Objective:

To establish a quality evaluation method of Erdi Xiaoke mixture based on HPLC fingerprint, multi-component determination and chemometric analysis.

Methods:

The separation was performed on a 30 ℃ thermostatic Acutfex PW-C18 (250 mm×4.6 mm, 5 μm) column, with the mobile phase comprising of acetonitrile -0.1% phosphoric acid flowing at 1.0 mL·min-1 in a gradient elution manner, and the detection wavelength was set at 285 nm to establish HPLC fingerprint of Erdi Xiaoke mixture. Contents of 11 components [5-hydroxymethyl furfural(5-HMF), salvianic acid A, chlorogenic acid, caffeic acid, calycosin-7-glucoside, acteoside, isochlorogenic acid B, isochlorogenic acid C, rosmarinic acid, salvianolic acid B and formononetin] in 12 batches of Erdi Xiaoke mixture were determined, and similarity evaluation, cluster analysis, principal component analysis and orthogonal partial least squares discriminant analysis were used to comprehensively evaluate the quality of Erdi Xiaoke mixture.

Results:

The 11 constituents showed good linear relationships within their respective ranges(r≥0.999 6), with average recovery rates of 95.3%-101.5%, and the RSDs were in the range of 1.1%-2.3%. There were 18 common peaks in the fingerprints for 12 batches of samples with the similarities above 0.960. The range of the content of the above 11 components in the 12 batches of samples were as follows as 43.40-65.70, 219.43-274.70, 119.00-164.78, 11.87-18.17, 42.80-56.86, 10.01-20.71, 8.21-14.73, 40.15-57.39, 43.29-60.29, 755.22-1 208.50, 2.47-4.44 μg·mL-1. And 12 batches of Erdi Xiaoke mixture could be clustered into three categories with some differences in sample quality among different batches. In addition, 5-HMF, isochlorogenic acid B and chlorogenic acid were the main potential markers affecting the quality of Erdi Xiaoke mixture.

Conclusion:

This method is stable, reliable, accurate and sensitive, and can be used to control and comprehensively evaluate the quality of Erdi Xiaoke mixture.

目的:

建立二地消渴合剂的HPLC指纹图谱、多成分含量测定和化学计量学相结合的质量评价方法。

方法:

采用Acutfex PW-C18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈-0.1%磷酸为流动相梯度洗脱,体积流量1.0 mL·min-1,柱温30 ℃,检测波长285 nm,建立二地消渴合剂的HPLC指纹图谱;对12批二地消渴合剂中的11个成分[5-羟甲基糠醛(5-HMF)、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B、芒柄花黄素]进行含量测定,并结合相似度评价、聚类分析,主成分分析及正交偏最小二乘-判别分析对二地消渴合剂质量进行综合评价。

结果:

所测11个成分在各自浓度范围内线性关系良好(r≥0.999 6),平均加样回收率为95.3%~101.5%,RSD为1.1%~2.3%。12批样品的HPLC指纹图谱中共确定18个共有峰,相似度均>0.960。12批样品中上述11个成分的含量的范围依次在43.40~65.70、219.43~274.70、119.00~164.78、11.87~18.17、42.80~56.86、10.01~20.71、8.21~14.73、40.15~57.39、43.29~60.29、755.22~1 208.50、2.47~4.44 μg·mL-1。12批二地消渴合剂可聚为3类,不同批次间质量存在一定差异,5-HMF、丹酚酸B和绿原酸是影响样品质量的主要潜在标志物。

结论:

该方法稳定可靠,准确灵敏,可用于二地消渴合剂的质量控制和综合评价。

目的:

建立二地消渴合剂的HPLC指纹图谱、多成分含量测定和化学计量学相结合的质量评价方法。

方法:

采用Acutfex PW-C18(250 mm×4.6 mm,5 μm)色谱柱,以乙腈-0.1%磷酸为流动相梯度洗脱,体积流量1.0 mL·min-1,柱温30 ℃,检测波长285 nm,建立二地消渴合剂的HPLC指纹图谱;对12批二地消渴合剂中的11个成分[5-羟甲基糠醛(5-HMF)、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B、芒柄花黄素]进行含量测定,并结合相似度评价、聚类分析,主成分分析及正交偏最小二乘-判别分析对二地消渴合剂质量进行综合评价。

结果:

所测11个成分在各自浓度范围内线性关系良好(r≥0.999 6),平均加样回收率为95.3%~101.5%,RSD为1.1%~2.3%。12批样品的HPLC指纹图谱中共确定18个共有峰,相似度均>0.960。12批样品中上述11个成分的含量的范围依次在43.40~65.70、219.43~274.70、119.00~164.78、11.87~18.17、42.80~56.86、10.01~20.71、8.21~14.73、40.15~57.39、43.29~60.29、755.22~1 208.50、2.47~4.44 μg·mL-1。12批二地消渴合剂可聚为3类,不同批次间质量存在一定差异,5-HMF、丹酚酸B和绿原酸是影响样品质量的主要潜在标志物。

结论:

该方法稳定可靠,准确灵敏,可用于二地消渴合剂的质量控制和综合评价。

Objective:

To establish a quality evaluation method of Erdi Xiaoke mixture based on HPLC fingerprint, multi-component determination and chemometric analysis.

Methods:

The separation was performed on a 30 ℃ thermostatic Acutfex PW-C18 (250 mm×4.6 mm, 5 μm) column, with the mobile phase comprising of acetonitrile -0.1% phosphoric acid flowing at 1.0 mL·min-1 in a gradient elution manner, and the detection wavelength was set at 285 nm to establish HPLC fingerprint of Erdi Xiaoke mixture. Contents of 11 components [5-hydroxymethyl furfural(5-HMF), salvianic acid A, chlorogenic acid, caffeic acid, calycosin-7-glucoside, acteoside, isochlorogenic acid B, isochlorogenic acid C, rosmarinic acid, salvianolic acid B and formononetin] in 12 batches of Erdi Xiaoke mixture were determined, and similarity evaluation, cluster analysis, principal component analysis and orthogonal partial least squares discriminant analysis were used to comprehensively evaluate the quality of Erdi Xiaoke mixture.

Results:

The 11 constituents showed good linear relationships within their respective ranges(r≥0.999 6), with average recovery rates of 95.3%-101.5%, and the RSDs were in the range of 1.1%-2.3%. There were 18 common peaks in the fingerprints for 12 batches of samples with the similarities above 0.960. The range of the content of the above 11 components in the 12 batches of samples were as follows as 43.40-65.70, 219.43-274.70, 119.00-164.78, 11.87-18.17, 42.80-56.86, 10.01-20.71, 8.21-14.73, 40.15-57.39, 43.29-60.29, 755.22-1 208.50, 2.47-4.44 μg·mL-1. And 12 batches of Erdi Xiaoke mixture could be clustered into three categories with some differences in sample quality among different batches. In addition, 5-HMF, isochlorogenic acid B and chlorogenic acid were the main potential markers affecting the quality of Erdi Xiaoke mixture.

Conclusion:

This method is stable, reliable, accurate and sensitive, and can be used to control and comprehensively evaluate the quality of Erdi Xiaoke mixture.

糖尿病是影响人类健康的主要代谢性疾病之一,近年来流行病学资料表明我国糖尿病总体患病率呈现出上升的趋势,已达11.2%,其中2型糖尿病(type 2 diabetes mellitus,T2DM)占90%以上[1]。2型糖尿病肾脏病(type 2 diabetic kidney disease,T2DKD)是由T2DM所导致的慢性肾脏病,主要临床表现为持续性的蛋白尿和肾脏功能减退,是导致终末期肾病(end-stage kidney disease,ESKD)和病人死亡的重要原因之一[2-3]。随着糖尿病总体患病率上升,T2DKD患病人数也在逐年增加[4]
二地消渴合剂是由熟地、生地、黄芪、丹参、麸炒白术、五味子、茵陈、天花粉、麦冬、丹参、郁金和山药共12味中药组成的复方制剂,是江苏省中医院内分泌科的临床经验方及代表方,方中熟地性温、滋阴益精,生地性寒、清热凉血,黄芪健脾益气,三者共为君药,山药补益脾脏、益肺生津,麦冬养阴润肺、益胃生津,五味子益气生津、补肾宁心,天花粉清热泻火、生津止渴,茯苓利水渗湿、宁心健脾,炒白术燥湿利水、健脾益气,茵陈清利湿热,全方健脾益气、养阴清热、利水渗湿、活血化瘀。对于T2DKD早期气阴两虚兼内热之基本病机具有显著改善作用,可显著降低T2DKD早期患者蛋白尿水平,改善患者临床症候[5-7]。二地消渴合剂为医疗机构制剂,现有质量标准较为简单,仅包含薄层鉴别等,未收载含量测定项,难以对该制剂进行整体质量评价。本研究采用HPLC法建立二地消渴合剂的指纹图谱并结合化学模式识别进行分析评价,同时测定其中11个化学成分的含量,以弥补二地消渴合剂成分分析研究的空白,为该制剂质量标准的完善和进一步的新药开发提供药学研究基础。
Waters e 2695型高效液相色谱仪配置PDA检测器(沃特世科技有限公司);Acutfex PW-C18色谱柱(250 mm×4.6 mm,5 μm,喆分医药科技有限公司);BP211D型十万分之一分析天平(赛多利斯公司);PL-S100TX数显超声波清洗机(康士洁超声波科技有限公司);Centrifuge 5430 R高速冷冻离心机(艾本德公司)。
对照品:5-羟甲基糠醛(5-HMF)(批号MUST-22102711,含量≥98%)、丹参素(批号MUST-22050920,含量98.38%)、毛蕊花糖苷(批号MUST-22112712,含量99.37%)、异绿原酸B(批号MUST-23012705,含量98.27%)、异绿原酸C(批号MUST-22081001,含量99.77%)、芒柄花黄素(批号MUST-23033005,含量99.03%)均购自成都曼斯特生物科技有限公司;绿原酸(批号110753-201314,含量96.6%)、咖啡酸(批号110885-200102,含量99.7%)均购自中国食品药品检定研究院;毛蕊异黄酮苷(批号J19HB174062,含量≥98%)、迷迭香酸(批号Y06A9K67402,含量≥98%)、丹酚酸B(批号N23HB202048,含量≥98%)均购自上海源叶生物科技有限公司。
乙腈、磷酸均为色谱纯,其余试剂为分析纯,水为超纯水。12批二地消渴合剂(批号20230107、20230109、20230218、20230222、20230311、20230313、20230326、20230403、20230414、20230419、20230510、20230523,编号S1~S12),均由江苏省中医院制剂部提供。熟地、生地、黄芪、丹参、麸炒白术、五味子、茵陈、天花粉、麦冬、丹参、郁金、山药12味中药饮片分购于马鞍山井泉中药饮片公司、贵州同德药业股份有限公司和安徽协和成药业饮片有限公司,经南京中医药大学附属医院钱芳主任中药师鉴定为正品,符合2020年版《中华人民共和国药典》相关要求。
天津喆分Acutfex PW-C18(250 mm×4.6 mm,5 μm)色谱柱,流动相为乙腈(A)-0.1%磷酸(B),梯度洗脱(0~9 min,5%A→10%A;9~13 min,10%A→12%A;13~20 min,12%A→15%A;20~40 min,15%A→25%A;40~53 min,25%A→27%A;53~71 min,27%A→68%A;71~77 min,68%A→80%A;77~82 min,80%A),检测波长285 nm,体积流量1.0 mL·min-1,柱温30 ℃,进样量5 μL。
精密称取对照品5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B和芒柄花黄素分别置于10 mL量瓶中,加适量甲醇溶解,超声(500 W,40 kHz)处理15 min,冷却后用甲醇稀释至刻度,得到质量浓度依次为2.00、1.02、1.13、0.86、1.45、1.08、0.83、1.30、1.10、1.64、1.14 mg·mL-1的各对照品储备液。精密吸取各对照品储备液适量,置于同一10 mL量瓶中,用甲醇定容,摇匀,即得混合对照品溶液(各成分质量浓度分别为120.0、152.4、113.0、25.9、86.8、21.6、25.0、65.1、55.2、492.0、20.7 μg·mL-1),平行制作2份。
精密量取二地消渴合剂2 mL于10 mL量瓶中,加甲醇适量,超声(500 W,40 kHz)处理15 min,冷却后甲醇稀释至刻度,14 000 r·min-1离心10 min,取上清液,即得。
参照二地消渴合剂制备工艺,分别制备缺黄芪的阴性样品,缺丹参的阴性样品,缺茵陈的阴性样品,缺五味子的阴性样品,缺生地和熟地的阴性样品,缺丹参和茵陈的阴性样品,缺五味子、熟地、麦冬和麸炒白术的阴性样品,按“2.2.2”项下方法处理,即得。
取样品S1适量,按“2.2.2”项方法制备供试品溶液,在“2.1”项色谱条件下连续进样6次,记录峰面积。以保留时间适中且分离度较好的毛蕊异黄酮苷峰作为参照峰,结果显示,各共有峰相对保留时间和相对峰面积的RSD分别在0.070%~0.38%和0.79%~2.4%,表明仪器精密度良好。
取样品S1适量,按“2.2.2”项方法制备供试品溶液,分别在室温放置0、2、4、8、16、24 h后,在“2.1”项色谱条件下进样测定,记录峰面积。以毛蕊异黄酮苷峰为参照峰,结果显示,各共有峰相对保留时间和相对峰面积的RSD分别在0.10%~0.29%和0.87%~2.6%,表明供试品溶液在24 h内稳定性良好。
取样品S1,按“2.2.2”项方法平行制备6份供试品溶液,在“2.1”项色谱条件下进样测定,测得5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B、芒柄花黄素含量的平均值分别为44.73、226.54、121.43、13.03、44.41、18.96、11.39、43.10、43.45、756.77、3.52 μg·mL-1。以毛蕊异黄酮苷峰为参照峰,各共有峰相对保留时间和相对峰面积的RSD分别在0.12%~0.19%和0.86%~2.4%,表明该方法重复性良好。
12批样品按照“2.2.2”项方法制备供试品溶液,在“2.1”项色谱条件下进样测定,得到样品色谱图,将色谱图依次导入《中药色谱特征图谱相似度评价系统》(2012版)软件进行分析,以样品S1的图谱作为参照图谱,采用平均数法,设定时间窗宽度为0.1 min,经多点校正后以Mark峰匹配生成对照指纹图谱(R)及HPLC特征指纹图谱。结果12批样品共标定18个共有峰,见图1。记录相似度评价结果,可知12批样品与对照指纹图谱的相似度分别为0.997、0.960、0.994、0.991、0.997、0.998、0.995、0.997、0.977、0.982、0.998、0.993,均>0.960,表明各批样品物质基础组成较为稳定。
将生成的对照谱图(R)与混合对照品及阴性样品溶液进行比对,共指认出11个色谱峰,分别为5-HMF(1号峰)、丹参素(2号峰)、绿原酸(5号峰)、咖啡酸(6号峰)、毛蕊异黄酮苷(8号峰)、毛蕊花糖苷(9号峰)、异绿原酸B(10号峰)、异绿原酸C(12号峰)、迷迭香酸(13号峰)、丹酚酸B(15号峰)、芒柄花黄素(18号峰)。其中,8、18号峰归属于黄芪,2号峰、13~17号峰归属于丹参,3号峰、5号峰、7号峰、10~12号峰归属于茵陈,4号峰归属于五味子,9号峰为生地和熟地共有成分,6号峰为丹参和茵陈共有成分,1号峰为五味子、熟地、麦冬和麸炒白术共有成分,见图2
使用SPSS 25.0软件对12批二地消渴合剂进行统计分析,将18个共有峰峰面积导入到软件,以平方欧氏距离作为测距,采用组间连接法对其进行聚类分析(cluster analysis,CA)。结果显示当平方欧氏距离为5时,12批样品可分为3类,其中S4、S10和S11为1类,S8和S9为1类,其余为1类,见图3。表明在相似度较高的情况下,不同批次间的样品成分含量具有一定差异性。
为进一步比较二地消渴合剂不同批次间的质量差异,将12批二地消渴合剂样品的18个共有峰峰面积导入到SPSS 25.0进行主成分分析(principal component analysis,PCA),提取特征值>1的成分为主成分,得到4个主成分,结果见表1。4个成分的累计方差贡献率为94.42%,能较好地代表样品中的大部分信息。运用SIMCA 14.0分析软件绘制主成分分析得分图,见图4。从得分图可以看出,12批样品散点分布可分为3类,其中S4、S10和S11为1类,S8和S9为1类,其余为1类,与CA结果一致。
为进一步比较12批二地消渴合剂化学成分的差异和寻找各批次含量之间的差异性标志化合物,将12批二地消渴合剂样品的18个共有峰峰面积导入到SIMCA 14.1软件中进行正交偏最小二乘-判别分析(partial least squares discrimination analysis,OPLS-DA),以变量重要性投影(variable importance in projection,VIP)>1.0为标准进行筛选,见图5。结果显示有3个成分VIP>1,按照从大到小的顺序依次为1号峰(5-HMF)、15号峰(丹酚酸B)、5号峰(绿原酸),表明这3个成分是二地消渴合剂不同批次间质量差异贡献较大的标志物,可作为其质量控制的重点关注对象。
取混合对照品溶液、供试品溶液(S1)及各阴性样品溶液,在“2.1”项色谱条件下进样测定。结果显示供试品溶液中各成分色谱峰分离度良好,保留时间与相应对照品一致,阴性无干扰,见图2,表明该方法专属性良好。
精密量取“2.2.1”项下的混合对照品溶液0.2、0.4、1.0、1.6、2.0、4.0、6.0 mL,分别置于10 mL量瓶中,甲醇稀释至刻度,在“2.1”项色谱条件下进样测定,记录峰面积。以质量浓度X为横坐标,对应色谱峰面积Y为纵坐标进行线性回归,计算回归方程,结果见表2,可知各成分在各自浓度范围内线性关系良好。
取样品S1适量,按“2.2.2”项方法制备供试品溶液,在“2.1”项色谱条件下连续进样测定6次,测得5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B和芒柄花黄素峰面积的RSD分别为1.6%、1.7%、1.1%、1.5%、1.3%、1.5%、1.0%、1.7%、1.2%、1.3%、0.94%,表明仪器精密度良好。
取样品S1适量,按“2.2.2”项方法制备供试品溶液,分别在室温放置0、2、4、8、16、24 h后,在“2.1”项色谱条件下进样测定,测得5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B和芒柄花黄素峰面积的RSD分别为0.79%、1.4%、1.3%、0.78%、2.0%、1.8%、1.8%、1.4%、1.3%、1.9%、1.7%,表明溶液在24 h内稳定性良好。
取样品S1适量,按“2.2.2”项方法平行制备6份供试品溶液,在“2.1”项色谱条件下进样测定,测得5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B和芒柄花黄素含量的平均值分别为44.46、227.31、121.93、12.93、44.62、18.52、11.90、42.89、43.39、756.67、3.34 μg·mL-1,RSD分别为2.9%、1.8%、1.9%、0.63%、1.9%、1.8%、1.8%、1.0%、1.5%、1.3%、1.9%,表明该方法重复性良好。
精密吸取已知各成分含量的样品S1 2.5 mL,加入混合对照品溶液(5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B和芒柄花黄素质量浓度分别为12.00、15.24、11.30、2.59、8.68、2.10、2.50、6.51、5.52、49.20、2.07 μg·mL-1)2.5 mL,甲醇定容至25 mL量瓶,按“2.2.2”项下方法平行制备6份供试溶液,在“2.1”项色谱条件下进样测定,计算回收率。结果,5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B和芒柄花黄素平均加样回收率分别为101.4%、97.8%、99.3%、101.5%、98.5%、96.3%、96.9%、95.3%、96.0%、95.4%、97.8%,RSD分别为2.2%、1.6%、2.3%、1.5%、2.0%、1.8%、1.1%、2.2%、1.3%、1.9%、2.1%。
取12批样品,每批样品按“2.2.2”项下方法平行制备3份供试品溶液,在“2.1”项色谱条件下进样测定,计算上述11个成分含量,结果见表3
二地消渴合剂方中熟地、生地、黄芪为君药,山药、麦冬、五味子、天花粉共为臣药,茯苓、炒白术、茵陈为佐药,丹参、郁金为使药。现代药理研究表明,黄芪中的黄酮类成分(如毛蕊异黄酮苷、芒柄花黄素)能够改善血管内皮细胞损伤,而血管内皮细胞损伤是糖尿病-肾病患者血管病变发展中的关键环节,提示该类成分可能是治疗糖尿病-肾病的潜在物质[8-9];苯乙醇苷类成分如毛蕊花糖苷是地黄中的主要成分之一[10],具有广泛的药理活性,如抗炎、抗氧化、抗细胞凋亡以及肾功能保护作用[11-12];茵陈中主要含有的机酸类成分(如绿原酸、咖啡酸、异绿原酸C、异绿原酸B),具有明显的抗菌消炎作用且水提液中含量较高[13];丹参中的丹酚酸类成分含量较高(如丹酚酸B、丹参素、迷迭香酸)且水溶性较好[14],能够显著抑制糖尿病-肾病模型小鼠的机体氧化应激反应和肾脏纤维化,减轻肾间系膜增生和肾小管间质损伤,起到改善糖尿病-肾病的作用[15-16];5-HMF在二地消渴合剂中含量较高,同时存在于熟地、五味子等多味药中,具有抗氧化[17]、降血糖[18]等生物活性。基于成分的含量和活性研究考虑,本研究选择5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B、芒柄花黄素共11个成分为二地消渴合剂的指标性成分。地黄中梓醇和地黄苷D在紫外区为末端吸收,难以检测,故未选作指标成分。
课题组前期使用PDA检测器在210~400 nm范围内对各成分进行波长扫描,异绿原酸B、异绿原酸C、绿原酸和咖啡酸在326 nm,5-HMF在285 nm,丹酚酸B在287 nm,丹参素在280 nm,迷迭香酸和毛蕊花糖苷在329 nm,毛蕊异黄酮苷和芒柄花黄素在250 nm处有最大吸收,通过对比不同检测波长下的谱图,结果显示在285 nm处11个成分均有较大吸收,基线平稳且杂质干扰峰较少,故作为检测波长。
考察了不同流动相体系(乙腈-0.1%磷酸、甲醇-0.1%磷酸、乙腈-0.1%甲酸)对色谱检测的影响,结果显示,有机相采用乙腈时,相较于甲醇,出峰数量多且色谱图基线较为平稳;水相采用含磷酸溶液时,相较于含甲酸溶液时,丹酚酸B、迷迭香酸、异绿原酸C和异绿原酸B几个有效成分的分离度更好。
二地消渴合剂中结构相似的成分较多,部分成分难以分离。本试验考察了不同色谱柱(喆分Acutfex PW-C18、Agilent Eclipse XDB-C18、Waters sunfire-C18)的分离效果,发现选用喆分Acutfex PW-C18色谱柱时峰形尖锐且对称性好,保留时间相近的迷迭香酸和异绿原酸C色谱峰可以得到有效分离。
指纹图谱相似度评价结果均>0.960,表明各批次样品物质基础组成较稳定。化学计量学分析将12批样品聚为3类,PCA得分图与CA结果一致,其中PCA得到的4个主成分可代表样品的大部分信息,OPLS-DA得出5-HMF、丹酚酸B和绿原酸可能是区分二地消渴合剂的差异性质量标志物,可加强对这3个关键成分的考察。11个指标成分含量测定结果的RSD在6.3%~20.3%,表明各批次间二地消渴合剂的质量存在一定的差异性,可能与原饮片产地加工、制剂的制备工艺、存储时间等因素有关[19],有待深入考察。
本研究建立了二地消渴合剂的HPLC指纹图谱,并对其中的5-HMF、丹参素、绿原酸、咖啡酸、毛蕊异黄酮苷、毛蕊花糖苷、异绿原酸B、异绿原酸C、迷迭香酸、丹酚酸B和芒柄花黄素共11个成分进行了含量测定方法研究,该方法准确度高且重复性强,可用于二地消渴合剂的质量控制。结合化学计量学分析,通过指纹图谱相似度评价、CA、PCA及OPLS-DA综合评价了不同批次二地消渴合剂的成分特征、含量差异及差异来源,可据此对二地消渴合剂指纹图谱共有模式进行长期稳定性考察,完善其质量标准,推动制剂制备工艺的“均一化”,从而保证临床用药的有效性和安全性。
