Objective:

To establish an HPLC method for simultaneous determination of seven components(narirutin, naringin, hesperidin, neohesperidin, naringenin, nobiletin and atractylenolide Ⅲ) in Zhizhu granules. And to provide reference for its quality evaluation by using chemometric analysis.

Methods:

The chromatographic column Waters Symmetry C18(250 mm×4.6 mm,5 μm) was adopted using HPLC-DAD. The mobile phase consisted of acetonitrile (A)-0.1% phosphoric acid (B) in gradient elution with flow rate of 1.0 mL·min-1, the detection wavelength were 220 nm (atractylenolide Ⅲ), 280 nm (narirutin, naringin, hesperidin, neohesperidin and naringenin) and 332 nm (nobiletin). The column temperature was set at 30℃ and the injection volume was 10 μL. Cluster analysis, principal component analysis and orthogonal partial least squares-discrimination analysis were used to distinguish the content determination results.

Results:

The resolution of each component in 24 batches of Zhizhu granules was good, and the linear relationship between concentration and peak area was good (r>0.999 9). The average recoveries were within 87.6%-114.3%. There were some differences in 24 batches of Zhizhu granules, and 4 different components were screened out, which were naringin, hesperidin, neohesperidin and naringin. The contents of narirutin, naringin, hesperidin, neohesperidin, naringenin, nobiletin and atractylenolide Ⅲ in 24 batches of Zhizhu granules samples were 0.378 4-1.380 1 mg·g-1, 5.125 8-18.137 6 mg·g-1, 0.283 9-1.195 8 mg·g-1, 4.490 3-22.585 0 mg·g-1, 0.022 5-0.349 8 mg·g-1, 0.063 3-0.211 4 mg·g-1 and 0.054 7-0.137 5 mg·g-1, respectively.

Conclusion:

The established method is accurate, reliable and reproducible, which can provide reference for the quality control of Zhizhu granules.

目的:

建立高效液相色谱法同时测定枳术颗粒中芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ 7个指标性成分的含量,同时结合化学计量学分析为其质量评价提供参考依据。

方法:

采用Waters Symmetry C18色谱柱(250 mm×4.6 mm,5 μm),流动相为乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱,流速1.0 mL·min-1,检测波长220 nm(白术内酯Ⅲ)、280 nm(芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素)和332 nm(川陈皮素),柱温30 ℃,进样体积10 μL。采用统计分析软件对含量测定结果进行聚类分析、主成分分析与正交偏最小二乘法判别。

结果:

24批枳术颗粒中各成分分离度较好,在考察的线性范围内浓度与峰面积呈良好的线性关系(r>0.999 9),平均回收率在87.6%~114.3%。24批枳术颗粒存在一定差异,筛选出4个差异性成分柚皮苷、橙皮苷、新橙皮苷和柚皮素。24批枳术颗粒供试样品中芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ的含量依次为0.378 4~1.380 1、5.125 8~18.137 6、0.283 9~1.195 8、4.490 3~22.585 0、0.022 5~0.349 8、0.063 3~0.211 4和0.054 7~0.137 5 mg·g-1

结论:

所建立的方法准确可靠,重复性强,可为枳术颗粒的质量控制提供参考依据。

目的:

建立高效液相色谱法同时测定枳术颗粒中芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ 7个指标性成分的含量,同时结合化学计量学分析为其质量评价提供参考依据。

方法:

采用Waters Symmetry C18色谱柱(250 mm×4.6 mm,5 μm),流动相为乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱,流速1.0 mL·min-1,检测波长220 nm(白术内酯Ⅲ)、280 nm(芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素)和332 nm(川陈皮素),柱温30 ℃,进样体积10 μL。采用统计分析软件对含量测定结果进行聚类分析、主成分分析与正交偏最小二乘法判别。

24批枳术颗粒中各成分分离度较好,在考察的线性范围内浓度与峰面积呈良好的线性关系(r>0.999 9),平均回收率在87.6%~114.3%。24批枳术颗粒存在一定差异,筛选出4个差异性成分柚皮苷、橙皮苷、新橙皮苷和柚皮素。24批枳术颗粒供试样品中芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ的含量依次为0.378 4~1.380 1、5.125 8~18.137 6、0.283 9~1.195 8、4.490 3~22.585 0、0.022 5~0.349 8、0.063 3~0.211 4和0.054 7~0.137 5 mg·g-1

结论:

所建立的方法准确可靠,重复性强,可为枳术颗粒的质量控制提供参考依据。

Objective:

To establish an HPLC method for simultaneous determination of seven components(narirutin, naringin, hesperidin, neohesperidin, naringenin, nobiletin and atractylenolide Ⅲ) in Zhizhu granules. And to provide reference for its quality evaluation by using chemometric analysis.

Methods:

The chromatographic column Waters Symmetry C18(250 mm×4.6 mm,5 μm) was adopted using HPLC-DAD. The mobile phase consisted of acetonitrile (A)-0.1% phosphoric acid (B) in gradient elution with flow rate of 1.0 mL·min-1, the detection wavelength were 220 nm (atractylenolide Ⅲ), 280 nm (narirutin, naringin, hesperidin, neohesperidin and naringenin) and 332 nm (nobiletin). The column temperature was set at 30℃ and the injection volume was 10 μL. Cluster analysis, principal component analysis and orthogonal partial least squares-discrimination analysis were used to distinguish the content determination results.

Results:

The resolution of each component in 24 batches of Zhizhu granules was good, and the linear relationship between concentration and peak area was good (r>0.999 9). The average recoveries were within 87.6%-114.3%. There were some differences in 24 batches of Zhizhu granules, and 4 different components were screened out, which were naringin, hesperidin, neohesperidin and naringin. The contents of narirutin, naringin, hesperidin, neohesperidin, naringenin, nobiletin and atractylenolide Ⅲ in 24 batches of Zhizhu granules samples were 0.378 4-1.380 1 mg·g-1, 5.125 8-18.137 6 mg·g-1, 0.283 9-1.195 8 mg·g-1, 4.490 3-22.585 0 mg·g-1, 0.022 5-0.349 8 mg·g-1, 0.063 3-0.211 4 mg·g-1 and 0.054 7-0.137 5 mg·g-1, respectively.

Conclusion:

The established method is accurate, reliable and reproducible, which can provide reference for the quality control of Zhizhu granules.

枳术颗粒由麸炒枳实、麸炒白术、荷叶制备而成,具有健脾消食、行气化湿等功效,临床上用于治疗慢性萎缩性胃炎[1]、残胃炎[2]、消化性溃疡[3-4]等胃肠道疾病。枳术颗粒系利用现代提取方法由枳术丸改剂型来的国家4类中药创新药,相比于丸剂,颗粒剂溶出快,药效发挥迅速,且服药依顺性更好,适合小儿与老人服用[5]。枳术颗粒所含化学成分丰富,但目前2020年版《中华人民共和国药典》的含量测定下仅测定柚皮苷及新橙皮苷,文献报道对枳术颗粒的成分检测多集中于枳实所含的黄酮类物质[6-8],检测种类较为单一。中药复方成分复杂,药效为多成分相互作用的结果,单一组分测定不利于中药复方制剂的整体质量控制[9-10]。麸炒白术在该方中用量为麸炒枳实的2倍,有先补脾胃之弱,后化脾胃之伤的功效,故该方适于脾胃虚弱的患者[11]。有文献报道,枳实与白术1∶2配伍时能够促进小鼠肠平滑肌收缩,有利于胃排空[12]。枳实在麸炒后促进胃肠蠕动的苷类成分含量降低,但与白术配伍后可明显改善这一现象,药材配伍保证了组方的健脾消食功效[13]。所以,枳实和白术作为组方中发挥药效的主要成分,同时控制其所含的活性成分更有利于枳术颗粒的药效评价。近年来,多组分含量测定及化学计量学分析结合的应用在中药及其制剂的质量评价中越来越受到重视[14]。本研究建立了高效液相色谱法同时测定麸炒枳实及麸炒白术中的7个活性成分,同时进行化学计量学分析,该方法简便、准确,重复性好,可为枳术颗粒质量的全面控制提供一定参考依据。
LC-40D岛津高效液相色谱仪,配置LC-40D二元泵、SIL-40C全自动进样器、CTO-40S柱温箱、SPD-M40检测器(岛津有限公司),ML104/02万分之一天平和XPR2百万分之一天平均购自梅特勒-托利多仪器(上海)有限公司,RODI-220A1纯水机(锐思捷水纯化技术有限公司),KH-500E超声波清洗仪(昆山禾创超声仪器有限公司)。
枳术颗粒(6 g·袋-1)24批,编号为S1~S24,相应的批号分别为190201、200301、200401、200501、200601、201001、201101、201102、201103、201104、210101、210201、210301、210302、210303、210304、220101、220201、220301、220401、220501、220502、220601、220602,均由南京中山制药有限公司提供。
对照品芸香柚皮苷(批号N2006002,含量99.27%),购自成都迈克斯医药科技有限公司;柚皮苷(批号110722-201714,含量93.4%)、橙皮苷(批号110721-202019,含量95.3%)、新橙皮苷(批号111857-201804,含量99.4%)、川陈皮素(批号112055-202102,含量100%)、白术内酯Ⅲ(批号111978-201501,含量99.9%),均购自中国食品药品检定研究院;柚皮素(批号100917,含量98%),购自上海融禾医药科技有限公司。色谱级乙腈(22075301)和甲醇(22075179)均购自安徽天地高纯溶剂有限公司;磷酸(C14276009)购自上海麦克林生化科技有限公司;甲醇(20220624)和乙醇(20220704)均购自国药集团化学试剂有限公司。
精密称取芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ的对照品适量,用甲醇溶解,配制成含上述7个成分分别为0.399 3、3.012 3、0.201 1、2.503 2、0.032 5、0.044 2和0.031 3 mg·mL-1的混合对照品母液。
枳术颗粒研细,精密称取1 g,精密加入甲醇溶液10 mL,密塞,称量,超声(功率300 W,频率50 kHz)处理30 min,放冷,再称量,用甲醇溶液补足减失的量,摇匀,0.22μm微孔滤膜滤过,取续滤液,即得。
按枳术颗粒制备方法制得缺枳实、白术的阴性样品,再按“2.1.2”项下方法制备阴性样品溶液。
Waters Symmetry C18色谱柱(250 mm×4.6 mm,5 μm),流动相为乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱(0~7 min,10%A→15%A;7~20 min,15%A→18%A;20~30 min,18%A→19%A;30~45 min,19%A→35%A;45~55 min,35%A→60%A;55~65 min,60%A→75%A;65~70 min,75%A→10%A;70~80 min,10%A),检测波长为220 nm(白术内酯Ⅲ)、280 nm(芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素)和332 nm(川陈皮素),流速1.0 mL·min-1,柱温30 ℃,进样体积10 μL。
分别精密吸取供试品溶液(S6)、混合对照品母液、阴性样品溶液各10 μL,按“2.2”项下色谱条件进样,该检测条件下待测成分色谱峰对称,与相邻色谱峰分离度R>1.5,且阴性无干扰,见图1
分别精密量取“2.1.1”项下母液0.3、0.5、1.0、1.5、2.0、2.5 mL,置5 mL量瓶中,用甲醇定容至刻度,得系列浓度对照品溶液(对照-1~对照-6),在“2.2”项色谱条件下进样测定。以对照品质量浓度(X,mg·mL-1)为横坐标,峰面积Y为纵坐标进行线性回归,见表1
取“2.3.2”项下对照-4溶液,按“2.2”项色谱条件下连续进样6次,记录峰面积。芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ峰面积的RSD分别为0.24%、0.24%、0.33%、0.25%、0.24%、0.24%、0.11%,说明仪器精密度良好。
取同一样品(S6),按“2.1.2”项方法制备供试品溶液,按“2.2”项色谱条件,分别于0、2、4、6、8、10、12、24 h进样,记录各成分峰面积。芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ峰面积的RSD分别为4.7%、1.0%、1.1%、1.1%、0.94%、0.91%、0.80%,说明供试品溶液在24 h内稳定。
取同一样品(S6),按“2.1.2”项方法平行制备6份供试品溶液,在“2.2”项色谱条件下进样测定。芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ的含量分别为0.910 0、7.734 6、0.458 8、6.502 8、0.084 5、0.098 8、0.077 0 mg·g-1,RSD分别为2.4%、2.4%、2.4%、2.4%、2.5%、2.2%、2.1%,说明该方法重复性良好。
取含量已知的枳术颗粒(S6)9份,精密称取0.5 g,分别按已知含量的80%、100%、120% 3个水平加入对照品,按“2.1.2”项方法制备供试溶液,按“2.2”项色谱条件进样测定,计算加样回收率及RSD。芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素、白术内酯Ⅲ的回收率在85.4%~116.6%,RSD在0.66%~3.2%,说明该方法准确度较好,见表2
取24批枳术颗粒,按“2.1.2”项方法制备供试品溶液,按“2.2”项色谱条件检测,计算各成分的含量。结果见表3
聚类分析是对测得的样本数据集进行分类,将相似程度相似的样本聚为一类的多元统计方法[15]。将含量测定结果用SPSS25.0软件进行分析,采用组间连接法,以欧氏距离为测度,结果见图2。当欧氏距离为15时,样品分为3类,其中S1~S6、S12和S13归为Ⅰ类,S7~S11和S14~S24归为Ⅱ类。
主成分分析通过数据降维来研究数据集的内在关系,在尽可能保留原有信息的条件下将多个指标转换成少数可反应原始大部分信息(70%~85%)的互相独立的主成分[16]。使用SPSS25.0对24批枳术颗粒含量测定结果进行主成分分析,提取特征值>1的主成分,得到2个主成分,累积方差贡献率为82.03%,可以解释样品的大部分信息。碎石图(图3)显示前2个成分陡峭,其他成分较平缓,说明前2个成分可以代表样品的大部分信息[17]。利用SIMCA14.0软件得到主成分得分图(图4),可看出24批样品分为3类,其中S1~S6归为Ⅰ类,S7~S11和S14~S24归为Ⅱ类,S12和S13归为Ⅲ类,与CA结果基本一致。
偏最小二乘判别分析是一种有监督的判别方法,因其集主成分分析、多元线性回归分析、典型相关分析为一体而使结果具有简洁直观的特点,被广泛应用于中药及其制剂的质量控制中[18]。采用SIMCA14.0软件进行PLS-DA分析,共提取4个主成分,模型解释率R2X为0.995,稳定性参数R2Y为0.732,预测能力参数Q2为0.539,均>0.5,说明该模型建立成功,具有较好的稳定性和预测力[19]。由得分散点图可以看出,除S1为异常值外,其余样本信息均落在95%置信区间内,剔除异常值后可分为2类,其中S2~S6、S12和S13归为Ⅰ类,S7~S11和S14~S24归为Ⅱ类,与CA、PCA结果基本一致,见图5。VIP图(图6)可直观反应各成分对影响组间差异的权重大小,VIP>1说明该组分对权重影响率高于50%,为差异标志物[20]。由VIP图提取4个差异标志物,按权重大小排序为3号峰(橙皮苷)、4号峰(新橙皮苷)、2号峰(柚皮苷)和5号峰(柚皮素)。其中,橙皮苷、新橙皮苷、柚皮苷可促进功能性消化不良大鼠的胃排空及小肠推进,柚皮素可促进正常小鼠的小肠推进[21]。所以,这些化合物可能是枳术颗粒发挥消食功效的重要成分,在生产中加强对这类成分的控制,可以更好地保证药品稳定发挥疗效。
本实验采用单因素法考察了不同提取时间(30、45和60 min),不同提取方式(加热回流和超声),不同提取溶剂(甲醇、70%甲醇水、乙醇和70%乙醇水)对有效成分提取的影响,最佳前处理方式为甲醇为溶剂超声30 min。
本实验对不同流动相种类(甲醇-水、乙腈-水和乙腈-0.1%磷酸水)及洗脱比例进行考察,最终确定使用乙腈-0.1%磷酸水进行梯度洗脱所得到的色谱图峰型对称,分离度较好。DAD 190~800 nm紫外区间进行全波长扫描,结果显示白术内酯Ⅲ的最大吸收波长为220 nm,芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素的最大吸收波长为280 nm,川陈皮素的最大吸收波长为332 nm。
本实验所测定的芸香柚皮苷、柚皮苷、橙皮苷、新橙皮苷、柚皮素、川陈皮素均属于麸炒枳实中的黄酮类成分,白术内酯Ⅲ来源于麸炒白术。现代药理学研究表明枳实中的黄酮类物质是治疗胃肠道疾病的主要成分[22],白术经麸炒后内酯类成分含量增加,健脾功效得到加强[23],故所选取的7个成分可作为评价枳术颗粒健脾消食功效的重要指标。含量测定结果表明所收集的2019年~2022年生产的24批次枳术颗粒中7个成分存在一定差异,造成差异的原因可能是饮片批次差异、生产过程中的质量控制差异。2020年版《中华人民共和国药典》规定枳术颗粒中每袋柚皮苷含量应不少于30 mg,新橙皮苷含量应不少于20 mg,可知各批次均符合标准。但现行标准中只规定了含量下限并无含量上限,可以临床实验品为依据,根据药效及含量规定合理的限度范围,不仅可以更好地规范生产工艺,也能为药品稳定发挥疗效提供保障[24]
CA和PCA结果表明,样品S1~S6与样品S7~S24间具有一定差异,推测造成批间差异的原因可能是药材的来源、产地及采收年份等因素[25]。PLS-DA结果提取了4个差异标志物,均来源于麸炒枳实,提示应更加注重控制麸炒枳实的药材质量。饮片是中药制剂的主要原料,投料会将饮片差异带到制剂中,因此确保投料环节饮片质量的稳定对中药制剂的稳定性尤为重要[26]。此外生产过程中的浓缩、醇沉单元及浸膏干燥等环节均会影响制剂质量,采用混批投料、标准化生产过程可进一步提高产品稳定性[24]。故生产过程中应重视控制投料饮片的批次差异、生产过程及储存方式等以保证样品批次间的均一性与稳定性。
本实验建立了一种枳术颗粒含量测定方法,可同时测定该产品中麸炒枳实及麸炒白术的有效成分,所建立的方法简便,准确度高,重复性良好,为更全面地评价枳术颗粒的质量提供了一定参考依据。
