Objective:

To evaluate the differences of aqueous extract protein content and kinds among Hudilong origins in order to provide reference for the identification and quality evaluation.

Methods:

The protein content of aqueous extract was determined, and the differences among the origins were compared by nonparametric tests. The protein fingerprints were established by SDS-PAGE, and the band matrix was constructed based on the molecular weight of the bands. The differences of the protein kinds among the 3 origins were compared and analyzed by CA and OPLS-DA.

Results:

There was no statistically significant difference in the aqueous extract protein content between the P. vulgaris and P. guillelmi, but both of them differed significantly from that of P. pectinifera (P<0.01). Aqueous extract protein content. from P. pectinifera was higher than from P. vulgaris or P. guillelmi. The results of SDS-PAGE showed that there were differences in the molecular masses and quantities of the protein bands of the 3 origins with relative molecular masses ranging from 10×103 to 250×103. The results of the CA and OPLS-DA showed that each of the origin of Hudilong could be differentiated. The differences of the protein relative molecular masses were 133.7×103, 10.4×103, 66.9×103, 51.9×103, 13.4×103, 7×103, 29.4×103, 14.6×103, 62.2×103, 81.1×103, 89.2×103, 7.7×103, and 101.7×103, respectively.

Conclusion:

There are differences in the aqueous extract protein among 3 origins of Hudilong. The P. vulgaris and P. guillelmi have high similarity in the aqueous extract protein components content and kinds. SDS-PAGE technique could be used to provide reference for the identification and quality evaluation.

目的:

对沪地龙3个基原间水溶性蛋白含量和种类的差异进行评价研究,为沪地龙的鉴定和质量评价提供参考。

方法:

测定沪地龙水溶性蛋白含量,应用非参数检验方法比较基原间含量差异;使用SDS-PAGE方法建立沪地龙水溶性蛋白指纹图谱,依据条带相对分子质量构建“条带矩阵”,应用聚类分析、正交偏最小二乘判别分析对3个基原间蛋白种类差异进行比较分析。

结果:

通俗环毛蚓、威廉环毛蚓水溶性蛋白含量无统计学差异,但二者均与栉盲环毛蚓存在显著性差异(P<0.01),栉盲环毛蚓水溶性蛋白含量高于二者。SDS-PAGE蛋白电泳显示沪地龙3个基原在相对分子质量10×103~250×103间蛋白谱带在相对分子质量和数量间存在差异;聚类分析、正交偏最小二乘判别分析结果表明,沪地龙各基原能明显区分,共有13个差异性蛋白成分,根据VIP大小差异相对分子质量依次为133.7×103、10.4×103、66.9×103、51.9×103、13.4×103、7×103、29.4×103、14.6×103、62.2×103、81.1×103、89.2×103、7.7×103、101.7×103

结论:

沪地龙基原间水溶性蛋白成分存在差异,通俗环毛蚓和威廉环毛蚓在蛋白含量和种类方面相似度高。SDS-PAGE方法可为沪地龙鉴定和质量评价提供参考。

测定沪地龙水溶性蛋白含量,应用非参数检验方法比较基原间含量差异;使用SDS-PAGE方法建立沪地龙水溶性蛋白指纹图谱,依据条带相对分子质量构建“条带矩阵”,应用聚类分析、正交偏最小二乘判别分析对3个基原间蛋白种类差异进行比较分析。

结果:

通俗环毛蚓、威廉环毛蚓水溶性蛋白含量无统计学差异,但二者均与栉盲环毛蚓存在显著性差异(P<0.01),栉盲环毛蚓水溶性蛋白含量高于二者。SDS-PAGE蛋白电泳显示沪地龙3个基原在相对分子质量10×103~250×103间蛋白谱带在相对分子质量和数量间存在差异;聚类分析、正交偏最小二乘判别分析结果表明,沪地龙各基原能明显区分,共有13个差异性蛋白成分,根据VIP大小差异相对分子质量依次为133.7×103、10.4×103、66.9×103、51.9×103、13.4×103、7×103、29.4×103、14.6×103、62.2×103、81.1×103、89.2×103、7.7×103、101.7×103

结论:

沪地龙基原间水溶性蛋白成分存在差异,通俗环毛蚓和威廉环毛蚓在蛋白含量和种类方面相似度高。SDS-PAGE方法可为沪地龙鉴定和质量评价提供参考。

Objective:

To evaluate the differences of aqueous extract protein content and kinds among Hudilong origins in order to provide reference for the identification and quality evaluation.

Methods:

The protein content of aqueous extract was determined, and the differences among the origins were compared by nonparametric tests. The protein fingerprints were established by SDS-PAGE, and the band matrix was constructed based on the molecular weight of the bands. The differences of the protein kinds among the 3 origins were compared and analyzed by CA and OPLS-DA.

Results:

There was no statistically significant difference in the aqueous extract protein content between the P. vulgaris and P. guillelmi, but both of them differed significantly from that of P. pectinifera (P<0.01). Aqueous extract protein content. from P. pectinifera was higher than from P. vulgaris or P. guillelmi. The results of SDS-PAGE showed that there were differences in the molecular masses and quantities of the protein bands of the 3 origins with relative molecular masses ranging from 10×103 to 250×103. The results of the CA and OPLS-DA showed that each of the origin of Hudilong could be differentiated. The differences of the protein relative molecular masses were 133.7×103, 10.4×103, 66.9×103, 51.9×103, 13.4×103, 7×103, 29.4×103, 14.6×103, 62.2×103, 81.1×103, 89.2×103, 7.7×103, and 101.7×103, respectively.

Conclusion:

There are differences in the aqueous extract protein among 3 origins of Hudilong. The P. vulgaris and P. guillelmi have high similarity in the aqueous extract protein components content and kinds. SDS-PAGE technique could be used to provide reference for the identification and quality evaluation.

地龙为钜蚓科动物参环毛蚓Pheretima aspergillum(E.Perrier)、通俗环毛蚓P. vulgaris Chen、威廉环毛蚓P. guillelmi(Michaelsen)或栉盲环毛蚓P. pectinifera Michaelsen的干燥体,前一种习称广地龙,后三者习称沪地龙[1]。地龙的蛋白类成分占干重的50%以上,所含种类丰富如抗微生物蛋白、钙调素结合蛋白、脂类蛋白、蚓激酶、纤溶酶、胶原酶、抗菌肽等,具有抗脑卒中、抗凝血、抗纤维化、抗菌等药理活性[2]。蛋白质作为动物重要的初生代谢产物之一,在生长、发育、代谢、能量转换、信号传导等过程中发挥关键作用,也具有重要的药理活性,如角蛋白[3]、水蛭蛋白[4]等。对地龙蛋白的研究多见于蛋白组分和特征多肽的研究[5-6],而对于地龙药材基原间蛋白差异研究报道较少[7]。沪地龙3个基原在生存环境、分布地域较为相近,其药材相似度高,传统鉴定方法难以区分[8],在临床与研究不作基原区分的现象时有发生[9-10],而且混用不利于质量稳定[11]。蛋白质大分子在物种体内具有稳定的含量和物种特异性,其差异在一定程度上能反映样品基原间、地域性、不同部位和加工方法的差异。
十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate-polyacrylamide gel electrophoresis,SDS-PAGE)是常用的蛋白质研究方法,将样品进行适当处理后,在适宜的电泳条件下对样品进行分析,结果以样品蛋白条带位置与标准相对分子质量蛋白位置进行比较,还可根据蛋白条带颜色深浅和条带宽度定量比较蛋白含量差异[3]。可用于中药的真伪鉴别[12-13]、特征蛋白研究[14]、炮制前后蛋白变化[15]、不同产地蛋白差异[16-17]等方面。本研究首先对沪地龙3个基原样品的水溶性蛋白含量进行测定,并对基原间含量差异进行比较;再应用SDS-PAGE方法获得沪地龙样品水溶性蛋白图谱,通过建立条带矩阵并应用聚类分析和正交偏最小二乘判别分析对沪地龙3个基原进行蛋白差异分析。
3020型酶标仪(赛默飞公司);DYY-6C型电泳仪电源,DYCZ-28C型电泳槽(北京六一仪器厂);ChemiScope6100型化学发光成像系统(上海勤翔公司);ME204E/02型万分之一电子分析天平(上海梅特勒-托利多公司);ZC-100S型超声波清洗机(常州恩培公司);HH-6型数显恒温水浴锅(上海邦西公司)。
BCA蛋白测定试剂盒,南京碧云天公司;15%Precast-GLgel Tris-Glycine电泳预制胶15孔,10×Tris-Glycine电泳缓冲液,双色10×103~250×103 Da TureColor预染蛋白Marker,考马斯亮蓝R-250,乙酸,十二烷基硫酸钠(SDS),β-巯基乙醇,三羟甲基氨基甲烷(Tris),均购自上海生工生物有限公司。
本实验所用沪地龙样品共计40批次,其中通俗环毛蚓16批次,威廉环毛蚓12批次,栉盲环毛蚓12批次。样品经海军军医大学张汉明教授鉴定,药材来源见表1
药材粉碎至粗粉,取1.0 g,精密称定,加入0.9%氯化钠溶液30 mL浸泡30 min,超声(300 W,40 kHz)提取30 min,温度控制在50 ℃以下,提取液8层纱布滤过,取续滤液经3 000 r·min-1离心5 min,取上清液,即得。
5×Loading Buffer:1 mol·L-1 Tris-HCL(pH 6.8)0.6 mL,50%甘油2.5 mL,10%SDS 2 mL,β-巯基乙醇0.5 mL,1%溴酚蓝1 mL,加水定容至10 mL。
脱色液:95%乙醇500 mL,冰乙酸160 mL,加水定容至2 000 mL。
染色液:考马斯亮蓝R-250 0.60 g,加脱色液定容至500 mL。
将BCA蛋白测定试剂盒(碧云天)中标准蛋白液用水进行梯度稀释,以蛋白浓度(X,μg·mL-1)为横坐标,以吸收度Y为纵坐标,得线性回归方程:
蛋白浓度在0~500 μg·mL-1与吸光度呈良好的线性关系。
先对样品进行蛋白浓度预测定,再根据结果将样品浓度调整到0~500 μg·mL-1范围内。16批通俗环毛蚓蛋白含量为44.19~106.08 mg·g-1,平均含量为68.54 mg·g-1;12批威廉环毛蚓蛋白含量为40.81~96.50 mg·g-1,平均含量为59.08 mg·g-1;12批栉盲环毛蚓蛋白含量为38.43~147.04 mg·g-1,平均含量为99.43 mg·g-1,见表2
根据“2.4”项检测结果将样品蛋白浓度稀释到1 μg·μL-1,取待测样品400 μL,5×Loading Buffer 100 μL置于1.5 mL离心管中,混匀后100 ℃水浴5 min,放置室温后5 000 r·min-1离心3 min。取上清液20 μL、marker 5 μL进行凝胶电泳。电泳条件采用恒压模式180 V,待指示剂距玻璃板下缘约1 cm时停止电泳,取出电泳胶。采用考马斯亮蓝染色法进行染色,经脱色后得蛋白电泳胶片,使用勤翔ChemiScope 6100凝胶成像系统拍照。对电泳图谱以从上至下顺序进行条带计数。结果通俗环毛蚓最多可得到18条条带(图1,1-B),威廉环毛蚓最多可得16条条带(图1,2-B),栉盲环毛蚓最多可得18条条带(图1,3-B)。
以各图谱的Marker比移值(Rf)为横坐标,相对分子质量的对数(lgM)为纵坐标建立标准曲线,得1-A标准曲线方程、2-A标准曲线方程和3-A标准曲线方程:
将蛋白条带比移值代入标准曲线方程,得到其相对分子质量。以蛋白谱带相对分子质量为参考,按从大到小的顺序排列,样品含有该条带记为“1”,不含有该条带记为“0”,建立条带矩阵(表3)。
应用SPSS 26.0对结果进行了方差齐性检验和正态分布检验,结果显著性均<0.05,即不满足ANOVA分析条件,因此选择非参数独立样本检验方法。应用Original Pro(2024版)得到非参数独立样本克鲁斯卡尔-沃利斯检验(K-W)检验结果P=0.021<0.05,表明3个基原间存在差异,应用Bonferroni法进行两两比较,结果通俗环毛蚓与栉盲环毛蚓间,威廉环毛蚓与栉盲环毛蚓间P均<0.01,即通俗环毛蚓与威廉环毛蚓间水溶性蛋白含量差异无统计学意义,通俗环毛蚓、威廉环毛蚓与栉盲环毛蚓间水溶性蛋白含量均存在显著性差异(图2)。
应用NTSYSpc2.10e软件对所得“条带矩阵”数据进行聚类分析。沪地龙3个基原中栉盲环毛蚓单独聚为1支,通俗环毛蚓和威廉环毛蚓聚为1支,而后二者又各单独聚为1支(除样品MV-HQYY-3和MV-KQYY-2),从整体上看,3个基原的蛋白谱带具有种内相似性和种间差异性,且种间差异大于种内差异(图3)。
先将沪地龙3个基原所得的“条带矩阵”数据导入软件SIMCA14.1,建立OPLS-DA分析总体模型Ml,以筛选3个基原间的差异性蛋白质(VIP>1);然后以2个基原为1组建立OPLS-DA分析模型M2(MV+MG)、M3(MV+AP)和M4(MG+AP),筛选基原间的差异性蛋白质。各模型的R2XR2YQ2均>0.67,见表4
通过200次置换检验对4个模型进行评价,均无过拟合现象,说明模型稳定且具有良好的解释及预测能力(图4)。在M1模型得分图中,沪地龙各基原在95%置信区间内,区分明显,单独聚集成簇(图5,1-A);在M2~M4模型得分图中,各基原分别位于2侧,说明不同基原间差异明显,可被区分(图5,2~4-A)。
以VIP>1为提取差异蛋白标准,沪地龙3个基原间差异蛋白相对分子质量依次为133.7×103、10.4×103、66.9×103、51.9×103、13.4×103、7×103、29.4×103、14.6×103、62.2×103、81.1×103、89.2×103、7.7×103、101.7×103图4-10,1-B);通俗环毛蚓与威廉环毛蚓间主要差异蛋白相对分子质量为133.7×103、10.4×103、66.9×103、89.2×103、101.7×103、11.8×103、8.8×103、7.7×103、16.4×103、35.4×103、27.1×103、24.1×103图4-10,2-B);通俗环毛蚓与栉盲环毛蚓间主要差异蛋白相对分子质量为51.9×103、13.4×103、7×103、29.4×103、14.6×103、62.2×103、81.1×103、21.7×103、7.7×103、10.4×103、66.9×103图4-10,3-B);栉盲环毛蚓与威廉环毛蚓间主要差异蛋白相对分子质量为133.7×103、51.9×103、13.4×103、7.7×103、7×103、101.7×103、29.4×103、14.6×103、89.2×103、62.2×103、81.1×103图5,4-B)。
蛋白质是生命组成和发挥功能的重要物质,地龙的蛋白含量可达一半以上,其含量多少在一定程度上能反映药材质量的高低。从对沪地龙3个基原水溶性蛋白含量结果看,各基原内样品批次间蛋白含量RSD在25.6%~32.4%,最高和最低相差可达2~3倍,推测其差异主要来自产地、加工方式、储存条件等因素的影响。对3个基原间蛋白含量统计分析结果表明,通俗环毛蚓与威廉环毛蚓在总体含量上不存在统计学差异,但二者均与栉盲环毛蚓存在显著性差异,而蛋白含量以栉盲环毛蚓最高。
SDS-PAGE图谱中蛋白条带的位置与其相对分子质量相关,与其蛋白构象无关,其颜色深浅与蛋白含量成正比。由图4-6可知,各基原可见5条明显主条带,颜色较深所占总蛋白含量比例较高,位于Marker的150×103、40×103、20×103、15×103、10×103附近。3个基原在相对分子质量20×103以下条带数目大多为5条条带,但条带位置存在明显差异,可作为鉴别依据;此外栉盲环毛蚓在20×103~35×103大多可见4条条带,70×103~100×103大多可见3条条带,此处与另外2个基原有明显差异,可作为区分栉盲环毛蚓的标志区。PLS-DA也结果表明,根据条带位置或相对分子质量可对3个基原进行有效区分。CA结果中通俗环毛蚓和威廉环毛蚓聚为1支,栉盲环毛蚓单独聚为1支,说明通俗环毛蚓与威廉环毛蚓在蛋白种类上较为相似,但二者均与栉盲环毛蚓存在差异,这与蛋白含量结果具有一致性。由上述可知,沪地龙3个基原SDS-PAGE图谱中条带位置与数量存在基原差异,而SDS-PAGE方法特征性强、归类准确,可对沪地龙进行鉴定。
本研究对地龙水溶性成分中的蛋白类成分进行了含量测定和SDS-PAGE方电泳分析,从分析结果来看,沪地龙3个基原中通俗环毛蚓和威廉环毛蚓蛋白含量方面不存在统计学差异,但二者均与栉盲环毛蚓存在显著性差异。水溶性蛋白种类方面,通俗环毛蚓和威廉环毛蚓间蛋白种类相似度高于栉盲环毛蚓。从本研究可知,沪地龙3个基原间水溶性蛋白成分存在差异,可为后续沪地龙研究和临床应用提供参考。
