Objective:

To establish an HPLC method for the simultaneous determination of eight components including salvianic acid A sodium,geniposidic acid,phellodendrine,salvianolic acid B,palmatine,berberine,atractylodin,tanshinone ⅡA in Shuangshi Tonglin capsules.

Methods:

The determination was performed on a Capcell Pak MG Ⅱ C18 column (250 mm×4.6 mm,5 μm) with the mobile phase consisting of acetonitrile-water(containing 0.1% phosphoric acid)-water(containing 0.1% phosphoric acid and 0.2% triethylamine,pH 6.0) in a gradient mode at a flow rate of 1.0 mL·min-1. The column temperature was set at 30 ℃,the injection volume was 10 μL,and the detection wavelength was 280 nm (0-13.4 min)for salvianic acid A sodium,238 nm (13.5-17.0 min) for geniposidic acid,280 nm (17.1-51.0 min)for phellodendrine,salvianolic acid B,345 nm(51.1.1-70.0 min)for palmatine,berberine,336 nm(70.1-78.0 min)for atractylodin and 268 nm(78.1-90.0 min)for tanshinone ⅡA.

Results:

Salvianic acid A sodium,geniposidic acid,phellodendrine,salvianolic acid B,palmatine,berberine,atractylodin,tanshinoneⅡA had good linearities in their respective linear ranges with all linear correlation coefficients ≥ 0.999 5 and RSDs of the precision,repeatability and stability less than 2.0%. The average recoveries were 101.3%,98.2%,97.3%,98.1%,96.6%,97.9%,98.7%,99.6%,with RSDs lower than 2.0%. The contents of the above eight components in three batches of samples was 4.47-5.51 mg·g-1,0.93-1.44 mg·g-1,1.00-1.07 mg·g-1,5.72-8.67 mg·g-1,0.64-0.69 mg·g-1,1.62-1.90 mg·g-1,0.055-0.088 mg·g-1,0.29-0.44 mg·g-1,respectively.

Conclusion:

The proposed method is rapid,sensitive and specific,which can be used for determination the contents of eitht components in Shuangshi Tonglin capsules.

目的:

建立HPLC法同时测定双石通淋胶囊中丹参素钠、京尼平苷酸、黄柏碱、丹酚酸B、巴马汀、小檗碱、苍术素、丹参酮ⅡA 8个成分的含量。

方法:

采用Capcell Pak MGⅡC18(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-0.1%磷酸水-水(含0.1%磷酸和0.2%三乙胺,pH=6.0),梯度洗脱,流速为1.0 mL·min-1,柱温为30 ℃,进样量为10 μL,检测波长分别为280 nm(0~13.4 min,丹参素钠)、238 nm(13.5~17.0 min,京尼平苷酸)、280 nm(17.1~51.0 min,黄柏碱和丹酚酸B)、345 nm(51.1.1~70.0 min,巴马汀和盐酸小檗碱)、336 nm(70.1~78.0 min,苍术素)和268 nm(78.1~90.0 min,丹参酮ⅡA)。

结果:

丹参素钠、京尼平苷酸、黄柏碱、丹酚酸B、巴马汀、小檗碱、苍术素、丹参酮ⅡA 8个成分在各自线性范围内线性关系良好,相关系数均≥0.999 5;精密度、稳定性、重复性试验的RSD均小于2.0%;平均回收率分别为101.3%、98.2%、97.3%、98.1%、96.6%、97.9%、98.7%、99.6%,RSD均小于2.0%。3批样品中上述8个成分的含量范围分别为4.47~5.51、0.93~1.44、1.00~1.07、5.72~8.67、0.64~0.69、1.62~1.90、0.055~0.088、0.29~0.44 mg·g-1

结论:

该方法快速、灵敏,专属性高,可用于同时测定双石通淋胶囊中8个成分的含量。

目的:

建立HPLC法同时测定双石通淋胶囊中丹参素钠、京尼平苷酸、黄柏碱、丹酚酸B、巴马汀、小檗碱、苍术素、丹参酮ⅡA 8个成分的含量。

方法:

采用Capcell Pak MGⅡC18(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-0.1%磷酸水-水(含0.1%磷酸和0.2%三乙胺,pH=6.0),梯度洗脱,流速为1.0 mL·min-1,柱温为30 ℃,进样量为10 μL,检测波长分别为280 nm(0~13.4 min,丹参素钠)、238 nm(13.5~17.0 min,京尼平苷酸)、280 nm(17.1~51.0 min,黄柏碱和丹酚酸B)、345 nm(51.1.1~70.0 min,巴马汀和盐酸小檗碱)、336 nm(70.1~78.0 min,苍术素)和268 nm(78.1~90.0 min,丹参酮ⅡA)。

结果:

丹参素钠、京尼平苷酸、黄柏碱、丹酚酸B、巴马汀、小檗碱、苍术素、丹参酮ⅡA 8个成分在各自线性范围内线性关系良好,相关系数均≥0.999 5;精密度、稳定性、重复性试验的RSD均小于2.0%;平均回收率分别为101.3%、98.2%、97.3%、98.1%、96.6%、97.9%、98.7%、99.6%,RSD均小于2.0%。3批样品中上述8个成分的含量范围分别为4.47~5.51、0.93~1.44、1.00~1.07、5.72~8.67、0.64~0.69、1.62~1.90、0.055~0.088、0.29~0.44 mg·g-1

结论:

该方法快速、灵敏,专属性高,可用于同时测定双石通淋胶囊中8个成分的含量。

Objective:

To establish an HPLC method for the simultaneous determination of eight components including salvianic acid A sodium,geniposidic acid,phellodendrine,salvianolic acid B,palmatine,berberine,atractylodin,tanshinone ⅡA in Shuangshi Tonglin capsules.

Methods:

The determination was performed on a Capcell Pak MG Ⅱ C18 column (250 mm×4.6 mm,5 μm) with the mobile phase consisting of acetonitrile-water(containing 0.1% phosphoric acid)-water(containing 0.1% phosphoric acid and 0.2% triethylamine,pH 6.0) in a gradient mode at a flow rate of 1.0 mL·min-1. The column temperature was set at 30 ℃,the injection volume was 10 μL,and the detection wavelength was 280 nm (0-13.4 min)for salvianic acid A sodium,238 nm (13.5-17.0 min) for geniposidic acid,280 nm (17.1-51.0 min)for phellodendrine,salvianolic acid B,345 nm(51.1.1-70.0 min)for palmatine,berberine,336 nm(70.1-78.0 min)for atractylodin and 268 nm(78.1-90.0 min)for tanshinone ⅡA.

Results:

Salvianic acid A sodium,geniposidic acid,phellodendrine,salvianolic acid B,palmatine,berberine,atractylodin,tanshinoneⅡA had good linearities in their respective linear ranges with all linear correlation coefficients ≥ 0.999 5 and RSDs of the precision,repeatability and stability less than 2.0%. The average recoveries were 101.3%,98.2%,97.3%,98.1%,96.6%,97.9%,98.7%,99.6%,with RSDs lower than 2.0%. The contents of the above eight components in three batches of samples was 4.47-5.51 mg·g-1,0.93-1.44 mg·g-1,1.00-1.07 mg·g-1,5.72-8.67 mg·g-1,0.64-0.69 mg·g-1,1.62-1.90 mg·g-1,0.055-0.088 mg·g-1,0.29-0.44 mg·g-1,respectively.

Conclusion:

The proposed method is rapid,sensitive and specific,which can be used for determination the contents of eitht components in Shuangshi Tonglin capsules.

双石通淋胶囊是在精研清代名医程钟龄“程氏萆薢分清饮”处方的基础上,结合中药现代药理研究,利用现代制药技术和工艺研制而成,由粉萆薢、关黄柏、败酱草、青黛、滑石、车前子、石菖蒲、茯苓、苍术、丹参组成[1],能够活血行气、清热解毒,具有消炎、镇痛和利尿的作用,有效缓解患者的热涩淋痛之症[2]。方中关黄柏具有清热燥湿,泻火除蒸,解毒疗疮之功效[3]153,主要含有生物碱、内酯、酚酸、萜、苯丙素等化学成分[4-5],其中生物碱类是关黄柏的主要活性成分,包含黄柏碱、巴马汀、小檗碱等[6],具有明显的抗菌、抗炎、调节心血管系统疾病、免疫调节、降血糖等药理作用[5];车前子具有清热利尿通淋、渗湿止泻、明目、祛痰之功效[3]69,主要含有多糖、苯乙醇苷、环烯醚萜、三萜、黄酮、甾醇及生物碱等类化学成分[7],具有利尿、消炎、降血糖、降血压、调血脂、抗氧化和调节免疫等药理作用[8];苍术具有燥湿健脾、祛风散寒、明目之功效[3]168,主要含有挥发油、糖、黄酮等类化学成分,具有抗炎、抗肿瘤、促进胃排空、抗胃溃疡等药理作用[9-12];丹参具有活血祛疲,通经止痛,清心除烦,凉血消痈之功效[3]77,主要含有丹参素、丹酚酸、丹酚酮等化学成分[13-14],具有抗炎、抗氧化、抗凝血、抗纤维化、抗肿瘤和保护脏器等药理作用[15]
双石通淋胶囊现行标准为国家食品药品监督管理局标准YBZ00352008,该标准采用薄层扫描法测定盐酸小檗碱的含量,测定误差较大,不能很好地对双石通淋胶囊中盐酸小檗碱的含量进行准确定量测定,目前未见双石通淋胶囊含量测定的其他报道。为有效控制该制剂的质量,本研究采用HPLC法同时测定制剂中丹参素钠、京尼平苷酸、黄柏碱、丹酚酸B、小檗碱、巴马汀、苍术素、丹参酮ⅡA8个成分的含量,为该制剂有效成分的测定及质量控制提供评价方法。
Agilent 1260型高效液相色谱仪(Agilent Technologies公司);CP224S型万分之一、BP210D型十万分之一电子分析天平(赛多利斯科学仪器北京有限公司);SK7200HP超声波清洗仪(上海科导超声仪器有限公司)。
对照品丹参素钠(批号110855-201915,含量以97.8%计)、京尼平苷酸(111828-201805,含量以98.1%计)、盐酸黄柏碱(111895-201805,含量以94.9%计)、丹酚酸B(111562-201917,含量以96.6%计)、盐酸巴马汀(110732-201812,含量以97.6%计)、盐酸小檗碱(110713-202015,含量以85.9%计)、苍术素(111924-201605,含量以99.8%计)、丹参酮ⅡA(110766-202022,含量以98.9%计)均购自中国食品药品检定研究院。乙腈为色谱纯,水为Milli-Q超纯水,其他所用试剂均为分析纯。3批双石通淋胶囊样品批号依次为221015、230207、230305,生产厂家为陕西摩美得气血和制药有限公司。
采用Capcell Pak MGⅡC18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈(A)-0.1%磷酸水溶液(B)-水(含0.1%磷酸、0.2%三乙胺,pH 6.0)(C),梯度洗脱,洗脱程序见表1,流速为1.0 mL·min-1,柱温为30 ℃,进样量为10 μL,检测波长分别为280 nm(0~13.4 min,丹参素钠)、238 nm(13.5~17.0 min,京尼平苷酸)、280 nm(17.1~51.0 min,黄柏碱和丹酚酸B)、345 nm(51.1.1~70.0 min,巴马汀和小檗碱)、336 nm(70.1~78.0 min,苍术素)和268 nm(78.1~90.0 min,丹参酮ⅡA)。
精密称取丹参素钠、京尼平苷酸、盐酸黄柏碱、丹酚酸B、盐酸巴马汀、盐酸小檗碱、苍术素、丹参酮ⅡA的对照品各适量,分别加75%甲醇水溶液适量使溶解,制成每1 mL含丹参素钠5.526 mg、京尼平苷酸1.452 mg、黄柏碱1.255 mg、丹酚酸B 9.902 mg、巴马汀0.743 7 mg、小檗碱2.565 mg、苍术素0.056 99 mg、丹参酮ⅡA 0.527 1 mg的单一对照品储备液。精密吸取上述对照品储备液各1.0 mL,置同一20 mL量瓶中,加75%甲醇水溶液稀释至刻度,摇匀,即得混合对照品溶液。
取10粒本品的内容物,混匀,研细,取约0.5 g,精密称量,置具塞锥形瓶中,精密加入75%甲醇水溶液25 mL,密塞,称量,超声(功率360 W,频率28 kHz)处理30 min,取出放冷,再称量,用75%甲醇水溶液补足减失的量,摇匀,滤过,取续滤液,即得。
按本品处方、制法,分别制备缺少丹参、车前子、关黄柏及苍术的阴性样品,按“2.2.2”项下方法制备各阴性样品溶液。
分别精密吸取“2.2”项下混合对照品溶液、供试品溶液及阴性样品溶液各10 μL,按“2.1”项色谱条件进行分析,结果见图1。供试品溶液色谱图中基线平稳,8个成分色谱峰峰形较好,且与相邻峰的分离度均≥1.5,阴性样品溶液在8个成分相对应的保留时间位置无色谱峰出现。
精密吸取“2.2.1”项的混合对照品溶液0.2、0.5、1、2、3、5 mL,分别置5 mL量瓶中,加75%甲醇水溶液稀释至刻度,摇匀,按“2.1”项色谱条件进行分析。分别以各成分峰面积A为横坐标,相应质量浓度C(μg·mL-1)为纵坐标进行线性回归,结果见表2
取“2.2.1”项下混合对照品溶液3 mL,置于5 mL量瓶中,加75%甲醇水溶液稀释至刻度,摇匀,按“2.1”项色谱条件,连续进样6次,测定各成分的峰面积。结果丹参素钠、京尼平苷酸、黄柏碱、丹酚酸B、巴马汀、小檗碱、苍术素、丹参酮ⅡA峰面积的RSD分别为1.2%、1.7%、1.4%、0.56%、0.94%、0.70%、1.1%、0.74%,表明仪器的精密度良好。
取批号为221015样品的供试品溶液,分别于制备后0、6、12、18、24 h进样分析,记录色谱图的峰面积。结果丹参素钠、京尼平苷酸、黄柏碱、丹酚酸B、巴马汀、小檗碱、苍术素、丹参酮ⅡA峰面积的RSD分别为0.99%、1.8%、0.67%、1.3%、0.20%、0.26%、0.31%、0.74%,表明供试品溶液在24 h内稳定性良好。
取批号为221015的样品适量,按“2.2.2”项下方法平行制备6份供试品溶液,进样分析,记录色谱图的峰面积,按标准曲线计算样品中8个成分的含量及RSD。结果样品中丹参素钠、京尼平苷酸、黄柏碱(按盐酸黄柏碱计)、丹酚酸B、巴马汀(按盐酸巴马汀计)、小檗碱(按盐酸小檗碱计)、苍术素、丹参酮ⅡA的平均含量分别为5.539、1.436、1.040、5.718、0.639 6、1.625、0.055 40、0.439 8 mg·g-1,RSD分别为1.8%、0.81%、1.1%、1.6%、1.1%、1.3%、0.62%、0.81%,表明该方法的重复性良好。
取批号为221015的样品的内容物适量,混匀,研细,精密称取0.25 g,分别加入“2.2.1”项下的单一对照品储备液0.26、0.26、0.20、0.15、0.22、0.16、0.25、0.21 mL,按“2.2.2”项下方法平行制备6份供试溶液,按“2.1”项下色谱条件进行测定,计算平均回收率和RSD,结果见表3
取3批样品(批号分别为221015、230207、230305),分别按“2.2.2”项下方法平行制备2份供试品溶液,按“2.1”项下色谱条件测定各成分峰面积,根据回归方程计算各成分的含量(其中本品每1 g含黄柏、小檗碱、巴马汀分别以盐酸黄柏碱、盐酸小檗碱、盐酸巴马汀计),结果见表4
应用二极管阵列检测器在200~400 nm对丹参素钠、京尼平苷酸、黄柏碱、丹酚酸B、巴马汀、小檗碱、苍术素、丹参酮ⅡA进行光谱扫描,结果表明丹参素钠等8个成分最大吸收波长从238~345 nm相差极大,因此,本文采用相关成分的最大吸收波长作为检测波长:丹参素钠280 nm、京尼平苷酸238 nm、黄柏碱和丹酚酸B 280 nm、巴马汀和小檗碱345 nm、苍术素336 nm、丹参酮ⅡA 268 nm。同时,经二极管阵列检测器分析,样品中的上述8个成分色谱峰均为纯的色谱峰。
考察了Kromasil C18(250 mm×4.6 mm,5 μm)、Capcell Pak MGⅡC18(250 mm×4.6 mm,5 μm)和Agilent C18(250 mm×4.6 mm,5 μm)色谱柱对8个待测成分的影响,以各成分与相邻色谱峰的分离度和峰形作为指标,最终选择Capcell Pak MGⅡC18色谱柱(250 mm×4.6 mm,5 μm)。
本试验对多种流动相系统进行了考察,以乙腈-0.1%磷酸进行梯度洗脱,结果样品中巴马汀色谱峰与小檗碱色谱峰分离度较差;以乙腈-0.2%三乙胺进行梯度洗脱,结果样品中巴马汀色谱峰与小檗碱色谱峰分离度大大增加,但丹参素钠、京尼平苷酸、丹酚酸B与相邻杂峰的分离较差。最终,采用乙腈-水(含0.1%磷酸)-水(含0.1%磷酸、0.2%三乙胺,pH 6.0)作为流动相,结果样品中8个待测成分与杂质峰分离较好,分离度均≥1.5。
考察了不同提取溶剂(乙醇、甲醇、75%甲醇水、50%甲醇水)、不同提取方法(超声、回流)、不同提取时间(20、30、45 min)以及不同溶剂量(20、25、50 mL)对成分提取的影响,发现75%甲醇水超声提取30 min时,各成分基本能被提取完全,故选择此条件作为供试品溶液的制备方法。
本实验建立了HPLC法同时测定双石通淋胶囊中8个成分的含量,方法准确,重复性高,3批次样品测定结果详见表4。但由于该制剂为独家品种,样本量较少,后期将继续收集样本积累数据,为双石通淋胶囊质量标准的制修订提供技术支持。
