Objective:

To establish an ion chromatography method for determination the content of sodium caprylate in human blood albumin products.

Methods:

The samples were precipitated with eluent, the suspension was centrifuged and filtered, the filtrate was injected to IC, and heptanoic acid was used as the internal standard. A Dionex InPacTM NS1 Analytical Column (250 mm×4mm, 10 μm) and a Dionex InPacTM NG1 Guard Column (35 mm×4 mm, 10 μm) were used,the flow rate was 1.0 mL·min-1. The conductivity detector and ASRS 300 membrane suppressor were used, and the regenerant solution was 5 mmol·L-1 tetrabutylsodium hydroxide solution; the column temperature was 30 ℃ and the injection volume was 25 μL.

Results:

The resolution between the peaks of sodium caprylate and the internal standard was greater than 1.5, and the linearity of concentration of sodium caprylate was good in the range of 0.38-2.52 mmol·L-1, r=0.999 5 (n=6). The RSD of the repeatability test was 1.1% (n=6). The average recovery was 97.4% and RSD was 1.8% (n=9). The limits of quantification and detection were 0.19 nmol and 0.09 nmol, respectively. The determination results of the content of sodium octanoate in 20 batches of human blood albumin samples from 7 enterprises at home and abroad ranged from 0.073-0.163 mmol·g-1.

Conclusion:

The method established in this study is simple to operate, accurate in results, high in sensitivity and good in repeatability, can be used for the determination of sodium caprylate content in human blood albumin products and provide a method guarantee for its quality control.

目的:

建立离子色谱法测定人血白蛋白制品中辛酸钠的含量。

方法:

样品加淋洗液[甲醇-乙腈-1.0 mmol·L-1盐酸(20∶42∶38)]沉淀蛋白,离心取上清液,过滤后直接进样,以庚酸为内标。采用Dionex InPacTM NS1分析柱(250 mm×4 mm,10 μm)与Dionex InPacTM NG1保护柱(35 mm×4 mm,10 μm),流速1.0 mL·min-1,ASRS 300膜抑制器,化学抑制,再生液为5 mmol·L-1四丁基氢氧化钠溶液,电导检测器检测,柱温30 ℃,进样量25 μL。

结果:

辛酸峰与内标峰间的分离度>1.5;辛酸钠在0.38~2.52 mmol·L-1范围内线性关系良好,r=0.999 5(n=6);重复性试验的RSD为1.1%(n=6);平均加样回收率为97.4%,RSD为1.8%(n=9);定量限与检测限分别为0.19 nmol与0.09 nmol;国内外7家企业20批人血白蛋白制品中辛酸钠含量范围为0.073~0.163 mmol·g-1

结论:

本研究建立的方法操作简便易行,结果准确,灵敏度高,重复性好,可用于人血白蛋白制品中辛酸钠含量的测定,为其质量控制提供方法保证。

目的:

建立离子色谱法测定人血白蛋白制品中辛酸钠的含量。

方法:

样品加淋洗液[甲醇-乙腈-1.0 mmol·L-1盐酸(20∶42∶38)]沉淀蛋白,离心取上清液,过滤后直接进样,以庚酸为内标。采用Dionex InPacTM NS1分析柱(250 mm×4 mm,10 μm)与Dionex InPacTM NG1保护柱(35 mm×4 mm,10 μm),流速1.0 mL·min-1,ASRS 300膜抑制器,化学抑制,再生液为5 mmol·L-1四丁基氢氧化钠溶液,电导检测器检测,柱温30 ℃,进样量25 μL。

结果:

辛酸峰与内标峰间的分离度>1.5;辛酸钠在0.38~2.52 mmol·L-1范围内线性关系良好,r=0.999 5(n=6);重复性试验的RSD为1.1%(n=6);平均加样回收率为97.4%,RSD为1.8%(n=9);定量限与检测限分别为0.19 nmol与0.09 nmol;国内外7家企业20批人血白蛋白制品中辛酸钠含量范围为0.073~0.163 mmol·g-1

结论:

本研究建立的方法操作简便易行,结果准确,灵敏度高,重复性好,可用于人血白蛋白制品中辛酸钠含量的测定,为其质量控制提供方法保证。

Objective:

To establish an ion chromatography method for determination the content of sodium caprylate in human blood albumin products.

Methods:

The samples were precipitated with eluent, the suspension was centrifuged and filtered, the filtrate was injected to IC, and heptanoic acid was used as the internal standard. A Dionex InPacTM NS1 Analytical Column (250 mm×4mm, 10 μm) and a Dionex InPacTM NG1 Guard Column (35 mm×4 mm, 10 μm) were used,the flow rate was 1.0 mL·min-1. The conductivity detector and ASRS 300 membrane suppressor were used, and the regenerant solution was 5 mmol·L-1 tetrabutylsodium hydroxide solution; the column temperature was 30 ℃ and the injection volume was 25 μL.

Results:

The resolution between the peaks of sodium caprylate and the internal standard was greater than 1.5, and the linearity of concentration of sodium caprylate was good in the range of 0.38-2.52 mmol·L-1, r=0.999 5 (n=6). The RSD of the repeatability test was 1.1% (n=6). The average recovery was 97.4% and RSD was 1.8% (n=9). The limits of quantification and detection were 0.19 nmol and 0.09 nmol, respectively. The determination results of the content of sodium octanoate in 20 batches of human blood albumin samples from 7 enterprises at home and abroad ranged from 0.073-0.163 mmol·g-1.

Conclusion:

The method established in this study is simple to operate, accurate in results, high in sensitivity and good in repeatability, can be used for the determination of sodium caprylate content in human blood albumin products and provide a method guarantee for its quality control.

人血白蛋白是从健康人的血液中提取、分离和制备而成的一种可直接注射到病人体内的血液制品,其主要的药理作用是增加血容量维持胶体渗透压[1],作为载体具有运输和解毒的作用[2],也可以作为氮源为组织提供营养供给,临床上主要用于肝硬化腹水、Ⅰ型或Ⅱ型肝肾综合征以及儿童急性呼吸窘迫综合征和烧伤造成的低血容量休克(12~16岁儿童)的治疗,还可用于自发性细菌性腹膜炎、胰腺炎和腹膜炎等疾病的辅助治疗[3-6]
人血白蛋白制品在生产过程中,需在60 ℃进行病毒灭活处理10 h[7-8]。为了防止蛋白质发生热变性和产生聚体,需在病毒热灭活时和制剂中添加适量的辛酸钠作为稳定剂。加入的辛酸钠含量过低会降低白蛋白的热稳定性形成过多的聚合体;加入的辛酸钠含量太高(每1 g蛋白质>0.180 mmol),则有可能导致白蛋白制品引起不良反应,如抑制血小板凝聚、引起低血糖症、抑制肝脏对长链脂肪酸的消除等[9-11],故应采用合适的方法准确测定人血白蛋白制品中辛酸钠的含量,以保证人血白蛋白制品的安全性和有效性。2020年版《中国药典》三部规定:人血白蛋白制品中,每1 g蛋白质中辛酸钠含量应为0.140~0.180 mmol,如与乙酰色氨酸混合使用,则每1 g蛋白质中应为0.064~0.096 mmol [12]
目前,关于人血白蛋白制品中辛酸钠含量测定的方法主要有气相色谱法[13-14]、反向高效液相色谱[11,15-16]、离子排斥色谱法[10]与萃取滴定法[17-18],但这些方法均需采用毒性有机试剂如三氯甲烷、正己烷、乙酸乙酯、石油醚、乙醚等萃取辛酸后再制成衍生物或直接测定,均存在样品前处理时间较长,操作步骤烦琐,检验周期长等缺点。离子色谱法是继高效液相色谱法后发展起来的液相分析技术,具有特异、灵敏、准确、环保无污染、自动化程度高等优势,适用于分析阴阳离子、有机酸、胺、醇类和氨基酸等离子型物质,在药物分析领域内已得到广泛应用[19],但在文献报道中,尚未见采用离子色谱法测定人血白蛋白制品中辛酸钠含量的报道。本文建立了离子色谱法测定人血白蛋白制品中辛酸钠的含量,并对国内外7家企业20批次人血白蛋白中的辛酸钠含量进行了测定。
Dionex ICS 6000型离子色谱仪,配备ASRS 300 4mm阴离子抑制器,电导检测器及变色龙7.1工作站(Thermo Fisher Scientific公司);Agilent 7890型气相色谱仪,配备FID检测器及Empower3色谱工作站(Waters公司);XS205型十万分之一电子天平(Mettler Toledo公司)。
10%四丁基氢氧化铵为分析纯(国药集团化学试剂有限公司);高氯酸、盐酸、三氯甲烷均为分析纯(上海凌峰化学试剂有限公司);甲醇与乙腈均为色谱纯(Fisher Scientific公司);超纯水由Mili-Q纯水机制备。
人血白蛋白样品来自国内外7家企业,共20批;辛酸标准品(批号MKCM7502,纯度99.6%)、庚酸标准品(批号BCBW6192,纯度99.9%),Sigma-Aldrich公司。
将甲醇-乙腈-1.0 mmol·L-1盐酸(20∶42∶38)作为淋洗液。
取庚酸标准品适量,精密称定,用淋洗液定量稀释制成每1 L中约含2.36 mmol的溶液,即得。
取辛酸标准品适量,精密称定,用淋洗液定量稀释制成每1 L中含辛酸约为25 mmol的溶液,即得。
精密量取“2.1.3”项下的对照品储备液适量,用内标溶液定量分别稀释,制成每1 L中含辛酸约为0.38、0.50、0.81、1.26、2.01、2.52 mmol的溶液,即得。
精密量取本品1.0 mL,置20 mL量瓶中,用内标溶液稀释至刻度,摇匀,于20 ℃ 3 800 r·min-1离心(20±5)min,取上清液,即得。
采用Dionex InPacTM NS1分析柱(250 mm×4 mm,10 μm)与Dionex InPacTM NG1保护柱(35 mm×4 mm,10 μm),以甲醇-乙腈-1.0 mmol·L-1盐酸(20∶42∶38)为淋洗液,ASRS 300膜抑制器,化学抑制,再生液为5 mmol·L-1四丁基氢氧化钠溶液,电导检测器检测,柱温30 ℃,流速1.0 mL·min-1,进样量25 μL。
精密量取“2.1”项下的空白溶液(淋洗液)、标准曲线溶液3、供试品溶液,分别按“2.2”项下的色谱条件进样测定,记录色谱图,见图1。结果表明空白溶液(淋洗液)不干扰主峰及内标峰的测定;标准曲线溶液3色谱图中,辛酸峰与庚酸峰间的分离度为4.9,辛酸峰的拖尾因子为1.0。
精密量取“2.1.4”项下的标准曲线溶液1~6,分别按“2.2”项下的色谱条件进样测定,记录色谱图。以质量浓度(X)为横坐标,辛酸峰面积与庚酸峰面积的比值(Y)为纵坐标,进行线性回归。结果显示辛酸在质量浓度0.38~2.52 mmol·L-1范围内线性关系良好,线性方程(n=6)为
精密量取“2.1.4”项下的标准曲线溶液1,用内标溶液逐级定量稀释,按“2.2”项下的色谱条件进样测定,以S/N=3或10时测得的辛酸量分别作为检测限和定量限,结果辛酸钠的定量限与检测限分别为0.19 nmol·L-1与0.09 nmol·L-1
取同一批人血白蛋白制品(批号A),按“2.1.5”项下的方法平行制备6份供试品溶液,按“2.2”下的色谱条件分别进样测定,记录色谱图。结果每1 g蛋白质中辛酸钠含量均值(n=6)为0.074 mmol,RSD为1.1%,表明方法的重复性良好。
由另一名分析人员在不同时间,按“2.3.4”项下方法进行重复性试验,结果每1 g蛋白质中辛酸钠含量均值(n=6)为0.075 mmol,RSD为1.1%;2次重复项试验每1 g蛋白质中辛酸钠含量均值(n=12)为0.076 mmol,RSD为1.7%。
精密量取“2.3.4”项下的供试品溶液,于室温条件下放置0、4、8、12、16、20与24 h,按“2.2”项下的色谱条件分别进样测定,结果辛酸峰面积与庚酸峰面积比值的RSD(n=7)为1.6%,表明供试品溶液于室温条件下放置24 h稳定性良好。
精密量取本品(批号A)1.0 mL,置50 mL量瓶中,共9份,分别精密加入“2.1.3”项下的对照品储备液0.6、1.0、1.4 mL,每个浓度平行配制3份,用内标溶液稀释至刻度,摇匀,分别作为80%、100%、120%的加样回收溶液。取上述加样回收溶液,按“2.2”项下色谱条件分别进样测定,计算回收率。结果80%、100%与120% 3个不同浓度的平均加样回收率(n=3)分别为98.3%、96.8%与97.2%,平均加样回收率为97.4%,RSD(n=9)为1.8%,表明方法的准确度良好。
在“2.1”项下色谱条件的基础上,分别改变Dionex InPacTM NS1分析柱批号(01617180、01914119、0201134)、再生液浓度(±1 mmol·L-1)、流速(±0.2 mL·min-1)、柱温(±5 ℃),其他色谱条件不变,考察对样品测定的影响。取“2.1”项下的标准曲线溶液1~6与供试品溶液,按上述条件分别进样测定,记录色谱图,结果见表1。结果表明,当各条件适当变动时,辛酸峰与庚酸峰分离度均>1.5;供试品溶液中辛酸钠含量结果基本一致,表明方法的耐用性良好。
取7家企业的20批人血白蛋白制品,分别按“2.1.5”项下的方法制备供试品溶液,按“2.2”项下的色谱条件分别进样测定,记录色谱图。辛酸钠含量测定结果见表2
采用离子色谱法测定辛酸,有2种分离机理可以选择。第1种是离子交换机理,固定相为阴离子交换树脂(色谱柱使用AS11-HC),淋洗液为0.04 mol·L-1氢氧化钾溶液,在这种分离机理下,辛酸虽可以获得合适的保留,但色谱峰峰型较差,拖尾严重,拖尾因子为2.04,色谱图见图2;第2种是本文建立的离子色谱法,交换机理为反相离子对机理,固定相是高交联度、高比表面积的中性无离子交换功能-的聚苯乙烯大孔树脂,流动相为甲醇-乙腈-1.0 mmol·L-1盐酸(20∶42∶38),在该色谱条件下,辛酸既可获得合适的保留,也可保持良好的峰形,拖尾因子为1.0,色谱图见图1
辛酸钠本身无紫外吸收,无法直接用通用紫外检测器进行检测[16],而辛酸的pKa为4.89,在电导检测器上可有较好的响应,灵敏度高,故检测器选择使用信噪比高且重现性好的电导检测器。
使用电导检测器时,淋洗液有很高的检测信号。抑制器可降低淋洗液的背景电导,增加被测离子的电导值,改善信噪比。常用的抑制模式有化学抑制、电抑制等。测定阴离子时,化学抑制使用酸作为再生液;电抑制使用外接水或电导池后流出的废液作为再生液。本方法的淋洗液为甲醇-乙腈-盐酸混合溶液,其在电抑制模式下会有较高的背景值,会干扰辛酸钠的测定,而在化学抑制模式下其无响应值,因此,为了降低背景电导值、提高灵敏度,可在色谱柱和检测器之间加入化学抑制器,使用再生液为5 mmol·L-1四丁基氢氧化钠溶液作为再生液。
2020年版《中国药典》三部通则“3111辛酸钠含量测定方法”为气相色谱法,为与之进行比较,本文还采用气相色谱法测定了7家企业20批人血白蛋白制品中的辛酸钠含量,结果见表1。统计学结果表明:2种方法最大相对平均偏差均不超过3%,检测结果无显著差异(P <0.05)。由此可见本文方法与法定方法在测定辛酸钠含量结果上具有高度的一致性,但法定方法样品前处理烦琐,需挥干萃取剂,检验周期长,且萃取剂三氯甲烷易挥发对人体有毒害作用,而本文建立的离子色谱法,无需使用三氯甲烷,样品仅需一步稀释后便可进样测定,前处理简单,检验周期短。
本文建立了一种人血白蛋白制品中辛酸钠含量测定的离子色谱法,该方法操作简便、环保、准确度和精密度好,灵敏度高,与2020年版《中国药典》三部通则“3111辛酸钠含量测定方法”相比,样品配制简单,无需使用有机试剂三氯甲烷,避免了挥发的三氯甲烷对人和环境的影响[20],缩短了试验时间,可作为中国药典收录的气相色谱法测定人血白蛋白制品中辛酸钠含量的有效补充,扩大先进检测技术在国家标准中的应用,为相关药品监督管理部门和药物研发生产单位提供参考,具有一定推广和应用价值。
