Objective:

To establish the fingerprint of Panax quinquefolius L. from Shandong by UPLC-PDA,and to simultaneously determine the contents of 16 ginsenosides.

Methods:

The chromatographic column was an Acquity UPLC BEH C18 column(50 mm×2.1 mm,1.7 μm),which was eluted with water-acetonitrile by gradient at a flow rate of 0.4 mL·min-1. The detection wavelength was 203 nm,the column temperature was 30 °C,and the injection volume was 2 μL. The Chinese Pharmacopoeia “Chinese Medicine Chromatography Fingerprint Similarity Evaluation System (2012 Edition)” was used for evaluation,and 42 batches of Panax quinquefolius L. from different habitats were compared with cluster analysis and principal component analysis.

Results:

The total content 16 ginsenosides in Panax quinquefolius L. from in Shandong were 19.73-58.07 mg·g-1,and the average value was (34.72±8.22) mg·g-1. The fingerprints of the ginsenosides in Panax quinquefolius L. from Shandong were established,and the similarities were above 0.90. And 10 common peaks constituted the characteristic peaks of Panax quinquefolius L.. Cluster analysis and principal component analysis showed that the contents of ginsenosides in Panax quinquefolius L. from Shandong were stable.

Conclusion:

The fingerprint of Panax quinquefolius L. established is highly characteristic,and the method issimple,which provides data support for the identification and quality control of Panax quinquefolius L..

目的:

利用超高效液相色谱-二极管阵列检测器(UPLC-PDA)法建立山东产西洋参皂苷类成分的指纹图谱,并同时测定16个单体皂苷的含量。

方法:

采用Acquity UPLC BEH C18(50 mm×2.1 mm,1.7 μm)色谱柱,以水-乙腈为流动相进行梯度洗脱,流速0.4 mL·min-1,检测波长203 nm,柱温30 ℃,进样量2 μL。运用《中国药典》“中药色谱指纹图谱相似度评价系统(2012版)”进行评价,结合聚类分析和主成分分析对42批不同产地的西洋参进行比较。

结果:

山东产西洋参中16个单体皂苷总量为19.73~58.07 mg·g-1,均值为(34.72±8.22 )mg·g-1。建立了西洋参皂苷类成分的指纹图谱,相似度均达到了0.90以上,确定了10个共有成分峰构成西洋参的特征峰,聚类分析和主成分分析表明该地区西洋参皂苷类成分具有较好的稳定性。

结论:

建立的西洋参药材指纹图谱特征性强,方法简便,为山东产西洋参的鉴定和质量控制提供数据支持。

目的:

利用超高效液相色谱-二极管阵列检测器(UPLC-PDA)法建立山东产西洋参皂苷类成分的指纹图谱,并同时测定16个单体皂苷的含量。

方法:

采用Acquity UPLC BEH C18(50 mm×2.1 mm,1.7 μm)色谱柱,以水-乙腈为流动相进行梯度洗脱,流速0.4 mL·min-1,检测波长203 nm,柱温30 ℃,进样量2 μL。运用《中国药典》“中药色谱指纹图谱相似度评价系统(2012版)”进行评价,结合聚类分析和主成分分析对42批不同产地的西洋参进行比较。

结果:

山东产西洋参中16个单体皂苷总量为19.73~58.07 mg·g-1,均值为(34.72±8.22 )mg·g-1。建立了西洋参皂苷类成分的指纹图谱,相似度均达到了0.90以上,确定了10个共有成分峰构成西洋参的特征峰,聚类分析和主成分分析表明该地区西洋参皂苷类成分具有较好的稳定性。

结论:

建立的西洋参药材指纹图谱特征性强,方法简便,为山东产西洋参的鉴定和质量控制提供数据支持。

Objective:

To establish the fingerprint of Panax quinquefolius L. from Shandong by UPLC-PDA,and to simultaneously determine the contents of 16 ginsenosides.

Methods:

The chromatographic column was an Acquity UPLC BEH C18 column(50 mm×2.1 mm,1.7 μm),which was eluted with water-acetonitrile by gradient at a flow rate of 0.4 mL·min-1. The detection wavelength was 203 nm,the column temperature was 30 °C,and the injection volume was 2 μL. The Chinese Pharmacopoeia “Chinese Medicine Chromatography Fingerprint Similarity Evaluation System (2012 Edition)” was used for evaluation,and 42 batches of Panax quinquefolius L. from different habitats were compared with cluster analysis and principal component analysis.

Results:

The total content 16 ginsenosides in Panax quinquefolius L. from in Shandong were 19.73-58.07 mg·g-1,and the average value was (34.72±8.22) mg·g-1. The fingerprints of the ginsenosides in Panax quinquefolius L. from Shandong were established,and the similarities were above 0.90. And 10 common peaks constituted the characteristic peaks of Panax quinquefolius L.. Cluster analysis and principal component analysis showed that the contents of ginsenosides in Panax quinquefolius L. from Shandong were stable.

Conclusion:

The fingerprint of Panax quinquefolius L. established is highly characteristic,and the method issimple,which provides data support for the identification and quality control of Panax quinquefolius L..

西洋参(Panax quinquefolius L.)又名花旗参、美国参,为五加科人参属多年生草本植物[1],在我国始载于《本草纲目拾遗》,清代《补图本草备要》中记载“西洋参,苦、甘、凉、味厚、气薄、补肺降火,生津液,除烦倦,虚而有火者相宜”。现代药理学研究表明,西洋参具有免疫调节[2]、抗抑郁[3]、抗肿瘤[4]、抗炎[5]、抗氧化[6]和降血脂[7]等多种药理作用。这些功能与西洋参中化学成分,尤其是生物活性显著的人参皂苷密切相关。人参皂苷是1类固醇类化合物,又称三萜皂苷,主要存在于人参属药材中,是人参属药材发挥药理作用的重要活性物质。人参皂苷Rb1、Rh1、Rh2、Rg1和Rg3可以诱导肿瘤细胞凋亡,抑制肿瘤细胞生长、增殖[8-9];人参皂苷Rb1、Rb2、Rb3、Rc、Rd、Rg3和Rh2具有抗氧化、抗心律失常的作用[10];人参皂苷Re、Rg1能调节神经中枢,促进RNA、DNA合成,人参皂苷Re抑制中枢神经,人参皂苷Rg1使中枢神经兴奋[11];人参皂苷Rg2具有抗休克作用,抑制心肌肥大,改善血液流变,可以用于治疗或改善心肌梗死,心律失常等心血管疾病[12]
自1975年我国西洋参引种成功以来,山东地区西洋参的生产和种植多为散户经营,缺乏系统化、规模化的管理,产品质量评价体系建设不完全,缺乏对于山东产西洋参化学成分的系统性研究,导致西洋参产品整体质量参差不齐。本研究对山东省威海市西洋参样品中皂苷类成分进行系统分析,为山东产西洋参的质量评价提供科学依据。
Acquity UPLC H-Class超高效液相色谱仪(PDA检测器,工作站Empower 3.0版本,Waters公司);752N紫外分光光度计(上海仪电分析有限公司);MS204S十万分之一电子分析天平(Mettler Toledo公司);EX 125 DZH万分之一电子天平(奥豪斯仪器有限公司);Milli-Q Advantage A10超纯水器(Millipore公司);WP-UP-WF-40微量分析型超纯水机(四川沃特尔水处理设备有限公司);DTC-8超声清洗机(湖北鼎泰恒胜超声波清洗设备有限公司);TGL-16G高速台式离心机(上海安亭科学仪器厂)。
本研究于2019年在山东省威海市12个乡镇23个村共42个采样点取样(样品编号为S1~S42),研究区域覆盖了山东省威海市主要的西洋参种植区域,占全省西洋参种植面积的90%以上。鲜西洋(采样地点见图1)参经吉林农业大学中药材学院李伟教授鉴定其为4年生五加科人参属植物西洋参Panax quinquefolius L.,42 ℃干燥粉碎,过筛(60目),备用。
对照品人参皂苷Rg1、Re、Rf、20(S)-Rh1、20(R)-Rh1、Rb1、Rc、F1、Rb2、Rb3、Rd、F2、20(S)-Rg3、20(R)-Rg3、20(S)-Rh2和20(R)-Rh2上海源叶生物科技有限公司,HPLC归一化法测定质量分数均≥98%。香兰素,上海源叶生物科技有限公司。甲醇、乙腈为色谱纯,Honeywell公司。正丁醇、甲醇、浓硫酸,来自北京化工厂。超纯水,由WP-UP-WF-40微量分析型超纯水机制备。
精密称取对照品人参皂苷Rg1、Re、Rf、20(S)-Rh1、20(R)-Rh1、Rb1、Rc、F1、Rb2、Rb3、Rd、F2、20(S)-Rg3、20(R)-Rg3、20(R)-Rh2和20(S)-Rh2 10 mg置于10 mL量瓶中,用80%甲醇定容至刻度,配制成质量浓度为1 mg·mL-1的混合对照品储备溶液。逐级稀释,得到质量浓度分别为1、10、20、50、100、200和500 μg·mL-1的系列混合对照品溶液,置于4 ℃的冰箱内,临用前经0.22 μm微孔滤膜滤过至进样瓶,即得。
分别取各西洋参样品约0.2 g,精密称量,置于5 mL离心管中,精密加入80%甲醇4 mL,称量,混匀,密封,室温下300 W,40 kHz超声提取30 min,取出,室温放冷,补足减失的量,以8 000 r·min-1离心5 min,取上清液,经0.22 μm微孔滤膜滤过,即得。
色谱柱:Acquity UPLC BEH C18柱(50 mm×2.1 mm,1.7 μm);流动相:水(A)-乙腈(B),梯度洗脱(0~6 min,13%B→22%B;6~16 min,22%B→38%B;16~18 min,38%B;22~23.5 min,40%B→45%B;23.5~24 min,45%B→58%B;24~30 min,58%B→62%B);流速:0.4 mL·min-1;进样量:2 μL;检测波长:203 nm;柱温:30 ℃。
取S1样品粉末,按“2.2”项下方法制备供试品溶液,按“2.3”项下色谱条件连续进样6次,记录指纹图谱,以人参皂苷Rb1色谱峰为参照峰,计算指纹图谱中各共有峰的相对保留时间和相对峰面积,结果各共有峰的相对保留时间和相对峰面积的RSD分别小于0.63%和1.1%。
取S1样品粉末6份,按“2.2”项下方法制备供试品溶液,按“2.3”项下色谱条件进行测定,记录指纹图谱,以人参皂苷Rb1色谱峰为参照峰,计算指纹图谱中各共有峰的相对保留时间和相对峰面积,结果各共有峰的相对保留时间和相对峰面积的RSD分别<3.1%和2.6%。
取S1样品粉末,按“2.2”项下方法制备供试品溶液,分别在供试品溶液制备后0、2、4、8、12、24 h按“2.3”项下色谱条件进行测定。以人参皂苷Rb1色谱峰为参照峰,结果表明各共有峰的相对保留时间和相对峰面积的RSD分别小于2.5%和小于1.8%。
取42批西洋参样品,分别按“2.2”项下方法制备供试品溶液,按“2.3”项下色谱条件进行测定,得到指纹图谱,再将得到的实验数据导入2012A版《中药色谱指纹图谱相似度评价系统》进行分析,设置S1为参考图谱,时间窗宽度设为0.2 min,采用多点校正后自动匹配指纹图谱,并以中位法生成对照指纹图谱(R),并对色谱峰进行指认。42批西洋参指纹图谱共检测到16个共有峰,经过与对照指纹图谱的比对,指认了其中10个色谱峰,分别为人参皂苷Re(2号峰)、20(S)-Rh1(3号峰)、Rb1(4号峰)、Rc(5号峰)、F1(6号峰)、Rb2(7号峰)、Rb3(8号峰)、Rd(9号峰)、20(S)-Rh2(12号峰)和20(R)-Rh2(13号峰),见图2。42批样品的相似度见表1,相似度均>0.900,表明42批西洋参的整体质量相对稳定,所建立指纹图谱的方法可用于西洋参的鉴定和质量控制。
精密吸取“2.1”项系列混合对照品溶液各2 μL,在“2.3”项色谱条件下测定,以对照品的峰面积(Y)对相应的质量浓度(X)进行线性回归,得回归方程,结果见表2。混合对照品溶液UPLC色谱图见图3
取20 μg·mL-1的混合对照品溶液,每次进样2 μL,连续进样6次,计算各成分峰面积的RSD,结果16个人参皂苷成分峰面积的RSD分别为1.2%、0.36%、0.15%、0.65%、0.14%、1.8%、1.6%、0.95%、1.0%、0.45%、0.24%、0.10%、0.11%、0.41%、0.67%和0.59%。
取6份S1样品粉末,精密称定,按“2.2”项下方法制备供试品溶液,分别进样2 μL,测定峰面积,计算12个人参皂苷类成分质量分数的RSD。结果12个人参皂苷Rg1、Re、20(S)-Rh1、Rb1、Rc、F1、Rb2、Rb3、Rd、F2、20(S)-Rh2、20(R)-Rh2的平均含量分别为1.15、10.17、0.37、14.02、2.25、0.22、0.54、0.94、1.85、0.22、0.07、1.07 mg·g-1,RSD分别为0.14%、1.8%、0.56%、2.6%、1.1%、0.80%、0.64%、2.1%、0.61%、1.1%、1.1%和0.39%。
取S1样品的供试品溶液,按“2.3”项下色谱条件分别于0、2、4、6、8、10、12和24 h进样2 μL,测定各成分峰面积,计算RSD。结果表明12个人参皂苷Rg1、Re、20(S)-Rh1、Rb1、Rc、F1、Rb2、Rb3、Rd、F2、20(S)-Rh2、20(R)-Rh2峰面积的RSD分别为0.74%、0.76%、0.46%、0.75%、0.24%、1.0%、0.44%、0.90%、0.16%、0.12%、0.57%和0.49%。
精密称取对照品人参皂苷Rg1、Re、20(S)-Rh1、Rb1、Rc、F1、Rb2、Rb3、Rd、F2、20(S)-Rh2、20(R)-Rh2各10 mg,分别置于10 mL量瓶中,80%甲醇定容至刻度线,配制成质量浓度为1 mg·mL-1的各对照品溶液。置于4 ℃的冰箱内,临用稀释即可。精密称取9份S1样品粉末各0.1 g,分别精密加入相当于样品中各待测成分含有量80%、100%和120%的各对照品溶液,按“2.2”项下方法制备供试品溶液,按“2.3”项下色谱条件进行测定,12个人参皂苷Rg1、Re、20(S)-Rh1、Rb1、Rc、F1、Rb2、Rb3、Rd、F2、20(S)-Rh2、20(R)-Rh2的平均加样回收率分别为99.1%、99.6%、102.9%、100.3%、99.2%、99.2%、100.7%、98.1%、98.6%、101.3%、99.6%和101.1%,RSD分别为0.54%、1.6%、0.22%、1.0%、0.64%、2.1%、1.6%、1.1%、0.91%、0.18%、0.67%和0.23%。
取42批西洋参样品,按“2.2”项下方法制备样品的供试品溶液,按“2.3”项下条件测定16个人参皂苷的含量,结果42批西洋参中人参皂苷Rg1、Re、20(S)-Rh1、Rb1、Rc、F1、Rb2、Rb3、Rd、F2、20(S)-Rh2和20(R)-Rh2的含量均值分别为(1.12±0.35)、(11.44±2.32)、(0.4±0.47)、(14.94±4.45)、(2.35±0.81)、(0.18±0.12)、(0.51±0.16)、(0.78±0.51)、(1.68±0.57)、(0.14±0.12)、(0.04±0.12)和(1.02±0.23) mg·g-1,42批西洋参中12个人参皂苷含量结果见图4
为了进一步比较评价42批样品的稳定性,采用化学计量学方法,以测得的12个成分的含量为变量,对42批西洋参进行分析。
采用SPSS23.0软件对42批西洋参中人参皂苷含量进行系统聚类分析,数据采用不转换方式,采用欧式距离和Ward linkage法[13-14],结果如图5所示。42批不同产地的样品以检出的12个人参皂苷含量作为聚类变量。当d=25时,可聚为2类:S14、S16~S26聚为1类;其余聚成1类。
主成分分析(principal component analysis,PCA)是常用的无监督分析方法,通过降低维度把相关性的指标转化为几个具有更高聚合度的主成分指标,可以最大限度地体现数据特征[15]。利用SPSS 26软件对西洋参的12个人参皂苷类成分进行PCA,得到12个成分的特征值和累计贡献率,见表3。根据特征值>1,提出的主成分有4个,其累积贡献率为80.492%,包含了42批西洋参指纹图谱的主要信息,可以有效地解释不同产地西洋参品质特性的总变异。载荷的绝对值越大,对主成分的贡献越大,矩阵结果见表4。人参皂苷Re、Rb1、Rc、F1、Rb2、Rd、F2在第1主成分中有明显的载荷值;人参皂苷20(S)-Rh1、F1在第2主成分中有明显的载荷值;人参皂苷20(S)-Rh1、20(R)-Rh2在第3主成分中有明显的载荷值;人参皂苷20(S)-Rh2、Rb3在第4主成分中有明显的载荷值。表明影响西洋参质量差异性的是多个成分,而不是单一成分。利用SIMCA14.1分析软件对其进行PCA,结果见图6。由PCA图可以看出,除S26和S41样品偏差较大,S14、S16~S24聚为1类;其余聚成1类,与聚类分析结果基本一致。由此说明,42批西洋参中皂苷类成分具有较好的稳定性,差异性不大。
本试验参照文献[16]的方法,考察了提取溶剂(甲醇、80%甲醇、60%甲醇、乙醇、80%乙醇、60%乙醇)、提取方法(加热回流法、超声提取法)、提取时间(30、60、90 min)、料液比(10:1、20:1、30:1)对西洋参药材提取效率的影响,发现加热回流法和超声提取法无显著性差异,以80%甲醇提取效率最高,且随提取时间和料液比的增加提取效率增加不显著,最终确定以80%甲醇作为提取溶剂,料液比10:1,超声(300 W,40 kHz)提取30 min。在指纹图谱的分析过程中,考察了流动相流速(0.3、0.4、0.5 mL·min-1)、不同有机相(乙腈、甲醇)对指纹图谱的影响,结果显示,流动相流速0.4 mL·min-1时,乙腈作为流动相获得的指纹图谱基线平稳,各个主要峰的分离度良好。在全波长扫描下,203 nm处色谱峰分离度良好,各峰吸收均匀,因此,最终选择203 nm作为检测波长。
42批西洋参中人参皂苷Rf、20(R)-Rh1、20(S)-Rg3、20(R)-Rg3在各样品中均未检出。西洋参中不含有人参皂苷Rf,其为人参的特有成分[17];人参皂苷20(R)-Rh1、20(S)-Rg3和20(R)-Rg3未检出,可能是其本身在西洋参中含量较低。检出的12个人参皂苷类成分含量之间具有一定差异性,其中以人参皂苷Rb1和Re含量最高,占皂苷总量的70%以上;人参皂苷Rc、Rd和Rg1和占皂苷总量的20%以上,其次依次为人参皂苷20(R)-Rh2、Rb3、Rb2、20(S)-Rh1、F1、F2、20(S)-Rh2。2020年版《中华人民共和国药典》(以下简称《中国药典》)对西洋参中人参皂苷Rg1、Re和Rb1的含量进行了规定,要求之和≥2.0%[1]。42批样品的不合格率为16.67%,不合格样品分别为S17、S18、S19、S21、S22、S23和S24。参考GB/T 36397-2018《西洋参分等质量》中对人参皂苷Rb1的含量要求≥2.0%,Rg1、Re之和≥0.6%。42批样品人参皂苷Rg1、Re之和均>0.6%,仅S28、S38、S40和S41样品中人参皂苷Rb1的含量>2.0%。42批中35批样品符合2020年版《中国药典》对人参皂苷含量的要求,但其人参皂苷Rb1的含量偏低。
2020年版《中国药典》规定西洋参一般为秋季采收,文献报道证明西洋参药材在秋季休眠期采收最佳,早则药势未成,晚则盛势已歇[18]。生长年限对西洋参中活性成分也有一定影响,李嘉欣等[19]对不同生长年限西洋参中皂苷含量研究表明,1~4年生西洋参中人参皂苷含量随生长年限增加而升高。西洋参的干燥温度越高干燥时间缩短,干燥温度>40 ℃时会引起人参皂苷损失,相较于低温度长时间干燥,高温度短时间干燥时西洋参中人参皂苷含量损失较低,因此一般采用较高温度(40~55 ℃)干燥。任何贮存条件都会引起西洋参中人参皂苷损失,贮存温度20 ℃时损失率在4.5%左右,当温度>30 ℃时,损失率达8%,温度越高损失率越高,在非冷藏储存成本较低的条件下人参皂苷的损失率较高是可以接受的。除以上因素外,产地是影响西洋参中活性成分的另一重要因素,陈军辉等[20]的研究表明,西洋参产地的经纬度、日照时间、无霜期、降水量、土壤特性等因素导致其活性成分含量呈现地域差异,不同产地甚至相同产地不同地点的西洋参中活性成分含量存在一定差异,即道地产地对西洋参活性成分含量存在较大影响。综上所述西洋参的质量差异性是由多因素、多条件、多成分共同作用的结果,并非单一因素或单一成分作用的结果。
本研究采用UPLC-PDA法对山东省42批西洋参中16个人参皂苷类成分进行测定,建立了西洋参UPLC指纹图谱,分析了其相似性,化学计量分析结果表明只有S26和S41这2个离群样品,其他40个样品的聚集均较好。结果表明山东地区的42批西洋参所含化学成分基本一致,所建立的指纹图谱可用于西洋参的鉴定与质量控制。
