Objective:

To investigate different batches of Runbi Tongqiao drops by fingerprint and multi-index component content determination method, and to provide basis for its quality evaluation.

Methods:

Acclaim TM-C18 chromatographic column(250 mm×4.6 mm, 5 μm) was used with methanol(A)-acetonitrile(B) -0.2% formic acid aqueous solution(C) as the mobile phase with gradient elution. The flow rate was 0.8-1.0 mL·min-1. The injection volume was 10 μL and the detection wavelength was 327 nm. The HPLC fingerprint of Runbi Tongqiao drops was established, and the quality consistency of Runbi Tongqiao drops was compared by stoichiometric method. The contents of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, luteoloside and isochlorogenic acid C were determined by HPLC.

Results:

A total of 14 common peaks were calibrated in the fingerprints of 10 batches of samples, and the similarities were ≥0.933. Seven common peaks of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, luteoloside and isochlorogenic acid C were identified by reference substances. The contents of seven components in Runbi Tongqiao drops were determined simultaneously, which were 0.131 5-0.133 8 mg·mL-1, 0.095 5-0.098 9 mg·mL-1, 0.087 4-0.090 1 mg·mL-1, 0.012 8-0.015 8 mg·mL-1, 0.010 3-0.013 7 mg·mL-1, 0.022 7-0.024 9 mg·mL-1 and 0.006 8-0.008 6 mg·mL-1, respectively.

Conclusion:

The established fingerprint of Runbi Tongqiao drops is stable and reliable, and the simultaneous determination method of multi-component content is simple and fast. It can be used for the quality control of Runbi Tongqiao drops, which lays a solid foundation for the follow-up study of Runbi Tongqiao drops.

目的:

采用指纹图谱、多指标成分含量测定方法研究不同批次润鼻通窍滴剂,为其质量评价提供依据。

方法:

采用Acclaim TM-C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇(A)-乙腈(B)-0.2%甲酸水溶液(C)为流动相,进行梯度洗脱,流速0.8~1.0 mL·min-1,进样量10 μL,检测波长327 nm。建立润鼻通窍滴剂的HPLC指纹图谱,利用化学计量方法比较润鼻通窍滴剂的质量一致性。采用HPLC法对新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C 7个成分进行多成分含量测定。

结果:

10批次样品指纹图谱共标定了14个共有峰,相似度均≥0.933,并利用对照品指认了新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C 7个共有峰。同步测定润鼻通窍滴剂中7个成分的含量,分别为0.131 5~0.133 8、0.095 5~0.098 9、0.087 4~0.090 1、0.012 8~0.015 8、0.010 3~0.013 7、0.022 7~0.024 9、0.006 8~0.008 6 mg·mL-1

结论:

建立的润鼻通窍滴剂指纹图谱稳定可靠,多成分含量同时测定方法简便快捷,可用于润鼻通窍滴剂的质量控制,为润鼻通窍滴剂的后续研究奠定了坚实的基础。

目的:

采用指纹图谱、多指标成分含量测定方法研究不同批次润鼻通窍滴剂,为其质量评价提供依据。

方法:

采用Acclaim TM-C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇(A)-乙腈(B)-0.2%甲酸水溶液(C)为流动相,进行梯度洗脱,流速0.8~1.0 mL·min-1,进样量10 μL,检测波长327 nm。建立润鼻通窍滴剂的HPLC指纹图谱,利用化学计量方法比较润鼻通窍滴剂的质量一致性。采用HPLC法对新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C 7个成分进行多成分含量测定。

结果:

10批次样品指纹图谱共标定了14个共有峰,相似度均≥0.933,并利用对照品指认了新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C 7个共有峰。同步测定润鼻通窍滴剂中7个成分的含量,分别为0.131 5~0.133 8、0.095 5~0.098 9、0.087 4~0.090 1、0.012 8~0.015 8、0.010 3~0.013 7、0.022 7~0.024 9、0.006 8~0.008 6 mg·mL-1

结论:

建立的润鼻通窍滴剂指纹图谱稳定可靠,多成分含量同时测定方法简便快捷,可用于润鼻通窍滴剂的质量控制,为润鼻通窍滴剂的后续研究奠定了坚实的基础。

Objective:

To investigate different batches of Runbi Tongqiao drops by fingerprint and multi-index component content determination method, and to provide basis for its quality evaluation.

Methods:

Acclaim TM-C18 chromatographic column(250 mm×4.6 mm, 5 μm) was used with methanol(A)-acetonitrile(B) -0.2% formic acid aqueous solution(C) as the mobile phase with gradient elution. The flow rate was 0.8-1.0 mL·min-1. The injection volume was 10 μL and the detection wavelength was 327 nm. The HPLC fingerprint of Runbi Tongqiao drops was established, and the quality consistency of Runbi Tongqiao drops was compared by stoichiometric method. The contents of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, luteoloside and isochlorogenic acid C were determined by HPLC.

Results:

A total of 14 common peaks were calibrated in the fingerprints of 10 batches of samples, and the similarities were ≥0.933. Seven common peaks of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, isochlorogenic acid B, isochlorogenic acid A, luteoloside and isochlorogenic acid C were identified by reference substances. The contents of seven components in Runbi Tongqiao drops were determined simultaneously, which were 0.131 5-0.133 8 mg·mL-1, 0.095 5-0.098 9 mg·mL-1, 0.087 4-0.090 1 mg·mL-1, 0.012 8-0.015 8 mg·mL-1, 0.010 3-0.013 7 mg·mL-1, 0.022 7-0.024 9 mg·mL-1 and 0.006 8-0.008 6 mg·mL-1, respectively.

Conclusion:

The established fingerprint of Runbi Tongqiao drops is stable and reliable, and the simultaneous determination method of multi-component content is simple and fast. It can be used for the quality control of Runbi Tongqiao drops, which lays a solid foundation for the follow-up study of Runbi Tongqiao drops.

润鼻通窍滴剂是贵州中医药大学第二附属医院研制的医疗机构制剂,主要由鹅不食草、鱼腥草、白芷、辛夷、炒苍耳子、防风、炙麻黄、山银花、菊花、石菖蒲、牡丹皮和薄荷组成。具有宣肺通窍、润鼻止痒、消肿利气的功效,主要用于治疗过敏性鼻炎及感染后鼻炎引起的阵发性鼻痒、鼻干、喷嚏、鼻胀塞、鼻黏膜干燥充血、清涕,甚至涕中带血等诸证。该制剂在贵州中医药大学第二附属医院临床使用疗效良好,为了保障该制剂使用的安全有效和质量可控,对其质量控制和评价方法尚待深入研究。因此,通过查阅相关文献[1-10],并根据前期研究结果,本文通过HPLC指纹图谱结合多成分同步含量测定方法,从定性和定量2个方面开展润鼻通窍滴剂的质量控制和质量评价研究,为完善该制剂的质量标准制定提供实验依据。
Thermo Ultimate 3000型高效液相色谱仪、二极管阵列(DAD)检测器(赛默飞世尔科技),AG135型十万分之一电子天平(上海梅特勒有限公司),SK5200H超声波清洗器(上海科导超声仪器有限公司,频率53 kHz,功率200 W)。
润鼻通窍滴剂10批(规格10 mL·支-1,批号20220401、20220402、20220403、20220404、20220405、20220406、20220407、20220408、20220409、20220410,编号S1~S10)为贵州中医药大学第二附属医院自制;对照品绿原酸(批号AZ221011851,纯度≥98%)、新绿原酸(批号AZ21102901,纯度≥98%)、隐绿原酸(批号AFBL2251,纯度≥98%)、异绿原酸A(批号AZ22042903,纯度≥98%)、异绿原酸B(批号AZ21092203,纯度≥98%)购自成都埃法生物科技股份有限公司,木犀草苷(批号PS000708,纯度≥98%)和异绿原酸C(批号PS001057,纯度≥98%)购自成都普思生物科技股份有限公司。甲醇(纯度99.99%,安徽天地高纯溶剂有限公司)、色谱纯乙腈(上海跃盛贸易有限公司);分析纯甲酸(天津科密欧化学试剂有限公司);水为怡宝纯净水。
鹅不食草(批号20220301)、鱼腥草(批号211116)、白芷(批号20220701)、辛夷(批号210628)、炒苍耳子(批号210327)、防风(批号20220701)、炙麻黄(批号211213)、山银花(批号210605)、菊花(批号20220601)、石菖蒲(批号211021)、牡丹皮(批号210715)和薄荷(批号103620210701),以上12味药材均购自贵州同济堂中药饮片有限公司,由贵州中医药大学第二附属医院主任药师高永跃鉴别为正品。
采用Acclaim TM-C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇(A)-乙腈(B)-0.2%甲酸水溶液(C)为流动相,梯度洗脱(0~2 min,5%A→18%A,1.0 mL·min-1;2~20 min,18%A→28%A,1.0 mL·min-1;20~30 min,28%A→42%A,1.0~0.80 mL·min-1;30~45 min,42%A,0.80 mL·min-1;45~60 min,42%A→93%A,0.80~1.0 mL·min-1),检测波长327 nm,进样量10 μL,柱温30 ℃。
分别精密称取对照品新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C适量,加甲醇溶解并制成浓度分别为1.000、1.000、1.001、0.202 0、0.199 0、0.200 0、0.202 0 mg·mL-1的单一对照品储备液。分别精密吸取上述对照品储备液加甲醇制成浓度分别为0.090 0、0.100 0、0.070 7、0.020 2、0.019 9、0.020 0、0.020 2 mg·mL-1的混合对照品溶液,备用。
取润鼻通窍滴剂5支混匀作为供试品溶液,取适量过0.45 μm微孔滤膜,即得。
分别取润鼻通窍滴剂中12味原药材,按照处方中对应的处方量比例进行取样,以润鼻通窍滴剂制剂工艺制得相应的单味药材样品溶液,取适量过0.45 μm微孔滤膜,即得。
按润鼻通窍滴剂处方比例和制备工艺,制备同时缺鱼腥草、山银花、菊花、炒苍耳子的阴性样品,过0.45 μm微孔滤膜,即得。
取S2号样品的供试品溶液适量,在“2.1”项色谱条件下连续进样6次,记录色谱峰,以2号新绿原酸色谱峰为参照峰(S),计算得到14个共有峰的相对保留时间RSD为0.045%~0.14%,峰面积比值的RSD为0.15%~1.9%,表明仪器精密度良好。
按“2.2.2”项下方法平行制备6份S2号样品的供试品溶液,在“2.1”项色谱条件下进样测定,记录色谱峰,以2号峰(新绿原酸)为参照峰,计算各共有峰相对保留时间的RSD在0.042%~0.26%,峰面积比值的RSD为0.18%~2.8%,表明该方法重复性良好。
取“3.1.2”项的供试品溶液1份,分别于0、4、8、12、16、24 h,在“2.1”项色谱条件下进样测定,记录色谱峰,以2号峰(新绿原酸)为参照峰,计算得各共有峰相对保留时间的RSD在0.035%~0.32%,峰面积比值的RSD在0.30%~2.5%,表明供试品溶液测定在24 h内稳定性良好。
分别取10批润鼻通窍滴剂(S1~S10)适量,同“2.2.2”项下制备供试品溶液,按“2.1”项下色谱条件进行HPLC分析,将样品的数据导入《中药色谱指纹图谱相似度评价系统(2004A版)》,设置S1为参照图谱,进行多点校正后自动匹配,得到10批润鼻通窍滴剂供试品的指纹图谱共有模式及对照指纹图谱(图1),以生成的对照图谱作为参照图谱(R),对10批样品进行相似度评价,并以2号峰作为参照峰计算各色谱峰的相对保留时间和相对峰面积的RSD。结果显示,S1~S10批润鼻通窍滴剂与对照图谱的相似度均>0.92,表明10批样品的差异较小,化学成分基本一致。14个共有峰的相对保留时间RSD为0.056%~0.20%,相对峰面积的RSD为0.48%~8.9%,表明不同批次润鼻通窍滴剂样品间的指纹图谱整体无差异,但色谱峰高或峰面积存在一定差异,可用于润鼻通窍滴剂定性鉴别。
在327 nm下共检出了14个共有峰,通过对照品比对指认出7个峰。其中峰2为新绿原酸、峰3为绿原酸、峰4为隐绿原酸、峰6为异绿原酸B、峰7异绿原酸A、峰9为木犀草苷、峰10为异绿原酸C,混合对照品溶液HPLC图谱见图2
依据紫外光谱信息和保留时间,对润鼻通窍滴剂与单味药的指纹图谱的相关峰进行比对,确定特征峰的来源归属,分别取12个原药材样品溶液,按“2.1”项下色谱条件进样测定,色谱图见图2,其中色谱峰1归属于鹅不食草;色谱峰2(新绿原酸)归属于鱼腥草、菊花;色谱峰3(绿原酸)归属于菊花、山银花、苍耳子;色谱峰4(隐绿原酸)归属于菊花、山银花;色谱峰5、9(木犀草苷)、11、13归属于菊花;色谱峰6(异绿原酸B)、7(异绿原酸A)归属于菊花、山银花;色谱峰8归属于鹅不食草、菊花;色谱峰10(异绿原酸C)归属于菊花、山银花;色谱峰12归属于白芷、菊花;色谱峰14归属于石菖蒲。由于薄荷、辛夷、炙麻黄、防风在处方中的量较低,且均含挥发性成分,在用水煎煮过程中容易损失,故在此条件下响应值低,未见其色谱峰;鱼腥草、鹅不食草、苍耳子、石菖蒲、白芷归属峰较少的原因,可能是煎煮过程中有大量成分挥发。
将10批润鼻通窍滴剂指纹图谱的14个量化共有峰峰面积导入SPSS 26.0软件,进行系统聚类,根据聚类分析图(图3)结果可知,10批供试品指纹图谱可分为2大类,其中S1、S2、S3、S10聚为1类,S4、S5、S6、S7、S8、S9聚为第二类,聚类结果显示二者在14个成分的含量上存在一定的差异,这可能与润鼻通窍滴剂生产工艺及药材批次间差异有关。
以10批润鼻通窍滴剂指纹图谱中14个共有峰的峰面积为变量,利用SPSS 26.0统计分析软件进行主成分分析(PCA),计算主成分特征值,累计方差贡献率,特征值>1的主成分有3个,累计方差贡献率为86.284%。色谱峰2、6、7、9、10在主成分1有较高的正载荷,色谱峰12、13、14、在主成分2有较高的正载荷,色谱峰3在主成分3上有较高的正载荷。分别以主成分1、2、3建立其三维坐标体系,由图4可知,S1、S2和S3可聚为1类;S4、S6聚为1类;同理S5、S10可聚为1类;S7、S8、S9较分散,各聚为1类。该分析结果与相似度评价、聚类分析结果有一定的差异。
分别吸取混合对照品溶液、供试品溶液、阴性样品溶液适量,按照“2.1”项下色谱条件分别进样,记录色谱图。结果显示,在新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷和异绿原酸C对照品相应位置上,供试品溶液中均有相同保留时间的色谱峰出现,且各相应色谱峰的紫外吸收(UV)光谱图与对照品的UV光谱图基本一致,而阴性样品溶液色谱图中在对应的位置上无色谱峰出现,说明阴性没有干扰,表明该方法专属性良好。结果见图5
分别精密吸取“2.2.1”项下的单一对照品储备液,加甲醇制成系列浓度的混合对照品溶液,按“2.1”项下色谱条件进样测定,记录峰面积,以对照品溶液的质量浓度X为横坐标,峰面积Y为纵坐标,绘制标准曲线,线性关系见表1
精密吸取“2.2.1”项下混合对照品溶液适量,按照“2.1”项下的色谱条件连续进样6次,记录峰面积。结果显示,新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C,各成分峰面积值的RSD(n=6)分别为0.57%、0.37%、0.40%、0.42%、0.71%、1.2%、0.57%,表明仪器精密度良好。
按“2.2.2”项下方法平行制备6份S2号样品的供试品溶液,在“2.1”项色谱条件下进样测定,记录峰面积。结果显示,新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C的平均含量分别为0.126 7、0.092 2、0.083 6、0.010 8、0.009 0、0.022 9、0.005 9 mg·mL-1,各成分含量的RSD(n=6)分别为0.43%、0.20%、1.0%、2.6%、2.9%、2.5%、2.3%。结果表明该方法的重复性良好。
取“4.4”项下供试品溶液1份,分别于0、4、8、12、16、24 h,在“2.1”项色谱条件下进样测定,记录峰面积。结果显示,新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C峰面积的RSD(n=6)分别为0.33%、0.33%、0.51%、0.30%、0.90%、1.4%、1.3%,表明供试品溶液在24 h内测定稳定性良好。
精密吸取6份润鼻通窍滴剂样品(S2)各5.0 mL,每份按1∶1的比例精密加入各对照品(新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C),按“2.2.2”项下方法平行制备6份供试品溶液,按“2.1”项下色谱条件进样分析。结果7个成分的平均加样回收率为93.3%~103.6%,RSD均<3%,该方法测定结果的准确度良好。
取10批润鼻通窍滴剂(S1~S10),依“2.2.2”项下方法制备供试品溶液,按“2.1”项下的色谱条件进样分析,含量测定结果见表2
试验对不同色谱柱(AQUASIL C18和Acclaim TM-C18),不同流动相(乙腈-0.3%乙酸水溶液、乙腈-0.2%乙酸水溶液、甲醇-0.2%磷酸水溶液、甲醇-0.2%甲酸水溶液、甲醇-乙腈-0.2%甲酸水溶液)的分离情况进行了考察。结果以Acclaim TM-C18色谱柱,流动相为甲醇-乙腈-0.2%甲酸水溶液时,峰数目更多,且分离度更高,响应值较高,表明该条件分离效果更佳。采用DAD检测器在225、285、327和345 nm 4个波长处比较供试品溶液色谱峰的响应情况,结果显示,大部分成分的色谱峰在327 nm下吸收较大、分离效果较优,故选择327 nm为检测波长。其次,考察了柱温在25、30和35 ℃时的分离效果,发现柱温在30 ℃时7个待测成分成分分离情况较好,且峰形较好,故选择30 ℃作为检测柱温。
通过查阅文献发现咖啡酰奎宁酸(绿原酸、新绿原酸、隐绿原酸)和二咖啡酰奎宁酸(异绿原酸A、异绿原酸B和异绿原酸C)化合物具有显著的镇咳、祛痰、抗炎、抗过敏[13],且对很多致病菌具有显著的抑制作用[14]。木犀草苷具有解热抗炎[15]、止咳[16]、抗病毒作用[17]。该指标与润鼻通窍滴剂的药效均有较强的关联性,其含量可客观反映该制剂的质量优劣。
本研究建立了润鼻通窍滴剂的HPLC指纹图谱方法,同时标定了多批样品中的14个共有峰,并指认出其中的7个成分,即新绿原酸、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、木犀草苷、异绿原酸C。10批润鼻通窍滴剂与对照图谱的相似度均>0.92,表明不同批次润笔通窍滴剂化学成分基本一致,但各成分的含量存在一定差异,可能与药材批次、制备工艺稳定性有关。14个共有峰的相对保留时间RSD为0.056%~0.20%,相对峰面积的RSD为0.48%~8.9%,表明不同批次润鼻通窍滴剂样品间的指纹图谱整体无差异,但色谱峰高或峰面积存在一定差异,可用于润鼻通窍滴剂定性鉴别。利用指纹图谱的色谱分离条件,建立了润鼻通窍滴剂中7个成分进行同步定量分析方法。本研究中润鼻通窍滴剂中新绿原酸的含量最高,可达0.133 8 mg·mL-1,绿原酸和隐绿原酸次之。
本研究以HPLC指纹图谱和多指标的含量测定相结合,通过对多批次润鼻通窍滴剂的定性鉴别和含量测定,为以多成分、多指标的润鼻通窍滴剂的质量标准建立提供了研究线索,并为润鼻通窍滴剂的深入开发奠定了坚实的基础。
