Objective:

To establish the quantitative analysis of multi-components by single marker(QAMS) method to simultaneously determine the contents of rutin,hyperoside,isoquercetin and quercetin in Zanthoxyli Pericarpium and Pericarpium Zanthoxyli Armati,and compare the differences in flavonoids contents.

Methods:

Methanol extract of Zanthoxyli Pericarpium was analyzed on an Agilent Eclipse Plus C18 (250 mm×4.6 mm,5 μm) chromatographic column. The mobile phase was acetonitrile(A)-0.1% formic acid(B) in a gradient elution. The flow rate was 1.0 mL·min-1. The column temperature was 35 ℃,and the detection wavelength was 360 nm. The injection volume was 10 μL. Using hyperoside as the internal standard substance,the relative correction factors for the other 3 components were calculate and the flavonoids contents in Zanthoxyli Pericarpium and Pericarpium Zanthoxyli Armati were determined.

Results:

The 4 components had good linear relationship within their respective ranges (r≥0.999 9). The average recovery rates of Zanthoxyli Pericarpium were 99.3%-105.1% (n=6),while thoes of Pericarpium Zanthoxyli Armati were 91.9%-99.8% (n=6). The relative average deviations of content of Zanthoxyli Pericarpium were 0.05%-3.37%,and the relative average deviations of content of Pericarpium Zanthoxyli Armati were 0.02%-3.28%. The results obtained by QAMS method were close to those obtained by the external standard method(ESM).

Conclusion:

This method of QAMS method is accurate and reliable. The total amount of flavonoids in Pericarpium Zanthoxyli Armati is significantly higher than that in Zanthoxyli Pericarpium. Which provides reference for the comprehensive evaluation of the quality research of Zanthoxyli Pericarpium and Pericarpium Zanthoxyli Armati.

目的:

建立一测多评(QAMS)法同时测定花椒和竹叶花椒中芦丁、金丝桃苷、异槲皮苷、槲皮苷的含量,比较黄酮成分含量差异。

方法:

花椒甲醇提取液分析,采用Agilent Eclipse Plus C18(250 mm×4.6 mm,5 μm)色谱柱,流动相为乙腈(A)-0.1%甲酸水(B),梯度洗脱,体积流量1.0 mL·min-1,柱温35 ℃,检测波长360 nm,进样量10 μL。以金丝桃苷为内标,计算其他3个成分的相对校正因子,测定花椒和竹叶花椒中4个黄酮成分的含量。

结果:

4个成分在各自范围内线性关系良好(r≥0.999 9),在花椒和竹叶花椒中的平均加样回收率(n=6)分别为99.3%~105.1%和91.9%~99.8%。花椒和竹叶花椒中4个黄酮成分含量的相对平均偏差分别在0.05%~3.37%和0.02%~3.28%,QAMS法所得结果与外标法(ESM)接近。

结论:

该方法准确可靠,花椒中黄酮总量显著高于竹叶花椒,可为全面评价花椒和竹叶花椒的质量研究提供参考。

目的:

建立一测多评(QAMS)法同时测定花椒和竹叶花椒中芦丁、金丝桃苷、异槲皮苷、槲皮苷的含量,比较黄酮成分含量差异。

方法:

花椒甲醇提取液分析,采用Agilent Eclipse Plus C18(250 mm×4.6 mm,5 μm)色谱柱,流动相为乙腈(A)-0.1%甲酸水(B),梯度洗脱,体积流量1.0 mL·min-1,柱温35 ℃,检测波长360 nm,进样量10 μL。以金丝桃苷为内标,计算其他3个成分的相对校正因子,测定花椒和竹叶花椒中4个黄酮成分的含量。

结果:

4个成分在各自范围内线性关系良好(r≥0.999 9),在花椒和竹叶花椒中的平均加样回收率(n=6)分别为99.3%~105.1%和91.9%~99.8%。花椒和竹叶花椒中4个黄酮成分含量的相对平均偏差分别在0.05%~3.37%和0.02%~3.28%,QAMS法所得结果与外标法(ESM)接近。

结论:

该方法准确可靠,花椒中黄酮总量显著高于竹叶花椒,可为全面评价花椒和竹叶花椒的质量研究提供参考。

Objective:

To establish the quantitative analysis of multi-components by single marker(QAMS) method to simultaneously determine the contents of rutin,hyperoside,isoquercetin and quercetin in Zanthoxyli Pericarpium and Pericarpium Zanthoxyli Armati,and compare the differences in flavonoids contents.

Methods:

Methanol extract of Zanthoxyli Pericarpium was analyzed on an Agilent Eclipse Plus C18 (250 mm×4.6 mm,5 μm) chromatographic column. The mobile phase was acetonitrile(A)-0.1% formic acid(B) in a gradient elution. The flow rate was 1.0 mL·min-1. The column temperature was 35 ℃,and the detection wavelength was 360 nm. The injection volume was 10 μL. Using hyperoside as the internal standard substance,the relative correction factors for the other 3 components were calculate and the flavonoids contents in Zanthoxyli Pericarpium and Pericarpium Zanthoxyli Armati were determined.

Results:

The 4 components had good linear relationship within their respective ranges (r≥0.999 9). The average recovery rates of Zanthoxyli Pericarpium were 99.3%-105.1% (n=6),while thoes of Pericarpium Zanthoxyli Armati were 91.9%-99.8% (n=6). The relative average deviations of content of Zanthoxyli Pericarpium were 0.05%-3.37%,and the relative average deviations of content of Pericarpium Zanthoxyli Armati were 0.02%-3.28%. The results obtained by QAMS method were close to those obtained by the external standard method(ESM).

Conclusion:

This method of QAMS method is accurate and reliable. The total amount of flavonoids in Pericarpium Zanthoxyli Armati is significantly higher than that in Zanthoxyli Pericarpium. Which provides reference for the comprehensive evaluation of the quality research of Zanthoxyli Pericarpium and Pericarpium Zanthoxyli Armati.

花椒为芸香科植物青椒Zanthoxylum schinifolium Sieb. et Zucc.或花椒Zanthoxylum bungeanum Maxim.的干燥成熟果皮,竹叶花椒为芸香科植物竹叶花椒Zanthoxylum armatum DC.的干燥成熟果皮。花椒味“辛”、性“温”,用于温中止痛,杀虫止痒[1],竹叶花椒亦具此功效[2]。花椒和竹叶花椒作为药食同源的传统中药,均含有挥发油、酰胺类、黄酮类、香豆素类等成分[2-7]
花椒Z. bungeanum Maxim.在我国具有两千多年的使用历史,早在秦汉时期就作为一种芳香防腐剂垫填内棺[8];《中华人民共和国药典》的另一基原青椒Z. schinifolium Sieb. et Zucc.与花椒的外观性状、化学成分、药理活性差异都较为明显,故不考虑纳入研究;竹叶花椒Z. armatum DC.最早记载于《本草图经》[9],历代本草关于功效描述与花椒基本一致,且外观性状、活性成分、药理作用都与花椒相似[10]。目前,市场上花椒和竹叶花椒不管是药用还是食用都远超于青椒,因此围绕花椒和竹叶花椒研究更有意义。黄酮类化合物是存在于植物药中的次生代谢物[11],花椒和竹叶花椒均具有良好的抗氧化活性[12-14],同时还具有抑菌[15]、抗炎镇痛[16]、抗抑郁[17]、保护心脑血管[18]等药理作用,这些都与黄酮成分相关。花椒和竹叶花椒的主要黄酮类成分具有相似性,均含有金丝桃苷、芦丁、槲皮苷等。目前的研究多集中于花椒和竹叶花椒中总黄酮含量[19-20],或黄酮类成分的HPLC含量测定[21-22],较少有采用一测多评(QAMS)法测定花椒和竹叶花椒黄酮成分含量的报道。QAMS法作为广泛应用于中药材质量控制的分析方法,可以实现单一对照品同时对多个物质成分含量测定[23]。本文共收集35批次花椒和26批次竹叶花椒,拟用QAMS法对花椒和竹叶花椒中的4个主要黄酮类成分进行含量测定,并与外标法(ESM)进行比较,评价该方法的有效性和准确性,深入挖掘含量测定结果,讨论花椒和竹叶花椒黄酮成分含量差异性,以期为花椒和竹叶花椒的质量研究提供参考。
Agilent 1260 Infinity Ⅱ型高效液相色谱仪(Agilent公司),Waters e2695-2998型高效液相色谱仪(Waters公司),十万分之一电子分析天平(梅特勒-托利多仪器有限公司),BS-110S电子分析天平(赛多利斯电子天平有限公司),KQ5200DE型超声波清洁器(昆山市超声仪器有限公司),纯水机(四川优普超纯科技有限公司)。
对照品:芦丁(批号 wkq20030203,纯度≥98%)、槲皮苷(批号 wkq23011612,纯度≥98%),购自四川省维克奇生物科技有限公司;金丝桃苷(批号 18070402,纯度≥98%),购自成都普菲德生物科技有限公司;异槲皮苷(批号 AFCK2052,纯度≥98%),购自成都埃法生物科技有限公司。乙腈为色谱纯(Fisher,批号 20231031),甲酸为分析纯(天津市津东天正精细化学试剂厂,批号 20230719)。
35批次花椒样品分别收集自四川、陕西、甘肃、山东、河北、山西等地,编号为HS1~HS35;26批次竹叶花椒样品分别收集自四川、贵州、重庆、云南、河南、河北等地,编号为ZS1~ZS26。经西南交通大学生命科学与工程学院宋良科副教授鉴定,分别为花椒Zanthoxylum bungeanum Maxim.和竹叶花椒Zanthoxylum armatum DC.。批次信息见表1
采用Agilent Eclipse Plus C18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈(A)- 0.1%甲酸水(B)为流动相,梯度洗脱(0~10 min,10%A;10~15 min,10%A→15%A;15~17 min,15%A→17%A;17~32 min,17%A;32~37 min,17%A→20%A;37~44 min,20%A;44~46 min,20%A→30%A;46~51 min,30%A;51~55 min,36%A→100%A),体积流量1.0 mL·min-1,柱温35 ℃,检测波长360 nm,进样量10 μL。在当前色谱条件下,各成分峰与相邻峰的分离度均>1.5。对照品及样品的色谱图见图1,对照品吸收光谱图见图2
精密称取芦丁、金丝桃苷、异槲皮苷、槲皮苷的对照品适量,加甲醇制成质量浓度分别为223 5、250 5、200 0、229 5 μg·mL-1的单一成分对照品储备液;各精密吸取0.3、0.4、0.5 mL,置于25 mL量瓶中,用甲醇稀释至刻度,摇匀,即得(浓度1:芦丁26.82 μg·mL-1、金丝桃苷30.06 μg·mL-1、异槲皮苷24.00 μg·mL-1、槲皮苷27.54 μg·mL-1;浓度2:芦丁35.76 μg·mL-1、金丝桃苷40.08 μg·mL-1、异槲皮苷32.00 μg·mL-1、槲皮苷36.72 μg·mL-1;浓度3:芦丁44.70 μg·mL-1、金丝桃苷50.10 μg·mL-1、异槲皮苷40.00 μg·mL-1、槲皮苷45.90 μg·mL-1)。
称取花椒和竹叶花椒粉末(过2号筛)约1.0 g,精密称定,分别置具塞锥形瓶,精密加入甲醇30 mL,称量,超声(功率200 W,频率40 kHz)辅助提取30 min,放冷,再称量,用甲醇补足减失的量,摇匀,0.45 μm微孔滤膜过滤,取续滤液,即得。
精密吸取“2.2.1”项下的对照品储备液0.1、1.0、2.0、5.0、8.0 mL,分别置于20 mL量瓶中,加甲醇稀释并定容至刻度,得到不同浓度梯度的混合对照品溶液(相当于0.5%、5%、10%、25%、40%对照品储备液浓度),按“2.1”项色谱条件进样分析,以各成分浓度为横坐标,峰面积为纵坐标,进行线性回归拟合,见表2。结果表明,花椒和竹叶花椒中的4个黄酮类成分质量浓度在相应的范围内线性关系良好。
精密吸取“2.2.1”项下的混合对照品溶液,按“2.1”项色谱条件连续进样6次,记录峰面积并计算其RSD,芦丁、金丝桃苷、异槲皮苷、槲皮苷峰面积的RSD分别为1.0%、0.87%、1.2%、1.6%,均<2.0%,表明仪器精密度良好。
取HS1和ZS1样品粉末,按“2.2.2”项方法分别制备6份花椒和竹叶花椒的供试品溶液,按“2.1”项色谱条件进样分析,记录峰面积并计算其含量RSD。花椒中芦丁、金丝桃苷、异槲皮苷、槲皮苷的平均含量分别为1.953、4.354、2.403、1.791 mg·g-1,RSD分别为1.2%、1.9%、1.7%、1.3%;竹叶花椒中芦丁、金丝桃苷、异槲皮苷、槲皮苷的平均含量分别为0.994、1.177、0.383、0.921 mg·g-1,RSD分别为0.98%、0.78%、1.6%、1.3%,RSD均小于2.0%,表明该方法重复性良好。
取HS1和ZS1样品粉末,按“2.2.2”项下方法分别制备花椒和竹叶花椒的供试品溶液,按“2.1”项色谱条件,分别于制备溶液后0、2、4、8、12、24 h进样分析,记录峰面积,结果花椒中芦丁、金丝桃苷、异槲皮苷、槲皮苷峰面积的RSD分别为0.97%、1.1%、1.9%、2.0%,竹叶花椒中芦丁、金丝桃苷、异槲皮苷、槲皮苷峰面积的RSD分别为0.99%、0.82%、1.6%、0.91%,RSD均小于2.0%,表明该方法稳定性良好。
取已知含量的HS1和ZS1样品粉末约0.5 g,各6份,精密称定,分别置于锥形瓶中,再精密加入适量的对照品,按“2.2.2”项下方法分别制备花椒和竹叶花椒的供试溶液,按“2.1”项色谱条件进样分析,计算加样回收率,结果见表34,花椒和竹叶花椒各成分加样回收率RSD符合要求,表明该方法准确性良好。
精密吸取“2.2.1”项3个不同浓度的混合对照品溶液,按“2.1”项色谱条件进样2、5、10、15、20 μL,记录各成分峰面积,以金丝桃苷为内标物,按照公式fkm为相对校正因子,wk为内标物浓度,Am为组分峰面积,wm为组分浓度,Ak为内标物峰面积)分别计算芦丁、异槲皮苷、槲皮苷的相对校正因子。结果见表5
取“2.2.1”项下的浓度3混合对照品溶液,按“2.1”项色谱条件进样分析,分别考察TC C18、Eclipse Plus C18、ZORBAX SB-C18等10种规格均为250 mm×4.6 mm,5 μm的色谱柱对相对校正因子的影响。结果见表6。各成分采用不同色谱柱的相对校正因子的RSD均<2.0%,表明不同色谱柱对相对校正因子的适用性较好。
取“2.2.1”项下的浓度3混合对照品溶液,按“2.1”项色谱条件进样分析,分别考察Agilent 1260和Waters e2695 2种高效液相色谱仪对相对校正因子的影响。结果见表7,表明不同高效液相色谱仪对相对校正因子的影响较小,重现性较好。
取“2.2.1”项下的浓度3混合对照品溶液,按“2.1”项色谱条件进样分析,考察柱温分别为25、30、35 ℃时对相对校正因子的影响,结果显示,各成分相对校正因子的RSD均<2.0%,表明在不同柱温下相对校正因子的适用性较好。结果见表8
取“2.2.1”项下的浓度3混合对照品溶液,按“2.1”项色谱条件进样分析,考察流速分别为0.8、1.0、1.2 mL·min-1对相对校正因子的影响,结果显示,各成分相对校正因子的RSD均<2.0%,表明在不同流速下相对校正因子的适用性较好。结果见表9
取“2.2.1”项下的浓度3混合对照品溶液,按“2.1”项色谱条件进样分析,以金丝桃苷为内标物,已知金丝桃苷的保留时间,计算芦丁、异槲皮苷、槲皮苷3个待测成分的相对保留时间,对待测成分进行峰定位。各成分保留时间范围:芦丁25.908~33.605 min、金丝桃苷27.173~35.138 min、异槲皮苷28.289~37.011 min、槲皮苷37.869~45.563 min。结果见表10,表明使用不同液相色谱仪和不同色谱柱重现性较好,花椒和竹叶花椒中3个待测黄酮成分的相对保留时间稳定性较好。
取35批花椒和26批竹叶花椒,按“2.2.2”项下方法制备供试品溶液,按“2.1”项色谱条件进样分析,记录色谱图信息。分别采用ESM和QAMS法测定4个黄酮成分的含量,结果花椒和竹叶花椒中4个黄酮成分含量的相对平均偏差分别在0.05%~3.37%和0.02%~3.28%,表明2种测定结果无显著差异,建立的QAMS法准确、可靠,可用于花椒和竹叶花椒黄酮成分含量测定。
以甲醇为空白,将混合对照品溶液在190~400 nm范围内进行紫外全波长扫描并记录紫外光谱图,发现金丝桃苷、芦丁、异槲皮苷和槲皮苷在360 nm处均有最大吸收,且色谱峰峰形较好,分离度较高,基线较为平稳,因此选择360 nm为4个黄酮成分的检测波长。本实验流动相条件同时考察了甲醇-水、乙腈-水、乙腈-0.1%磷酸水、甲醇-0.1%磷酸水、甲醇-0.1%甲酸水、乙腈-0.1%甲酸水,最终发现乙腈-0.1%甲酸水梯度洗脱效果最好,确定以乙腈-0.1%甲酸水进行梯度洗脱,流速1.0 mL·min-1,柱温35 ℃,梯度洗脱时间为55 min。
考察了超声和回流2种提取方式,发现黄酮含量提取差异不大,考虑超声提取方式较为便捷,因此选择超声辅助提取作为样品提取方式。实验还考察了甲醇、乙醇、20%甲醇、40%甲醇、60%甲醇、80%甲醇作为溶剂进行提取,发现甲醇提取时4个黄酮成分含量较高。考察了超声时长为20、30、40、50、60 min对黄酮含量的影响,发现超声时长为30、40、50、60 min时黄酮含量差别不大,将成分提取效率高、方法稳定性较好作为指标,最终以色谱图信息作为结果,确定供试品溶液制备方法。因此,选择甲醇为提取溶剂,超声辅助提取时间30 min,此时各成分出峰情况较好,可作为供试品溶液制备方法。
金丝桃苷属于黄酮醇苷类化合物,性质稳定,在花椒和竹叶花椒中峰面积最高,因此选择金丝桃苷为内标物符合QAMS法技术指南要求[24]。建立金丝桃苷与芦丁、异槲皮苷和槲皮苷3个黄酮成分的相对校正因子,通过比较不同高效液相色谱仪、不同色谱柱和色谱条件变化对相对校正因子的影响,以及在同一色谱条件下待测成分与内标物相对保留时间的影响。结果表明,各相对校正因子均具有良好的重复性,待测成分与内标物相对保留时间较为稳定。ESM和QAMS法测定结果表明二者无显著差异,方法准确、可靠,实现了花椒和竹叶花椒中4个主要黄酮成分低成本的含量测定,同时QAMS法可用于花椒和竹叶花椒药材的质量评价。
实验采用同一采收期的花椒和竹叶花椒,更有利于分析黄酮成分含量。比较花椒和竹叶花椒的黄酮成分总量,由图3可知,花椒黄酮成分总量显著高于竹叶花椒。若将黄酮成分作为质量评价指标之一,花椒质量整体上优于竹叶花椒。比较不同产地花椒黄酮成分,陕西、甘肃、山西等地金丝桃苷含量较高;芦丁在黄酮成分中含量最低,含量差异达11倍,总体含量值以甘肃最高,山东最低;异槲皮苷含量各产地整体分布均匀;槲皮苷含量以HS25(四川盐源)、HS26(四川汉源)最低,与其他产地含量差异明显,可能与贮藏时间有关。由此可见,甘肃、陕西的花椒黄酮成分含量较高,若以黄酮成分作为评价标准之一,主产区甘肃和陕西花椒总体质量较好。竹叶花椒黄酮成分含量差异远低于花椒,同样以芦丁含量差异大,最高值ZS25(四川汉源)和最低值ZS12(重庆江津)差异值达2.6倍,低于花椒;金丝桃苷、异槲皮苷、槲皮苷含量整体分布良好;总体上竹叶花椒以主产区云南昭通黄酮成分含量最高,整体质量水平较为均一。以上结果表明不同产地的花椒和竹叶花椒黄酮成分含量差异较大,黄酮成分含量一定程度上能够评价出花椒和竹叶花椒的质量。将含量数据导入SIMCA-P14.0软件进行分析,建立偏最小二乘判别分析(OPLS-DA)模型,结合变量重要性投影(VIP)法,以VIP贡献值大于1为条件进行筛选,结果金丝桃苷、异槲皮苷、槲皮苷VIP值均>1,表明金丝桃苷、异槲皮苷、槲皮苷可能是花椒和竹叶花椒之间主要的黄酮类含量差异性成分(图4~6),这对花椒和竹叶花椒的质量比较研究具有借鉴意义。
花椒和竹叶花椒由于外观性状和物质基础相似,具有同等研究价值,黄酮类成分作为花椒和竹叶花椒抗氧化等活性作用的关键成分具有重要研究意义。本研究建立花椒和竹叶花椒黄酮类成分QAMS分析方法,实现了对花椒和竹叶花椒中芦丁、金丝桃苷、异槲皮苷、槲皮苷4个主要黄酮成分的含量测定,该方法准确,具有良好的可信度。含量测定结果发现,花椒和竹叶花椒4个黄酮成分含量差异较大,深入挖掘数据信息发现,金丝桃苷、异槲皮苷、槲皮苷可能是花椒和竹叶花椒之间主要的黄酮类含量差异性成分,该结果有助于为花椒和竹叶花椒质量比较研究提供参考,为更加全面评价花椒和竹叶花椒药材及饮片质量提供参考依据。花椒和竹叶花椒成分复杂,含有挥发油、酰胺、生物碱等多种化学成分[325],本文只研究黄酮类成分,其他成分尚未涉及,下一步会结合气相色谱-质谱(GC-MS)联用和液相色谱-质谱(LC-MS)联用分析花椒和竹叶花椒挥发性成分以及其他化学成分,全面分析2种基原花椒成分,为2种基原花椒质量控制研究奠定基础。
