Objective:

To establish an HPLC method for the determination of potentially genotoxic impurity E, impurity I, and 2-chloromethyl-4-methoxy-3,5-dimethlpyridine in esomeprazole sodium.

Methods:

The chromatographic conditions were as follows: impurity E, YMC-Triart C18 column (250 mm×4.6 mm, 5 μm), mobile phase A 0.05 mol·L-1 monopotassium phosphate buffer, mobile phase B was acetonitrile, the gradient elution program was used at the flow rate of 1.0 mL·min-1, the detection wavelength was 302 nm and the column temperature was 30 ℃. Impurities I, Agilent Microspher C18 column (100 mm×4.6 mm, 3 μm), mobile phase A was water-phosphate buffer (pH 7.6)-acetonitrile(80∶10∶10), mobile phase B was acetonitrile-phosphate buffer (pH 7.6)-water (80∶1∶19), the flow rate was 1.0 mL·min-1, the detection wavelength was 302 nm and the column temperature was 30 ℃. 2-chloromethyl-4-methoxy-3,5-dimethlpyridine, GL Inertsil ODS-3 column (250 mm×4.6 mm, 5 μm), mobile phase A was 0.01 mol·L-1 disodium phosphate solution (pH 6.5), mobile phase B was acetonitrile, the flow rate was 1.0 mL·min-1, the detection wavelength was 265 nm and the column temperature was 30 ℃.

Results:

The linear ranges of impurity E, impurity I and 2-chloromethyl-4-methoxy-3,5-dimethlpyridine were 0.025 1-0.200 7, 0.020 2-0.302 7, 0.126 6-2.110 0 μg·mL-1. The LOQ of impurity E, impurity I, 2-chloromethyl-4-methoxy-3,5-dimethlpyridine were 0.50, 0.40, 2.53 ng, and the LOD were 0.15, 0.12, 0.84 ng. The average recovery rate ranged from 96% to 104%, and the RSD was less than 2%. No potential toxic impurities were detected in the samples.

Conclusion:

The method has the advantages of good repeatability, high precision, high accuracy and good linearity, and the analysis method are simple and efficient.

建立HPLC法测定艾司奥美拉唑钠中潜在基因毒性杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶。

方法:

杂质E色谱条件:YMC-Triart C18色谱柱(250 mm×4.6 mm,5 μm),流动相A为0.05 mol·L-1磷酸二氢钾缓冲液,流动相B为乙腈,梯度洗脱,流速为1.0 mL·min-1,检测波长为302 nm,柱温为30 ℃;杂质I色谱条件:Agilent Microspher C18色谱柱(100 mm×4.6 mm,3 μm),流动相A为水-磷酸盐缓冲液(pH 7.6)-乙腈(80∶10∶10),流动相B为乙腈-磷酸盐缓冲液(pH 7.6)-水(80∶1∶19),梯度洗脱,流速为1.0 mL·min-1,检测波长为302 nm,柱温为30 ℃;2-氯甲基-3,5二甲基-4-甲氧基吡啶色谱条件:GL Inertsil ODS-3色谱柱(250 mm×4.6 mm,5 μm),流动相A为0.01 mol·L-1磷酸氢二钠溶液(pH 6.5),流动相B为乙腈,流速为1.0 mL·min-1,检测波长为265 nm,柱温为30 ℃。

结果:

杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶的线性范围分别为0.025 1~0.200 7、0.020 2~0.302 7、0.126 6~2.110 0 μg·mL-1,定量限分别为0.50、0.40、2.53 ng,检测限分别为0.15、0.12、0.84 ng。平均回收率都在96%~104%,RSD均<2%。样品中均未检出潜在毒性杂质。

结论:

该方法检测杂质的重现性好,精密度高,准确度高,线性良好,分析方法简便高效。

目的:

建立HPLC法测定艾司奥美拉唑钠中潜在基因毒性杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶。

方法:

杂质E色谱条件:YMC-Triart C18色谱柱(250 mm×4.6 mm,5 μm),流动相A为0.05 mol·L-1磷酸二氢钾缓冲液,流动相B为乙腈,梯度洗脱,流速为1.0 mL·min-1,检测波长为302 nm,柱温为30 ℃;杂质I色谱条件:Agilent Microspher C18色谱柱(100 mm×4.6 mm,3 μm),流动相A为水-磷酸盐缓冲液(pH 7.6)-乙腈(80∶10∶10),流动相B为乙腈-磷酸盐缓冲液(pH 7.6)-水(80∶1∶19),梯度洗脱,流速为1.0 mL·min-1,检测波长为302 nm,柱温为30 ℃;2-氯甲基-3,5二甲基-4-甲氧基吡啶色谱条件:GL Inertsil ODS-3色谱柱(250 mm×4.6 mm,5 μm),流动相A为0.01 mol·L-1磷酸氢二钠溶液(pH 6.5),流动相B为乙腈,流速为1.0 mL·min-1,检测波长为265 nm,柱温为30 ℃。

结果:

杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶的线性范围分别为0.025 1~0.200 7、0.020 2~0.302 7、0.126 6~2.110 0 μg·mL-1,定量限分别为0.50、0.40、2.53 ng,检测限分别为0.15、0.12、0.84 ng。平均回收率都在96%~104%,RSD均<2%。样品中均未检出潜在毒性杂质。

结论:

该方法检测杂质的重现性好,精密度高,准确度高,线性良好,分析方法简便高效。

Objective:

To establish an HPLC method for the determination of potentially genotoxic impurity E, impurity I, and 2-chloromethyl-4-methoxy-3,5-dimethlpyridine in esomeprazole sodium.

Methods:

The chromatographic conditions were as follows: impurity E, YMC-Triart C18 column (250 mm×4.6 mm, 5 μm), mobile phase A 0.05 mol·L-1 monopotassium phosphate buffer, mobile phase B was acetonitrile, the gradient elution program was used at the flow rate of 1.0 mL·min-1, the detection wavelength was 302 nm and the column temperature was 30 ℃. Impurities I, Agilent Microspher C18 column (100 mm×4.6 mm, 3 μm), mobile phase A was water-phosphate buffer (pH 7.6)-acetonitrile(80∶10∶10), mobile phase B was acetonitrile-phosphate buffer (pH 7.6)-water (80∶1∶19), the flow rate was 1.0 mL·min-1, the detection wavelength was 302 nm and the column temperature was 30 ℃. 2-chloromethyl-4-methoxy-3,5-dimethlpyridine, GL Inertsil ODS-3 column (250 mm×4.6 mm, 5 μm), mobile phase A was 0.01 mol·L-1 disodium phosphate solution (pH 6.5), mobile phase B was acetonitrile, the flow rate was 1.0 mL·min-1, the detection wavelength was 265 nm and the column temperature was 30 ℃.

Results:

The linear ranges of impurity E, impurity I and 2-chloromethyl-4-methoxy-3,5-dimethlpyridine were 0.025 1-0.200 7, 0.020 2-0.302 7, 0.126 6-2.110 0 μg·mL-1. The LOQ of impurity E, impurity I, 2-chloromethyl-4-methoxy-3,5-dimethlpyridine were 0.50, 0.40, 2.53 ng, and the LOD were 0.15, 0.12, 0.84 ng. The average recovery rate ranged from 96% to 104%, and the RSD was less than 2%. No potential toxic impurities were detected in the samples.

Conclusion:

The method has the advantages of good repeatability, high precision, high accuracy and good linearity, and the analysis method are simple and efficient.

艾司奥美拉唑钠属于抑制胃酸分泌药,是胃壁细胞中质子泵的特异抑制剂[1-4],在胃壁细胞泌酸微管中的高酸条件下可以转化为活性模式,抑制该部位的质子泵活动,对于基础胃酸和受刺激后胃酸的分泌都有抑制作用。可以用于治疗急性上消化道出血[5-7],急性消化性溃疡出血[8],改善重症急性胰腺炎患者肠黏膜屏障功能[9]等疾病。
杂质是指在药品生产、运输、贮藏等过程中产生或引入的影响药物纯度的物质。基因毒性杂质(genotoxic impurities,GTIs)是指能引起遗传毒性的杂质[10-11],极微量水平即能诱发DNA突变,具有重大的遗传毒性安全风险,需要重点控制[12]。根据艾司奥美拉唑钠的制备工艺,分析了其杂质来源,主要有工艺杂质和降解杂质。艾司奥美拉唑钠有关物质的HPLC检测方法已经收载于2020年版《中华人民共和国药典》(以下简称《中国药典》),包括一部分杂质的质量标准[13],但并未收载杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶(基因毒性杂质)的检测方法。参考ICH(M7)中毒理学关注阈值(TCC,1.5 μg·d-1)作为基因毒性杂质的可接受限度[14],按照注射用艾司奥美拉唑钠日最大服用剂量40 mg计算,则计算限度为37.5 μg·g-1。为了实现对这几个基因毒性杂质的质量控制,利用HPLC法进行测定。
李锐等[15]采用质谱法对艾司奥美拉唑钠基因毒性杂质杂质E、杂质I进行检查,王璐等[16]采用质谱法对艾司奥美拉唑钠中基因毒性杂质2-氯甲基-3,5二甲基-4-甲氧基吡啶进行检查。本文采用更加简便的方法、更普及的检测设备进行3个基因毒性杂质的检测。
Agilent 1260高效液相色谱仪(Agilent公司);CPA225D电子天平(精度0.001 g,Sartorius公司);AUW220电子天平(精度0.1 mg,岛津公司);YMC Triart C18色谱柱(YMC公司)、Agilent Microspher C18色谱柱(Agilent公司);GL Inertsil ODS-3色谱柱(GL公司);Waters Symmetry C18色谱柱(Waters公司)。
样品:艾司奥美拉唑钠原料(批号170601、170701、170702、C037-220006,四川海思科制药有限公司);乙腈(色谱纯,Merck公司);杂质E对照品(批号ESE20130129,纯度98.57%,上海诺星医药科技有限公司);杂质I对照品(批号95.16.08.04,纯度99.10%,LGC公司);2-氯甲基-3,5二甲基-4-甲氧基吡啶盐酸盐对照品(批号02322CEV,纯度99.7%,Sigma公司);磷酸二氢钾、氢氧化钾、磷酸二氢钠及磷酸氢二钠(分析纯,国药集团化学试剂有限公司)。
采用YMC-Triart C18色谱柱(250 mm×4.6 mm,5 μm),流动相A为0.05 mol·L-1磷酸二氢钾缓冲液(1.25 g·mL-1氢氧化钾溶液调pH 7.6),流动相B为乙腈,梯度洗脱(0~5 min,85%A;5~20 min,85%A→70%A;20~30 min,70%A;30~31 min,70%A→50%A;31~36 min,50%A;36~37 min,50%A→85%A;37~50 min,85%A),流速为1.0 mL·min-1,检测波长为302 nm,进样量为20 μL,柱温为30 ℃。
采用Agilent Microspher C18色谱柱(100 mm×4.6 mm,3 μm),流动相A为水-磷酸盐缓冲液(pH 7.6)(每1 000 mL中含0.005 2 mol磷酸二氢钠与0.032 mol磷酸氢二钠)-乙腈(80∶10∶10),流动相B为乙腈-磷酸盐缓冲液(pH 7.6)-水(80∶1∶19),梯度洗脱(0~10 min,100%A→80%A;10~30 min,80%A→0%A;30~31 min,0%A→100%A;31~45 min,100%A),流速为1.0 mL·min-1;检测波长为302 nm,进样量为20 μL,柱温为30 ℃。
采用GL Inertsil ODS-3色谱柱(250 mm×4.6 mm,5 μm),流动相A为0.01 mol·L-1磷酸氢二钠溶液(pH 6.5),流动相B为乙腈,梯度洗脱(0~20 min,60%A;20~21 min,60%A→20%A;21~35 min,20%A;35~36 min,20%A→60%A;36~45 min,60%A),流速为1.0 mL·min-1,检测波长为265 nm,进样量为20 μL,柱温为30 ℃。
取样品适量,精密称定,加50%乙腈溶液(稀释剂1)溶解并定量稀释制成每1 mL中约含2.5 mg的艾司奥美拉唑钠溶液,摇匀,作为杂质E供试品溶液。
精密称取杂质E对照品适量,用稀释剂1溶解并定量稀释制成每1 mL中约含1 μg的溶液,摇匀,作为杂质E对照品储备液,精密量取1.00 mL,置10 mL量瓶中,用稀释剂1稀释至刻度,摇匀,作为杂质E对照品溶液(0.1 μg·mL-1)。
精密称取样品25.00 mg,置10 mL量瓶中,加入杂质E对照品储备液1 mL,用稀释剂1溶解并稀释至刻度,摇匀,作为杂质E系统适用性溶液。
取样品适量,精密称定,用流动相A[水-磷酸盐缓冲液(pH 7.6)(每1 000 mL中含0.005 2 mol磷酸二氢钠与0.032 mol磷酸氢二钠)-乙腈(80∶10∶10)](稀释剂2)溶解并定量稀释制成每1 mL中约含5 mg的艾司奥美拉唑钠溶液,摇匀,作为杂质I供试品溶液。
另精密称取杂质I对照品适量,用稀释剂2溶解并定量稀释制成每1 mL中约含2 μg的溶液,摇匀,作为杂质I对照品储备液,精密量取1.00 mL,置10 mL量瓶中,用稀释剂2稀释至刻度,摇匀,作为杂质I对照品溶液(0.2 μg·mL-1)。
精密称取样品50.00 mg,置10 mL量瓶中,加入杂质I对照品储备液1 mL,用稀释剂2溶解并稀释至刻度,摇匀,作为杂质I系统适用性溶液。
取样品适量,精密称定,用40%乙腈溶液(稀释剂3)溶解并定量稀释制成每l mL中约含25 mg的艾司奥美拉唑钠溶液,摇匀,作为2-氯甲基-3,5二甲基-4-甲氧基吡啶供试品溶液。
精密称取2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品适量,用稀释剂3溶解并定量稀释制成每1 mL中含2-氯甲基-3,5二甲基-4-甲氧基吡啶10 μg的溶液,摇匀,作为2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品储备液,精密量取1.00 mL,置10 mL量瓶中,用稀释剂3稀释至刻度,摇匀,作为2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品溶液(1 μg·mL-1)。
精密称取样品250.00 mg,置10 mL量瓶中,加入上述对照品储备液1 mL,用稀释剂3溶解并稀释至刻度,摇匀,作为2-氯甲基-3,5二甲基-4-甲氧基吡啶系统适用性溶液。
取稀释剂1~3,“2.2”项制备的3个成分的对照品溶液、供试品溶液和系统适用性溶液,分别按照拟定的色谱条件进行杂质定位。结果这3个杂质均能够很好地分离,系统适用性溶液样品中其他杂质均不干扰检测,色谱条件的专属性良好(图1)。
取杂质E对照品适量,精密称定,分别用稀释剂1溶解并制成质量浓度为0.025、0.05、0.08、0.10、0.15、0.20 μg·mL-1的系列浓度杂质E对照品溶液;取杂质I对照品适量,精密称定,分别用稀释剂2溶解并制成质量浓度为0.02、0.04、0.10、0.15、0.20、0.30 μg·mL-1的系列浓度杂质I对照品溶液;取2-氯甲基-3,5二甲基-4-甲氧基吡啶的对照品适量,精密称定,分别用稀释剂3溶解并制成质量浓度为0.12、0.20、0.40、0.80、1.0、2.0 μg·mL-1的系列浓度2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品溶液。按照“2.1”项的色谱条件分别进行测定,以溶液质量浓度C(μg·mL-1)为横坐标,相对应的峰面积A为纵坐标分别作图,进行线性回归,求得回归方程(表1)。
分别取杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶的对照品溶液并逐步稀释进样,信噪比约3∶1的浓度作为检测限,以信噪比约10∶1的浓度作为定量限。结果见表1。各色谱条件均能有效满足杂质检出的要求。
进样精密度:取3个杂质的对照品溶液,每种溶液分别连续进样6针,记录色谱图。杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶峰面积的RSD(n=6)分别为1.1%、2.3%、0.060%,表明进样精密度良好。
重复性试验:精密称取样品25.00 mg,置10 mL量瓶中,加入杂质E对照品储备液1 mL,用稀释剂1溶解并稀释至刻度,摇匀,平行制备6份,分别进样,记录色谱图,杂质E重复性试验回收率在99.0%~101.2%,平均回收率为100.1%,RSD(n=6)为0.98%;精密称取样品50.00 mg,置10 mL量瓶中,加入杂质I对照品储备液1 mL,用稀释剂2溶解并稀释至刻度,摇匀,平行制备6份,分别进样,记录色谱图,杂质I重复性试验回收率在100.0%~103.2%,平均回收率为102.0%,RSD(n=6)为1.2%;精密称取样品250.00 mg,置10 mL量瓶中,加入2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品储备液1 mL,用稀释剂3溶解并稀释至刻度,摇匀,平行制备6份,分别进样,记录色谱图,2-氯甲基-3,5二甲基-4-甲氧基吡啶重复性试验回收率在97.0%~99.2%,平均回收率为97.6%,RSD(n=6)为0.98%。结果表明测定方法的重复性均良好。
中间精密度试验:更换不同试验人员,按重复性试验方式进行测定,记录色谱图。杂质E中间精密度试验回收率(n=6)在99.5%~101.8%,平均回收率为100.5%,RSD(n=6)为0.97%;杂质I中间精密度试验回收率(n=6)在100.5%~103.7%,平均回收率为101.9%,RSD(n=6)为1.4%;2-氯甲基-3,5二甲基-4-甲氧基吡啶中间精密度试验回收率(n=6)在96.5%~100.8%,平均回收率为98.0%,RSD(n=6)为1.6%。结果表明中间精密度均良好。
取“2.2”项下配制的各对照品溶液和各供试品溶液,按照“2.1”项下方法进样,测定3个杂质的峰面积以考察溶液稳定性。
杂质E对照品及供试品溶液分别于0、2.5、10、14 h进样,结果14 h内杂质E对照品溶液峰面积RSD(n=4)为1.2%,供试品溶液14 h内均未检出杂质E。表明杂质E对照品溶液与供试品溶液14 h内稳定性良好。
杂质I对照品及供试品溶液分别于0、2、4、8、12 h进样,结果12 h内杂质I对照品溶液峰面积RSD(n=5)为1.2%,供试品溶液12 h内均未检出杂质I。表明杂质I对照品溶液与供试品溶液12 h内稳定性良好。
2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品及供试品溶液分别于0、2、4、6、8 h进样,结果8 h内2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品溶液峰面积RSD(n=5)为0.15%,供试品溶液8 h内均未检出2-氯甲基-3,5二甲基-4-甲氧基吡啶。表明2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品溶液与供试品溶液8 h内稳定性良好。
取杂质E对照品适量,精密称定,用稀释剂1制成每1 mL中含杂质约1 μg的杂质E对照品溶液,再取样品9份,按50%、100%、150%限度浓度加入杂质E对照品溶液,进行杂质回收率试验;取杂质I对照品适量,精密称定,用稀释剂2制成每1 mL中含杂质约2 μg的杂质I对照品溶液,再取样品9份,按50%、100%、150%限度浓度加入杂质I对照品溶液,进行杂质回收率试验;取2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品适量,精密称定,用稀释剂3制成每1 mL中含杂质约10 μg的2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品溶液,再取样品9份,按50%、100%、150%限度浓度加入2-氯甲基-3,5二甲基-4-甲氧基吡啶对照品溶液,进行杂质回收率试验。结果见表2
采用3个色谱条件下系统适用性溶液的配制方式制备供试品溶液,以“2.1”项的色谱条件为基础,进行部分调整(表3),用以考察色谱条件微小变化下杂质的回收率情况。色谱条件部分微调后,杂质E回收率均在98.8%~100.2%,杂质I回收率均在100.8%~103.2%,2-氯甲基-3,5二甲基-4-甲氧基吡啶回收率均在97.2%~98.4%,表明耐用性良好。
按照“2.1”色谱条件,对多批样品(批号170601、170701、070702、C037-220006)进行测定,记录色谱图,结果多批样品中均未检出杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶。
本研究通过对流动相组成、pH、洗脱方式及色谱柱的筛选来确定杂质检测的色谱条件,在确定检测杂质E的色谱条件时,探索了3种色谱条件,但以Waters Symmetry C18为色谱柱,水或者0.02 mol·L-1磷酸二氢钾缓冲液为流动相A时不能很好地分离杂质峰和样品峰,故而选择YMC-Triart C18色谱柱,0.05 mol·L-1磷酸二氢钾缓冲液作为流动相A,可以实现样品与杂质E的良好分离,且稀释剂1与样品中其他杂质不干扰杂质E的测定。在确定检测2-氯甲基-3,5二甲基-4-甲氧基吡啶的色谱条件时,发现梯度洗脱比等度洗脱相能更好分离样品与杂质,稀释剂3及样品中其他杂质不干扰2-氯甲基-3,5二甲基-4-甲氧基吡啶的测定。
在选择测定波长时,取杂质对照品溶液采用高效液相色谱仪(HPLC-DAD)在190~400 nm波长处进行测定,记录色谱图,通过紫外光谱确定各个杂质的吸收波长,发现杂质E和杂质I在302 nm处有较大吸收,2-氯甲基-3,5二甲基-4-甲氧基吡啶在265 nm处有较大吸收,因此将这几个波长作为相应杂质的检测波长。
本研究采用高效液相色谱法,对杂质E、杂质I、2-氯甲基-3,5二甲基-4-甲氧基吡啶进行检测,为了确定方法的可靠性,进行了专属性试验、线性考察、中间精密度试验、检测限和定量限确定、准确度试验和耐用性试验,该方法具有重现性好、精密度高、准确度高、线性良好等优点,并且比质谱法操作更加简单,可以用于药品的杂质检测和质量控制,保证药品质量安全。
