Objective:

To establish an HPLC method for simultaneous determination of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, forsythiaside A, 3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid, forsythin, andrographalide and dehydroandrographalide in compound Shuanghua tablets.

Methods:

The samples were extracted with 50% methanol solution under ultrasonic condition, and were performed on Agilent Zoabax SB-C18 column (250 mm×4.6 mm, 5 μm) by gradient elution of acetonitrile -0.15% phosphoric acid solution at a flow rate of 1.0 mL·min-1. The column temperature was 30 ℃, the injection volume was 10 μL, and the detection wavelength were set at 327 nm (detecting neochlorogenic acid,chlorogenic acid, cryptochlorogenic acid, forsythiaside A,3,4-O-dicaffeoylquinic acid,3,5-O-dicaffeoylquinic acid and 4,5-O-dicaffeoylquinic acid) and 226 nm (detecting forsythin, andrographalide and dehydroandrographalide).

Results:

The linear ranges of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid,forsythiaside A,3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid, forsythin, andrographalide and dehydroandrographalide were 4.071-40.71 μg·mL-1(r=0.999 9), 20.16-201.6 μg·mL-1(r=0.999 9), 4.730-47.30 μg·mL-1(r=0.999 9), 4.536-45.36 μg·mL-1(r=0.999 8), 1.817-18.17 μg·mL-1(r=0.999 9), 2.266-22.66 μg·mL-1(r=0.999 7), 3.321-33.21 μg·mL-1(r=0.999 9), 3.462-34.62 μg·mL-1(r=0.999 6), 2.111-21.11 μg·mL-1(r=0.999 9) and 2.290-22.90 μg·mL-1(r=0.999 7), respectively. The average recoveries (n=6) were 101.3%(2.1%),103.0%(1.5%),100.9%(2.0%),101.1%(2.0%),98.4%(1.6%),102.2%(2.4%),98.2%(1.3%),97.8%(2.0%),99.0%(2.0%) and 96.4%(1.1%), respectively. The contents of the 10 components in 4 bacthes of Fufang Shuanghua tablets were in the range of 1.685-2.649 mg·g-1, 12.202-13.780 mg·g-1, 2.415-2.594 mg·g-1, 1.340-1.919 mg·g-1, 0.501-0.791 mg·g-1, 0.891-1.342 mg·g-1, 1.299-2.105 mg·g-1, 1.147-1.504 mg·g-1, 0.654-0.694 mg·g-1, 0.846-1.151 mg·g-1, respectively.

Conclusion:

This simple accurate reproducible method can be used for the quality control and evaluate of compound Shuanghua tablets.

目的:

采用高效液相色谱(HPLC)技术,建立同时测定复方双花片中新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、连翘苷、穿心莲内酯和脱水穿心莲内酯10个成分含量的方法。

方法:

复方双花片样品用50%甲醇水超声提取,采用Agilent Zorbax SB-C18(250 mm×4.6 mm,5μm)色谱柱分析,流动相为乙腈-0.15%磷酸水溶液,梯度洗脱,流速1.0 mL·min-1,柱温30 ℃,进样量10 μL,检测波长327 nm(检测新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸)和226 nm(检测连翘苷、穿心莲内酯、脱水穿心莲内酯)。

目的:

采用高效液相色谱(HPLC)技术,建立同时测定复方双花片中新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、连翘苷、穿心莲内酯和脱水穿心莲内酯10个成分含量的方法。

方法:

复方双花片样品用50%甲醇水超声提取,采用Agilent Zorbax SB-C18(250 mm×4.6 mm,5μm)色谱柱分析,流动相为乙腈-0.15%磷酸水溶液,梯度洗脱,流速1.0 mL·min-1,柱温30 ℃,进样量10 μL,检测波长327 nm(检测新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸)和226 nm(检测连翘苷、穿心莲内酯、脱水穿心莲内酯)。

结果:

新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、连翘苷、穿心莲内酯和脱水穿心莲内酯质量浓度分别在4.071~40.71 μg·mL-1r=0.999 9)、20.16~201.6 μg·mL-1r=0.999 9)、4.730~47.30 μg·mL-1r=0.999 9)、4.536~45.36 μg·mL-1r=0.999 8)、1.817~18.17 μg·mL-1r=0.999 9)、2.266~22.66 μg·mL-1r=0.999 7)、3.321~33.21 μg·mL-1r=0.999 9)、3.462~34.62 μg·mL-1r=0.999 6)、2.111~21.11 μg·mL-1r=0.999 9)和2.290~22.90 μg·mL-1r=0.999 7)与峰面积呈良好的线性关系;平均回收率(RSD)(n=6)分别为101.3%(2.1%)、103.0%(1.5%)、100.9%(2.0%)、101.1%(2.0%)、98.4%(1.6%)、102.2%(2.4%)、98.2%(1.3%)、97.8%(2.0%)、99.0%(2.0%)和96.4%(1.1%);4批样品中上述10个成分的含量分别为1.685~2.649 mg·g-1、12.202~13.780 mg·g-1、2.415~2.594 mg·g-1、1.340~1.919 mg·g-1、0.501~0.791 mg·g-1、0.891~1.342 mg·g-1、1.299~2.105 mg·g-1、1.147~1.504 mg·g-1、0.654~0.694 mg·g-1、0.846~1.151 mg·g-1

结论:

所建立的方法简单准确,可用于复方双花片的质量控制与评价。

Objective:

To establish an HPLC method for simultaneous determination of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, forsythiaside A, 3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid, forsythin, andrographalide and dehydroandrographalide in compound Shuanghua tablets.

Methods:

The samples were extracted with 50% methanol solution under ultrasonic condition, and were performed on Agilent Zoabax SB-C18 column (250 mm×4.6 mm, 5 μm) by gradient elution of acetonitrile -0.15% phosphoric acid solution at a flow rate of 1.0 mL·min-1. The column temperature was 30 ℃, the injection volume was 10 μL, and the detection wavelength were set at 327 nm (detecting neochlorogenic acid,chlorogenic acid, cryptochlorogenic acid, forsythiaside A,3,4-O-dicaffeoylquinic acid,3,5-O-dicaffeoylquinic acid and 4,5-O-dicaffeoylquinic acid) and 226 nm (detecting forsythin, andrographalide and dehydroandrographalide).

Results:

The linear ranges of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid,forsythiaside A,3,4-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid, forsythin, andrographalide and dehydroandrographalide were 4.071-40.71 μg·mL-1(r=0.999 9), 20.16-201.6 μg·mL-1(r=0.999 9), 4.730-47.30 μg·mL-1(r=0.999 9), 4.536-45.36 μg·mL-1(r=0.999 8), 1.817-18.17 μg·mL-1(r=0.999 9), 2.266-22.66 μg·mL-1(r=0.999 7), 3.321-33.21 μg·mL-1(r=0.999 9), 3.462-34.62 μg·mL-1(r=0.999 6), 2.111-21.11 μg·mL-1(r=0.999 9) and 2.290-22.90 μg·mL-1(r=0.999 7), respectively. The average recoveries (n=6) were 101.3%(2.1%),103.0%(1.5%),100.9%(2.0%),101.1%(2.0%),98.4%(1.6%),102.2%(2.4%),98.2%(1.3%),97.8%(2.0%),99.0%(2.0%) and 96.4%(1.1%), respectively. The contents of the 10 components in 4 bacthes of Fufang Shuanghua tablets were in the range of 1.685-2.649 mg·g-1, 12.202-13.780 mg·g-1, 2.415-2.594 mg·g-1, 1.340-1.919 mg·g-1, 0.501-0.791 mg·g-1, 0.891-1.342 mg·g-1, 1.299-2.105 mg·g-1, 1.147-1.504 mg·g-1, 0.654-0.694 mg·g-1, 0.846-1.151 mg·g-1, respectively.

Conclusion:

This simple accurate reproducible method can be used for the quality control and evaluate of compound Shuanghua tablets.

复方双花片为国家食品药品监督管理局国家药品标准[WS3-178(Z-178)-2002(Z)]收录的制剂,由金银花、连翘、穿心莲、板蓝根4味药材加工而成,具有清热解毒,利咽消肿之功效,临床用于风热外感所致的感冒,症见发热、头痛、鼻塞流涕等[1]。方中金银花、连翘清热解毒、消肿散结,共为君药,金银花主要活性成分有绿原酸、咖啡酰奎宁酸等酚酸类[2-4],连翘中主要活性成分为连翘苷和连翘酯苷A[5-7];穿心莲清热解毒、凉血消肿,为臣药,主要活性成分为穿心莲内酯和脱水穿心莲内酯[8-10];板蓝根清热解毒、凉血利咽,为佐药,主要活性成分为(RS)-告依春[11-13],全方共奏清热解毒、消肿散结、利咽止痛之功效。
中药复方制剂组成复杂,其临床疗效往往是多个成分协同作用的结果。复方双花片现行质量标准仅将绿原酸作为含量测定的指标成分,质控指标单一,相关研究报道较少。杨帅等[14]采用HPLC法测定复方双花片中穿心莲内酯和脱水穿心内酯含量,任莹等[15]采用HPLC法测定复方双花片中绿原酸、咖啡酸等5个成分的含量,均不能全面反映该制剂的内在质量。本文采用HPLC法,同时测定该制剂中新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、异绿原酸B、异绿原酸A、异绿原酸C、连翘苷、穿心莲内酯和脱水穿心莲内酯10个成分的含量,以期为全面评价复方双花片的质量和后期质量标准的提高提供参考。
Agilent 1260型高效液相色谱仪,配置自动进样器、四元泵、柱温箱和二极管阵列检测器(DAD)(Agilent公司),XSE-205十万分之一电子天平(梅特勒公司),Milli-Q超纯水处理系统(Millipore公司),KQ-500DE超声清洗器(昆山超声仪器有限公司)。
对照品新绿原酸[批号(下同)DSTDX001504,含量(下同)98.80%]、隐绿原酸(DSTDY003501,98.85%)和3,4-二-O-咖啡酰奎宁酸(DSTDX003703,99.00%)购自德思特生物科技有限公司,绿原酸(110753-201817,96.8%)、连翘酯苷A(111810-202108,96.2%)、3,5-二-O-咖啡酰奎宁酸(111782-201807,94.3%)、4,5-二-O-咖啡酰奎宁酸(111894-202104,95.1%)、连翘苷(110821-201816,95.1%)、穿心莲内酯(110797-202010,99.6%)、脱水穿心莲内酯(110854-202111,98.9%)购自中国食品药品检定研究院。色谱纯乙腈和甲醇(Merck公司),分析纯甲酸、冰乙酸和磷酸(国药集团化学试剂有限公司),实验用水为超纯水。复方双花片(批号220702、220405、211201、220301,陕西康惠制药股份有限公司,0.62 g·片-1,有限期2年)。

分别精密称取对照品新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、连翘苷、穿心莲内酯和脱水穿心莲内酯8.24、20.83、9.57、9.43、7.34、9.61、13.97、7.28、8.48、9.26 mg,各置20 mL量瓶中,加50%甲醇水溶解并稀释至刻度,摇匀,制成各单一对照品储备液。分别依次量取对照品储备液1、2、1、1、0.5、0.5、0.5、1、0.5、0.5 mL,置同一10 mL量瓶中,振摇,加50%甲醇水定容,摇匀,即得质量浓度新绿原酸40.71 μg·mL-1、绿原酸201.6 μg·mL-1、隐绿原酸47.30 μg·mL-1、连翘酯苷A 45.36 μg·mL-1、3,4-二-O-咖啡酰奎宁酸18.17 μg·mL-1、3,5-二-O-咖啡酰奎宁酸22.66 μg·mL-1、4,5-二-O-咖啡酰奎宁酸33.21 μg·mL-1、连翘苷34.62 μg·mL-1、穿心莲内酯21.11 μg·mL-1和脱水穿心莲内酯22.90 μg·mL-1的混合对照品溶液。
取复方双花片适量,除去薄膜衣,研细,精密称取0.6 g,置锥形瓶中,精密加入50%甲醇水50 mL,称量,超声处理(功率500 W,频率45 kHz)20 min,放冷,再称量,用50%甲醇水补足减失的量,摇匀,过0.45 μm滤膜,即得。
按复方双花片处方比例和制备工艺,分别制成缺金银花、连翘、穿心莲的阴性样品,取适量按“2.2.2”项下方法制成各阴性样品溶液。
取“2.2”项下3种溶液适量,按“2.1”项下色谱条件进样分析,记录色谱图。结果,10个目标峰分离效果良好,分离度均>1.5,且阴性无干扰,方法专属性良好,见图1
分别精密吸取“2.2.1”项下混合对照品溶液1、2、4、6、8、10 mL,分别置10 mL量瓶中,加50%甲醇水定容,得6个系列浓度溶液,按“2.1”项下色谱条件进样测定,记录各成分峰面积。以质量浓度(X,μg·mL-1)为横坐标,峰面积(Y)为纵坐标进行线性回归,得回归方程和线性相关系数(r),结果见表1。另取混合对照品溶液适量,用50%甲醇水逐步稀释,按“2.1”项下色谱条件进样测定,以信噪比(S/N)为3∶1时的浓度为检测限(LOD),S/N为10∶1时的浓度为定量限(LOQ),结果见表1
取“2.2.1”项下混合对照品溶液适量,用50%甲醇水稀释2倍,按“2.1”项下色谱条件,连续进样6次,测定各成分的峰面积。结果新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、连翘苷、穿心莲内酯和脱水穿心莲内酯色谱峰峰面积的RSD(n=6)分别为0.16%、0.17%、0.15%、0.54%、0.20%、0.18%、0.13%、0.24%、0.28%和0.27%,表明仪器的精密度良好。
取批号220702样品制备的供试品溶液,分别在0、4、8、16、24、30 h依次进样,按“2.1”项下色谱条件测定各成分峰面积。结果新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、连翘苷、穿心莲内酯和脱水穿心莲内酯峰面积的RSD分别为0.53%、0.93%、1.2%、1.2%、1.2%、1.2%、0.51%、0.79%、1.7%和0.70%,表明供试品溶液在30 h内稳定性良好。
取批号220702的样品适量,除去薄膜衣,按“2.2.2”项下的方法制备6份供试品溶液,进样10 μL,测定峰面积。结果新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、连翘苷、穿心莲内酯和脱水穿心莲内酯含量的平均值分别为2.697、13.68、2.645、1.940、0.575 7、1.199、1.556、1.541、0.697 7、0.853 8 mg·g-1,RSD(n=6)分别为0.84%、1.3%、1.5%、1.5%、1.4%、1.7%、1.3%、2.0%、2.2%、1.4%,表明该方法重复性良好。
取含量已知的同一批次(批号220702)复方双花片样品,除去薄膜衣,研细,精密称取0.3 g,共6份。按1∶1的比例精密加入各对照品溶液(0.407 1 mg·mL-1的新绿原酸2 mL、1.034 mg·mL-1的绿原酸4 mL、0.401 3 mg·mL-1的隐绿原酸2 mL、0.392 9 mg·mL-1的连翘酯苷A 1.5 mL、0.350 0 mg·mL-1的3,4-二-O-咖啡酰奎宁酸0.5 mL、0.375 3 mg·mL-1的3,5-二-O-咖啡酰奎宁酸1 mL、0.468 6 mg·mL-1的4,5-二-O-咖啡酰奎宁酸1 mL、0.456 9 mg·mL-1的连翘苷1 mL、0.419 5 mg·mL-1的穿心莲内酯0.5 mL、0.249 6 mg·mL-1的脱水穿心莲内酯1 mL),加50%甲醇水补足至50 mL,按“2.2.2”项下方法制备供试溶液,再按“2.1”项下方法进样测定,记录各成分峰面积。采用外标双点校正法计算各成分含量,再计算平均回收率,结果见表2
取4批复方双花片样品,除去薄膜衣,按“2.2.2”项下方法每批分别制备2份供试品溶液,再按“2.1”项下色谱条件进样测定,记录峰面积,采用外标双点校正法计算绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸、连翘苷、穿心莲内酯和脱水穿心莲内酯的含量,结果见表3
中药复方制剂所含成分具有不同的最大吸收波长,而采用某一波长时,难以确保所测成分被检测出或获得最大吸收,从而影响定量结果。因此,本实验采用DAD检测器对混合对照品在200~400 nm波长范围内进行全波长扫描,并采集光谱图。结果新绿原酸、绿原酸、隐绿原酸最大吸波长为326 nm,连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸在最大吸收波长为328 nm,连翘苷、穿心莲内酯、脱水穿心莲内酯分别在228、226、252 nm有最大吸收,而脱水穿心莲内酯在226 nm也有较大吸收。所以,本实验选择327 nm作为新连翘酯苷A、3,4-二-O-咖啡酰奎宁酸、3,5-二-O-咖啡酰奎宁酸、4,5-二-O-咖啡酰奎宁酸的检测波长,226 nm作为连翘苷、穿心莲内酯、脱水穿心莲内酯的检测波长。
考察了以甲醇-水、乙腈-水为流动相体系,发现10个成分峰形较差,当添加甲酸、冰乙酸、磷酸时,10个色谱峰峰形均有改善,以磷酸为最佳,当磷酸浓度为0.15%时,再增加磷酸浓度,10个成分色谱峰峰形无明显改善。考察了色谱柱Ⅰ[Waters symmetry-C18(4.6 mm×250 mm,5 μm)]、色谱柱Ⅱ[phenomenex Luna-C18(4.6 mm×250 mm,5 μm)]和色谱柱Ⅲ[Agilent Zorbax-C18(4.6 mm×250 mm,5 μm)]对10个待测成分分离的影响,以各成分与相邻色谱峰的分离度和峰形为判定指标,连翘酯苷A在其他色谱柱中与相邻色谱峰有重叠或分离较差现象,在色谱柱Ⅲ中分离效果为最好。
预试验首先考察了不同提取溶剂(乙醇、70%乙醇水、50%乙醇水、甲醇、70%甲醇水、50%甲醇水)对各成分提取的影响,发现以50%甲醇水为溶剂,各成分提取率为最高,且峰形为最佳。考察超声提取(考察时间10、20、30、40 min)和回流提取(30、40、50、60 min)2种提取方式,发现超声提取用时较少,且在20 min内各成分基本上提取完全,故确定为供试品溶液制备方法。
预试验还考察了复方双花片中佐药板蓝根活性成分(RS)-告依春作为含量测定的指标性成分,发现其含量较低,故未纳入本次试验含量测定中。从表3可知,4批样品中各成分比较稳定,色谱图中以4,5-二-O-咖啡酰奎宁酸色的峰面积适中,可以在今后研究中,以此峰为参考峰,计算其他各峰相对校正因子,采用一侧多评法测定复方双花片中10个成分含量,可以减少标准物质的使用和提高工作效率。本次研究因缺少验证仪器设备,故未开展相关研究,在今后工作中,将加强和院校合作,开展相关领域研究。
本实验建立了HPLC法同时测定复方双花片中10个成分的含量,能较全面地反映该制剂的质量。结果显示各成分含量范围波动较小,质量较为稳定,但只收集了4批样品,样本量偏少,尚不能制定含量控制限度,后续将继续收集样本,以期制定各成分的限量值。
