Objective:

To establish an HPLC method for determination of the impurity profile in furosemide and its injection. At the same time,applying this method to detect and analyze products from many domestic and foreign enterprises,to evaluate the status quo of impurity control in API and injection,and the correlation of major degraded impurities in injection with prescription and packaging materials.

Methods:

A YMC Hydrosphere C18 column(250 mm×4.6 mm,3 μm)was used,and the mobile phase was 0.05% TFA solution (pH 2.23)-methanol-acetonitrile at the flow rate of 1.0 mL·min-1,gradient elution. Detection wavelength was set at 238 nm and 277 nm and column temperature was 30 ℃. Impurity reference was used for localization and the relative retention time of each impurity was calculated. The known impurities were calculated using the principal component self-control method with correction factor,the unknown impurities were determined by principal component without correction factor.

Results:

The method for the determination of 11 kinds of known impurities,the potential genotoxic impurity in furosemide,its injection was established,the separation degree of all impurities met the requirements,and the sources of the impurities were identified by forced degradation test. The detected quantity of degraded impurity C and degraded impurity G in the injections produced by 2 to 3 companies exceeded the identification limit in ICH by 0.2%,the amount of potential genotoxic impurity furfural detected in the injection of 5 enterprises was much higher than that of the reference preparation,other impurities were the same as the reference preparation.

Conclusion:

The established HPLC method can be used for rapid detection and analysis of impurities in furosemide and its injection. The impurity control level of injection produced by only 2 domestic enterprises is basically consistent with the reference preparation,others need to optimize prescription and packaging materials. This study can provide reference for improving the safety of furosemide injection and evaluating the quality consistency of generic drugs.

目的:

建立HPLC法研究呋塞米及其注射液的杂质谱,同时检测和分析来源于国内外多家企业的产品,评价该产品的杂质控制现状,以及注射液的主要降解杂质与处方、包装材料的相关性。

方法:

采用YMC Hydrosphere C18(250 mm×4.6 mm,3 μm)色谱柱,以0.05%三氟乙酸溶液(pH 2.23)-甲醇-乙腈为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长238 nm和277 nm,柱温30 ℃。计算各杂质与呋塞米的相对保留时间(RRT),已知杂质采用加校正因子(f)的主成分自身对照法计算,未知杂质为不加校正因子的主成分自身对照法。

结果:

建立了呋塞米及其注射液中11个已知杂质和1个潜在基因毒杂质的检测方法,各杂质分离度均符合要求,通过强制降解试验归属了各杂质来源。2~3家企业注射液中降解杂质C和降解杂质G的检出量比ICH的鉴定限度大0.2%,5家企业注射液中潜在基因毒杂质糠醛的检出量远高于参比制剂。

结论:

建立的HPLC方法可用于呋塞米及其注射液中杂质的快速检测分析,国内只有2家企业生产的注射液杂质控制水平基本与参比制剂一致,其他企业需优化处方和包装材料,本研究可为提高呋塞米注射液的安全性和仿制药质量一致性评价研究提供参考。

目的:

建立HPLC法研究呋塞米及其注射液的杂质谱,同时检测和分析来源于国内外多家企业的产品,评价该产品的杂质控制现状,以及注射液的主要降解杂质与处方、包装材料的相关性。

方法:

采用YMC Hydrosphere C18(250 mm×4.6 mm,3 μm)色谱柱,以0.05%三氟乙酸溶液(pH 2.23)-甲醇-乙腈为流动相,梯度洗脱,流速1.0 mL·min-1,检测波长238 nm和277 nm,柱温30 ℃。计算各杂质与呋塞米的相对保留时间(RRT),已知杂质采用加校正因子(f)的主成分自身对照法计算,未知杂质为不加校正因子的主成分自身对照法。

结果:

建立了呋塞米及其注射液中11个已知杂质和1个潜在基因毒杂质的检测方法,各杂质分离度均符合要求,通过强制降解试验归属了各杂质来源。2~3家企业注射液中降解杂质C和降解杂质G的检出量比ICH的鉴定限度大0.2%,5家企业注射液中潜在基因毒杂质糠醛的检出量远高于参比制剂。

结论:

建立的HPLC方法可用于呋塞米及其注射液中杂质的快速检测分析,国内只有2家企业生产的注射液杂质控制水平基本与参比制剂一致,其他企业需优化处方和包装材料,本研究可为提高呋塞米注射液的安全性和仿制药质量一致性评价研究提供参考。

Objective:

To establish an HPLC method for determination of the impurity profile in furosemide and its injection. At the same time,applying this method to detect and analyze products from many domestic and foreign enterprises,to evaluate the status quo of impurity control in API and injection,and the correlation of major degraded impurities in injection with prescription and packaging materials.

Methods:

A YMC Hydrosphere C18 column(250 mm×4.6 mm,3 μm)was used,and the mobile phase was 0.05% TFA solution (pH 2.23)-methanol-acetonitrile at the flow rate of 1.0 mL·min-1,gradient elution. Detection wavelength was set at 238 nm and 277 nm and column temperature was 30 ℃. Impurity reference was used for localization and the relative retention time of each impurity was calculated. The known impurities were calculated using the principal component self-control method with correction factor,the unknown impurities were determined by principal component without correction factor.

Results:

The method for the determination of 11 kinds of known impurities,the potential genotoxic impurity in furosemide,its injection was established,the separation degree of all impurities met the requirements,and the sources of the impurities were identified by forced degradation test. The detected quantity of degraded impurity C and degraded impurity G in the injections produced by 2 to 3 companies exceeded the identification limit in ICH by 0.2%,the amount of potential genotoxic impurity furfural detected in the injection of 5 enterprises was much higher than that of the reference preparation,other impurities were the same as the reference preparation.

Conclusion:

The established HPLC method can be used for rapid detection and analysis of impurities in furosemide and its injection. The impurity control level of injection produced by only 2 domestic enterprises is basically consistent with the reference preparation,others need to optimize prescription and packaging materials. This study can provide reference for improving the safety of furosemide injection and evaluating the quality consistency of generic drugs.

呋塞米(结构式见图1)化学名为2-[(2-呋喃甲基)氨基]-5(氨磺酰基)-4-氯苯甲酸,别名呋喃苯胺酸[1-4]。呋塞米注射液由原研企业Sanofi Aventis Us LLC于1968年在美国上市,商品名“Lasix”;国内由西安风华药业首先研发并上市。2020年1月,国家局发布呋塞米注射液参比制剂,2mL:20 mg规格的注射液持证商为Sanofi-Aventis GmbH和Sanofi-Aventis France,属于未进口原研药品。
呋塞米及其注射液现均收载于2020年版《中华人民共和国药典》(简称《中国药典》),其有关物质仅对未知单个杂质和总杂质进行控制,且采用不加校正因子的主成分自身对照法[5],未控制已知杂质;BP 2022收载了杂质A、B、C、D、E、F 6个杂质的结构式,并对杂质C和杂质D进行了单独控制[6]。呋塞米注射液稳定性欠佳[7],光照、高温和氧化条件均可加速其降解,且光降解杂质糠醛含基因毒警示结构[8],各国药典均未对其控制。目前,尚无文献报道对呋塞米及其注射液中杂质的系统研究。本研究结合购买到的12个杂质对照品和原料药破坏试验,建立HPLC梯度洗脱分析方法,归属各杂质来源,并测定10家企业的原料药和注射液,探索杂质与产品的贮存环境、处方、包装材料的相关性,为提高呋塞米注射液的安全性和仿制药质量一致性评价研究提供参考。

呋塞米对照品(批号100544-201102,中国食品药品检定研究院,含量99.7%);各杂质对照品信息见表1
呋塞米(国内H企业(CPC-004-M180204),印度I企业(I0038304),呋塞米注射液(来自国内A(D42201051)、B(34220208)、C(2202261)、D(2203032)、E(72204161)、F(22022827)、G(2112021-1)等7家生产企业,规格2 mL:20 mg),呋塞米注射液参比制剂(LASIX公司,批号J0485),规格2 mL:20 mg)。
乙腈、甲醇为色谱纯,默克公司;三氟乙酸为分析纯,上海麦克林生化科技股份有限公司;纯化水为自制。
采用YMC Hydrosphere C18(250 mm×4.6 mm,3 μm)色谱柱,以0.05%三氟乙酸溶液(pH为2.23)(A)-甲醇(B)-乙腈(C)为流动相,梯度洗脱程序见表2,检测波长238 nm(杂质A~杂质K)和277 nm(糠醛),柱温30 ℃,流速1.0 mL·min-1,进样体积20 μL。
精密称取对照品呋塞米10.26 mg和糠醛10.14 mg,置同一100 mL量瓶中,加甲醇溶解并稀释至刻度,摇匀,作为混合对照品储备液Ⅰ;精密量取混合对照品储备液Ⅰ5 mL,置100 mL量瓶中,加50%乙腈溶液稀释至刻度,摇匀,作为混合对照品溶液Ⅰ。再分别精密称取对照品呋塞米10.69 mg、杂质A 10.15 mg、杂质B 10.43 mg、杂质C 10.44 mg、杂质D 10.05 mg、杂质E 10.10 mg、杂质F 10.32 mg、杂质G 11.25 mg、杂质H 10.18 mg、杂质I 10.44 mg、杂质J 10.66 mg和杂质K 10.45 mg,置同一100 mL量瓶中,加甲醇溶解并稀释至刻度,摇匀,作为混合对照品储备液Ⅱ;精密量取混合对照品储备液Ⅱ 5 mL,置100 mL量瓶中,加50%乙腈溶液稀释至刻度,摇匀,作为混合对照品溶液Ⅱ。精密量取混合对照品储备液Ⅰ和Ⅱ各5 mL,置100 mL量瓶中,加50%乙腈溶液稀释至刻度,摇匀,作为混合对照品溶液。
分别取呋塞米原料和呋塞米注射液各适量,加50%乙腈溶液稀释制成每1 mL中含呋塞米1 mg的溶液,滤过,即得。
取杂质A适量,置50 mL量瓶中,加入“2.2.2”项下供试品溶液5 mL,加50%乙腈溶液制成每1 mL约含呋塞米和杂质A各0.1 mg的混合溶液。
精密量取“2.2.2”项下供试品溶液1 mL,置100 mL量瓶中,用50%乙腈溶液稀释至刻度,摇匀,即得。
将辅料根据各企业的最大处方量按照“2.2.2”项供试品溶液的制备方法配制空白辅料溶液,滤过,即得。
分别取“2.2.1”项混合对照品溶液、“2.2.3”项系统适用性溶液和“2.2.5”项空白辅料溶液,按“2.1”项色谱条件测定,结果混合对照品溶液和系统适用性溶液中各色谱峰与相邻色谱峰在双波长下均可满足分离度要求,空白辅料溶液无干扰。
利用强制降解试验进一步验证该方法的专属性。取呋塞米原料(H企业,批号CPC-004-1903014)6份,每份约50 mg,分别置50 mL量瓶中,分别对6份样品进行不处理、加0.1 mol·L-1盐酸溶液2 mL室温放置6 h、加0.1 mol·L-1氢氧化钠溶液2 mL室温放置6 h、加30%过氧化氢溶液2 mL室温放置6 h、加50%乙腈溶液5 mL水浴(60 ℃)加热6 h和加50%乙腈溶液5 mL紫外灯(254 nm)照射6 h等操作,酸碱破坏样品各加入0.1 mol·L-1氢氧化钠溶液或0.1 mol·L-1盐酸溶液2 mL中和,各破坏样品加50%乙腈溶液稀释至刻度,摇匀,滤过,按“2.1”项色谱条件分析测定,结果在不同破坏条件下,各降解产物之间均能达到良好分离,见图2
精密量取“2.2.1”项混合对照品储备液Ⅰ和Ⅱ各适量,分别置10 mL量瓶中,加50%乙腈溶液稀释至刻度,摇匀,即得含呋塞米和各杂质浓度约为0.1~20 μg·mL-1的线性溶液。按“2.1”项色谱条件进样分析,以质量浓度为横坐标,峰面积为纵坐标,进行线性回归得回归方程。以各杂质的保留时间与呋塞米主峰的保留时间之比计算相对保留时间(RRT),以呋塞米与各杂质标准曲线斜率之比计算相对校正因子(f),结果见表3
取“2.2.1”项混合对照品溶液Ⅰ和Ⅱ,以S/N为3时的对照品浓度作为检测限,以S/N为10时的对照品浓度作为定量限,结果见表3
取“2.2.1”项混合对照品溶液Ⅰ和Ⅱ,按“2.1”项色谱条件连续进样6次,测得呋塞米及其12个杂质保留时间的RSD(n=6)均为0,峰面积RSD(n=6)均小于1.0%,仪器精密度良好。
取呋塞米注射液(E企业,批号72202161),按照“2.2.2”项供试品溶液的配制方法和“2.2.4”项对照溶液的配制方法分别平行配制供试品溶液和对照溶液各6份。照“2.1”项色谱条件进样,按加校正因子(如表3所示)的主成分自身对照法计算各杂质含量,结果杂质C含量分别为0.24%、0.23%、0.24%、0.24%、0.23%、0.23%,RSD=2.3%;杂质B含量均为0.01%;杂质F含量均为0.01%;杂质A含量均为0.01%;杂质D含量均为0.02%;糠醛含量均为0.006%;各杂质总和分别为0.30%、0.30%、0.30%、0.31%、0.30%、0.29%,RSD=2.1%。
取“2.3.3”项下供试品溶液,在样品温度为10 ℃时,分别于0、1、2、4、7、10 h,按“2.1”项色谱条件进样,测得供试品溶液在温度为10 ℃时,各杂质含量稳定无变化,表明供试品溶液在10 h内稳定性良好。
精密量取混合对照品溶液Ⅰ和Ⅱ各80、100和120 μL,分别置100 mL量瓶中,每个浓度平行制备3份,共9份;再精密量取呋塞米注射液(E企业,批号72202161)10 mL,分别置于上述9个100 mL量瓶中,加50%乙腈溶液稀释至刻度,摇匀,制得高中低3个浓度的供试品溶液,照“2.1”项色谱条件进样,按外标法计算回收率,12种已知杂质的平均回收率为91.8%~100.0%,见表4
采用“2.1”项色谱条件测定10家企业产品中的有关物质,已知杂质按照加校正因子的主成分自身对照法计算,未知杂质按照不加校正因子的主成分自身对照法计算,结果如表5所示。
由检测结果可知,杂质A、杂质E、杂质H和杂质I在原料药和注射液中均未检出,其余8个杂质有不同程度的检出,其中杂质C在E、F和G企业的注射液中含量相对较高,杂质G在F和G企业的注射液中检出量较高;除B企业注射液中糠醛含量略高于参比制剂外,其他注射液企业中糠醛含量相较于参比制剂高出一个数量级。
根据呋塞米注射液溶液性质,优化了制备供试液的溶剂,重新建立了有效分离呋塞米及其12个杂质的HPLC方法。
呋塞米呈弱酸性易溶于碱性溶液中,因此呋塞米注射液呈碱性,pH在8.0~9.0。在酸性环境下呋喃苯胺酸容易游离产生沉淀,而且生成沉淀的速度与数量随pH值的下降而增加[9]。依据标准检验中发现在制备供试液时加入溶剂(取冰醋酸22 mL,加乙腈-水(1:1)至1 000 mL,混匀)的瞬间有白色絮状沉淀生成。另外,呋塞米在高浓度的乙酸溶液中,见光会很快地产生杂质C、糠醛等降解杂质,检验中需严格避光操作,对操作环境要求严格[9-10],因此现行标准中的溶剂不适用于本品供试液制备。经查阅文献资料和实验对比研究,最终选择50%乙腈溶液作为溶剂。
现行标准采用水-四氢呋喃-冰醋酸(70:30:1)作为流动相,对比2020年版《中国药典》和EP 9.0的流动相,分别尝试了磷酸二氢钠溶液、磷酸溶液、磷酸二氢钾溶液和0.05%三氟乙酸溶液,最终选择分离效果最佳的0.05%三氟乙酸溶液作为色谱系统水相,以0.05%三氟乙酸溶液-乙腈-甲醇作为梯度洗脱的流动相。
2020年版《中国药典》有关物质测定波长为272 nm,BP 2013和EP 9.0采用238 nm,而USP 40-NF 35采用254 nm和272 nm双波长进行测定。本研究发现在238 nm波长处各杂质检测灵敏度更高,检出杂质种类数也更多;糠醛的最大吸收波长为277 nm,而在238 nm处吸收较弱(图4),综上,最终选择双波长测定本品的有关物质[11],238 nm测定除糠醛外的其他所有杂质,277 nm波长测定糠醛。
分别对呋塞米及其注射液进行酸、碱、氧化、高温和光照破坏试验,结果原料药和注射液在光照、氧化和高温条件下均不稳定,其中杂质C、杂质B、杂质G、杂质K和糠醛增加明显;结合注射液的稳定性试验,对各杂质进行归属(表6)。破坏试验表明注射液中降解杂质的产生主要受生产和储运过程中高温、光照和氧化等因素的影响,其中光照影响最大。
表5可以看到,呋塞米中各已知杂质检出量均低于质控限度0.15%,同时其他未知最大单个杂质远低于鉴定限度0.10%[12]。大部分企业注射液中杂质含量较低,除了杂质C、杂质G和糠醛外,其他杂质检出量与参比制剂基本相同;F和G 2家企业制剂中杂质C和杂质G均超出了鉴定限度0.2%,其他未知最大单杂检出量远低于鉴定限度0.2%[13]
糠醛为潜在基因毒杂质,除A、B 2家企业和参比制剂外,其余5家仿制企业注射液中糠醛含量均高于文献报道中按照注射液的最大使用量计算糠醛的质控限度0.019 7%[8],其中E和F企业最高;2家企业原料药,仅1家检出糠醛,检出量较低。
综上,呋塞米原料药杂质控制较好,国内大部分企业注射液杂质控制水平与参比制剂存在较大差距,需进一步改进。
根据杂质测定结果,对比各企业处方,E、F和G企业处方中含有参比制剂未使用的抗氧剂无水亚硫酸钠和亚硫酸氢钠,课题组开展了原辅料相容性试验,结果在高温条件下亚硫酸氢钠和无水亚硫酸钠会加速呋塞米的降解,尤以杂质C含量升高最为明显(表7),未加入任何抗氧剂的原料药在高温条件下杂质C含量反而增长缓慢,说明亚硫酸氢钠和亚硫酸钠在高温条件下与呋塞米不相容。
E和G企业注射液中杂质G含量远高于其他企业,处方显示2家企业所用辅料的种类一致,均采用亚硫酸氢钠作为抗氧剂。由表7可知,呋塞米与亚硫酸氢钠的混合物在高温和光照条件下放置10 d后,杂质G含量迅速升高;在光照放置10 d后,糠醛含量也迅速升高,表明亚硫酸氢钠与呋塞米不相容。
鉴于未加抗氧剂处方的稳定性反而优于加入抗氧剂的处方,且注射液属于高风险剂型,选择辅料的种类应越少越好[14],呋塞米注射液处方中添加抗氧剂无水亚硫酸钠和亚硫酸氢钠不合理[15]。根据质量源于设计(QbD)理念,建议相关企业优化处方,对产品质量从源头设计进行较为系统的控制。
杂质C、杂质G和潜在基因毒杂质糠醛均属于光降解杂质,除受上述处方因素影响之外,光稳定性试验表明包装材料也直接影响杂质含量的高低。参比制剂采用棕色的中硼硅玻璃安瓿瓶,而国内仿制制剂企业均采用透明的低硼硅玻璃安瓿瓶,对主药基本没有光保护作用,建议国内企业采用与参比制剂一致的包装材料。
综上,本研究采用高效液相色谱法建立了呋塞米及其注射液中11个已知杂质和1个潜在基因毒杂质的有关物质检查方法,对呋塞米注射液的杂质控制现状,主要降解杂质与处方和包装材料之间的相关性进行了分析研究,可为后续呋塞米注射液的安全性提高和仿制药的质量一致性评价提供参考。
