Objective:

To establish a quality consistency assessment method to evaluate the consistency of product quality of Liuwei Dihuang concentrated pills (LDCP) among different manufacturers.

Methods:

Firstly,high performance liquid chromatography (HPLC) was used to determine the content of the six index components in LDCP,and analyze the content differences between different batches of the same manufacturer and the current product quality of different manufacturers. Secondly,quality consistency parameters,i.e.,intra-batch content consistency differences (PA),inter-batch content consistency differences (PB),and fingerprint similarity (PC),were constructed to assess the consistency of product quality among the different manufacturers. And lastly,the consistency parameters were taken as the variables and subjected to the principal component analysis (PCA) to classify the consistency of the LDCP samples of the seven manufacturers to be fitted and differentiated.

Results:

The contents of the six index components in thirty-five batches of LDCP samples from seven manufacturers totaled 1.48-2.99 mg per pill,the RSDs of the contents of different components were 4.9%-29.7%,and the consistency parameters of the seven products were 4.2%-15.1% for PA,26.4%-49.5% for PB,and 92.9%-98.2% for PC. There were some differences in the homogeneity of contents in samples from different manufacturers,and the contents of the product varied significantly between batches,with P value of 64.5-75.8,indicating that the difference in the consistency of samples from different manufacturers was relatively small. But under certain conditions,the seven manufacturers can be classified into three categories,with B and J as a category,Z and R as a category,and X,F,and S as a category.

Conclusion:

This study provides a simple and effective method for monitoring and distinguishing the quality consistency of commercially available LDCP products,and the experimental results can provide a reference for the sample quality homogeneity of LDCP manufacturers.

目的:

建立质量一致性评价方法评价不同厂家间六味地黄浓缩丸质量一致性。

方法:

首先采用高效液相色谱测定六味地黄浓缩丸中6个指标成分含量,分析同厂家不同批次和不同厂家产品的含量差异;其次构建质量一致性参数即批内含量一致性差异(PA)、批间含量一致性差(PB)和指纹图谱相似率(PC),以评估不同厂家产品质量均一性;最后以一致性参数为变量,运用主成分分析(PCA)对7个厂家的六味地黄浓缩丸样品的一致性进行拟合与区分。

结果:

7个厂家的35批次六味地黄浓缩丸样品的6个指标成分含量总计为1.48~2.99 mg·丸-1,不同成分含量RSD为4.9%~29.7%,7家产品的PA为4.2%~15.1%,PB为26.4%~49.5%,PC为92.9%~98.2%,不同厂家间样品含量均一性存在一定差异,且产品的批次间含量差异值明显,P为64.5~75.8,表明不同厂家样品一致性差异较小,但在一定条件下,7个厂家样品可划分为三类,B、J为一类,Z、R为一类,X、F、S为一类。

结论:

本研究提供了一种简单有效的方法用于评估和区分市售六味地黄浓缩丸产品的质量一致性,可为六味地黄浓缩丸生产厂家的样品质量均一性评价提供参考。

目的:

建立质量一致性评价方法评价不同厂家间六味地黄浓缩丸质量一致性。

方法:

首先采用高效液相色谱测定六味地黄浓缩丸中6个指标成分含量,分析同厂家不同批次和不同厂家产品的含量差异;其次构建质量一致性参数即批内含量一致性差异(PA)、批间含量一致性差(PB)和指纹图谱相似率(PC),以评估不同厂家产品质量均一性;最后以一致性参数为变量,运用主成分分析(PCA)对7个厂家的六味地黄浓缩丸样品的一致性进行拟合与区分。

结果:

7个厂家的35批次六味地黄浓缩丸样品的6个指标成分含量总计为1.48~2.99 mg·丸-1,不同成分含量RSD为4.9%~29.7%,7家产品的PA为4.2%~15.1%,PB为26.4%~49.5%,PC为92.9%~98.2%,不同厂家间样品含量均一性存在一定差异,且产品的批次间含量差异值明显,P为64.5~75.8,表明不同厂家样品一致性差异较小,但在一定条件下,7个厂家样品可划分为三类,B、J为一类,Z、R为一类,X、F、S为一类。

结论:

本研究提供了一种简单有效的方法用于评估和区分市售六味地黄浓缩丸产品的质量一致性,可为六味地黄浓缩丸生产厂家的样品质量均一性评价提供参考。

Objective:

To establish a quality consistency assessment method to evaluate the consistency of product quality of Liuwei Dihuang concentrated pills (LDCP) among different manufacturers.

Methods:

Firstly,high performance liquid chromatography (HPLC) was used to determine the content of the six index components in LDCP,and analyze the content differences between different batches of the same manufacturer and the current product quality of different manufacturers. Secondly,quality consistency parameters,i.e.,intra-batch content consistency differences (PA),inter-batch content consistency differences (PB),and fingerprint similarity (PC),were constructed to assess the consistency of product quality among the different manufacturers. And lastly,the consistency parameters were taken as the variables and subjected to the principal component analysis (PCA) to classify the consistency of the LDCP samples of the seven manufacturers to be fitted and differentiated.

Results:

The contents of the six index components in thirty-five batches of LDCP samples from seven manufacturers totaled 1.48-2.99 mg per pill,the RSDs of the contents of different components were 4.9%-29.7%,and the consistency parameters of the seven products were 4.2%-15.1% for PA,26.4%-49.5% for PB,and 92.9%-98.2% for PC. There were some differences in the homogeneity of contents in samples from different manufacturers,and the contents of the product varied significantly between batches,with P value of 64.5-75.8,indicating that the difference in the consistency of samples from different manufacturers was relatively small. But under certain conditions,the seven manufacturers can be classified into three categories,with B and J as a category,Z and R as a category,and X,F,and S as a category.

Conclusion:

This study provides a simple and effective method for monitoring and distinguishing the quality consistency of commercially available LDCP products,and the experimental results can provide a reference for the sample quality homogeneity of LDCP manufacturers.

六味地黄丸是常用滋阴补肾类中成药,由熟地黄、酒萸肉、山药、茯苓、泽泻、牡丹皮六味中药组成,处方中熟地黄为君药,酒萸肉、山药均为具有补益作用的臣药,泽泻、牡丹皮、茯苓均为具有泻下作用的佐药,三补三泻六味合用达到滋阴补肾作用。六味地黄浓缩丸(Liuwei Dihuang concentrated pills,LDCP)具有携带和服用方便等特点,是目前市场上同系列产品中销售额最大的主流品种。
目前,对LDCP的质量差异研究有很多,如詹国平等[1]按照2015年版《中华人民共和国药典》(简称《中国药典》)规定项对不同厂家的产品质量进行分析,发现不同厂家样品均已达到国家标准所规定的基本要求,但丹皮酚、马钱苷含量上存在明显差异;夏昀卿等[2]将指纹图谱技术和“组分结构”理论联合使用模式对LDCP的质量进行控制与评价,发现不同厂家生产的各批次样品指纹图谱相似度、牡丹皮组分结构的构成要素的含量以及量比关系差别较大;冯霞霞等[3]对LDCP中马钱苷、莫诺苷和丹皮酚含量检测,对比不同厂家之间的样本间存在明显差异;张小博[4]按照2010年版《中华人民共和国药典》的质量标准对市售LDCP样品常规检查及测定含量考察其质量,发现7个厂家共25批样品的水分、溶散时限及含量均符合规定,但不同批次间的含量数值有较大波动、不同厂家间溶散时限差异较大;封亮[5]通过HPLC色谱法对药材与成品制剂比较,发现不同厂家成品之间存在一定质量差异,Ji等[6]建立LDCP的指纹图谱,结合化学计量学分析,发现多数厂家样品之间一致性较差,Bo等[7]建立五波长全融合指纹图谱和多组分定量相结合的方法将22批次LDCP质量一致性进行评价,这些研究方法对监控LDCP质量控制具有重要参考意义。然而,以往研究在样本收集时随机性较大,缺乏对同厂家产品的批次和批次间样品质量的细致考察,评价结果仅能给出品种质量差异大小的宏观概念[8],无法追溯厂家内部与厂家之间产品质量一致性的影响因素。
本研究建立了一种操作简便的对LDCP中6个指标成分定性定量的HPLC检测方法,在样本选择方面将成药质量与生产过程相关联,收集确定数量的各自厂家的相同批号(n1=5)、不同批号(n2=5)的样品,从批内一致性(PA)、批间一致性(PB)、指纹图谱相似率(PC)3个维度评估7个厂家35批次LDCP的质量一致性,揭示不同厂家产品质量差异,同时采用主成分分析方法对不同生产厂家样品进行质量一致性分类区分,实验结果为规范和促进该产品质量提升提供数据参考。
ME204T/02型万分之一天平(梅特勒-托利多仪器有限公司);XSR105/A型十万分之一天平(梅特勒-托利多公司);1260型高效液相色谱仪(Agilent公司,包括G1311B型四元梯度泵,G1329B型自动进样器,G1316A型柱温箱,G4212A型二极管阵列检测器);KQ-250DB超声波清洗仪(昆山市超声仪器有限公司)。
纯净水(广州屈臣氏食品饮料有限公司);色谱级乙腈(上海星可高纯溶剂有限公司);色谱级磷酸(北京迈瑞达科技有限公司);分析级甲醇(天津富宇精细化工有限公司)。
对照品没食子酸(BCY-000550,纯度≥98%),马钱苷(批号BCY-000532,纯度≥98%),莫诺苷(批号BCY-000536 ,纯度≥98%),芍药苷(批号BCY-000094,纯度≥98%),丹皮酚(批号BCY-000262 ,纯度≥98%)均购于江西佰草源生物科技有限公司,5-羟甲基糠醛(批号PS020460,纯度≥95%)购买于成都普思生物科技股份有限公司。LDCP分别来自7个不同生产厂家,详细信息见表1
分别取没食子酸、5-羟甲基糠醛、莫诺苷、马钱苷、芍药苷、丹皮酚的对照品适量,用50%甲醇溶解,分别配制成每1 mL含34.4、94.8、27.2、41.4、8.1、44.2 μg的混合对照品溶液,备用。
取本品数粒研成粉末,精密称定约0.5 g,置具塞锥形瓶中,精密加入50%甲醇25 mL,称量,超声(功率250 W,频率40 kHz)提取20 min,冷却至室温,用50%甲醇补足减失的量,摇匀,经0.45 μm微孔滤膜滤过,取续滤液,即得。
采用Waters HSS T3(250 mm×4.6 mm,5 μm)色谱柱,流动相为0.08%磷酸(A)-乙腈(B),梯度洗脱(0~3 min,95%A;3~12 min,95%A→90%A;12~25 min,90%A→78%A;25~30 min,78%A→72%A;30~32 min,72%A→50%A,32~40 min,50%A→15%A,40~45 min,15%A),流速为0.8 mL·min-1,柱温为40 ℃,检测波长为216、232、238、274、284 nm,进样体积为10 μL。238 nm检测波长下HPLC色谱图如图1
精密称取没食子酸11.57 mg、5-羟甲基糠醛15.62 mg、莫诺苷11.34 mg、马钱苷10.28 mg、芍药苷13.03 mg、丹皮酚16.17 mg于25 mL量瓶中,加入50%甲醇溶解并定容至刻度,精密量取上述混合对照品溶液1 mL,分别稀释1、2、4、8、16、32、64倍,滤过,按“2.1.3”项下色谱条件分别进样测定,以浓度为横坐标,峰面积为纵坐标,分别得到5个成分浓度与峰面积之间的线性回归方程,且在相应范围内各成分的回归系数均不小于0.999 1,表明5个成分的校准曲线具有良好的线性相关性,结果如表2所示。
取200705号样品,按“2.1.2”项方法制备供试品溶液,按“2.1.3”项下色谱条件,连续进样6次,结果没食子酸、5-羟甲基糠醛、莫诺苷、马钱苷、芍药苷、丹皮酚峰面积的RSD分别为0.10%、0.10%、0.10%、0.10%、1.9%、0.20%,表明仪器精密度良好。
选择200705样品的一供试品溶液分别在0、2、4、8、12、24 h时进样,计算没食子酸等6个成分峰面积的RSD,结果没食子酸、5-羟甲基糠醛、莫诺苷、马钱苷、芍药苷、丹皮酚的RSD分别为0.20%、1.3%、2.6%、2.4%、1.7%、0.10%,表明该供试品溶液24 h内稳定性良好。
取200705批号样品平行制备6份供试品,按“2.1.3”项下色谱条件,分别进样然后计算没食子酸等6个成分含量的RSD,结果显示没食子酸、5-羟甲基糠醛、莫诺苷、马钱苷、芍药苷、丹皮酚的RSD分别为0.50%、0.90%、1.3%、1.6%、1.7%、1.2%,结果表明该实验方法重复性良好。
精密称取没食子酸等6个成分的对照品适量,制成每1 mL含没食子酸399.2 μg、5-羟甲基糠醛554.8 μg、莫诺苷356.8 μg、马钱苷444.8 μg、芍药苷162.4 μg和丹皮酚495.2 μg的混合溶液。精密称取已知含量的200705号样品0.25 g,按“2.1.2”项方法制备供试品共6份。按1:1分别精密加入混合对照品溶液,按“2.1.3”项色谱条件测定,分别计算6个成分的加样回收率及RSD,6个成分的回收率结果介于99.4%~102.2%之间,结果见表3
35批次不同样品的6个指标成分总量范围为1.48~2.99 mg·丸-1(如表4所示),其中莫诺苷和马钱苷总计在0.37~1.18 mg·丸-1(莫诺苷0.14~0.71 mg·丸-1,马钱苷0.19~0.47 mg·丸-1),丹皮酚含量0.32~0.81 mg·丸-1,没食子酸含量在0.24~0.53 mg·丸-1,5-羟甲基糠醛含量在0.09~1.26 mg·丸-1,芍药苷含量在0.05~0.20 mg·丸-1。按照2020年版《中国药典》[10]要求,LDCP含莫诺苷和马钱苷的总量计,每丸不得少于0.37 mg、含丹皮酚每丸不得少于0.32 mg,则所有样品均为合格产品。另外,通过对比各厂家样品含量(),发现各成分含量存在不均一性,7个厂家的莫诺苷与马钱苷和含量的RSD为6.8%~23.2%、丹皮酚RSD为6.0%~19.5%、没食子酸的RSD为7.2%~20.8%、芍药苷的RSD为8.7%~29.7%,5-羟甲基糠醛的RSD为4.9%~26.6%。
将来自相同厂家的5个不同批号样品的相同成分含量的均值代表该厂家该成分的含量()水平(结果如表4图2所示),发现不同厂家间相同成分含量差异明显:5-羟甲基糠醛差异最大,Z厂家平均含量最高(1.02 mg·丸-1),S厂家平均含量最低(0.12 mg·丸-1),2个厂家相差8倍有余;芍药苷含量差异次之,Z厂家(0.17 mg·丸-1)与X厂家(0.05 mg·丸-1)两者含量差4倍,莫诺苷与马钱苷和的含量最高的F厂家(1.06 mg·丸-1)与最低的R厂家(0.44 mg·丸-1)差2.4倍;丹皮酚平均含量差约2倍,没食子酸平均含量差异最小(S厂家0.48 mg·丸-1,R厂家0.31 mg·丸-1,极值含量差1.5倍)。由此也体现出质量一致性控制的必要性。
为考察同一品种样品质量一致性结果,本实验从含量、指纹图谱2个方面进行评价分析,引用批内一致性差异值(PA)、批间一致性差异值(PB)、指纹图谱相似率(PC)3个参数。
PA n1 个样品的相同指标成分含量之间的一致性,PB n2 个样品的相同指标成分含量之间的一致性。测定并计算出多个样品相同成分的含量,该成分含量极差值与均值的百分比即为该成分的PA/B。(注:①PA/B=(含量极大值-含量极小值)/含量平均值×100%,②当所测指标为非单一成分时,一致性差异值(PA/B)等于各指标成分一致性差异值的均值[9])。
PC是同厂家不同批号样品之间的指纹图谱相似率,将相同厂家不同批号LDCP供试品按照“2.1.3”项色谱条件进样分析,选取238 nm波长下图谱作为指纹图谱,将各色谱图导入《中药色谱指纹图谱相似度评价系统》(2.0版),采用中位数法,时间窗宽度为0.1 min,经多点校正、全峰匹配后生成的图谱为参照图谱,计算各图谱与参照图谱的相似度,取最小值的百分值即为PC
以7个厂家的3个一致性参数为变量,采用SIMCA-P 13.0软件对7厂家以主成分特征值和方差累计贡献率进行PCA,以新提取的主成分为变量,统计7个厂家样品分类。
表5所示,PA在4.2%~15.1%,F、S、X厂家的PA值均小于10%,即相同批号不同包装单位的样品6个成分的含量相对极差较小,反映出在原料确定情况下该厂家的设备工艺稳定性良好,B、J、Z、R4个厂家PA>10%,相比于F、S、X厂家,这几个厂家需要提高制剂工艺稳定性;7个厂家PB在26.4%~49.5%,S、X厂家的PB值约小于30%,但B、J 2个厂家PB>40%,相比较可知B、J 2个厂家样品因制药原料质量、设备工艺影响因素表现出质量均一性较差;7个厂家Pc值在92.9%~98.2%之间,即所有厂家内部样品相似度较好,且相互之间差异较小。
基于定性定量一致性参数可知:批次间一致性较批次内一致性差异大,说明各生产厂家在设备工艺稳定性一定的前提下,药材原料对成品质量影响更大,即当前确保产品质量均一保证原料质量是关键;F、S、X 3个厂家样品PAPB值均较其余4个厂家小,且PC值均高于96%,表明这几个厂家生产设备工艺与原料药材相对稳定从而表现出样品质量一致性较好。
7厂家样品PCA分析,散点得分图如图3所示(2个主成分累计贡献率为93.8%),如图可见,7个厂家零散分布于3个坐标象限,B、J分布于第一象限且距离较近,说明这2个厂家样品质量相似,F、S、X 3个厂家分布于第二象限且互相之间距离较近,说明3个厂家间质量差异小,剩余Z、R 2个厂家分布于第四象限坐标轴处且与其他厂家距离较远,说明这几个厂家间存在一定质量差异。
为将以上7个厂家样品质量进行量化表征,在PCA模式下对新提取的主成分p1、主成分p2与3个一致性参数变量因子进行关联统计,2个主成分因子对3个变量参数的表达公式分别为p1=0.698PA +0.709PB+0.103PC和p2=-0.175PA+0.029PB+0.984PC,依据各厂家3个变量参数实际数值分别计算各厂家新的主成分因子p1、p2。2个主成分因子在坐标轴上的贡献率分别为0.602和0.336,根据其在坐标轴上的贡献率,设置一致性区分因子(P)计算公式,即P=0.602p1+0.336p2,参数统计如表6所示,根据P值,7个厂家可分为三类,B、J 2个厂家P>70,可分为一类,Z、R 2个厂家65<P<70,可以分为一类,X、F、S 3个厂家P值均近似等于65,可以分为一类;现有7个厂家样品P值相接近,但在一定条件下仍然可以分为3个不同种类,说明现有厂家间存在一定程度的质量差异。
药典规定LDCP以每丸含酒萸肉以莫诺苷和马钱苷的总量计,含牡丹皮以丹皮酚计,由测定结果发现,3个成分在符合药典要求底线前提下,表现出一定的差异性,这种结果也证实了建立一种区别于药典检测指标成分的含量检测方法,对于判别、区分同一品种不同厂家样品之间质量差异的必要性。本研究在药典规定的含量测定指标基础上另加5-羟甲基糠醛、没食子酸、芍药苷3个质控指标成分,且5-羟甲基糠醛为方中君药熟地黄加工过程产物,监控其含量可以评估熟地黄质量。有报道称5-羟甲基糠醛含量过高对人体内脏具有损害作用[11],对5-羟甲基糠醛进行合理控制非常重要;没食子酸是臣药酒萸肉的有效成分之一、芍药苷是牡丹皮中的有效成分之一,2个成分均具有抗菌、抗肝损伤作用[12-13],虽非药典指标,但对评价药品质量亦具有补充作用。
研究过程中对样品进行了包括提取溶剂、料液比、提取时间在内的前处理单因素变量考察,确定最优参数;样品指纹图谱检测波长选取238 nm,是因为该波长下各成分响应较好,有较好的峰形和分离度;采用含量一致性与整体相似性评价,既监督了定性成分,又兼顾了定量指标,定量与定性相结合,既能评价真伪,又能判断优劣[14]PAPBPC 3个参数在一定程度上反映生产工艺与原料对成药质量的影响[8],具有一定科学性。本研究构建产品一致性评价模型,实现从35批次样品的多指标成分含量结果,到7个生产厂家的批内、批间含量一致性及指纹图谱相似率参数,再到一致性区分因子的逐步代表性简化,最终以P值反映不同厂家样品质量一致性差异,实验结果对于区分药品质量差异具有参考意义。
中医药高质量发展的关键核心是建立科学合理的中药质量评价体系和高质量的评价标准,以实现对中医药产业的正向引导[15]。随着“健康中国”战略的实施,人民的健康保健意识不断提升,侧面带动了具有滋补功效类中药大品种如六味地黄丸等销售额的逐年攀升,其品质影响人民的健康,因此建立合适高效的分析评价方法区分相同品种的质量差异具有重要意义。本研究为市售LDCP产品质量一致性评价提供了参考实例和科学数据,期望能推进中成药质量提升,保障中成药的质量安全稳定。
