验,试剂的特异性、稳定性、重现性均非常好,灵敏度为琼脂糖凝胶电泳的10倍。并且解决了PCR扩增成败的关键即特异性引物的设计问题,PCR基因检测预混液中含有最佳浓度、最佳退火温度、循环次数的特异性引物。扩增程序:94 ℃预变性5 min;95 ℃ 30 s,59 ℃ 30 s,72 ℃ 30 s循环次数20次;充分延伸72 ℃ 5 min。节省了引物设计、订购合成、筛选反应条件的时间至少1周。三模块优化:制备了紫河车对照药材DNA克隆液,直接反映了紫河车独一无二DNA序列,并且以重组质粒的形式长期储存,取之不尽用之不竭,省时省力省钱省药材。四模块优化:采用免疫胶体金试纸条进行结果检测,不需要贵重仪器配合,2~3 min即可出现可视化结果,快速灵敏,与琼脂糖凝胶电泳相比较,至少节省2 h。本方法为单重PCR-试纸条,一次只能鉴定紫河车的真伪,后续将开展多重PCR-试纸条,实现紫河车的高通量掺假鉴定。
本研究具有PCR技术特异性高,灵敏度强的特点,具有免疫胶体金试纸条低成本,可视化优势,具有试剂盒规范化、标准化特征。在普通实验室、普通操作人员即可完成紫河车的真伪鉴定,从源头上控制中药材紫河车的质量,对于保障临床用药的有效性及安全性,保障中药材、中成药的生产和中药产业的健康发展都具有重要意义。
