Objective:

To study the preparation and quality control of 3,5-dihydroxy-7,4’-dimethoxyflavone candidate chemical reference substance from Zhuang medicine Amomum paratsaoko S. Q. Tong & Y. M. Xia.

Methods:

3,5-dihydroxy-7,4’-dimethoxyflavone was separated and purified consecutively by silica gel, recrystallization as well as preparative HPLC. The structure of 3,5-dihydroxy-7,4’-dimethoxyflavone was identified by IR, UV, NMR and other comprehensive spectrum analytical methods with MS spectrum. Its purity was determined by HPLC and TLC, the ash content was determined by incandescent residue method, and the content of 3,5-dihydroxy-7,4’-dimethoxyflavone was calculated by mass balance method. The method for quality analysis of 3,5-dihydroxy-7,4’-dimethoxyflavone candidate reference materials was investigated by HPLC.

Results:

The mean value determined by HPLC of 3,5-dihydroxy-7,4’-dimethoxyflavone was 98.85%, the ash value was 0.02%, and the content of 3,5-dihydroxy-7,4’-dimethoxyflavone calculated by mass balance method was 99.83%. The established analytical method was specific and precise.

Conclusion:

3,5-Dihydroxy-7,4’-dimethoxyflavone which was prepared in this study met the quality standard requirements of chemical reference substance, and could be used as a reference substance for quality control of medicinal materials and their preparations such as Amomum paratsaoko S. Q. Tong & Y. M. Xia and others. The analysis method was accurate and reliable, which provided a scientific basis for the research and formulation of quality standard of this reference substance.

目的:

从壮药拟草果中制备纯化3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品并建立其质量分析方法。

方法:

采用硅胶柱色谱、重结晶和高效制备液相色谱等,从拟草果中制备纯化得到高纯度的3,5-二羟基-7,4’-二甲氧基黄酮,通过红外、紫外、核磁共振等综合波谱解析方法及质谱确定其化学结构,采用薄层色谱法、高效液相色谱法检测色谱纯度,炽灼残渣法测定灰分,质量平衡法计算3,5-二羟基-7,4’-二甲氧基黄酮的含量。采用高效液相色谱法对建立的3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品质量分析方法进行方法学考察。

结果:

3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品用高效液相色谱法面积归一化法测定的含量均值为99.85%,炽灼残渣法测得灰分值为0.02%,以质量平衡法计算的含量为99.83%。建立的含量测定方法专属性强,精密度良好。

结论:

制备得到的3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品符合化学对照品的质量标准要求,可作为拟草果及其他含有此成分的药材与制剂质量控制的对照品。建立的质量分析方法准确、可靠,可为该对照品质量标准研究与制定提供科学依据。

目的:

从壮药拟草果中制备纯化3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品并建立其质量分析方法。

方法:

采用硅胶柱色谱、重结晶和高效制备液相色谱等,从拟草果中制备纯化得到高纯度的3,5-二羟基-7,4’-二甲氧基黄酮,通过红外、紫外、核磁共振等综合波谱解析方法及质谱确定其化学结构,采用薄层色谱法、高效液相色谱法检测色谱纯度,炽灼残渣法测定灰分,质量平衡法计算3,5-二羟基-7,4’-二甲氧基黄酮的含量。采用高效液相色谱法对建立的3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品质量分析方法进行方法学考察。

结果:

3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品用高效液相色谱法面积归一化法测定的含量均值为99.85%,炽灼残渣法测得灰分值为0.02%,以质量平衡法计算的含量为99.83%。建立的含量测定方法专属性强,精密度良好。

结论:

制备得到的3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品符合化学对照品的质量标准要求,可作为拟草果及其他含有此成分的药材与制剂质量控制的对照品。建立的质量分析方法准确、可靠,可为该对照品质量标准研究与制定提供科学依据。

Objective:

To study the preparation and quality control of 3,5-dihydroxy-7,4’-dimethoxyflavone candidate chemical reference substance from Zhuang medicine Amomum paratsaoko S. Q. Tong & Y. M. Xia.

Methods:

3,5-dihydroxy-7,4’-dimethoxyflavone was separated and purified consecutively by silica gel, recrystallization as well as preparative HPLC. The structure of 3,5-dihydroxy-7,4’-dimethoxyflavone was identified by IR, UV, NMR and other comprehensive spectrum analytical methods with MS spectrum. Its purity was determined by HPLC and TLC, the ash content was determined by incandescent residue method, and the content of 3,5-dihydroxy-7,4’-dimethoxyflavone was calculated by mass balance method. The method for quality analysis of 3,5-dihydroxy-7,4’-dimethoxyflavone candidate reference materials was investigated by HPLC.

Results:

The mean value determined by HPLC of 3,5-dihydroxy-7,4’-dimethoxyflavone was 98.85%, the ash value was 0.02%, and the content of 3,5-dihydroxy-7,4’-dimethoxyflavone calculated by mass balance method was 99.83%. The established analytical method was specific and precise.

Conclusion:

3,5-Dihydroxy-7,4’-dimethoxyflavone which was prepared in this study met the quality standard requirements of chemical reference substance, and could be used as a reference substance for quality control of medicinal materials and their preparations such as Amomum paratsaoko S. Q. Tong & Y. M. Xia and others. The analysis method was accurate and reliable, which provided a scientific basis for the research and formulation of quality standard of this reference substance.

化学对照品是中药质量标准研究、质量检测和质量控制的实物对照。中药化学对照品的研究,是中药标准化、现代化研究的一个非常重要的部分,对于产品的质量评价,特别是在药品生产的质量控制中,中药化学对照品起着重要的作用,是中药质量控制的基础与核心。目前,部分中药药材及其制剂化学成分不明或无化学对照品,无法阐明其作用和化学物质基础,则无法进行有效的质量控制研究,难以进入国际药品市场,对中药发展形成了阻碍。因此,开展中药化学对照品研究,制备高纯度的化学对照品并对其进行质量标准规范化研究,是中药现代化发展的必由之路,对于推进中药标准化具有较高的学术价值和重要的现实意义[1]
3,5-二羟基-7,4’-二甲氧基黄酮是黄酮醇类化学成分,存在于姜科[2-6]、菊科[7-8]、唇形科[9]、桦木科[10]、蔷薇科[11]、凤仙花科[12]、伞序蕨科[13]等多种植物中,具有较好的抗菌作用[14]。课题组前期研究发现,3,5-二羟基-7,4’-二甲氧基黄酮是壮药拟草果(Amomum paratsaoko S. Q. Tong & Y. M. Xia)的主要活性成分之一[15-16],在拟草果药材中的含量约为0.4 mg·g-1[17],可作为药材及其制剂的指标性成分。目前尚未有相应的国家药品标准物质规定,国内外对3,5-二羟基-7,4’-二甲氧基黄酮作为中药化学对照品的系统研究也未见报道。
本研究以姜科豆蔻属植物拟草果果实为原料,提取分离得到3,5-二羟基-7,4’-二甲氧基黄酮,对3,5-二羟基-7,4’-二甲氧基黄酮的候选化学对照品进行制备纯化和质量控制研究,为其作为中药化学对照品以及含有此成分的药材和制剂的质量标准研究与制定提供科学基础和保证。制备出的3,5-二羟基-7,4’-二甲氧基黄酮还可用于开展药理活性研究,对研制新药和开发其他医药用途具有重要意义。

薄层色谱及提取分离用石油醚、乙酸乙酯、正丁醇、甲醇等试剂均为分析纯(广东光华科技股份有限公司);液相甲醇、乙腈为色谱纯(赛默飞世尔科技公司);水为超纯水。3,5-二羟基-7,4’-二甲氧基黄酮由实验室分离纯化得到,纯度>98%。
拟草果果实于2017年9月采集于广西那坡,经广西中医药研究院中药资源研究所黄云峰副研究员鉴定为姜科豆蔻属植物拟草果(Amomum paratsaoko S. Q. Tong & Y. M. Xia)。
取干燥拟草果果实10 kg,粉碎,用甲醇冷浸提取3次,每次加入甲醇100 L,浸泡7 d,滤过,合并滤液,回收甲醇,得甲醇提取浸膏480 g。甲醇提取浸膏用1 L水分散后,再经5倍体积的石油醚(60~90 ℃)、乙酸乙酯、正丁醇依次萃取,每个溶剂萃取4次,静置,合并有机相层,减压回收溶剂,得到石油醚萃取物(35 g)、乙酸乙酯萃取物(163 g)、正丁醇萃取物(62 g)。乙酸乙酯萃取物经硅胶柱色谱(200~300目),用石油醚-乙酸乙酯(30∶1~0∶100)梯度洗脱,收集石油醚-乙酸乙酯(3∶1)洗脱含3,5-二羟基-7,4’-二甲氧基黄酮的流分,薄层色谱(TLC)检测合并,浓缩,重结晶,就得到3,5-二羟基-7,4’-二甲氧基黄酮的粗结晶(4.0 g)。
粗结晶采用高效制备液相色谱[Ecosil C18 Extend半制备色谱柱(250 mm×10 mm,5 μm),甲醇-水(85∶15),tR=17.0 min]进行制备纯化,收集含3,5-二羟基-7,4’-二甲氧基黄酮流分,经高效液相色谱检测[Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm,5 μm),流动相甲醇-水(80∶20),流速5.0 mL·min-1],合并相同保留时间且纯度>98%以上的流分,减压浓缩,真空干燥后得到纯度>98%以上的3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品(3.0 g),从药材中制得率约为0.03%。
制备所得候选化学对照品为黄色针状结晶(甲醇),溶于乙酸乙酯、氯仿、丙酮、乙醇、甲醇,不溶于水。UV(CH3OH)λmax:350 nm。νKBrmax(cm-1):3 310(-OH);1 658(C=O);1 594、1 555、1 455、1 417(苯环);1 259、1 154(C-O-C);879、833(取代基)。HR-ESI-MS m/z:315.085 24 [M+H]+(理论值:315.086 86,C17H15O6),推导其分子式为C17H14O61H-NMR(400 MHz,DMSO-d6)δ:12.43(1H,s,5-OH),9.65(1H,s,3-OH),8.17(2H,d,J=9.0 Hz,H-2’,6’),7.11(2H,d,J=9.0 Hz,H-3’,5’),6.74(1H,d,J=2.2 Hz,H-8),6.34(1H,d,J=2.1 Hz,H-6),3.86(3H,s,4’-OCH3),3.84(3H,s,7-OCH3);13C-NMR(100 MHz,DMSO-d6)δ:176.0(C-4),164.7(C-7),160.3(C-8a),160.1(C-5),155.9(C-4’),146.4(C-2),136.3(C-3),129.1(C-2’,6’),123.0(C-1’),113.8(C-3’,5’),103.9(C-4a),97.3(C-6),91.8(C-8),55.8 (7-OCH3),55.2(4’-OCH3)。以上述数据与文献[8,10]报道基本一致,故确证该化合物为3,5-二羟基-7,4’-二甲氧基黄酮,化学结构式见图1
取3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品适量,精密称量,用甲醇制成100 μg·mL-1的溶液,在同一硅胶G板上,按不同的点样量梯度点样,点样量分别为2、4、6、8、10 μg(图2中依次为1~5)。分别用石油醚-丙酮-冰醋酸(4∶1∶0.1)、石油醚-乙酸乙酯-冰醋酸(6∶1∶0.1)和环己烷-三氯甲烷-冰醋酸(3∶1∶0.3)3个系统展开,取出,晾干,喷以5%三氯化铝溶液显色,置紫外灯(365 nm)下观察。在3种不同系统溶剂中的5个不同点样量浓度的薄层色谱中,均为单一的黄绿色荧光斑点,无其他杂质斑点,薄层色谱图见图2
取3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品适量,精密称量,加甲醇溶解,配制成1 mg·mL-1的供试品溶液,0.22 μm微孔滤膜,滤过,取续滤液。使用Agilent1260高效液相色谱仪,Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm,5 μm),进样量为20 μL,流速为1.0 mL·min-1,分别在3个不同系统测试条件下测定。系统测试条件一:流动相为甲醇-0.1%磷酸水溶液(80∶20),检测波长266 nm;系统测试条件二:流动相为甲醇-0.1%磷酸水溶液(80∶20),检测波长350 nm;系统测试条件三:流动相为乙腈-0.1%磷酸水溶液(60∶40),检测波长350 nm。3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品在3个色谱条件下的液相色谱图(见图3)均为单一峰,色谱峰纯度较好,峰面积归一化法计算其相对百分含量为98%以上。
取供试品溶液,按“2.3.2”项系统测试条件二,注入高相液相色谱仪进行检测。峰纯度检查液相色谱图见图4,三维图谱见图5,紫外吸收光谱及5点紫外光谱重合图见图6。由图4~6可知,3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品液相色谱图为单一峰,最大吸收波长为350 nm,色谱峰上选择的5个点紫外光谱完全重合,表明为单一物质峰。
在测定条件下,吸取供试品溶液20 μL,注入液相色谱仪,采用系统测试条件二和三分别记录色谱图至主成分的出峰保留时间的2.5倍以上,用面积归一化法计算含量,结果系统测定对照品含量均在98%以上,除溶剂峰外,杂质峰面积总和结果均<2.0%。结果见表1图7
Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm,5 μm),流动相为甲醇-0.1%磷酸水溶液(75∶25),流速1.0 mL·min-1,检测波长350 nm,进样量20 μL。
取3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品适量,精密称量,加甲醇制成50.5 μg·mL-1的对照品储备液。分别精密吸取对照品储备液1.0、2.0、3.0、4.0、5.0、6.0 mL,置10 mL量瓶中,加甲醇至刻度,摇匀,0.22 μm微孔滤膜滤过,取续滤液,即得系列浓度的对照品溶液。分别精密吸取系列浓度的对照品溶液20 μL,按“2.4.1”项下的色谱条件,每个浓度进样2次,测定峰面积值。以进样量(μg)为横坐标,峰面积积分值为纵坐标,求得线性回归方程:
结果表明:3,5-二羟基-7,4’-二甲氧基黄酮对照品质量浓度在5.05~30.3 μg·mL-1呈现良好的线性关系。
精密度:取同一供试品溶液,依法连续进样6次,按“2.4.1”项下的色谱条件进行测定,记录3,5-二羟基-7,4’-二甲氧基黄酮色谱图及峰面积积分值,计算含量。RSD为0.071%,表明仪器的精密度良好。
重现性:称取3,5-二羟基-7,4’-二甲氧基黄酮6份,精密称量,按“2.3.2”项下方法制备供试品溶液,按“2.4.1”项下的色谱条件进行测定,记录3,5-二羟基-7,4’-二甲氧基黄酮的色谱图和峰面积积分值,计算含量。RSD为0.093%,说明方法重现性良好。
稳定性:取同一供试品溶液,在室温下放置1、2、3、6、8、24 h。依次进样测定,记录3,5-二羟基-7,4’-二甲氧基黄酮的色谱图和峰面积积分值,计算含量。结果RSD为0.12%,表明该供试品溶液在24 h内稳定。
采用3种不同厂家、不同品牌的色谱柱Agilent ZORBAX SB-C18(250 mm×4.6 mm,5 μm)、Waters XBridge RP-C18(250 mm×4.6 mm,5 μm)、Thermofisher Hypersil GOLD(250 mm×4.6 mm,5 μm),按“2.4.1”项下的条件,分别对保留时间、理论板数、分离度和杂质分离效果进行测定。3,5-二羟基-7,4’-二甲氧基黄酮峰与其他杂质峰达到基线分离,分离度>1.5,按3,5-二羟基-7,4’-二甲氧基黄酮峰计算理论板数≥4 000时,可满足测定要求,见表2
2个条件的流动相系统(甲醇-0.1%磷酸水溶液和乙腈-0.1%磷酸水溶液)检出的3,5-二羟基-7,4’-二甲氧基黄酮,采用峰面积归一化法计算峰纯度分别为99.85%、99.86%,杂质峰面积总和结果均<2.0%(见“2.3.4”)。采用0.1%自身对照法[18]测得3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品色谱纯度为99.85%(n=2),杂质总含量均<2.0%。
参照2020年版《中华人民共和国药典》(四部通则0841)炽灼残渣检查法,测得3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品炽灼残渣为0.02%。
参照2020年版《中华人民共和国药典》(四部通则9103)引湿性试验方法,测得3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品增重百分率为1.5%,略有引湿性。计算公式:增重百分率=(m3-m1)/(m 2-m 1)×100%,其中,m1为恒温干燥后具塞玻璃称量瓶的质量,m2为供试品加具塞玻璃称量瓶的质量,m3为恒温恒湿条件下24 h供试品加具塞玻璃称量瓶的质量。
采用质量平衡法[19]计算3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品纯度为99.83%。计算公式:候选化学对照品含量=色谱峰纯度×(100%-炽灼残渣%)。
本研究分离纯化的3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品,经过3个展开剂溶剂系统和5个不同浓度的梯度点样,TLC检测为单一斑点,未见有其他的杂质斑点;在2个色谱系统2个波长检测HPLC液相色谱图均为单一峰,且纯度均达到98.0%以上,杂质总量<2.0%;采用高效液相色谱法建立的3,5-二羟基-7,4’-二甲氧基黄酮候选化学对照品含量测定方法,经方法学考察具有专属性强、精密度良好、准确可靠的特点,符合国家药品标准工作手册关于化学对照品的要求[20]。研究结果可为3,5二羟基-7,4’-二甲氧基黄酮化学对照品的质量标准提出与制定提供基础资料,为研制新药和开发新药标准提供技术支持,还可为拟草果及其他含3,5-二羟基-7,4’-二甲氧基黄酮的药材与制剂的质量控制提供依据。
