Objective:

To evaluate the quality of the Japanese encephalitis inactivated vaccine strain adapted in human diploid cells (ZFB-3).

Methods:

The virus strain was identified by identification test to determine whether it was Japanese encephalitis virus strain,the virus titer was tested in mice brain to evaluate the adaptability and proliferation ability of the virus seed on human diploid cells (ZFB-3),the sterility test,mycoplasma test,and external virus factor test was conducted for any external contamination.

Results:

The virus strain was proved as Japanese encephalitis virus and had been well adapted to human diploid cells (ZFB-3) with high virus titer. The virus strain was free from contamination by bacteria,fungi,mycoplasma or exogenous viral factors.

Conclusion:

The quality of Japanese encephalitis inactivated vaccine strain adapted in human diploid cells (ZFB-3) meets the requirements and can be applied to vaccine production.

目的:

对人二倍体细胞(ZFB-3)乙型脑炎灭活疫苗毒种进行质量评价。

方法:

通过鉴别试验对毒种进行鉴别,以鉴别是否为乙脑病毒;通过脑内滴定法对毒种进行病毒滴度检测,以评估其在人二倍体细胞(ZFB-3)上的适应性和增殖能力;对毒种进行无菌检查、支原体检查、外源病毒因子检查,以检测是否有外源性污染。

结果:

证明毒种为乙脑病毒,且具有较高的病毒滴度,在人二倍体细胞(ZFB-3)上已较好地适应,且无细菌、真菌、支原体、外源病毒因子污染。

结论:

人二倍体细胞(ZFB-3)乙型脑炎灭活疫苗毒种质量符合要求,可应用于疫苗的生产。

目的:

对人二倍体细胞(ZFB-3)乙型脑炎灭活疫苗毒种进行质量评价。

方法:

通过鉴别试验对毒种进行鉴别,以鉴别是否为乙脑病毒;通过脑内滴定法对毒种进行病毒滴度检测,以评估其在人二倍体细胞(ZFB-3)上的适应性和增殖能力;对毒种进行无菌检查、支原体检查、外源病毒因子检查,以检测是否有外源性污染。

结果:

证明毒种为乙脑病毒,且具有较高的病毒滴度,在人二倍体细胞(ZFB-3)上已较好地适应,且无细菌、真菌、支原体、外源病毒因子污染。

结论:

人二倍体细胞(ZFB-3)乙型脑炎灭活疫苗毒种质量符合要求,可应用于疫苗的生产。

Objective:

To evaluate the quality of the Japanese encephalitis inactivated vaccine strain adapted in human diploid cells (ZFB-3).

Methods:

The virus strain was identified by identification test to determine whether it was Japanese encephalitis virus strain,the virus titer was tested in mice brain to evaluate the adaptability and proliferation ability of the virus seed on human diploid cells (ZFB-3),the sterility test,mycoplasma test,and external virus factor test was conducted for any external contamination.

Results:

The virus strain was proved as Japanese encephalitis virus and had been well adapted to human diploid cells (ZFB-3) with high virus titer. The virus strain was free from contamination by bacteria,fungi,mycoplasma or exogenous viral factors.

Conclusion:

The quality of Japanese encephalitis inactivated vaccine strain adapted in human diploid cells (ZFB-3) meets the requirements and can be applied to vaccine production.

流行性乙型脑炎(简称乙脑)是由乙脑病毒引起的一种急性神经系统传染病,每年全球报道病例约68 000例,病死率约为30%。幸存者中30%~50%会出现严重的神经系统和精神方面后遗症[1]。随着我国乙脑疫苗的大规模接种和疾病防控手段的提高,近些年乙脑的发病率不断下降,2022年发病率降至约千万分之一。
市场上的乙脑疫苗有3种:乙脑减毒活疫苗(SA14-14-2)、乙脑灭活疫苗和乙脑黄热嵌合疫苗。目前使用的乙脑减毒活疫苗为原代地鼠肾细胞生产,乙脑灭活疫苗多为Vero细胞生产,国内外的使用证明二者均具有很好的安全性和有效性[2-15]
近年来,除Vero细胞乙脑疫苗外,人二倍体细胞乙脑疫苗也开始研发。人二倍体细胞是指正常人体组织在体外培养的细胞,其来源于正常人胚胎,在传代过程中保持二倍体核型。ZFB-3细胞属于人二倍体细胞系,为安徽智飞龙科马生物制药有限公司自建的人胚肺成纤维细胞。本文对该公司生产的乙脑灭活疫苗毒种进行质量评价研究。
人二倍体细胞(ZFB-3)乙脑灭活疫苗毒种为安徽智飞龙科马生物制药有限公司生产。
攻击用乙脑病毒强毒株P3株由中国食品药品检定研究院(简称中检院)保存,金黄色葡萄球菌购自北京三药科技开发公司,乙脑阳性血清参考品(批号:250008-201404)由中检院保存。
Vero细胞、ZFB-3细胞、MRC-5细胞由中检院保存。SPF级昆明(KM)小鼠,性别不限,体质量7~9 g,购自中检院动物繁育中心。
乙脑抗体阴性小牛血清购自杭州四季青生物工程材料有限公司,甲基纤维素覆盖物购自Sigma公司。DMEM培养基购自Gibco公司,硫乙醇酸盐培养基和胰酪大豆胨液体培养基、支原体肉汤及半流体培养基、精氨酸支原体肉汤及半流体培养基购自北京三药科技有限公司,二苯甲酰胺荧光染料购自Sigma公司,Thermo 3111二氧化碳培养箱购自热电公司,OLYMPUS CKX53倒置显微镜购自奥林巴斯公司,NUAIR-NU-437-600S生物安全柜购自纽艾尔公司,Eppendorf 5810R离心机购自艾本德公司,BS-31 55L水浴摇床购自杰奥特公司。
用含2%牛血清的DMEM培养基将乙脑疫苗毒种做10倍系列稀释,取10-4~10-8稀释度的病毒液与乙脑抗体阴性小牛血清等量混合为对照组,10-1~10-5稀释度的病毒液与乙脑特异性免疫血清等量混合为试验组,于37 ℃水浴90 min。试验组和对照组每稀释度分别接种体质量为7~9 g昆明小鼠6只,每只脑内注射0.03 mL,观察14 d判定结果,3 d内死亡者不计。
结果显示,阳性血清中和后毒种脑内注射小鼠后均存活。计算乙型脑炎灭活疫苗毒种病毒中和指数为≥64 565,结果远高于标准规定>500。具体结果如表1
用含2%牛血清的DMEM培养基将乙脑疫苗毒种做10倍系列稀释。将体质量为7~9 g昆明小鼠分为5组,每组5只。取10-5~10-9稀释度病毒液脑内接种小鼠,每稀释度注射5只,每只0.03 mL,观察14、3 d内死亡者不计。Reed-Muench法计算LD50。结果测得乙型脑炎灭活疫苗毒种病毒滴度为7.7 lgLD50·mL-1。具体情况见表2
分别取装量为15 mL的硫乙醇酸盐培养基2支和装量为10 mL的胰酪大豆胨液体培养基1支,每支培养基接种供试品1.5 mL,取1支硫乙醇酸盐培养基置30~35 ℃培养14 d,另1支硫乙醇酸盐培养基和1支胰酪大豆胨液体培养基置20~25 ℃培养14 d。同时取硫乙醇酸盐培养基2支,分别置30~35 ℃和20~25 ℃培养14 d,取胰酪大豆胨液体培养基1支置20~25 ℃培养14 d作为阴性对照。以1份硫乙醇酸盐培养基培养14 d后的样品以金黄色葡萄球菌为阳性对照菌。结果接种毒种的培养基培养14 d后无菌生长,阳性对照培养第1 d即有菌生长。
培养法:分别取4支支原体肉汤培养基和精氨酸支原体肉汤培养基,再分别取2支相应的半流体培养基,每支培养基接种乙脑疫苗毒种0.5 mL。另取4种培养基各1支作为阴性对照。置于(36±1)℃培养21 d。于接种后的第7 d分别取2支首次接种毒种的培养基进行次代培养,每支培养基分别转种至相应的支原体半流体培养基及支原体肉汤培养基各2支,置于(36±1)℃培养21 d。
DNA染色法:Vero细胞消化后,以2×105细胞/瓶接种于25 cm2细胞培养瓶中,培养过夜。在1瓶中加入乙脑特异性抗血清中和后的毒种1 mL,在另1瓶加入无抗生素培养液为阴性对照。接种后的细胞瓶置于37 ℃,5% CO2培养箱中培养3~5 d。将培养到期后的细胞培养物刮下,制成细胞悬液。培养到期前1 d,将Vero细胞消化,稀释至细胞浓度为4×104 cells·mL-1,以0.5 mL每孔接种6孔培养板,每孔补加2.5 mL无抗生素培养基,于37 ℃,5% CO2培养箱中培养过夜。取1 mL制备的细胞悬液加至相应标记的孔中,同时设置支原体阳性对照孔,将板置于37 ℃,5% CO2培养箱中培养3~5 d。培养结束后,吸出板中液体,加入固定液5 mL,放置5 min,吸出固定液,再加入5 mL固定液固定10 min,吸出固定液,使盖玻片在空气中干燥。每孔中加入二苯甲酰胺荧光染料工作液5 mL,加盖,室温染色30 min,吸出染液,用5 mL蒸馏水洗涤3次,吸出水,使盖玻片在空气中干燥。取载玻体做成封片。用荧光显微镜观察结果。
结果显示,支原体检查培养法和指示细胞培养法(DNA染色法)中,阳性对照均为支原体阳性,而阴性对照和毒种均为支原体阴性,证明毒种无支原体污染。
采用动物试验法和细胞培养法进行病毒外源因子检查。
动物试验法:取乙脑疫苗毒种5 mL加入 乙脑特异性免疫血清溶液5 mL,混匀,37 ℃水浴90 min。吸取中和后样品,分别对15~20 g小鼠和出生后24 h内乳鼠进行腹腔和脑内注射,各注射10只,小鼠每只腹腔注射0.5 mL,脑内注射0.03 mL;乳鼠每只腹腔注射0.1 mL,脑内注射0.01 mL;小鼠观察21 d,乳鼠观察14 d。解剖所有在24 h后死亡或有患病体征的小鼠或乳鼠,直接肉眼观察其病理改变,并取有病变的相应组织和脑、脾制备成悬液,通过脑内和腹腔接种另外至少5只15~20 g小鼠或出生后24 h内乳鼠,小鼠观察21 d,乳鼠观察14 d。接种24 h内小鼠或乳鼠死亡超过20%,试验无效。
细胞培养法:(1)非血吸附病毒检查:分别取长满单层的ZFB-3细胞、MRC-5细胞、Vero细胞各6个T25细胞瓶,弃去细胞培养液,每瓶加入细胞维持液3.5 mL,接种中和后的供试品1.5 mL,同时取ZFB-3细胞、MRC-5细胞、Vero细胞各2瓶,每瓶加入细胞维持液5 mL,作为阴性对照。置35 ℃、5% CO2培养箱培养。接种后连续观察14 d,接种第7 d更换细胞维持液1次。(2)血吸附病毒检查:接种后第7 d和第14 d,分别取每种细胞培养物2瓶进行血吸附检查。用0.25%鸡和豚鼠红细胞混合悬液2 mL覆盖于细胞表面,1瓶于2~8 ℃放置30 min,另1瓶于20~25 ℃放置30 min,吸弃多余红细胞后显微镜下观察红细胞吸附情况。
结果显示,外源病毒因子检查动物试验法中,所有接种小鼠观察21 d均健存,所有乳鼠观察14 d均健存;细胞培养法中,接种中和后毒种的细胞培养物均未见细胞病变,未见血吸附情况。
国内生产的乙脑灭活疫苗毒种均为乙脑病毒P3株,P3株于1949年分离自北京患者的脑组织,后经鼠脑传代至53代,即P3株-鼠脑53代(P3-Smb53),其免疫原性较好,抗原谱较广[16]。因其之前一直在鼠脑传代,若使用其在细胞基质上生产疫苗,需首先进行适应。只有较好地适应增殖后,才能用于疫苗生产。本研究评价的毒种为P3株在人二倍体细胞ZFB-3上的适应株。
乙脑灭活疫苗毒种的滴度检测采用的方法为脑内毒力滴定。因小鼠对乙脑病毒非常敏感,因此在对乙脑病毒研究之初,即使用小鼠来评价乙脑病毒毒力和病毒滴度。本研究发现,人二倍体细胞ZFB-3乙脑病毒毒种滴度较高,证明乙脑病毒在ZFB-3细胞上已较好适应,但病毒滴度略低于乙脑病毒P3株在Vero细胞上的传代适应株,因此在疫苗生产中产量可能略低于Vero细胞乙脑灭活疫苗。
鉴别试验是用于鉴别毒种是否为乙脑病毒的一种方法,研究采用特异性的乙脑中和血清将乙脑病毒中和后,比较中和前后的病毒滴度来计算中和指数。若病毒为乙脑病毒,未中和病毒的滴度远高于中和后病毒的滴度,中和指数较高;若病毒非乙脑病毒,则乙脑特异性血清难以将其中和,因而中和后病毒的滴度与未中和病毒的滴度相差不大,则中和指数较低。因此可判断毒种是否为乙脑病毒毒种。本研究中毒种的鉴别试验中和指数符合规定,但为进一步保证毒株的正确性,可考虑在药典中增加序列测定对毒种进行质量控制。但目前绝大多数企业都在内控中对毒种的序列进行了测定。
无菌检查用于评价毒种是否被细菌或真菌污染。支原体检查用于评价毒种是否被支原体污染。毒种二者均为阴性,证明毒种无细菌、真菌和支原体污染。
外源病毒因子是用于评价毒种是否被外源的病毒因子污染。研究采用动物试验法和细胞培养法进行了外源病毒因子的检测,结果均符合规定。证明毒种相关的外源的病毒因子污染。
本研究对乙脑灭活疫苗P3毒种在人二倍体细胞ZFB-3上的适应株进行了质量评价,研究发现,其鉴别试验中和指数符合规定,证明毒种为乙脑病毒。毒种病毒滴度较高,在ZFB-3细胞上适应较好,毒种无细菌、真菌、支原体及外源病毒因子污染,可应用于乙脑灭活疫苗生产。
