Objective:

To establish a method for the simultaneous determination of six components (rutin, naringin, neohesperidin, quercetin, purpurin and mollugin) in Huazhi tablets and Huazhi capsules by HPLC.

Methods:

The assay was performed on an Agilent ZORBAX SB-Aq (150 mm×4.6 mm, 5 μm) and the sample was eluted by mobile phase consisting of acetonitrile(A) -0.1% phosphoric acid(B) with a gradient at a flow rate of 1.0 mL·min-1 and the column temperature was 30 ℃. The detection wavelengths were set at 250 nm for rutin during 0-20 min, 283 nm for naringin and neohesperidin during 20-37 min, and 250 nm for quercetin, purpurin and mollugin during 37-60 min.

Results:

Rutin, naringin, neohesperidin, quercetin, purpurin and mollugin exhibited good linearity(r>0.999 0) in the ranges of 37.26-1 863.20 μg·mL-1, 11.27-563.70 μg·mL-1, 9.58-479.04 μg·mL-1, 1.92-95.90 μg·mL-1, 0.52-25.83 μg·mL-1 and 0.90-45.10 μg·mL-1, respectively. The average recoveries of the above mentioned six components in Huazhi tablets (n=6) were 103.2%(RSD=1.4%), 99.5%(RSD=1.7%), 97.7%(RSD=1.2%), 95.2%(RSD=1.1%), 104.2% (RSD=1.2%) and 104.2%(RSD=0.80%), respectively, and the average recoveries of the above mentioned six components in Huazhi capsules (n=6) were 102.5% (RSD=1.3%), 96.9%(RSD=0.48%), 97.1%(RSD=1.1%), 96.9%(RSD=0.78%), 102.3%(RSD=1.4%) and 101.8%(RSD=1.2%), respectively. The content ranges (mean ± SD) of rutin, naringin, neohesperidin, quercetin, purpurin and mollugin in the 17 batches of Huazhi tablets and Huazhi capsules were 31.14-98.25(44.33±15.65) mg·g-1, 5.12-20.85(12.96±5.85) mg·g-1, 4.03-17.00(10.14±5.17) mg·g-1, 0.63-7.17(1.97±1.49) mg·g-1, 0.23-1.32(0.57±0.28) mg·g-1 and 0.67-1.72(1.08±0.29) mg·g-1, respectively.

Conclusion:

The established method can simultaneously determine six components in Huazhi tablets and Huazhi capsules and can provide a reference for the quality control of Huazhi tablets and Huazhi capsules.

目的:

建立高效液相色谱波长切换法测定化痔片和化痔胶囊中6个成分(芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素)的含量。

方法:

目的:

建立高效液相色谱波长切换法测定化痔片和化痔胶囊中6个成分(芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素)的含量。

方法:

采用Agilent ZORBAX SB-Aq色谱柱(150 mm×4.6 mm,5 μm),以乙腈(A)-0.1%磷酸(B)为流动相,梯度洗脱,流速1.0 mL·min-1,柱温30 ℃,检测波长250 nm(0~20 min,芦丁)、283 nm(20~37 min,柚皮苷、新橙皮苷)、250 nm(37~60 min,槲皮素、羟基茜草素、大叶茜草素)。

结果:

芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素的质量浓度分别在37.26~1 863.20 μg·mL-1、11.27~563.70 μg·mL-1、9.58~479.04 μg·mL-1、1.92~95.90 μg·mL-1、0.52~25.83 μg·mL-1和0.90~45.10 μg·mL-1与峰面积呈现良好的线性关系(r>0.999 0);化痔片中上述6个成分的平均加样回收率(RSD)(n=6)分别为103.2%(1.4%)、99.5%(1.7%)、97.7%(1.2%)、95.2%(1.1%)、104.2%(1.2%)和104.2%(0.80%);化痔胶囊中上述6个成分的平均加样回收率(RSD)(n=6)分别为102.5%(1.3%)、96.9%(0.48%)、97.1%(1.1%)、96.9%(0.78%)、102.3%(1.4%)和101.8%(1.2%)。17批次化痔片和化痔胶囊中芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素的含量范围(平均值±标准偏差)分别为31.14~98.25(44.33±15.65)mg·g-1、5.12~20.85(12.96±5.85)mg·g-1、4.03~17.00(10.14±5.17)mg·g-1、0.63~7.17(1.97±1.49)mg·g-1、0.23~1.32(0.57±0.28)mg·g-1和0.67~1.72(1.08±0.29)mg·g-1

结论:

本研究所建立的方法可同时测定化痔片和化痔胶囊中6个主要活性成分的含量,可为化痔片及化痔胶囊的质量控制提供参考。

Objective:

To establish a method for the simultaneous determination of six components (rutin, naringin, neohesperidin, quercetin, purpurin and mollugin) in Huazhi tablets and Huazhi capsules by HPLC.

Methods:

The assay was performed on an Agilent ZORBAX SB-Aq (150 mm×4.6 mm, 5 μm) and the sample was eluted by mobile phase consisting of acetonitrile(A) -0.1% phosphoric acid(B) with a gradient at a flow rate of 1.0 mL·min-1 and the column temperature was 30 ℃. The detection wavelengths were set at 250 nm for rutin during 0-20 min, 283 nm for naringin and neohesperidin during 20-37 min, and 250 nm for quercetin, purpurin and mollugin during 37-60 min.

Results:

Rutin, naringin, neohesperidin, quercetin, purpurin and mollugin exhibited good linearity(r>0.999 0) in the ranges of 37.26-1 863.20 μg·mL-1, 11.27-563.70 μg·mL-1, 9.58-479.04 μg·mL-1, 1.92-95.90 μg·mL-1, 0.52-25.83 μg·mL-1 and 0.90-45.10 μg·mL-1, respectively. The average recoveries of the above mentioned six components in Huazhi tablets (n=6) were 103.2%(RSD=1.4%), 99.5%(RSD=1.7%), 97.7%(RSD=1.2%), 95.2%(RSD=1.1%), 104.2% (RSD=1.2%) and 104.2%(RSD=0.80%), respectively, and the average recoveries of the above mentioned six components in Huazhi capsules (n=6) were 102.5% (RSD=1.3%), 96.9%(RSD=0.48%), 97.1%(RSD=1.1%), 96.9%(RSD=0.78%), 102.3%(RSD=1.4%) and 101.8%(RSD=1.2%), respectively. The content ranges (mean ± SD) of rutin, naringin, neohesperidin, quercetin, purpurin and mollugin in the 17 batches of Huazhi tablets and Huazhi capsules were 31.14-98.25(44.33±15.65) mg·g-1, 5.12-20.85(12.96±5.85) mg·g-1, 4.03-17.00(10.14±5.17) mg·g-1, 0.63-7.17(1.97±1.49) mg·g-1, 0.23-1.32(0.57±0.28) mg·g-1 and 0.67-1.72(1.08±0.29) mg·g-1, respectively.

Conclusion:

The established method can simultaneously determine six components in Huazhi tablets and Huazhi capsules and can provide a reference for the quality control of Huazhi tablets and Huazhi capsules.

化痔片及化痔胶囊均收录于国家药品标准中,二者处方相同,均由槐米、茜草、枳壳、三棱、三七5味中药组成,具有清热、凉血、止血、行气散瘀的作用,临床用于内痔、外痔、混合痔、内外痔血栓等痔疮的治疗[1-3]。处方中槐米的主要活性成分是黄酮类化合物,其中芦丁、斛皮素均具备良好的止血作用[4-5];茜草具有凉血,止血,祛瘀等功效,所含大叶茜草素为萘醌类有效成分,羟基茜草素为蒽醌类有效成分[6-9];三七中含皂苷类、黄酮类、氨基酸等多个成分,其中人参皂苷类、三七皂苷类、三七素等成分具有活血、止血、止痛等功效[10-12];枳壳临床上常用于胃下垂、脱肛、子宫脱垂等病症,新橙皮苷、柚皮苷等黄酮类成分是其主要活性成分之一[13-16]。目前有关化痔片和化痔胶囊中组分含量测定的研究较少,马灵珍[17-18]分别建立了化痔胶囊中柚皮苷和新橙皮苷、羟基茜草素和大叶茜草素的含量测定方法。胡晓莲等[19]建立了UPLC法测定化痔片中3个皂苷成分的含量,另有文献报道了芦丁的含量测定方法[20-22]
化痔片及化痔胶囊为组方制剂,处方中的枳壳、茜草,市场上混伪品较多,现行标准含量测定项下仅测定芦丁1个成分,不能很好地控制该制剂的质量。本研究选择处方中槐米、茜草、枳壳的主要活性成分作为检测指标,建立HPLC法同时测定芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素的含量,以期为建全该制剂质量控制方法提供参考。
Agilent 1290高效液相色谱仪,配置二极管阵列检测器(Agilent公司);Agilent ZORBAX SB-Aq(250 mm×4.6 mm,5 μm)色谱柱(Agilent公司);XM-400ES型超声波清洗器(小美超声仪器有限公司);XS205DU十万分之一电子分析天平(梅特勒-托利多公司);Milli-Q超纯水系统(Millipore公司)。
对照品芦丁(批号100080-202012,纯度91.6%)、柚皮苷(批号110722-202116,纯度93.5%)、新橙皮苷(批号111857-202305,纯度99.6%)、槲皮素(批号100081-201610,纯度99.1%)、羟基茜草素(批号111898-202004,纯度98.8%)和大叶茜草素(批号110884-202107,纯度99.5%)均购自中国食品药品检定研究院,乙腈和甲醇(色谱纯,赛默飞世尔公司),水为超纯水(自制),甲醇和磷酸(分析纯,国药集团化学试剂有限公司)。
化痔片和化痔胶囊均为市售样品,12批化痔片来自5个生产厂家(厂家1:批号20200601、20230203、20230404、20230911、20231001;厂家2:批号20230401、20230603、20240101、20240102;厂家3:批号20220824;厂家4:批号221202;厂家5:批号230301,样品编号S1~S12),5批化痔胶囊来自1个生产厂家(批号210112、211030、221005、2304171、2306011,样品编号S13~S17)。
取对照品芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素适量,精密称量,加甲醇制成每1 mL含芦丁0.4 mg、柚皮苷0.1 mg、新橙皮苷0.1 mg、槲皮素20 μg、羟基茜草素5 μg、大叶茜草素10 μg的混合对照品溶液。
取样品粉末(片剂除去包衣,研细;胶囊剂取内容物)各约0.25 g,精密称量,置具塞锥形瓶中,精密加入甲醇25 mL,称量,超声(300 W,40 kHz)提取30 min,放置至室温,再称量,用甲醇补足减失的量,摇匀,过0.22 μm微孔滤膜,取续滤液,即得。
分别按片剂和胶囊剂的处方工艺和比例,分别制成缺槐米、缺茜草、缺枳实的阴性样品,按“2.1.2”项下方法,制备各阴性样品溶液。
采用Agilent ZORBAX SB-Aq(250 mm×4.6 mm,5 μm)色谱柱,以乙腈为流动相A,以0.1%磷酸溶液为流动相B,梯度洗脱(0~30 min,14%A→15%A;30~40 min,15%A→32%A;40~52 min,32%A→62%A;52~55 min,62%A→93%A;55~60 min,93%A),柱温30 ℃,流速1.0 mL·min-1,进样体积5 μL,检测波长为250 nm(0~20 min时,检测芦丁)、283 nm(20~37 min时,检测柚皮苷、新橙皮苷)、250 nm(37~60 min时,检测槲皮素、羟基茜草素、大叶茜草素)。
取混合对照品溶液、供试品溶液、各阴性样品溶液,按“2.2”项下色谱条件进样测定,结果见图1图2。由此可知,供试品溶液、混合对照品溶液中各成分色谱峰保留时间一致,各待测成分峰与相邻峰分离度均>1.5,片剂与胶囊剂阴性样品均无干扰,表明该方法专属性良好。
分别取对照品芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素适量,精密称定,加甲醇制成质量浓度分别为9.316、1.879、1.996、0.959、0.861、0.902 mg·mL-1的单一对照品储备液。分别精密吸取各对照品储备液适量,用甲醇制成质量浓度分别为1 863.20、563.70、479.04、95.90、25.83、45.10 μg·mL-1的混合对照品储备液。精密吸取混合对照品储备液0.2、0.4、1、2、4、6、8、10 mL,分别置于10 mL量瓶中,加甲醇定容,得8个系列浓度溶液,按“2.2”项下色谱条件进样测定。以各成分的峰面积Y为纵坐标,对应的浓度X为横坐标,进行线性回归,结果见表1,可见6个成分在各自浓度范围内线性关系良好。另取混合对照品储备液适量,用甲醇逐级稀释,按“2.2”项下色谱条件测定,以3倍信噪比对应的质量浓度作为检测限,以10倍信噪比对应的质量浓度作为定量限,结果见表1
精密吸取“2.3.2”项的混合对照品储备液,用甲醇稀释5倍,按“2.2”项下色谱条件进样6次,记录峰面积。结果芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素色谱峰面积的RSD(n=6)分别为0.13%、0.20%、0.10%、0.13%、0.74%和0.14%,表明仪器精密度良好。
取化痔片S1和化痔胶囊S13供试品溶液,分别于1、2、4、6、12、24 h进样测定。结果化痔片中芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素峰面积的RSD(n=6)分别为0.46%、1.3%、0.45%、0.58%、0.47%和0.66%;化痔胶囊中6个成分峰面积的RSD(n=6)分别为0.49%、0.47%、1.5%、0.72%、0.33%和0.51%,表明供试品溶液在24 h内稳定性良好。
按照“2.1.2”项下方法分别制备6份化痔片S1供试品溶液和6份化痔胶囊S13供试品溶液,在“2.2”项色谱条件下进样测定。结果化痔片中6个成分芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素的平均含量(n=6)分别为40.69、8.67、8.12、2.27、0.24、0.86 mg·g-1,RSD分别为1.9%、1.6%、1.8%、1.4%、2.0%和0.60%;化痔胶囊中上述6个成分的平均含量(n=6)分别为40.87、7.44、5.71、1.55、0.22、0.79 mg·g-1,RSD分别为1.6%、1.2%、1.1%、1.1%、1.3%和1.4%,表明该方法的重复性良好。
取已知含量的化痔片S1和化痔胶囊S13样品的粉末各6份,每份约0.125 g,精密称定,分别置具塞锥形瓶中,精密加入芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素对照品溶液适量,按照“2.1.2”项下方法制备供试溶液,进样分析,计算回收率,结果见表2
取12批化痔片及5批化痔胶囊样品,按“2.1.2”项下方法制备供试品溶液,在“2.2”项色谱条件下进样测定,以外标法计算芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素的含量,结果见表3
6个成分在甲醇中的溶解性较好,故采用甲醇作为提取溶剂;考察超声和回流2种提取方式的提取效率,结果回流30 min与超声30 min提取效果接近;在超声提取条件下考察溶剂体积(25、50 mL),发现25 mL时各成分基本提取完全。因此采用甲醇溶液25 mL,超声30 min的提取方式,简单又能达到较好的提取效果。
由于所测6个成分为2类不同类型的化合物(组1:芦丁、柚皮苷、新橙皮苷、槲皮素为黄酮类;组2:羟基茜草素、大叶茜草素为蒽醌类),各成分之间紫外最大吸收波长差别较大,需要在不同吸收波长下进行测定。本实验采用DAD检测器采集各成分在190~400 nm波长范围内的光谱图,发现芦丁、槲皮素、羟基茜草素、大叶茜草在250 nm处均有较强的紫外吸收,故选择250 nm作为检测波长同时检测以上4个成分;柚皮苷、新橙皮苷在283 nm处均有较强的紫外吸收,故选择283 nm作为二者的检测波长。
本研究考察了不同比例的乙腈-磷酸水溶液和乙腈-醋酸水溶液作为流动相体系,结果显示采用乙腈-0.1%磷酸水溶液时,待测成分的分离度和峰形较好。由于6个待测成分极性跨度较大,采用等度洗脱无法对所有化合物进行分离,故采用梯度洗脱方式,6个成分在60 min内获得较好的分离。考察了Symmetry Shield AQ-C18色谱柱(250 mm×4.6 mm,5 μm)、Agilent ZORBAX SB-Aq色谱柱(150 mm×4.6 mm,3.5 μm)和YMC Hydrosphere C18色谱柱(250 mm×4.6 mm,5 μm)对各成分色谱峰分离度的影响,结果发现Agilent ZORBAX SB-Aq色谱柱分离效果最好,在该系统下6个成分得到较好的保留和分离。
本研究建立的HPLC法可同时测定化痔片和化痔胶囊中6个主要活性成分的含量,方法简便、准确且重复性好。从含量测定结果来看,6个成分的含量波动范围较大,可能与中药材来源广泛、制药设备与制备工艺的差异等有关。所测芦丁、柚皮苷、新橙皮苷、槲皮素、羟基茜草素和大叶茜草素为处方中槐米、枳壳、茜草3种药材的主要活性成分,能较全面地反映该制剂的质量。同时在一定程度上,可通过这6个成分的含量,排除制剂中易混淆药材枳壳和茜草的投料风险,排除用药安全风险,为化痔片及化痔胶囊的质量控制提供参考。
