Objective:

To identify the risks and provide a scientific basis for the formulation of the limit standards by the determination and risk assessment of pesticide residues in Yixintong tablets and hawthorn leaf extracts.

Methods:

The samples were extracted with acetonitrile by high speed homogenater, and purified by solid - phase extraction(SPE) column of envi-carb/NH2. 262 pesticides in hawthorn leaf extracts and Yixintong tablets were detected by GC-MS/MS and LC-MS/MS. Moreover, the acute and chronic intake risks were calculated using the deterministic risk assessment model.

Results:

25 and 31 types of pesticides were detected in hawthorn leaf extracts and Yixintong tablets, respectively. 18 types of pesticides were detected in all of them. The contents were 0.004-1.457 mg·kg-1, 0.007-2.1 mg·kg-1, respectively. The results of risk assessment revealed that the short-term hazard index(HIa) of hexaconazole was 1 and the risk was needed to be paid more attention. HIa and long-term hazard index(HIc) of other pesticides detected in Yixintong tablets and hawthorn leaf extracts were much lower than 1, indicating an acceptable risk.

Conclusion:

In the present study, the pesticide residues were explored, and their health risks were assessed;therefore our study provided technical support for the improvement of risk assessment applicable to traditional Chinese medicines (TCMs) and the formulation of relevant pesticide residue limit standards.

目的:

通过对益心酮片及其原料山楂叶提取物的农药残留情况进行测定及风险评估,明确相关品种的风险,并为相关限量标准的制定提供科学依据。

方法:

以乙睛为溶剂,高速匀浆提取,经氨基石墨化碳小柱净化。GC-MS/MS和LC-MS/MS方法对山楂叶和山楂叶提取物中262种农药进行检测。并采用确定性评估模式,分别计算其短期和长期摄入风险。

结果:

山楂叶提取物和益心酮片中分别检出农药25种和31种,检出共有农药18种,残留量范围分别为0.004~1.457 mg·kg-1和0.007~2.1 mg·kg-1。风险评估结果表明,除了己唑醇的短期危害指数(HIa)为1,风险需要进一步被关注外,益心酮片及其原料山楂叶提取物中其他检出农药的HIa和长期危害指数(HIc)均低于1,风险可接受。

结论:

本研究探索中药制剂益心酮片及其原料中农药残留情况,并评估其风险,为完善符合中药使用特点的风险评估技术体系及相关农药残留限量标准的制订提供技术支持。

目的:

通过对益心酮片及其原料山楂叶提取物的农药残留情况进行测定及风险评估,明确相关品种的风险,并为相关限量标准的制定提供科学依据。

方法:

以乙睛为溶剂,高速匀浆提取,经氨基石墨化碳小柱净化。GC-MS/MS和LC-MS/MS方法对山楂叶和山楂叶提取物中262种农药进行检测。并采用确定性评估模式,分别计算其短期和长期摄入风险。

结果:

山楂叶提取物和益心酮片中分别检出农药25种和31种,检出共有农药18种,残留量范围分别为0.004~1.457 mg·kg-1和0.007~2.1 mg·kg-1。风险评估结果表明,除了己唑醇的短期危害指数(HIa)为1,风险需要进一步被关注外,益心酮片及其原料山楂叶提取物中其他检出农药的HIa和长期危害指数(HIc)均低于1,风险可接受。

结论:

本研究探索中药制剂益心酮片及其原料中农药残留情况,并评估其风险,为完善符合中药使用特点的风险评估技术体系及相关农药残留限量标准的制订提供技术支持。

Objective:

To identify the risks and provide a scientific basis for the formulation of the limit standards by the determination and risk assessment of pesticide residues in Yixintong tablets and hawthorn leaf extracts.

Methods:

The samples were extracted with acetonitrile by high speed homogenater, and purified by solid - phase extraction(SPE) column of envi-carb/NH2. 262 pesticides in hawthorn leaf extracts and Yixintong tablets were detected by GC-MS/MS and LC-MS/MS. Moreover, the acute and chronic intake risks were calculated using the deterministic risk assessment model.

Results:

25 and 31 types of pesticides were detected in hawthorn leaf extracts and Yixintong tablets, respectively. 18 types of pesticides were detected in all of them. The contents were 0.004-1.457 mg·kg-1, 0.007-2.1 mg·kg-1, respectively. The results of risk assessment revealed that the short-term hazard index(HIa) of hexaconazole was 1 and the risk was needed to be paid more attention. HIa and long-term hazard index(HIc) of other pesticides detected in Yixintong tablets and hawthorn leaf extracts were much lower than 1, indicating an acceptable risk.

Conclusion:

In the present study, the pesticide residues were explored, and their health risks were assessed;therefore our study provided technical support for the improvement of risk assessment applicable to traditional Chinese medicines (TCMs) and the formulation of relevant pesticide residue limit standards.

山楂叶为蔷薇科山楂属植物山里红(Crataegus pinnatifida Bge. var. major N. E. Br.)或山楂(Crataegus pinnatifida Bge.)的叶,我国东北、华北、广东、广西等地均有种植,药用资源十分丰富。早在东晋就有关于山楂叶“茎叶煮汁,洗漆疮”的记载,历来为药食两用之佳品[1-3]。益心酮片是以山楂叶提取物为原料而制成的成方制剂,收录于2020年版《中华人民共和国药典》,具有活血化瘀、理气通脉的功效[4]。山楂叶在种植过程中,由于其病虫害高发,其防治以使用化学农药为主。对山楂叶提取物及其制剂农药残留情况进行测定并进行风险评估可为中药农药监管和农药留限量标准制、修订提供科学合理的线索。
本研究从全国范围内抽取益心酮片样品16批次,此外,为了考察其原料农药残留情况,收集了6批山楂叶提取物。选择262种农药作为检测目标,分别对益心酮片和山楂叶提取物的农药残留检测结果进行分析。并采用确定性评估模式探索性地对样品中农药的短期和长期摄入暴露量分别进行计算。通过分析和风险评估,明确中药制剂益心酮片及其原料中农药残留情况,确定应重点关注的农药种类,以期为中药中风险评估体系的完善及相关残留限量标准的制、修订提供有益参考。
益心酮片样品来源于2020年国家评价性抽验样品,收集于24个省市,从中随机抽取16批次,涵盖全国范围内益心酮片的全部生产企业,其剂型包括薄膜衣片和糖衣片2种。此外,通过药材市场、药店等渠道收集了6批山楂叶提取物样品。
精密称取样品粉末5 g,加入氯化钠1 g,置100 mL试管中,精密加入乙腈50 mL,匀浆处理3 min,离心(12 000 r·min-1)3 min,分取上清液,沉淀再加乙腈45 mL,匀浆2 min,离心(12 000 r·min-1)3 min,合并2次提取的上清液,50 ℃减压浓缩至3~5 mL,用乙睛转移并定容至10 mL量瓶中,摇匀,取4 mL用氨基石墨化碳小柱净化,洗脱液为含1%醋酸的乙腈-甲苯(3∶1)溶液25 mL。收集洗脱液,浓缩至约2 mL,用乙腈定容至10 mL,即得。
采用GC-MS/MS弹性石英毛细管柱(30 m×0.25 mm,0.25 μm) DB17 ms,载气为高纯氨气,柱流速1.3 mL·min-l,进样口温度240 ℃,高压不分流进样。升温程序为初始温度60 ℃,保持1 min;以30 ℃·min-1升至120 ℃;以10 ℃·min-1升至160 ℃;以2 ℃·min-1升至230 ℃;以15 ℃·min-1升至300 ℃,保持6 min;以20 ℃·min-1升至320 ℃,保持3 min。EI测定条件:70 eV,离子源温度200 ℃,接口温度250 ℃。
采用CORTECSTM UPLC Cis(150 mm×2.1 mm,1.6 μm)色谱柱,以0.1%甲酸水(含5 mmol·L-1甲酸铵)为流动相A,以95%乙腈水溶液(含5 mmol·L-1甲酸铵、0.1%甲酸)为流动相B,梯度洗脱(0~0.2 min,70%A;0.2~10 min,70%A→0%A;10~12 min,0%A;12~13 min,0%A→70%A;13~17 min,70%A),流速0.3 mL·min-1,柱温40 ℃,进样量1 μL。电喷雾离子源(ESI),正离子扫描模式;监测模式为多反应监测(MRM);毛细管电压2.50 kV;离子源温度150 ℃;去溶剂温度550 ℃;锥孔气150 L·h-1;去溶剂气900 L·h-l;气帘气138 kPa;碰撞气48 kPa。
此方法共有262种农药指标(总计271个化学单体),在1~100 mg·L-1浓度范围内线性关系良好,相关系数(r)>0.999;重复进样6次,精密度RSD<5.0%;且多数指标灵敏度达到或低于0.01 mg·kg-1;在0.01、0.05、0.1 g·L-1 3个水平进行回收率及重复性考察,得出此方法大部分的农药回收率在70%~120%,RSD≤15%,符合痕量多残留分析技术要求。
参照课题组前期研究制定的《中药中外源性有害残留物安全风险评估技术指导原则》[6]分别对益心酮片及其原料山楂叶提取物中检出的农药残留对人体健康的风险进行评估。农药的健康指导值即每日允许摄入量(ADI)和急性参考剂量(ARfD)从WHO官网农药数据库获得[7]。分别按公式Exp=(EF×Ed×IR×C)/(AT×W)计算其短期暴露和长期暴露情况下样品中农药残留的暴露量(Exp,μg·kg-1),公式中EF为暴露频率,Ed为中药一生的暴露年限,IR为样品的日服用剂量(g),C为样品中的农药残留量(mg·kg-1);W为人体体质量,一般按60 kg计。
对于短期暴露量的计算,EF×Ed/AT=1;IR为最大日服用剂量;C为样品中农药残留量的最大值。对于长期暴露量的计算,IR为平均日服用剂量;C为样品中农药残留量的平均值;根据国家食品安全风险评估中心在全国11个省份范围内20 917名调查者的有效消费调查问卷,EF的P 95分位值为每年90 d,Ed为20 a[8],AT为平均寿命天数=365×70=25 550 d;分别根据2020年版《中华人民共和国药典》对于益心酮片和山楂叶提取物项下日用量的规定代入Exp=(EF×Ed×IR×C)/(AT×W)计算其短期暴露量(Expa,μg·kg-1)和长期暴露量(Expc,μg·kg-1)。
短期危害指数(HIa)的计算公式为HIa=(Expa×100)/ARfD,长期危害指数(HIc)的计算公式为HIc=(Expc×100)/ADI,公式中100为安全因子,表示每日由中药材及其制品中摄取的农药残留量不大于日总暴露量(包括食物和饮用水)的1%。若危害指数HI≤1,样品中农药的健康风险较低;若HI>1,风险应予以关注。
采用LC-MS/MS和GC-MS/MS法检测益心酮片中262种农药残留的结果如表1所示,山楂叶提取物检测结果如表2所示。
综合表12结果,山楂叶提取物及其制剂益心酮片中均检出的共有农药24种,包括多菌灵、噻虫嗪、吡虫啉、啶虫脒、多效唑、腈菌唑、戊唑醇、甲基硫菌灵、氟硅唑、除虫脲、氟环唑、己唑醇、百克敏、噻嗪酮、毒死蜱、哒螨灵、克菌丹、三唑酮、三唑醇A、三唑醇B、噻呋酰胺、丙环唑、氟氯氰菊酯、氯氰菊酯、氰戊菊酯、苯醚甲环唑。从检出农药的种类来分析,山楂叶提取物中检出农药25种,益心酮片中检出农药31种,制剂益心酮片中检出农药比山楂叶提取物中检出的农药种类增加了6种。其中,除甲氰菊酯外,16批益心酮片中30种农药的检出率在20%以上。其中多菌灵、吡虫啉、啶虫脒、烯酰吗啉、腈菌唑、戊唑醇、氟硅唑、己唑醇、三唑醇a、三唑醇b、噻呋酰胺、丙环唑、苯醚甲环唑、嘧菌酯的检出率高达100%。2020年版《中华人民共和国药典》未对山楂叶提取物及益心酮片中农药残留作出限量规定。按照《GB 2763-2019食品安全国家标准食品中农药最大残留限量》标准中对于部分药用植物的限量标准,6批益心酮片中多菌灵、3批益心酮片中丙环唑以及2批山楂叶提取物中丙环唑不符合要求。因此,不能忽视制剂中的农药残留量,对于制剂中农药残留的风险评估很有必要。
益心酮片及其原料山楂叶提取物中短期和长期风险评估结果如表34所示。从表中结果可见,除了己唑醇的HIa为1,风险需要进一步被关注外,益心酮片及其原料山楂叶提取物中其他检出的农药的HIa和HIc均远低于1,风险可接受。
本研究对16批益心酮片及其原料山楂叶提取物样品中的262种农药的残留水平进行测定,从检出农药种类来分析,山楂叶提取物中检出农药25种,益心酮片中检出农药31种。不能忽视制剂中的农药残留量。国际上对于污染物(包括农药残留)常用的风险评估方法包括WHO推荐的危害指数法(HI法)以及US EPA推荐的靶标危害系数法(THQ)和致癌风险法(CR)等[9-14]。本研究采用HI法(包括短期危害指数法和长期危害指数法)分别对益心酮片及其原料山楂叶提取物中检出的农药残留量进行评估。风险评估结果显示,益心酮片及其原料山楂叶提取物中农药检出率较高,但大多处于痕量残留水平,总体安全风险可控。需要进一步加强对于己唑醇的监测,关注其短期风险。HI法是国际通用的风险评估方法,也是近年来兴起的在中药外源性有害残留物风险评估方面使用最广泛的方法[15-16]。本研究结合课题组前期的调查研究结果,将人类平均寿命、中药使用频率、暴露年限等参数系统地应用于益心酮片集原料中农药残留的风险评估,形成的数据对风险管理者提供有益的参考,并为完善符合中药使用特点的风险评估技术体系及相关农药残留限量标准的制修订提供技术支持。
