Objective:

To establish the first batch of national drug reference standard of L-methionine sulfoxide.

Methods:

The molecular structure of methionine sulfoxide was confirmed by the methods of IR,NMR and high resolution MS. Moreover,the moisture,residual solvents,residual ignition and related substances of L-methionine sulfoxide were determined,and then,its hygroscopicity,homogeneity and stability were also observed. Finally,mass balance method,HPLC and quantitative NMR were applied to determine and calculate its contents.

Results:

The results of experiments showed the structure of L-methionine sulfoxide were confirmed,its contents of moisture,residual ignition,metal elements and related substances were 0.23%,0.20%,10.87 μg·g-1 and 5.09%. The residual solvents were not detected in the samples. The final content of methionine sulfoxide was 94.5% by mass balance method. The contents of this batch of L-methionine sulfoxide were uniform,and it is stable in short-terms.

Conclusion:

The first batch of national drug reference standard of L-methionine sulfoxide was established,and it is used for the inspection for related substances of amino acid drug,which is helpful in normalizing drug quality and guaranteeing drug safety.

目的:

建立首批L-甲硫氨酸亚砜国家药品对照品。

方法:

采用红外吸收光谱仪、核磁共振波谱仪及高分辨质谱仪对L-甲硫氨酸亚砜对照品原料进行结构确证,并检查其水分、炽灼残渣、金属元素含量、残留溶剂、有关物质,采用质量平衡法、液相色谱法及定量核磁法对其含量进行测定和计算,采用质量平衡法对其含量进行赋值,最后考察了其引湿性、均匀性和稳定性。

结果:

试验结果确证了L-甲硫氨酸亚砜的结构,水分为0.23%,炽灼残渣为0.20%,金属元素含量为10.87 μg·g-1,有关物质为5.09%,未检出残留溶剂。以质量平衡法赋值首批L-甲硫氨酸亚砜对照品的含量为94.5%。本批次对照品含量均匀,短期稳定性试验结果证明L-甲硫氨酸亚砜短期内稳定。

结论:

首批L-甲硫氨酸亚砜国家药品对照品的研制成功,可用于氨基酸类药品有关物质的检查,规范相关药品质量,保证用药安全。

目的:

建立首批L-甲硫氨酸亚砜国家药品对照品。

方法:

采用红外吸收光谱仪、核磁共振波谱仪及高分辨质谱仪对L-甲硫氨酸亚砜对照品原料进行结构确证,并检查其水分、炽灼残渣、金属元素含量、残留溶剂、有关物质,采用质量平衡法、液相色谱法及定量核磁法对其含量进行测定和计算,采用质量平衡法对其含量进行赋值,最后考察了其引湿性、均匀性和稳定性。

结果:

试验结果确证了L-甲硫氨酸亚砜的结构,水分为0.23%,炽灼残渣为0.20%,金属元素含量为10.87 μg·g-1,有关物质为5.09%,未检出残留溶剂。以质量平衡法赋值首批L-甲硫氨酸亚砜对照品的含量为94.5%。本批次对照品含量均匀,短期稳定性试验结果证明L-甲硫氨酸亚砜短期内稳定。

结论:

首批L-甲硫氨酸亚砜国家药品对照品的研制成功,可用于氨基酸类药品有关物质的检查,规范相关药品质量,保证用药安全。

Objective:

To establish the first batch of national drug reference standard of L-methionine sulfoxide.

Methods:

The molecular structure of methionine sulfoxide was confirmed by the methods of IR,NMR and high resolution MS. Moreover,the moisture,residual solvents,residual ignition and related substances of L-methionine sulfoxide were determined,and then,its hygroscopicity,homogeneity and stability were also observed. Finally,mass balance method,HPLC and quantitative NMR were applied to determine and calculate its contents.

Results:

The results of experiments showed the structure of L-methionine sulfoxide were confirmed,its contents of moisture,residual ignition,metal elements and related substances were 0.23%,0.20%,10.87 μg·g-1 and 5.09%. The residual solvents were not detected in the samples. The final content of methionine sulfoxide was 94.5% by mass balance method. The contents of this batch of L-methionine sulfoxide were uniform,and it is stable in short-terms.

Conclusion:

The first batch of national drug reference standard of L-methionine sulfoxide was established,and it is used for the inspection for related substances of amino acid drug,which is helpful in normalizing drug quality and guaranteeing drug safety.

L-甲硫氨酸亚砜(L-methionine sulfoxides)由L-甲硫氨酸氧化而来,是含有甲硫氨酸成分药物中的一种常见杂质[1-2],会对药物的质量安全产生影响[3-5]。甲硫氨酸氧化后会失去原有活性,导致甲硫氨酸实际含量降低,影响药效[6];蛋白中的甲硫氨酸残基也会被氧化成亚砜残基,据文献报道,这种改变会影响蛋白质本身的生物活性[7-11],因此也会对相关蛋白质药物的药效产生影响。同时,越来越多的研究表明,甲硫氨酸和L-甲硫氨酸亚砜以及相关组成的蛋白质在生物体内发挥着不同的作用[12-15],因此,甲硫氨酸氧化为L-甲硫氨酸亚砜这一变化过程会对很多药物质量产生影响。L-甲硫氨酸亚砜也是氨基酸注射液等临床用药或相关生物制品质控中的重要检查对象。为规范产品质量,减少不良反应的发生,满足企业生产需求,同时也为相关药物研发或治疗方法的开发提供基础对照,研制L-甲硫氨酸亚砜国家对照品具有重要意义。本文对L-甲硫氨酸亚砜原料进行了质量分析,建立了首批L-甲硫氨酸亚砜国家药品对照品。
XPE205电子天平,d=0.01 mg,Mettler Toledo公司;UV-2600紫外-可见分光光度计,Shimadu公司;Alpha Ⅱ红外光谱仪,Bruker公司;QE高分辨质谱仪,Thermo Fisher公司;LC-40液相色谱仪,Shimadu公司;MP230K pH计,Mettler Toledo公司;Metrohm 890水分测定仪,Metrohm公司;7890B+7697A顶空进样-气相色谱仪,Agilent公司;Optima 7000 ICP-OES,PE公司;350 ICP-MS,PE公司;Advantage动态水分吸附仪,SMS公司;Flash Smart氮分析仪,Thermo Fisher公司;AVANCE NEO 600M核磁共振仪,Bruker公司。
L-甲硫氨酸亚砜原料(批号0615-RD-0010)购自广州佳途科技股份有限公司;L-甲硫氨酸亚砜(98.0%)、L-甲硫氨酸砜(99.1%)对照品购自USP;18种必须氨基酸及脯氨酸对照品(批号140624-201506)、马来酸内标对照品(批号510137-201601,纯度99.8%),中国食品药品检定研究院;钴、锂、砷、镉、汞、铅、铜、钒、镍、锑、铯等12种金属元素标准溶液购自中国计量科学研究院,质量浓度均为1 000 μg·mL-1;甲醇、乙腈、二氯甲烷、丙酮等有机溶剂均为色谱纯,购自ThermoFisher公司;重水购自Sigma公司;磷酸、硫酸、醋酸、磷酸盐等无机试剂购自国药集团化学试剂有限公司。
目视供试品为淡黄色粉末,无特殊异味。
取本品适量,置动态水分吸附分析仪中测定,增重为2.18%,归类为有引湿性(2%~15%)。
采用溴化钾压片法制样,分别取适量供试品和对照品加溴化钾研细后压片制样,利用傅里叶变换红外光谱仪扫描,次数为32,分辨率为4.0,得到供试品和对照品的红外光吸收图谱,如图1所示,二者红外吸收光谱一致。
分别称取适量L-甲硫氨酸亚砜对照品和供试品,用重水配制成质量浓度约为10 mg·mL-1的溶液,置核磁管中测试,结果如图2所示,对照品与供试品的氢谱、碳谱图谱一致。
称取适量供试品,用乙腈-水(1:1)配制成质量浓度约为0.1 μg·mL-1的供试品溶液,用高分辨质谱负离子模式扫描,喷雾电压设置为3.5 kV,鞘气流速为3 030 kPa,辅助器流速为1 010 kPa,辅助气温度320 ℃,毛细管电压15 V,毛细管温度320 ℃,分辨率为70 000。结果如图3所示,[M-H]-峰的质荷比为164.037 40,其理论计算值应为164.038 69,误差为δ 7.86,说明测量值与理论值吻合。
取供试品约20 mg,置10 mL顶空瓶中,压盖密封,采用卡式炉加热库仑滴定方法测定,卡式炉温度为120 ℃,平行测定6次,空白2次,最后测得平均水分含量为0.23%。
将坩埚750 ℃炽灼冷却后称量,准确称取供试品约1.0 g,置于恒重后的坩埚中炽灼完全后冷却称量,经测定,供试品炽灼残渣含量为0.20%。
精密称定供试品约200 mg,置于20 mL顶空瓶中,加无水硫酸钠0.2 g及水1 mL后压盖密封,剧烈震荡促进分散,后用顶空进样-气相色谱法进行测定,顶空温度为90 ℃,时间为30 min,进样口200 ℃,色谱柱为DB-624,50 ℃保持6 min,以40 ℃·min-1后升至220 ℃保持2 min,FID检测器,温度为280 ℃。结果无溶剂峰检出,表明供试品未检出残留溶剂。
精密称定供试品约25 mg,用2%稀硝酸溶解并定容至50 mL备用,用2%稀硝酸稀释钾元素的标准溶液得到质量浓度分别为1、5、10、20 μg·mL-1的系列溶液,建立标准曲线,以2%稀硝酸溶液为空白溶液,采用电感耦合等离子体发射光谱法测定,外标法定量,结果测得供试品中钾、钙、钠的含量分别为0.01%、0.05%、0.05%。
将12种金属元素的标准溶液用5%硝酸配制成系列浓度的对照溶液(不同元素浓度不同),分别加金元素的标准溶液。取供试品适量,精密称定,加硝酸2.5 mL消解后加水稀释,加金元素标准溶液(使得金的终浓度为0.1 μg·mL-1),将以上溶液用电感耦合等离子体质谱法测定,对照品中钴含量0.06 μg·g-1、砷含量0.36 μg·g-1、镉含量0.02 μg·g-1、汞含量0.04 μg·g-1、铅含量0.11 μg·g-1、铜含量9.07 μg·g-1、镍含量·1.15 μg·g-1、锑含量0.03 μg·g-1、铯含量0.03 μg·g-1,总量为10.87 μg·g-1
分别对氨基酸类和非氨基酸类的有关物质进行考察。氨基酸类有关物质采用氨基酸分析仪,离子交换柱-茚三酮衍生法进行检查,非氨基酸类有关物质采用C18柱紫外检测进行检查。
氨基酸类有关物质检查,使用Skyam LCAK06/Na柱分离,枸橼酸缓冲液-硼酸盐缓冲液-甲醇洗脱体系进行洗脱,检测波长为570 nm(一级氨基酸)和440 nm(二级氨基酸),系统适应性为18种氨基酸及脯氨酸混合溶液,采用主成分自身对照法计算,结果表明氨基酸类有关物质主要为甲硫氨酸砜(含量4.25%),部分为谷氨酸(含量0.08%),二者总含量为4.33%,其他氨基酸未检出,如图45所示。
非氨基酸类有关物质检查,采用乙腈-磷酸水溶液体系洗脱,经试验,甲硫氨酸砜和谷氨酸不存在末端吸收,故检测波长采用205 nm。结果表明,非氨基酸类有关物质总量为0.76%,其中单个最大有关物质含量为0.53%,如图4所示。最后确定,本批次甲硫氨酸有关物质总含量为5.09%,单个最大含量有关物质为甲硫氨酸砜,含量为4.25%,最大单个未知杂质含量为0.53%。
分别用氮分析法、核磁定量法和质量平衡法来测定和计算供试品的含量。氮分析法中左右炉温分别为950 ℃和840 ℃,柱温50 ℃,载气流速140 mL·min-1,参比流速100 mL·min-1,氧气流速250 mL·min-1,分析时间450 s,注氧时间15 s,扣除L-甲硫氨酸砜和谷氨酸,测得甲硫氨酸亚砜含量为94.6%。称取样品14.72 mg,马来酸内标14.39 mg,用氘代DMSO溶解后,置核磁测定管中上机测定,扣除L-甲硫氨酸砜,测得含量94.7%。根据水分、炽灼残渣及HPLC纯度的测定结果,以质量平衡法计算,供试品(L-甲硫氨酸亚砜对照品)的HPLC法含量=(1-水分-炽灼残渣)×HPLC纯度×100%,结果为94.5%。3种方法测得的结果基本一致,最终采用质量平衡法赋值,为94.5%。
随机取供试品24份,每份约20 mg,用水溶解并定容至50 mL量瓶中,供液相色谱分析用,采用“2.2.5”项下试验方法进行测定。对目标峰面积与称样量的比值进行单因素方差分析,P值为0.78,表明各取样间无明显差异,供试样均匀性较好。
取适量供试品置于40 ℃,75%相对湿度的环境条件下,分别避光储存0、5、10 d,使用液相色谱测定不同处理时间下样品中L-甲硫氨酸亚砜含量及杂质的变化(方法同均匀性),测定结果(主成分自身对照法)显示,0、5、10 d处理的主成分含量分别为95.32%、95.31%、95.53%,杂质含量分别为4.68%、4.69%、4.47%,表明储存期内无明显变化,短期稳定。
对供试品作为首批L-甲硫氨酸亚砜对照品进行标定,对其化学结构确证、纯度及杂质、理化性质等进行了考察,并采用氮分析法、质量平衡法及核磁共振法多种方法对其含量进行测定。此外,参考2020年版《中华人民共和国药典》四部通则<9901国家药品标准物质制备指导原则>,对本批待标品的均匀性和稳定性进行考察。最终通过WHO推荐的标准物质定值法,即质量平衡法,以扣除水分、炽灼残渣、残留溶剂后的HPLC纯度对本批对照品含量进行赋值。经国家药品标准物质专家委员会审定,批准其作为L-甲硫氨酸亚砜检查用国家药品对照品,其含量为94.5%。
